在试管中对 4 5株产气荚膜梭菌A型菌 (Cl .PerfringensA型 )进行了初步筛选 ,用于制作仔猪肠毒血症疫苗的菌株。这 4 5株全部能产生致死性、坏死性、溶血性的α 毒素 ,其中 35 %可产生肠毒素 ,4 2 .5 %能形成芽孢。C5 7- 1株虽然不产生肠毒素 ,也不形成芽孢 ,但其毒性最强 ,在 1ml培养液中含有 10 0MLD。由于猪肠毒血症是由α 毒素引起的 ,与肠毒素无关 ,所以被初步选定为制作仔猪肠毒血症疫苗进行动物体内试验时的备用菌株 相似文献

1临床症状
肠毒血症多呈急性或最急性。犊牛出生后精神、食欲尚好,发病突然,多在生后2~10日龄犊牛,其特征是膨胀、脱水和排出红色粘性粪便;病程短,死亡快。 相似文献

为治疗产气荚膜梭菌感染引起的疾病,研究制备了产气荚膜梭菌多价高效抗毒素血清。试验采用C、D型产气荚膜梭菌标准菌株,制备了高浓度外毒素和灭活疫苗,作为免疫原多次免疫绵羊,通过间接ELISA法监测绵羊抗体水平变化,采用小鼠中和试验检验绵羊抗毒素血清保护效果。结果表明:制备的高效价抗C、D型产气荚膜梭菌毒素血清每0.1 m L血清能中和400个C型毒素对小鼠的MLD和600个D型毒素MLD,有良好的应用前景。 相似文献

In order to interpret the molecular structure and biological characteristics of Clostridium perfringens alpha-toxin (CPA), the CPA251-370 gene was cloned and the 120 amino acid carboxy terminal of CPA (CPA251-370) was obtained. The secondary and three-dimensional (3D) structures of CPA251-370 were predicted. The secondary structure of CPA251-370 consisted primarily of 35.48% β-sheets and 44.35% random coils. Compared with the CPA toxin consisting of 10 α-helices and eight β-sheets, the 3D structure of CPA251-370 only contained eight β-sheets. The circular dichroism (CD) spectrum detection showed that the CD spectrum of CPA251-370 changed slightly compared with the CD spectrum of CPA. Biological activity assays showed that CPA251-370 had lost the phospholipase C (PLC) activity and haemolytic activity of CPA. More importantly, the mice immunized with CPA251-370 were protected against a challenge with 1 MLD C. perfringens type A strain C57-1. This study laid a solid foundation for explaining the relationship between molecular structure and biological characteristics of CPA in the future. 无菌采取内蒙古通辽市某羊场病死羊肠道内容物、肝脏和肺脏,进行细菌的分离培养。将从十二指肠内分离到的1株疑似致病菌株进行生化试验、小鼠致病性试验,再将其通过魏氏梭菌多重PCR试验、魏氏梭菌ELISA试验及16S rRNA PCR试验进行鉴定。将PCR产物进行测序并进行了16S rRNA基因的进化树分析。结果显示,经细菌生化试验、多重PCR试验、ELISA试验和16S rRNA试验均证实此分离株为A型产气荚膜梭菌;进化树分析显示该菌与序列号为HQ808749.1(美国)的A型产气荚膜梭菌遗传距离最近。结果表明,该羊病例所分离的致病菌为A型产气荚膜梭菌。 相似文献

旨在探索产气荚膜梭菌外毒素造成机体炎性损伤及免疫调控紊乱的毒性机制,为产气荚膜梭菌病的致病机理研究提供理论基础。将C型产气荚膜梭菌强毒株C59-2培养上清腹腔注射BALB/c小鼠,采集小肠样本进行转录组测序,筛选差异表达基因,并对其进行GO功能注释和KEGG通路富集分析。结果显示,共获得40.99 Gb有效碱基,筛选后共得到795个差异表达基因,其中,229个基因表达上调,566个基因表达下调,对随机选取的10个基因进行q-PCR验证,其相对表达量与转录表达谱一致。GO功能注释主要涉及G蛋白偶联核苷酸受体活性和G蛋白偶联嘌呤核苷酸受体活性等。KEGG通路富集分析发现,主要富集在TNF信号通路、IL-17信号通路、p53信号通路、FOXO信号通路、Toll样受体信号通路、NF-κB信号通路等。产气荚膜梭菌外泌毒素侵入机体,会激活TNF等炎性信号通路,进而造成肠道发生炎性损伤甚至坏死。 相似文献
