Antibodies to Leptospira interrogans serovars hardjo and pomona were present in 8.3% and 0.5% of sera respectively, from adult female cattle in Alberta surveyed in 1984-85. Criterion for a positive serum sample was 50% agglutination at 1/100 dilution in the microscopic agglutination test. A positive herd contained one or more cows with positive serum. Prevalences were calculated on sample sizes that would give 80-95% reliability. Hardjo antibody prevalences and hardjo-positive herd prevalences were 0-53.9% and 0-83.3%, respectively, among 65 municipalities surveyed. Pomona prevalences by comparison were 0-3.4% and 0-11.7% respectively. Hardjo had increased significantly since 1980-82, and antibodies were found throughout the province. Pomona occurred mainly in southeastern Alberta, where it was isolated from cattle, swine and skunks. Hardjo was isolated only from cattle and it was found in many areas. Antibodies to icterohaemorrhagiae were present in 0.4% of sera from parts of Alberta surveyed in 1980; evidence of the presence of leptospires related to this serovar in bovine and porcine urinary tracts was obtained by immunofluorescence.  相似文献

Abortions occurred in 18% of 131 beef cows and heifers during two months, on a farm in southern Saskatchewan. The losses began two weeks after acute febrile illness and agalactia in a dairy cow to which the beef herd had been exposed. A diagnosis of Leptospira interrogans serovar pomona infection was made on the basis of serology in cows and the finding of leptospires in fetal tissues by fluorescent antibody test. Tentative diagnosis of infectious bovine rhinotracheitis delayed treatment and prophylaxis until infection attained high intensity in the herd and severe losses to the farmer occurred. Abortions ceased after vaccination against pomona and oxytetracycline treatment of pregnant cows, although chronic debility followed the acute phase of the disease in some cows. Recrudescence of infection was suspected four months later, when acute agalactia occurred in one cow and debility in calves and cows was recurring. Pomona infection was not proven, but dihydrostreptomycin treatment and revaccination were applied to the whole herd. Seroconversion and IgM antibody continued to indicate a persistent source of infection and susceptibility in a minority of the population one year after onset. The source of the original infection is believed to have been a carrier beef cow, or a dairy cow which was leptospiruric at the time of contact with the beef herd. With the exception of one aborted calf, no evidence of pomona infection was found outside the farm, in cattle or wild mammals tested serologically within a radius of 30 km, during one year following the outbreak.  相似文献

The results of laboratory examination of 20460 specimens for the diagnosis of rabies by the standard fluorescent antibody and mouse inoculation tests over a five year period are presented. Specimens were received from British Columbia, Alberta, Saskatchewan, Manitoba, and the Yukon and Northwest Territories. Of those examined, 9.96% were positive. The main reservoirs of rabies were skunks, bats, and foxes. During this observation period a rabies epizootic occurred in skunks in Saskatchewan. When both tests were performed, the fluorescent antibody test was found to agree with the mouse inoculation test in over 99% of cases.  相似文献

A survey of 930 ovine sera and kidneys from 33 sheep was conducted to assess the rate of leptospiral infection in sheep slaughtered in Alberta. Sera were tested for the presence of agglutinins to indigenous serovars of Leptospira interrogans. Kidneys with gross lesions were examined for the presence of leptospires by means of an indirect fluorescent antibody test (FAT) and by culture. Antibodies to serovars pomona and hardjo were present at rates of 1.0% and 0.4%, respectively, in sheep from Saskatchewan, Alberta and British Columbia. Sera from 120 feedlot lambs shipped from Oregon reacted to serovars pomona, hardjo and grippotyphosa at rates of 1.7%, 61.7% and 59.1%, respectively. Fluorescent antibody test detected serovars (presumptively) hardjo in 52% of Oregon feedlot lambs and grippotyphosa in 32% of the same group, a finding supported by the isolation of both these serovars from a pool of two fluorescent antibody test-positive kidneys. The grippotyphosa strain was highly virulent for hamsters, producing intense icterus and death. Leptospires, presumptively serovar grippotyphosa were demonstrated by fluorescent antibody test in one Alberta lamb kidney. The possibility of spreading leptospirosis by movement of breeding stock through public facilities and by assembling lambs in feedlots is discussed.  相似文献

The prevalence of reactors to different serovars of Leptospira interrogans was determined in 117 swine premises in Quebec. A total of 926 sera were tested, using the microscopic agglutination method, against six leptospiral serovars: pomona, icterohaemorrhagiae, grippotyphosa, hardjo, canicola and ballum. Of the sera tested, 30 (3.2%) reacted to one of the three following serovars: pomona (2%), icterohaemorrhagiae (1.1%) and ballum (0.1%).  相似文献

A macroscopic plate test was found to be reliable and convenient for detection of Leptospira serotype pomona antibodies in bovine sera. However, the procedure was unreliable for detecting L. serotype canicola antibodies because of false positive reactions. An indirect immunofluorescence test and the microscopic agglutination test provided comparable results and they effectively detected serotype pomona antibodies in bovine sera.  相似文献

Fourteen abortions, stillbirths and neonatal deaths occurred over an interval of one month in crated sows in a farrow-to-finish swine operation. Leptospira interrogans serovar pomona (kennewicki by DNA analysis) was demonstrated by culture and fluorescent antibody test to be present in sows, piglets, boars, feeders, drinking water and skunks on the premises.

Antibody was found in all clinically affected sows, at serum dilutions ranging from 1/800 to 1/25,600, and in all breeding boars at titers from 1/50 to 1/1600. Pomona antibody was present in 118 sow sera collected nine months before the outbreak, at a prevalence of 21.3%. Parvovirus infection in fetuses was intercurrent with the leptospirosis epizootic, despite vaccination for the former. Environmental contamination, feedback through skunks via drinking water, and dissemination through the piggery are discussed.

A total of 123 rabies virus isolates from various geographical areas in Canada were characterized by a panel of 43 anti-nucleocapsid monoclonal antibodies. Four major antigenic groups are found in terrestrial mammals: "Canadian Arctic" from Ontario, Quebec and the Northwest Territories; "south-eastern Georgian Bay" from Ontario; "south mid-central skunk" from Alberta, Saskatchewan and Manitoba; and "Brook's, Alberta skunk" from a restricted area in Alberta. Bat isolates can be divided into 4 major antigenic groups: "B-1" in Eptesicus fuscus from Ontario; "B-2" in a variety of bat species from British Columbia eastward into Ontario; "B-3" in Myotis spp. from Ontario and New Brunswick; and "B-4" in E. fuscus from Alberta and Saskatchewan. A single case of bat to horse transmission of rabies virus is recorded. These street isolates are compared with isolates of fixed virus. Epidemiological aspects are discussed.  相似文献

Leptospirosis occurs enzootically over most of Southern Ontario. Leptospira pomona is the serotype most commonly found in outbreaks. Antibodies to L. pomona occur frequently in the sera of deer in wilderness areas. The geographic location of leptospirosis presents a pattern which closely parallels the distribution of Paleozoic bedrock. By contrast, L. pomona infection is absent from areas underlain by Precambrian bedrock. Comparisons of water chemistry, soil type, habitat, and host and pathogen availability in these two geologically distinct environments have not defined the mechanisms involved in the disease pattern. Leptospires resembling saphophytic strains occur widely, regardless of bedrock type. High titers to L. biflexa, a saprophytic serotype, were found frequently in deer sera from a Precambrian area which was surveyed intensively. Antibodies to L. hardjo and L. sejroe occur in many bovine sera from a predominantly Precambrian area where Paleozoic outliers are numerous. Colloidal clay is common to leptospiral habitats. A microenvironment structured by the surface activity of clay is likely to be a key ecological factor in the landscape epizootiology of leptospirosis. In Ontario, bedrock composed of limestone and dolomite formed in the Paleozoic era appears to be a reliable ecological marker for Leptospira pomona infection.  相似文献

An eight-year-old city-dwelling Cairn Terrier was presented to a veterinary hospital in acute renal failure with evidence of hepatic insufficiency. The dog was treated symptomatically over three days, during which time vomiting was largely controlled, but it became jaundiced as hepatic insufficiency worsened. Leptospira pomona was demonstrated in large numbers by immunofluorescent staining of urinary sediment. It was isolated and its identity confirmed as L. pomona genotype kennewicki. The source of the infection was thought to be raccoons.

Sera from 474 blood samples submitted for diagnostic purposes to two clinical pathology laboratories in southern Ontario were examined with the microscopic agglutination test for antibodies to selected leptospiral serovars. Of the sera tested, 39.2% reacted at titers ≥1:100 with one or more serovars, the majority of all sera (26.2%) reacting at low titers to canicola or icterohaemorrhagiae, or both. These reactions likely resulted from vaccination. A smaller proportion reacted to other serovars tested: autumnalis (3.8%), bratislava (8.2%), grippotyphosa (1.9%), hardjo (3.0%), and pomona (3.2%). Among dogs reacting to these latter serovars (other than bratislava), many had broadly cross-reacting and relatively high titers. One dog with a titer of 1:800 to pomona had had a disease typical of leptospirosis two years previously. Three other dogs with high titers to autumnalis, bratislava, or mixed serovars had clinical histories compatible with leptospirosis.

We suggest that leptospiral bacterins for dogs in Ontario be broadened to include at least serovars autumnalis and pomona.

A serological survey of 213 randomly chosen dairy farm residents in the Manawatu showed that 66 (34%) of the people who milked cows had leptospiral titres ≤ 1:24 by the Microscopic Agglutination Test. Forty-eight (72.7%) of these people had titres to hardjo, while 29 (43.9%) had titres to pomona. Dual hardjo/pomona titres occurred in 12 people. Ballum and Copenhageni accounted for 8% of the titres found. Women milkers and farm residents who did not milk were all serologically negative. A third of the seropositive milkers had a history of clinical leptospirosis. Other factors which significantly correlated with leptospiral titres included the time spent in the dairy shed during milking, the wearing of shorts, the keeping of pigs for sale, and the number of years the individual had been working on a dairy farm. The type of milking shed and the size of the herd were interrelated and both showed strong trends towards a correlation with serological prevalence.  相似文献

A total of 1745 healthy cattle from 295 farms in Saskatchewan and Alberta was tested by ELISA for antibodies to four viruses. Antibodies to infectious bovine rhinotracheitis (IBR) virus were found in 37.8% of sera (59.5% of properties), to parainfluenza 3 (PI3) virus in 93.9% of sera (99.7% of properties), to bovine respiratory syncytial (BRS) virus in 78.5% of sera (86.6% of properties), and to bovine viral diarrhea (BVD) virus in 40.6% of sera (66.7% of properties)

The prevalence of PI3 viral antibodies among Saskatchewan cattle was not affected by district of origin, breed, sex, age, or vaccination practices, though BRS viral antibodies appeared less frequent in young, male, and unvaccinated animals. Antibodies to IBR and BVD viruses were less prevalent in the Prince Albert/Tisdale districts and in young, male, and unvaccinated animals, but were more common in Holstein cattle. Antibodies to IBR virus appeared less frequent in Herefords. Antibodies were more prevalent in cattle which had been vaccinated against IBR, BRS, and BVD virus infections.

The relatively small number of cattle sampled from Alberta had a similar prevalence of antibodies to PI3 and BRS viruses to that seen in cattle in Saskatchewan, though IBR and BVD prevalence rates were lower.

The results of laboratory examination of 22,503 specimens for the diagnosis of rabies by the standard fluorescent antibody and mouse inoculation tests over a seven year period are presented. Specimens were received from British Columbia, Alberta, Saskatchewan, the Yukon and Northwest Territories. Of the 1,445 positive cases, 10.50% involved human contact. The main reservoirs of rabies were skunks, bats and foxes. The reliability of the fluorescent antibody test for the diagnosis of rabies was reaffirmed by agreement with the mouse inoculation test in over 99% of cases.  相似文献

The results of combined epidemiological, clinical, serological, bacteriological and histopathological studies following an outbreak of disease caused by L. pomona on a farm stocked with cattle, sheep, pigs, goats and horses maintained for experimental purposes, are reported.

The incidence of infection was high in horses, cattle and pigs. A few low titres were seen in sheep. The goats were not infected. Apart from a single bovine abortion all the clinical symptoms observed occurred in pregnant sows. Seven of these aborted or gave birth to stillborn pigs within a six week period.

Fifteen species of wildlife were trapped or shot on the farm during the year following the outbreak. L. pomona was isolated from four skunks and a porcupine. Epidemiological studies indicated that wildlife reservoir hosts were the primary source of infection for the domestic livestock.

Leptospiruria and the serological response were studied in a group of eight infected sows. Microscopic agglutination titres of 102 or less could not be associated with leptospiruria and the duration of leptospiruria was found to range from a few weeks to over two years in individual sows. Direct dark-field examination of urine proved superior to guinea-pig inoculation as a method of detecting leptospiruria and it is suggested that the former technique could be adopted with advantage as a routine aid to diagnosis.

This study investigated the disease status of Saskatchewan’s feral wild boar population. Whole carcasses, tissue samples, and/or serum from 81 hunter-killed boars from Saskatchewan were submitted to the Canadian Wildlife Health Cooperative (CWHC) between 2009 and 2014. Serological tests were negative for PRRS, H1N1, and H3N2 swine influenza, PCV-2, and TGE/PRCV in 22/22 boars and for Toxoplasma gondii and Mycoplasma hyopneumoniae in 20/20 boars. Of 20 boars whose sera were tested 20 were positive for Actinobacillus pleuropneumoniae, with 7 positive for, among other strains, serotype 14; 16 were positive for Lawsonia intracellularis, 1 was positive and 6 were suspicious for Salmonella spp. Polymerase chain reaction tests were negative for PRRS and PCV2 in 58/58 boars and positive for Torque teno virus in 1/8 boars. Digestion assays were negative for Trichinella spp. in 22/22 boars. The high seroprevalence of A. pleuropneumoniae serotype 14 is noteworthy as this serotype has not been previously reported in North America.  相似文献

Subsequent to a cross-sectional serological survey of Manawatu dairy farm workers, a case-control study was carried out to investigate the correlation between titres to leptospiral serovars in workers and those in cattle in their herds. A total of 52 herds was investigated, 25 of which were ‘high risk’ where milkers had titres of 1:96 or greater, and 27 were case-controls where milkers had no detectable agglutinin titres at a minimum serum dilution of 1:24.

The serological prevalence of titres to hardjo in cattle on ‘high risk’ farms (76.5%) was significantly higher (P<0.05) than on the case-control farms (60.0%). The geometric mean titres of seropositive cattle on ‘high risk’ farms were also significantly higher (P<0.0l) than in the cattle from the case-control farms, especially in the younger cohorts. These findings suggest that there was active endemic hardjo infection in the two- to three-year-old cattle on the ‘high risk’ farms.

Titres to pomona were demonstrated in only 5.2% of the cattle from both types of farm. Workers with titres to pomona tended to be from farms on which stock, especially calves, were bought-in and pigs were kept.

Conventional measures for protecting milkers from contact with infected urine appeared to be ineffective and it is concluded that prevention of leptospirosis in dairy farm workers can only be achieved by elimination of infection in the herd by vaccination of cattle.  相似文献

Capsular polysaccharide antigens of serotypes 1, 2, 5 and 7 of Actinobacillus pleuropneumoniae were used in enzyme-linked immunosorbent assays (ELISAs) to test sera from experimentally infected and field pigs. Specific reactions were found in sera of experimental pigs with antigens of serotypes 1, 5 and 7 whereas the serotype 2 antigen was cross-reactive. A 1:200 serum dilution was used for testing of 300 sera from 21 swine herds in southern Ontario. Cases of pleuropneumonia had occurred in 11 of these herds, but not in the others. The negative cut-off value was the mean optical density at 405 nm (OD405) + three standard deviations (SD) for 16 negative reference sera. Sera from four pigs naturally infected with Actinobacillus suis were tested and found to react to varying degrees with each of the antigens. Therefore a second cut-off value was determined as the mean OD405 + 2 SD for the A. suis sera. Sera which, in the ELISA produced OD readings above the latter cut-off were considered positive for antibodies to A. pleuropneumoniae; those which were lower than the former cut-off were considered negative. Readings between the two cut-off values may have been due to low positive titers or cross-reactivity, possibly with A. suis, and could not be used to predict pleuropneumonia. Of the pleuropneumonia-free herds, none had positive reactors to serotypes 5 or 7, whereas one and two herds had positive reactors to serotypes 1 and 2, respectively. Of the pleuropneumonia positive herds, six had positive reactors to serotype 1, one to serotype 2, four to serotype 5, and eight to serotype 7.  相似文献

The results of laboratory examination of 18,086 specimens for the presence of rabies antigen by the fluorescent antibody and mouse inoculation tests over a ten year period are presented. The submissions were received from British Columbia, Alberta, Saskatchewan, the Yukon and the Northwest Territories. Of those examined, 10.74% were positive: however, the incidence of rabies varied widely in the specimens and species submitted, depending on their origin. The principal wildlife reservoirs of the disease appear to be skunks, foxes and bats. A correlation of almost 99% was obtained between the fluorescent antibody test and the mouse inoculation test, indicating that the diagnostic procedures used were highly reliable in identifying rabies-infected animals.  相似文献

Investigations were carried out in 1975, 1976 and 1977 in 16 dairy herds where leptospiral abortions were suspected and in five other herds where clinical disease was not present. Both Leptospira interrogans serovars pomona and hardjo were isolated from cattle in herds with leptospirosis, but only pomona was recovered from those that had aborted. There was no evidence that hardjo caused clinical disease in dairy cattle in the Waikato district.

It was found that 73% of the cows that aborted and 19% of other animals in the same herds had microscopic agglutination test titres to pomona of 1:2 000 or greater. By contrast, only 2% of cattle in herds without clinical evidence of leptospirosis had such titres. One cow retained a titre of 1:2 000 or greater to pomona for 7 months; titres of this order had a shorter duration in other cows. Leptospiruria occurred in 50% of cows that had aborted and in 9% of in-contact cows in the same herds. Only 0.7% of cows had leptospiruria in the herds with no clinical disease. Ten of 35 cows shedding pomona still had leptospiruria one month later.

It was concluded that clinical leptospirosis should be diagnosed by testing a sample of the herd, rather than just individual cows, because of the variability and persistence of leptospiruria and serological titres in cows with and without clinical signs. Although hardjo is common in cattle in the Waikato district, it was not found to cause abortion in cattle.  相似文献

Controlling rabies in skunk populations is an important public health concern in many parts of the United States due to the potential for skunk rabies outbreaks in urban centres and the possible role for skunks in raccoon rabies variant circulation. Oral rabies vaccination (ORV) programmes have supported wildlife rabies control efforts globally but using ORV to control rabies in skunk populations has proven more challenging than with other target species, like foxes, coyotes and raccoons. A review of published studies found that some ORV constructs are immunogenic in skunks and protect against virulent rabies virus challenges, especially when delivered by direct installation into the oral cavity. However, in field ORV programmes using currently available vaccine‐bait formats and distribution methods targeting other rabies reservoir species, skunks often fail to seroconvert. Field effectiveness of ORV in skunks appears to be limited by poor bait uptake or inadequate ingestion of vaccine rather than from poor vaccine efficacy. Observations of captive skunks revealed vaccine spillage when handling and biting into baits such that modification of bait formats might improve field effectiveness. In addition, a dose–response relationship between bait distribution density and post‐baiting seroconversion among skunks was observed across the limited number of field studies. Additional research is needed to identify opportunities to modify ORV baits and distribution strategies to improve the viability of ORV as a rabies control strategy in skunks.  相似文献
