本文采用内涂OV-1固定液的石英毛细管气相色谱柱和FID检测器,以磷酸三苯酯为内标物,对胺菊脂、苯醚氰菊酯和氯菊酯三种组分同时进行定量分析。本方法中胺菊酯、苯醚氰菊酯和氯菊酯变异系数分别为0.418%、0.964%和0.494%,平均回收率为100.43%、99.86%和100.03%,线性相关系数为0.9997、0.9997和0.9998。  相似文献

For testing the susceptibility of the head louse to insecticides impregnated bunches of polyamide fibres, whose diameter resembled that of human hair (0.1 mm) were used. In this manner a low mortality of control lice collected from infected persons was obtained (2.9% after 16 hours of exposure). Even after 25 years of DDT use no apparent resistance could be demonstrated in 25 tested louse populations and cross resistance to permethrin in 7 tested populations. The diagnostic concentration for resistance to pp'-DDT was established at 1%. Of the tested insecticides the highest toxicity was shown by malathion (LC50 = 0.000 018%), pirimiphos-methyl had on average a twice lower toxicity, trichlorphon 104 times lower, tetramethrin 1220 times lower, permethrin 2.5 times lower and pp'-DDT 630 times lower toxicity.  相似文献

BACKGROUND: Environmental pollutants, including metals, pesticides and other organics, pose serious risks to many aquatic organisms. The acute toxicities to zebrafish (Brachydanio rerio Hamilton & Buchanan) were determined for two organophosphorus insecticides, four pyrethroid insecticides and 50:50 binary mixtures. RESULTS: At 24, 48, 72 and 96 h after treatment, LC50 of permethrin, tetramethrin, bifenthrin, etofenprox, dichlorvos and phoxim to zebrafish were 0.0052–0.0025, 0.0782–0.0460, 0.0065–0.0032, 0.0969–0.0791, 51.3–13.0 and 1.28–0.469 mg L?1 respectively. LC50 of permethrin + dichlorvos, permethrin + phoxim, tetramethrin + dichlorvos, tetramethrin + phoxim, bifenthrin + dichlorvos, bifenthrin + phoxim, etofenprox + dichlorvos and etofenprox + phoxim were 0.0082–0.0046, 0.0078–0.0042, 0.264–0.124, 0.141–0.121, 0.0251–0.0154, 0.0154–0.0087, 0.396–0.217 and 0.213–0.0391 mg L?1. CONCLUSION: Toxicity levels of all pyrethroid insecticides to the zebrafish were high or very high. The organophosphate dichlorvos showed low toxicity, but phoxim showed high or intermediate toxicities to zebrafish, and the toxicities of binary mixtures of permethrin and dichlorvos or phoxim, bifenthrin and dichlorvos or phoxim and etofenprox and phoxim (48, 72 and 96 h exposure) were very high. The toxicities of binary mixtures of tetramethrin and dichlorvos or phoxim, etofenprox and dichlorvos and etofenprox and phoxim (24 h exposure) were high. Two substrains of Aedes aegypti, already resistant to DDT and pyrethroids, were further selected using either DDT or permethrin by mass exposure of the females only. DDT selection over 14 generations raised the resistance to DDT so far that no accurate LC50 values could be determined. Selection with permethrin raised the tolerance to an irregular plateau 7–10 times the original. DDT selection in the adults raised the DDT resistance of the larvae, but this could be partly overcome using a dehydrochlorinase inhibitor. The resistance to pyrethroids was increased but tolerance of dieldrin, malathion and propoxur compounds was little changed. Permethrin selection of the adults raised resistance to pyrethroids more than DDT selection but also increased DDT resistance. Similar patterns were found for the larval insects. A strain from Demerara in Guyana showed both DDT and pyrethroid resistance, including strong resistance to pyrethrins together with dieldrin and propoxur. It was concluded that two major independent resistance mechanisms existed in the selected strains, a dehydrochlorinase affecting DDT alone, and an unknown mechanism, probably nerve insensitivity (kdr) affecting both DDT and pyrethroids.  相似文献

Insecticide resistance in the bedbug Cimex hemipterus was investigated using 4211 bedbugs collected from three districts of Sri Lanka. Insecticide bioassays were carried out with discriminating dosages of deltamethrin, permethrin, DDT, malathion, and propoxur. Activity levels of insecticide metabolizing enzymes and the insecticide target site acetylcholinesterase were monitored using biochemical assays. Percentage survivals after DDT, malathion, and propoxur exposure were 41-88%, 18-64%, and 11-41%, respectively. For deltamethrin and permethrin, KT50/KT90 (time to knock-down 50%/90% of the population) values were 0.5-24/1.0-58 and 1.3-10/2.5-47 h, respectively. Both elevated esterase and malathion carboxylesterase mechanisms were present in bedbug populations. Monooxygenase levels were heterogeneous. Organophosphate and carbamate target site acetylcholinesterase, was insensitive in 29-44% of the populations. High DDT resistance was probably due to glutathione S-transferases. Malathion carboxylesterases are mainly responsible for high malathion resistance. High tolerance to both DDT and pyrethroids suggests the presence of ‘kdr’ type resistance mechanism in one population.  相似文献

The field strain of Anopheles stephensi, the main malaria vector in south of Iran, was colonized in laboratory and selected with DDT and dieldrin in two separate lines for 3 generations to a level of 19.5- and 14-fold for DDT and dieldrin resistance, respectively. Synergist tests with chlorofenethol (DMC) and piperonyl butoxide (PBO) on the selected strains indicated that dehydrochlorination and oxidative detoxification might be the underlying mechanisms involved in the resistance to dieldrin and DDT in selected strains. DDT selection decreased susceptibility to DDT and pyrethroids including lambdacyhalothrin, permethrin deltamethrin and cyfluthrin. The result also showed that selection with dieldrin caused negative and positive cross-resistance to pyrethroid and fipronil, respectively. Based on these results, it can be concluded that besides metabolic resistance mechanisms, other factors such as mutation in γ aminobutyric acid (GABA) and voltage-gated sodium channels (Kdr) might be involved.  相似文献

Insecticide resistance in the German cockroach can be mediated by a number of mechanisms, the most common being enhanced enzymatic metabolism. Seven field-collected strains of German cockroach, Blattella germanica (L.) with various levels of resistance to pyrethroids, five out of which were also cross-resistant to DDT were used in this study. The investigation of possible mechanisms responsible for permethrin resistance was carried out using the synergists PBO, DEF and DMC and biochemical assays, including general esterases, glutathione S-transeferases and monooxygenases assays, using an automated microtitre plate reader. PBO and DEF, the inhibitors of cytochrome p450 monooxygenases and general esterases, respectively, affected permethrin resistance to varying degrees depending on the strain. DDT resistance in five strains were not completely eliminated by the synergist DMC, an inhibitor of glutathione S-transferase enzymes, suggesting that a further non-metabolic resistance mechanism such as kdr-type may be present. This suggestion was further supported by GST assay data, where a little elevation in GST activity was detected in only two strains. The synergist data supported by biochemical assays implicated that cytochrome p450 monooxygenases or hydrolases are involved in permethrin resistance in some strains. However, these results implicated both enhanced oxidative and hydrolytic metabolism of permethrin as resistance mechanism in the other strains. The results of synergist and biochemical studies implicated that all the field-collected permethrin resistant strains have developed diverse mechanisms of resistance, although these strains have been collected from the same geographic area. The change in resistance ratios of some strains by using PBO or DEF is discussed. It is of interest to note that because resistance to permethrin was not completely eliminated by DEF and PBO, it is likely that one or more additional mechanisms are involved in permethrin resistance in every strain studied.  相似文献

The initiation of flight from loss of tarsal contact (flight reflex responses) and the tergotrochanteral muscle (TTM) responses evoked by brain stimulation were analyzed during carbofuran, permethrin, deltamethrin, and DDT poisoning in the house fly. Blockage of the flight reflex by LD50 doses of permethrin or deltamethrin was rapid, but the effects of DDT on the flight reflex took hours to develop. In addition, carbofuran treatment induced spontaneous flight in blocked preparations by an action in the central nervous system. This result suggests that pyrethroid blockage of the flight reflex was due to an action on sensory nerves, since the central flight program and its associated efferent systems were functionally intact. The relevance of this finding in terms of pyrethroid knockdown is discussed. The TTM response was unaffected by permethrin or deltamethrin both early and late in the poisoning process, possibly because the evoked TTM response does not involve peripheral sensory nerves, which seem to be important sites of pyrethroid action early in poisoning. Carbofuran induced repetitive firing and blockage of the TTM response within 1 hr, but normal responses were observed late in poisoning, which is consistent with the reversibility of carbamate inhibition of acetylcholinesterase. DDT caused no change in the evoked TTM response until bursts were recorded about 15 hr after treatment; this was another example of a slowly developing DDT effect. The protracted development of various DDT actions was concordant with a hypothesis of reduced efficacy at a proposed target site, viz., the sodium channels of nerve membranes.  相似文献

Synergists were used to diagnose possible mechanisms of permethrin resistance in permethrin-selected strains of the tobacco budworm, Heliothis virescens (F.). In addition to permethrin, these strains of the tobacco budworm were resistant to α-cyano-pyrethroid insecticides, organophosphorus insecticides and DDT. The monooxygenase-inhibiting prop-2-ynyl aryl ethers were the only effective synergists of permethrin among 16 candidates tested. The most effective synergist was 1,2,4-trichloro-3-(2-propynyloxy)benzene. Piperonyl butoxide, a common monooxygenase-inhibiting synergist in other species and tobacco budworm strains, was inactive. These results suggested the presence and contribution of an unusual monooxygenase in the enzymatic detoxication of permethrin. DDT cross-resistance, which was not synergized, and broad pyrethroid cross-resistance supported previous evidence for target site insensitivity as a second pyrethroid-resistance mechanism in these strains. The actions of S,S,S-tributyl phosphorotrithioate (TBPT) and triphenyl phosphate (TPP) suggested that hydrolytic detoxication, important in methyl parathion-resistance tobacco budworm strains, had little or no role in conferring pyrethroid resistance in these strains.  相似文献

A survey in SE England showed that seven out of 17 populations of glasshouse whitefly were resistant to DDT. Resistance to malathion occurred in 15 populations and to resmethrin in eight. Mecarbam and methiocarb gave good kills in screening tests but some insect growth regulators gave variable results. The synthetic pyrethroid permethrin was slightly more effective against a population of whitefly resistant to both DDT and malathion than against a susceptible population. Parthenogenetic females from a population susceptible to DDT and malathion produced almost entirely male progeny. An unsuccessful attempt was made to lower the resistance level of a DDT/malathion resistant strain by introducing these males to resistant females.  相似文献

Comparisons with standard susceptible insects showed that a strain of Tribolium castaneum, with a specific resistance to malathion and its carboxylic ester analogues, had no cross-resistance to topical applications of natural pyrethrins. Another strain of T. castaneum, showing resistance to many organophosphorus (OP) insecticides, was cross-resistant to pyrethrins ( × 34) and eight synthetic pyrethroids also applied topically; least cross-resistance occurred with resmethrin ( × 2.2), bioresmethrin ( × 3.3) and phenothrin ( × 4.0). Generally larger resistance factors were recorded with formulations synergised by piperonyl butoxide (PB). The greatest cross-resistance encountered was with unsynergised tetramethrin ( × 338). Apart from tetramethrin, factors of synergism did not exceed 5.7 with either the susceptible or multi-OP resistant strains. PB antagonised six of the nine pyrethroids against the multi-OP resistant strain. Antagonism also occurred with two of these six, permethrin (cis: trans ratio 1:3) and 5-prop-2-ynylfurfuryl ( 1RS)-cis,trans-chrysanthemate (‘Prothrin’), against the susceptible strain. Considering only formulations without the synergist, the most effective compounds against the susceptible strain, relative to pyrethrins, were bioresmethrin (2.7) and permethrin (2.4). Similarly with the multi-OP resistant strain the most effective compounds were bioresmethrin (28), resmethrin (14) and permethrin (6.6). Thus the LD50 (the dose required to kill 50% of the test species) for bioresmethrin against the resistant strain (0.14 μg) only slightly exceeded the LD50 for pyrethrins against the susceptible strain (0.12 μg).  相似文献

A sample of houseflies initially collected from a pig farm and found to be resistant to bendiocarb, DDT, gamma-HCH, pyrethrins + piperonyl butoxide (PB), tetrachlorvinphos and trichlorfon, was tested for resistance to knockdown by other toxicants. At the KD50 response level, resistance factors were obtained for: permethrin (× 141), deltamethrin (×96), bioresmethrin + PB (×37), resmethrin + PB (×33), fenitrothion (×94), bromophos (×58), iodofenphos (×42), pirimiphos-methyl (×30), dichlorvos (×22), dimethoate (×9), diazinon (×8), methomyl (×4) and methomyl + PB (×4). The slopes of the dose–response lines were lower for the farm strain than for a susceptible strain. This resulted in an increase of resistance factors at the KD95 level by an average of × 1.6. The houseflies on the farm could not be controlled using space sprays of pyrethrins + PB, although resistance to this toxicant was only ×12. Samples of 24 house fly (Musca domestica L.) populations were collected from animal farms in Hungary in 1990 and kept in the laboratory to determine their susceptibility to different types of insecticide: organochlorines, organophosphates, carbamates, pyrethroids, macrocyclic lactone and insect growth regulators. The adulticides were tested with topical bioassay in all 24 populations, the larvicides were studied with treated larval medium in 16 populations. The data were expressed as LD50 and LC50 values (ng fly ?1 and mg kg ?1 larval medium respectively). The percentages of populations which had resistance ratios > 10 at LD50 or LC50 were: 63% to DDT, 50% to methoxychlor, 13% to lindane, 83% to malathion, 63% to trichlorfon, 4% to propetamphos, 96% to dioxacarb, 46% to propoxur, 4% to methomyl, 13% to pyrethrum, 96% to bioresmethrin, 63% to permethrin, 58% to cypermethrin, 79% to SK-80, 79% to deltamethrin, 38% to invermectin, 0% to diflubenzuron, 0% to cyromazine. Correlation analysis showed a high degree of positive correlation among the adulticides except for ivermectin, bioresmethrin and SK-80. No cross-resistance was found between the larvicides and the conventional adulticides. Differences of insecticide resistance levels among the populations surveyed were studied by principal component and factor analysis. A fairly good relationship between resistance status and control practices used on farms was revealed. The populations originating from those farms where the application of adulticides had been frequent or regular and where high resistance was shown to most chemicals could be separated from the others.  相似文献

用FAO推荐的点滴法测定了上海、广州和江西等地小菜蛾(Plutella xylostetla L.)的抗药性。测定结果表明,上海和广州的小菜蛾(DBM)对现有的各类杀虫剂,如DDT、有机磷、氨基甲酸酯和拟除虫菊酯都发生了抗药性,其中以拟除虫菊酯的抗药性尤为突出,如溴氰菊酯(均>10414倍)、氰戊菊酯(2103和>3569倍)和氯菊酯(245和1533倍)。用浸渍法监测了上海地区小菜蛾对有机磷(1979-1987年)和拟除虫菊酯(1981-1987年)的扰药性。上海地区的DBM对敌敌畏、乐果和乙酰甲胺磷的抗性分别为8.1、10.1和16.6倍;从1981-1986年,DBM对氰戊菊酯,溴氰菊酯,氯菊酯、氯氰菊酯和氟氰戊菊酯的抗性分别为406.4、66.5、38.2、78.5和313.5倍,在1990年它们的抗性都有不同程度降低。各种解毒酶抑制剂TBPT,PB,TPP,DEM和DMC的活体测定表明,DBM的抗性涉及多功能氧化酶(MFO)、谷胱甘肽转移酶和酯酶,可能还涉及神经敏感度降低。  相似文献

Permethrin resistance in the human head louse, Pediculus capitis De Geer (Anopulura: Pediculidae), has been reported worldwide, is associated with the knockdown phenotype, and elicits cross-resistance to DDT and the pyrethrins. Two point mutations, T929I and L932F, in the voltage-sensitive sodium channel α-subunit gene are responsible for permethrin resistance as a resistant haplotype (kdr-like). We have optimized a serial invasive signal amplification reaction (SISAR) protocol for the detection of these mutations using PCR amplified DNA fragments. SISAR distinguished all genotypes with high accuracy in a head louse population from Texas that was heterogeneous in terms of permethrin sensitivity. Using SISAR, resistance-conferring mutations are detected in a high throughput format, facilitating the efficient monitoring of permethrin resistance allele frequency in field populations.  相似文献

The variation in tolerance to diflubenzuron [1-(4-chlorophenyl)-3-(2,6-difluorobenzoyl)urea] was examined in fourth instar larvae of seven strains of Aedes aegypti, some of which were resistant to DDT and permethrin. The difference between the least and the most tolerant to diflubenzuron was approximately two-fold. There was no correlation with resistance to the other insecticides. A DDT-resistant strain (T8) was selected 10 times (during 12 generations) with diflubenzuron. The LC50 to diflubenzuron had increased 3.3 times by the S8 generation but there was no further increase in later generations despite further selection. Associated with this increase, a marked decrease in resistance to DDT was observed but no change in permethrin tolerance. A genetically enriched strain (Hotchpotch) was synthesised from 35 strains of different geographic origin and crossed to the selected T8 strain before subsequent generations were selected five times with diflubenzuron. This procedure resulted in an 8 to 12-fold increase in the LC50 value over that for unselected T8, accompanied by a decrease in the slope of the log dose against probit mortality line.  相似文献

BACKGROUND: The Marin strain of Culex pipiens Say is a pyrethroid‐resistant population that was collected in Marin County, California, in 2001 and subsequently maintained in the laboratory under regular permethrin exposure. RESULTS: In this study, two cDNAs, CpGSTd1 and CpGSTd2, encoding glutathione S‐transferase (GST) were cloned from Cx. pipiens Marin. Phylogenetic analysis of the deduced amino acid sequences, CpGSTD1 and CpGSTD2, of these genes indicated that they belong to the Delta class of insect GSTs. The nucleotide and deduced amino acid sequences of CpGSTd1 and CpGSTd2 were 59 and 48% identical respectively. CpGSTD1 and CpGSTD2 were expressed in Escherichia coli and purified by affinity chromatography. The recombinant GSTs exhibited unique selectivity towards the general GST substrates 1‐chloro‐2,4‐dinitrobenzene (CDNB) and 1,2‐dichloro‐4‐nitrobenzene (DCNB), and also differed in their sensitivity to known inhibitors of GSTs. CpGSTD1 exhibited peroxidase activity with cumene hydroperoxide, while CpGSTD2 appeared to lack this activity. CpGSTD1 was able to metabolize 1,1,1‐trichloro‐2,2‐bis(4‐chlorophenyl)ethane (DDT), while DDT metabolism by CpGSTD2 was not detectable. CpGSTD1 and CpGSTD2 showed no detectable metabolism of permethrin. Gene expression of CpGSTd1 and CpGSTd2 in Marin mosquitoes was elevated about twofold in comparison with that found in a pyrethroid‐sensitive mosquito strain. CONCLUSION: The results indicate that CpGSTD1 and CpGSTD2 have unique biochemical characteristics, but they do not appear to play major roles in permethrin resistance in Marin mosquitoes. The cytochrome P450-dependent monooxygenases (P450s) are an important enzymatic system that metabolizes xenobiotics (e.g., pesticides), as well as endogenous compounds (e.g., hormones). P450-mediated metabolism can result in detoxification of insecticides such as pyrethroids, or can be involved in the bioactivation and detoxification of insecticides such as organophosphates. We isolated (from the JPAL strain) a permethrin resistant strain (ISOP450) of Culex pipiens quinquefasciatus, having 1300-fold permethrin resistance using standard backcrossing procedures. ISOP450 is highly related to the susceptible lab strain (SLAB) and the high resistance to permethrin is due solely to P450-mediated detoxification. This is the first time in mosquitoes that P450 monooxygenase involvement in pyrethroid resistance has been isolated and studied without the confounding effects of kdr. Resistance in ISOP450 is incompletely dominant (D = +0.3), autosomally linked, and monofactorally inherited. It is expressed in the larvae, but not in adults. Cross-resistance to pyrethroids lacking a 3-phenoxybenzyl moiety (tetramethrin, fenfluthrin, bioallethrin, and bifenthrin) ranged from 1.5- to 12-fold. ISOP450 had only limited (6.6- and 11-fold) cross-resistance to 3-phenoxybenzyl pyrethroids with an α-cyano group (cypermethrin and deltamethrin, respectively). Examination of cross-resistance patterns to organophosphate insecticides in ISOP450 showed an 8-fold resistance to fenitrothion, while low, but significant, levels of negative cross-resistance were found for malathion (RR = 0.84), temephos (RR = 0.73), and methyl-parathion (RR = 0.55). The importance and uniqueness of this P450 mechanism in insecticide resistance is discussed.  相似文献

In May 2001 a sample of Culex pipiens pipiens variety molestus Forskål from Marin County, California, collected as larvae and reared to adults, was found to show reduced resmethrin and permethrin knock‐down responses in bottle bioassays relative to a standard susceptible Cx pipiens quinquefasciatus Say colony (CQ1). Larval susceptibility tests, using CQ1 as standard susceptible, indicated that the Marin mosquitoes had LC50 resistance ratios of 18.3 for permethrin, 12 for deltamethrin and 3.3 for pyrethrum. A colony of Marin was established and rapidly developed higher levels of resistance in a few generations after exposure to permethrin as larvae. These selected larvae were shown to cross‐resist to lambda‐cyhalothrin as well as to DDT. However, adult knock‐down time in the presence of permethrin, resmethrin and pyrethrum was not increased after increase in tolerance to pyrethroids as larvae. Partial and almost complete reversion to susceptibility as larvae was achieved with S, S, S‐tributylphosphorotrithioate and piperonyl butoxide (PBO), respectively, suggesting the presence of carboxylesterase and P450 monooxygenase mediated resistance. Insensitive target site resistance (kdr) was also detected in some Marin mosquitoes by use of an existing PCR‐based diagnostic assay designed for Cx p pipiens L mosquitoes. Carboxylesterase mediated resistance was supported by use of newly synthesized novel pyrethroid‐selective substrates in activity assays. Bottle bioassays gave underestimates of the levels of tolerance to pyrethroids of Marin mosquitoes when compared with mortality rates in field trials using registered pyrethroid adulticides with and without PBO. The effects of organophosphorus insecticide trichlorfon, surfactant sodium dodecyl sulphate (SDS), and the mixture of trichlorfon and SDS on the antioxidant defense system and acetylcholinesterase (AChE) in Tilapia nilotica were assessed in vitro. Various concentrations of trichlorfon (0, 0.0001, 0.001, 0.01, 0.1 and 1 g/L) and SDS (0, 0.0625, 0.125, 0.25, 0.5, 1 g/L) were incubated with homogenate of liver and muscle, respectively, at 25 °C for 0, 30, 60 and 90 min. Two concentrations of mixture of trichlorfon and SDS (0.0001 g/L trichlorfon + 0.5 g/L SDS, 0.1 g/L trichlorfon + 0.5 g/L SDS) and 0.0001 g/L trichlorfon, 0.1 g/L trichlorfon, 0.5 g/L SDS and control, were incubated simultaneously with homogenate of liver and muscle, respectively, at 25 °C for 60 min. After incubation, the content of reduced-glutathione (GSH) and the activity of superoxide dismutase (SOD), catalase (CAT) and glutathione S-transferase (GST) in homogenate of liver were determined, and the activities of AChE in homogenate of muscle were also measured.Treatment with trichlorfon caused a significant concentration-dependent and time-related inhibition of AChE activity at all treatment concentrations and times since trichlorfon is a cholinesterase inhibitor. For the same trichlorfon treatment, an apparent decrease in GSH content was found in concentration of 0.01, 0.1, 1 g/L, whereas no significant alteration in antioxidant enzyme activity were found at all experiment concentrations and times, which might indicate that antioxidant enzymes have not involved in the metabolism of trichlorfon. The depletion of GSH might indicate that ROS could be involved in the toxic effects of trichlorfon. Exposure of SDS can inhibit activities of AChE, GST and CAT at concentrations of 0.5 and/or 1 g/L, which could be due to the denaturing process of SDS to the enzymes. For the mixture exposure of trichlorfon and SDS, the effect of the mixture of 0.0001 g/L trichlorfon and 0.5 g/L SDS on inhibition of AChE shows synergistic other than simple additive of trichlorfon and SDS. The combined effects of chemicals and detergents deserve to be particularly noted. It should be noted that the toxicity experiments were made in tissue homogenates instead of whole organisms. The responses against the toxic compounds will not be the same in both systems.  相似文献
