The apple cultivar Honeycrisp exhibits genetic resistance to apple scab. The characterization of the macroscopic and microscopic responses on leaves infected by the pathogen Venturia inaequalis is described. The macroscopic resistance reactions observed in ‘Honeycrisp’, its parent ‘Keepsake’, and grandparents ‘Frostbite’ and ‘Northern Spy’ ranged from 0 (no reaction) to chlorotic flecking, stellate chlorosis, necrotic flecking, and sporulation. No hypersensitive response was observed. The resistance response occurred as early as 7 days post inoculation (dpi) at the same time that susceptible plants exhibited macroscopic signs of the disease. The resistance reactions were similar in the progeny population of ‘Honeycrisp’ × ‘Twin Bee Gala’, although they were delayed to 10–14 dpi possibly due to variable greenhouse conditions. This population segregated 3 resistant:1 susceptible, which suggests the presence of two genes in ‘Honeycrisp’ and agrees with the finding that different responses within ‘Honeycrisp’ to mixed inoculum are due to differential recognition of pathogen effectors.  相似文献

Disease incidence and severity was studied for winter wheat variety Bezostaya 1 and susceptible checks based on data from international nurseries from 1969 to 2010 and from 51 countries across major winter wheat production regions totalling 1,047 reports. The frequency of leaf rust and stripe rust occurrence was stable over time with marked increases in severity in 2001–2010 especially in Europe and Central and West Asia. Substantial global reductions in stem rust occurrence were recorded and attributed primarily to use of resistance genes although the recent emergence of race Ug99 makes wheat more vulnerable. The occurrence of powdery mildew remained globally stable over time. It was the most important foliar disease in Western and Southern Europe, where the frequency was very high for all time periods coupled with slight increases in severity during 2001–2010. The durable resistance of variety Bezostaya 1 to all four diseases was demonstrated in the study using comparisons of disease severities of Bezostaya 1 and the most susceptible entries. The Lr34/Yr18/Pm38 pleiotrophic set possessed by Bezostaya 1 is currently an important target for selection because it is now amenable to molecular selection. Increased use of genes like Lr34 combined with strategies to minimize cultivation of extremely susceptible varieties will contribute to long term maintenance of low and non-damaging disease levels. The durable disease resistance of Bezostaya 1, combined with its adaptability and good end-use quality, was a significant reason for its huge impact in agriculture over the last 50?years.  相似文献

Fusarium head blight (scab) epidemics of wheat occurred in uawy (Northern Poland) during 1998 and in Wielkopolska (West) and in Southern regions of Poland in 1999. Four species were identified in wheat heads with scab symptoms: Fusarium culmorum, Fusarium graminearum, Fusarium avenaceum and Microdochium nivale. A significant increase in the frequency of F. graminearum (between 23% and 38%), was observed, compared to about 10% during the previous decade. The mycotoxins deoxynivalenol (DON), nivalenol (NIV) and moniliformin (MON) in amounts up to 24.3, 14.2 and 1.72mgkg–1respectively, were identified in kernels samples.  相似文献

In 1993–1996, the virulence of regional populations of the wheat powdery mildew pathogen (Erysiphe graminis DC f. sp. tritici Marchal) from the Czech Republic, Austria, Hungary and Slovakia against 13 resistance genes was investigated. The populations differed mainly at the regional level. Populations from the Czech Republic, mainly from the western regions, showed higher values of virulence against the Pm4b gene. Lower frequency of virulence against Pm4b was found in Austria, and the lowest value was observed in Hungary. The differences in frequencies of virulence against Pm4a and Pm4b showed a similar geographic pattern across the four countries: a continuous decline from west to east and from north to south. Virulence against Pm2 decreased in all countries considered; virulence to pm5, Pm6, Pm8 and Mli was high throughout. Genes and gene combinations that can ensure a relatively effective biological protection against this pathogen across Central Europe at present are Pm3b, Pm2+Mld and Pm1+2+9. Czech and Slovak populations were the most complex: virulence complexity reached a maximum in Slovakia in 1994. A similar evolution, though less significant, was observed in the Czech Republic. Data on complexity of isolates suggest that Central European populations of wheat powdery mildew tend to reach an intermediate level representing the optimal number of virulence genes. Cucurbit downy mildew, caused by the oomycete Pseudoperonospora cubensis, is a devastating, worldwide-distributed disease of cucurbit crops in the open field and under cover. This review provides recent data on the taxonomy, biology, ecology, host range, geographic distribution and epidemiology of P. cubensis. Special attention is given to host-pathogen interactions between P. cubensis and its economically-important cucurbit hosts (Cucumis sativus, C. melo, Cucurbita pepo, C. maxima, and Citrullus lanatus); pathogenic variability in P. cubensis at the species, genus, and population levels; and, differentiation of pathotypes and races. Genetics and variability of host resistance and cellular and molecular aspects of such resistance are considered. A focus is given to methods of crop protection, including prevention and agrotechnical aspects, breeding for resistance—classical and transgenic approaches, chemical control and fungicide resistance. Novel technologies in biological and integrated control are also discussed. This review also summarizes the most important topics for future research and international collaboration.  相似文献

Two-way effects of herbicide on rice resistance and multiplication and feeding of the rice brown planthopper (BPH), Nilaparvata lugens (Stål) were studied. Four of 11 herbicides tested, butachlor, metolachlor, oxadiazon and bentazone, had significantly stimulating effects on multiplication and feeding of BPH. Multiplication times (N 1/N 0) of BPH on the rice treated with the four herbicides were 674, 367, 322 and 311, respectively, while that of N. lugens on the untreated plants was 159. The results of measurements of honeydew production by BPH indicated that the feeding rates of the brown planthoppers increased when the insects fed on the plants treated with bentazone, quinclorac, oxadiazon and quinclorac. Survival rates of N. lugens first to third-instars rose on the rice plants treated with butachlor, bentazone, but that of the fourth and fifth instars were not affected. Furthermore, all of those herbicides tested led to a decline of rice resistance, i.e. the injury scales of the treated plants were larger than that of the control under the same pest stress. Based on their effects on rice resistance and on BPH, the herbicides tested can be divided into two categories, one affecting rice resistance and BPH multiplication, the other affecting rice resistance only.  相似文献

Downy mildew of lettuce (Bremia lactucae) is a serious disease. An alternative to chemicals is the application of disease resistance inducers. The aim of this study was to test whether DL--amino butyric acid (BABA) and Phytogard® (K2HPO3) could induce resistance in downy mildew susceptible plants. Aqueous solutions of BABA (0, 10, 20, 30, 50, 80, 100mM) and Phytogard® (0.0, 5.8, 29.0, 40.6, 58.0 and 87.0ppm) were sprayed on seven-day-old seedlings 0, 3, 7 and 15 days before or 1–3 days after inoculation with B. lactucae. Results obtained showed that Phytogard®- and BABA-induced resistance was dose-dependent. At 40.6ppm for Phytogard® and 10mM for BABA, complete protection was obtained. Both compounds had a curative effect and the induced resistance lasted for at least 15 days. It was also shown that both compounds induced systemic resistance in lettuce against downy mildew. Phytogard® at 40.6ppm completely inhibited spore germination while BABA at 20mM did not. Pathogenesis related (PR) protein analysis showed that BABA induced weak accumulation of PR-2, but not PR-1, PR-5 and PR-9. Phytogard® induced none of these proteins. The use of these two compounds to protect lettuce from B. lactucae is discussed.  相似文献

The erythrocyte, due to its role as O2 and CO2 transporter, is under the constant exposure to reactive oxygen species and oxidative stress. The objective of this study was to investigate the ability of 2,4-D to induce oxidative stress in blood of male wistar rats. Rats were randomly divided into four groups: a control group and three treated groups receiving by gavage 15, 75 and 150 mg, respectively, of 2,4-D/kg/BW/day for 28 days. Results showed that 2,4-D caused significant negative changes in the investigated biochemical parameters. In fact, 2,4-D exposition strongly increases LDH, by contrast, there is a statistically significant decrease in Hgb levels. The malondialdehyde level was significantly increased in 2,4-D treated groups. Fatty acid composition of the erythrocytes was also significantly changed with 2,4-D exposure, in favor of the peroxidation of polyunsaturated fatty acids. Furthermore, antioxidant enzyme (SOD, CAT, GPx, and GR) activities in erythrocytes were significantly decreased. Thus, our results indicated the potential effects of 2,4-D to cause oxidative stress in rat erythrocytes. Therefore, at higher doses, 2,4-D may play an important role in the development of vascular disease via lipid peroxidation and oxidative stress.  相似文献

Abstract: The adult male of dracunculoid nematode Ichthyofilaria argentinensis Incorvaia, 1999 (Guyanemidae) is described for the first time based on specimens found in the swimbladder of its type host, Merluccius hubbsi Marini (Merlucciidae), caught off the coast of Buenos Aires, Argentina (western Atlantic Ocean). In addition, the males of Ichthyofilaria bergensis (Wülker, 1930) K?ie, 1993 are redescribed from specimens collected from the body cavity and visceral surface of Molva macrophthalma (Rafinesque) (Lotidae) caught in the western Mediterranean Sea, off the coast of Sardinia. Light and scanning electron microscopy examinations revealed some new morphological features for the genus, such as a pair of deirids located near the end of muscular oesophagus, the body wall conspicuously twisted immediately anterior to the cloaca, the presence of a copulatory plate, one pair of adcloacal papillae and a pair of phasmids situated on the posterior half of the tail. On the basis of this material, the generic diagnosis of Ichthyofilaria is modified to include some of these newly observed features, as well as to indicate the absence of spicules. The diagnosis of Guyanemidae is extended to include that a copulatory plate and/or two spicules may be present as characteristics for this family.  相似文献

Samara is the reproductive organ (seed) for many tree species in arid land in northwestern China. It is ecologically important in population development due to its dispersal function. However, information on its photosynthesis and effect of environmental stresses on its photosynthesis is still very limited. In the present study, responses of photosystem II (PSII) activity in samara and leaf of Siberian maple to short-term chilling/freezing and subsequent recovery potential were comparatively investigated by using polyphasic fluorescence test. The samara had more efficient photosynthesis (Fv/Fm and PIABS) and more efficient electron transport (φEo) but lower energy dissipation (DIo/RC) than leaf. Generally, the PSII performance and the electron transport for both samara and leaf were inhibited under low temperature stress, accompanied by an increase of energy dissipation in PSII reaction centers (RCs). PSII of both samara and leaf was not markedly affected by chilling and could acclimate to chilling stress. Short-term freezing could completely inhibit PSII activity in both samara and leaf, indicated by the drop of values of Fv/Fm, PIABS, φEo to zero. PSII functional parameters of short-term dark frozen samara could be largely recovered whereas those of frozen leaf could not be recovered. The higher tolerance of samara to short-term low temperature stress than leaf is of great ecological significance for seed development, population establishment of Siberian maple.  相似文献

A 60% azadirachtin-A concentrate has been obtained through repeated precipitation with hexane from a methanolic solution of a 20% concentrate. Azadirachtin-A (90%) has been obtained by medium-pressure liquid chromatography of the 60% concentrate with an RP-18 column and a methanol + water (1 + 1 by volume) solvent system. Catalytic hydrogenation of the 60 and 90% azadirachtin concentrates yielded the corresponding tetrahydroazadirachtin concentrates. Dihydroazadirachtin and tetrahydroazadirachtin formed during the first 5 h of hydrogenation were identified by LC-ESI-MS on the basis of their unique mass fragmentation pattern. The efficacy of tetrahydroazadirachtin concentrates in inhibiting the feeding and growth of Helicoverpa armigera (Hübner) larvae has been compared with that of azadirachtin concentrates. They were in general more active and deterred larvae from feeding at all concentrations. Tetrahydroazadirachtin-A (90%) and azadirachtin-A (90%) with respective IC(50) values of 280 and 390 mg L(-1) were effective as insect growth regulators, while tetrahydroazadirachtin-A (90%) displayed higher antifeedant activity (AI(50) 14 mg L(-1)) against the test insect.  相似文献

The full-length cDNA sequence of γ-aminobutyric acid (GABA) receptor alpha2 subunit gene was cloned from Spodoptera exigua, an economically important pest species in China, through rapid amplification of cDNA ends (RACE) and polymerase chain reaction using primers based on the 5′ and 3′-ends of the mRNA. The gene (named as SeGABARα2) was 1620 bp long, with an open reading frame of 1500 bp encoding a predicted GABA receptor protein of 499 amino acids with a molecular weight of 55 kDa. The predicted GABA receptor protein shared 96.19%, 82.16%, 74.30%, 73.95%, 73.42%, 71.40%, 67.11% and 65.13% identity with other GABA receptors proteins isolated from Heliothis virescens, Plutella xylostella, Aedes aegypti, Laodelphax striatellus, Lucilia cuprina, Musca domestica, Drosophila simulans and Drosophila melanogaster, at amino acid level, respectively. The developmental changes and tissue-specificity of the relative mRNA expression levels of SeGABARα2 gene were investigated in S. exigua. The highest expression level was observed in adult and lowest was in the first instar, there was a stable expression level during the developmental period from the second instar to the adult. Expression levels were 1.41-, 1.57-, 1.45-, 1.80-, 1.82- and 1.93-fold higher in the second instar, third instar, fourth instar, fifth instar, pupa and adult than in the first instar larva, respectively. The relative mRNA expression levels of SeGABARα2 gene were 3.51-fold higher in head and 1.77-fold higher in thorax than in abdomen  相似文献

Mesta (Hibiscus sabdariffa) is an important bast fiber crop. In August 2011, there was an outbreak of a phytoplasma-like disease on H. sabdariffa in different villages of the northern coastal mesta-growing region of Andhra Pradesh, India, covering mainly two districts – Srikakulam and Vijayanagaram. The infected plants showed characteristic symptoms such as phyllody and reddening of leaves. PCR with P1/P7 universal primer pair of 16 S rDNA yielded amplicons of 1850 bp from all symptomatic mesta leaf samples similar to samples of brinjal little leaf (phytoplasma positive reference control). However, asymptomatic samples were not amplified. Multiplex nested-PCR showed simultaneous amplification of DNA fragments with phytoplasma specific primers, viz., P1/P7 universal primer pair of 16 S rDNA, nested primer pair R16F2n/R2, uvrB and DegV gene-specific uvrB-degVF/R primer generating amplicons of 1850 bp, 1200 bp and 1023bp, respectively. However, SecY-map gene specific primer SecY-mapF/R was not amplified. The 1023 bp nucleotide sequence of uvrB and DegV gene of the phytoplasma was deposited in the GenBank (NCBI) with the accession no. JX975061. NCBI BLASTn analysis of the 1023 bp products showed that the phytoplasma strain belonged to elm yellows group (16SrV-D). This is the first report that Hibiscus sabdariffa is infected by a phytoplasma and we named it mesta phyllody disease (MPD).  相似文献
