Norway spruce has shown needle yellowing and defoliation symptoms on manysites of the Bohemian Forest in north central Austria since theearly 1980s. A forest amelioration experiment was set up toevaluate the response of spruce to magnesite based fertilizerswith different solubilities. Response variables were a crownvigor index, diameter growth, nutrient contents in needles,soil solution chemistry, and soil chemistry. Soil solution atfertilized plots showed an immediate response in base cationconcentrations, nitrate concentrations and pH-values.Fertilization increased the pool of exchangeable cations in theupper 15 cm of the soil and reduced levels of exchangeable Al. Themagnesium and calcium content of the needles of dominant trees wasgreatly improved. Fertilization has increased the mineralizationrate of soil organic matter. Diameter growth, as a measure of treevigor, showed a pronounced positive response with a time delay of5 years. Our results demonstratethat magnesite based fertilizers are a viable option to improvethe nutritional status of spruce forests on granitic bedrock.  相似文献

The lability and mobility of Zn(II)–, Cd(II)–, Pb(II)–, and Cu(II)–humic acid complexes were studied using diffusive gradients in thin films (DGT). A unique feature of this research was (1) the use of DGTs with diffusive layer thicknesses ranging from 0.4 to 2.0 mm to study lability and mobility of Zn(II)–, Cd(II)–, Pb(II)–, and Cu(II)–humic acid complexes, combined with (2) the application of a competing ligand exchange (CLE) method using Chelex 100, the same chelating resin that is used in DGT, to study the kinetic speciation. The CLE experiments were run immediately after the completion of the DGT experiments, thereby allowing effects of the competing ligand to be separated from the effects introduced by the use of the polyacrylamide gel that is used in DGT. The results indicate that Zn(II) and Cd(II) tend to form more labile and more mobile complexes with humic acid than Pb(II) or Cu(II). The dissociation rate constants of Zn(II), Cd(II), and Pb(II) were found to increase with the ionic potential of the metal, suggesting that the binding between some trace metals and humic acid has a significant covalent component. Furthermore, the results suggest that the Eigen mechanism may not be strictly obeyed for metals such as Cu(II) which have high rate constants of water exchange, k w. Consequently, the markedly slow kinetics of Cu(II)-HA species suggests that the usual equilibrium assumption may not be valid in freshwaters.  相似文献

The aim was to characterize dissolved organic matter in soils under different tree species. Molecular size distribution and chemical composition of dissolved organic carbon and nitrogen were determined in water extracts from humus layers and mineral soils taken from silver birch ( Betula pendula Roth.), Norway spruce ( Picea abies (L.) Karst.) and Scots pine ( Pinus sylvestris L.) stands. Concentrations of tannins and 15 phenolic acids in the humus layers were measured. Per unit of organic matter, the concentrations of dissolved organic C and N were larger in birch and spruce humus layers than in the pine humus layer. In the underlying mineral soil, the concentrations of dissolved organic C were similar at all sites, but the concentration of dissolved organic N was greater in spruce and pine soils than in birch soil. In all soils, the 10–100 kDa fraction was the most abundant molecular size group and hydrophobic acids the most abundant chemical group of dissolved organic C. In all humus layers, hydrophobic acids and hydrophilic bases were the major components of dissolved organic N. There were only minor differences in the concentrations of total tannins in the humus layers under different tree species. Small-molecule tannins (about < 0.5 kDa) were most abundant in the birch humus, and large-molecule tannins in the pine humus. Coniferous humus contained more ferulic and p -coumaric acids than did the birch humus. The concentrations of 3,4 and 3,5-dihydroxybenzoic acid, vanillic acid and 4-hydroxybenzoic acid were similar in all soils.  相似文献

Nitrogen cycling in two Norway spruce (Picea abies (L.) Karst.) ecosystems in the ARINUS experimental watershed areas Schluchsee and Villingen (Black Forest, SW Germany) and initial effects of a (NH4)2SO4 treatment are discussed. Although N reserves in the soils are similar and atmospheric N input is the same low to moderate level characteristic for many forested areas in SW Germany, N export by both seepage and streamwater differs considerably. At Villingen, deposited N is almost totally retained in the ecosystem, whereas at Schluchsee N export is the order of the input. This is explained by differences in forest management history. The Villingen site had been subject to excessive biomass export (e.g., litter raking) leading to unfavorable microbial transformations in the soil. In contrast, as a ‘relic’ of the former beech stand, the Schluchsee site is characterized by high biological activity in the soil with vigorous nitrification despite low pH values. Accordingly, the two ecosystems responded differently to the additional N input (150 kg NH4 + -N ha?1 as (NH4)2SO4). Nitrification starting immediately in the Schluchsee soils led to continued Al mobilization and leaching of basic cations and NO3 ?. The availability of Mg, already deficient before treatment, further decreased due to Mg leaching and marked N uptake by the stand. In contrast, most of the added N in Villingen was immobilized in the soil. Hence, uptake by the stand and leaching of NO3 ? and cations was correspondingly lower than at Schluchsee. The results emphasize the problems associated with the definition of generally applicable values for ‘critical loads’ of N deposition.  相似文献

Mycetophilids is a species-rich insect group for which the ecological requirements in temperate forests are poorly understood. This study of mycetophilids was based on trap samples from 15 oak-dominated sites in the boreonemoral zone of southern Sweden. Species richness and composition were analysed in relation to environmental variables at a local and at larger scales (multiple regression), and compared to results from similar studies in spruce-dominated sites in the boreal zone of Norway (PCA and two-sample t tests). Regressions showing a dominance of regional factors over local in-site variables agree with species-richness models assuming that local communities most often are unsaturated. Precipitation (inter-correlated with elevation) was the strongest factor for explaining the variation in species-richness, which is consistent with previous results indicating that mycetophilids are disfavoured by drought. In addition to precipitation, the area of mixed forest with high biodiversity values (woodland key habitats and protected areas) was a positive factor for species-richness, probably because such habitats combine elements of both coniferous and deciduous forests. PCA ordination revealed a clear separation of the species composition between boreal and boreonemoral forests. Species-richness in boreal forest was significantly higher than in boreonemoral forest, indicating a preference for boreal habitats in many of the species. For mycetophilids and other drought-sensitive insects, it is suggested that (partial) cutting in some dense successional oak stands should be avoided, and that some invading spruces should be tolerated.  相似文献

In April-October 1990-95 soil sciarids emerging in two Danish barley fields were recorded by means of emergence traps. In 1990-92 the fields were conventionally tilled, in the autumn of 1992 one of the fields was abandoned as annual set-aside (1993) and in 1994-95 most of the field was recropped. In the cropped and uncropped fields 480-527 ind. m−2 and 369-433 ind. m−2 emerged, respectively. Ten sciarid species were recorded, eight of which are well-known farmland inhabitants. Apparently, abandonment affected Scatopsciara atomaria negatively, whereas Corynoptera perpusilla was encouraged. The predominant species were probably bivoltine or trivoltine. In Lycoriella castanescens, Scatopsciara atomaria, Corynoptera perpusilla and Bradysia trivittata the female proportion increased significantly during the season. It is assumed that the primary sex ratio of the species is spanandrous. The seasonal change in sex ratio might arise from the unusual modes of reproduction demonstrated in Sciaridae. In Lycoriella castanescens, Bradysia trivittata and Corynoptera perpusilla a significant seasonal increase in wing length was recorded, whereas the wing length of Scatopsciara atomaria and Bradysia rufescens decreased significantly during the season. This suggests a seasonality in the production of large, potentially migrant females. In arable fields sciarid life history traits such as sex ratio, adult biometry and fecundity might be facultative.  相似文献

In this study, N-(2-aminoethyl)salicylaldimine bonded silica gel was synthesized and characterized using Fourier transform infrared and C, H, N elemental analysis. The analytical conditions such as the pH and volume of the solution, flow rates of the sample solution and the type of eluent to achieve the simultaneous preconcentration of Cu(II), Ni(II), Cd(II) and Zn(II) were optimised using the modified silica gel loaded column using a solid phase extraction technique. Samples (50?C500?ml) containing metal ions at optimal pH of 8 were passed through the column filled with the modified silica gel at 7?ml min?1 and then elution was achieved using 5?ml of 0.25?M HCl. The concentrations of metal ions in the eluates were determined using flame atomic absorption spectrometry (FAAS). The effects of matrix ions were also studied and none of the major ions interfered to the proposed method. The accuracy of the developed method was validated using a certified reference water sample (Ontario Lake water, NWTMDA-54.4). The method was successfully applied to the analysis of various natural water samples. The adsorption capacities of the modified silica gel for Cu(II), Ni(II), Cd(II) and Zn(II) ions were determined and found to be 0.332, 0.261, 0.130 and 0.375?mmol g?1, respectively.  相似文献

Citrus essential oils are widely used in the food, cosmetics, and pharmaceutical industries, so the determination of heavy metals content is of great importance to guarantee their quality. The present work deals with the quantification of Cd(II), Cu(II), Pb(II), and Zn(II) in different varieties of citrus essential oils, using derivative potentiometric stripping analysis. Two different metals extraction procedures, involving concentrated hydrochloric acid treatment and acid-alcoholic dissolution, are tested on lemon, mandarin, sweet orange, and bergamot essential oils, and they give very similar results. Cd(II), Cu(II), Pb(II), and Zn(II) recovery tests spanned from 95 to 100.50%, providing evidence that metals quantification remained unaffected by the cleanup steps of the two procedures. The repeatability of the hydrochloric acid extraction method, applied on different varieties of essential oils, is >95.00% for Cd(II), Cu(II), Pb(II), and Zn(II), whereas the repeatability of the acid-alcoholic dissolution method is >93.00% for Cu and Cd only in lemon oil. Detection limits obtained for the four analytes, using both procedures, ranged from 0.10 to 0.98 ng g(-)(1) in lemon, mandarin, sweet orange, and bergamot essential oils.  相似文献

Eggs of dippersCinclus cinclus from a chronically acidified area in Southern Norway were compared with eggs from a non-acidified area in Central Norway. There were no differences in egg size, as measured by volume, weight, length and calculated surface area, between the two areas. Eggshells were 7.0% lighter and 6.1% thinner, as measured by the Ratcliffe index and 7.0% as measured by the eggshell index (shell weight/surface area) in Southern Norway than in Central Norway. The Ratcliffe and eggshell indices were highly correlated. Scanning electron micrographs showed that the palisade layer of eggshells of eggs from the acidified area was 10.7% thinner than that of eggshells of eggs from the non-acidified area. Eggshell vapour permeability was not significantly influenced by area. Since the moderately lower thickness in Southern Norway was not accompanied by higher vapour permeability, this indicates that the reduced eggshell thickness did not cause desiccation of dipper eggs in the acidified area. The possibility of underestimating the environmental effects of acidification on dippers is discussed.  相似文献

过氧化物酶体增殖物激活受体(peroxisome proliferators-activated receptors,PPARs)是核激素受体超家族的一员,分α、β/δ、γ3个亚型,是调节细胞生长和分化的重要因子.为了解PPARs各成员在鸭(Anas platyrhynchos)骨骼肌发育中的不同作用,本研究克隆了鸭PPARα、PPARβ和PPARγ基因启动子区,预测其共同转录因子结合位点,用qRT-PCR检测PPARs及共同转录因子在鸭胚及出生后骨骼肌发育过程中的表达,比较表达模式并进行聚类分析.结果表明,扩增出鸭PPARα、PPAEβ和PPARγ启动子序列分别为2 526、1 631和2 942 bp,GenBank登录号分别为KX845431、KX845432和KX845433.鸭PPARs启动子区存在典型的CAAT-box、TATA-box顺式作用元件,预测存在共同的特异性蛋白1(specificity protein 1,Sp1)、核因子κB(nuclear factor kappa B,NF-κB)和CCAAT增强子结合蛋白(CCAAT/enhancer binding protein alpha,C/EBP-α)转录因子结合位点.PPARs成员在鸭胸肌、腿肌组织中均有表达.在胸肌中,转录因子NF-κB与PPARβ基因的表达模式一致;在腿肌中,Sp1、NF-κB、PPARβ和PPARγ的表达模式一致.PPARs各成员均参与了骨骼肌的发育调控,PPARβ作用可能更大;PPAPβ、PPARγ在鸭骨骼肌发育过程中的功能可能相似;此外,NF-κB可能调控PPARβ在骨骼肌中的表达.研究结果为PPARs基因的表达和功能鉴定提供理论依据.  相似文献

Ethylendiamintetraacetic acid (EDTA) is persistent in the environment. The presence of EDTA in soil may alter the mobility and transport of Zn, Cd and Ni in soils because of the formation of water soluble chelates, thus increasing the potential for metal pollution of natural waters. Mobility of metals is related to their extractability. To investigate metal extractability affected by EDTA, Zn, Cd and Ni were added to Vertisol and Alluvial soil at rates of 50, 2 and 5 mg kg-1, respectively. Both natural and metal amended soils were treated with Na2EDTA at rates of 0; 0.2 and 0.5 mg kg-1. After five months of incubation soil samples were extracted with 0.1 N HCl, 0.005 M DTPA + 0.01 M CaCl2 + 0.1 M TEA (0.005 M Diethylenetriaminepentaacetic acid + 0.01 M Calcium cloride + 0.1 M Triethanolamine) and 1 M Mg(NO3)2, the latter of which extracts the exchangeable from of metald (Zn, Cd and Ni).

According to experiment results, Zn, Cd and Ni in all extraction increased with increasing rates of EDTA in the natural and metal amended soils.  相似文献

The labeling of products containing genetically modified organisms (GMO) is linked to their quantification since a threshold for the presence of fortuitous GMOs in food has been established. This threshold is calculated from a combination of two absolute quantification values: one for the specific GMO target and the second for an endogenous reference gene specific to the taxon. Thus, the development of reliable methods to quantify GMOs using endogenous reference genes in complex matrixes such as food and feed is needed. Plant identification can be difficult in the case of closely related taxa, which moreover are subject to introgression events. Based on the homology of beta-fructosidase sequences obtained from public databases, two couples of consensus primers were designed for the detection, quantification, and differentiation of four Solanaceae: potato (Solanum tuberosum), tomato (Solanum lycopersicum), pepper (Capsicum annuum), and eggplant (Solanum melongena). Sequence variability was studied first using lines and cultivars (intraspecies sequence variability), then using taxa involved in gene introgressions, and finally, using taxonomically close taxa (interspecies sequence variability). This study allowed us to design four highly specific TaqMan-MGB probes. A duplex real time PCR assay was developed for simultaneous quantification of tomato and potato. For eggplant and pepper, only simplex real time PCR tests were developed. The results demonstrated the high specificity and sensitivity of the assays. We therefore conclude that beta-fructosidase can be used as an endogenous reference gene for GMO analysis.  相似文献

A method of DNA analysis has been developed to verify the authenticity of grouper (Epinephelus guaza), wreck fish (Polyprion americanus), and Nile perch (Lates niloticus) fillets. A short fragment (208 bp) of the mitochondrial 12S rRNA gene was amplified by the polymerase chain reaction and analyzed by single-strand conformation polymorphism to get species-specific patterns of single-stranded DNA (ssDNA). DNA strands were separated by native polyacrylamide gel electrophoresis and visualized by silver staining. Discrimination among the three fish species studied was possible, because each one expressed a specific ssDNA pattern.  相似文献

Hessian fly, Mayetiola destructor (Say), is the major pest of wheat in North Africa. In Morocco, durum wheat (Triticum turgidum L. subsp. durum (Desf). Husn.) losses due to this pest have been estimated at 32%. Genetic resistance is the only economical and practical means of controlling this insect. Field and greenhouse screening of durum wheat genotypes resulted in the identification of one source of resistance to Hessian fly in Morocco. This is the first source of durum wheat Hessian fly-resistance identified in Morocco. This source of resistance expresses a medium level of antibiosis against first-instar Hessian fly larvae; about 25% of the larvae survive on resistant plants. The deployment of varieties that allow for larval survival on resistant plants should reduce selection for biotype development. This source of resistance is being used by CIMMYT/ICARDA and Moroccan breeders to develop resistant durum wheat varieties.  相似文献

Chlorophenols are potentially harmful pollutants that are found in numerous natural and agricultural systems. Plants are a sink for xenobiotics, which occur either intentionally or not, as they are unable to eliminate them although they generally metabolize them into less toxic compounds. The metabolic fate of [ (14)C] 4-chlorophenol (4-CP), [ (14)C] 2,4-dichlorophenol (2,4-DCP), and [ (14)C] 2,4,5-trichlorophenol (2,4,5-TCP) was investigated in lettuce, spinach, and radish to locate putative toxic metabolites that could become bioavailable to food chains. Radish plants were grown on sand for four weeks before roots were dipped in a solution of radiolabeled chlorophenol. The leaves of six-week old lettuce and spinach were treated. Three weeks after treatments, metabolites from edible plant parts were extracted and analyzed by high performance liquid chromatography (HPLC) and characterized by mass spectrometry (MS), and nuclear magnetic resonance spectroscopy (NMR). Characterization of compounds highlighted the presence of complex glycosides. Upon hydrolysis in the digestive tract of animals or humans, these conjugates could return to the toxic parent compound, and this should be kept in mind for registration studies.  相似文献

Atlantic salmon are severely affected by acidification in Norway. Water quality criteria for the salmon have to be based on the most sensitive stage, the smolt stage. The sensitivity to acidic water increases enormously during smolting, the seawater tolerance being especially vulnerable. Even moderately acidic water (pH about 6) with low inorganic monomeric aluminium (LA1) concentrations (<20μg. L?1) and short-term episodes may be harmful. Mixing zones in limed or unlimed rivers may also represent a problem for seaward migrating smolts. In limed salmon rivers, the national liming goal has been increased to pH 6.5 during smolting (1 February to 1 July) and to 6.2 the rest of the year as a result of our experiments. In contrast to what has been found for brown trout, salmon strains originating from watercources undergoing acidification were not more tolerant than those from non-acidic watercourses. At the moment no such “tolerant” strains are available for restocking limed rivers in Norway.  相似文献

A separation and preconcentration procedure was developed for the determination of trace amounts of Cd(II), Cu(II), Ni(II), and Pb(II) in water and food samples using Amberlite XAD-2 fuctionalized with a new chelating ligand, 3-(2-nitrophenyl)-1H-1,2,4-triazole-5(4H)-thione (Amberlite XAD-2-NPTT). The chelating resin was characterized by Fourier transform infrared spectroscopy (FT-IR) and used as a solid sorbent for enrichment of analytes from samples. The sorbed elements were subsequently eluted with 10 mL of 1.0 M HNO(3), and the eluates were analyzed by inductively coupled plasma-atomic emission spectrometry. The influences of the analytical parameters including pH, amount of adsorbent, eluent type and volume, flow rate of the sample solution, volume of the sample solution, and effect of matrix on the preconcentration of metal ions have been studied. The optimum pH for the sorption of four metal ions was about 6.0. The limits of detection were found to be 0.22, 0.18, 0.20, and 0.16 μg L(-1) for Cd(II), Cu(II), Ni(II), and Pb(II), respectively, with a preconcentration factor 60. The proposed method was applied successfully for the determination of metal ions in water and food samples.  相似文献
