The search for renewable and abundant sources of antioxidants has recently focused on agricultural byproducts, especially promising due to their natural origins and low costs. In particular, plant raw materials are sources of important compounds such as dietary fiber, carotenoids, tocopherols, and polyphenolics, which are mostly discarded during harvesting and processing. Among these vegetal crops, red chicory is attractive because of the large quantity of its byproducts (residues as leaves and stems); moreover, there is no information on its role as a food and feed ingredient. In this study, red chicory leaf residue was evaluated as a natural substitute for synthetic antioxidants for the food and feed industry. After lyophilization, a red chicory extract (RC) was characterized for its phenolic profile and its oxidative stability as compared to BHT. RC was shown to reduce lipid peroxidation of different oils in the Rancimat test. In addition, the antioxidant property of RC was studied in a model system by evaluating the Saccharomyces cerevisiae response to oxidative stress by means of gene expression. In this analysis, the RC extract, added to the yeast culture prior to oxidative stress induction, exhibited a pleiotropic protective effect on stress responsive genes.  相似文献

Prevention of oxidative rancidity in rice bran during storage.   总被引:5,自引:0,他引:5  
The effect of microwave heat on lipoxygenase (LOX) activity in rice bran under various storage conditions was examined. Raw rice bran from the long-grain variety Lemont was adjusted to 21% moisture content and heated in a microwave oven at 850 W for 3 min. Raw and microwave-heated rice bran samples were packed in zipper-top bags or vacuum packs and stored at room temperature (25 degrees C) or in the refrigerator (4-5 degrees C) for 16 weeks. Samples were analyzed for LOX activity at 4-week intervals. LOX activity did not significantly change from its initial value at week 0 for zipper-top and vacuum-packed samples while stored at 4-5 degrees C for 12 weeks, but decreased at week 16. Vacuum packing did not show a significant impact on LOX activity during 16 weeks of storage. Microwave-heated samples stored in the refrigerator did not show significant change in LOX activity for up to 12 weeks but showed a significant (p < 0. 05) decrease at 16 weeks. Results showed that oxidative rancidity of rice bran could be prevented by microwave heating the samples, packing in zipper-top bags, and storing at 4-5 degrees C for up to 16 weeks.  相似文献

Cocoa is a rich source of flavanols and procyanidin oligomers with antioxidative properties, providing protection against oxidation and nitration. The present study investigated the potential protective effect of a polyphenolic extract from cocoa on cell viability and antioxidant defenses of cultured human HepG2 cells submitted to oxidative stress induced by tert-butylhydroperoxide (t-BOOH). Pretreatment of cells with 0.05-50 microg/mL of cocoa polyphenolic extract (CPE) for 2 or 20 h completely prevented cell damage and enhanced activity of antioxidant enzymes induced by a treatment with t-BOOH. Moreover, lower levels of GSH caused by t-BOOH in HepG2 cells were partly recovered by a pretreatment with CPE. Increased reactive oxygen species (ROS) induced by t-BOOH was dose-dependently prevented when cells were pretreated for 2 or 20 h with CPE. These results show that treatment of HepG2 in culture with CPE (within the physiological range of concentrations) confers a significant protection against oxidation to the cells.  相似文献

Fractionation of the polyphenols constituting a food grade lingonberry extract (Vaccinium vitis-idaea) highlighted a composition more complex than described until now in the berry. Procyanidins B1, B2, and A2 were identified by UPLC/ESI-MS(2) along with the presence of other flavanol oligomers. Processing induced the release of large amounts of aglycones for ferulic acid, p-coumaric acid, and quercetin. The described anthocyanic composition of lingonberry was completed with hexoside derivatives of peonidin, petunidin, malvidin, and delphinidin. Besides confirmation of in vitro antioxidant activity, in vivo study was performed on rats fed a diet inducing oxidative stress. Supplementation with lingonberry extract significantly decreased the total oxidant status and favorably affected antioxidant defense enzymes in red blood cells and liver. A drop in the serum reduced glutathione level was also prevented, and uric acid was maintained at low level, confirming the antioxidant activity of the extract (5% proanthocyanidins) from a dosage of 23 mg/kg of body weight.  相似文献

The Chinese herb DongChong-XiaCao originating from Cordyceps sinensis is widely used as a traditional medicine in China for treatment of a wide variety of diseases. The extracts of Cordyceps sinensis (CSE) and Cordyceps militaris (CME) are well-known for their biological effects. In the present study, the antioxidant efficiency of CME and CSE in protecting lipid, protein, and low-density lipoprotein (LDL) against oxidative damage was investigated. CME and CSE showed weakly inhibitory effect on liposome oxidation, that of CME being superior to that of CSE. As for the protein oxidation model system, the inhibitory effect of CME on protein oxidation was inferior to that of CSE. CME and CSE at 1.0 mg/mL showed 50.5 and 67.1% inhibition of LDL oxidation, respectively. The contents of bioactive ingredients cordycepin and adenosine in CME are higher than those of CSE; however, both cordycepin and adenosine showed no significant antioxidant activity as determined by the Trolox equivalent antioxidant capacity method. Polyphenolic and flavonoid contents are 60.2 and 0.598 microg/mL in CME and 31.8 and 0.616 microg/mL in CSE, respectively, which may in part be responsible for their antioxidant activities. In addition, a polysaccharide present in CME and CSE displayed antioxidant activity, which suggested that the activity might be derived partly from polysaccharides of CME and CSE. The tendency to scavenge the ABTS(*)(+) free radical and the reducing ability of CME and CSE display concentration-dependent manners, suggesting that CME and CSE may be potent hydrogen donators. On the basis of the results obtained, the protective effects of CME and CSE against oxidative damage of biomolecules are a result of their free radical scavenging abilities.  相似文献

Phenolic compounds in extra virgin olive oil (EVOO) have been associated with beneficial effects for health. Indeed, these compounds exert strong antiproliferative effects on many pathological processes, which has stimulated chemical characterization of the large quantities of wastes generated during olive oil production. In this investigation, the potential of byproducts generated during storage of EVOO as a natural source of antioxidant compounds has been evaluated using solid-liquid and liquid-liquid extraction processes followed by rapid resolution liquid chromatography (RRLC) coupled to electrospray time-of-flight and ion trap mass spectrometry (TOF/IT-MS). These wastes contain polyphenols belonging to different classes such as phenolic acids and alcohols, secoiridoids, lignans, and flavones. The relationship between phenolic and derived compounds has been tentatively established on the basis of proposed degradation pathways. Finally, qualitative and quantitative characterizations of solid and aqueous wastes suggest that these byproducts can be considered an important natural source of phenolic compounds, mainly hydroxytyrosol, tyrosol, decarboxymethyl oleuropein aglycone, and luteolin, which, after suitable purification, could be used as food antioxidants or as ingredients in nutraceutical products due to their interesting technological and pharmaceutical properties.  相似文献

Peanut roots as a source of resveratrol   总被引:12,自引:0,他引:12  
A potent antioxidant, resveratrol (3,4',5-trihydroxystilbene), was extracted using 80% methanol from peanut roots (Arachis hypogaea L.), isolated with a solid-phase extraction column, purified by a semipreparative HPLC, and identified with 1H NMR and MS. The highest and lowest resveratrol contents in the peanut roots of 2000 fall and 2001 spring crops were 1.330 and 0.130 mg/g and 0.063 and 0.015 mg/g, respectively. When the dehydrated peanut root powders of spring and fall crops were combined and cooked with pork-fat patties (1%, w/w) and the separated oils were stored at 60 degrees C for conjugated diene hydroperoxide (CDHP) determination, CDHP contents of the control oils increased after 3 days of storage, whereas the contents in the peanut root-treated oils of spring and fall crops did not increase after 9 and 15 days of storage, respectively. It is of merit to find that peanut roots, usually left in the field as agricultural waste, contain resveratrol and bear potent antioxidative activity.  相似文献

This study investigated the effect of a natural tomato extract (TE) on cataract formation in two animal models. A TE containing 5% lycopene was included in the diet of diabetic sand rats at 0.2%, and Sprague Dawley rats were fed a high-galactose diet (30 g/100 g of diet), supplemented with either the lycopene-rich extract at concentrations of 0.2, 0.4, and 0.8% or BHT (0.2%). TE had no significant effect on plasma glucose levels or cataract development in sand rats; however, in rats maintained on a diet rich in galactose, both BHT and TE decreased cataract incidence, and grades were lower than in control animals. In addition, lens protein and reduced glutathione levels were higher and aldose reductase activity was lower than in the control group. The results suggest that antioxidants act as protective agents when oxidative stress is a primary cause of cataract formation but may be less effective in preventing cataracts in hyperglycemic animals.  相似文献

The antiradical capacity (radical scavenger capacity, RSC) of anthocyanin-based fruit extracts prepared in the laboratory (black chokeberry, black-thorn, and strawberry) was studied by using the 2, 2-diphenyl-1-picrylhydrazyl radical (DPPH(*)). To determine their RSC, the second-order rate constant (k(2)) for the oxidation of these extracts by DPPH(*) was calculated. The value of k(2) was compared to that used in the food industry as natural (alpha-tocopherol) or synthetic (butylated hydroxytoluene (BHT) and butylated hydroxyanisole (BHA)) antioxidants, as well as for a commercial elderberry concentrate and a synthetic colorant (Ponceau 4R). The k(2) values ((mg/mL)(-)(1) s(-)(1)), in methanol at 25 degrees C, were 1.87, 0.7, 0.42, 0.2, 0.05, 0.03, and 0.008 for alpha-tocopherol, black chokeberry, BHA, black-thorn, BHT, strawberry, and elderberry, respectively. Ponceau 4R lacked RSC. Therefore, these natural colorants proved to be a combined source of color and RSC for food material.  相似文献

A peroxidase isozyme (BP) was purified to homogeneity from broccoli stems ( Brassica oleraceae var. maraton) discarded from industrial processing wastes. BP specific activity was 1216 ABTS [2,2'-azinobis(3-ethylbenzthiazoline-6-sulfonic acid)] units/mg, representing 466-fold that of crude extract. BP is a monomeric glycoprotein containing 16% carbohydrates, with a molecular mass of 49 kDa and an isoelectric point close to 4.2. From kinetic data it showed a two-substrate ping-pong mechanism, and the catalytic efficiency measured as the rate-limiting step of free BP regeneration was 3.4 x 10(6) M(-1) s(-1). The ABTS K m value was 0.2 mM, which was about 20 times lower than that reported for acidic commercial horseradish peroxidase (HRP). Assessment of BP secondary structure showed 30% helical character, similar to HRP and cytochrome c peroxidase. BP lost only 25% activity after 10 min of heating at 55 degrees C and pH 6; it was stable in the pH range from 4 to 9 and showed an optimum pH of 4.6 using ABTS as substrate. BP was active on substrates normally involved in lignin biosynthesis, such as caffeic and ferulic acids, and also displayed good catechol oxidation activity in the presence of hydrogen peroxide. Reverse micellar extraction was successfully used as potential large-scale prepurification of broccoli peroxidase, achieving a purification factor of 7, with 60% activity yield. Stems from the broccoli processing industry have a high potential as an alternative for peroxidase purification.  相似文献

Abstract

Numerous fungicide applications are often needed to control powdery mildews in greenhouse-grown crops, and consequently there is an increasing demand for alternative means to control them. In commercial greenhouses, attack of powdery mildew caused by Podosphaera xanthii in cucumbers and P. pannosa in cut roses was controlled by weekly sprays of C-Pro CE601, a dilution of grapefruit seed extract (GSE). The raw material of GSE, delivered by Citricidal®, is a glycerolic extract from seed and juiceless pulp of grapefruits (Citrus paradise). In both cultures C-Pro at 2000 ppm achieved as good control as treatments with penconazole or triforine. The mean level of attack by powdery mildew on leaves of cut roses one week after the last of three weekly treatments, was 9.8, 12.9 and 40.5% for penconazole, C-Pro (2000 ppm), and untreated, respectively. In cucumbers, there was less difference between the untreated control and the different treatments, but for all times of assessments, C-Pro and triforine were better than untreated (p=0.001). There was no significant difference in cucumber between C-Pro and triforine. Phytotoxicity symptoms such as leaf twisting and rolling in roses (cv. Jade and Cezann) and dark green leaves and indications of growth retardation in cucumbers (cv. Ventura) were observed at the higher concentrations used (3000–4000 ppm). Thin layer chromatography of C-Pro CE601 has previously shown presence of benzethonium chloride, a synthetic antiseptic agent. Further research is necessary to determine if the observed effects of the product tested are due to natural compounds or are merely due to a synthetic preservative agent added. Our results revealed that C-Pro might be a supplement or even a replacement to some of the current fungicides used against powdery mildew.  相似文献

Winter legumes can serve dual purposes in no-tillage cropping systems. They can provide a no-tillage mulch, and supply a considerable quantity of N for thesummer crops. Cotton (Gossypium hirsutum L.) was no-tillage planted into crimson clover (Trifolium incarnatum L.), common vetch (Vicia sativa L.), and fallowed soil for two years to determine the effects of winter legume mulches on growth, yield, and N fertilizer requirements. The legumes were allowed to mature and reseed prior to planting cotton. The winter legumes produced no measurable changes in soil organic matter, N, or bulk density, but water infiltration was more rapid in the legume plots than in the fallowed soil. In the fallow system, 34 kg ha?1 N fertilizer was required for near maximum yields. In the clover plots, yields without N fertilizer were higher than when N (34 and 68 kg ha?1) was applied. In the vetch plots, cotton yields were highest without N fertilizer the first year, but yields were increased with 34 kg ha?1 N the second year because of a poor vetch seed crop and a subsequently poor legume stand. In the clover plots, a 20–30% cotton seedling mortality occurred in one year, but this stand reduction apparently did not affect cotton yields. Winter legume mulches can provide the N needs for no-tillage cotton without causing an excessive and detrimental quantity of N in sandy soils naturally low in soil N (0.04%). Unless the reseeding legume systems are maintained for at least 3 years, the legumes do not, however, provide an economical N source for cotton when N fertilizer requirementsare low (34 kg ha?1 in this study). A possible disadvantage of the system for reseeding legumes is that cotton planting is delayed 4–6 weeks beyond the normal planting date, which can reduce yields in some years.  相似文献

Crimson clover (Trifolium incarnatum L.) plus hairy vetch ( Vicia villosa Roth), red clover (Trifolium pratense L.), white clover (Trifolium repens L.), red clover plus white clover, and bermudagrass (Cynodon dactylon [L.] Pers.) were evaluated as cover crops for pecans. Crimson clover plus hairy vetch supplied the equivalent of 101 to 159 kg nitrogen (N)/ha. Red clover plus white clover supplied up to 132 kg N/ha. Either white clover or red clover alone were less effective in supplying N than when grown together. Soil Kjeldahl‐N was usually not affected or increased using the legumes compared to fertilized bermudagrass sod. Soil nitrate (NO3) concentrations during October were occasionally higher in unfertilized legume plots than in bermudagrass plots with March‐applied N.  相似文献

Soil types or map units are considered to be taxonomically adjacent if they differ in only one criterion, defined by an arbitrary threshold value. By treating soil types as nodes of a graph and taxonomic adjacency as the graph edges connecting nodes, algebraic graph theory can be used to produce a measurement of the uncertainty in a soil map associated with arbitrary classification boundaries between soil types. The largest eigenvalue of the adjacency matrix of a graph, the spectral radius, is an indication of network complexity. A larger spectral radius indicates a more complex network, and a greater degree of uncertainty or potential error associated with taxonomic adjacency. Benchmark values of spectral radius for cases of no taxonomic adjacency, including a single pair of adjacent soils, a chain or cycle‐type graph structure and a fully connected graph, are established so that taxonomic adjacency indices based on the spectral radius can be established. Examples are shown from two contrasting USA soil landscapes in the Ouachita Mountains, Arkansas, and the coastal plain of North Carolina, using both US Soil Taxonomy and the world reference base. The taxonomic adjacency indices are also useful in assessing soil richness and pedodiversity, with smaller values indicating a greater likelihood that identified soils represent distinct entities.  相似文献
