The addition of a yeast culture food supplement (DVAqua from Diamond V. Mills, Cedar Rapids, IA, USA) was evaluated during feeding of McConaughy strain rainbow trout Oncorhynchus mykiss. Two trials were run consecutively, starting with initial feeding, and were replicated over 2 years. The yeast culture was added to commercial trout starter at levels of 0.125 and 0.25 g kg?1. The number of rainbow trout that died during the first 4 weeks of rearing was significantly reduced in the tanks receiving either of the yeast culture‐supplemented diets compared with the tanks receiving a control starter diet. Ending tank weights and gain were significantly and consistently greater in the tanks fed the 0.25 g kg?1 yeast culture diet. Individual fish weights were also significantly heavier in the fish‐fed yeast culture containing diets in each year of the study at the end of 27 days. In the trials from 4 to 8 weeks postinitial feeding, mortality rates were significantly less only in the tanks receiving 0.25 g kg?1 yeast culture supplementation. Individual fish lengths and weights at the end of the second trial were significantly greater in the tanks receiving either level of yeast culture compared with the control diet only in 1 year. Total tank weights and feed conversion were not significantly different between any of the treatments. The inclusion of DVAqua yeast culture provided significant increases in trout survival and growth through the first 4 weeks of feeding, with continued benefits for the next 4 weeks.  相似文献

In this study, the effect of gonadotropin‐releasing hormone agonist (GnRHa) injection on milt production in spent rainbow trout was investigated. On day 0, 25 newly matured male rainbow trout (Oncorhynchus mykiss) were stripped manually, and sperm quantity (vol: mL fish?1) and quality, spermatocrit (%), sperm count (cell mL?1), motile sperm percentage and motility duration (s) were evaluated. After stripping, fish were randomly divided into five groups: intact; sham (injected with propylene glycol as a hormone vehicle); and groups receiving 4, 8 or 16 μg kg?1 BW of [d ‐Ala6 Des‐Gly10] mGnRHa. On day 7, the fish were stripped again and the same sperm characteristics as on day 0 were measured. At the beginning of the experiment, there were no significant differences in any of the sperm quantity characteristics between groups. On day 7, expressible milt volume was significantly reduced compared with day 0 (P<0.05, t‐test) in the intact and sham groups but milt quality remained the same (P>0.05, t‐test). The present study shows that GnRHa injection with a concentration as low as 4 μg kg?1 BW after first stripping could prevent a significant reduction in milt quantity collected 7 days later without any adverse effects on sperm quality.  相似文献

The effect of finishing extensive farming period, to reduce fat content and manipulate the fatty acid profile of fish muscle, was evaluated in rainbow trout. Fish were stocked in an artificial lake, in which fish were fed only on naturally available nutrients with no supply of artificial feed, for different lengths of time from 0 to 120 days. No weight loss was noted during the whole finishing period while total length increased from 228±7 to 269±3 mm and the condition factor decreased from 1.41±0.04 to 0.89±0.02. The total fat content of the fillets decreased considerably from 4.7±0.6% at the beginning to 2.4±0.4% and 0.7±0.2% after 45 and 120 days respectively. Fillet fatty acid composition was affected by the time of stocking in the extensive farm. In contrast to the reduction in C18:1n‐9, C18:2n‐6, total monounsaturated fatty acid and total n‐6 percent values, an increase in the C20:5n‐3, C22:6n‐3, total polyunsaturated fatty acid and total n‐3 percent values was observed. It was shown that while other finishing strategies for salmonids have some disadvantages, the extensive culture system seems to be a potentially useful tool for increasing the general quality of the end product.  相似文献

The efficacy of adenoviral vectors for gene delivery into fish cells, both in vitro and in vivo, was evaluated. Vectors utilized were of human adenovirus serotype 5 (Ad), which are commonly used in human clinical trials, but have not been assessed for gene delivery to fish. Because nothing is known about Ad receptors in fish, both an Ad (Ad5Luc1) with natural tropism for the coxsackie and adenovirus receptor (CAR), as well as an infectivity enhanced Ad (Ad5LucRGD) were included within this study. Gene expression was detected in cell lines using either vector. The levels seen with Ad5LucRGD were much higher than for Ad5Luc1 in most lines except CHSE-214. Transduction of CHSE-214 cells with Ad5Luc1 could be blocked with an excess of a competitive inhibitor, suggesting that these cells possess a CAR homologue thatmediates attachment of Ad, similar to that seen in mammalian cells. In vivo gene delivery was attempted by several methods, with significant expression seen only via intramuscular injection, although infection efficiency was low. Thus it was observed that several teleost cell lines are capable of being infected and one cell line expressed a human serotype adenoviral receptor homologue that aids in Ad infection. Additionally, in vivo studies indicated that muscle tissue of rainbow trout could be infected with Ad vectors, suggesting an alternative gene delivery strategy for this animal.  相似文献

The deposition of natural, optically active, astaxanthin fatty acid esters in rainbow trout ( Oncorhynchus mykiss ) was studied. Mono-esterified and di-esterified (3 S ,3' S ) astaxanthin were purified from the green microalga Haematococcus pluvialis and incorporated into extruded diets and compared with diets containing synthetic racemic astaxanthin (Carophyll Pink) and a total carotenoid extract from the alga. All sources of astaxanthin achieved >4 mg kg−1 in the white muscle after 6 weeks feeding. No significant difference ( P  > 0.05) between the deposition of astaxanthin or total carotenoid for the different diets was observed. Other xanthophylls, namely lutein, zeaxanthin and idoxanthin were found in the white muscle of rainbow trout fed all diets and together accounted for 10–14% of total carotenoid. Astaxanthin was deposited in the white muscle in the stereochemical form administered in the diet, i.e. racemic astaxanthin for Carophyll Pink and ˜100% (3 S ,3' S )-astaxanthin for the algal sources. In contrast, epimerization of (3 S ,3' S ) astaxanthin from the alga was observed for the astaxanthin esters deposited in the skin of rainbow trout, with a ratio close to 1.0:2.0:1.0 (3 S ,3' S :3 R ,3' S :3 R ,3' R ).  相似文献

The aim of this study was to investigate the effect of dietary lactoferrin (Lf) on growth, haematology and non‐specific immune response of rainbow trout Oncorhynchus mykiss (Walbaum). Fish were fed an experimental diet containing 0 (as control), 50, 100, 200 and 400 mg Lf kg?1 diet twice daily for 8 weeks and sampled at 2, 4, 6 (immune function and growth), and 8 weeks (immune function, haematology and growth). Statistical analyses revealed no significant effects of dietary Lf on growth performance (specific growth rate, weight gain, feed conversion ratio, feed intake and condition factor) or haematological parameters (red and white blood cell count, haemoglobin, haematocrit, serum iron and total iron binding capacity [TIBC]). Among the serum non‐specific immune parameters, lysozyme activity increased significantly in fish fed 100, 200, or 400 mg Lf kg?1 feed for 8 weeks, whereas haemolytic complement activity increased in fish fed 100 and 400 mg Lf kg?1 diet after 6 weeks. The antiprotease activity increased in groups fed 100, 200 or 400 mg Lf kg?1 diet after 8 weeks. However, no significant effect was observed on serum peroxidase level. It can be concluded that feeding of rainbow trout on the diet supplemented with 100 mg kg?1 or higher for 8 weeks enhances the non‐specific immune response.  相似文献

Long‐day photoperiods are considered as an effective managerial tool in manipulating of reproduction and somatic growth in a number of fish species. In this study, the effects of three different artificial photoperiods on the gonadal development and somatic growth of rainbow trout (Oncorhynchus mykiss L.) were investigated. Two years‐old immature female rainbow trout (279.94 ±2.25 g) were exposed to three artificial photoperiod regimes of 24L:0D, 18L:6D and 6L:18D and natural light (NL) regime for 5 months. The highest gonadosomatic indices were recorded in NL and 6L:18D groups while the rates were significantly lower in fish maintained under 18L:6D and 24L:0D photoperiods (< 0.05). Mean oocyte diameters in fish exposed to 24L:0D and also to 18L:6D were significantly lower than the 6L:18D and NL groups. Photoperiods with 24L:0D and 18L:6D regimes resulted in significantly higher mean final weights and specific growth rates (SGR) than NL regime. The highest mean final weight (635.45 ± 16.19 g) and SGR (1.03 ± 0.04% day?1) were obtained under 24L:0D photoperiod. Fish exposed to 24L:0D and 18L:6D showed the highest condition factor as 1.44 ± 0.01 and 1.44 ± 0.02 respectively, when compared with the NL (1.27 ± 0.01) and 6L:18D (1.34 ± 0.02) groups. Basically, the results suggested that continuous artificial lightning can be used as an influential factor in delaying gonadal development and enhancing somatic growth in rainbow trout during gonadal growth phase.  相似文献

ABSTRACT:   Thermal tolerance was studied in a rainbow trout strain successively selected through high temperature breeding at 20–27°C since 1966 in Miyazaki Prefecture, Japan. The hatching rate and fry mortality at high temperatures were examined in the selected strain along with normal strains cultured at water temperature of 9–17°C. The hatching rate of embryos fertilized at either 10 or 14°C and subsequently subjected to high temperatures in the blastula or neurula stage of the selected strain, was marginally higher than that of the normal strain counterparts. The upper 50% lethal temperatures (LT50) for embryos in the early segmentation, blastula and neurula stages of the selected strain were also higher than those of the normal strain counterparts. Death temperatures and LT50 of fries acclimated to 20°C of the selected strain were significantly higher than those of the normal strains. However, no difference in the critical thermal maximum was detected between the different strains. These results suggest that the selected strain of rainbow trout established by selecting successively for many generations at high temperatures acquired a degree of thermal tolerance.  相似文献

Genetic variation fuels selective change in natural and captive populations. In establishing a broodstock for selective improvement, the level of genetic diversity is an important consideration because it provides an indication of the scope for selective progress. Three domesticated strains of rainbow trout, Oncorhynchus mykiss (Walbaum), were examined at nine polymorphic microsatellite loci to assess detectable levels of allelic diversity and heterozygosity within and differentiation among the strains. A total of 126 alleles were observed to segregate into unique multilocus genotypes for each of the 152 individuals assayed. There was an average heterozygosity of 71.5% at these nine loci, and an average of 14 alleles at a locus. Each locus was represented by alleles unique to at least two of the three strains. Deviations from Hardy–Weinberg expectations of genotype frequencies were detected in each strain. Subsequent analysis indicated sub‐structuring within strains leading to Wahlund effects that caused these deviations. Significant differences in genotype frequencies and pairwise FST values demonstrated that all strains were unique. The overall FST of 0.089 provides additional evidence of unique genetic diversity present in each strain, and agrees well with the degree of genetic variation found in rainbow trout across broad geographical ranges. The genetic diversity contributed by each population suggests that there is greater scope for selective improvement of numerous traits within a synthetic strain combining these three strains than within any individual strain.  相似文献

A feed budget model, which enables farmers to adapt a general model to local conditions, was tested on rainbow trout, Oncorhynchus mykiss (Walbaum). The model has two major components; component 1 is used to calculate the energy required for one unit of biomass gain (g or kg) and component 2 estimates the daily growth increment (g or kg). Both components can be adjusted in relation to prevailing environmental conditions and, together, they express the daily energetic requirement of the specific species or strain. Fish fed using the model as a guide had a high rate of growth and good feed conversion. The model accurately predicted growth over the experiment, and is presented as a means of calculating the daily feed budget in fish farming. Growth of fish fed using self-feeders is presented for comparison with the results obtained using the model.  相似文献

The aim of this study was to assess the metabolic and digestive enzyme activities in rainbow trout, Oncorhynchus mykiss Walbaum fed dietary supplements. The biometric indices and the following enzymes activities were measured: acid protease and pepsin from the stomach homogenates, alkaline phosphatase (AP) from the small intestine and the brush border membrane, total proteolytic enzymes and trypsin from the small intestine and liver/pancreas. Dietary garlic induced a higher pepsin activity in the stomach than lipopolysaccharide (LPS) and ginger. Conversely, dietary ginger and LPS showed a significant difference (P > 0.05) in acid protease activity compared to the controls. There was not any significant difference observed between treated groups and the controls for hepato‐pancreas proteolytic enzyme activity, except for trypsin. AP activity increased significantly with dietary garlic, ginger and LPS compared to the controls.  相似文献

Jatropha curcas is a multipurpose and drought‐resistant tree, widespread throughout tropics and subtropics. Its seeds are rich in oil and protein. It is being promoted as a biofuel plant. Jatropha kernel meal obtained after oil extraction is an excellent source of protein [580–660 g kg?1 crude protein (CP)]. However, the presence of toxic and antinutritional constituents restricts its use in fish feed. Jatropha kernel meal was detoxified. Using Oncorhynchus mykiss juveniles, a 12‐week experiment was conducted to evaluate the nutritional quality of detoxified Jatropha kernel meal (DJKM). Oncorhynchus mykiss juveniles (36, av. wt. 4.2 ± 0.4 g) were distributed into three groups with 12 replicates and fed isonitrogenous (CP 450 g kg?1) diets: control [fish meal (FM)–based protein], J50 and J62.5 (50% and 62.5% of fishmeal protein replaced by DJKM). Growth performance, apparent lipid conversion, hepatosomatic index, nutrients and energy digestibilities; efficiency of digestible nutrients and energy; and glucose and creatinine levels in blood were statistically similar (P > 0.05) for control and J50 but were higher (P < 0.05) than for J62.5. Opposite trend was observed for phosphorus, sodium, albumin and total protein in blood. Feed conversion ratio, protein efficiency ratio, protein productive value, energy retention, gastro somatic index, muscle lipid peroxide; and red and white blood cells, haematocrit, haemoglobin, mean corpuscular volume, mean corpuscular haemoglobin, mean corpuscular haemoglobin concentration; alkaline phosphatase and alanine transaminases activity, globulin, total bilirubin, blood urea nitrogen, calcium and potassium in blood and lysozyme activity in serum did not differ significantly among the groups. Intestinal enzyme activities; and plasma and muscle cholesterol levels for control were higher (P < 0.05) than for DJKM groups, while opposite trend was observed for relative intestinal length (mm g?1). Conclusively, performance of J50 group was similar to control group and, thus, is recommended as the optimal diet for rainbow trout.  相似文献

Fish growth is manifested by a number of measurable physical changes. We have developed a sensitive method for monitoring the growth rate of fish fed at four different planes of nutrition. This technique consists in measuring expression levels of myosin RNA isolated from the muscle of experimental animals. Using PCR (polymerase chain reaction) primers, and a fluorescence-labelled single-stranded DNA probe that hybridizes specifically to a region within the myosin mRNA of rainbow trout Oncorhynchus mykiss (Walbaum), we were able to detect differences in the relative level of myosin expression between groups of fish. This method also allows for the determination of absolute expression levels when reactions are performed with standards consisting of known levels of in vitro-transcribed myosin RNA. With the proper equipment, this novel procedure can be performed rapidly on large numbers of individuals, and with the procurement of non-invasive muscle biopsies the same experimental animal could theoretically be sampled multiple times throughout the course of the study. This new method could be used to measure differences in muscle synthesis in fish associated with various nutrient intake levels, environmental parameters, life-history stages and health status.  相似文献

The efficient use of feed for growth and meat production is important for all animal production industries including aquaculture. Residual feed intake (RFI) is an alternative measure of feed efficiency that has been widely used in livestock production. Residual feed intake was calculated as the difference between intake observed and intake predicted on the basis of a bioenergetics model; a low RFI indicates greater efficiency. Residual feed intake offers some advantages as a selection criterion for improving production efficiency over traditional feed efficiency statistics because it is not a ratio and it typically has a larger coefficient of variation. The RFI of individually reared rainbow trout progeny from six different genetic cross‐types was examined for genetic variation. Proximate analysis and nitrogen retention were also evaluated to determine if differences in RFI correlate to differences in body composition and nutrient retention and varied by cross‐type. Differences between cross‐types indicated a genetic component for RFI, with the most efficient fish of approximately 160 g consuming 0.99 g less and inefficient fish consuming 0.05 g more feed per day than expected. Lower RFI was associated with higher growth rates (r=?0.38, P<0.05) and greater nitrogen retention (r=?0.82 P<0.001).  相似文献

The covariation between diploid and triploid progenies from common breeders was investigated in various progeny-testing experiments where either dams or sires were sampled from rainbow trout, Oncorhynchus mykiss (Walbaum), stocks. Triploidiza tion was found to frequently reduce the performance in the traits studied: body length and weight, growth, coefficient of condition and pyloric caeca number. Triploidization also generated significant interactions with the parental breeding value. These interactions were caused in part by the familial variance not being the same in triploids as in diploids, but also by actual ranking differences between diploid and triploid familial performances. However, the effect of these interactions was minor as compared with the amount of variation common to both ploidy levels (genetic correlations averaged 0.7–0.9). Therefore, selection of diploid breeders appeared efficient enough for improving triploid progeny, unless family selection methods including triploid progeny testing were preferred for other reasons. Lastly, it was observed that variances from maternal origin tended to be larger in triploids, whereas variances from paternal origin tended to be smaller, as compared with diploids. This point was discussed referring to the genetic make-up of triploids and in the scope of dams and sires for selective breeding.  相似文献
