The efficacy of vaccination with Newcastle disease (ND) La Sota and R2B (Mukteswar) modified live strain vaccines was determined by experimental challenge and with ND La Sota vaccine under field conditions in Nepal. Booster vaccination with ND La Sota vaccine after a primary vaccination with ND La Sota vaccine, induced a geometric mean titre (GMT) of 5.0 log2 haemagglutination inhibition (HI) units, compared to a GMT of 6.0 log2 HI units following booster vaccination with R2B vaccine 1 month after primary vaccination with ND La Sota vaccine. Both vaccines provided 100% protection against challenge with a local field ND strain. Furthermore, booster vaccination with ND La Sota vaccine induced protective levels of antibody after field use in villages in Jhapa, and no outbreaks of ND occurred during the study period. The ND La Sota modified live vaccine is immunogenic and efficacious and is a suitable vaccine for use in vaccination programmes in village chickens in the rural areas of Nepal.

Antibody responses in indigenous village and commercial chickens vaccinated with 12 thermostable Newcastle disease (ND) vaccine and protection levels against challenge with a virulent field isolate were determined. The antibody response of village chickens vaccinated by eye drop revealed that 30, 60 and 90 days after primary vaccination, the mean log2 HI titres were 6.1, 5.4 and 3.6, respectively, whereas for commercial chickens, the antibody response after 14, 30 and 90 days were 8.2, 5.1 and 4.2, respectively. Village chickens vaccinated orally via drinking water had mean log2 HI titres of 3.4 after 30 days. After booster vaccination, the mean HI titre was 5.4 and 3.3 after 30 and 60 days post-secondary vaccination (i.e. 60 and 90 days after primary vaccination). Antibody response of mean log2 HI titres of 2.6 was recorded 30 days after primary vaccination orally through food; 30 and 60 days after secondary vaccination (i.e. 60 and 90 days after primary vaccination), mean log2 HI titres were 5.3 and 3.2, respectively. All commercial and village chickens vaccinated by eye drop survived the challenge trial whereas village chickens vaccinated through drinking water and food had protection levels of 80% and 60% 30 days after primary vaccination, respectively. However, 30 days after booster vaccination, the protection level was 100%. At 60 days after secondary vaccination, the protection level dropped again to 80% for chickens vaccinated orally. All control chickens used in the challenge trials developed clinical ND and died 3-5 days after inoculation with the virulent virus. Supported by laboratory findings, I2 strain of NDV seemed to be avirulent, immunogenic and highly protective against virulent isolates of NDV. The aim of this study was to develop a strategy to control Newcastle disease (ND) in free ranging village chickens using the Nobilis ND Inkukhu vaccine (Intervet South Africa). The study was conducted at Thibella village in Qwa-Qwa, South Africa from April 2001 to October 2002. Three different routes of vaccination (administration via eye-drop, drinking water and feed) were investigated. The haemagglutination inhibition (HI) test was conducted monthly in order to measure the antibody response of village chickens after immunization against Newcastle disease. Using a South African isolate of velogenic ND virus, challenge trials were conducted to determine the efficacy of the vaccine. A questionnaire was provided to evaluate perceptions of farmers on vaccinations. The eye-drop vaccination route produced the highest HI titres ranging between 2.7 and 4.4, followed by the drinking water vaccination route with titres ranging between 2.3 and 4.0. The lowest titres were from the feed vaccination route which ranged between 1.6 and 3.0. Following the challenge, the entire control group died on the third and fourth day after infection. However, 70% of the chickens immunized by using either the eye-drop or drinking water route survived the challenge. Only 20% of the chickens from the group immunized through the feed route survived. Evidently both the eye-drop and drinking water routes were efficient in preventing disease. Necropsies showed that vaccinated chickens had mild lesions whilst control chickens had severe lesions compatible with Newcastle disease. The efficacy of the vaccine using either of the routes can be enhanced by administration of booster vaccinations at 3-month intervals during the first year of a vaccination campaign and then at 6-month intervals from the second year onwards. Since 1997, severe outbreaks of Newcastle disease (ND) in geese in many regions throughout China have resulted in high morbidity and mortality, and great economic loss to farmers; however, no licensed, specific vaccine is yet available for this disease in China. In this study, goslings were immunized with different combinations and dosages of several commercial ND vaccines including La Sota vaccine, Mukteswar vaccine, recombinant live vaccine against avian influenza (AI) and ND (rL-H5 strain), and inactivated ND oil-emulsion vaccine (La Sota strain). The protective effects were evaluated based upon the level of antibody response and the degree of protection against the goose-origin virulent NDV strain. The result showed that two doses (i.e., one more than that for chicken) of La Sota vaccine priming, followed by 2-5 doses of Mukteswar vaccine boosting 2-3 weeks later, not only induced higher HI antibody levels, but also conferred longer-lasting protection. This immunization procedure can be recommended for prevention of ND in geese.  相似文献

1-day-old SPF chickens and commercial Jingfen chickens were vaccinated with IBD immune complex(IC) vaccine, NDV La Sota vaccine were inoculated simultaneously every one week and every two weeks.NDV La Sota immunization alone was as the control group.At the 2nd, 3rd, 4th and 5th week post inoculation, blood samples were taken and the ND HI antibody were tested.Experimental chickens were challenged with high virulent NDV at the 5th week post inoculation, the protective rate of each group was calculated.The results showed the ND HI antibody were not significant different in the combined immunization of IBD IC vaccine priming and NDV La Sota vaccine boost and NDV La Sota vaccine alone immunization (P>0.05).The results indicated that IBD IC vaccine has no immunosuppression on NDV La Sota vaccine in SPF chickens and commercial Jingfen chickens.  相似文献

鸡新城疫CHR 株冻干疫苗中试产品3 批以及La Sota 株冻干疫苗( 符合质量要求) , 以5 种免疫途径: 肌注、滴鼻、喷雾、饮水、拌料分别接种11 日龄雏鸡。每批疫苗每种途径接种1 组, 20 只/ 组, 共20 组。雏鸡群免疫前血凝抑制抗体(HI) 效价为1∶2-13 , 于14 d 、28 d 后再测HI抗体水平, 并分别用10 000 鸡胚半数致死量(ELD50) 新城疫标准强毒北京株攻击。从而检测2 种疫苗用5 种免疫途径免疫后在不同时间的近期免疫效力。结果表明, 近期内CHR 株疫苗HI抗体呈上升趋势, 而La Sota 株疫苗HI抗体先升高后下降; CHR 株疫苗以饮水和拌料途径效果较好, 其它3 种途径效果相似。接种后14 d 攻毒, 2 种疫苗均能较好保护强毒攻击; 28 d 后, CHR 株保护率高于La Sota 株。  相似文献

BackgroundThe commercially available Newcastle disease (ND) vaccines were developed based on Newcastle disease virus (NDV) isolates genetically divergent from field strains that can only prevent clinical disease, not shedding of virulent heterologous virus, highlighting the need to develop genotype-matched vaccinesObjectivesThis study examined the efficacy of the NDV genotype-matched vaccine, mIBS025 strain formulated in standard vaccine stabilizer, and in carboxymethyl sago starch-acid hydrogel (CMSS-AH) following vaccination via an eye drop (ED) and drinking water (DW).MethodsA challenge virus was prepared from a recent NDV isolated from ND vaccinated flock. Groups of specific-pathogen-free chickens were vaccinated with mIBS025 vaccine strain prepared in a standard vaccine stabilizer and CMSS-AH via ED and DW and then challenged with the UPM/NDV/IBS362/2016 strain.ResultsChickens vaccinated with CMSS-AH mIBS025 ED (group 2) developed the earliest and highest Hemagglutination Inhibition (HI) NDV antibody titer (8log2) followed by standard mIBS025 ED (group 3) (7log2) both conferred complete protection and drastically reduced virus shedding. By contrast, chickens vaccinated with standard mIBS025 DW (group 5) and CMSS-AH mIBS025 DW (group 4) developed low HI NDV antibody titers of 4log2 and 3log2, respectively, which correspondingly conferred only 50% and 60% protection and continuously shed the virulent virus via the oropharyngeal and cloacal routes until the end of the study at 14 dpc.ConclusionsThe efficacy of mIBS025 vaccines prepared in a standard vaccine stabilizer or CMSS-AH was affected by the vaccination routes. 将试验鸡分成3个试验组和1个对照组。A组鸡接种鸡新城疫系疫苗,B组鸡接种油乳剂灭活苗,C组鸡接种鸡新城疫系疫苗,并在接种疫苗后第3、4、5、6、7、9、11、13、15、20、25d采取各组鸡血并分离血清,检测HI抗体。结果表明,接种系疫苗的组,HI抗体效价均值从4.67log2上升到10log2,接种后第5d开始上升,接种后第11d达到峰值,持续6d保持高滴度抗体水平。接种系疫苗的组,HI抗体效价均值从4.67log2上升到7log2,接种后第4d开始上升,接种后第9d达到峰值。接种油乳剂灭活苗的组,HI抗体效价均值从4.67log2上升到9.33log2,接种后第5d开始上升,接种后第11d达到峰值,持续16d保持高滴度抗体水平。系疫苗HI抗体效价上升快,效价高,较适合于紧急接种,油乳剂灭活苗HI抗体效价可在高水平维持较长时间,较适合于预防接种。  相似文献

用NDV分离株及La Sota株为抗源液,经福尔马林灭活后,与油佐剂混合,分别制成分离株灭活苗、La Sota株灭活苗及分离株与La Sota株二价灭活苗。将这三种灭活疫苗分别免疫SPF鸡后,均获得100%抵抗NDV分离株及F48株强毒攻击的保护力;而用这3种灭活苗与La Sota活苗单独或联合使用,免疫带有ND母源抗体的普通鸡后,3种灭活苗的免疫效力不同,分离株灭活苗与价灭活苗对NDV分离株攻击的免疫保护效力明显优于La Sota灭活苗;灭活苗与活苗同时使用,其免疫效力明显优于单独使用灭活苗或活苗。  相似文献

A lymphocyte transformation microassay was used to study cell mediated immunity (CMI) in chickens following primary and secondary vaccination with inactivated oil emulsion infectious bronchitis (IB) vaccine and subsequent challenge with Massachusetts-41 (M-41). Humoral immunity was monitored for comparison, using the haemagglutination inhibition (HI) microassay. Positive stimulation indices (2 to 2.7 after primary and 2 to 4.8 after secondary vaccination) were lower and HI titres were higher than those previously reported following primary and secondary vaccination with live IB vaccines. The highest HI titres appeared in birds which had received the inactivated vaccine as a secondary vaccination. Challenge of vaccinated and revaccinated birds resulted in strong HI and weak CMI secondary responses. There was no correlation between CMI and HI antibody production. Monitoring egg production and clinical signs showed that a high level of protection against challenge resulted from revaccination with an inactivated oil adjuvant vaccine.  相似文献

The formulation and evaluation of trehalose nano-organogels for storage and oral delivery of Newcastle disease (ND) strain I-2 vaccine to chickens were carried out in this study. Trehalose sugar was blended with vegetable oil to form nano-organogels where trehalose also acted as a stabilizer against thermal inactivation of I-2 ND virus. Results from infectivity titration assay indicated that the titre of 107.5 EID50/0.1 mL was maintained after 12 weeks of storage of nano-organogel I-2 vaccine at ambient room temperature. Serology results showed that 33% chickens which were vaccinated with nano-organogel I-2 vaccine after 14 days had HI antibody titres of ≥ 3.0 log2 with GMT of 2.3. Moreover, results showed 100% of chickens vaccinated with nano-organogel I-2 vaccine had the mean antibody titres of 3.4 and 3.7 log2 at 21 and 28 days after vaccination, respectively. All vaccinated chickens (100%) survived the challenge of virulent ND virus whereas all unvaccinated chickens succumbed to challenge and died of signs consistent with ND. The findings from this study showed that the nano-organogel I-2 vaccine was stable at room temperature, safe and produced protective antibody response in vaccinated chickens. Moreover the nano-organogel I-2 vaccine was used for oral administration and hence is suitable for mass vaccination. However, optimization of the formulation of trehalose nano-organogel vaccine is required in order to achieve its application potentials.  相似文献

Prevalence studies have shown that almost 100% of free-range chickens are infected with a wide range of parasites. The infections are mostly subclinical in nature, resulting in production losses and occasionally mortality. Newcastle disease (ND), on the other hand, results in high mortality rates during epidemics. ND is a limiting factor for increasing poultry production in many tropical countries, where frequent reports indicate vaccination failures. The aim of our study was to investigate the influence of helminths on the antibody response after vaccination against Newcastle disease of free-range chickens naturally infected with parasites. Sixty chickens were divided into six groups, of which three were vaccinated against ND with a live De Soto vaccine, while the other three remained non-vaccinated. One group within the vaccinated groups and the one within the non-vaccinated group was kept naturally infected with helminth parasites, while the other two groups in each set were dewormed with fenbendazole and niclosamide, and one of each of these groups was subsequently infected with Ascaridia galli. After vaccination, all the groups were followed for 5 weeks and their antibody titres were determined weekly using a HI test. All the birds were finally challenged 4 weeks after vaccination with a virulent velogenic ND virus obtained from a field outbreak. All the vaccinated chickens seroconverted and had high antibody levels after 3 weeks, but these dropped to low levels at 4 weeks after vaccination. After challenge, the antibody titres rose in the dewormed groups but not in the parasite-infected groups. After 5 weeks, all the parasite-infected animals had significantly lower antibody titres than the dewormed animals. All the vaccinated chickens survived the challenge infection, emphasizing the importance of the cellular immune response. Further studies are needed to examine the effects of the parasitic infection on protection against ND over a longer period.  相似文献

A locally-produced Newcastle disease (ND) I-2 thermostable vaccine of embryo-infective dose (EID50) 10(8.5) per ml was administered to 100 laboratory chickens in four test groups, each of 25 birds. It was given by the eye-drop method, in drinking water, in drinking water freshly medicated with levamisole, or using millet grains as a vaccine carrier. A fifth control group consisting of 25 birds received the heat-sensitive La Sota vaccine (EID50 10(9) per ml) by the eye-drop method. The immunological responses were monitored by the enzyme-linked immunosorbent assay (ELISA) ND antibody technique using serum samples collected from 18 birds in each group at 3-week intervals for 3 months. The overall mean ND antibody log(10) titres and percentage positivities were 3.1, 88%; 2.9, 70%; 3.0, 83%; 3.2, 87% and 3.3, 87%, respectively. The use of water alone or medicated with levamisole for vaccine administration produced significantly lower ND antibody titres only in the first 3 weeks. The immunogenicity shown by the I-2 vaccine as a potential vaccine is discussed in relation to free-range poultry management conditions in Uganda.  相似文献

Bivalent Newcastle disease (ND)/infectious bursal disease (IBD) and trivalent ND/IBD/infectious bronchitis (IB) inactivated oil emulsion vaccines were prepared in the laboratory and evaluated under field conditions. Broiler breeder parent chickens previously vaccinated with live vaccines were inoculated with commercial monovalent ND and experimental bivalent or trivalent oil emulsion vaccines. The commercial vaccine induced a higher initial ND haemagglutination inhibition (HI) response than the experimental vaccines but, by 34 weeks after vaccination, the mean ND HI levels were not significantly different in any of the three flocks. All three vaccines provided sufficient ND immunity to protect against the clinical disease and egg production losses. The IBD responses of both flocks vaccinated with oil emulsion vaccine were similar to each other and only slightly lower than those flocks vaccinated with monovalent IBD oil emulsion vaccine in earlier experiments. Six weeks after vaccination, sufficient immunity was transferred to protect all the progeny against IBD challenge up to 33 days of age and some of them up to 45 days of age. Thirty-four weeks after vaccination of the parents with oil emulsion vaccine, the progeny were totally immune up to 27 days of age and some of them were immune until 37 days. Application of oil emulsion vaccines in bivalent or trivalent form did not impair the responses of the chickens to the monovalent components.  相似文献

The efficacy of green-coloured (GC) I-2 Newcastle disease vaccine was determined in the present study. I-2 vaccine was mixed with a green coloured dye and stored at 4°C for 6 months while assayed for the virus infectivity at a monthly interval. Chickens were vaccinated with the GC vaccine by eye drop. Serum samples were collected from all birds before and after vaccination at weekly interval for 4 weeks and tested for haemagglutination-inhibition (HI) antibody against Newcastle disease virus (NDV). These chickens were challenged with NDV virulent strain four weeks after vaccination. The results showed that there was no difference between the infectivity titres of GC and uncoloured vaccines. However, chickens vaccinated with GC vaccine produced higher HI antibody titres than chickens vaccinated with uncoloured vaccine. Results from the challenge trial showed that all vaccinated chickens survived whereas all unvaccinated chickens died. The consequence of cryptosporidiosis on the immune response of vaccinated chickens against Newcastle disease and/or avian influenza was studied by using 240, 1 day old, male, white Hy-Line chicks and divided into 8 groups and subgroups. Each group or subgroup was consisting of 30 chicks (15?×?2 replicates). The first and second groups were kept as unvaccinated control, G1uninfected and G2 infected. G3, G4 and G5 contained 2 subgroups A&B (G3A, G3B, G4A, G4B, G5A and G5B). Chicks of subgroup A were vaccinated only while chicks of subgroup B were infected and vaccinated. These chicks were orally inoculated with 5?×?105 oocysts of Cryptosporidium baileyi (C. baileyi) at 2 days of age. Chickens were vaccinated intraocular with live Newcastle disease (ND) vaccine (Hitchner on day 7th and LaSota on day 17th of chicken life) (G3) or vaccinated by subcutaneous route with Volvac®- H5N2- AI vaccine on day 10 of chicken life (G4). Last group (G5) was infected similarly and vaccinated with ND and AI vaccines with the same day, dose and route of vaccination for each one. Random blood samples were collected for 3 weeks post-vaccination for investigation of humoral immune response against Newcastle and/or avian influenza vaccines by the haemagglutination inhibition (HI) test. The results showed that H5N2 vaccine at day 10 of chicken life is effective in chickens indicated by the geometric mean of HI titer against AI virus. The findings of this study showed that the infection with Cryptosporidia in the broiler chicken has a depressive effect on the immune status of the birds vaccinated against ND and/or AI vaccination. Moreover, the obtained protection rates against challenge with virulent ND virus observed to be parallel to the results of HI- test. Also, by using 2 different antigens (one commercial and field prepared antigen) to avian influenza virus, lower Geometric mean (GM) HI titer were appeared in infected and vaccinated group than vaccinated group only. A study of the relative lymphoid organs weight such as bursa of Fabricius from the experimental chicks indicated that those organs were comparable between the groups infected-vaccinated and vaccinated only. Non significant variations in final live weight between uninfected control and infected groups were indicated. Also, H5N2-AI vaccination at 10 days old did not affect the final live weight. ND and/or AI Vaccination could not be a substitute to application of good hygienic measures and fecal examination of the birds especially for protozoal diseases such as cryptosporidiosis. It could be concluded that cryptosporidiosis could be one cause of ND and/or AI vaccination failure in poultry farms.  相似文献

以禽病原性大肠杆菌O18、O78分离株制成超声波裂解铝佐剂灭活苗免疫14日龄鸡,以相同或不同外膜蛋白型(Outer membrane protein pattern,OMP型)的O18、O78分离株攻毒。结果表明:O78血清相同和不同OMP型分离株间能获得最大保护;O18血清型相同OMP型分离株间获得最大保护,而不同OMP型分离株间不能保护;上述两个血清型的分离株间不论OMP型是否相同,均缺乏保护。以间接ELISA试验、间接血凝试验分别测定了试验鸡临攻毒前针对大肠杆菌OMPs和脂多糖(Lipopolysaccharide,LPS)的抗体。结果表明:免疫组鸡血清上述两种抗体明显高于攻毒对照组;在免疫组,存活鸡临攻毒前血清中上述两种抗体滴度恒高于死亡鸡,但除3个组外,多数组差异不显著。攻毒对照组这一关系不稳定。结果说明:禽大肠杆菌疫苗的免疫保护,主要与O血清型有关,部分与OMP型有关,如O18分离株,免疫保护性抗原含OMPs,LPS等多个抗原表位。  相似文献
