An 8-week feeding trial was conducted in a recycling water system at 28±1°C to investigate carbohydrate to lipid ratio (CHO:L ratio) in African catfish Clarias gariepinus (12.32±0.04g). Five isonitrogenous (40% crude protein) and isoenergetic (20kJg–1 gross energy (GE)) fishmeal based diets with varying carbohydrate to lipid (CHO:L g/g) ratios of 0.74, 1.13, 1.66, 2.47 and 3.42 for diets 1–5, were tested, respectively. The diets containing a fixed protein to energy ratio (P:E ratio) of 20-mg proteinkJ–1 GE were fed to triplicate groups of 20 fish (per 30-L tank). Fish were fed 5% of their body weight per day adjusted fortnightly. Diet 1, containing 14% carbohydrate and 21% lipids with a CHO:L ratio of 0.74 produced the poorest (P<0.05) growth rates, feed and protein efficiency. Increasing carbohydrate content in the diets to 27% concomitant with a reduction in lipid content to 16% with a CHO:L ration of 1.66 of diet 3 significantly improved (P<0.05) growth rates, feed and protein efficiency. A further increase in dietary carbohydrate up to 38% and a decrease in lipids levels to 11% with a CHO:L ratio ranging from 1.66 to 3.42 (diet 3 – 5) did not significantly improve the fish performance. Apparent net protein utilisation (ANPU) of fish fed diet 4 was higher (P<0.05) than for diets 1–3 but did not differ from diet 5. Higher lipid deposition (P<0.05) in whole body and liver were observed with decreasing dietary CHO:L ratios as increasing lipid levels. Whole body protein and liver glycogen content, digestive enzyme activities (protease and lipase) and histological examination of intestine and liver of fish fed varying CHO:L diets did not show any discernible changes among the dietary treatments. However intestinal -amylase activity increased (P<0.05) with increasing dietary carbohydrate levels. This study revealed that African catfish can perform equally well on diets containing carbohydrate ranging from 27 to 38% of the diet, with lipid content ranging from 16 to 11% or at CHO:Lg/g ratio of 1.7–3.4.  相似文献

In fish, sex steroids initiate and/or accelerate the maturation of the brain-pituitary-gonad axis. In order to obtain information on the steroid milieu during the pubertal development of male African catfish, we have monitored the conversion of [3H]-pregnenolone and [3H]-androstenedione by testis and [3H]-pregnenolone by interrenal tissue fragmentsin vitro. Pubertal development occurs between two and six months of age. Testicular development proceeds through four main stages that are characterised histologically by the presence of spermatogonia (stage I), spermatogonia and spermatocytes (stage II), spermatogonia, spermatocytes and spermatids (stage III), and all germ cells including spermatozoa (stage IV). 11β-Hydroxyandrostenedione and cortisol were the main products of testes and interrenal tissue, respectively, in all stages of the pubertal development; a change in the steroidogenic pattern was not observed during this period. The interrenal tissue displayed no significant conversion of [3H]-pregnenolone to 11-oxygenated androgens. Blood plasma was analyzed for the presence of five androgens; testosterone, 11β-hydroxytestosterone, 11β-hydroxyandrostenedione, androstenetrione, and 11-ketotestosterone. 11-Ketotestosterone was the quantitatively dominating androgen in the circulation at all stages of development, which was more pronounced in stages III and IV. The obvious differences between thein vitro andin vivo results, namely 11β-hydroxyandrostenedione being the main testicular productvs. 11-ketotestosterone dominating in the blood, may indicate that 11β-hydroxyandrostenedione is converted to 11-ketotestosterone at extratesticular sites.  相似文献

11-ketotestosterone (OT) is a typical androgen of male teleost fish, but information on the question if it is involved in the feedback regulation of pituitary gonadotropin II (GTH-II) secretion is controversial. We have therefore studied the effects of OT on gonadotropin releasing-hormone (GnRH) stimulated GTH-II secretion in male African catfish Clarias gariepinus). In vivo experiments were carried out with intact and castrated fish. OT plasma levels were increased by implantation of silastic capsules containing 11-ketoandrostenedione (OA) which is converted to OT in both intact and castrated fish. When intact males received OA- or blank-capsules, treatment with salmon gonadotropin releasing-hormone analogue (Des-Gly10-D-Arg6-sGnRH-NEt; 0.2 μg sGnRHa/kg body weight) elevated the plasma GTH-11 levels in both groups. However, the levels were about 2 times higher in blank- than in OA-implanted fish. When castrated fish received either blank-or OA-capsules, sGnRHa treatment led to plasma GTH levels significantly higher than in sham-operated fish. However, there was no difference between the blank- or OA-implanted castrates, though OA implantation led to a restoration of OT plasma levels. This suggests that replacement ofOT is insufficient to reverse castration-induced effects. In vitro experiments were carried out with pituitary tissue fragments using a static culture system. The tissue remained sensitive to sGnRHa (5 × 10?9M) for 4 days after the beginning of incubation. Preincubation of pituitary tissue for 24 hours with 25 ng OT/ml medium (80 nM) completely abolished the stimulatory effect of sGnRHa on GTH-II secretion. Tritiated OT was not metabolized by pituitary tissue during 6 hours of incubation. We conclude that 11-ketotestosterone, a quantitatively prominent and non-aromatizeable circulating androgen participates, at least in part by direct action on the pituitary, in the negative feedback regulation of GnRH-stimulated GTH-II secretion in male African catfish.  相似文献

A quality cost analysis was performed with the cooperation of six full-time African catfish, Clorias gariepinus, farmers in The Netherlands. The relative distribution of the quality costs does not fit a common quality cost model. Possible explanations for this observation are the absence of (1) feedback from customers and (2) well-defined quality criteria. The results show that it is likely that a considerable reduction in quality costs can be achieved with respect to sorting and rewatering. A further reduction is possible if agreement on quality criteria for African catfish can be reached.  相似文献

Isolated gonadotrops of the African catfish,Clarias gariepinus, were incubated with dopamine (DA) and/or catecholestrone and the activity of the enzyme catechol-O-methyltransferase (COMT) was determined by measuring the methylated products. From the apparent Km values for DA and catecholestrone of 0.4–1.3 M and 17.9–25.2 M respectively, it was concluded that catecholestrone is a better substrate for the enzyme COMT, compared to DA. Moreover, the methylation of DA is inhibited by comparatively low concentrations of catecholestrone.  相似文献

The objective of this study was to assess whether high‐pressure injection of air into the brain of African catfish (Clarias gariepinus) could render the animal unconscious and insensible immediately and permanently. In the study, 48 African catfish with a live weight of 900–1900 g were restrained and equipped with EEG and ECG electrodes and then stunned. The catfish were stunned mechanically using a captive needle pistol. The pressure to shoot the needle was 8 bar and that to inject the air was 3 bar for 1.5 s. The catfish behaviour was observed during and after stunning. τ and δ waves and spikes, which precede a stoppage in brain activity as measured on the EEG, were used as indices for the measurement of immediate induction of unconsciousness and insensibility In 23 of 42 fish, an iso‐electric line was observed after an average of 13.4 s, while in the remaining fish the τ and δ waves and spikes remained on the EEG during the recording period. In all cases, the ECG showed an irregular heart rate with fibrillation and extra systolae. Moreover, the configuration showed ischaemia. Before the captive needle stunning, free‐swimming fish (n=7) explored the tank for an average of 21±12 s before lying down at the bottom. After stunning, they showed clonic uncoordinated swimming movements. The movements stopped after an average of 38±50 s. In another group (n=7) that was stunned and subsequently placed in ice water, clonic cramps were observed in two out of seven animals. When taking into account the number of animals with a reliable EEG (n=42) and using 95% confidence intervals, it was concluded that at least 93% of the catfish were effectively stunned using a correctly positioned captive needle pistol. Furthermore, it is recommended to immobilize the stunned fish by chilling, as the post‐stun clonic cramps may hinder gutting and filleting.  相似文献

A high rate of sibling cannibalism is one of the principal obstacles in the rearing of larvae and juveniles of the African catfish Clarias gariepinus. This paper examines the underlying behavioural components of agonistic behaviour under culture conditions. Ten day old catfish larvae were stocked at 30 larvae l–1 in three tank designs with different surface areas and equal volumes. Stocking densities were 1.2, 0.6 and 0.3 fish cm–2 bottom surface. Growth did not differ between treatments. The highest mortality over 30 days was recorded at the medium density (0.6 larvae l–1). Fish stocked at this density showed the highest rate of aggression, while there was no difference in aggression between the highest and the lowest stocking densities. The results indicate that stocking density should be at least as high as 1.2 larvae cm–2 bottom surface area to obtain high production at best survival rates.  相似文献

Six fishmeal‐based diets of two protein levels (330 and 430 g kg?1), each with three lipid levels (40, 80 and 120 g kg?1) resulted in P/E ratios ranging from 15.5 to 21.3 mg protein kJ?1 gross energy (GE) were fed to 20 fish (per 30‐L tank) in triplicate. Fish were fed 50 g kg?1 of their body weight per day adjusted fortnightly. Significantly higher (P < 0.05) growth rates and feed conversion efficiency were evident in fish fed with higher protein diet. The highest growth rate was found by fish fed 430 g kg?1 protein, 21.2 kJ?1 GE with a P/E ratio of 20.5 mg protein kJ?1 GE. Significantly indifferent (P > 0.05) values of protein utilization were found in‐between the both (higher and lower) protein diets. Higher lipid deposition (P < 0.05) in whole body and liver was observed with increasing dietary lipid level at each protein diet and as higher (P < 0.05) for the lower protein diets. Liver glycogen tended to decrease with increasing gross energy at each protein diet and higher protein diet showed comparatively lower values (P > 0.05). Digestive enzyme activities (protease and lipase) and histological examination of intestine and liver of fish fed varying P/E diets found no significant differences in response to experimental diets. The study reveals that African catfish C. gariepinus performed best the diet containing 430 g kg?1, 21.2 kJ g?1 and 20.5 mg protein kJ g?1 GE protein, gross energy and P/E ratio, respectively.  相似文献

Vitellogenin is a female-specific calcium-binding glycolipophosphoprotein synthesized in the hepatocytes of fishes. Its synthesis can be induced in fishes of either sex by estradiol or by xenoestrogens. To study the in vitro synthesis of vitellogenin, different culture conditions were set up using the hepatocytes of Clarias gariepinus. The present study reports on a non-enzymatic procedure for isolation and culture of hepatocytes from the liver of the catfish Clarias gariepinus, in order to study the effects of estradiol on vitellogenin synthesis in vitro. The procedure employs chelating properties of ethylenediamine tetracetic acid to achieve cell viability in excess of 95%. Equal numbers of isolated cells were incubated in different culture media viz. RPMI F1640, Medium-199, and Williams’ Medium E. At 36 h, cell attachment and monolayer formation is faster in M-199 and Williams’ Medium E than in RPMI. In order to study the effects of estradiol on vitellogenin synthesis, the isolated hepatocytes were seeded in Williams’ Medium E in 24-well cell culture plates. 17 β-estradiol (E2) was introduced in the culture plates at different concentrations and for different time periods. The media were assayed for vitellogenin using competitive ELISA. Vitellogenin appeared in the medium after 48 h of incubation with 10−5 M estradiol whereas after 72 h of incubation 5×10−7 M E2 could elicit the synthesis.  相似文献

Semen of the African catfish, Clarias gariepinus (Burchell, 1822), was investigated with respect to its cellular composition, sperm cell density, maturation grade, motility and fertility. Storage conditions were tested, whereby sperm viability was assessed by measurement of the motility after activation and by fertility tests. Testicular semen differed in its composition, i.e. the sperm density and numbers of spermatids, according to the maturity grade of the testis. Two semen types could be distinguished: semen type I was characterized by high sperm densities and low numbers of spermatids and semen type II had lower sperm densities and higher numbers of spermatids. Two semen types did not differ in motility and fertility (when adjusted for differences in sperm density). During storage, the sperm viability was influenced by the sodium concentration of the storage medium, temperature, membrane stabilizers as bovine serum albumen (BSA) or hen egg yolk, antibiotics and oxygen. Semen viability was maintained best when it was diluted at a ratio of 1:5 in storage solution (150 mmol L?1 NaCl, 2.5 mmol L?1 KCl, 1 mmol L?1 CaCl2, 1 mmol L?1 MgSO4, 20 mmol L?1 Tris (pH 8.5) and 0.5% BSA or 0.5% hen egg yolk) and stored at 4 °C. Oxygen gassing and addition of antibiotics (1 mg mL?1 gentamycine sulphate) to the storage solution affected the two semen types in different ways. Antibiotics had no effect on type I semen, but had a positive effect on type II semen. Oxygen gassing had a positive effect on type I semen but a negative effect on type II semen.  相似文献

Effect of experimentally induced thyroxine overdose on the testis and seminal vesicles was studied in the air-breathing catfish, Clarias gariepinus during the preparatory and the pre-spawning phase. The present study revealed a marked reduction in testosterone level in serum, testis and seminal vesicles (SV). Histological examination showed a considerable reduction in the number of spermatozoa/spermatids in the seminiferous tubular lumen as well as depletion of fluid in the loculi of SV. SDS-PAGE analysis of SV fluid proteins demonstrated a significant decrease in the level of a ~27 kDa protein in thyroxine treated fishes. Successive exposure to aquaculture‐related stressors may compromise the allostatic capacity of African catfish and lead to allostatic overload and poor welfare. Therefore, we tested the effect of (i) feeding during the light or dark phase, (ii) density (51 fish per 140 L versus 51 fish per 43 L) and (iii) altered available resting space/shelter on stress handling were studied in African catfish (Clarias gariepinus). Available resting space/shelter was manipulated by providing PVC‐tubes in the tanks. Growth, baseline stress load (cortisol and osmolality), metabolism (glucose, lactate and non‐esterified fatty acids) and aggression (measured by skin scar incidence) were assessed. Upon completion of the experimental period, we determined the adaptive capacity of the fish by the response to air exposure. We show that night‐feeding enhances growth and lowers feed conversion ratio compared to day‐feeding. No effects of density were found for baseline stress hormones, basal metabolism or aggression. Low density seems to stimulate aggressive behaviour after air exposure. Unexpectedly, providing PVC‐tubes under high‐density conditions increased aggression, raised baseline cortisol levels and decreased basal metabolism as well as growth; also the endocrine response to air exposure was stronger. We argue that these effects relate to a reduction in available space, i.e. a further increase in density, or to an increased competition for shelter. Overall, our data suggest that night‐feeding optimizes growth and that care should be taken in using objects as shelter/enrichment for African catfish.  相似文献

Asian catfish, Clarias batrachus, were fed semi-purified basaldiets containing 0, 0.1, 0.5, 1, 3 and 5 mg biotin kg–1diet for 60 days. Fish fed the control diet (no biotin) showed(P < 0.05) higher mortality, lower weight gain, specificgrowth rate (SGR), feed efficiency ratio (FER) and protein efficiencyratio (PER) than in fish fed diets supplemented with biotin. The highestweight gain, SGR, FER and PER were noticed in fish fed 1 mg biotinkg–1, followed by 0.5, 5, 3 and 0.1 mg biotinkg–1, except for PER (followed by 0.5, 5, 0.1 and 3 mgbiotin kg–1). Quadratic analysis showed that the optimumdietary biotin requirements for maximal weight gain, PER and PER were2.49, 2.54 and 2.52 mg kg–1, respectively. Liver biotinconcentrations were influenced by levels of biotin in the diet.Concentration of liver biotin increased as level of dietarysupplementation increased and no biotin was detected in the liver of thecontrol fish. Liver pyruvate carboxylase and acetyl CoA carboxylaseactivities were higher in fish fed biotin-supplemented diets than incontrols. Biotin concentrations, pyruvate carboxylase and acetyl CoAcarboxylase activities in liver associated with normal growth rangedfrom 10.59 to 10.66 g g–1, 147.97 to 148.18 units mgprotein–1 and 12.76 to 12.78 units mg protein–1, respectively. Biotin deficiency symptoms such as anorexia, darkskin colour and convulsions were observed in fish fed the control diet.The optimum dietary biotin requirement for maximal growth of C.batrachus is about 2.49 mg kg–1 diet.  相似文献

An 8‐week feeding trial was conducted in a recycling water system at 28 ± 1 °C to investigate compensatory growth and body composition in African catfish Clarias gariepinus (13.05 ± 0.05 g). A fishmeal‐based diet containing 350 g kg?1 protein and 17.5 kJ g?1 gross energy was fed to triplicate groups of 20 fish (per 30‐L tank). Fish were fed the diet either to satiation or feed restricted in six feeding schedules as satiation 56 days; restricted 28 days + satiation 28 days; restricted 14 days + satiation 14 days; restricted 7 days +satiation 7 days; restricted 3 days + satiation 4 days; and restricted 2 days + satiation 2 days. The restricted regime was achieved by offering fish 1% (maintenance ration) of their body weight per day adjusted after fortnightly weighing. African catfish showed partial compensatory growth under alternating periods of feed restriction and satiation feeding with significantly different values (P < 0.05) from fish fed in satiation throughout. However, significantly indifferent (P > 0.05) values of feed, protein, lipid and energy utilization were found under alternating periods of feed restriction and satiation feeding. Significantly higher (P < 0.05) feed intake was observed in treatment with satiation throughout than those in other treatments. All the feeding schedules showed no significant differences (P > 0.05) on body composition, organ indices, eviscerated carcass composition, viscera lipid and liver lipid. These studies reveals that C. gariepinus showed partial compensatory growth responses at alternating periods of restricted and satiation feeding.  相似文献

The present study investigated the qualitative amino acid (AA) requirements of larval African catfish Clarias gariepinus. Yolk-sac larval AA profiles were measured at different temperatures and also in animals reared at 28 °C fed Artemia nauplii or an experimental dry diet. The AA profile of C. gariepinus larvae changed during ontogeny, especially before the start of exogenous feeding. The AA profiles of the food items (yolk, Artemia and the dry diet) differed considerably from that of the larvae. No selective absorption of yolk AA was detected. Higher temperatures led to increased absorption and depletion rates of AA, and also to a higher retention efficiency of yolk nutrients. However, changes in temperature did not induce preferential absorption or depletion of individual AA, and caused only small variations in the AA profile. Depletion rates of individual AAs varied, possibly due to differences between larval and yolk AA profiles, and also to changes in the larval AA profile during ontogeny. There was little regulation of catabolism of individual AA in yolk-sac and starved larvae, and no sparing of essential AA.  相似文献

The influence of ontogeny and nutrition on metabolic enzyme activities in larvae of the African catfish, Clarias gariepinus, was studied. After start of exogenous feeding, the larvae were reared for 10 days under three different nutritional conditions: Artemia nauplii, a dry starter diet, and starvation. The live feed gave the best growth (96 mg within 10 days) whereas the dry diet resulted in low growth (33 mg). This growth difference was reflected in larval RNA and DNA concentrations, but not in the levels of soluble protein. Enzymes representing the following aspects of metabolism have been analysed: NADPH generation (G6PDH, ME), glycolysis (PFK, PK), gluconeogenesis (FDPase), amino acid catabolism (GOT, GPT) and oxidative catabolism (CS). All enzymes were present from the start of exogenous feeding onwards, but their maximum specific activities displayed different developmental patterns. In catfish larvae fed on Artemia, G6PDH and ME activities steadily increased with age and weight of the larvae. CS levels remained, after an immediate enhancement upon onset of exogenous feeding, on a rather stable plateau. The amino acid-degrading enzymes GOT and GPT showed maximum levels at days 3–5 of feeding or at a body weight of 10–20 mg, but decreased thereafter. Activities of PFK, PK and FDPase showed low initial levels, and increased significantly with age and size. Based on the ontogenetic patterns of metabolic enzymes, in C. gariepinus larvae an early and a late developmental phase can be distinguished. During the early phase, the glycolytic and gluconeogenetic capacities are low, whereas they are enforced during the later phase. The oxidative capacity is high both during the early and the late phase. The metabolic changes in catfish development coincide with other major ontogenetic events, e.g., alterations of muscle organization, gill morphology, respiration and stomach structure and function. Rearing catfish larvae on a dry diet instead of Artemia partly altered the developmental pattern described: The ontogenetic elevation of CS, PFK and FDPase was delayed and the early peak in GOT and GPT activities was not realized. Four distinct forms of native gonadotropin‐releasing hormone (GnRH) and two newly designed analogues were tested for their in vivo activity to induce ovulation in African catfish. The effects of these peptides on ovulatory parameters were compared with those of carp pituitary and [d ‐Ala6, Pro9‐NEt]‐mammalian GnRH analogue (mGnRHa), two tested ovulation‐inducing agents in African catfish. Assessment of ovulation was carried out by determining the ovulation ratio and the relative quantity of egg produced. From the results of the experiments, the order of potency of the native GnRH peptides is summarized as chicken GnRH‐II (cGnRH‐II) >salmon GnRH (sGnRH) >mammalian GnRH >chicken GnRH‐I (cGnRH‐I). Chicken GnRH‐II was as potent as mGnRHa while cGnRH‐I was totally ineffective. The new d ‐Orn6‐cGnRH‐II and d ‐Orn6‐sGnRH with a substitution at position 6 with d ‐isomer residue were as potent as the most extensively used mGnRHa, indicating that the position 6 modification might be more crucial than the substitution at the C‐terminal. On the basis of our results, the potential use and incorporation of cGnRH‐II and sGnRH for the development of more generic spawning induction therapies are suggested.  相似文献

11-Ketotestosterone (11-KT) is an important plasma androgen in male African catfish. The quantitatively predominating androgen produced by the testis, however, is 11-hydroxyandrostenedione (OHA). Our working hypothesis to explain this mismatch assumed that OHA is converted to 11-KT at extratesticular sites. First, we examined the in vivo metabolism of [3H]-OHA in mature males after sham-operation or removal of either the testes (TC), the seminal vesicles (SVC), or both (TSVC) by analysing the pattern of circulating [3H]-androgens two hours after intravenous injection of [3H]-OHA. [3H]-OHA was converted to [3H]-11-KT to the same extent in all groups, indicating that neither ablation of testes nor of seminal vesicles, or both attenuates this conversion. We then examined the in vitro metabolism of [3H]-OHA by several types of tissues. Liver and seminal vesicle tissue were found to produce significant amounts of [3H]-11-KT. The conversion capacity in vivo was assessed by injecting TSVC-castrated males with increasing doses of radioinert OHA, followed by the quantification of OHA and 11-KT plasma levels. Saturation of the conversion capacity was not reached but the 11-KT production capacity is at least 80 ng per ml of plasma per hour. Moreover, liver fragments were tested for their OHA to 11-KT conversion capacity in vitro using increasing concentrations of radioinert OHA. The 11-KT producing increased with time and OHA concentration. The production rate was 90 pg 11-KT mg-1 liver h-1. Considering the results of the surgical experiments and the fact that the total hepatic mass by far exceeds that of the seminal vesicles, we conclude that the hepatic conversion is of primary relevance in vivo.  相似文献

Juvenile African catfish, Clarias gariepinus (Burchell), of mean initial weight 15 g, were fed practical diets containing fresh or rancid oil (1:1 cod liver:corn oil) supplemented with either 20 or 100 mg all-rac-α-tocopheryl acetate per kg dry diet, at 0.03 × body weight per day for 8 weeks. After this time, catfish had grown by at least four times in body weight. Significant ( P < 0.05) inter-treatment differences in final body weight were noted. Clarias fed low-tocopherol: oxidized-oil diets performed least well with regard to growth, though elevated dietary vitamin E partially abrogated this effect. Growth of fish fed fresh-oil diets did not benefit from increased dietary α-tocopherol content. Muscle, liver, plasma, heart and spleen all responded significantly ( P < 0.05) to dietary vitamin E dose. Inclusion of oxidized oil in catfish diets decreased tissue α-tocopherol concentration. Hepatic α-tocopherol concentration (μg α-tocopherol per g liver) was observed to be lowered by 90% by the rancid oil diets. When fish previously fed fresh-oil diets were switched to oxidized: low-tocopherol diets, hepatic α-tocopherol concentration was significantly ( P < 0.05) lowered within 2 weeks. The results highlight the importance of dietary oil quality in modulating tissue α-tocopherol concentrations in African catfish.  相似文献
