To identify markers for the everbearing gene in strawberries, 199 F1 progeny plants were produced from a cross between ‘Ever Berry’ (a Japanese everbearing strawberry) and ‘Toyonoka’ (a Japanese Junebearing strawberry) as the experimental population. The results of flowering tests produced 97 everbears and 102 Junebears. The chi‐square test gave a goodness of fit for the expected ratio of 1 : 1 for everbears to Junebears, suggesting the inheritance of the everbearing trait is controlled by a monogenic dominant gene. RAPD analyses on this trait were carried out using ‘Ever Berry’ and ‘Toyonoka’. Seventy‐one primers, which produced 89 polymorphic fragments between the two parents, were identified from a total of 175 primers. Five markers relating to the everbearing trait were selected from 26 of the 199 progeny plants. The remaining 173 seedlings were analysed with these five markers and a linkage map was constructed using all of the 199 F1 progeny plants. The length of this linkage group is 39.7 cM. The closest markers found, OPE07‐1 and OPB05‐1, are respectively mapped at 11.8 and 15.8 cM on each side of the everbearing gene.  相似文献

The objective of this research was to determine which strawberry germplasm could be used to combine late flowering with reduced susceptibility to the strawberry blossom weevil, Anthonomus rubi. Seven cultivars and three breeding lines were compared for their susceptibility to A. rubi, by introducing adult weevils onto detached inflorescences. No consistent differences were detected but this contradicted observations from field trials. Five late‐flowering strawberry genotypes were crossed in a half‐diallel programme and progeny were tested for their susceptibility to A. rubi by using detached inflorescences and in a field trial where plants were exposed to a natural infestation. There was good correspondence between the results from the two methods and heritable differences were detected in both experiments, with the additive genetic variance being the more important. The cultivars ‘Idea’ and ‘Alice’ were identified as promising parents for reduced susceptibility while progeny from ‘Sophie’ were most likely to be susceptible.  相似文献

Summary Random amplified polymorphic DNA (RAPD) markers have been successfully employed to analyse the genetic diversity among cultivated and subspontaneous accessions of Coffea arabica. The narrow genetic base of commercial cultivars was confirmed. On the other hand, a relatively large genetic diversity was observed within the germplasm collection demonstrating the importance of collecting missions. Results suggested an East-West differentiation in Ethiopia, the primary centre of diversification of C. arabica. The large heterosis effect reported in intergroup hybrids could be related to such genetic differentiation. RAPD method appeared to be effective in resolving genetic variations and in grouping germplasm in C. arabica.  相似文献

Characterisation of fifteen commercial varieties of Chrysanthemum was carried out through RAPD analysis. Varieties could be distinguished from each other and the level of similarity between varieties seemed to be not very high. In vitro cultures were establish from four varieties and were subjected to different proliferation conditions. Five individuals from each variety and treatment were analysed using RAPD at the beginning of the treatment and after a month of culture. Variation was detected at both stages of the culture period. The rate of variation found showed differences between varieties, but no significant difference was found between culture conditions. In order to ensure the genetic diversity of the Olea europaea complex,it is necessary to characterize the cultivated varieties and the wildpopulations. In the present study, we focused on the olives growing on twoMediterranean islands, Corsica and Sardinia. On these two islands, there areolives with many denominations, as well as forests of oleasters. Here, it wasproposed to determine the relationships among cultivated and wild olives.Some Italian denominations were studied in addition to assess the influenceof the mainland on the two islands in this respect.The 59 RAPD markers obtained showed the existence of manysynonymous, and homonymous. A dendrogram was constructed using theUPGMA method and a FCA was carried out. The results of these twoanalyses showed the existence of a genetic divergence between the oleastersand the cultivated varieties. They suggest that some of the Corsicanvarieties were probably selected from local wild forms, contrary to theSardinian varieties. They also show that there are feral forms growing onboth islands, which result from hybridization between oleasters andvarieties.  相似文献

The growing number of candidate varieties presented every year at each national Plant Variety Protection Office and their decreasing genetic variability forces strategies to be adopted that will reduce costs without losing rigour when deciding about the acceptance or rejection of a candidate variety. Molecular markers have been envisaged as a reliable tool to establish differences, but can molecular markers be used for assessing distinctness? A comparison between a molecular and a morphological characterization of 41 seed samples belonging to 36 cucumber varieties and including several external controls has been carried out to investigate the applicability of molecular markers in the context of distinctness, uniformity and stability (DUS) tests and the protection of new varieties. Both types of character indicate the same relationships among main groups, Dutch and Beth‐Alpha types, and Gerking and Slice types, the correlation between distance matrices being only 0.6489. Varieties considered uniform for morphological traits proved not to be so for molecular markers. Therefore, molecular characterization is not offering the same estimates of uniformity and relatedness between varieties as does morphological characterization. External controls were used to establish distinction limits with morphological and molecular dendrograms to focus on those varieties, which were suspiciously similar. The results with cucumber show that molecular identity could be used to assess any lack of distinctness and so corroborate the morphological assessment of candidate varieties. The morphological trait ‘type’ provides distinct characterization of varieties and the molecular characterization of candidate varieties could be used to design better field experiments to assess distinction within each group of morphological cucumber type.  相似文献

Genetic variation among 43 date palm (Phoenix dactylifera L.) accessions, including 37 accessions from Morocco and 6 cultivars from Iraq and Tunisia, was studied using Random Amplified Polymorphic DNA (RAPD) markers. The pre-screening of 123 primers on four genotypes allowed selection of 19 primers which revealed polymorphism and gave reproducible results. All 43 analysed genotypes were distinguishable by their band patterns. RAPD technology therefore appears very effective for identifying accessions of date palm. RAPD-based genetic distance was used to determine the relationships between the accessions. The grouping-association identified by cluster analysis was rather weak. However, morphologically similar varieties clustered together. A relatively low polymorphism and a lack of evident organisation are observed among the date palm varieties grown in Morocco. This could be related to the mode of introduction and maintenance of the Moroccan date palm germplasm involving limited foundation germplasm, exchange of cultivars between plantations, and periodic development of new recombinant cultivars following sexual reproduction. DNA from thirty-six cymbidium cultivars was examined using polymerase chain reaction (PCR) to determine the efficiency of randomly amplified polymorphic DNA (RAPD) markers in identifying cultivars and determining levels of genetic variability. A total of 132 RAPD markers, 78% of which were polymorphic, were produced from 15 10mer arbitrary primers. All the cultivars were distinguishable when a number of primers was considered. One cultivar, Blue Smoke ‘Green Meadow’ could be distinguished from all the rest based only on lack of the OPA5-370 fragment. Genetic distances among the cultivars were estimated based on the amount of band sharing and ranged from 0.08–0.50 with an average of 0.29. Cluster analysis of genetic distance estimates grouped siblings together with each other and parents with offsprings, thereby agreeing with known parentage information and corroborating isozyme data obtained from a separate study. The possible application of the observed polymorphism and variation to cymbidium breeding is discussed. Variation in Capsicum annuum revealed by RAPD and AFLP markers   总被引:16,自引:0,他引:16  
Genetic relationships were examined among thirty-four pepper (Capsicum annuum) cultivars of different types. Two types of PCR-based markers were used, RAPD and AFLP, and their relative effectiveness was compared. A dendrogram based on RAPD markers separated the large-fruited sweet cultivars from the small-fruited pungent peppers, and the former group showed less divergence than the latter. The percentage of polymorphic markers was lower for AFLP than for RAPD markers (13 and 22% respectively). However, AFLP primers amplified on average six times more products than RAPD markers. The average numbers of polymorphic products per primer were 1.6 and 6.5 for RAPD and AFLP primers, respectively, i.e., AFLP primers were four times more efficient than RAPD primers in their ability to detect polymorphism in pepper. While four blocky type cultivars were indistinguishable by RAPD, two AFLP primer pairs were sufficient to distinguish the four cultivars from each other. The potential use of the Randomly Amplified Polymorphic DNA (RAPD) technique for characterization and assessment of genetic relationships was investigated in nineteen walnut (Juglans regia L.) genotypes used as parents or released as cultivars from the breeding program of the University of California at Davis. Most of the 72 decamer primers used yielded scorable amplification patterns based on discernable bands. The results obtained produced a unique fingerprint for each of the walnut genotypes studied. Cluster analysis separated the 19 walnut genotypes into two main groups whose differences were related to their pedigree. Genotypes sharing common parents tend to group together and with at least one of the parents. Thus, RAPD markers can detect enough polymorphism to differentiate among walnut genotypes, even among closely related genotypes, and the genetic similarity based on RAPDs appears to reflect the known pedigree information. RAPD technology can be useful in current walnut breeding programs, allowing the identification of new cultivars as well as the assessment of the genetic similarity among genotypes which will help in selecting the best parents to obtain new genetic combinations. This research was conducted to study the genetic diversity in safflower (Carthamus tinctorius L.) using agro-morphological traits and RAPD markers. Sixteen selected lines derived from landraces growing in various agro-climatic regions of Iran along with four exotic genotypes were evaluated in a randomized complete block design with three replications under field conditions. Days to emergence, days to initial flowering, days to flowering, days to maturity, plant height, branches per plant, capitula per plant, seeds per capitulum, 1,000-seed weight, seed yield per plant, seed yield, and reaction to powdery mildew (Leveillula taurica Arnaud) were evaluated in this study. Genetic diversity of the genotypes was assessed by RAPD markers. The results indicated significant differences among genotypes for the agro-morphological traits and clustering based on these traits classified the genotypes into five groups. Analysis of the RAPD markers revealed 15 polymorphic primers out of 50 used primers. Based on RAPD data, the highest genetic similarity was observed between the cultivars of “AC Sunset,” “AC Sterling” from Canada and the lowest relatedness observed between a local breeding line “E2428” and genotype “GE62923” from Germany. Cluster analysis based on RAPD markers and 54% coefficient of similarity divided the genotypes into five distinct groups. Comparing the clusters based on agro-morphological traits with those from molecular markers showed slight similarities. The finding of high genetic variation for agro-morphological traits and polymorphism at DNA level reveal that agronomic traits can be improved by selection programs.  相似文献

Oryza malampuzhaensis, one of the tetraploid taxa of the genusOryza, is geographically restricted to Western Ghats of South India. Its taxonomic status is not well established and is generally treated as a tetraploid race of O. officinalis. Sixty-three morphological traits and 262 random amplified polymorphic DNA (RAPD) markers generated by 23 random decamer primers were used to assess the genetic relationship between O. malampuzhaensis and O. officinalis. Pair wise comparisons based on both RAPDs and morphological traits revealed 60% genetic distance between the two taxa and was significantly higher (p <0.01) than the corresponding intra-specific distances. Cluster and principal component analysis (PCA) of genetic distance estimations based on RAPDs and morphological traits clearly differentiated the two taxa. High frequency of discrete O. malampuzhaensis specific RAPDs (21%) and the significantly higher (p < 0.05) mean number of amplification products per individual in O. malampuzhaensis observed in the study reflect its allopolyploid nature. Low genetic diversity within O. malampuzhaensis revealed by RAPD analysis indicates the recent origin of this taxa. The RAPD analysis further revealed the possibility that the putative ‘C’ genomeprogenitor of O. malampuzhaensis is a close relative of O. officinalis. In addition, amplification products diagnostic to O. malampuzhaensis were identified. The results of the present study clearly demonstrated that the O. malampuzhaensis is a distinct entity, and support the recent conclusion that O. malampuzhaensis has diverged enough to deserve species status. Estimates of genetic diversity can be based on different types of data. The aim of this research were to study genetic diversity among Croatian wheat cultivars by random amplified polymorphic DNA (RAPD) markers, morphological traits and pedigree records; to analyse differences between wheat cultivars from two breeding centres; and to evaluate usability of RAPD markers for estimation of genetic diversity among wheat cultivars in comparison with morphological traits and pedigree record data. Studies were conducted on 14 wheat cultivars and breeding lines from two breeding centres in Croatia. For the RAPD analysis 36 primers were screened and the 14 most polymorphic ones yielded 341 polymorphic bands. Twelve morphological traits were used for morphological analysis. Pedigrees were composed of seven generations of ancestors. RAPD markers showed a high level of polymorphism among the cultivars examined and the breeding lines. No significant correlations were observed among the methods tested.  相似文献

A survey of the genetic diversity among the major cuban rice cultivars was conducted using isozyme, RAPD and AFLP markers. Polymorphisms were detected for esterases, peroxidases, alcohol dehydrogenases and polyphenoloxidases systems; 21 RAPD primers and four AFLP primer combinations. Heterozygosity arithmetic mean value (Hav(p)), the effective multiplex ratio (EMR) and the marker index (MI), were calculated for isozyme, RAPD and AFLP markers. The mean value of genetic similarity among the different varieties was 0.92 for isozyme, 0.73 for RAPD and 0.58 for AFLP analyses. Thus, AFLP were able to detect polymorphisms with higher efficiency than RAPD (+15%) and isozyme (+34%). Data from the isozyme, RAPD and AFLP analyses were used to compute matrices of genetic similarities. The efficiency of the UPGMA for the estimation of genetic relatedness among varieties was supported by cophenetic correlation coefficients. The resulting values indicated that the distortion level for the estimated similarities was minimal. The correlation coefficients obtained by the Mantel matrix correspondence test, which was used to compare the cophenetic matrices for the different markers, showed that estimated values of genetic relationship given for isozyme and RAPD markers (r = 0.89), as well as for AFLP and RAPD markers (r = 0.82) were properly related. However, AFLP and isozyme data showed only moderate correlation (r = 0.63). Although the genetic variability found among the different cultivars was low, both RAPD and AFLP markers proved to be efficient tools in assessing the genetic diversity of rice genotypes. The Lupinus genus includes a number of important crop species. The use of defined nucleotide sequences for the analysis of genetic diversity among these species has revealed modest levels of diversity. The aim of this study was to access AFLP, ISSR and RAPD markers to evaluate the genetic diversity among L. albus, L. angustifolius, L. cosentinii, L. hispanicus, L. luteus, L. mutabilis, L. pilosus and L. polyphyllus. Unexpectedly, low levels of genetic similarity were found (ranging from 0.205 to 0.432), regardless of the type of molecular marker used. Nevertheless, these techniques consistently showed a greater genetic similarity between L. pilosus and L. cosentinii, L. mutabilis and L. polyphyllus and among L. luteus, L. hispanicus and L. angustifolius, clearly separating the Old World from the New World species. Such low genetic similarity among Lupinus spp. is most unlikely to be due to differences in coding sequences but could be the result of a long diverging process concerning non‐coding regions, which would represent a very important proportion of these genomes.  相似文献

The potential use of RAPD markers for taxonomic studies in Malus was investigated using 18 accessions of wild species and 27 apple cultivars. 29 preselected random decamer primers were applied to three sets of Malus genotypes. Random amplified polymorphic DNA (RAPD) ‘fingerprints’ were analysed for polymorphic amplification fragments, and coefficients estimating genetic similarity were calculated on the basis of about 50 polymorphic RAPD loci in each set of genotypes. Cluster analysis by an unweighted pair-group method with arithmetic averages (UPGMA) revealed that, in the cultivars, the molecular classification was in good agreement with the known lineage. A dendrogram generated for the wild species gave relationships that were, in principle, in accordance with the known phylogenetic information. Closely related species from section I were clearly distinguishable from those of sections III and IV. On the molecular level, a high degree of genetic diversity was found among both different apple cultivars and wild species of the genus Malus. The results gave additional evidence for the hypothesis that M. pumila and M. sylvestris were involved in the origin of the cultivated apples.  相似文献

The applicability of random amplified polymorphic DNA (RAPD) markers in the cultivated rubber tree, Hevea, was evaluated using 43 decamer oligonucleotide primers in a set of 24 clones selected in different South-East Asian countries. A total of 220 0.35–3.5 kb DNA fragments were amplified, of which 111 were polymorphic. Of these, 80 fragments (RAPD markers) which were repeatable and clearly scorable across all genotypes were used to estimate genetic distances among the clones tested. The estimated genetic distances ranged from 0.05 (RRII 308 and PB 5/51) to 0.75 (RRIC 100 and SCATC 88–13). A mean genetic distance of 0.5 indicates a rather high genetic variability among the tested clones. As expected, because of the breeding history of Hevea, UPGMA cluster analysis and Principal Coordinate Analysis (PCoA) indicated the absence of a distinct geographical grouping. The possible application of RAPD markers for clone identification and also for analysis of genetic relationships among Hevea clones is discussed.  相似文献

Forty-one apple (Malus × domestica Borkh.) cultivars were screened for RAPD (Random Amplified Polymorphic DNA) and AFLP(Amplified Fragment Length Polymorphism) markers. RAPD analysis was performed with 35 arbitrary 10-mer primers, selected from 60 primers tested (kits A, C and E, Operon Technologies, Inc.). Of a total of 362bands observed, 208 (57.5%) were polymorphic. Three-hundred-and-eighty-one AFLP fragments were obtained with 8primer combinations, of which 218 (57.2%) were polymorphic. Cultivars differentiated through mutation were included in this study and showed identical patterns when analysed with both RAPD and AFLP analysis. The estimated genetic relationships were correlated (r = 73.7%) between the analysis with the two different markers. UPGMA analysis was performed and dendrograms were constructed using either the data apart from each(RAPD and AFLP) method or combined in a single joint matrix. The relationships among the forty-one studied cultivars were basically consistent with the known lineage and geographic origins of the cultivars. The four Portuguese cultivars included in this study clustered together and diverged from the other cultivars. Apparently they constitute an independent genetic pool, which could be of interest for apple plant breeders. Our objectives were to improve the rate of haploid plant regeneration through increasing the rate of callus initiation on the anthers and sustaining shoot regeneration frequency, and to analyze the field population of anther culture origin by morphological and molecular methods. Regarding the callus initiation, the most responsive clones were ‘N-90’(59%) in P. nigra and ‘D-29’ (75%) in P. deltoides. The rate of shoot regeneration and number of shoots/calli ranged from 4%–79% and 1–9, respectively. From the 208 rooted plants 8 haploid, 179 diploid, 4 tetraploid and 17 aneuploid plants were found. In the field population, the haploid plants could be easily identified by their retarded development and morphological characteristics (size and shape of the leaves, strong branching, etc). Several diploid plants showed depressed developmental and morphological traits similar to the haploid ones. Three traits (growth rate, leaf blade length and shape of leaf base) of the 6 different morphological characteristics measured were in correlation with the ploidy level within the poplar field population.
