A practical dilution method for determining the degree of susceptibility of aquatic Vibrio and Aeromonas spp. to antibacterial agents is described. The method utilizes the differential ability of these two genera to ferment glucose. Acidification produced by the bacterial metabolism is demonstrated by the color change of an indicator (bromthymol blue) from blue-green to bright yellow. Where bacteria are inhibited by a sufficient concentration of the antibacterial drug tested, no color change occurs. The accurate quantitative results obtained with this method facilitate proper therapy for diseased fish, shrimps and oysters. The same method can in principle be applied to any other glucose-fermenting bacteria, when challenged with non-acidic antibacterial agents.  相似文献

Abstract. The minimum inhibitory concentrations (MICs) of 29 antimicrobial agents were determined for some representative Aeromonas salmonidda, Yersinia ruckeri and Vibrio isolates. Eleven of them, including three widely used in clinical practice, were further tested against 28–36 recent field isolates of A. salmonidda. The fluoroquinolones were inhibitory at very low concentrations with 50% of A. salmonidda isolates having a MIC of enro-floxacin ≤0·02μg ml-1. The distribution of MICs to the fluoroquinolones and to oxolinic acid were similar indicating possible cross resistance. Amoxycillin, florfenicol, minocycline, rifampicin and the potentiated sulphonamides had a single peak MIC distribution with values of ≤1·25μg ml-1. Minimum inhibitory concentrations of the fluoroquinolones were four or more times greater at 10°C compared with 22°C; this was not so with amoxycillin, florfenicol or the sulphachlorpyridazine-trimethoprim (S-TP) combination. A. salmonicida was rapidly killed at 22°C by the fluoroquinolones and S-TP whereas oxolinic acid and amoxycillin killed within 24h. Florfenicol was bacteriostatic. The therapeutic potential of some of these antibiotics is considered.  相似文献

Experiments were conducted to evaluate different prophylactic methods to control the bacterial load in brine shrimp, Artemia, hatching. The first experiment evaluated three treatments to control Vibrio spp. during the Artemia hatching: microalgae (Chaetoceros calcitrans), probiotic (Bacillus spp.), and antibiotic (Florfenicol). In the second experiment, Artemia metanauplius were enriched in distinct treatments with C. calcitrans, probiotic, and emulsion rich in docosahexaenoic and eicosapentaenoic fatty acids. Enriched Artemia metanauplius and nauplii (control) were offered to white shrimp, Litopenaeus vannamei, postlarvae (PL7–PL19). Presumptive Vibrio were quantified in Artemia, PL, and rearing water. Microalgae and probiotic were effective to control Vibrio spp. in Artemia nauplii. The enrichment process increased the Artemia bacterial load but did not affect Vibrio load in L. vannamei.  相似文献

对18株分离自患病乌鳢的气单胞菌进行了6大类共11种常用抗菌药物的耐药性研究。药敏试验结果显示,这18株菌对磺胺类药物--甲氧嘧啶、复方新诺明产生了高达100%的耐药性,对红霉素、阿米卡星、四环素也有一定的耐药率(50%~90%),而对喹诺酮类3种药物--恩诺沙星、环丙沙星、诺氟沙星则耐药率较低,并存在多重耐药的现象。通过对喹诺酮类、四环素类、氨基糖苷类、氯霉素类等4大类抗生素15种耐药基因进行检测,发现ant、qnr S、cat检出率较高,分别为50.0%、44.4%、38.9%,而qnr B、cat B基因则未检出。同种细菌不同分离株表现出不同的耐药性,结果表明,生产用药时有必要针对病原进行药敏试验,筛选敏感药物进行疾病防治。  相似文献

In Iceland the rearing temperature of salmonid fish, mostly Atlantic salmon, is generally below 10°C. Infections most often characterized by shallow skin lesions that may cause considerable mortality have been endemic in the country since rearing in salt water started. A variety of Vibrio spp. has been isolated from the diseased fish. In the present study, a total of 58 strains isolated from salmonid fish and 21 reference strains were subjected to a numerical taxonomy study. The results show that two phena are most common in the diseased fish; one of them includes V. marinus, whereas the other does not include a reference strain but has similarities to V. logei. Challenge tests showed that a representative strain of the phenon that included V. marinus is pathogenic for Atlantic salmon with an LD50 lower than 3.5 × 103.  相似文献

在18份鲍鱼养殖厂的各种水样和不同生长期的鲍样品中,共分离出17株弧菌:溶藻性弧菌11株,副溶血弧菌3株,霍乱弧菌2株,拟态弧菌1株。其中,溶藻性弧菌分布比较广,存在于各种样品;副溶血弧菌主要分布于鲍的肠道内;而非O1 O139群霍乱弧菌和拟态弧菌则分布于源头水中。鲍养殖水体中细菌总数的计数可以作为监测细菌性病害的指标之一。副溶血弧菌在鲍肠道内的高检出率提示鲍与副溶血弧菌的关系值得深入研究。  相似文献

杂色鲍养殖环境中致病性弧菌分布调查   总被引:3,自引:0,他引:3  
从深圳不同的杂色鲍人工养殖场采集并分类环境生物样品。应用TCBS平板分离并作生理生化初步鉴定弧菌,平板涂布法统计水样和各类生物每mL(或每g)所含弧菌总数,并应用基于16S~23SrDNA间区序列设计的4种水产病原弧菌特异性引物进行PCR,定性检测各类环境样品所携带致病性弧菌的分布状况。结果显示,弧菌广泛存在于杂色鲍养殖环境中,且杂色鲍养殖池池水中的弧菌密度大于进水口海水。在养殖环境生物中,不同环境生物中每克生物所携带的弧菌数差别很大,其中盘管虫、海蟑螂、等足类所携带的弧菌数最多,而海鞘携带的弧菌数最少。在常见的4种致病性弧菌的检测结果上,创伤弧菌和溶藻弧菌阳性率均为3.4%。通过研究弧菌在杂色鲍养殖环境中弧菌的分布特征,为杂色鲍养殖中流行性弧菌病的预防提供科学依据。  相似文献

Members of the genus Aeromonas are emerging human pathogens, causing a variety of extra‐intestinal, systemic and gastrointestinal infections in both immunocompetent and immunocompromised persons. Aeromonas virulence is multifaceted and involves surface‐associated molecules, motility, biologically active extracellular products and biofilm formation. Aeromonads, isolated from diverse freshwater fish species as well as sea water, were screened for biofilm formation, with varying physicochemical parameters including temperature, agitation and nutrient availability. Motility, cell surface characteristics (auto‐aggregation, hydrophobicity and S layer), and extracellular virulence factor production (haemolysis, proteolysis, DNase production) were also assessed to identify potential associations with the biofilm phenotype. Biofilm formation was influenced by environmental conditions, with isolates preferentially forming biofilms in nutrient‐rich media at 30 °C, although strong biofilm formation also occurred at 37 °C. Strong biofilm formation was observed for Aeromonas culicicola isolates following exposure to nutrient‐rich conditions, while Aeromonas allosaccharophila isolates preferred nutrient‐poor conditions for biofilm formation. Source‐/species‐specific correlations, ranging from weak to strong, were observed between biofilm formation and motility, cell surface characteristics and/or extracellular virulence factor production. Understanding the specific mechanisms by which Aeromonas species adhere to abiotic surfaces may aid in preventing and/or treating disease outbreaks in aquaculture systems and could lead to effective eradication of these fish pathogens.  相似文献

Aeromonas spp. are primary and opportunistic fish pathogens that are responsible for severe economic losses in aquaculture, and are exacerbated by their increasing incidence of antimicrobial resistance. Phytochemicals are being explored as alternatives to conventional antimicrobial agents since they have destressing, growth‐promoting, immune‐stimulating, and antimicrobial properties. The susceptibility of 93 aquatic Aeromonas spp. and six Plesiomonas shigelloides isolates (from cultured catfish, koi carp, tilapia, and seawater) to three phytochemicals, viz.: cinnamaldehyde (10–1250 µg/mL), vanillin (5–250 µg/mL), and crude Kigelia africana fruit extracts (4–10 mg/mL ethyl acetate, dichloromethane, methanol, and hexane) were assessed using the disk diffusion assay and compared to ampicillin and tetracycline, using activity indices. No zones of inhibition were obtained with 10 µg/mL of cinnamaldehyde, or with 5–250 µg/mL of vanillin. However, varying degrees of inhibition were observed with higher concentrations of cinnamaldehyde, as well as with the K. africana methanol extract. Cinnamaldehyde (≥500 µg/mL) and the K. africana methanol extract displayed better antimicrobial activity against study isolates in comparison to vanillin and ampicillin. They appear to be promising and sustainable phytochemicals that might be useful as alternatives to the antimicrobial agents currently in use in aquaculture.  相似文献

根据弧菌属细菌(Vibrio spp.)共有的rpo A基因序列,比对和设计了一对PCR引物,建立了能够快速而准确地检测弧菌属细菌的通用PCR检测方法,该方法对靶标DNA的检测灵敏度为58 fg/μL,对菌液的检测灵敏度为2.7×102cfu/m L,能够区分该属细菌与其它属的细菌,具有极高的特异性。使用建立的方法对分离自南美白对虾的病原菌进行了检验,并结合16S r DNA序列分析,结果显示待检测病原均为弧菌属的细菌,与测序结果一致,说明该检测方法可用于弧菌属细菌的检测和诊断。  相似文献

Juvenile Atlantic halibut (~100 mg, Hippoglossus hippoglossus) were exposed to Vibrio proteolyticus, a Vibrio spp. isolate, Photobacterium damselae ssp. damselae and five different isolates of Aeromonas salmonicida ssp. achromogenes via an hour‐long bath immersion to ascertain their variation in pathogenicity to this fish species. Results were analysed using Kaplan–Meier survival analysis. Analysis of the data from challenges using A. salmonicida ssp. achromogenes revealed three survival values of zero and a spread of values from 0 to 28.43. Challenges using a Vibrio spp isolate, Vproteolyticus and P. damselae resulted in Kaplan–Meier survival estimates of 31.21, 50.41 and 57.21, respectively. As all bacterial species tested could induce juvenile halibut mortalities, they must all be considered as potential pathogens. However, the degree of pathogenicity of A. salmonicida is isolate dependent.  相似文献

Abstract  Atlantic salmon, Salmo salar L., reared from two Baltic strains were released around the islands Bornholm and Møn in the Baltic Sea between 1995 and 1999. A total 600 000 reared salmon were released from net pens using the delayed release technique, keeping the salmon in net pens for approximately 3 months after smolting, and 208 000 were released directly from the hatchery. Of these, 15 958 were tagged with Carlin tags. Additionally, 65 300 coded wire tagged salmon were released as delayed release salmon close to Bornholm in 2000. Recaptures from the five years of Carlin tagged releases varied between 2.8% and 21.2% (average 13.1%). Most recaptures were from within the Baltic Sea (average 98%), but some were recaptured outside the Baltic Sea, either in the sea (1%) or in fresh water (1%). Recaptures outside the Baltic Sea and in fresh water were higher for releases at Møn in the western part of the Baltic, than releases at Bornholm. Straying rates from the releases into six rivers on the Swedish west coast were estimated using information from capture in traps and sport and broodstock fisheries. The proportion of straying salmon in rivers on the Swedish west coast was about 3.8% of the salmon run, but with large variations between rivers. Releases were discontinued because of possible deleterious effect on the local wild salmon populations.  相似文献
