Twenty-nine of 200 (14.5%) ewes on a farm in Cobleskill, NY aborted or had dead lambs during the lambing seasons of 1985 and 1986. Thirteen of 15 ewes that aborted had high Toxoplasma gondii antibody titers (1,024), via the modified agglutination test, and T gondii was isolated from the tissues of 1 fetus. In the 1987 lambing season, 5 ewes aborted, but not because of T gondii infection. Toxoplasma gondii antibodies were detected in 73.8% of sera obtained from 592 ewes in January 1987, indicating enzootic toxoplasmosis on this farm. Seropositivity increased with age; 40.2% of 1-year-old ewes had detectable antibody vs 89.2% of 2-year-old ewes.  相似文献

Summary Serological investigation by the indirect haemagglutination test forToxoplasma antibodies was carried out in a flock of dairy goats. Antibody titres ranging from 1:4 to 1:1,024 were found in 34% of 371 apparently healthy animals. High antibody titres of 1:256 suggestive of recent infection were noted in 18 (4.9%) animals including 13 nannies. The occurrence of a number of non-brucella abortions associated with high serological titres and the isolation of the parasite from the brain tissues of an aborted foetus suggestedToxoplasma as a probable aetiologic factor. Sera having significant levels of haemagglutinating antibodies (1:128) were also found positive to the card agglutination test. Epidemiological aspects and the public health potential of subclinical toxoplasmosis in dairy goats are discussed.

Resumen Se realizaron investigaciones serológicas de toxoplasmosis mediante la prueba indirecta de hemaglutinación en un rebaño de cabras lecheras. Se encontraron títulos de 1:4 a 1:1024 en 34% de 371 animales aparentemente sanos. Títulos de 1:256 sugestivos de infecciones recientes, se encontraron en 18 (4.9%) animales, incluyendo 13 hembras paridas. La ocurrencia de un significativo número de abortos no asociados con brucela, títulos serológicos altos de toxoplasmosis y el aislamiento del parásito del cerebro de fetos abortados, sugiere que eltoxoplasma es el factor etiológico más importante. Se discuten aspectos epidemiológicos de importantcia para la salud pública, relacionados con el potencial de la toxoplasmosis (subclínica) en cabras lecheras.

Résumé Des investigations sérologiques par la méthode de l'agglutination indirecte pour rechercher des anticorps toxoplasmiques ont été faites sur un troupeau de chèvres laitières. Des anticorps ont été mis en évidence chez 34 p.100 des 371 animaux apparemment sains à des titres allant de 1:4 à 1:1024. Des titres élevés d'anticorps atteignant 1:256 ont été relevés chez 18 (soit 4,9 p.100) des animaux comprenant 13 chèvres, ce qui fait penser à une infection récente.L'existence de nombre d'avortements non-brucelliques associés avec des titres sérologiques élevés et l'isolement du parasite des tittus cérébraux d'un foetus avorté suggerent que les Toxoplasmes en ont probablement été le facteur étiologique. Des sérums ayant des taux significativement élevés d'anticorps agglutinants (1:128) ont également été reconnus comme positifs par le test d'agglutination en EAT. Les aspects épidémiologiques et le danger potentiel pour l'homme de ces cas subcliniques de toxoplasmose chez les chèvres laitières sont discutés.

Background

Toxoplasma gondii is a major problem for the sheep industry as it may cause reproduction problems. The importance of T. gondii in Norwegian goat herds is uncertain, but outbreaks of toxoplasmosis in dairy goat farms have been recorded. The aim of this study was to describe the prevalence of T. gondii infection in Norwegian dairy goats by using serology.

Findings

Goat serum originally collected as part of two nationwide surveillance and control programmes between 2002 and 2008 were examined for T. gondii antibodies by using direct agglutination test. In total, 55 of 73 herds (75%) had one or more serologically positive animals, while 377 of 2188 (17%) of the individual samples tested positive for T. gondii antibodies.

Conclusions

This is the first prevalence study of T. gondii infection in Norwegian goats. The results show that Norwegian goat herds are commonly exposed to T. gondii. Nevertheless, the majority of goat herds have a low prevalence of antibody positive animals, which make them vulnerable to infections with T. gondii during the gestation period.  相似文献

Little information is available about the seroprevalence of Toxoplasma gondii and Neospora caninum infections in goats in Romania and even in Europe. During 2007-2010, 735 serum samples were collected from dairy goats located in 4 historical regions (Cri?ana, Maramure?, Transylvania and Muntenia) of Romania. Sera were analyzed for T. gondii and N. caninum antibodies (IgG type) by enzyme-linked immunosorbent assay (ELISA) using two commercial kits (Chekit Toxotest Antibody ELISA and Chekit Neospora caninum Antibody ELISA; Idexx-Bommeli, Switzerland). Three hundred and eighty-eight out of 735 (52.8%) goats presented T. gondii antibodies and 12 out of 512 (2.3%) goats had N. caninum antibodies. The high seroprevalence of T. gondii suggests that infection with this parasite is common in dairy goats in Romania, and less common the infection with N. caninum. This is the first time that infection with N. caninum in goats has been reported in Romania and the first extended study on seroepidemiology of T. gondii.  相似文献

Serum samples from 1,000 dairy goats from northwest United States (1982 to 1984) were examined for Toxoplasma gondii antibodies by a modified agglutination test. Toxoplasma gondii antibody titers were less than 1.40 for 779 goats, 1.40 for 153 goats, and greater than or equal to 1:400 for 68 goats. Seroprevalence increased with age of goats; 3.7% of 54 six-month-old goats were seropositive (greater than or equal to 1:40) vs 17.8% of 218 one-year-old goats.  相似文献

During the lambing season of 1983/1984, 8 of 44 purebred Hampshire ewes on a farm in Knoxville, Md had reproductive problems. In at least 4 of these ewes, the problem was attributed to toxoplasmosis. Necrosis and Toxoplasma gondii tachyzoites were found in placental specimens from 3 ewes. Agglutinating antibody to T gondii, at a titer of 1:80, was found in pleural fluids of both fetuses aborted from 1 ewe; this ewe had an antibody titer of 1:6,400 at the time of abortion. In another ewe, the diagnosis was confirmed by the isolation of T gondii from the placenta and 1 of her lambs. Of numerous free-roaming adult cats on the farm, 16 were trapped, euthanatized, and examined for T gondii. Agglutination antibody to T gondii, at titers of 1:4 to 1:64, was found in serum samples from all the cats. Toxoplasma gondii was isolated from the brain and skeletal muscles of 9 of the cats, and from the feces of 1 cat. Blood samples obtained from all 78 sheep on the farm 6 months after the episode of abortion were examined for antibody to T gondii. Agglutinating antibody titers to T gondii were less than 1:16 in 46 sheep, 1:16 in 16, 1:64 in 12, 1:256 in 2, 1:024 in 1, and 1:4,096 in 1. Analyses of serologic data in sheep of various age groups suggested that the Toxoplasma infection was acquired sporadically, probably from feed contaminated with oocysts.  相似文献

This study assessed the effectiveness of a mutant strain of Toxoplasma gondii (RH strain) lacking the mic1 and mic3 genes (Mic1-3KO) against Toxoplasma abortion in sheep. Ewes were inoculated subcutaneously with 105 Mic1-3KO tachyzoïtes in three independent experiments. Following vaccination, Mic1-3KO induced a mild febrile response and serum IgG antibodies, which persisted throughout the experiments. Tissue cysts formed in the sheep, but were not, under our experimental conditions, infectious when given orally. Ewes were mated two months after vaccination and were orally challenged with the PRU strain of T. gondii at mid-gestation (400 oocysts in Experiments 1 and 2; 100 oocysts in Experiment 3). Challenge of vaccinated pregnant ewes resulted in a slight febrile response, whereas unvaccinated ewes developed a more severe, characteristic febrile response of longer duration. After challenge, all unvaccinated ewes aborted whereas 62%, 91% and 64% (Experiments 1, 2 and 3 respectively) of the lambs from vaccinated ewes were viable, with no clinical signs of infection. Mic1-3KO was as effective as S48, the strain used as a live vaccine for sheep (Toxovax®). A dose of 105 Mic1-3KO tachyzoites was sufficient to induce protection (versus a dose of 2 × 106). Both subcutaneous and intraperitoneal injections were effective. Moreover, preliminary results showed the potential of Mic1-3KO to reduce the development of tissue cysts in lambs born to vaccinated ewes. This study demonstrates that Mic1-3KO is a potent vaccine candidate.  相似文献

Nineteen does (female goats) were dosed with 500,000 oocytes of Hammondia hammondi prior to breeding. At about 90 days of gestation these, and 18 uninoculated does were challenged with 25,000 Toxoplasma gondii oocysts. The 19 H. hammondi--inoculated does produced 26 live and one dead kid (newborn goat). The 18 does not given H. hammondi produced 12 live and 19 dead kids. However, examination of all of the kids by isolation of T. gondii in mice, serology and histology revealed that they were all infected with T. gondii. Thus, while H. hammondi "vaccination" is protective against the deleterious effects of T. gondii on pregnant does, perhaps by reducing the severity of placental lesions, it does not prevent foetal infection.  相似文献

Food-borne parasitic diseases, such as toxoplasmosis, are increasingly becoming a global food safety concern. A cross-sectional study was conducted to investigate seroprevalence and risk factors of Toxoplasma gondii seropositivity in apparently healthy, unvaccinated dairy goat flocks reared under mixed smallholders, northern Tanzania between April and October 2011. Flock- and animal-level data were collected using a questionnaire. Sera (n?=?337) collected from goats aged ≥6 months and from 102 flocks, respectively, were analyzed using modified Eiken latex agglutination test. A flock was classified as T. gondii seropositive if at least one animal tested positive. Titers considered diagnostically significant (≥1:16) were detected in 19.3 % of goats and 45.17 % of flocks, respectively. The antibody levels ranged from 1:16 to 1:2,048 and among the seropositive goats, the proportion of high antibody levels (≥1:2,048), suggestive of acute infection, was 1.5 %. The study revealed that goats raised in Babati are at a lower risk of acquiring T. gondii infection (P?=?0.00209) than those which are raised in Arumeru district. The prevalence of T. gondii antibody was significantly higher in crossbred (24.7 %) and Saanen (24.4 %) breed goats than in local (14.3 %) and Toggenburg (12.1 %) and in females than in males (P?=?0.043). No significant difference was observed among goats kept under various husbandry practices. The relatively high seroprevalence detected in this study suggests that toxoplasmosis may be posing a significant animal and human health risk and that the consumption of goat meat may play a role in the transmission of the disease to humans.  相似文献

An outbreak of foot lameness in a dairy herd of 170 goats is described. The herd, that was kept mostly indoors, was attended between December 2006 and November 2007. During this period, 15% of the goats showed lameness with foot lesions and 24% of the goats showed lameness without foot lesions. The foot lesions consisted of cracks and erosions on the horn of the bulbs of the heel that in most cases extended along the internal side of the axial hoof wall. The clinical diagnosis was interdigital dermatitis. Annual milk production was lower in lame goats compared to those that were not lame (P < 0.05). Furthermore, annual milk yield was significantly affected by the type of foot disease present and was significantly lower in cases with foot lesions than in the animals that were not lame (P < 0.05) and also in the lame goats without foot lesions (P < 0.05).  相似文献

Tissues of 20 2-3-month-old kids and 17 2-4-year old does fed Toxoplasma gondii oocysts were examined histologically. Kids were necropsied between 3 and 50 days post-inoculation (DPI) and does were necropsied between 10 and 422 DPI. Lesions were seen in tissues of all young goats fed 1000-100,000 oocysts and consisted of acute enteritis, necrosis of mesenteric lymph nodes and encephalomyelitis. Selenium is an essential part of the enzyme glutathione-peroxidase (GSH-Px) and plays an important role in the intracellular aspecific immune defence. Reference values for blood levels of GSH-Px are not available for dairy goats. The EU has authorized the addition of selenium (as E), in the form of sodium selenite or sodium selenate, to animal feeds, to a maximum of 0.5 mg selenium/kg complete feed. Dairy goats given feed containing the maximum level of selenium (0.5 mg/kg) had GSH-Px levels of more than 1000 U/g Hb. The reference values for GSH-Px in cattle, horses, and pigs are between 120 and 600 U/g Hb. Newborn kids had GSH-Px levels between 350 and 400 U/g Hb, comparable with those ofnewborn calves. In conclusion, the addition of selenium to feeds for dairy goats in amounts authorized by the EU leads to blood GSH-Px levels that are substantially higher than those in other species, such as horses, cattle, and pigs. Thus the maximum level of supplemental selenium in feeds for dairy goats should be less than 0.5 mg/kg.  相似文献

The focus of this work is to determine the distribution and identify species of Eimeria parasites of dairy goats in the livestock of the National Goat and Sheep Research Center in Sobral, State of Ceará, Northeast Brazil. Results showed the presence of multiple species in 196 of 215 analyzed samples (91.2%). Fifty five out of these were from kids (28%) and 141 from adult goats (72%). Eight different Eimeria species were identified and their prevalence in the herd was: Eimeria alijevi Musaev, 1970 (26.7%), E. arloingi (Marotel, 1905) Martin, 1909 (20.6%), E. hirci Chevalier, 1966 (18%), E. ninakohlyakimovae Yakimoff & Rastegaieff, 1930 (16.2%), E. jolchijevi Musaev, 1970 (8.7%), E. christenseni Levine, Ivens & Fritz, 1962 (6%), E. caprovina Lima, 1980 (2.8%) and E. caprina Lima, 1979 (1%). Moreover, E. ninakohlyakimovae showed higher prevalence in kids (97%), followed by E. arloingi and E. alijevi (88%). On the other hand, E. alijevi (77%) was more common in adult goats followed by E. hirci (74%) and E. ninakohlyakimovae (70%). The species E. caprina had low frequency in both kids (27%) and adult goats (13%). Data indicated that infection was relatively common among kids and adult goats. The implementation of a routine diagnostic strategy can be useful in maintaining Eimeria populations under monitoring and will enable the determination of its potential impact on dairy goat herds in Northeast Brazil.  相似文献
