为了解中国江苏省及周边地区猪场断奶仔猪多系统衰竭综合征(PMWS)相关病原的流行情况,本研究采用PCR方法,对2014年1月至2015年5月采自江苏、安徽及浙江等地猪场的125份健康猪样品和261份发病猪样品分别进行猪圆环病毒2型(PCV2)、猪繁殖与呼吸综合征病毒(PRRSV)、猪细小病毒(PPV)、输血性传播病毒(TTV)和类猪圆环病毒P1的检测。结果显示,所有样品PCV2、PRRSV、PPV、TTV1、TTV2和P1的阳性率分别为39.38%、21.76%、3.11%、15.80%、16.32%和10.10%,其中混合感染主要存在于PMWS的发病猪群,以PCV2与TTV2 (15.32%)和PCV2与PRRSV (11.87%)的混合感染为主。结果表明,江苏省及周边地区猪场普遍存在PMWS相关病原的混合感染现象,加大了PMWS相关病原的防控难度。  相似文献

A comparative serologic and virologic study was performed in pigs from 5 herds with postweaning multisystemic wasting syndrome (PMWS) and 2 herds without PMWS in Quebec. In each herd, 60 blood samples were collected at 4-wk intervals from pigs from 3 to 23 wk of age. The serum was evaluated for the presence of antibodies to porcine circovirus type 2 (PCV2) and porcine reproductive and respiratory syndrome virus (PRRSV), as well as for the presence of nucleic acid of PCV2, PRRSV, and porcine parvovirus (PPV), by means of the polymerase chain reaction (PCR). Serologic profiles for PCV2 were very similar in 6 of the 7 herds, including the 2 without PMWS, and were characterized by a gradual decrease in antibody titres from 3 until 11 wk of age, followed by seroconversion at 15 wk, and high PCV2 antibody titres thereafter in all pigs. Only starting at 11 to 15 wk of age could PCV2 viremia be detected, except in 1 herd, in which clinical signs were observed at 6 to 7 wk of age. A PCV2 viremia could be detected within the same pigs for a minimum of 8 wk, and the virus could still be detected in 41% of the serum samples obtained at 23 wk of age. The antibody level did not appear to influence the occurrence of disease, since titres were similar in pigs in the herds with or without PMWS. Infection with PRRSV, as demonstrated by PCR and seroconversion, preceded that of PCV2 by at least 1 mo in both types of herd. Both PRRSV and PCV2 were detected in some pigs in 5 of the 7 herds, including 1 herd without PMWS. Porcine parvovirus could be detected in serum by PCR in 2 herds with PMWS after the onset of clinical signs and also in 1 herd without PMWS. Genomic analysis of PCV2 strains identified in the herds without PMWS indicated complete or very high homology (99.4% to 100%) with the PCV2 strains identified in 4 herds with PMWS. In our field study, the triggering of PMWS in the herds could not be linked to coinfection with either PRRSV or PPV or to the use of a specific immunostimulant, such as vaccines, or to particular genomic differences between the PCV2 strains identified.  相似文献

随着规模化养猪业的发展,传染性疾病越来越多,混合感染或多重感染十分普遍,给疾病的诊断和预防带来很大困难。本文将我们实验室2010年分别采自福建、广西、河南、上海、内蒙、浙江、江苏和山西等地发病猪场的185份病料,通过RT-PCR和PCR方法对其进行猪繁殖与呼吸综合征病毒(Porcine reproductive and respiratory syndrome virus,PRRSV)、猪繁殖障碍的猪瘟病毒(Classical swine fever virus,CSFV)、猪圆环病毒2(Porcine circovirus type 2,PCV2)、猪细小病毒(Porcine parvovirus,PPV)、猪伪狂犬病毒(Pseudorabies virus,PRV)、细环病毒2(Torque teno virus 2,TTV2)等病毒检测。结果表明在所检的病料中PRRSV、PCV2和TTV2的阳性率比较高,分别为49.2%、62.2%和95.1%。有些地方TTV2的阳性率高达100%;同时,还存在很普遍的PRRSV与PCV2、PRRSV与TTV2、PCV2与TTV2等混合感染,混合感染率分别为27.6%、45.4%、58.4%;以及PRRSV、PCV2与TTV2的三重感染率为24.9%。同时对部分PRRSV阳性病料的PRRSV GP5和nsp2基因分别进行测序,分析测序结果表明病料中的PRRSV的GP5和nsp2序列与PRRSV HuN4株的相应序列的亲缘关系分别在98.2%和96.2%以上,且在nsp2区域都存在30个不连续氨基酸的缺失,说明现在临床中所流行的PRRSV可能仍是与高致病性PRRSV基因型相似的病毒。通过本文可以及时了解当前养猪场的病毒性疾病的流行情况,为猪场病毒性疾病的防控提供依据。  相似文献

为了解我国猪群中猪输血传播病毒(TTV)与猪圆环病毒(PCV)混合感染情况,本研究采用PCR方法对采自14个省280份病料样品进行检测,结果显示,TTV1和TTV2阳性检出率分别为51.8%和28.2%,TTV1与TTV2混合感染率为18.2%;PCV1和PCV2阳性率分别为41.1%和37.5%,PCV1与PCV2混合感染率为19.6%;PCV2阳性样品中TTV1和TTV2混合感染率达75.0%。此外,对2株TTV1基因组进行测序,与GenBank中登录的6个TTV1序列比对,相似性结果为67.3%~95.1%;对4株TTV2基因组测序,与GenBank登录的4条TTV2序列比对,相似性为84.7%~90.4%;将TTV1与TTV2进行序列比对,相似性仅为44.0%。TTV1和TTV2基因高变区分别位于520 nt~2 594 nt和720 nt~2 170 nt;TTV1基因保守区位于1 nt~520 nt和2 595 nt~2 800 nt;而TTV2基因保守区位于1 nt~719 nt和2 171 nt~2 800 nt。以上结果表明,我国猪群中存在TTV与PCV2的混合感染,并且两种病毒混合感染引起...  相似文献

根据GenBank_h发表的猪圆环病毒2型(Porcinecircovirustype2,PCV2)基因组序列和TTV(Torquetenovirus)1、2型的UTR序列设计合成引物,建立了分别用于检测PCV2和TTV1、TTV2的PCR及巢式PCR方法。应用建立的PCR方法对送检的广东、福建和江西等7个省份258份血液和组织样品进行了PCV2、TTV1和TTV2的检测,确定猪群中PCV2与TTV1和/或TTV2混合感染情况。结果表明,94份样品表现为PCV2和TTV1的混合感染,占样品总数的36.4%;193份样品表现为PCV2和TTV2的混合感染,占74.8%;另外,还有一些样品为三重感染,占34.5%。由此可以看出,猪群中PCV2和/或TTV1和/或TTV2的混合感染很普遍。  相似文献

应用PCR技术,结合,临床观察、病理解剖对福建省不同地区断乳多系统衰竭综合征(PMws)发病猪群采集的病料进行多重病原检测与分离。结果表明50份病料中猪圆环病毒2型(PCV2)、猪繁殖与呼吸综合征病毒(PRRSV)、猪瘟病毒(CSFV)、猪伪狂犬病病毒(PRV)阳性率分别为40%、26%、4%、0,不同地区各病原检出率差别不大。病毒性混合感染以PCV2和PRRSV为主,感染率为22%;PCV2与CFSV混合感染率为4%;PRRSV与CFSV混合感染率为4%;PCV2、CFSV、PRRSV三种病原混合感染率为4%。研究结果表明福建大部分地区猪群PMWS的发生是PCV2、PRRSV、CSFV等多种病原混合感染的结果,其主要病原为PCV2,该病的发生没有明显地域性。  相似文献

Currently virus surveillance in swine herds is constrained by the cost-effectiveness and efficiency of sampling methods. The objective of this study was to assess the value of using oral fluids collected by barn personnel as a method of surveillance based on PCR testing. Approximately 12,150 pigs in 10 wean-to-finish barns on 10 farms were monitored for the presence of porcine circovirus type 2 (PCV2), porcine reproductive and respiratory syndrome virus (PRRSV), influenza A virus (IAV), and Torque teno virus genogroups 1 (TTV1) and 2 (TTV2) by sampling oral fluid specimens. Oral fluid samples were collected from 6 pens at each site starting at the time of pig placement (~3 weeks of age) and continuing thereafter at 2-week intervals for a period of 18 weeks. Data were analyzed both on a pen basis and barn basis. Overall, 508 (85%) samples were positive for PCV2, 73 (12%) for PRRSV, 46 (8%) for IAV, 483 (81%) for TTV2, and 155 (26%) for TTV1 during the study period. The estimated arithmetic means of the quantitative PCR-positive oral fluids for PCV2, PRRSV, and IAV were 1×10(4.62), 1×10(4.97), and 1×10(5.49)per ml. With a single exception, all barns were positive for PCV2 and TTV2 at every sampling point in the study. Virus detection varied among barns, particularly for IAV and PRRSV. The pen level, cumulative distribution of agent combinations between all 10 barns were statistically different. The most commonly observed patterns were PCV2+TTV2 (239 pen samples, 40%), PCV2+TTV1+TTV2 (88 pen samples, 15%), and PCV2 alone (66 pen samples, 11%). This "proof-of-concept" project showed that a variety of viruses could be detected either intermittently or continuously in pig populations and demonstrated that barn herd virus status is highly variable, even among barns in the same production system. Oral fluid sampling is a promising approach for increasing the efficiency and cost effectiveness of virus surveillance in swine herds.  相似文献

This work aimed to detect and study natural co-infection of Circoviridae torque teno virus (TTV) and porcine circovirus 2 (PCV2) in the swine reproductive apparatus. Semen and organs from 17 boars were tested by nested and real-time PCR. PCV2 was amplified from semen (47%), lymph nodes (84.6%) and testicles (35.3%). TTV2 was amplified from 16/17 testis and 13/13 lymph nodes. TTV1 DNA was detected in fewer testicle samples (2/17), which were also TTV2 positive. Analyzed ovaries, follicular fluid and uteri of 83 culled sows showed TTV2, TTV1 and PCV2 from 49.3%, 30.1% and 6.0% of the sows, respectively. Sperm analysis indicated insignificant differences between PCV2 and TTVs positive and negative boars. The most frequent pathologic lesion in sows was endometritis (28.9%), but this was unassociated with PCV2 or TTVs detection. These findings question the importance of PCV2 and TTV2 natural co-infection in the pathology of porcine reproductive failures.  相似文献

为了解细环病毒(Torque teno virus,TTV)在广西猪群中的感染情况,本研究运用Nest-PCR方法,对2009—2011年采自广西140个规模猪场的156份血液、流产胎儿及肺脏、脾脏、肾脏、肝脏、淋巴结等组织样品进行检测,并对阳性样品的非编码区(UTR)进行克隆测序及遗传进化分析;同时对部分样品进行猪圆环病毒2型(PCV2)、猪繁殖与呼吸综合征病毒(PRRSV)、典型猪瘟病毒(CSFV)、伪狂犬病病毒(PRV)检测及细菌的分离鉴定。结果发现,广西猪群中TTV总感染率达到93.6%,TTV2的感染率(76.9%)不仅明显高于TTV1(16.7%),且毒株间遗传变异较大。TTV多与PCV2和PRRSV混合感染,且以与PRRSV混合感染率更高(64.29%)。猪群中存在2重、3重,甚至4重TTV与其他病毒的混合感染。临床上TTV与细菌的混合感染(或细菌继发感染)以链球菌和副猪嗜血杆菌多见。本研究证实在广西猪群中存在TTV感染,且存在普遍的TTV与PRRSV、PCV2和CSFV混合感染。  相似文献

广西规模猪场猪呼吸道疾病综合征的病原学调查   总被引:1,自引:0,他引:1  
为了解猪呼吸道疾病综合征(PRDC)在广西规模场猪群中的感染状况,采用PCR和RT-PCR方法,对2007~2009年期间采自广西13个市65个规模猪场的281份疑似PRDC感染组织样品进行了12种病原体检测。结果显示:73.85%(48/65)的猪场和71.87%(202/281)的组织样品为PRDC感染,其中,被猪繁殖与呼吸综合征病毒(PRRSV)、猪圆环病毒2型(PCV2)、猪瘟病毒(CSFV)、猪链球菌(SS)、猪伪狂犬病毒(PRV)、败血性波氏杆菌(Bb)、猪附红细胞体(E-su is)、猪流感病毒(SIV)、产毒素多杀性巴氏杆菌(T+PM)、副猪嗜血杆菌(HP)、猪细小病毒(PPV)和猪传染性胸膜肺炎放线杆菌(APP)感染(包括混合感染)的阳性样品分别占49.11%(138/281)、34.52%(97/281)、9.60%(27/281)、8.19%(23/281)、6.41%(18/281)、6.41%(18/281)、6.05%(17/281)、6.14%(15/281)、4.63%(13/281)、3.91%(11/281)、0.36%(1/281)和0.00%(0/281);单纯感染占28.11%(79/281);混和感染占43.77%(123/281)。二重、三重和四重混合感染分别占28.11%(79/281)、12.46%(35/281)和3.20%(9/281)。其中,以PRRSV+PCV2(+其他)混合感染形式较多见,占所有混合感染样品的52.03%(64/123)。细菌性病原体感染样品占20.28%(57/281),其中19.22%(54/281)为混合感染。可见:广西地区规模猪场普遍存在PRDC感染,其中PRRSV和PCV2是引起PRDC的主要病原;感染类型复杂多样,以PRRSV+PCV2(+其他)混合感染最为常见。  相似文献

根据GenBank登录的猪伪狂犬病毒(PRV)、猪圆环病毒2型(PCV2)和猪繁殖与呼吸综合征病毒(PRRSV)的参考基因序列,设计3对引物分别用于扩增PCV2的ORF2基因、PRV的gE基因、PRRSV的N基因的目的片段,通过优化反应中各个影响因素,建立了PRV、PCV2、PRRSV的多重PCR(mPCR)检测方法。敏感性和特异性的结果表明,该方法对这3种病毒的最低核酸检出量分别为32.5(PRV)、25.2(PCV2)、35.9pg(PRRSV)。该方法对猪流感病毒(SIV)、猪圆环病毒1型(PCV1)、大肠杆菌、猪瘟病毒(CSFV)、猪流行性腹泻病毒(TGE)等病毒的检测结果均为阴性。200份临床样品的多重PCR结果表明,PCV2感染率为80%(160/200),PRV感染率为21%(42/200),PRRSV的感染率为78%(156/200)。200份临床样品主要为PCV2和PRRSV混合感染,阳性率达56.0%(112/200)。该方法的建立对这3种病毒病的早期快速检测和指导临床实践具有十分重要的意义。  相似文献

Fourteen diseased pigs from four farms in which there had been an outbreak of salmonellosis were investigated. Granulomatous inflammation with depletion of lymphocytes was observed in the swollen lymph nodes in these pigs. Antigens to porcine circovirus type 2 (PCV2) were immunolabeled in the lesions along with detection of viral DNA as PCV2 by polymerase chain reaction (PCR). In addition, antigens to porcine reproductive respiratory syndrome virus (PRRSV) were immunodetected in the lungs and Salmonella Choleraesuis was isolated from the affected pigs. The nine salmonellosis affected pigs, five (55.6%) with salmonellosis and PMWS concurrently infected with PRRSV were much higher than those infected with salmonellosis and PMWS (22.2%) or with salmonellosis and PPPRV (22.2%).  相似文献

为了解2018年广西猪群重要疫病流行情况,试验采集广西各地的病死猪组织样品及病猪腹泻拭子,应用多重实时荧光定量RT-PCR检测猪瘟病毒(CSFV)、猪繁殖与呼吸综合征病毒(PRRSV),应用多重实时荧光定量PCR检测猪伪狂犬病病毒(PRV)、猪圆环病毒1型(PCV1)、猪圆环病毒2型(PCV2)及猪圆环病毒3型(PCV3),应用多重RT-PCR检测猪流行性腹泻病毒(PEDV)、猪德尔塔冠状病毒(PDCoV)、猪传染性胃肠炎病毒(TGEV)和猪轮状病毒(PRoV)。结果显示,所检测的694份组织样品中,CSFV、PRRSV、HP-PRRSV、PRV、PCV1、PCV2、PCV3的阳性率分别为11.10%、18.88%、7.20%、5.19%、2.45%、67.00%和5.76%;2种病原混合感染率为41.21%,3种病原混合感染率为4.32%,其中PRRSV和PCV2混合感染率最高。所检测的792份肠内容物及拭子腹泻样品中,PEDV、PDCoV、TGEV、PRoV的阳性率分别为9.72%、5.81%、1.77%和6.31%;2种病原混合感染率为5.30%,其中PEDV和PRoV混合感染率最高。结果表明,当前多种重要病毒性疫病仍在广西猪群发生和流行,并且多重感染普遍存在,应进一步加强监测和防控。  相似文献

The objectives of this study were to determine if coinfection of segregated early weaned (SEW) pigs with porcine circovirus type 2 (PCV2) and porcine parvovirus (PPV) induces an increase in the incidence of post-weaning multisystemic wasting syndrome (PMWS) compared to singular PCV2 infection, and to determine if vaccination against PPV protects pigs against PMWS associated with PCV2/PPV coinfection in SEW pigs. Seventy, 3-week-old, SEW pigs were randomly assigned to one of the five groups. Pigs in group 1 (n = 14) served as the negative controls, group 2 pigs (n = 14) were inoculated with PCV2, group 3 pigs (n = 12) were inoculated with PPV, groups 4 (n = 16) and 5 (n = 14) pigs were inoculated with both PCV2 and PPV. Pigs in groups 1-3 and 5 were vaccinated with two doses of a killed parvovirus-leptospira-erysipelothrix (PLE) vaccine prior to inoculation. The PCV2/PPV-coinfected pigs (groups 4 and 5) had significantly (P < 0.05) higher and more persistent fevers than the singular PCV2-infected pigs. One pig in each of the coinfected groups developed clinical disease (fever, respiratory disease, jaundice, weight loss) consistent with PMWS. Lymphoid depletion was significantly (P < 0.05) more severe in the dually-infected pigs at 42 days post-inoculation (DPI). Vaccinated, coinfected pigs (group 5) remained viremic significantly (P < 0.05) longer and had higher copy numbers of genomic PCV2 DNA in sera at 28, 35, and 42 DPI compared to the unvaccinated coinfected pigs (group 4). PPV-viremia was detected only in the unvaccinated group 4 pigs. PLE-vaccination prevented PPV-viremia but did not prevent clinical PMWS or reduce the severity of lymphoid depletion in PCV2/PPV-coinfected pigs. Evidence of increased incidence of clinical PMWS due to vaccination was not observed in this model.  相似文献

A field-based case-control study was conducted to assess the strength of association of porcine circovirus type 2 (PCV2) and some major swine viruses with postweaning multisystemic wasting syndrome (PMWS). Cases were defined as individual pigs with a clinical history of progressive weight loss and histopathological lesions characteristic of PMWS. Controls were pigs without clinical signs and histopathological lesions typical of PMWS. A total of 31 cases and 56 controls was identified from diagnostic submissions. Serum and various tissues were collected from all animals and assayed for PCV, porcine reproductive and respiratory syndrome virus (PRRSV), porcine parvovirus, porcine enterovirus types 1-3, swine influenza virus, porcine respiratory coronavirus, transmissible gastroenteritis virus, porcine endogenous retrovirus, porcine lymphotropic herpesvirus type 1, and bovine viral diarrhea virus. The proportion of case and control pigs positive for each virus was determined and statistically compared for determining the strength of the association that each virus had with PMWS individually or in combinations. Porcine circovirus type 2 had the strongest association (OR = 9.3, P = 0.006) with PMWS among the viruses tested for. Risk for PWMS was much higher (OR = 31.2, P = 0.0009) if the animal was concurrently infected with PCV2 and PRRSV, suggesting that development of PMWS may be enhanced by cofactor(s). Because PCV2 was also found in 62.5% of the controls, PCV2 from 5 cases and 4 controls were selected and genetically compared. No significant genetic difference was observed between PCV2 from PMWS and control pigs.  相似文献

In this study, 60 pigs with clinical signs of post-weaning multisystemic wasting syndrome (PMWS) from 20 different pig herds and 180 control pigs (without clinical signs of PMWS) were examined to get more insights into the frequencies of porcine circovirus 2 infections and the presence of co-infections in pigs with and without clinical signs of PMWS in the Netherlands. Porcine circovirus type 2 was detected in 100% of the pigs with clinical signs of PMWS by virus isolation and/or PCR and in 50% of the pigs from PMWS-free herds. There was an association between the levels of infectious PCV2 and/or PCV2 DNA load and the severity of clinical signs as described for PMWS. A high variation in PCV2 antibody titres was found in the clinically affected pigs, and 27% of these pigs did not mount PCV2 antibody titres higher than 1:200. A concurrent infection of PCV2 and porcine reproductive and respiratory syndrome virus (PRRSV) was found in at least 83% of the pigs with clinical signs of PMWS and in 35% of the pigs from PMWS-free herds. Co-infections of European- and American-type PRRSV were detected only in PMWS herds and in one control herd with a history of PMWS clinical signs.  相似文献

Porcine reproductive and respiratory syndrome virus (PRRSV) and porcine circovirus type 2 (PCV2) are pathogens, which can significantly affect the swine industry worldwide. Field surveys suggest that simultaneous PRRSV and PCV2 infection is common in pigs. The objective of this study was to measure the changes in peripheral blood leukocyte subpopulations in piglets co-infected experimentally with PRRSV and PCV2, in order to analyze the synergistic influence of co-infection on the immune system. Changes in peripheral blood leukocyte subpopulations were systematically measured by flow cytometry (FCM). The levels of antibodies to PRRSV and PCV2 were detected by indirect Enzyme-Linked ImmunoSorbent Assay (ELISA) and the indirect fluorescent antibody test (IFA), respectively. Serum viral loads were measured using real-time PCR. The results showed that piglets co-infected with PRRSV and PCV2 exhibited slower generation and lower levels of antibodies to PRRSV and PCV2, and increased amounts and a prolonged presence of both PRRSV and PCV2 in serum, in comparison to the piglets infected with either virus alone. The major finding in our study was that the total and differential leukocyte counts, including white blood cells (WBCs), monocytes, granulocytes and lymphocytes (T, B and NK cells, as well as T-cell subpopulations), dramatically decreased early during co-infection with PRRSV and PCV2 for about two weeks, in contrast with animals singly infected with either PRRSV or PCV2. These results suggest that PRRSV and PCV2 co-infection results in a synergistic decrease in immune cells in the peripheral blood of piglets. These data contribute to the understanding of the immunosuppressive effects resulting from PRRSV and PCV2 co-infection in pigs.  相似文献
