高羊茅光周期调控基因CONSTANS的克隆与分析   总被引:1,自引:1,他引:1  
本研究以坪用型"黔草1号"高羊茅为实验材料,基于多年生黑麦草CONSTANS(CO)基因序列设计引物,利用3'RACE和5'RACE方法扩增CO基因。结果表明,从高羊茅中分离出CO基因,命名为FaCON-STANS。经序列分析,cDNA全长1557bp,编码376个氨基酸,具有完整的开放阅读框(ORF,166~1296bp),氨基端具有B-box型锌指结构域,碳端具有CCT(CO,CO-like,TOC1)结构域。进一步的同源性分析表明,Fa-CONSTANS蛋白与禾本科植物的CO蛋白亲缘关系较近,与其它植物的亲缘关系较远。  相似文献

A field experiment was conducted on sandy loam acidic soil to study the effect of nutrient managements on light interception, photosynthesis, growth, biomass production and yield of Indian mustard [Brassica juncea (L.) Czern & Coss.]. Plant height, number of branches per plant, number of siliqua per plant, number of seeds per siliquae, 1000‐seed weight, seed and oil yield of Indian mustard improved at 100 % recommended rates of NPK (N‐P‐K at 80‐17.2‐33.2 kg ha?1) + 10 t ha?1 farmyard manure (FYM) (T3) compared with 100 % NPK rate (T2). It was also at par with 100 % NPK + 10 kg ha?1 borax + 20 kg ha?1 ZnSO4 (T6) and 50 % NPK + 10 t ha?1 FYM +10 kg ha?1 borax + 20 kg ha?1 ZnSO4 (T10). The rate of photosynthesis increased due to appropriate nutrient management treatments (T3, T6 or T10) with concomitant increase in photosynthetically active radiation, internal CO2 concentration and rate of transpiration and decrease in stomatal resistance. Consequent upon the higher rate of photosynthesis, dry‐matter accumulation increased. The crop receiving nutrient treatment T3 or T6 maintained higher light interception ratio (LIR), leaf area index (LAI), biomass production, crop growth rate (CGR) and net assimilation rate (NAR) that resulted in greater rate of photosynthesis, harvest index and seed yield. Similarly, T10 was equally efficient in registering greater LIR, LAI, CGR, NAR and seed yield of mustard. The average seed yields were 1692, 1683 and 1668 kg ha?1 in T3, T6 and T10, respectively, and these three treatments were significantly superior to T2 (1332 kg ha?1), control (723 kg ha?1) and other treatments. Significantly greater seed oil contents of 41.30, 40.60 and 41.07 % were recorded in T3, T6 and T10, respectively. Thus, significant improvement due to appropriate combination of NPK, FYM, borax and ZnSO4 was observed for uptake of nutrients.  相似文献

为合理利用多肽有机肥,采用田间试验方法研究了多肽有机肥对豇豆产量品质和土壤物理性质的影响。结果表明:在南海和增城2个试验点,多肽肥的施用均增加了豇豆的产量,降低了硝酸盐含量,提高了维生素C含量,而在白云区试验点,施用多肽肥对豇豆的生长作用并不明显;在3个试验点,多肽肥的施用对土壤比重的作用并不大;在白云区试验点,多肽肥与无机肥配合降低了土壤容重,土壤总孔隙度提高,但是在南海里水试验点,多肽肥与无机肥配合降低了土壤容重,但是土壤孔隙度并未提高,在增城试验点,多肽肥与无机肥配合并未降低土壤容重,总孔隙度并未增加。在3个试验点,施多肽肥均提高了土壤持水量。总之,多肽肥对土壤物理性质的提高效果优于鸡粪有机肥,且在砂质土壤上的效果优于粘土。  相似文献

A field experiment was conducted to evaluate the effect of long-term exogenous sucrose supply on aspects of corn plant physiology and development during the grain filling period. Concentrated sucrose solution was supplied to corn ( Zea mays L.) stems by an injection technique. This injection technique delivered pressurized solutions through syringe needles sealed to the stem with latex. The pressure was applied to the syringe plunger with ceramic construction bricks. Solutions containing sucrose at 0, 150, and 300 g L−1 were injected over a 32 day period encompassing the duration of the active grain filling period. The primary ears of plants injected with sucrose produced approximately 50% more kernels and 30% more grain weight than those injected with distilled water. The injected internode was also considerably heavier (56%) for plants receiving 300 g sucrose L−1 than plants receiving distilled water. For all measured variables, plants injected with distilled water were either not different from or had larger values than the non-injected controls. After one week sucrose injection caused photo-synthetic inhibition (as measured by chlorophyll fluorescence) in the leaf just above the ear and the ear leaf, and this was more severe for plants receiving 300 g sucrose L−1 than for those receiving 150 g sucrose L−1. For sucrose injected plants, the increased size of the primary ear was concomitant with a large decrease in grain production by the secondary ear and an overall decrease in per plant grain production. These results suggest that the mechanisms for signalling between sinks (the primary and secondary ears) and the primary sink and the source (leaves) are different.  相似文献

Increasing production of wheat from a limited water supply can result from efficient irrigation and nutrient management. A 3‐year field experiment was conducted at the Indian Institute of Soil Science, Bhopal, to study the growth, yield, seasonal evapotranspiration (ET) and water use efficiency (WUE), and the water–yield relationship of wheat in a soybean–wheat cropping system on vertisols. Three levels of irrigation, viz. I0, no post‐sowing irrigation; I1, two irrigations [crown root initiation (CRI) and flowering stage]; and I2, three irrigations (CRI, maximum tillering and flowering stage) and three nutrient management treatments, viz. F0, control (without fertilizer/manure); F1, 100 % NPK (100–21.5–24.9 kg ha?1); and F2, 100 % NPK + farmyard manure (FYM‐10 t ha?1) were tested in a split‐plot design with three replication. It has been established (through anova ) that the year effect was rather negligible and the interaction effects of irrigation and nutrient management on the growth parameters, ET, yield components, yield and WUE were significant. Plant height, progressive leaf area index, dry matter accumulation and crop growth rate were higher in I2F2, and I2F1 and I1F2 were statistically at par. The seasonal ET increased significantly with the increase in water supply in every nutrient treatment and it was highest in I2F2 and lowest in I0F0. The highest grain yield was obtained in I2F2; and a similar yield was recorded in I3F1 and I2F2. This shows a strong interaction effect between irrigation and nutrients. Yield components, viz. number of ears m?2, number of grains ear?1 and 1000‐grain weight were significant. The higher number of ears m?2 containing greater number of grains with relatively heavier weights appeared to have contributed to the higher yield in I1F2, I2F1 and I2F2. The highest WUE obtained in I0F2 did not correspond to the highest yield and maximum ET, but a WUE of 10.43 kg ha?1 mm?1 in the I2F2 combination corresponded with the highest yield and the seasonal ET requirement was 391.8, which was 137 % greater than the water use at maximum WUE. The ET–grain yield relationship was linear, with a lowest regression slope (i.e. marginal WUE) and elasticity of water production (Ewp) in F0 and a considerably higher slope and Ewp in F1 and F2. As the Ewp is positive and close to one in 100 % NPK treatment, the scope of improving WUE and yield with only inorganic fertilizer is very little, and relatively greater scope exists in the integrated management of organic manure and inorganic fertilizer. The results suggest that integrated nutrient management (100 % NPK + FYM) in conjunction with three irrigations maximized yield of wheat with concomitant improvement in ET and WUE under limited water availability.  相似文献

以苎麻[Boehmeria nivea (L.) Gaud.]栽培品种芦竹青为材料,经MseⅠ酶切并与相应接头连接,然后与生物素标记的探针(CT)15杂交,再用链亲和素包被磁珠(Dynabeads M-280)捕捉杂交产物,以21碱基接头寡核苷酸为引物经PCR扩增获得了双链目的片段,回收300~1 000 bp DNA片段,然后克隆到pMD18-T载体上,转化至DH5α中,这样就构建了苎麻富含(GA)n微卫星的部分基因组文库。随机挑取36个克隆,以锚定简单重复序列为引物对其进行PCR筛选。测序分析了22个阳性克隆,每个克隆都含有微卫星DNA,阳性克隆率为61.1%,微卫星种类以与探针互补的(GA/CT)n为主,占83.3%,同时还存在(TG/AC)n和三碱基、六碱基为单元构成的苎麻微卫星,如(GAC)n、(ACG)n、(TCT)n、(TCG)n、(GAA)n、(CCGACG) n、(GAGAAA)n等。最后以18个微卫星位点设计了18对苎麻微卫星特异引物。GA/CT重复单元的长度从7到40范围内变化,平均为19.2,显示了它们将产生良好多态性,为一种强有力的遗传标记。苎麻微卫星DNA标记可广泛应用于苎麻基因型鉴定,基因和QTL分析,分子标记辅助育种,系谱分析等。  相似文献

Accumulations of high molecular weight glutenin subunits (HMW‐GS) and glutenin macropolymer (GMP) in wheat grains are important indicators of grain quality. Two wheat cultivars, Yangmai 158 (shading tolerant) and Yangmai 11 (shading intolerant) which contains the same subunit pairs of 7 + 8 and 2 + 12, were used to evaluate the impacts of shading on HMW‐GS accumulation and GMP concentration in the grain. Three shading levels were implemented from jointing to maturity, i.e. S1, S2 and S3, in which the plants received 8 %, 15 % and 23 % less radiation of the control (S0), respectively. The initial formation of HMW‐GS was pre‐dated by shading. The rapid HMW‐GS accumulation duration was shortened, and the accumulation rate during late grain filling period was lowered in the two relatively severe shaded treatments (S2 & S3). Thus, the total HMW‐GS accumulation in single grain at maturity was lower in S2 and S3 than in the control (S0). However, concentrations of HMW‐GS and GMP at maturity increased because of the reduced single grain weight in S2 and S3, as compared to S0. In contrast, the low density shading (S1) prolonged the rapid accumulation duration of HMW‐GS, hence increased the accumulation and concentration of HMW‐GS in the grains. Consequently, S1 reduced falling number and SDS‐sedimentation volume, while shortened dough development time (DDT) and dough stability time (DST). In contrast, S2 and S3 increased falling number, wet‐gluten concentration and SDS‐sedimentation volume, and lengthened the DDT and DST. In addition, the fluctuations in accumulations of HMW‐GS and GMP and most quality traits because of shading in Yangmai 158 were less than Yangmai 11. The interrelations between HMW‐GS accumulation, GMP concentration and quality of grain and dough were further discussed.  相似文献

Summary Reactions of 13 grasses to Meloidogyne naasi varied with species; ryegrasses, fescues and their hybrids were generally susceptible and cocksfoot and timothy resistant. Marked variation in host resistance levels occurred between genotypes within cultivars.Selection of single plants, followed by tests on replicate tillers, identified resistant and susceptible genotypes in both Italian and perennial ryegrass cultivars. Resistant plants had few nematode-induced galls and fewer females and eggs than susceptibles. There was more or less continuous variation, with many genotypes intermediate between extremes of resistance and susceptibility. Selected resistant and susceptible genotypes are of use in assessing variation in nematode populations and as controls for breeding and selection programmes.  相似文献
