干扰素(IFN)具有抗病毒、抗肿瘤和免疫调节功能,具备较好的临床应用前景。为了解棘腹蛙干扰素的基因信息,本试验明确了棘腹蛙Ⅰ型IFN基因ORF编码长度为561 bp,编码的蛋白质含有1个信号肽和103aa的保守功能结构域。遗传进化分析显示,棘腹蛙Ⅰ型IFN进化相对保守;其蛋白质结构主要由α螺旋构成,细胞表面受体结合能力相对保守。将目的片段装入原核表达载体pET32a并转化BL21(DE3)进行蛋白表达与纯化,获得了预期蛋白。本研究证实了棘腹蛙Ⅰ型IFN属于相对保守的典型Ⅰ型干扰素;为后期深入挖掘棘腹蛙IFN的生物学功能、促进广谱抗病毒生物制剂研发提供了参考。 相似文献

IGF-Ⅰ能够促进细胞的增殖分化和代谢过程,而其表达是受STAT介导的,该通路最终受到生长激素(GH)的调控。生长激素与生长激素受体结合激活JAK家族,使得STAT家族成员,特别是STAT5磷酸化。磷酸化的STAT5进入细胞核内,与IGF-Ⅰ基因上的STAT5结合位点结合,激活胰岛素样生长因子-Ⅰ(IGF-Ⅰ)的表达,IGF-Ⅰ再通过血液循环到达机体的局部组织,促进组织细胞的生长和分化。作者对动物生长轴中STAT和IGF-Ⅰ的组成、功能以及STAT调控IGF-Ⅰ表达的分子机理的研究进展作一综述。 相似文献

本研究旨在探讨不同泌乳相关激素和生长因子对奶牛乳腺上皮细胞增殖的影响及其与细胞外基质主要成分层黏连蛋白的关系。将正常的荷斯坦泌乳期奶牛乳腺上皮细胞进行体外培养,在未包被或包被层黏连蛋白的条件下,以MTT法检测催乳素(PRL)、牛生长激素(GH)、类胰岛素生长因子-1(IGF-Ⅰ)、类胰岛素生长因子-2(IGF-Ⅱ)对细胞增殖作用的影响。在层黏连蛋白包被条件下,进行血清恢复的同时添加不同泌乳相关激素和生长因子,GH、IGF-Ⅰ有促进细胞增殖的作用(P<0.05),PRL、IGF-Ⅱ有维持细胞存活的作用(P<0.05);无血清时,几种激素和生长因子单独添加均无明显促增殖效应(P>0.05)。无基质条件下,与血清联合使用时,PRL、GH、IGF-Ⅰ、IGF-Ⅱ均对细胞生长有不同程度促进作用(P<0.05);无血清时,仅IGF-Ⅰ使细胞增殖速率显著加快(P<0.05)。层黏连蛋白作为培养基质对于体外培养的泌乳乳腺上皮细胞生长速度没有显著促进作用,但有利于PRL和IGF-Ⅱ发挥促存活作用。PRL、GH、IGF-Ⅰ、IGF-Ⅱ对细胞增殖和存活有促进作用,但需要与血清中其他成分协同才能充分发挥作用,其中IGF-Ⅰ促增殖能力最强。 相似文献

The long-term prognosis after surgical resection of canine insulinoma is poor. Signs of hypoglycaemia often recur soon after surgery because tumour tissue has only been resected partially and/or functional (micro-)metastases were present. Using quantitative real-time PCR, the expression of 16 target genes was compared between primary canine insulinomas and their corresponding metastases. There was significantly higher expression of genes encoding for growth hormone (GH) and insulin-like growth factor-1 (IGF-1) in metastases compared to their primary tumours. Immunohistochemical examination of proteins of the GH-IGF-1 axis revealed expression of GH, IGF-1 and GH receptor (GHR) in both primary insulinomas and metastases. Immunohistochemical staining for IGF-1 was significantly higher in metastases compared to primary tumours. 相似文献

本试验旨在探讨饲粮不同非纤维性碳水化合物和中性洗涤纤维比(NFC/NDF)条件下,诱导奶山羊发生亚急性瘤胃酸中毒(SARA)过程中瘤胃上皮细胞胰岛素样生长因子Ⅰ(IGF-Ⅰ)及胰岛素样生长因子Ⅰ受体(IGF-ⅠR)的基因表达量的变化。选用12只泌乳期关中奶山羊为试验动物,试验分4期进行,每期15 d,依次饲喂NFC/NDF分别为1.02(Ⅰ期)、1.24(Ⅱ期)、1.63(Ⅲ期)、2.58(Ⅳ期)的4种饲粮,以逐渐增加饲粮精料的方式诱导奶山羊发生SARA,并采用实时定量PCR(qRT-PCR)法对瘤胃上皮细胞中IGF-Ⅰ及IGF-ⅠR的基因表达量的变化进行相对定量分析。结果表明:随着饲粮NFC/NDF的升高,IGF-Ⅰ及IGF-ⅠR的基因表达量均出现不同程度的增加,Ⅱ期、Ⅲ期和Ⅳ期IGF-Ⅰ的基因表达量分别是Ⅰ期的1.70、2.71和9.61倍,IGF-ⅠR的基因表达量分别是Ⅰ期的1.88、3.09和10.19倍。饲粮NFC/NDF为2.58(即SARA期)时,与Ⅰ期相比,IGF-Ⅰ和IGF-ⅠR的基因表达量均出现极显著增加(P<0.01)。结果提示,以提高饲粮NFC/NDF的方法逐渐诱导SARA,IGF-Ⅰ及IGF-ⅠR的基因表达量显著提高,SARA发生后,它们的表达量有大幅增加。 相似文献

Serum concentrations of growth hormone (GH) and insulin-like growth factor-1 (IGF-1) were determined in 5 calves in the same lineage with growth retardation. They had normal appetites, activities, body proportion, and laboratory test results. Calves with growth retardation had higher serum GH concentrations and lower serum IGF-I concentrations. 应用Real-time PCR方法检测不同浓度(0、1、10和100 ng/mL)胰岛素样生长因子-Ⅰ(IGF-Ⅰ)处理后的体外培养奶牛乳腺上皮细胞乳蛋白和脂肪合成相关基因mRNA的相对表达量。结果表明,添加不同浓度IGF-Ⅰ对体外培养的奶牛乳腺上皮细胞IGF-Ⅰ受体基因(IGFIR)、IGF-Ⅰ结合蛋白-3基因(IGFBP3)、α-s1-酪蛋白基因(CSN1S1)和κ-酪蛋白基因(CSN3)mRNA的相对表达丰度均无显著影响(P>0.05)。随着IGF-Ⅰ添加浓度的增加,β-酪蛋白基因(CSN2)、乙酰辅酶A羧化酶基因(ACACA)、脂肪酸合成酶基因(FASN)和脂肪酸结合蛋白-3基因(FABP3)mRNA的相对表达丰度显著上调(P<0.05)。提示,IGF-Ⅰ作为一种重要细胞因子参与调节乳腺上皮细胞乳蛋白和乳脂肪相关基因mRNA的表达。 相似文献

The somatotropic axis plays a key role in the co-ordination of protein and energy metabolism during postnatal growth. This review discusses the complexity of the regulation of protein and energy metabolism by the somatotropic axis using three main examples: reduced nutrition, growth hormone (GH) treatment and insulin-like growth factor-1 (IGF-1) treatment. Decreased nutrition leads to elevated GH secretion, but it reduces hepatic GH receptor (GHR) number and plasma levels of IGF-1; it also changes the relative concentrations of IGF binding proteins (IGFBPs) in plasma. GH treatment improves the partitioning of nutrients by increasing protein synthesis and decreasing protein degradation and by modifying carbohydrate and lipid metabolism. However, these well-established metabolic responses to GH can change markedly in conditions of reduced nutritional supply or metabolic stress. Short-term infusion of IGF-1 in lambs reduces protein breakdown and increases protein synthesis. However, long-term IGF-1 administration in yearling sheep does not alter body weight gain or carcass composition. The lack of effect of IGF-1 treatment can be explained by activation of feedback mechanisms within the somatotropic axis, which lead to a reduction in GH secretion and hepatic GHR levels. The somatotropic axis has multiple levels of hormone action, with complex feedback and control mechanisms, from gene expression to regulation of mature peptide action. Given that GH has a much wider range of biologic functions than previously recognized, advances in research of the somatotropic axis will improve our understanding of the normal growth process and metabolic disorders. 相似文献

为研究玉米-豆粕型日粮中赖氨酸水平对70~90日龄生长肉兔生产性能、血清类胰岛素生长激素浓度及其基因表达的影响。选择100只体重接近、前期饲养相同的70日龄新西兰肉兔为试验动物,随机分成5组,以赖氨酸含量分别为0.55%、0.65%、0.75%、0.85%和0.95%的5种日粮为试验处理,进行为期20 d的饲养试验。结果表明,随日粮赖氨酸水平的增加,90日龄生长肉兔生长性能组间差异不显著(P>0.05);血清胰岛素、生长激素、类胰岛素生长激素浓度显著提高(P<0.05),分别在0.85%,0.75%和0.95%达到最大值;肝脏和肌肉IGF-Ⅰ相对表达量也明显增加(P<0.05)。综上所述,70~90日龄生长肉兔血清IGF-Ⅰ浓度和基因表达受日粮赖氨酸水平的影响。 相似文献

选取21日龄断奶长白仔猪37头,按表皮生长因子(EG F)、胰岛素样生长因子-Ⅰ(IG F-Ⅰ)、基础日粮、自然哺乳随机分为4组,每组3重复,每个重复3头。EG F和IG F-Ⅰ的剂量为17.86μg/d,研究其对早期断奶仔猪免疫功能的影响。结果表明:EG F组和IG F-Ⅰ组脾淋巴细胞转化率均极显著(P<0.01)高于基础日粮组,空肠后段肠黏膜SIgA含量极显著(P<0.01)高于基础日粮组而且EG F组脾淋巴细胞转化率高于IG F-Ⅰ组。EG F组小肠各段黏膜上皮间淋巴细胞数极显著(P<0.01)高于基础日粮组;IG F-Ⅰ组空肠和回肠黏膜上皮间淋巴细胞数极显著(P<0.01)高于基础日粮组;EG F组回肠黏膜上皮间杯状细胞数极显著(P<0.01)高于基础日粮组;IG F-Ⅰ组回肠黏膜上皮间杯状细胞数极显著(P<0.01)低于自然哺乳组。表皮生长因子和胰岛素样生长因子-Ⅰ能提高早期断奶仔猪的免疫功能。 相似文献
