AIM: To investigate the effect of DEK downregulation on the apoptosis of gastric carcinoma SGC-7901 cells, and to explore its associations with NF-κB signaling pathway and apoptosis related proteins. METHODS: SGC-7901 cells with different treatments were divided into 3 groups including untreated group, control siRNA group and DEK siRNA group. The expression of DEK at mRNA and protein levels in the SGC-7901 cells was detected by real-time PCR and Western blot. The cell apoptosis was examined by flow cytometry. Furthermore, the activities of caspase-3 and caspase-9 in the SGC-7901 cells were investigated by Caspase-Glo®-3/9 kit. Finally, the expression of key regulatory protein p65 of NF-κB signaling pathway and apoptosis-related proteins Bcl-2 and Bax in the SGC-7901 cells was investigated by Western blot. RESULTS: Compared with untreated group and control siRNA group, the expression of DEK at mRNA and protein levels was significantly downregulated in DEK siRNA group (P<0.05). In addition, the ratios of early phase apoptosis and total apoptosis in DEK siRNA group were markedly higher than those in untreated group and control siRNA group (P<0.05). Most notably, the decrease in p65 and Bcl-2 proteins, increase in Bax protein and the increases of caspase-3 and caspase-9 activities were observed in DEK siRNA group. CONCLUSION: Downregulation of DEK mediates cell apoptosis of gastric carcinoma may be tightly associated with NF-κB signaling pathway. 相似文献

AIM: To investigate the effect of histone deacetylase 1 (HDAC1) silencing on apoptosis of squamous cell carcinoma of skin. METHODS: Skin squamous cell carcinoma A431 cells were transfected with HDAC1 small interfering RNA (HDAC1 siRNA) or small interfering RNA negative control (siRNA NC). The expression levels of HDAC1 in transfected cells were detected by RT-PCR and Western blot. The cell viability was measured by MTT assay, and the apoptosis was analyzed by flow cytometry. The protein levels of STAT3, p-STAT3 and cleaved caspase-3 were determined by Western blot. The inhibitor of STAT3 signaling pathway was used to treat the A431 cells transfected with HDAC1 siRNA. The cell viability was detected by MTT assay, the apoptosis was analyzed by flow cytometry, and the protein levels of STAT3, p-STAT3 and cleaved caspase-3 were determined by Western blot. RESULTS: HDAC1 siRNA inhibited the expression of HDAC1 at mRNA and protein levels in the A431 cells. After interfering with the expression of HDAC1, the cell viability and the protein level of p-STAT3 in the cells decreased, while the apoptotic rate and the protein level of cleaved caspase-3 in the cells were increased. After treatment with the inhibitor of STAT3 pathway, the viability of A431 cells transfected with siRNA and the protein level of p-STAT3 decreased, while the apoptotic rate and the protein le-vel of cleaved caspase-3 in the cells were increased. CONCLUSION: Interference with HDAC1 expression may regulate the STAT3 signaling pathway to inhibit the viability of skin squamous cell carcinoma cells, thus promoting the apoptosis of squamous cell carcinoma of skin. 相似文献

AIM: To investigate whether indomethacin induces gastric cancer cell apoptosis through Akt/GSK3β/NAG-1 pathway.METHODS: Gastric cancer cell line MGC-803 was used in the study. Cell viability was measured by MTT method. Hoechst 33258 nuclear staining and flow cytometry analysis were used to determine apoptosis. The protein expression level was examined by Western blotting. RESULTS: Indomethacin induced MGC-803 cell apoptosis via caspase-dependent pathway. Indomethacin inhibited Ser473-Akt and Ser9-GSK3β phosphorylation and up-regulated the expression of non-steroidal anti-inflammatory drug-activated gene-1 (NAG-1). Inhibition of PI3K or Akt alone also increased NAG-1 expression. Moreover, the effect of indomethacin on NAG-1 expression was abolished by pretreatment of the cells with GSK3β inhibitor SB216763. CONCLUSION: Indomethacin induces gastric cancer cell apoptosis through Akt/GSK3β/NAG-1 pathway. 相似文献

AIM:To investigate the effects of nodosin on apoptosis of human hepatocellular carcinoma HepG2 cells. METHODS:HepG2 cells were treated with nodosin at different concentrations (1.25 μmol/L, 2.5 μmol/L, 5 μmol/L, 10 μmol/L and 20 μmol/L) for 24 h. The morphological changes of the HepG2 cells were observed by Hoechst 33258 staining and electron microscopy. The apoptotic rates were analyzed by flow cytometry. The mRNA expression of apoptotic protease-activating factor-1 (Apaf-1) was detected by RT-qPCR. The protein levels of pro-caspase-3, caspase-3 and cleaved caspase-3 were determined by Western blot. RESULTS:HepG2 cells showed obvious cell shrinkage and nucleus drift when treated with nodosin as the concentration was increased. Many apoptotic bodies were observed in 5 μmol/L, 10 μmol/L and 20 μmol/L nodosin groups. The mRNA expression of Apaf-1 was increased in 5 μmol/L, 10 μmol/L and 20 μmol/L nodosin groups as compared with control group (P<0.05). The protein levels of pro-caspase-3, caspase-3 and cleaved caspase-3 were increased with the increasing dose of nodosin (P<0.05). CONCLUSION:Nodosin induces the apoptosis of HepG2 cells. This effect was related to increasing Apaf-1 mRNA expression and subsequently promoting the activation of caspase-3. 相似文献

AIM:To explore the effect of dengue virus type 2 (DENV-2) infection on the change of mitochondrial membrane potential (Δψm) in EA.hy926 cells. METHODS:The inhibitory effect of DENV-2 infection on EA.hy926 cell growth was examined by MTT assay. The changes of Δψm were analyzed by flow cytometry or observed under fluorescence microscope with JC-1 staining. The activity of caspase-9 was measured by a colorimetric kit. RESULTS:Infection of DENV-2 for 24 h, 36 h and 48 h inhibited the viability of EA.hy926 cells. After DENV-2 infection, the changes of Δψm in EA.hy926 cells were observed. Compared with the normal control cells, Δψm in DENV-2-infected EA.hy926 cells was notably decreased. The activity of caspase-9 increased at early stage after infection of DENV-2 and maintained at a high level at least to 48 h. CONCLUSION:DENV-2 infection decreases the mitochondrial membrane potential and increases the activity of caspase-9 in EA.hy926 cells in the early stage of proliferation, thus promoting the process of apoptosis. 相似文献

AIM: To study the effect of forkhead box protein M1 (FoxM1) silencing on apoptosis of oral squamous-cell carcinoma. METHODS: Oral squamous-cell carcinoma SCC9 cells were infected with FoxM1-shRNA lentivirus or negative control lentivirus. The silencing effect was measured by RT-qPCR and Western blot. The changes of cell viability effect was measured by MTT saay. The cell colony formation ability was measured by plate experiment. Flow cytometry was used to analyze the changes of apoptotic rate. Western blot was used to measure the protein levels of cleaved caspase-3 and cleaved caspase-9 in the cells. The changes of mitochondrial membrane potential were measured by JC-1 method. Western blot was used to measure the protein level of cytochrome C in the mitochondria and cytoplasm. RESULTS: Infection with FoxM1-shRNA lentivirus successfully reduced the expression of FoxM1 in oral squamous-cell carcinoma cells (P<0.05). Negative control lentivirus had no effect on the expression level of FoxM1 in the cells. The cell viability was reduced by FoxM1 silencing, and the ability of cell colony formation was also decreased. The apoptotic rate and the protein levels of cleaved caspase-3 and cleaved caspase-9 were all increased (P<0.05), and the mitochondrial membrane potential was decreased. The protein level of cytochrome C in the cytoplasm was increased, while the protein level of cytochrome C in the mitochondria was decreased (P<0.05). CONCLUSION: Silencing of FoxM1 induces the apoptosis of oral squamous-cell carcinoma cells by decreasing the mitochondrial membrane potential and promoting the release of cytochrome C from mitochondria. 相似文献

AIM: To study the effect of epigallocatechin gallate (EGCG) on the growth and apoptosis of ova-rian cancer cell line SKOV-3 and its molecular mechanism. METHODS: SKOV-3 cells were treated with different concentrations of EGCG (0~50 μmol/L), SRT1720 (1 μmol/L) or EX527 (1 μmol/L) for 24 h. The cell activity was evaluated by CCK-8 assay. The apoptosis of SKOV-3 cells was analyzed by flow cytometry. The mRNA expression of Bcl-2 and Bax was detected by real-time PCR. SIRT1 deacetylase fluorometric assay kit was used to detect the activity of SIRT1. The protein levels of SIRT1 and acetylated P53 (Ac-P53) were determined by Western blot. RESULTS: EGCG or EX527 decreased the deacetylase activity and protein expression of SIRT1, and increased the level of Ac-P53 in a dose-dependent manner. Moreover, SRT1720 abrogated the effects of EGCG on the activity, apoptosis and SIRT1-P53 pathways in ovarian cancer cell line SKOV-3. CONCLUSION: EGCG inhibits the activity and induces apoptosis of ovarian cancer cell line SKOV-3 by regulating SIRT1-P53 pathways. 相似文献

AIM: To investigate whether quercitrin induces apoptosis of gastric cancer cell line SGC7901 by inhibition of PI3K/AKT signaling pathway. METHODS: The human gastric cancer SGC7901 cells were selected as the research object. The cytotoxicity of quercitrin was detected by MTT assay, and IC50 value of quercitrin was calculated. The SGC7901 cells were divided into control group, quercitrin group (incubated with 200 μmol/L quercitrin), insulin-like growth factor-1 (IGF-1) group (incubated with 100 μg/L IGF-1) and quercitrin+IGF-1 group (incubated with 200 μmol/L quercitrin and 100 μg/L IGF-1). After 48 h, the apoptosis of SGC7901 cells was analyzed by flow cytometry, and the protein levels of cleaved caspase-3, p-AKT (Ser473), AKT, p-PI3K (Tyr508) and PI3K were determined by Western blot. RESULTS: The viability of SGC7901 cells was significantly decreased as the concentration of quercitrin increased, starting at 100 μmol/L (P<0.05). The IC50 value of quercitrin for 48 h was 275.40 μmol/L. After treatment with 200 μmol/L quercitrin for 48 h, the apoptosis rate and the protein level of cleaved caspase-3 in quercitrin group were significantly increased (P<0.05), and the phosphorylated levels of AKT and PI3K were significantly decreased compared with control group (P<0.05). Treatment with quercitrin and IGF-1 inhibited the effect of quercitrin on SGC7901 cells compared with quercitrin group. CONCLUSION: Quercitrin may induce apoptosis of gastric cancer cell line SGC7901 by inhibiting the activation of PI3K/AKT signaling pathway. 相似文献

AIM:To study the effects of fucoidan sulfate on the apoptosis of gastric cancer cells, and to explore its relationship with histone deacetylase 1 (HDAC1). METHODS:The effects of fucoidan sulfate on the viability of gastric cancer BGC-823 cells were measured by MTT assay. The cells were divided into si-HDAC1 group, si-NC group and fucoidan+pcDNA 3.1-HDAC1 group and fucoidan+pcDNA 3.1 group. The transfection were performed by liposome method. The apoptosis was analyzed by flow cytometry. The protein expression of HDAC1, Bcl-2 and Bax was determined by Western blot. The mRNA expression of HDAC1 was detected by RT-qPCR. RESULTS:Compared with the control cells, the inhibitory rates of the cell viability by fucoidan sulfate (0.2, 0.4, 0.6, 0.8 and 1.0 g/L) were increased signi-ficantly (P<0.05). The appropriate concentration was 0.6 g/L. Compared with control group, the expression of HDAC1 at mRNA and protein levels in fucoidan sulfate group was significantly down-regulated, while the apoptotic rate was significantly increased (P<0.05). Knock-down of HDAC1 expression significantly up-regulated the apoptotic rate, while over-expression of HDAC1 reversed the apoptosis-promoting effect of fucoidan sulfate on the BGC-823 cells. CONCLUSION:Fucoidan sulfate promotes apoptosis of gastric cancer cells. The mechanism may be related to the direct inhibition of HDAC1, which provides support for the clinical application of fucoidan sulfate. 相似文献

AIM:To investigate the molecular mechanisms of apoptosis and to elucidate the apoptosis signaling pathway triggered by etoposide in Jurkat human leukemia cells. METHODS:Apoptosis was detected using annexin V-FITC and propidium iodide (PI) staining, respectively, and annexin V-FITC positive cells and hypodiploid cells were analyzed by flow cytometry. Mitochondrial membrane potential (△Ψm) was detected using 3, 3-dihexyloxycarbocyanine iodide [DiOC6(3)] staining and △Ψm low cells were analyzed by flow cytometry. Preparation of cytosolic extracts and isolation of mitochondria were completed by centrifugation. Western blotting analysis was used to evaluate the level of cytochrome c, caspase-3, and poly (ADP-ribose) polymerase (PARP) expression. RESULTS:Etoposide induced apoptosis showing phosphatidylserine externalization and DNA fragmentation in a time-dependent manner and the apoptosis could be inhibited by a broad caspase inhibitor benzyloxycarbonyl-Val-Ala-Asp-fluoromethylketone (zVAD.fmk). Collapse of △Ψm induced by etoposide preceded DNA fragmentation and phosphatidylserine externalization. In contrast, it was not blocked by zVAD.fmk. Etoposide caused cytochrome c release from mitochondria into cytosol, subsequent activation of caspase-3 (32 kD) presented with an intermediate (20 kD) and its active product (17 kD), and cleavage of full-length PARP (116 kD) into the so-called apoptotic 85 kD fragment. CONCLUSION:Etoposide-induced Jurkat cell apoptosis is initiated through mitochondria signaling pathway with cytochrome c release into cytoplasm and caspase is the ultimate executioner of cell apoptosis. 相似文献

AIM: To investigate the role of NF-κB in apoptosis induced by 10-hydroxycamptothecin (HCPT) in human breast carcinoma cells. METHODS: The cell growth inhibition was measured by MTT assay. Agarose gel electrophoresis was performed for cell apoptosis. Western blotting was performed for protein xpression. DIG-EMSA was conducted to determine the DNA-binding activation of NF-κB. RESULTS: HCPT had a remarkable effect of growth inhibition on breast cancer cells. After exposed to HCPT, the breast cancer cells presented some morphologic features of apoptosis including cell shrinkage, nuclear condensation and DNA fragmentation. NF-κB was activated in cancer cells treated with HCPT but not activated in IκBα-mutant cells and apoptosis was inhibited in IκBα-mutant cells treated with HCPT.CONCLUSION: HCPT may induce apoptosis and activation of NF-κB in a Bcap-37 cell line. Inhibition of NF-κB activation attenuates apoptosis induced by HCPT. 相似文献

AIM: To investigate whether sphingosine kinase 1 (SPK1) interferes with apoptosis of Lewis lung cancer (LLC) cells by regulating the Bcl-2/Bax pathway. METHODS: The SPK1 gene siRNA eukaryotic expression vector was constructed, and transfected into the LLC cells. The transfected LLC cells was observed under a fluorescence microscope. The apoptotic rate of LLC cells after transfection was analyzed by flow cytometry. The expression levels of SPK1, Bcl-2 and Bax in LLC cells after transfection were detected by Western blot. The protein levels of Bax and Bcl-2 were measured by ELISA. RESULTS: Transfected LLC cells emitted green fluorescence under a fluorescence microscope. Apoptosis in siRNA-SPK1 group was significantly higher than that in siRNA-SPK1-Neg group (P<0.01). Western blot analysis showed that the expression of Bax in siRNA-SPK1 group was significantly higher than that in siRNA-SPK1-Neg group, and the expression of Bcl-2 was lower than that in siRNA-SPK1-Neg group. The ELISA results showed that the protein level of Bax in siRNA-SPK1 group was significantly higher than that in siRNA-SPK1-Neg group (P<0.01), and the protein level of Bcl-2 in siRNA-SPK1 group was significantly lower than that in siRNA-SPK1-Neg group (P<0.01). CONCLUSION: The expression of SPK1 in LLC cells is related to the apoptotic rate. SPK1 may interfere with the apoptosis of LLC cells via Bcl-2/Bax pathway. 相似文献

AIM:To investigate the effect of TRIM29 gene expression silencing on the apoptosis and PI3K/AKT signaling pathway in human nasopharyngeal carcinoma 5-8F cells. METHODS:The 5-8F cells were divided into blank group, negative control (NC) group (transfected negative control siRNA) and si-TRIM29 group (transfected TRIM29 specific siRNA). The viability of the 5-8F cells transfected with si-TRIM29 for 0~96 h was measured by CCK-8 assay. The apoptotic rate and the protein levels of TRIM29, cleaved caspase-3, cleaved caspase-9, Bcl-2, Bax, t-AKT and p-AKT in the 5-8F cells transfected with si-TRIM29 for 48 h were determined by flow cytometry and Western blot, respectively. PI3K/AKT signal specific inhibitor LY294002 at 10 μmol/L and si-TRIM29 alone or in combination were treated with the 5-8F cells, and the cells were divided into blank group, LY294002 group and LY294002+si-TRIM29 group. The apoptotic rates in the 3 groups were detected by flow cytometry. RESULTS:The protein expression of TRIM29 in the 5-8F cells transfected with TRIM29 siRNA was significantly lower than that in blank group (P<0.05). Compared with blank group, the cell viability was significantly decreased, the apoptotic rate was significantly increased, the protein levels of cleaved caspase-3, cleaved caspase-9 and Bax were significantly increased, and the protein levels of Bcl-2 and p-AKT were significantly decreased in si-TRIM29 group (P<0.05). The apoptotic rate in LY294002 group was higher than that in blank group, while that in LY294002+si-TRIM29 group was even higher than that in LY294002 group (P<0.05). CONCLUSION:Silencing of TRIM29 gene expression induces apoptosis of nasopharyngeal carcinoma 5-8F cells by inhibiting PI3K/AKT signaling pathway. 相似文献

AIM:To explore the effect of low density lipoprotein from the patients of diabetes mellitus (DM-LDL) on the activation of caspase-12 an important molecule in endoplasmic reticulum stress (ERS)-associated apoptotic pathway, in the murine macrophages, and to clarify the underlying molecular mechanisms of apoptosis. METHODS:Murine macrophage RAW264.7 was exposed to DM-LDL (25, 50 and 100 mg/L), normal low density lipoprotein (n-LDL, 50 mg/L), or tunicamycin (TM, 4 mg/L) for 24 h. Additionally, RAW264.7 macrophages were precultured with 4-phenylbutyric acid (PBA, 5 mmol/L) for 1 h and then exposed to DM-LDL (100 mg/L) for 24 h. The cell viability and apoptosis were detected by MTT assay and flow cytometry with Annexin V-FITC/propidium iodide staining, respectively. Lactate dehydrogenase (LDH) activity in the media was measured by assay kit. The protein level of caspase-12 was determined by Western blot. RESULTS:Similar to TM (an ERS inducer), treatment with DM-LDL caused significantly decrease in the viability and increase in LDH activity in the media and apoptotic rate of the RAW264.7 macrophages (P<0.05). Additionally, DM-LDL induced activation of caspase-12 especially at the dose of 50 and 100 mg/L (P<0.01). However, the ERS inhibitor PBA protected RAW264.7 macrophages from DM-LDL-induced decrease in viability and increase in LDH activity and apoptosis (P<0.05). Furthermore, PBA attenuated DM-LDL-induced activation of caspase-12 (P<0.05). CONCLUSION:DM-LDL may induce apoptosis in RAW264.7 macrophages, and the mechanism may be related to the activation of caspase-12. 相似文献

AIM: To investigate whetier resveratrol induces apoptosis of human ovarian cancer SKOV3 cells through Sirt3-SOD2-ROS pathway. METHODS: SKOV3 cells were cultured in vitro and treated with resveratrol at 0, 2.5, 5, 10, 20, 40 and 80 mg/L for 24 h. The inhibitory effect of resveratrol on the viability of SKOV3 cells was measured by MTT assay. SKOV3 cells were randomly divided into blank control group, 10 mg/L resveratrol group, 20 mg/L resveratrol group and 40 mg/L resveratrol group. After 24 h of treatment, Hoechst 33342 staining and confocal microscopy were used to observe the nuclear changes. The protein levels of silent mating type information regulation 2 homolog 3 (Sirt3), superoxide dismutase 2 (SOD2), Bcl-2, Bax and cleaved caspase-3 were determined by Western blot. RESULTS: Treatment with resveratrol at 2.5, 5, 10, 20, 40 and 80 mg/L for 24 h significantly reduced the viability of SKOV3 cells. The observation by confocal microscopy showed that the nucleus of SKOV3 cells was markedly condensed and heavily stained with the increase in the concentration of resveratrol. Compared with blank control group, the red fluorescence intensity of ROS in different concentrations of resveratrol groups was significantly reduced. The results of Western blot showed that the protein levels of Sirt3, SOD2, Bax and cleaved caspase-3 in resveratrol groups were significantly higher than those in control group, while the protein expression of Bcl-2 was significantly lower than that in control group (P <0.05). CONCLUSION: Resveratrol induces apoptosis of SKOV3 cells by regulating Sirt3-SOD2-ROS pathway. 相似文献

AIM: To investigate the effects of ubiquitin-specific peptidase 9, X-linked (USP9X) down-regulation on apoptosis and invasion ability in gastric carcinoma cells, and to explore its possible molecular mechanisms. METHODS: USP9X small interfering RNA (siRNA) and control siRNA were used to be transfected into gastric carcinoma AGS cells. The cells were divided into 3 groups, including untreated AGS group, control siRNA group and USP9X siRNA group. The expression of USP9X at mRNA and protein levels in the AGS cells with different treatments was determined by real-time PCR and Western blot. The cell viability was analyzed by CCK-8 assay. Flow cytometry and Boyden chamber were employed to examine the apoptosis and invasion ability of the AGS cells. RESULTS: USP9X siRNA significantly down-regulated the expression of USP9X at mRNA and protein levels in the AGS cells. Down-regulation of USP9X markedly induced apoptosis and reduced invasion ability of the gastric carcinoma AGS cells. Notably, down-regulation of USP9X significantly reduced the protein expression of Mcl-1 and MMP-2, but markedly increased the protein level of Bax. CONCLUSION: USP9X may be a key regulator for apoptosis and invasion in gastric carcinoma. 相似文献

AIM:To investigate the effect of erianin on the viability and apoptosis of human lung cancer A549 cells and its possible mechanism. METHODS:A549 cells and BEAS-2B cells were cultured in vitro and treated with erianin at 10, 20, 40, 80 and 160 nmol/L for 48 h. The cell viability was measured by CCK-8 assay. The apoptosis and reactive oxygen species (ROS) were analyzed by flow cytometry. The activity of superoxide dismutase (SOD) was detected by WST-8 method, and the content of malondialdehyde (MDA) was detected by barbituric acid method. The protein levels of nuclear factor E2-related factor 2 (Nrf2), NAD(P)H:quinone oxidoreductase-1 (NQO1), heme oxygenase-1 (HO-1), p38 MAPK, p-p38 MAPK, caspase-3 and cleaved caspase-3 were determined by Western blot.RESULTS:Erianin remarkably reduced the viability of A549 cells in a dose-dependent manner (P<0.05) with IC50 at 52.64 nmol/L. Erianin also induced apoptosis (P<0.05), increased ROS level and MDA content (P<0.05), diminished SOD activity (P<0.05), and down-regulated the protein levels of Nrf2, NQO1 and HO-1 (P<0.05), in a dose-dependent manner. Meanwhile, erianin up-regulated the levels of p-p38 MAPK and cleaved caspase-3 (P<0.05), and these effects were inhibited by N-acetyl-L-cysteine and SB203580 (P<0.05).CONCLUSION:Erianin may induce apoptosis of human lung cancer A549 cells most likely via inhibiting SOD activity and down-regulating the protein levels of Nrf2, NQO1 and HO-1, thus resulting in an increase in ROS and activation of p38 MAPK. 相似文献
