An 8-year-old castrated male Golden Retriever was evaluated for decreased appetite, lethargy, and labored breathing of 1-week duration. Bilateral pulmonary infiltrates, hepatomegaly, and splenomegaly were present. Results of a CBC revealed marked leukocytosis (62,600/microL; reference interval 4000-15,500/microL) and large numbers of atypical cells (30,700/microL) with abundant cytoplasm. There was no concurrent anemia, neutropenia, or thrombocytopenia. Morphology of the atypical cells was most consistent with a histiocytic origin. Similar cells were identified in bone marrow aspirates, and were morphologically suggestive of the macrophage variant of disseminated histiocytic sarcoma. However, flow cytometry of the abnormal circulating cells revealed CD1c, CD11c, and major histocompatibility complex (MHC) Class II expression without expression of CD11d or lymphoid markers, consistent with myeloid dendritic antigen-presenting cells. At necropsy, the splenic architecture was effaced by neoplastic histiocytes that were also infiltrating lung, liver, an abdominal lymph node, myocardium, an bone marrow. Immunohistochemistry of the splenic neoplastic cells confirmed dendritic cell origin (CD1c+, CD11c+, MHC II+, no expression of CD11d and lymphoid markers). To the authors' knowledge, this is the first report of canine dendritic cell leukemia-in this instance accompanied by marked tissue infiltration.  相似文献

A mononuclear cell leukemia in Sprague-Dawley rats is described in which liver, spleen, and lung involvement was a constant feature. The cell was 16 to 25 microns in diameter with round, oval or indented nucleus, and the cytoplasm contained bright red granules. This is the first report of such a leukemia in Sprague-Dawley rats and a comparison with mononuclear cell leukemia in other strains of laboratory rats is made.  相似文献

An unusual case diagnosed as connective tissue-type mast cell leukemia with marked mastocyte infiltration into visceral organs in a seven-year-old female Curly-Coated retriever is presented. Acute circulatory collapse, emesis, diarrhea, abdominal enlargement, icterus, cyanosis, dyspnea, pulmonary edema, hepatomegary, ascites, and right ventricular enlargement were observed. Hematologic and biochemical examinations revealed mast cell leukemia, mature neutrophilia, monocytosis, thrombocytopenia, hemolytic hyperbilirubinemia, hyperhistaminemia, renal and hepatic injuries. Mast cells were distributed systemically, but predominantly in the diaphragm and liver with a large mass among the serosa of ileum, cecum and colon. Mast cells were stained intensely by both safranin and berberine sulfate.  相似文献

Total and differential leucocyte counts (lymphocytes, neutrophils, monocytes, eosinophils and basophils) were measured in 140 stallions, 101 mares and 25 foals of Lipizzan breed. The values fell in the normal ranges for warm-blooded horses. Differences between mares and stallions were not significant with the exception of foals, having higher white blood cell, neutrophil, lymphocyte, monocyte and basophil values in females than in males. Foals exhibited an age-related increase of total leucocyte count during the first 4 months of life, accompanied by a decrease in neutrophil and increase in lymphocyte and eosinophil counts. In mares and stallions, the total number of leucocytes, lymphocytes, monocytes and basophils significantly decreased but the number of neutrophils and eosinophils remained almost unchanged with age gain.  相似文献

【目的】 筛选鸡胸腺中免疫相关的环状RNA (circular RNA,circRNA),研究circRNA在鸡免疫调节中的作用。【方法】 采用高通量测序平台Illumina PE150对黄羽肉鸡和山地乌骨鸡的胸腺组织进行转录组测序,用生物信息学分析软件find_circ和CIRI识别circRNA分子,鉴别其基本结构特征。通过差异显著性分析筛选差异表达circRNA,再经过GO功能和KEGG通路富集分析筛选出免疫相关circRNA,用IRESfinder和PFAM软件进行circRNA编码潜能预测,并用实时荧光定量PCR进行验证。【结果】 circRNA在鸡胸腺中有丰富的表达,共识别到8 015个circRNAs,其中115个circRNAs在胸腺组织中的表达量呈显著差异(log2|FoldChange|>1,P<0.05),在黄羽肉鸡中上调表达基因54个,下调表达基因61个(以山地乌骨鸡为对照)。GO功能富集结果显示有104个GO条目与免疫相关,涉及23个差异表达cricRNAs,占比高达20%(23/115)。KEGG通路分析结果显示,差异表达circRNA来源基因集中在细胞因子-细胞因子受体相互作用、RIG-Ⅰ样受体信号通路、Toll样受体信号通路等7个免疫相关通路中,共涉及7个差异表达circRNAs。有5个circRNAs在GO功能和KEGG通路富集分析中重复出现:circ_0002478(IL-1R1)、circ_0003208(MAPK11)、circ_0001674(STX8)、circ_0005105(RIPK2)和circ_0003590(BRAF),推测它们可能在免疫系统调节中扮演着重要角色。实时荧光定量PCR结果显示,5个circRNAs在2个鸡品种胸腺组织中的表达差异与测序结果一致,circ_0002478(IL-1R1)不仅在两品种间差异极显著(P<0.01),而且具有编码多肽潜能,它可能以多种方式参与免疫调节。【结论】 鸡胸腺组织中有丰富的circRNA,在与免疫相关的circRNA中circ_0002478(IL-1R1)可能通过多种方式参与免疫调节,可作为鸡免疫力研究的重点关注靶点。  相似文献

A 5‐year‐old male neutered Bernese Mountain Dog was presented for cutaneous plasmacytoma, which was treated by surgical excision. Four months later, the dog developed multiple skin masses, hyphema, pericardial and mild bicavitary effusions, myocardial masses, and marked plasmacytosis in the peripheral blood. Circulating plasma cells expressed CD34 and MHC class II by flow cytometry. Immunocytochemistry demonstrated that these cells were strongly positive for multiple myeloma oncogene 1/interferon regulatory factor 4 (MUM‐1) and weakly to moderately positive for Pax5. The dog was hypoglobulinemic but had a monoclonal IgA gammopathy detected by serum immunofixation electrophoresis. The PCR analysis of antigen receptor gene rearrangements (PARR) by fragment analysis using GeneScan methodology revealed that plasmacytoid cells in the original cutaneous plasmacytoma and peripheral blood had an identical immunoglobulin heavy chain gene (IgH) rearrangement, indicating that both populations were derived from the same neoplastic clone. Canine cutaneous plasmacytoma rarely progresses to a malignant form and plasma cell leukemia is rarely diagnosed in the dog. This report describes a case of cutaneous plasmacytoma progressing to plasma cell leukemia with a rapid and aggressive clinical course. This report also highlights the utility of flow cytometry, immunocytochemistry, immunofixation electrophoresis, and PARR by fragment analysis using GeneScan methodology in the diagnosis of this hematopoietic neoplasm.  相似文献

A 16‐year‐old, Irish Draft mare was admitted to the referring veterinarian for an annual health check. A mild generalized lymphadenomegaly was noted. Rectal palpation and transrectal ultrasonographic examination revealed prominent mesenteric lymph nodes. A transcutaneous abdominal ultrasonographic evaluation was unremarkable. A CBC revealed a marked leukocytosis (63.06 × 103/μL) and lymphocytosis (58.2 × 103/μL) due to increased numbers of small lymphocytes. No evidence of anemia or thrombocytopenia was found and neutrophil counts were low‐normal. Cytologic examination of fine‐needle aspirates of multiple lymph nodes and a bone‐marrow aspirate revealed the presence of a monomorphic population of small lymphocytes similar to those observed in the peripheral blood, suggesting a leukemic small cell lymphoma (SCL) or chronic lymphocytic leukemia (CLL). As the lymphadenomegaly and peripheral blood lymphocytosis were present simultaneously, the distinction between these 2 conditions was not possible. Immunophenotyping by immunocytochemistry and flow cytometry of the lymphoid cells in peripheral blood determined a T‐cell phenotype. As the horse was clinically stable, no treatment was initiated, but regular examinations were undertaken. A CBC repeated 120 days after the diagnosis showed a marked lymphocytosis (157.6 × 103/μL) with no evidence of anemia or other cytopenias. The horse was euthanized 194 days after the initial diagnosis. Histopathology and immunohistochemistry of submandibular lymph nodes and bone marrow confirmed the diagnosis of leukemic SCL or CLL, and a T‐cell phenotype. SCL and CLL are rare in horses; previous immunohistochemical studies determined that the T‐cell phenotype is predominant. To the authors' knowledge, this is the first report of the combined use of immunocytochemistry and flow cytometry in a horse with leukemic SCL or CLL.  相似文献

A 7‐year‐old Shetland Sheepdog was presented with anorexia. A CBC indicated thrombocytopenia and neutropenia. Bone marrow cytology revealed that 67.7% of all nucleated cells (ANC) were anaplastic large mononuclear cells. These cells were confirmed to be of B‐cell origin based on IgH rearrangement, immunohistochemical, and flow cytometric analysis. Microscopic examination revealed that the neoplastic cells had intranuclear inclusions resembling Dutcher bodies. Immunohistochemistry confirmed that the intranuclear inclusions were immunopositive for IgG antibodies. The periodic acid–Schiff reaction was negative for the presence of polysaccharides and related substances. Although the dog achieved complete remission with a multi‐drug chemotherapy protocol, it ultimately died because of tumor progression and acute renal insufficiency on day 201. This is the first known case of canine acute B‐cell leukemia with intranuclear inclusions resembling Dutcher bodies.  相似文献

百里香(Thymus mongolicus)是一种兼具香化、绿化、美化功能的芳香草本植物,为探究百里香叶片挥发性物质释放种类和释放量与物候期变化间的关系,以期为百里香挥发性物质的进一步研究提供数据支撑与理论依据,本研究采用顶空固相微萃取-气相色谱-质谱联用(HS-SPME-GC-MS)技术结合外标法,检测百里香叶片在不同物候期释放的挥发性物质,分析挥发性物质的成分与含量,探索百里香挥发性物质的释放节律。结果表明:4个物候期的百里香叶片中共检测出69种挥发性物质,分属于烯烃类、醇类、酯类、芳香烃类、酮类和其他类化合物,不同物候期百里香叶片释放的挥发性物质的种类和含量表现出差异性。挥发性物种种类表现为生长旺盛期(53种)>开花期(50种)>结果期(43种)>衰败期(28种)。挥发性物质含量表现为开花期(163.90μg·g-1)>结果期(157.21μg·g-1)>生长旺盛期(104.47μg·g-1)>衰败期(75.42μg·g-1),总体呈现先升高后降低的趋势。不...  相似文献
