Mixed-sex groups of three-month-old Finn Dorset and Dorset Horn lambs were given complete diets containing either high or low protein. A trickle infection with Haemonchus contortus of 200 larvae, given three times a week, began one month later for a 17 week period. Erythrokinetic, metabolic, digestibility and balance studies were conducted with the male lambs during weeks 12 to 14 of the infection. Clinical signs of haemonchosis became evident only in the low protein diet group. The infection was characterised by severe macrocytic anaemia, hypoproteinaemia, hypoalbuminaemia, reduced survival, high egg counts and a significantly greater worm burden. In the high protein diet group many lambs developed resistance to further infection in contrast to the low protein group in which no resistance developed. Live-weight gain was not affected by infection in the high protein diet group but decreased in the lambs given the low protein diet compared with uninfected controls. However, infection did not appear adversely to affect the digestive efficiency, nitrogen retention or iron absorption in either dietary group although loss of appetite occurred in the low protein group. Examination of a sample joint from the carcases showed infection was associated with a marked reduction in muscle mass in lambs given the low protein diet relative to the control animals.  相似文献

Infection of Suffolk lambs with Dictyocaulus filaria larvae beginning at two months of age with one larva per day, six days per week, and doubling every four weeks to reach a final infection rate of 64 larvae per day gave rise to a pattern of larval excretion in the faeces which approximated to that seen in naturally infected Suffolk lambs in Midlothian in 1974. Several of the lambs were still infected with adult D filaria and were excreting larvae in their faeces 28 weeks after the infection began. However, when some of the lambs were challenged at this time, those which had received the previous escalating infection were resistant to the challenge infection.  相似文献

The influence of dietary protein supplementation on resistance to haemonchosis was examined in Hampshire down lambs fed either a basal diet or a diet supplemented with soyabean. At seven months of age the lambs were challenged with an initial loading dose of Haemonchus contortus, followed by a trickle infection three times a week. Blood and faecal samples were collected three times a week and bodyweights were recorded weekly. After 10 weeks the lambs were slaughtered and their worm burdens and carcase composition determined. Although their mean worm burdens were similar, the lambs given the basal diet had higher faecal egg counts, lower packed red cell volumes and lower concentrations of total plasma protein and plasma albumin than the lambs given the supplemented diet. The dietary supplementation also improved the carcase composition of the lambs.  相似文献

Serum protein values were determined in 6 lambs given 1 dose of 200 metacercariae of Fasciola gigantica. Over the 22 weeks of the experiment, total serum protein values in these lambs were only moderately increased over those of control lambs (n = 6). The increase was mainly associated with the change in alpha2-globulin and gamma-globulin fractions. The highest value of gamma-globulin (39.03%) of total serum protein was observed at 18 weeks after infection. Mean albumin concentration in the exposed lambs decreased considerably, starting in week 8 and persisting until the end of the experiment. The lowest level (39.28%) was seen at week 16.  相似文献

The whole blood clotting time progressively increased from the second to the eighth week in lambs receiving a primary infection of Dictyocaulus filaria larvae. However, in vaccinated lambs it remained unaffected. Levamisole hydrochloride was ineffective when given 4 days after infection but restored the blood clotting time to near normal soon after treatment when it was given 30 days after infection.Deceased 23 October 1987  相似文献

Six-month-old red Maasai lambs were more resistant than Dorper lambs to repeated infections at one to two week intervals with 1000 Haemonchus contortus infective larvae. Resistance after infection was assessed by means of faecal egg counts, packed cell volumes, eosinophil counts, total serum protein concentrations and mortality rates. The weight gains of the infected animals were only marginally lower than those of their uninfected controls, most probably because of their significantly higher feed consumption, and evidently the infected lambs were not utilising all of the extra feed for growth. This absence of anorexia in spite of the infection was probably due to the palatability of the high protein diet fed to the lambs.  相似文献

This study describes responses to artificial infection with Haemonchus contortus in ewes and lambs of 50% Dorset, 25% Rambouillet, and 25% Finn-sheep ancestry and provides estimates of genetic parameters for measures of parasite resistance. One hundred ninety-eight ewes out of 64 sires, and 386 lambs out of 25 sires were evaluated in autumn and spring of 2 yr. Ewes were dewormed shortly after weaning their lambs and lambs were dewormed at about 120 d of age. One week after deworming, ewes and lambs were dosed with approximately 10,000 infective larvae of H. contortus. After infection, BW, fecal egg counts (FEC), and packed cell volume (PCV) were measured weekly for 7 wk in lambs kept in drylot and fortnightly for 11 wk in ewes on pasture. Summary traits were defined as initial PCV, mean BW across all times, and means for PCV (MPCV) and log-transformed FEC (MLFEC) at wk 3 to 7 after infection for lambs and wk 3 to 11 after infection for ewes. Ewes and lambs did not lose weight overall in any year or season, but there was no consistent effect of year or lambing season on mean LFEC or mean PCV during infection in either ewes or lambs. Yearling ewes were less resistant to infection than older ewes, with lower PCV (P < 0.05) and higher LFEC (P < 0.05). During infection, PCV was positively correlated with BW and negatively correlated with LFEC in both ewes and lambs. In lambs, heritabilities were 0.39 (P < 0.01) for PCV, 0.10 (P < 0.05) for LFEC across all measurement times, and 0.19 (P < 0.01) for three measures of LFEC taken at the peak of infection. Heritability estimates for ewes were 0.15 (P < 0.05) for PCV and 0.31 (P < 0.01) for LFEC. Repeatabilities for LFEC and PCV across measurement times were moderate in ewes and lambs. Correlations between dam and lamb records for MLFEC were generally low, suggesting different mechanisms of resistance in lambs and nonlactating ewes. Ewes with higher genetic merit for growth as lambs were less resistant to infection as adults, but genetic merit for fertility and prolificacy were not related to parasite resistance. Lambs with higher genetic merit for body weight were more resistant to infection. Selection for resistance to H. contortus is therefore possible and should not adversely affect growth of lambs and fertility of ewes in this production environment.  相似文献

The efficacy of leucocyte extract (LE) and sulphadimidine in preventing coccidiosis in naturally infected lambs on pasture was evaluated in 3 separate experiments, whereas the prophylactic effect of levamisole was studied in 1 of the experiments. LE prepared from ewes immune to coccidia (Eimeria spp.) was administered either intravenously or intraperitoneally to young lambs 7, 5, or 2 days before they were turned out on pastures contaminated with coccidia. In all experiments, LE failed to transfer protective immunity to the lambs against the first coccidial infection on pasture. The LE preparations used apparently had an immunosuppressive effect, which resulted in more severe clinical signs of coccidiosis in the recipients. The lambs given LE showed a higher incidence of diarrhoea, a poorer weight gain, a higher mortality, and a higher oocyst output than the untreated control lambs. In lambs treated with sulphadimidine at 200 mg/kg on days 12, 13, and 14 after turnout there was a reduced severity of the coccidial infections in all experiments. The sulphadimidine-treated lambs had better weight gains and passed fewer oocysts than the controls during the third and fourth week after turnout, but some of them developed diarrhoea. Lambs treated with levamisole at 2 mg/kg 2 days before turnout, at turnout, and 2 days after turnout were more severely affected by the first coccidial infection on pasture than the controls.To study the lambs’ immunity against a heavy challenge infection with coccidia as compared with their immunity against the natural reinfection on pasture, some of the lambs from the original groups (untreated, sulphadimidine-treated, LE-treated) were each inoculated with 2 mill. Eimeria spp. oocysts about 6 weeks after turnout. The oocyst counts of the challenged lambs, except the LE-treated lambs, increased to a new peak 19–20 days after challenge. The challenge infection caused a softening of the faeces and a marked depression in weight gain in all challenged groups of lambs, mainly between days 10 and 17 after challenge. The lambs were thus only partially immune to coccidia after the first coccidial infection on pasture. The lambs treated with either LE or sulphadimidine in connection with the first coccidial infection on pasture were not appreciably more susceptible to the challenge infection than the untreated lambs.  相似文献

Groups containing six lambs that previously had been infected five times each week for four, eight or 12 weeks, either with 1000 Trichostrongylus vitrinus third stage larvae (L3) or 1000 T vitrinus L3 and 2500 L3 Teladorsagia (previously Ostertagia) circumcincta were challenged, along with six worm-free control lambs, with 3000 75Se-labelled T vitrinus L3. The lambs were killed 10 days after challenge. Those given a dual infection for four and eight weeks had significantly lower total T vitrinus numbers than monospecifically infected lambs. Significantly fewer radiolabelled larvae were recovered from the dual infected group killed after four weeks exposure to infection than from lambs infected monospecifically. The negative interaction between these two nematode species appears to be physiologically mediated and may be an important factor in accounting for differences in their seasonal prevalence.  相似文献

In the lungworm-endemic areas of Kashmir, 6-10 week old lambs of Karnah and Kashmir Merino breeds were vaccinated with two doses of 50 kR gamma-irradiated larvae of Dictyocaulus filaria, given a month apart. Assessed on the basis of reduced prevalence and significantly lower faecal larval output over an eight-month observation period, vaccinated lambs showed a high degree of resistance to naturally acquired D. filaria infection. The results also show that vaccination against D. filaria provided some degree of protection against infection with other lungworm species.  相似文献

Neonatal and weaner Gulf Coast Native (Native) lambs were studied to determine whether an immunological basis underlies their natural resistance to Haemonchus contortus infection. Neonatal Native lambs (n = 8) and weaner Native lambs (n = 15) were randomly assigned to a treatment or a control group. Lambs in the treatment group received dexamethasone by intramuscular injection three times a week for 10 weeks (neonatal) and 15 weeks (weaners). All lambs were monitored for fecal egg count (FEC), blood packed cell volume (PCV), and white blood cell differential counts on a weekly basis for the duration of the studies. Neonatal lambs were kept on pasture with their dams and weaner lambs were dewormed at weaning and kept in pens where they received trickle infections. Serum antibody titers to H. contortus whole worm antigen (WWA) were determined using ELISA. Lymphocyte proliferation assays on peripheral blood mononuclear cells (PBMC) were done to assess lymphocyte function. All lambs were vaccinated with killed Brucella abortus strain 19 to assess the effect of dexamethasone treatment on antibody response. All lambs were necropsied at the end of each study to recover the contents of the gastrointestinal tract for nematode enumeration and identification. The results showed that mean FEC and mean PCV of the treatment group was significantly higher and lower, respectively, than in the control group in both neonatal and weaner lambs from weeks 6 and 5, respectively. At necropsy, total nematode count was significantly higher in treatment groups than in the control groups. Serum antibody titers to H. contortus WWA were significantly lower in treated groups than in control groups. Treatment groups showed a consistent depression in lymphocyte percentage being significantly lower from week 6 in both neonatal and weaner lambs. No differences were found in the response of PBMC to mitogen stimulation between the groups. Lambs in the control groups showed strong positive brucellosis card tests and the treatment groups did not. Dexamethasone treatment resulted in depression of the immune response and loss of natural resistance of Native lambs to H. contortus infection. The results of these studies suggest that some aspects of the immune response may underlie the natural resistance of Native sheep to H. contortus infection.  相似文献

An epizootic of abortions, weak lambs, stillbirths, and congenital arthrogryposis-hydranencephaly occurred in a sheep flock in West Texas. The outbreak began during the first week of January 1987 and continued through the third week of February 1987. Lambs born after February 1987 were not affected. A high incidence of antibodies to Cache Valley virus (Texas 7856 isolate) was demonstrated in the ewes' serum and in precolostral serum from affected lambs. No virus was isolated from tissues and body fluids of the affected lambs. The clinical, pathological, and immunological features of the epizootic were similar to those reported in Akabane virus infection in sheep. Although serological findings strongly suggest Cache Valley virus as the etiological agent of this outbreak, transmission studies are needed.  相似文献

The performance of growing lambs dosed daily for 12 weeks with 2500 Trichostrongylus vitrinus larvae (I) or given no larvae (control, C) was compared at two levels of phosphorus intake; 2.75 gP/kg dry matter (normal, NP) and 1.88 pgP/kg dry matter (low, LP). Both low phosphorus and T vitrinus infection reduced dry matter intake and growth rate and the combined effect of low phosphorus and infection were additive. Plasma hypophosphataemia (less than 60 mg per litre) occurred in groups CLP, ILP and INP from week 3, the values gradually returning to normal levels in INP lambs by the end of the trial. T vitrinus infection did not depress plasma phosphorus concentration below that seen in the low phosphorus controls. Both low phosphorus intake and T vitrinus infection reduced the density and mineralisation of bone, the most severely affected bones occurring in groups ILP lambs. T vitrinus infection reduced the content and concentration of phosphorus in the rumen and the effect was greater at the lower intake of phosphorus. Infection also reduced the proportion of phosphorus in the rumen liquid phase. The number of T vitrinus recovered from the intestine was higher and villous atrophy more extensive in lambs from group ILP. It is suggested that development of 'resistance' may be delayed on a low phosphorus intake.  相似文献

Fifteen worm-free lambs (two-and-a-half to three months or four to four-and-a-half months old) were infected with 3500 or 4000 Ostertagia ostertagi larvae on five days each week for six weeks, and their performance compared to that of controls. Eleven lambs were killed after eight weeks and four were challenged with O circumcincta to determine whether any cross resistance had developed. A feature of the O ostertagi infection was the considerable variation in response. Overall liveweight gain was lowered by 24 per cent in the two-and-a-half to three-month-old infected lambs, mainly due to reductions of 27 to 40 per cent in four of the seven lambs. There was no consistent effect in the older lambs. The worm populations consisted mainly of early fourth stage larvae and developing worms, but a small percentage reached sexual maturity and these produced a low faecal egg count (1 to 63 eggs per gram). Numerous intraluminal refractive crystals were present in the gut of both adult worms and developing stages, possibly reflecting degenerative changes. Hypertrophy of the abomasal mucosa with patchy loss of differentiation was a feature of the infection, and in four lambs serum pepsinogen concentrations were elevated. Exposure to O ostertagi did confer some protection against challenge with O circumcincta in that worm counts were reduced to about 60 per cent of those in controls, although no increase was observed in the numbers of arrested larvae. The successful passage of O ostertagi through young lambs could be important in mixed or alternate grazing systems by providing a reservoir of infection for the alternate host.  相似文献

Abstract

CASE HISTORY AND CLINICAL FINDINGS: Two White Dorper lambs from the North Island of New Zealand, 2 and 4 weeks of age, were presented with large skin flaps hanging from the flanks, separation of skin from the subcutis over mobile joints, and de-gloving injuries of the limbs and tail. The lambs were subject to euthanasia on humane grounds.

PATHOLOGICAL FINDINGS: Large skin tears with associated haemorrhage, periarticular S/C oedema and generalised skin fragility were observed in both lambs at post-mortem examination. Histology of the affected skin revealed diffuse hyalinisation of dermal collagen compared with control lambs, protein-filled peri-adnexal clefts and areas of deep dermal and S/C granulation tissue consistent with previous separation of skin from the subcutis. Analysis of hair follicles, collected from one of the lambs, using a commercially available genetic test in Australia was consistent with the lamb being homozygous for the mutation responsible for ovine dermatosparaxis.

DIAGNOSIS: Likely dermatosparaxis.

CLINICAL RELEVANCE: These findings strongly suggest that the mutation responsible for dermatosparaxis in White Dorper sheep is present in New Zealand. Dermatosparaxis should be considered when investigating skin fragility in lambs with White Dorper genetics. Confirmation of the disorder is possible through genetic analysis of hair follicles.  相似文献

Groups of seven lambs were infected on five days each week for 11 weeks with either 1000 or 2000 Trichostrongylus vitrinus larvae alone or concurrently with 1500 or 2500 Ostertagia circumcincta larvae and their growth performance compared to that of worm-free controls. The cumulative liveweight gain of the infected groups was significantly lower than that of the controls but the gain of the concurrently infected lambs did not differ significantly from the respective T vitrinus group. There were no significant differences in the numbers of T vitrinus or O circumcincta recovered from the single and concurrently infected groups, although there was a tendency for lower mean T vitrinus burdens at the higher level of ostertagia/trichostrongylus infection. The distribution of T vitrinus along the small intestine was similar in single and concurrently infected lambs. The reason for the lack of a marked additive effect on growth rate with concurrent infection is discussed in relation to changes in the concentration of two plasma constituents and lesions in the gastrointestinal tract.  相似文献

The frequent appearance of patent infestations of Strongyloides papillosus in lambs under one week is observed, and it is suggested that either the short generation interval or pre-natal infection is the cause. Pathogenic effects associated with heavy infestations in young lambs consisted of anorexia, loss of weight, diarrhoea and a moderate anaemia. The lambs were usually very weak, unable to stand and laying prostrate. Death usually followed at this stage and three such lambs examined showed severe enteritis. There was also severe dermatitis noticed around the flanks and abdomen. The epidemiology studies which were based on worm egg counts taken at monthly intervals for a twelve-month period showed high counts in October and November, intermediate counts in May to August and low counts from February to April. The egg counts per g of faeces ranged from 100 to 18,000.  相似文献

AIMS: To determine whether: a) using a controlled-release anthelmintic capsule (CRC) instead of a programme of 5 oral drenches administered at 3-4 week intervals, would delay the development of anti-parasite immunity in lambs; b) the use of ivermectin instead of albendazole, administered either as a CRC or as a programme of 5 oral drenches, would delay the development of anti-parasite immunity in lambs; c) lambs treated with CRCs would have higher liveweight gains than lambs drenched orally 5 times at 3-4 week intervals, and; d) delayed onset of anti-parasite immunity is associated with reduced liveweight gains in the period following anthelmintic treatment. METHODS: Three field trials were conducted, 1 on a research farm and 2 on commercial sheep farms, in which groups of 30 lambs were treated with either a CRC containing albendazole, a CRC containing ivermectin, 5 oral drenches with albendazole, or 5 oral drenches with ivermectin, administered at 3-4 week intervals. Liveweights and faecal nematode egg counts (FECs) were recorded in all trials. Immunoglobulin-G (IgG) antibody levels to Ostertagia circumcincta and Trichostrongylus colubriformis adult and larval antigens were measured in Trials 1 and 3, and fleece weights and resistance of animals to nematode challenge infection were measured in Trial 1. RESULTS: CRC-treated lambs had higher levels of antibodies to O. circumcincta infective-stage larvae (L3) than orally drenched lambs in Trial 3, but no other immunological differences due to mechanism of delivery were detected. Antibody levels were lower in lambs treated with ivermectin than albendazole, as a CRC or oral drench in Trial 1, but this was not associated with any measurable effects on FEC or productivity. No significant differences (p>0.05) were detected between drench types (albendazole vs ivermectin) or delivery mechanisms (CRC vs oral drenching) in any of the production parameters measured, in any of the trials. Albendazole-CRCs failed to control FECs in all 3 trials. CONCLUSIONS: Although some differences between treatments in antibody levels were detected these were not associated with measurable differences in level of parasitism or productivity of lambs. CRC use did not appear to offer substantial gains in productivity over a structured programme of 5 oral drenches administered at 3-4 week intervals.  相似文献

The effect of a bronchodilator (or ciprenaline sulphate) and intravascular oxygen releaser (sodium dihydrogen orthophosphate) on the host in experimental Dictyocaulus filaria infection was studied. Fifteen male lambs of Dorset-Muzaffarnagri breed, aged 4-6 months, were divided into four groups of four (infected bronchodilator), four (infected i.v. O2 releaser), four (infected untreated controls) and three (uninfected controls). The administration of i.v. O2 releaser helped in increasing the length of useful patency, estimated total larval production and survival rate of D. filaria producer lambs. The administration of I.V. O2 releaser and bronchodilator helped in efficiently restoring the altered values of blood pH, haemoglobin, packed cell volume, erythrocyte sedimentation rate and osmotic fragility of erythrocytes to near normal levels. However, the blood clotting time and level of lactate dehydrogenase activity remained altered and followed a course typical of ovine dictyocauliosis.  相似文献

Background

Anaplasma phagocytophilum (formerly Ehrlichia phagocytophila) is an obligate intracellular bacterium causing the disease tick-borne fever (TBF) in domestic ruminants. An effective vaccine against the infection has been demanded for livestock by sheep farmers and veterinary practitioners for years.

Findings

In the present study, we immunized lambs with an inactivated suspension of 1 × 108 killed A. phagocytophilum organisms mixed with adjuvant (Montanide ISA 61VG; Seppic). Twelve 9-months-old lambs of the Norwegian White Sheep breed were used. A full two-dose series of immunization was given subcutaneously to six lambs with a 4 week interval between injections. One month after the last immunization, all lambs were challenged with the homologous viable variant of A. phagocytophilum. After challenge, all lambs showed clinical responses for several days, although the immunized lambs reacted with an anamnestic response, i.e. significant reduction in infection rate and a significantly higher antibody titer.

Conclusion

Immunization with inactivated A. phagocytophilum did not protect lambs TBF.  相似文献
