To understand nutrient cycling in soils, soil processes and microorganisms need be better characterized. To determine whether specific trophic groups of fungi are associated with soil enzyme activity, we used soil imprinting to guide mm-scale sampling from microsites with high and low phosphatase activities in birch/Douglas-fir stands. Study 1 involved sampling one root window per site at 12 sites of different ages (stands); study 2 was conducted at one of the stem-exclusion stands, at which 5 root windows had been installed. Total fungal and ectomycorrhizal (EM) fungal terminal-restriction fragment length polymorphism (TRFLP) fingerprints differed between high-and low-phosphatase activity microsites at 8 of 12 root windows across 12 sites. Where differences were detected, fewer EM fungi were detected in high-than low-phosphatase activity microsites. Using 5 root windows at one site, next-generation sequencing detected similar fungal communities across microsites, but the ratio of saprotrophic to EM fungal reads was higher in high-phosphatase activity microsites in the two windows that had low EM fungal richness. In windows with differences in fungal communities, both studies indicated that EM fungi were less successful than saprotrophic fungi in colonizing fine-scale, organic matter-rich microsites. Fine-scale sampling linked with in situ detection of enzyme activity revealed relationships between soil fungal communities and phosphatase activity that could not be observed at the scales employed by conventional approaches, thereby contributing to the understanding of fine-scale phosphorus cycling in forest soils.  相似文献

Low supply of the nutrients nitrogen (N) and phosphorus (P) limit plant growth and spreading, and increase the plant-microbial nutrient competition in subarctic and arctic regions. We investigated the mycorrhizal community structure of a polar shrub willow (Salix polaris) and the microbial turnover in its rhizosphere to explore the adaptation of a mycorrhizal plant in the subarctic tundra. The ectomycorrhizal colonisation ranged from 35 to 64% of the fine root tips and decreased with an increasing soil C/N ratio. In total, 16 ectomycorrhizal morphotypes were found under S. polaris (eight to 13 morphotypes per site, five morphotypes at all four sites). Cenococcum sp. was the most common EM fungus (32% of the ectomycorrhizal fine roots). The abundance of Cenococcum sp. increased with an increasing organic matter content and N/P ratio in the soil. Arbuscular mycorrhizal colonisation of S. polaris was absent or less than 1% of the fine root length. Microbial biomass P accounted for 21–75% of the organic soil P and 6–49% of the total soil P. Microbial biomass P, alkaline and acid phosphatase activities in the rhizosphere increased with increasing soil N concentration. We conclude that a higher N supply decreases the diversity in the mycorrhizal community on polar willows and increases the role of P turnover from the soil microbial biomass for the nutrient supply.  相似文献

The succession of responsible microbiota during the composting process of rice straw (RS) was studied for 145 d in relation to the changes in the organic constituents of RS. During the composting process, the C / N ratio of RS decreased from 56 to 22. On a C basis, the relative contents of lipid, water-soluble organic matter (WSOM), hemicellulose, cellulose, and lignin fractions in RS changed from 5.6, 8.9, 32.9, 17.9, and 34.0%, to 7.3, 5.8, 30.7, 3.8, and 51.1%, respectively, indicating that the cellulose fraction was mainly decomposed in the composting process. Biomass C accounted for 18.3% (on day 75 when the total amount of phospholipid fatty acids (PLFAs) reached a peak) and 11.5% (at the end of composting) of the total C of RS under the composting process. As for PLFAs, the biomarkers of fungi and Gram-negative bacteria predominated in the RS material used. At the thermophilic stage (the first 2 weeks), biomarkers of Gram-positive bacteria and actinomycetes predominated. After the thermophilic stage, biomarkers of other Gram-positive bacteria became dominant. Finally, at the curing stage, the proportion of the biomarkers of Gram-negative bacteria and eukaryotes increased, indicating the co-contribution of Gram-positive and Gram-negative bacteria and fungi in the decomposition process at this stage. The trans / cis ratio of 16 : 1ω7 PLFA of RS under the composting process ranged from 0.18 to 0.30, indicating that the composting process of RS prepared a significantly lower environmental stress (p < 0.01) compared to the decomposition of RS in a submerged paddy soil.  相似文献

The species of an ectomycorrhizal (ECM) community were investigated in a temperate oak forest by morphotyping and ITS rDNA sequencing. Thirty-six ECM morphotypes were found at the site. The niche effect (as organic soil, mineral soil or dead woody debris artificially introduced in the soil) on the ECM community structure and on the potential catabolic activities of the most abundant morphotypes was studied. The morphotypes in each niche were subjected to enzymatic tests developed for hydrolytic and oxidative enzymes involved in the decomposition of organic compounds. The ECM community structure varied widely depending on the soil horizon or habitat patch. The species richness was higher in the A1 horizon than in the other niches. Different ECM species had different activity patterns for the eight enzymatic tests while co-occurring in the same niche. Catabolic activities also changed within species between niches. Dead woody debris were extensively colonized by two saprotrophic fungi (Megacollybia platyphylla and Armillaria sp.) and, in this particular niche, ECM morphotypes predominantly belonged to the genera Lactarius and Tomentella. These morphotypes showed high chitinase activities. This study suggested also that some ECM fungi could obtain nutrients via the chitin degradation of dead or live saprobes.  相似文献

In this study we found that Penicillium spp. exhibiting P-solubilizing activity are common both on and in the roots of wheat plants grown in southern Australian agricultural soils. From 2,500 segments of washed and surface-disinfested root pieces, 608 and 223 fungi were isolated on a selective medium, respectively. All isolates were screened for P solubilization on solid medium containing hydroxyapatite (HA); 47 isolates (5.7%) solubilized HA and were identified as isolates of Penicillium or its teleomorphs. These isolates were evaluated for solubilization of Idaho rock phosphate (RP) in liquid culture. Penicillium bilaiae strain RS7B-SD1 was the most effective, mobilizing 101.7 mg P l–1 after 7 days. Other effective isolates included Penicillium simplicissimum (58.8 mg P l–1), five strains of Penicillium griseofulvum (56.1–47.6 mg P l–1), Talaromyces flavus (48.6 mg P l–1) and two unidentified Penicillium spp. (50.7 and 50 mg P l–1). A newly isolated strain of Penicillium radicum (KC1-SD1) mobilized 43.3 mg P l–1. RP solubilization, biomass production and solution pH for P. bilaiae RS7B-SD1, P. radicum FRR4718 or Penicillium sp. 1 KC6-W2 was determined over time. P. bilaiae RS7B-SD1 solubilized the greatest amount of RP (112.7 mg P l–1) and had the highest RP-solubilizing activity per unit of biomass produced (up to 603.2 g P l–1 mg biomass–1 at 7 days growth). This study has identified new isolates of Penicillium fungi with high mineral phosphate solubilizing activity. These fungi are being investigated for the ability to increase crop production on strong P-retaining soils in Australia.  相似文献

对西北地区5个酿酒葡萄赤霞珠(Vitis vinifera L. cv. Cabernet Sauvignon)葡萄园根际土壤060 cm土层的AM真菌空间分布进行了研究。结果表明,葡萄根系可形成丛枝菌根,且侵染率较高,最高达79%; 在西北地区的5个样地中共分离出AM真菌4属22种,其中球囊霉属(Glomus)15种,无梗囊霉属(Acaulospora)4种,盾巨孢囊霉属(Scutellospora)2种,巨孢囊霉属(Gigaspora)1种。5个样地孢子密度大小顺序为: 陕西泾阳(JY)>山西永济(YJ)>陕西杨凌(YL)>宁夏银川(YC)>甘肃莫高(MG)。各样地葡萄根际土壤中AM真菌种的丰富度不同,陕西泾阳地区最高; 分布于葡萄根际的AM真菌按种类多少排序的属依次是: 球囊霉属无梗囊霉属盾巨孢囊霉属巨孢囊霉属,球囊霉属占据的比例保持着绝对优势; 根内球囊霉、摩西球囊霉、地表球囊霉在不同样地中均为优势菌株,副冠球囊霉,集球囊霉,细凹无梗囊霉是多数样地中的稀有种类。研究表明,葡萄与AM真菌具有良好的共生关系,二者协同进化产生了具有生态环境特异性的菌根真菌多样性; 葡萄根际存在较为丰富的丛枝菌根真菌资源,可供进一步开发利用。  相似文献

Taro (Colocasia esculenta (L.) Schott), cultivated in Vêtuboso, a village of northern Vanuatu, Melanesia, was surveyed to: (1) assess the extent of morphological and molecular variation being maintained by growers at the village level and, (2) compare this diversity with the diversity found in the crops in Vanuatu. Ethnobotanical data were combined with AFLP analysis to elucidate possible sources of variation. Folk assessment of variation is based on: (a) morphological characteristics (11 characters), (b) names and (c) classification according to habitat, uses, origin and agronomic adaptation. This 3-fold approach allowed growers to differentiate 96 morphotypes, all of which are given distinct vernacular names. AFLP fingerprints successfully differentiated all these 96 morphotypes which do not present a significant intra-clonal variation. But genetic results showed no clear groupings according to geographic origin or habitat of morphotypes and stated that the diversity found within the village was comparable with the overall diversity found in Vanuatu. Local nomenclature and stories associated with each cultivar suggested three sources of diversity: introductions (38%), somatic mutations (15%) and sexual recombinations (48%). AFLP results confirm folk beliefs about origin at least for three pairs of mutants. The 11 so-called wild forms analysed by AFLP were suggested to be feral, escapes from domestication. A dynamic in situ conservation strategy (DISC), favouring a broadening of the national genetic base, was discussed for taro.  相似文献

Weedy rice is a complex of Oryza morphotypes widely distributed in commercial rice fields, which interfere with rice cultivation, seed production, industrial processing and commercialization of this crop in several countries. The objective of this study was to characterize the weedy rice complex of Costa Rica by comparing it with the cultivated and wild rice species found in the country. A collection of weedy rice accessions, representative of the morphotypes found in the country, was established and characterized. Their morphometric relationships were established by comparing 27 morphological traits with commercial rice cultivars, landraces and wild Oryza species and by performing a multivariate analysis. Twenty-one weedy rice morphotypes were identified among 735 weedy accessions by using a three-digit code based on seed characters. Three principal components (PCs) explained 66.25% of the variation observed. The first PC accounted for 36.21% of the variation and separated CCDD genome type Oryza latifolia and O. grandiglumis from AA genome species O. sativa, O. glumaepatula, O. rufipogon and O. glaberrima. The second (18.9%) and third (11.14%) PCs separated the weedy morphotype groups from the AA genome species O.sativa, O. glaberrima and O. rufipogon. The weedy morphotypes were scattered between the indica commercial rice varieties, the cluster landraces–glaberrima and O. rufipogon. Additionally, a group of morphotypes showed intermediate characteristics between O. sativa and O. rufipogon, suggesting that hybridization could have taken place in the past between these species. None of the morphotypes collected in Costa Rica clustered with the allotetraploids CCDD species or O. glumaepatula.  相似文献

The goat willow (Salix caprea) belongs to the typical pioneer vegetation at former ore mining sites in Middle Europe as a result of its high metal-tolerance. Ectomycorrhizal (EM) fungi are known to contribute essentially to the tolerance of plants against abiotic stress. We investigated the EM associations of S. caprea at three former silver-mining sites around Freiberg (Saxonia, Germany) after 33 years of revegetation. The EM colonisation ranged from 3 to 36% of the fine root tips. We found 14 different EM types and characterised them morphologically as well as anatomically. The fungal partners were identified using LSU and ITS sequences. They belonged to four families: Thelephoraceae, Cortinariaceae, Tricholomataceae and Tuberaceae. Thelephoraceae, with 53–91%, comprised the largest portion of the total EM colonisation at all test sites and sampling dates. Thelephoraceae produced mainly contact exploration types of ectomycorrhiza. Tricholoma cingulatum was the only EM fungus which was found as ectomycorrhiza and sporocarp. We suppose that the low exploration of the majority of EM associations of S. caprea, in combination with high portions of metal-chelating organic acids containing fungal partners, contributes essentially to the high tolerance of this tree species against high metal concentrations in the soil.  相似文献

本试验研究了化肥减施配施EM微生物菌剂对鲜食玉米产量、品质和土壤肥力的影响。共设置不施加任何肥料和菌剂(CK)、全量化肥(依据当地化肥施用量,CF)、减施40%化肥(RCF)、减施40%化肥和灌施加喷施EM菌剂(RCF+EM)、减施40%化肥和灌施加喷施清水(RCF+W)5个处理。结果表明,RCF+EM处理的玉米产量高于CF处理,但未达到显著水平;玉米可溶性糖、可溶性蛋白和维生素C含量都高于CF处理,其中可溶性蛋白含量达到显著水平,增幅为11.79%;RCF+EM处理下的土壤pH、有机质、速效养分和微生物碳氮均高于CF处理,其中pH、速效钾和微生物氮达到显著水平,增幅依次是20.08%、15.81%、59.96%;RCF+EM处理的土壤真菌数量显著低于CF处理,而细菌、放线菌数量则显著高于CF处理,增幅分别达到153.42%和124.17%。综上,减施化肥配施EM菌剂对使鲜食玉米产量无显著影响,但可显著提高果实品质,改善土壤肥力、提高土壤微生物菌群数量。  相似文献

Summary Vesicular-arbuscular mycorrhizal (VAM) fungi improve plant growth in marginal soils. This study was conducted to determine the effects of three species of VAM fungi on plant nutrition in two cultivars of corn (Zea mays L.) and one of sunflower (Helianthus annus L.). Plants were grown in pot cultures under controlled (greenhouse) conditions in a soil high in K, Mg, and P, and low in Ca and N, and were supplied with amounts of VAM-fungal inocula in which equal numbers of infective propagules had previously been determined. Analysis of variance showed highly significant main effects and interactions due to both factors (plant and fungus) for N, P, Ca, and Mg. For K, only plant effects were significant (P<0.043). The uptake of nutrients was selectively enhanced or inhibited by one or the other VAM fungus relative to non-VAM control plants. In sunflower, N concentration was markedly enhanced (73%) by the mixed inoculum of the three fungi, even though individual effects were not significant. Evaluation of leaf nutrient analyses by the Diagnosis and Recommendation Integrated System (DRIS) revealed the utility of this system to rank nutritional effects by VAM fungi in an order of relative nutrient deficiency. The effect of dual inoculation on three local cultivars (Miss Kelly, Portland Red, Round Red) of red kidney beans (Phaseolus vulgaris, L.) with four strains of Rhizobium leguminosarum bv. phaseoli and three species of vesicular-arbuscular mycorrhizal (VAM) fungi was examined in a clay loam soil. Rhizobial strains B 17 and B 36, each paired with Glomus pallidum or G. aggregatum, were the most effective pairings for cv. Miss Kelly. Inoculation of Miss Kelly with any of these pairings significantly (P=0.05) increased growth, number of nodules, nodule dry weight, mycorrhizal colonization, and shoot N and P content than other pairings. The growth response by cv. Portland Red was significantly improved by pairings of B 36 or B 17 with any of the three VAM fungi. For both cultivars (Miss Kelly and Portland Red), CIAT 652 or T 2 paired with VAM fungi did not give a positive growth response. In contrast, for cv Round Red the T 2 rhizobial strain in combination with any of the three VAM fungi showed a significant (P=0.05) growth improvement in all parameters. Our results suggest that while dual inoculation of VAM fungi and rhizobia significantly improves the growth response by red kidney beans, the best pairings of VAM fungus and rhizobia for each cultivar need to be carefully selected.  相似文献

Summary Applications of elemental sulfur (S°) increase the phytoavailability of P in alkaline high-Ca soils through the production of H2SO4 which is induced by microbial oxidation of S. Concentrated S bands, allowing time release acidity, may aid in maintaining the phytoavailability of both residual and fertilizer P. Greenhouse and field studies were therefore conducted to determine the effectiveness of band-applied S° for increasing the phytoavailability of residual and fertilizer P for spring wheat (Triticum aestivum L.) and corn (Zea mays L.). We also used inoculation of S° with Thiobacillus thioparus to determine whether it is necessary or helpful in alkaline soils to initiate acidification. Treatments were inoculation, S°, S° + inoculation, triple superphosphate, triple superphosphate + S°, triple superphosphate + inoculation, and triple superphosphate + S° + inoculation applied to three alkaline soils: Typic Argiborolls, Borollic Calciorthids, and Ustollic Haplargids. P availability was determined by plant uptake of P, NaHCO3-extractable P, dry-matter yield, grain yield and grain-protein production, and on available-P index (NaHCO3-extractable P, post-harvest, + plant uptake of P). Application of S° with triple superphosphate gave a significantly higher available-P index than triple superphosphate alone on all three soils. Inoculation of S° with T. thioparus increased soil acidity; however, in some cases this treatment eliminated the beneficial effect of S°. Grain yields of wheat increased with S° applications at one of two field sites. These results suggest that applying S° with triple superphosphate may be an effective means of increasing soil P availability and the efficiency of superphosphate fertilizers on alkaline high-Ca soils.  相似文献

Summary Variation of the seed storage protein phaseolin was analysed in a collection of 100 accessions of Phaseolus lunatus L. using one-dimensional SDS/PAGE. Cultivated small-seeded genotypes belonging to Sieva and Potato morphotypes and intermediate Sieva-Potato and Sieva-Big Lima morphotypes showed the M (Mesoamerican) pattern, confirming their origin in the same gene pool. Cultivated Big Lima morphotypes showed the A (Andean) pattern, confirming that they belong to a distinct gene pool.  相似文献

The fungicide Vitavax RS and the seed polymer Extender were sequentially applied to canola seed to determine their effect on the fungal inoculum, Penicillium bilaiae. In the laboratory, the fungicide was applied at the recommended rate of 23.4 ml kg–1, and the polymer was applied by the manufacturer. Significant death of P. bilaiae occurred after 2.5 days on the Vitavax RS-treated seed and after 1.5 days with the seed treated with both Vitavax RS and Extender. In field treatments, the seed was sown at six sites in the fall just prior to freeze-up. Results from the laboratory experiment indicate that there should be no harmful effects of the Vitavax RS and Extender on the P. bilaiae at the time of seeding because the seeds were sown within 2 h of inoculation. The environmental extremes in 2001 and 2002 resulted in large variations in the data. P. bilaiae did not increase yield under these conditions.  相似文献

试验以樱桃番茄为材料,研究减施化肥配施不同微生物菌剂(EM和春语)对樱桃番茄产量、品质和土壤肥力的影响。结果表明:减施化肥配施EM菌剂处理(RCF+EM)的樱桃番茄产量高于全量化肥处理(CF),但差异未达显著水平(P0.05),RCF+EM处理番茄可溶性固形物、可溶性蛋白和可溶性糖含量显著高于CF处理(P0.05),增幅依次为31.05%、27.82%、62.73%;RCF+EM处理的土壤pH、有机质、速效养分和微生物生物量碳氮含量均高于CF处理,其中p H、碱解氮和微生物生物量碳差异达到显著水平(P0.05),增幅依次为13.07%、20.83%、66.10%。与EM菌剂相比,春语微生物菌剂的效果总体较差。因此,减施化肥配施EM菌剂可提高樱桃番茄产量和果实品质,对土壤肥力也有良好的改善作用。  相似文献

Summary Field experiments were carried out to determine the effects of single and mixed inoculations with Rhizobium and vesicular-arbuscular mycorrhiza (VAM) on nodulation, symbiotic N2 fixation and yield of soybeans in six Taiwan subtropical-tropical sites. Inoculation with Rhizobium alone significantly increased nodulation, nodule weight and nitrogenase activity of nodules in three out of six experimental fields, and affected soybean yields in the range –13% to + 134%. Inoculation with VAM fungi alone did not have a significant effect on nodulation and nitrogenase activity. Mycorrhiza inoculation affected soybean yields in the range –13% to + 65%, but only the yield increases at one out of six sites with N application were statistically significant. Mixed inoculation with Rhizobium and mycorrhiza affected yields in the range –8% to + 145% A synergistic effect from mixed inoculation of Rhizobium-mycorrhiza on soybean yields was found in one out of six experimental fields. The yield response to N application (40 kg N ha–1) in these six paddy-field trials was not significant. These results suggest that single or mixed inoculation of rhizobia can greatly assist soybean grain yields and can replace N fertilizers.  相似文献

Effects of soil amendment with crabshell chitin on the growth of white clover (Trifolium repens L.) and perennial ryegrass (Lolium perenne L.), and on populations of soil bacteria, fungi, and plant-parasitic and free-living nematodes were investigated in a pot trial. Five soil samples were collected from Te Puke (Paengaroa Shallow Sand, a Typic Hapludand) and five from Hamilton (Bruntwood silt loam, an Aquic Hapludand), New Zealand. Subsamples of each soil were either amended with chitin or unamended and planted with white clover and ryegrass. The ryegrass shoot weight in amended soil was greater (P<0.01), most probably due to N mineralised from chitin. A significantly lower (P<0.01) root: shoot ratio of ryegrass in the amended soil also suggested improved N availability, and therefore less root mass was needed to support a given shoot mass. A reduction in nodulation was observed in 12-day-old white clover seedlings (P<0.05) and also in 6-week-old seedlings (P<0.01). The shoot weight of white clover was significantly lower (P<0.05) in amended soil, possibly due to phytotoxic effects of chitin. Chitin increased (P<0.01) the populations of bacteria and fungi by 13-fold and 2.5-fold, respectively. The cyst nematode of white clover, Heterodera trifolii, was significantly reduced in chitin-amended soil, possibly due to increased levels of chitinase produced by rhizosphere microorganisms. Two other plant-parasitic nematodes, Pratylenchus spp. and Tylenchus spp., were also reduced in ryegrass roots and in soil as a result of the chitin amendment. However, the total number of free-living nematodes increased 5.4-fold in amended soil.  相似文献

The colonization and diversity of arbuscular mycorrhizal fungi (AMF) associated with the rhizosphere of tea [Camellia sinensis (L.) O. Kuntze] growing under ‘natural’ as well as ‘cultivated’ conditions in the Kumaun region of Uttaranchal Himalaya (India), during the periods of active growth and dormancy were investigated. Root and rhizosphere soil samples, collected from both the ecosites (natural and cultivated), were monitored for root colonization. While the percent root colonization was quite high (77.66 ± 4.40 and 86.40 ± 3.02%, in the natural and cultivated tea, respectively) during the period of active growth in both the ecosites, relatively higher colonization (97.33 ± 0.78 and 98.13 ± 0.80%, in the natural and cultivated tea, respectively) was recorded during the period of dormancy. The rhizosphere of cultivated tea bushes was found to be dominated by Glomus morhpotypes (88.89% of the total isolates) along with three morphotypes of Acaulospora; occurrence of 35 morphotypes belonging to four genera viz. Acaulospora (11.43%), Gigaspora (11.43%), Glomus (68.57%) and Scutellospora (8.57%) was recorded in the rhizosphere of tea plants from the natural ecosite. A total of 51 AMF morphotypes were detected. Shannon–Weaver index of diversity was higher (1.80 ± 0.13 and 2.05 ± 0.10 during periods of active growth and dormancy, respectively) at the species level for the natural ecosite over its counterparts from the cultivated ecosite. Values for the diversity indices of natural and cultivated ecosites did not show much variation in the period of dormancy. These data suggest that collectively, various cultural practices negatively affect AMF diversity at the genus level in tea plantations of the colder regions.  相似文献

This work explores the links between ecogeographical barriers and clines, and the boundaries of infraspecific biological entities. The distribution of the three subspecies and eight morphotypes of the Neanurinae Deutonura deficiens (Collembola) are mapped and commented based on 570 records. The species inhabits a wide range of forest habitats from the north of Portugal to southeastern France. The subspecies and morphotypes constitute homogeneous geographic sets arranged as a mosaic of closely related forms. A few overlapping zones between subspecies or morphotypes have been detected, some of them having the character of hybrid zones; on the whole however, overlap is small or rare and allopatry or parapatry is the general pattern. Current range boundaries of subspecies and morphotypes were found to be usually not determined by major hydrographical and topographical barriers, with the noticeable exception of the Rhône River. At subspecies level, the morphological cline of increasing tuberculization correlated to the bioclimatic gradient from Mediterranean to Atlantic climates, already recorded by Deharveng (1984. Polymorphisme et polytypisme morphologiques chez quelques Neanurinae européens. Rev. Ecol. Biol. Sol. 21, 533–562), is confirmed on the present data set. Conversely, ecological boundaries, known to constrain species distribution in many living groups of the region, do not fit the contact zones between the different morphotypes of the three subspecies of D. deficiens. The eight recognized forms appear to be at different degrees of reproductive isolation, illustrating active speciation process possibly inherited from recent ecogeographical events. A more detailed sampling on contact zones and a robust phylogenetic hypothesis are needed at this stage for progressing in our understanding of this complex pattern.  相似文献
