Two hundred and twenty rainbow trouts (IBW: 700 g) were randomly allotted to four tanks, with a male/female ratio of 0.56. Fish were fed for 168 d with four experimental diets containing herring oil, cod liver oil and coconut oil with the following inclusion rates: diet A: 12-1-0% respectively; diet B: 6-1-6%; diet C: 0-1-12%; diet D: 0-0-13%. Irrespective of the dietary treatment, weight gains of broodstocks were high (> 3 g/d) and FCR below 2. No significant difference was observed concerning the total amount of eggs spawn, egg average weight (82.5 mg/egg) and lipid content (5.4 mg/egg). However, the fatty acid profile of eggs was significantly affected by the dietary treatments. The content of unsaturated fatty acids, particularly the n-3 fatty acid series (EPA and DHA) significantly decreased with increasing levels of coconut oil in the diet.  相似文献

This paper reports on inclusion of Spirulina platensis (SP) in rainbow trout (Oncorhynchus mykiss) fish diet and its effect on the fish tissue fatty acids (FAs) composition. Fish in triplicate groups with average initial body weight of 75 ± 0.23 g and age of about 7 months were fed a control diet with no supplements and with three experimental diets supplemented with 2.5, 5 and 7.5 weight percent (wt%) of SP for 12 weeks. At the end of experiment, the fillet FA compositions were evaluated. It was found that the composition of FAs in fish fillet was significantly (P < 0.05) affected by inclusion of SP in diets. As compared to the control sample, feeding the fish supplemented particularly with low levels of SP improved the quality of FAs by decreasing undesirable saturated FAs (SFAs) content from 20.03 to 17.93 % and increasing the level of some beneficial long-chain highly unsaturated FAs namely eicosapentaenoic acid (C20:5 n-3) from 6.47 to 7.27 %, docosapentaenoic acid (C22:5 n-3) from 1.42 to 1.56 % and docosahexaenoic acid (C22:6 n-3) from 1.26 to 1.35 %. Further increase in the SP inclusion level generally did not lead to desirable results. The SP inclusion in the fish diet had no significant impact on overall contents of n-3 and n-6 polyunsaturated FAs in fish fillet. However, the total highly unsaturated FAs (HUFAs) and n-3 HUFAs levels, UFAs/SFAs ratio and n-3/n-6 index were increased from 11.25, 10.65, 3.98 and 0.57 to 12.13, 11.56, 4.40 and 0.62 %, respectively, as a result of 5 wt% SP loading. In general, it can be concluded that SP inclusion up to 5 wt% of loading in rainbow trout culture can be beneficial in terms of FAs compositions of the fish fillet.  相似文献

Rainbow trout (Oncorhynchus mykiss) skin cell cultures were obtained by trypsinization of the tissue and grown in Leibovitz L-15 medium. Lipid class compositions, and fatty acid profiles of total lipids and individual phospholipid classes were determined at different times of culture. The metabolism of polyunsaturated fatty acids (PUFA) was investigated by incubating primary cultures after 7 and 14 days with [1-14C]18:2n-6 and [1-14C-]18:3n-3. The change in morphology between epithelial-like primary cultures and fibroblastic-like secondary subcultures was accompanied by alterations in the lipid composition. Polar lipids became predominant by 14 days in culture. The relative proportions of phosphatidylcholine (PC), the most abundant phospholipid, phosphatidylinositol and cholesterol increased significantly, while sphingomyelin decreased. Saturated fatty acids, 18:1n-9, n-6 and n-9PUFA were more abundant in total lipid in cultures at 14 days and 4 months than in cells initially isolated which contained higher percentages of longer chain monoenes and n-3PUFA. The changes in fatty acid composition with time in culture were observed in all the major phospholipid classes. Rainbow trout skin cells in culture desaturated and elongated both 18:2n-6 and 18:3n-3, with 20:4n-6 and 20:5n-3 being the most abundant products, respectively. PC presented the highest incorporation of radioactivity, especially following incubation with 18:3n-3. Lipid metabolism in general increased with the age of primary cultures, with both the amount of C18 PUFA incorporated and metabolized by desaturation/elongation significantly increased in 14 day cultures compared to 7 day cultures. Product/precursor ratios calculated for both n-6 and n-3 fatty acids showed that, while 6 desaturase activity was increased significantly with cell age, 5 desaturase activity was more affected by the fatty acid series, with 18:3n-3 being more readily transformed to 20:5n-3 than 18:2n-6 to 20:4n-6. Further desaturation of 20:5n-3 to hexaenes was low. Overall, the data suggested that the trout skin cell cultures were more similar to mammalian skin fibroblasts than mammalian epidermal/keratinocyte cultures.  相似文献

The aim of this study was to determine the effects of replacing fish oil (FO) with laurel seed oil (LSO), as an alternative plant lipid source in diets on the growth and fatty acid composition of rainbow trout (Oncorhynchus mykiss; 111.47 ± 0.2 g mean individual weight). At the end of the feeding trial, survival was 100% in all treatments. No significant differences were seen in growth between the dietary groups (P > 0.05). The protein, lipid and ash contents were not significantly different among the groups (P > 0.05); however, there was a significant difference in protein and ash content between the treatment groups and the initial, and between the 50LSO group and the initial group, respectively (P < 0.05). The viscerosomatic index (VSI) and hepatosomatic index (HSI) values were not affected by increasing LSO percentages in the diets. The n‐6 polyunsaturated fatty acid (PUFA) concentration increased with increasing LSO levels in the diets. In contrast, the n‐3 PUFA levels decreased with increasing LSO levels in the diets. The liver and muscle were used for the analysis of fatty acids. The highest level of eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) concentrations was recorded in fillet of fish fed the FO diet and the lowest in those fed the 50LSO diet. However, EPA and DHA ratios in the liver of fish fed the 75LSO diet were higher than those in fillet of fish fed the FO and 50LSO diets. No significant differences were seen in fatty acid composition between the dietary groups (P > 0.05). Based on the results of growth performance and fatty acid composition of the experimental fish in this study, it can be concluded that the 75% concentration of laurel seed oil performed best among the diets tested in the experiment.  相似文献

This study evaluated the effects of diets containing 0, 25, 50, 75 and 100 g kg?1 Spirulina platensis on proximate composition, fatty acid profile and lipid peroxidation of rainbow trout (Oncorhynchus mykiss). Supplementation of S. platensis did not change moisture and protein contents, but fish fed 50 and 100 g kg?1 S. platensis had lower muscle lipid content than those fed control diet (< 0.05). Fish fed 100 g kg?1 of S. platensis contained lower percentages of saturated and monounsaturated fatty acid and a higher percentage of polyunsaturated fatty acid than those fed control diet (< 0.05). The n‐3/n‐6 ratio of the fatty acid increased and muscle atherogenic and thrombogenic indices were significantly decreased as the dietary supplement of S. platensis increased. Furthermore, lipid peroxidation of the fillet significantly decreased with increasing dietary S. platensis at 4 °C and at ?20 °C (< 0.05). The results of this study show that supplementation of S. platensis to the diet improves muscle quality of the rainbow trout.  相似文献

This study aimed to investigate the effects of dietary crude palm oil (CPO) on fatty acid metabolism in liver and intestine of rainbow trout. Triplicate groups of rainbow trout for 10 weeks at 13 °C were fed on diets in which CPO replaced fish oil (FO) in a graded manner (0–100%). At the end of the trial, fatty acid compositions of flesh, liver and pyloric caeca were determined and highly unsaturated fatty acid (HUFA) synthesis and fatty acid oxidation were estimated in isolated hepatocytes and caecal enterocytes using [1‐14C]18:3n‐3 as substrate. Growth performance and feed efficiency were unaffected by dietary CPO. Fatty acid compositions of selected tissues reflected the dietary fatty acid composition with increasing CPO resulting in increased proportions of 18:1n‐9 and 18:2n‐6 and decreased proportions of n‐3HUFA, 20:5n‐3 and 22:6n‐3. Palmitic acid, 16:0, was also increased in flesh and pyloric caeca, but not in liver. The capacity of HUFA synthesis from 18:3n‐3 increased by up to threefold in both hepatocytes and enterocytes in response to graded increases in dietary CPO. In contrast, oxidation of 18:3n‐3 was unaffected by dietary CPO in hepatocytes and reduced by high levels of dietary CPO in enterocytes. The results of this study suggest that CPO can be used at least to partially replace FO in diets for rainbow trout in terms of permitting similar growth and feed conversion, and having no major detrimental effects on lipid and fatty acid metabolism, although flesh fatty acid compositions are significantly affected at an inclusion level above 50%, with n‐3HUFA reduced by up to 40%.  相似文献

The use of macroalgae as an additional component in animal feeding has been studied. However, information on how it could influence muscle composition of fish body is scarce. This study evaluates four diets with different macroalgae inclusion levels (0%, 1.5%, 3% and 6%) to test the effect on body fatty acid composition of rainbow trout. Tanks with a volume of 600 L were stocked with 60.6 ± 7.9 g fish at a density of 45 individuals tank?1 and fed for 124 days. At the end of the experiment there were not significant differences (P<0.05) in muscle proximate composition among fish fed the different treatments. However, it was determined that inclusion of 3% and 6% of macroalgae meal resulted in a significant increase (P<0.05) of polyunsaturated fatty acids (PUFAs) in muscle. In summary, macroalgae meal in rainbow trout diets do not enhance the quantity of protein and lipid contents at muscle level but an addition of 3–6% might contribute to increase the level of PUFAs, specially EPA, DHA and LIN. Thus, use of macroalgae meal might help to increase lipid quality content in the final product due the beneficial effects of PUFAs for human health.  相似文献

Rainbow trout (186 g) were fed three test diets where the lipid source (150 g kg?1) was either menhaden oil (MO), pollock oil (PO) or canola oil (CO) for eight weeks to an average weight of 370 g. The CO group was then divided into two groups, one continuing on the CO diet and the other switched to the PO diet (CO–PO). After nine additional weeks of feeding, the average fish weight approximately doubled (719–749 g). No significant differences were found in average final weight or fillet yield among dietary treatment groups. Fatty acid profiles of fillets from trout fed MO, PO or CO‐supplemented diets reflected the fatty acid profiles of the added oils, whereas the fatty acid profile of fillet from trout in the CO–PO group exhibited values similar to those of fish fed PO. The ratio of ω3 : ω6 FA was nearly 2.5 times higher in fillets from the CO–PO group compared to the CO group. Sensory analysis showed that panelists preferred CO‐fed fillets over those fed MO, PO, or CO–PO. Phase‐feeding CO and PO reduced fish oil use and resulted in fillets with double the content of Eicosapentaenoic acid (EPA) and Docosahexaenoic acid (DHA) over CO‐fed fish, similar to levels in MO‐fed fish.  相似文献

Rainbow trout Oncorhynchus mykiss triploids are regularly produced in fish farms to improve growth because the triploid females do not develop ovaries during the reproductive cycle. In this study, the tissue fatty acid allocations in triploid and diploid females were compared during the reproductive cycle to determine whether the ploidy influences the fatty acid profile of fish produced in aquaculture. The ovaries, liver, and white muscle fatty acid contents of diploid and triploid females were analyzed during the reproductive cycle. Diploid females tend to accumulate more polyunsaturated fatty acids than triploids during some phases of the reproductive cycle, and this profile was compensated by an increase in saturated and monounsaturated fatty acids in triploid females. Arachidonic acid (C20:4n6) was the main n6 polyunsaturated fatty acid in the ovaries of diploid females during the most advanced phases of the reproductive cycle, and docosahexaenoic acid (C22:6n3) was the main n3 polyunsaturated fatty acid. In triploid females, the percentage of both of these polyunsaturated fatty acids was lower than in diploid females during the most advanced phases of the reproductive cycle. In general, the lack of ovary development altered the hepatic synthesis of some fatty acids, mainly monounsaturated and polyunsaturated fatty acids, decreasing the content of the main fatty acids in the white muscle and, consequently, the mobilization of these specific fatty acids to the ovaries.  相似文献

Uncoupling proteins are mitochondrial anions transporters that dissociate respiration from ATP synthesis through proton leaks. Uncoupling protein 2 reportedly plays a role in several physiological processes such as energy partitioning, nutrition, and fatty acid metabolism. The mRNA expression of rainbow trout UCP2 genes (UCP2A and UCP2B) was monitored during embryogenesis and early larval development. Both genes were recruited early and displayed similar steadily decreasing patterns from fertilization until hatching. The expression of UCP2A and UCP2B appeared significantly differentiated after hatching and during the yolk sac absorption, with UCP2A displaying higher expression. We suggest that UCP2 expression profiles in the rainbow trout embryo could be associated with the utilization of lipids as a source of energy during development.  相似文献

This study examined how muscle metabolic organization varied during an annual cycle in which rainbow trout (Oncorhynchus mykiss) were held in outdoor holding ponds in which they were exposed to natural changes in temperature (range 0.2 to 15.6°C) and photoperiod. We examined the activities of glycolytic and mitochondrial enzymes in red and white muscle to evaluate whether trout enhance their capacity for lipid and carbohydrate oxidation during cold-acclimization. When assayed at habitat temperature, the enzyme activities generally increased in spring to reach a maximum in summer followed by a decrease in the fall. This led to significantly higher activities at warm than cold periods for all enzymes measured in red muscle and all but one in white muscle. The activities at 10°C provided little evidence for compensatory adjustments of aerobic capacity. Particularly in red muscle, enzyme levels at 10°C were generally lower during cold than warm periods. The variation of enzyme activities throughout the cycle was not due to changes in protein concentration, as the same responses were observed when activities were expressed per g wet mass or per mg protein. Although the aerobic capacity did not increase with cold-acclimatization, the relative capacity for lipid oxidation was higher in winter than in summer trout. In contrast, the relative capacity for aerobic glycolysis was higher in summer than in winter trout. Thus, the metabolic capacities of trout muscle undergo seasonal reorganization.  相似文献

The effect of dietary inclusion of whole grain white lupin (Lupinus albus) on growth performance, histology, muscle fatty acid composition and nutrient digestibility was investigated in an 11‐week growth and a 4‐week digestibility trial with rainbow trout (initial body weight of 54.0 ± 6.2 and 181.9 ± 3.4 g respectively). Four experimental extruded diets were formulated to contain 0%, 30%, 40% and 50% of whole grain lupin and fed to triplicate groups of fish twice a day until apparent satiation. Faeces were collected daily from each digestibility tank by decantation. No significant trends were observed with respect to growth, feed utilization, apparent digestibility coefficients or whole‐body composition (P>0.05). Conversely, increasing levels of dietary lupin led to significant decreases in the Hepatosomatic index (R2=0.75, P<0.05) and slight lipid infiltration into hepatocytes and enterocytes. Muscle fatty acid compositions were slightly affected by the dietary treatment. Polynomial regression of dietary inclusion of lupin and muscle fatty acid concentrations showed an increase in C18:1n‐9, C18:2n‐6 and C18:3n‐3 and a decrease in C20:5n‐3 with increasing dietary lupin level. These results demonstrated that whole grain lupin can be included up to 50% in commercial rainbow trout diets without negative effects.  相似文献

Edwardsiella tarda was isolated from naturally infected rainbow trout (Oncorhynchus mykiss) in raceway culture on a commercial fish farm where the fish were kept at a density of 110 kg m?3 and at a water temperature of 14°C and with a photoperiod of 13 h light:11 h dark. The clinical signs of diseased fish (150 ± 20 mm standard length) were anorexia and lethargy. The most striking lesions in the fish were in the liver. There were hyperaemia and haemorrhages; on histopathological examination, the liver displayed inflammatory infiltrate in portal area, focal necrosis, dilatation of blood sinus and activation of sinusoidal cells. Infection experiments, performed 2 years after isolation of the original culture of E. tarda, were carried out under laboratory conditions at water temperatures of 15, 18 or 24°C. All experimental fish (common carp, Prussian carp, tench), intraperitoneally injected with 8 × 106 cells, demonstrated a total resistance to E. tarda.  相似文献

This experiment intended to assess the effect of sesame (SO), sunflower (SFO) and linseed (LO) oils on growth performance, fatty acid composition of fillet and liver or flesh quality traits of rainbow trout. Fish fed different four iso‐nitrogenous and iso‐lipidic experimental feeds. The control feed contained only fish oil as the primary lipid source. The fillet eicosapentaenoic acid and docosahexaenoic acid levels were the highest in fish fed control feed. In contrast, the liver eicosapentaenoic acid level was the highest in fish fed LO feed. Fish fed SFO feed had the highest level of total n?6 fatty acids in fillet and liver. Fish fed SO feed had the highest level of 18:1 n?9 fatty acid in fillet and liver. During the 12 days refrigerated storage period at 1°C, thiobarbituric acid (TBA) and pH values gradually increased in all dietary groups. However, trimethylamin nitrogen (TMA‐N) values increased in all dietary groups between days 0 and 9 during the storage period. Generally, pH value in fillets of control group was slightly higher than the other fish groups during 12 days refrigerated storage. Nevertheless, the chemical indicators of spoilage, TBA, TMA‐N and pH values were in the limit of acceptability for human consumption. Results of growth performance and chemical tests in the present study showed that sesame, linseed and sunflower oils could be used in feeds for rainbow trout. Moreover, we concluded that further researches should be carried out on the partial replacement of fish oil by sesame oil in rainbow trout nutrition.  相似文献

This study investigated the effects of different soya oil products on growth, nutrient digestibility, and fatty acid composition of muscle in rainbow trout. The products’ ingredients were soap stock, fat powder, crude oil, and refined soya oil as a control. Four experimental diets were formulated by addition of 12 % of each of the four ingredients to a basal diet. Replacement of refined soya oil by the soya oil products did not influence growth performance until 8 weeks (P > 0.05). This replacement, however, reduced fat digestibility in rainbow trout (83.7 vs. 73.3–79.8 %; P < 0.05). Dietary inclusions of fat powder and crude oil led to larger values of protein digestibility than those of refined soya oil and soap stock (P < 0.05). The compositions of saturated and monounsaturated fatty acids of the muscle were similar, but linoleic acid concentration (LA, 18: 2n-6) increased in fish muscle fed refined soya oil compared to the other diets (37 vs. 30–31 %; P < 0.05). Arachidonic acid (ArA, 20: 4n-6) concentration was higher (P < 0.05) in fish fed both fat powder and soap stock than those fed the other diets. In conclusion, refined soya oil can be replaced by soya oil products in rainbow trout diet with no negative impacts on the growth. However, fish fed on diet containing refined soya oil had higher concentration of LA and lower density of ArA. It appears that rainbow trout is capable of digesting soya oil products though the digestibility rate reduced slightly with an increase in saturated fatty acid content.  相似文献

The aim of the present study was to determine the effects of the use of astaxanthin alternate feeding on rainbow trout pigmentation in term of astaxanthin serum concentration, muscle colour and astaxanthin muscle retention. Four hundred and fifty rainbow trout were fed the same basal diet supplemented with two different astaxanthin levels, 100, and 200 mg astaxanthin kg? 1 of diet, hereafter designated as AX100 and AX200, respectively. An additional astaxanthin-free (AX0) diet was used. The experimental treatments were as follows: (1) REF = AX100 diet at each meal each day, served as reference; (2) SD1 = AX100 diet at each meal the first day followed by AX0 diet at each meal every second day; (3) SD2 = AX100 diet and AX0 diet in alternate meals each day; (4) R2 = AX200 diet and AX0 diet in alternate meals each day; (5) R4 = AX200 diet at the first meal the first day followed by AX0 diet at the second meal the first day and at each meal every second day. Fish were fed the experimental feeding schedule for 42 days.At the end of the experiment there were no significant differences among fish fed the different feeding schedules in term of final mean weight, specific growth rate and feed efficiency ratio. SD2 fish group displayed the highest (P < 0.05) astaxanthin serum concentration and the R4 fish group the lowest one. REF and R2 fish groups showed similar astaxanthin serum concentrations. Muscle chroma showed the most pronounced effect. It increased significantly for all fish groups during the experiment. At the end of the experiment REF and R2 fish groups displayed higher values than SD1 and R4 fish groups. Muscle astaxanthin concentrations increased significantly during the experiment whatever the astaxanthin feeding schedule. At the end of the experiment, the highest muscle astaxanthin concentration was recorded for R2 fish group while the lowest was noted for R4 fish group. Except for SD1 and R4 fish groups, muscle astaxanthin retention decreased significantly during the experiment. At the end of the experiment, muscle astaxanthin retention coefficients for SD2 fish group were significantly higher than those for REF fish group. The results reported here provide further evidence of the potential applicability of alternate astaxanthin feeding on rainbow trout pigmentation. Extending the optimisation of the SD2 treatment will therefore be subject for future studies. Its application could result in cost saving in the fish farming industry.  相似文献

Extreme acid-base imbalances in salmonids have been shown to impact on gastrointestinal motility. Changes in pH are known to affect cardiac and skeletal muscle in fish and mammalian intestinal smooth muscle preparations. This study was conducted to determine if the contractility of rainbow trout intestinal muscle was sensitive to pH fluctuations and to begin the investigation into the mechanisms by which pH affected contractility. Isolated duplicate or triplicate segments of rainbow trout intestine were suspended in organ baths containing modified Krebs-Henseleit solution. The pH was adjusted by varying the concentration of CO2 aerating the solution and with HCl or lactic acid. Contractility was determined as response to the administration of 5-hydroxytryptamine (5-HT, serotonin), KCl, and transmural stimulation within the pH range of 6.3 to 8.5. Optimum pH for proximal segments was 7.85 while the range was much wider for distal segments. Decreasing the pH with CO2 resulted in a greater inhibition of smooth muscle contractility than when the pH was decreased in 100% O2 with either HCl or lactic acid, particularly when the tissues are stimulated electrically. This effect was ameliorated as the fish attained sexual maturity, although the mechanisms involved were not clear. The Cl-/HCO 3 - exchanger appeared to be involved in recovery from acidotic stress, particularly at pH levels below 6.5. The role played by the Na+/H+ exchanger is still unclear. Although contractility was inhibited in the presence of the amiloride analog EIPA (5-(N-ethyl-N-isopropyl)amiloride), whether the inhibition was due to blockage of ion transport or to cytosolic effects is yet to be determined.  相似文献

Information on the metabolism of exogenous histamine in fish is of much concern regarding the effect of dietary histamine on fish. Histamine catabolic enzymes, diamine oxidase and histamine N-methyl transferase were measured in the tissues of rainbow trout. Diamine oxidase was detected in the stomach, pylorus caeca and intestine. Histamine N-methyl transferase was detected only in the liver.A change in the contents of histamine and its metabolites was observed in the tissues of rainbow trout after oral administration of histamine. A large amount of imidazole acetic acid was observed in the serum, kidney, liver and muscle. On the other hand, 1-methyl histamine was detected only in the liver. Histamine and its metabolites, imidazole acetic acid and 1-methyl histamine were metabolized and diminished within 48 hr in all tissues.These results showed that histamine was metabolized by two metabolic routes in rainbow trout. One is the main pathway producing imidazole acetic acid by intestinal diamine oxidase and the other is the complementary pathway producing 1-methyl histamine by liver N-methyl-transferase.  相似文献
