Although avian species are known to be susceptible to infection with Mycobacterium spp. organisms, much remains unknown about the susceptibility of birds to infection with M. bovis. The objective of this current study was to determine if wild turkeys (Meleagris gallopavo) can be infected with M. bovis when inoculated by the oral or intratracheal route. Six turkeys were orally inoculated and another six were inoculated via the trachea with a high dose of M. bovis, 1 x 10(5) CFU/ml. Six turkeys were sham-inoculated controls. Two turkeys from each treatment group were sacrificed on days 30, 60, and 90 postinoculation. There were no gross or microscopic lesions consistent with mycobacteriosis in the 23 inoculated turkeys over the 90-day duration of this study. Fecal cultures were also consistently negative for M. bovis when sampled before inoculation and on days 1, 30, and 60 postinoculation. Two intratracheally inoculated turkeys were positive for M. bovis in visceral tissues at 30 days postinoculation. However, this finding was only indicative of passive persistence of mycobacteria in the tissues and not of infection, as there were no attendant lesions or clinical compromise to support infection. Thus, it can be concluded that young wild turkeys are resistant to infection with M. bovis and, therefore, pose minimal threat as reservoir or spillover hosts for this organism. 相似文献

During fattening, the bodyweight of modern broad-breasted turkeys increases considerably within a very short space of time. In particular, the breast muscles increase disproportionately. This leads to a disadvantageous distribution in weight, and as a consequence, to a disturbed leg position and skeletal deformations like antitrochanteric degeneration, tibial dyschondroplasia, bending, twisting and rotation of the tibia, osteochondrosis, osteomyelitis, rickets, and epiphyseolysis of the femoral head increases. This cases of degenerative joint disease cause severe pain in humans and there are indications that this is also true for turkeys. The purpose of this study was to determine if behaviour indicative of such pain in turkeys of the B.U.T. Big 6 breeding line could be attenuated by administering a quick-acting analgesic, butorphanol. Twelve pairs of turkeys were tested at the ages of 7 and 12 weeks. One bird in each pair received an analgesic opioid injection, while the other one received a control injection of physiologically balanced saline solution. The time the birds spent putting weight on their legs, i.e., 'walking' and 'standing' and the distance covered by the birds were recorded during the 30 min periods before and after the application of the drug. At week seven the treated birds spent significantly more time putting weight on their legs than control birds. At week 12, the same tendency was observed. No significant differences were found in the distances covered by the animals. It is concluded that fattening turkeys reduce the time they are putting weight on their legs because these behaviours may be associated with pain. 相似文献

Thirteen lectins were used to characterize lectin-binding specificity of glycoconjugates on sections of formalin-fixed lung and trachea from seven normal turkeys, two turkeys with acute pneumonia, and two turkeys with chronic pneumonia. Neuraminidase was used to digest sialic acid residues. One N-acetylgalactosamine-binding lectin and two N-acetylgalactosamine/galactose-binding lectins stained the apical membrane and cytoplasm of multifocal cells that lined air atria and hyperplastic granular cells. Other lectins in these groups stained ciliated cells of the trachea and bronchi and air capillary epithelial cells. Sialic acid residues were on apical surfaces of ciliated and nonciliated tracheal and bronchial lining cells, air capillary epithelial cells, and vascular endothelial cells. Mannose/glucose-binding lectins stained reticular and elastic fibers in the lamina propria of trachea, primary and secondary bronchi, and the tunica adventitia of arteries and veins. By transmission electron microscopy, colloidal gold-Arachis hypogaea (peanut agglutinin) labeled microvilli on the apical surface of mature granular cells. The L-fucose-binding lectin, in addition to several other lectins, stained nonspecifically in both trachea and lung. These studies show that granular cells that line air atria can be identified with lectins of N-acetylglucosamine and N-acetylgalactosamine/galactose groups, and that apical surfaces of epithelial cells and endothelial cells in the trachea and lung express terminal sialic acid residues. 相似文献

1. The influence of age at thyroidectomy on reproductive activity of male turkeys was investigated. 2. Thyroidectomy of 53-week-old male domestic turkeys that had been reproductively active for 25 weeks resulted in undetectable plasma thyroxine concentrations, termination of semen production and testicular regression. Daily light-dark treatment was 14L:10D before and after thyroidectomy. 3. Thyroidectomy of 28-week-old, reproductively inactive, males maintained on a daily light-dark cycle of 8L:16D resulted in increased semen volumes following photostimulation with 14L:10D at 32 weeks of age. Semen quality was not affected, while plasma thyroid hormone concentrations were undetectable. The moult and termination of semen production that occurred in response to changing the daily light-dark cycle from 14L:10D to 8L:16D near the end of the study were inhibited by thyroidectomy. 4. The requirement for the thyroid glands in the reproductive processes of male turkeys changes either with age or after an extended period of reproductive activity. 相似文献

Viscera of 49 wild turkeys (Meleagris gallopavo) collected in the spring of 2001 and 23 wild turkeys collected in the fall and winter of 2001-02 from 12 counties in eastern Kansas were examined for enteric helminths. Four cestode species, two trematode species, one nematode species, and one acanthocephalan species were identified. Two cestode and two trematode species present in the spring sample also were present in the fall and winter sample. Parasite prevalence was similar to previous studies of enteric helminths of wild turkeys except for the low numbers of nematode species and individuals recovered in the present study. 相似文献

Two experiments were conducted to investigate the differences in susceptibility to foot pad dermatitis (FPD) of two medium-heavy lines of turkeys, and whether FPD is painful, by detailed analysis of behaviour in birds with and without analgesic treatment (betamethasone). Turkeys housed on dry litter in the first experiment generally had more frequent bouts of different behaviours that were of shorter duration than birds on wet litter. T-patterns (behavioural sequences) were more frequent, varied and complex on dry than on wet litter. Betamethasone-injected birds of line B, but not breed A, had shorter resting and longer standing durations on wet litter than saline-injected birds. In the second experiment, turkeys on wet litter given saline stood less and rested more than all other treatment groups, suggesting that they experienced pain that was alleviated in birds receiving betamethasone. Turkeys on dry litter had more frequent, varied and complex patterns of behaviour than turkeys on wet litter and birds kept on intermediate litter wetness. Betamethasone provision increased pattern variety regardless of litter treatment. Turkeys with low FPD scores transferred to wet litter and given saline injections had a longer total duration of resting and shorter duration of standing compared to betamethasone-treated birds. Low FPD birds transferred to wet litter had a similar number of patterns and total pattern occurrence as high FPD birds transferred to dry litter. Betamethasone increased pattern variety and frequency compared to saline injections whereas overall pattern complexity was similar. It was concluded that wet litter affects the behaviour of turkey poults independently of FPD and that betamethasone may also change the behaviour of turkeys. There was some evidence from analgesic treatment and T-pattern analyses that FPD was painful. However, there was no evidence of differences in susceptibility to FPD of the two commercial hybrids. 相似文献

Nuclear inclusion bodies typical of the adenovirus group were widespread in in the spleen and other tissues of 8-week-old turkeys with severe respiratory disease and concomitant evidence of colisepticemia. Adenoviral virions were seen in affected nuclei of splenic tissue and in negatively stained preparations of ground spleen. In splenic tissue, inclusions were most prominent in reticular cells and macrophages in the periarterial lymphoid sheaths, the red pulp and the marginal zones of the periarteriolar reticular sheaths. Marked reticuloendothelial hyperplasia, lymphoid atrophy and granulocytic splenitis characterized the splenic changes. There were inclusions in the respiratory tract, intestinal tract, liver, kidney and pancreas. Inoculation of young turkeys, especially when immunosuppressed, resulted in evidence of infection and respiratory disease. The viruses produced cytopathic changes in primary turkey kidney cell cultures but did not affect embryonating chicken eggs. Concentrated viral suspensions induced precipitin lines in agar gel immunodiffusion tests with known antisera against known turkey adenoviruses but did not show an antigenic relationship to chicken adenoviruses. 相似文献

1. The photopic spectral sensitivity of domestic ducks and turkeys was determined using an operant psychophysical technique. Spectral sensitivity was determined over a range of specified wavelengths, including UVA, between 326 < lambda < 694 nm and the results were directly compared with human spectral sensitivity measured under similar experimental conditions. 2. Domestic ducks and turkeys had similar spectral sensitivities to each other, and could perceive UVA radiation, although turkeys were more sensitive to UVA than ducks. For both species, peak sensitivity was between 544 < lambda < 577 nm, with reduced sensitivity at lambda = 508 and 600 nm. Both ducks and turkeys had a very different and broader range of spectral sensitivity than the human subjects tested. 3. Spectral sensitivity and UVA perception in these avian species are discussed in relation to their visual ecology and the mechanisms controlling neural processing of colour information. 相似文献

To test the hypothesis that walls of air capillaries are a site for Escherichia coli to pass the air-blood barrier, fimbriated and nonfimbriated strains of E. coli were inoculated intratracheally into 18-day-old turkeys. Venous blood was cultured, and turkeys were necropsied from 0.5 to 8 hours post-inoculation. Lungs were processed for histopathology and electron microscopy. E. coli 078 was identified ultrastructurally using rabbit anti-lipopolysaccharide antibody and protein A-colloidal gold. All birds developed bacteremia; there was no significant difference between groups given fimbriated or nonfimbriated bacteria. Bacteria adhered to the plasma membrane of air capillary epithelial cells and were seen within vacuoles of portions of these cells that lined the fornices of air capillaries. Bacteria were also seen in the basement membrane at the basal surface of air capillary epithelial cells and, rarely, in vacuoles of subjacent endothelial cells. Infected granular and non-granular cells that lined air atria were necrotic 4 hours post-inoculation. Bacteria were within the overlying trilaminar substance and between reticular fibers of the interstitial stroma and pleura at 30 minutes post-infection and thereafter. Thus, the pulmonary air capillaries are a site for entrance of E. coli into the pulmonary blood capillaries, but fimbriae play little or no role in passage across the air-blood barrier. 相似文献

Heart, aorta, and sciatic arteries were collected from 157 wild male turkeys (Meleagris gallopavo silvestris) by hunters in 9 states during the spring 1983 and 1984 hunting seasons. In descending order of extent and severity, intimal vascular changes were observed in the left sciatic artery, aorta at the celiac region, cranial abdominal portion of the aorta, sciatic bifurcation, caudal abdominal portion of the aorta, coronary arteries, and thoracic portion of the aorta. Only the aorta from the celiac region and right sciatic artery had significant differences (P less than 0.05) among turkeys from various locations. Turkeys from Indiana had significantly (P less than 0.05) larger plaque scores in the celiac region than did those from Alabama, Missouri, and South Carolina. Turkeys from Indiana also had significantly (P less than 0.05) greater plaque scores in the right sciatic artery than did turkeys from Arkansas. When all tissues were considered, tissues from turkeys from Michigan had the highest plaque scores and those from Iowa had the lowest. Plaque scores for turkeys from Michigan were significantly (P less than 0.05) higher overall (including all blood vessels) than were plaque scores for turkeys from Alabama, Arkansas, Iowa, and South Carolina. Few significant (P less than 0.05) correlations were detected among plaque scores in turkeys from within states of origin (geographic location). Also, only a few significant (P less than 0.05) correlations were determined between age or body weight and atherosclerosis for blood vessels from turkeys within various states. 相似文献

1. Sexual size dimorphism (SSD) is a common phenomenon in animals. Rensch’s rule states that larger species generally exhibit a higher male to female body size ratio than smaller ones. 2. Domesticated animals offer excellent opportunities for testing predictions of the functional explanations of Rensch’s rule and this was tested in a meta-analysis of SSD in 38 breeds of domestic geese compared among themselves and with their wild relatives (subfamily Anserinae, 35 species). 3. Domestic geese and wild Anser species taken together supported Rench’s rule but the wild species did not. 4. Genetic parameters for production and feed efficiency traits in the Orlopp line of turkeys were estimated to determine breeding goals and future potential of the line in a long-term genetic improvement programme. 2. Body weight, egg production and fertility traits were recorded and feed conversion ratio (FCR) was assessed from 16–20 weeks of age. 3. Moderate heritabilities were found for feed intake and body weight gain (0.25 to 0.31). Average FCR was 3.14, with heritability of 0.10. Body weight, breast conformation score and egg production traits showed moderate heritabilities (0.22 to 0.52), while both fertility and hatch of fertile eggs were low (0.04 and 0.09, respectively). 4. Genetic correlations between breast confirmation score, 10- and 18-week body weights were moderate, 0.50 and 0.45, respectively. Aspergillus fumigatus remains a major respiratory pathogen in birds and treatment is still difficult. We challenged different groups of few-day-old turkeys via intratracheal aerosolisation with increasing concentrations (10 5 up to 10 8) of conidia using a MicroSprayer ® device. The fungal burden was assessed by real-time PCR, galactomannan dosage, CFU counting and histopathological evaluation in order to provide a comparison of these results within each inoculum groups. Significant mortality, occurring in the first 96 h after inoculation, was only observed at the highest inoculum dose. Culture counts, GM index and qPCR results on the one hand and inoculum size on the other hand appeared to be clearly correlated. The mean fungal burden detected by qPCR was 1.3 log 10 units higher than the mean values obtained by CFU measurement. The new model and the markers will be used to evaluate the efficacy of antifungal treatments that could be used in poultry farms. 相似文献

To show hormonal differences between male turkeys with yellow semen syndrome (YSS) and white, normal semen (WNS), the expression of aromatase, oestrogen receptor α (ERα), and oestrogen receptor β (ERβ) as well as testosterone and oestradiol concentrations in YSS and WNS testes, epididymis, and ductus deferens were examined.2. To measure gene expression levels of aromatase and oestrogen receptors (ERs), three complementary techniques (real-time PCR, Western blot, and immunohistochemistry) were used, whereas steroid hormone levels were determined radio-immunologically.3. Upregulation of aromatase and ERα mRNAs in YSS testes ( P < 0.05; P < 0.01), epididymis ( P < 0.001; P < 0.001), and ductus deferens ( P < 0.05; P < 0.01) compared to those of WNS tissues was detected. Significant increases in the levels of aromatase and ERα proteins were detected in YSS testes ( P < 0.001; P < 0.05), epididymis ( P < 0.001; P < 0.001), and ductus deferens ( P < 0.001; P < 0.05). The expression of ERβ mRNA and protein level was upregulated in the testes ( P < 0.05; P < 0.01) and epididymis ( P < 0.001; P < 0.01) but not in ductus deferens where it was downregulated ( P < 0.01; P < 0.01). Increased intensity of immunoreactive proteins in YSS versus WNS reproductive tissues corroborated gene expression results.4. Testosterone concentration diminished in YSS epididymis ( P < 0.05) and ductus deferens ( P < 0.05), but not in the testes, remaining at high level ( P < 0.05) compared to WNS values. Concomitantly, increased oestradiol concentration was found in YSS testes ( P < 0.05) and epididymis ( P < 0.05) but decreased in the ductus deferens ( P < 0.05).5. From the published literature, this study is the first to demonstrate the ability for androgen aromatisation in the turkey reproductive tissues and to show the cellular targets for locally produced oestrogens. The data suggested that the androgen/oestrogen ratio is a mechanistic basis for amplification of differences between turkeys with white and yellow semen and that these results can have a relevance in applied sciences to widen the knowledge on domestic bird reproduction. 相似文献

1. Economic requirements have encouraged farmers to select broad-breasted strains of turkeys (Meleagris gallopavo). These birds are very heavy (more than 30 kg), have pectoral hypertrophy and are frequently lame. Traditional turkeys, closer to wild birds, weigh 9 kg on average. 2. Gaits, which are motor pattern expressions, seem to be linked to species morphology. However, no significant differences are evident between the gaits of traditional and broad-breasted strains. 3. Yet, posterior views of the walk of both strains show lateral oscillations, which are very slight in the traditional one but very marked in the broad-breasted one. 4. In the latter, modification of the centre of gravity path may increase the energetic cost of walking and be associated with problems of lameness. 5. Therefore, if the genetic pool permits an increase in weight of broad-breasted turkeys, then extrinsic factors, such as gravity constraints, may limit this evolution. 相似文献

Vertebrate long bones are formed by endochondral ossification, a process accompanied by changes in extracellular matrix synthesis and remodeling, performed mainly by the matrix metalloproteinases (MMP). The temporal/spatial expression patterns of 5 members of the MMP family known to be important for endochondral ossification were studied, for the first time, in the turkey growth plate during embryonic and juvenile stages. The expression of MMP-2 was detected in the proliferative zone, MMP-3, MMP-9, and MMP-13 in cells lining the blood vessels; MMP-13 was also detected in hypertrophic chondrocytes. The MMP-16 expression was detected in the reserve zone of the growth plate. These results present a detailed survey of turkey MMP, serving as a data source (atlas) for further studies in this subject. 相似文献
