Vegetative propagation techniques are recognized as indispensable tools for mass multiplication of important multipurpose trees adopted in different agroforestry systems. Albizia procera, one among important species, is difficult to propagate commercially either by stem / root cuttings or layering. A study was undertaken to develop procedure for its in vitro regeneration through organogenesis. Explants collected from 15±2 yr-old mature plus trees and from 15 days old juvenile seedlings were regenerated with exogenous application of different hormones. Epicotyl and hypocotyl explants excised from juvenile seedlings showed higher callusing than axillary bud and shoot tip explants derived from mature trees. Benzylaminopurine (BA) at 3 μg/l was most effective, which induced hundred percent callusing in epicotyl and hypocotyl explants in 1/2 Murashige and Skoog (MS) medium. Callus originated from axillary buds and apical shoot tips of mature trees failed to form organs, however callus derived from epicotyl and hypocotyl explants proliferated and formed de novo shoots and leaflets. A concentration of 3 μg/l of BA was found effective for shoot proliferation. Shoots grew vigorously in 2 μg/l gibberellic acid (GA3) treatment and rooted in 1/2 MS medium supplemented with indole-3-butyric acid (IBA) and indole-3-acetic acid (IAA). Rooting was most successful on medium supplemented with 6 μg/l IBA alone on which 93.3% of the shoots formed roots. Sand or vermiculite supplemented with 4 ml of yoshida solution proved as best hardening media, which recorded 70-80% survival of plantlets. One year old tissue culture raised plants had comparatively more height, collar diameter, biomass, and root shoot ratio than plants raised from cuttings and seeds of the same age. The procedures enumerated provide a basis for the development of in vitro techniques for rapid multiplication of A. procera. Shoot anatomical features viz., number of vessel elements, width and length of vessel and fibre elements were studied in relation to juvenility/maturity of teak (Tectona grandis Linn. f.). The findings show significant differences in these features, which indicates that variation in shoot anatomy can be used as a reliable indicator of juvenility/maturity in teak. The effect of age of donor plants and application of auxins on adventitious root formation in single noded leafy stem cuttings of teak was also investigated. Aging of donor trees suppressed rooting and sprouting of cuttings, but increased callus formation at the base of cuttings. Per cent rooting and per cent sprouting in cuttings decreased as age of donor plants increased from 2-months to 15-years and up to 30-years. IBA treatment promoted per cent rooting as well as per cent sprouting, NAA promoted only rooting and it suppressed sprouting. The effect of auxin treatment on rooting and sprouting of teak stem cuttings varied with the age of donor plants. Auxin and its concentration, which suppressed rooting and sprouting in cuttings of younger donor plants, actually promoted rooting when cuttings were taken from more mature donor plants. It appears that the auxin requirement for causing and promoting rooting in teak cuttings increased with increasing age of donor plants. The higher auxin requirement for causing and promoting rooting in cuttings of older trees may be due to a decrease in the content on endogenous auxins or decreased sensitivity of aging tissues to rooting promoters and /or accumulation of inhibitory substances which inhibit rooting.  相似文献

We developed procedures for in vitro cloning of Cedrus atlantica Manetti and C. libani A. Rich explants from juvenile and mature plants. Explant size was one determinant of the frequency of axillary bud break in both species. Shoot tips and nodal explants mainly developed calli, whereas bud sprouting occurred in defoliated microcuttings cultured on a modified Murashige and Skoog medium without growth regulators. Isolation and continuous subculture of sprouted buds on the same medium allowed cloning of microcuttings from C. atlantica and C. libani seedlings and bicentennial C. libani trees, thus providing a desirable alternative for multiplying mature trees that have demonstrated superior characteristics. We also report adventitious bud differentiation from isolated embryos of C. atlantica. Neither auxin treatments nor other methods tested, including infection with Agrobacterium rhizogenes, were effective in inducing root initiation.  相似文献

Variations in tree ring growth of Quercus suber L. were analysed using dendrochronological techniques on cork oak discs from trees harvested in the cork producing region of Alentejo, Portugal. A tree-ring chronology containing a strong common signal and covering the period from 1970 to 1995 was build for ca. 30-year-old cork oaks never submitted to cork harvesting using 14 trees that crossdated satisfactorily out of 30 sampled trees. The tree ring indices correlated positively with September temperature (r = 0.48, P < 0.05) and very strongly with precipitation totals from previous October until current February (r = 0.82, P < 0.001) showing that the water stored in the soil during the autumn and winter months prior to the growing season has a primordial effect on the growth of the given season. The effects of cork harvesting were analysed by comparing mean ring width, mean annual vessel area, vessel density (n°vessels/mm2), and vessel coverage (percentage of transverse surface occupied by vessels) between three mature cork oak trees and three young trees, for the period from 1987 to 1996, corresponding to the growth between two consecutive cork removals in the case of mature trees. In 1988, 1989 and 1996 (corresponding to the first and second years after cork removal, and 1996 to a year of cork removal), the ratios between ring widths of young versus mature trees was twice that for the rest of the period. However, an effect of cork removal indicated by eventual alterations in vessel size and distribution in the wood rings corresponding to the years 1988, 1989 and 1996 in the mature cork oaks was not observed.  相似文献

Juvenile (2 year old trees) and mature hardwood (15 year old trees) cuttings of Robinia pseudoacacia and Grewia optiva were tested for their capacity to form roots. Cuttings were prepared in spring, monsoon and winter seasons and treated with different concentrations (250, 500 and 750 mg/l) of IBA and NAA. These were planted in a mist chamber maintained at 25±1°C with relative humidity >70%. Juvenile cuttings of both species rooted significantly better than mature hardwood cuttings in all three seasons, and the age effect was more pronounced in auxin treated cuttings. The highest rooting in juvenile (83.3%) and mature (66.6%) cuttings was observed with the NAA (500 mg/l) treatment in R. pseudoacacia during the spring season. In G. optiva, IBA (250 mg/l) in the monsoon season was most effective and yielding a maximum of 80% and 70% rooting in juvenile and mature cuttings, respectively. Auxin treatments also significantly enhanced the number of roots, root length, leaf number and leaf area. Statistical analysis of data revealed that interactions between age, season and treatments were significant at P <0.05 level for R. pseudoacacia and non-significant for G. optiva.  相似文献

Black locust (Robinia pseudoacacia L.) is one of the most important stand-forming tree species in Hungary and its importance is increasing in many countries. Black locust plants are commonly produced by two methods, by seed and by root cuttings. Tissue culture propagation can be considered as a relatively new method. Growing trees from seed is a relatively simple method for reliably producing seedlings on a large scale under a variety of circumstances. Mechanization of the method is easy and the production cost is relatively low. Propagation from root cuttings and tissue culture are valuable for reproduction of superior individuals or varieties. By applying these methods, superior traits of the selected trees can be preserved in the clones. Recent experiments demonstrated that micropropagated trees could be successfully transplanted into soil, hardened and grown in the field. Ficus species are multipurpose trees well known by rural populations in Sahelian and Sudanian zones of Africa. Their uses are well documented but their amenability for vegetative propagation has not been extensively studied. This paper compares the rooting ability of stem and aerial root cuttings from thirteen Ficus species found in West Africa. It highlights the differences between species belonging to the sub-genera Sycomorus and Urostigma. The former show no capacity to propagate from cuttings whereas the latter, with epiphytic development, can be propagated by cuttings, although this capacity varies among species. Thus, F. thonningii, F. leprieurii and F. ovata are easily propagated, while F. platyphylla and F. elasticoides are propagated with difficulty. The rooting capacity also varies depending on the cutting material used. It decreases in the following order: long leafless hardwood cuttings (pole) > nodal cuttings > apical cuttings. Rooting potential increases when the cuttings are harvested towards the end of the dry season(March to May). Aerial root can be used for cuttings in all species of the sub-genus Urostigma. The capacity of root cuttings to regenerate is greatest when cuttings are collected at the beginning of the dry season (November). In this case, wound-induced adventitious roots arise at the basal end of the cutting while de novo buds are developed from the cambium at the distal end. The subsequent morphological development is identical to that of a stem cutting. These results clarify and allow the optimal use of the knowledge and methods developed by the indigenous people of the Sahel and could assist and promote fig tree (Ficus sp.) domestication in the dry tropics.This revised version was published online in November 2005 with corrections to the Cover Date.  相似文献

We examined the effect of zeatin on the formation of shoot buds from explants and callus tissues derived from stem segments of Actinidia polygama Miq. (matatabi or silver vine). Stem and petiole segments cultured on combined broad-leaved tree medium and woody plant medium (BW medium) supplemented with zeatin for 40 days formed many shoot buds. Callus tissues derived from the stem segments and subcultured on BW medium supplemented with 6-benzyladenine and 1-naphthaleneacetic acid formed shoot buds when the medium contained 13.7µM zeatin. BW medium containing low concentrations of sucrose strongly induced the formation of shoot buds from the callus.  相似文献

In vitro clonal multiplication of Pterocarpus santalinus L. was achieved using mature nodal explants of a 10-year-old elite quality tree. Combinations of serial transfer technique and incorporation of antioxidants (250 mg/l L-ascorbic acid and 50 mg/l citric acid) into the culture medium helped to minimize medium browning and improve explant survival during shoot sprouting. About 70% of explants were sprouted on Murashige and Skoog (MS) liquid medium containing 4.4 μM 6-benzyladenine (BA). The explant harvest period also influenced the bud break and shoot sprouting in nodal explants. The combination of 4.4 μM BA and 2.2 μM thidiazuron (TDZ) was found to be the most suitable growth regulator for obtaining the highest percentage of nodal segment sprouting (74%–75%), the number of secondary shoots per primary shoot (two or three), the shoot length (5–6 cm), the number of new nodal segments generated per active explant (four or five), and the multiplication coefficient (3.5) within 6 weeks. Repeated subculturing of nodal explants obtained from shoot cultures enabled continuous production of healthy axillary shoots. At the end of the sixth passage, about 90% of nodal explants produced five or six healthy green shoots, each being about 6.6 cm long with six or seven nodes. Multiplication coefficient was also increased from the first subculture (5.4) to the sixth subculture (8.3). The best rooting response was achieved on solidified half-strength MS medium supplemented with 4.9 μM indole-3-butyric acid (IBA). About 70% of the micropropagated plantlets were established successfully in 20-cm pots containing a mixture of soil and farmyard manure (4 : 1 ratio) and formed new leaflets.  相似文献

The cutting propagation technique ofPicea koraiensis was carried under automatic spraying of total sunlight by adopting the same year epicormic branches from different trees with different age treated by different hormones in different concentration and time and the orthogonal and randomized block was designed. The results were as follows: (l)The rooting rate of the cutting reached at least 87 percent, when the cuttings were from the seed tree with the age below 25 years old and the cuts of the cuttings base were in vertical angle and on the fine sand medium. (2) The sequence of effective factors affecting the rooting rate were the style of cut, the age of seed tree and the type of medium. [目的]建立16年生卷荚相思优树组培快繁技术体系。[方法]以16年生卷荚相思优树的当年新生枝条带腋芽茎段为材料,对卷荚相思外植体进行消毒、初代培养、增殖培养、生根培养和移植等。[结果]表明:通过对16年生卷荚相思成年优树采条进行扦插,以扦插苗建立采穗圃,选取采穗圃中当年生健康无病虫害枝条的中段为外植体,最佳消毒方式为75%的酒精处理0.5 min和0.1%的升汞处理18 min,其存活率达69.33%,芽诱导率达86.67%;最佳初代培养基为改良MS+蔗糖40 g·L-1,出芽率为91.33%。最佳增殖培养基为MS+6-BA 0.5 mg·L-1+NAA 0.1 mg·L-1+蔗糖30 g·L-1,35 d增殖倍数可达3.50倍;最佳生根培养基为1/2 MS+IBA 0.25 mg·L-1+NAA 0.5 mg·L-1+蔗糖40 g·L-1,15 d生根率为96.11%;将生根苗移植至以沙为基质的营养杯中,存活率为71.11%。[结论]研究解决了16年生卷荚相思成年优树外植体污染率高和芽诱导率低等问题,建立的组培快繁技术体系对今后加快卷荚相思良种选育及优质苗木大量扩繁具有重要作用。  相似文献

Juniperus species (juniper) are resistant to drought and have medicinal properties. In order to investigatethe condition of juniper reproduction and prevent a decline in its area, largely caused by problems in natural regeneration, the asexual reproduction of juniper has received special attention besides its sexual propagation. Among the six species of juniper in Iran, the most abundant are Juniperus polycarpos and J. excelsa. Juniperus polycarpos is ambi-sexual and J. excelsa unisexual. We show that collecting seeds in September provided for better germination than in November. In sexual propagation 82.0% and in asexual (graft) 47.6% success were achieved. Compared to a mixture of water, lime and hormones, a natural treatment of only water had better results. Diurnal change of temperature increased germination of juniper seeds over a constant temperature. Aphyllous stem cuttings from mature and old trees are less successful in rooting than leafy cuttings of juniper saplings. A treatment with Aaterra significantly reduced the percentage of rooted cuttings and total root length. Aliette, Fongarid, Octave, Benomyl, Delsene M and Maneb fungicides significantly increased the percentage of rooted cuttings but did not affect total root length. Autumn explants formed more calli than spring explants. Successful multiplication was achieved by treating buds with a mixture of 0.7% polyvinyl pyrrolidone (PVP) and 2% sucrose and cultured in a basal MS or WRC medium, with a maximum number of new buds. In consideration of extracted material of juniper species in remedy of infectious, fungus, contagious diseases as well as for drought resistance and longevity of this species in severe arid environmental conditions and various industrial applications, reproduction and seed production by micro propagation and biotechnology are necessary.  相似文献

[目的]松材线虫病是我国对森林危害和威胁最严重的病害,控制其传播媒介昆虫松褐天牛是防治松材线虫病的主要手段。作者发现的松褐天牛深沟茧蜂(Iphiaulax monochamusi Yang)是寄生松褐天牛中老龄幼虫的重要天敌。为明确松褐天牛深沟茧蜂的寄生率与寄主树木、寄主和环境因子的关系,开展了本研究。[方法]通过解剖46株松褐天牛危害致死的马尾松,调查了松褐天牛深沟茧蜂寄生率与马尾松、松褐天牛和环境因子之间的关系。[结果]调查研究表明:寄主树木的高度、胸径和树龄对松褐天牛深沟茧蜂寄生率没有显著影响,松褐天牛的数量对松褐天牛深沟茧蜂寄生率亦没有显著影响,而松褐天牛幼虫龄期和松褐天牛幼虫在树干上的位置显著影响松褐天牛深沟茧蜂寄生率。松褐天牛深沟茧蜂寄生率与松褐天牛龄期呈显著正相关,其偏好寄生3~5龄幼虫;就天牛在其危害寄主树木上的位置而言,松褐天牛深沟茧蜂偏好寄生马尾松主干上部和在韧皮部危害的松褐天牛幼虫,其对位于马尾松树干上部的寄主幼虫寄生率最高,达27.38%,对在韧皮部生活、危害的寄主幼虫寄生率为20.18%,显著高于位于木质部生活的寄主幼虫寄生率(5.46%)。逐步回归分析表明:影响松褐天牛深沟茧蜂寄生率的关键环境因子是寄主树木的坡位和其它天敌寄生率,其中,坡位与松褐天牛深沟茧蜂呈显著正相关,其它天敌寄生率与松褐天牛深沟茧蜂寄生率呈显著负相关。[结论]以上研究初步明确了松褐天牛深沟茧蜂的寄生率与寄主树木、寄主害虫和天敌之间的关系,为今后利用该重要天敌控制松褐天牛打下了基础。  相似文献

Aging of the donor tree decreased adventitious root formation in shoot cuttings of Tectona grandis Linn. f. (teak). Exogenous application of auxins, i.e., α-naphthalene acetic acid (NAA) and indole-3-butyric acid (IBA) has a significant positive effect on the percentage of rooting. The maximum percent rooting was obtained with 4,000 ppm IBA as compared to other treatment. Significant increase in root number was recorded in shoot cuttings treated with 4,000 ppm NAA. The overall rooting response was better in the treatment with IBA rather than with NAA. Further periodic samples (0, 10, 20, and 30 days) were taken to assess the total soluble sugar, starch, protein, and peroxidase (PER) activity in the rooting zone of shoot cuttings of teak during adventitious root formation. Application NAA and IBA to shoot cuttings resulted in an increase in the level of total soluble sugar, starch, protein, and PER-activity in the rooting zone. The stored carbohydrates were utilized during adventitious root formation. Hence, total soluble sugar and starch contents of cuttings, irrespective of age of donor plants, decreased with the passage of time in cuttings planted for rooting. Significant fluctuations were observed in the protein content of cuttings during the time of root induction. There was an increase in the protein content with the passage of time from the day of planting up to its 20th day, followed by a sharp decline in the protein content of cuttings at the 30th day of planting, irrespective of the age of donor plants or the treatment of cuttings with auxins. Irrespective of donor plant age, PER-activity in the cuttings increased from the day of their planting for rooting up to the 20th day, and then declined at its 30th day of planting. It was interesting to note that PER-activity remained higher at all stages in the cuttings of 2-month-old seedlings which rooted profusely as compared to the cuttings of 15- and 30-year-old donor plants those rooted poorly. This study suggested that the exogenously applied NAA and IBA at different concentration seems to activate sugar metabolism for release of energy, protein and PER-activity which are necessary for cellular division and differentiation during adventitious root primordium initiation or development in the rooting zone of shoot cuttings.  相似文献

Pollen dispersal was estimated in two test plots in a hinoki (Chamaecyparis obtusa) seed orchard using a chloroplast DNA marker, the spacer region between thetrnD andtrnY genes, and SSCP (single strand conformation polymorphism). In Plot 1, 2,020 seeds from 40 trees within 30 m of the marker tree were analyzed using the PCR-SSCP method. In Plot 2, 1,850 seeds from 37 trees were analyzed in the same manner. The results revealed that the maximum pollen dispersal distance in the two plots exceeded 25 m. Pollen dispersal appeared to be inversely proportional to the distance from the marker tree. The effective pollen dispersal was suggested to be less than about 20 m in a mature hinoki seed orchard. Adjacent trees had an excessive influence when the pollen density was increased by artificial flower stimulation. Therefore, it was suggested that seed production better resembles ideal random mating when carried out as naturally as possible. In conclusion, the SSCP chloroplast DNA marker was a useful tool for amassing basic information on pollen management in seed orchards of coniferous species.  相似文献

A study was conducted to identify and explore indigenous knowledge relating to fodder trees and silvopastoral management systems of small-scale farmers in seasonally dry areas of Jamaica. The objectives of the study were to: 1) explore farmers indigenous knowledge relating to fodder trees and silvopastoral management systems in a subtropical dry climate; 2) examine pastoral land-use systems and technologies in current use; and 3) seek to integrate this information into silvopastoral tree fodder systems. Data were gathered via observation and semistructured interviewing. A snowball sampling strategy was used to purposively select all small-scale farmers who had cattle in the Green Park valley for interview.Of forty farmers interviewed, 37 males (92.5%) and 3 females (7.5%) were primary caretakers of cattle. Twenty-seven of 40 farmers (68%) raised cattle to generate income. During extended droughts farmers are forced to look for alternatives to desiccated, overgrazed pasture grasses, including: 1) local travel to harvest sugar cane tops and to cut Guinea grass (Panicum maximum); 2) purchase of bag feed; and 3) harvest and use of tree fodder. Preferred fodder trees were identified as Bacedar (Guazama ulmifolia), Guango (Albizia saman), Breadnut (Brosimum alicastrum) and Quickstick (Gliricidia sepium).Recommended silvopastoral management systems include fodder hedgerows, three strata forage systems, and living fences. In addition, production of indigenous fodder tree species, introducing improved tree fodder species, and planting improved pasture grasses concurrent with cash trees is suggested.  相似文献

The effect of two tree species, karité (Vitellaria paradoxa) and néré (Parkia biglobosa) on soil condition, development and production of cotton, sorghum and pearl millet, main crops in South Mali, has been investigated.The soil under the trees is slightly richer (organic matter content and several cations) compared to adjacent tree-less sites. Also tree-specific effects exist.Soil enrichment, however, is mainly a matter of redistribution of locally available nutrient resources.Of the six associations studied, only karité-cotton shows no tree-induced reduction in crop production. All other associations suffer greatly from reduced crop outputs caused by the trees, usually in the order of 60%.There are at least two reasons that explain tree-induced yield reduction. Because of increased humidity in the immediate surroundings of trees, in both soil and air, lower numbers of crop plants survive up to maturity, presumably because plants are attacked by fungi. Secondly, crop plants that mature show reduced output because of inter-specific competition for light and nutrients.In case of néré, in order to minimize the trees' negative influence lopping is advised. Such practice, however, is less suitable in case of karité, because of its assumed lower potential to regenerate, as compared to néré.  相似文献

The frequency of open-pollinated male-sterile seedlings from a male-sterile mother tree in Cryptomeria japonica D. Don was determined in 765 open-pollinated seedlings. Pollen was not formed by 29 out of the 765 seedlings. To find pollen donor candidates for 29 male-sterile seedlings, a male-sterile tree was crossed with five individuals in the neighborhood of the mother tree. Analysis of microspores in male flowers of F1 seedlings revealed that one of the five individuals had a heterozygous male-sterility gene. Paternity tests were conducted on the 29 male-sterile seedlings using eight kinds of CAPS markers in C. japonica. The pollen parents of 29 male-sterile seedlings were not only a tree in the neighborhood of the mother tree, but also other trees. Since it was found that the pollen parents of four of the male-sterile seedlings were not in the study plot, it was inferred that C. japonica trees that possess the male-sterility gene grow in other places and may be widely distributed.  相似文献

On fertile alluvial soils on the lakeshore plain of Malawi, maize (Zea mays L.) yields beneath canopies of large Faidherbia albida (synAcacia albida) trees greatly exceed those found beyound tree canopies, yet there is little difference in soil nutrients or organic matter. To investigate the possibility that soil nutrient dynamics contribute to increased maize yields, this study focused on the impact of Faidherbia albida on nitrogen mineralization and soil moisture from the time of crop planting until harvest. Both large and small trees were studied to consider whether tree effects change as trees mature.During the first month of the rainy season, a seven-fold difference in net N mineralization was recorded beneath large tree canopies compared to rates measured in open sites. The initial pulse beneath the trees was 60 g N g–1 in the top 15 cm of soil. During the rest of the cropping cycle, N availability was 1.5 to 3 times higher beneath tree canopies than in open sites. The total production of N for the 4-month study period was 112 g N g–1 below tree canopies compared to 42 g N g–1 beyond the canopies. Soil moisture in the 0–15 cm soil layer was higher under the influence of the tree canopies. The canopy versus open site difference grew from 4% at the beginning of the season to 50% at the end of the cropping season.Both N mineralization and soil moisture were decreased below young trees. Hence, the impact of F. albida on these soil properties changes with tree age and size. While maize yields were not depressed beneath young F. albida, it is important to realize that the full benefits of this traditional agroforestry system may require decades to develop.  相似文献

Tests were conducted to assess rootability of red alder softwood cuttings from shoots of young trees and epicormic sprouts of mature individuals. Ortets were 3 to 34 years old, and treatments with and without indole-3-butyric acid (IBA) were compared. The extent of rooting and root vigor on the cuttings varied greatly among ortets and treatments. Among nonterminal cuttings, the best overall rooting success, with and without IBA, was obtained with cuttings of the youngest ortet. Average rooting of terminal and nonterminal cuttings of one clone were 100% and 64%, respectively. Although cuttings of some trees rooted well without IBA, the IBA treatment appeared to be generally beneficial. For all nonterminal cuttings of all clones, best results (80.0%/x) were obtained with a 10-sec dip in 2,000 or 4,000 ppm IBA. Vigorous planting stock was obtained from the rooted cuttings of all clones. Using epicormic sprouts from mature trees of proven performance will presumably allow propagation of superior trees and establishment of new plantations with improved stock.  相似文献
