Current methods for obtaining venous blood from the reproductive organs of livestock often have a low rate of success or involve intensive surgical procedures that may impair ovarian function. Therefore, the caudal vena cava was catheterized via the lateral saphenous vein to determine the feasibility of using this method for chronic sampling of blood draining from the reproductive organs of ewes (n = 6), cows (n = 6), and gilts (n = 7). Blood samples were collected at 2-cm (ewes and gilts) or 5-cm (cows) intervals during insertion of catheters. Correct placement, defined as the position at which plasma concentrations of progesterone or estrogen were at least threefold greater than in jugular venous plasma, varied among species and among animals within species. It seemed, however, that a majority of catheters would be placed correctly if secured at 48 to 52 cm in ewes, 52 cm in gilts, and 90 to 100 cm in cows. Saphenous vein catheters were secured for sequential sampling of vena caval blood during the follicular phase of ewes (n = 25), cows (n = 4), and gilts (n = 5). Catheters remained patent for the duration of sampling in all individuals. Concentrations of estrogen in jugular and vena caval plasma were correlated (ewe P less than .0003; cow P less than .0001; gilt P less than .0001). Profiles of progesterone and estrogen revealed an episodic pattern of secretion in vena caval but not jugular plasma. Catheterization of the vena cava via the saphenous vein is a relatively simple and noninvasive method for obtaining blood containing uterine and ovarian hormones before their metabolism.  相似文献

Twelve horses of various breeds and either sex were anesthetized with xylazine and ketamine injected into a median or lateral thoracic vein. During anesthesia, with the horse in sternal recumbency, a 14-gauge, 8.9 cm catheter was inserted into each jugular vein by using aseptic technique. Guaifenesin in water (100 mg/kg or a maximum dose of 50 grams) was infused into one jugular vein and an equal volume of 0.9% saline solution was infused into the other jugular vein. Seven horses received 10% guaifenesin, and five horses received 5% guaifenesin. The catheters were removed before the horses recovered from anesthesia. The horses were euthanatized approximately 48 hours later, and the jugular veins were removed for histologic examination. Adherent thrombus material was observed in all veins exposed to 10% guaifenesin and in one vein exposed to 5% guaifenesin. No evidence of thrombus was observed in four veins infused with 5% guaifenesin or in those infused with saline solution. These findings are of particular significance with horses at increased risk for thrombosis or thrombophlebitis.  相似文献

Before injection of diuretic drugs, blood pressures were determined in the cranial superficial epigastric veins (milk vein) and jugular veins of 15 Holstein cows with udder edema at parturition and of 15 healthy (control) cows at parturition. Cows with udder edema had a significant (P less than or equal to 0.05) mean increase in cranial superficial epigastric venous pressure at parturition, compared with that of control cows. After IV administration of 500 mg of furosemide in cows with udder edema, the mean cranial superficial epigastric venous blood pressure significantly (P less than or equal to 0.05) decreased within 5 minutes, remained at the decreased pressure for 90 minutes, and then returned to near pretreatment pressures by 210 minutes after furosemide injection; however, furosemide did not significantly affect the jugular venous blood pressure of these cows. Furosemide injection did not significantly affect the cranial superficial epigastric or jugular venous blood pressure in the control cows. After IV administration of hydrochlorothiazide (250 mg), acetazolamide (500 mg), or 50% dextrose (500 g), the cranial superficial epigastric and jugular venous blood pressures in cows with udder edema at parturition were not significantly different from those in cows without udder edema at parturition during the 210-minute evaluation period after injection of the drugs.  相似文献

Experience with vascular pathology and vascular surgery, especially with the use of grafts, is limited in horses. Only one case of a venous aneurysm has been described in the horse, but without successful surgical correction. This case report describes the treatment of a 3-year-old female pony presented with an intermittent localised swelling of the left jugular vein after a previous thrombophlebitis. Ultrasonographic examination revealed a large jugular vein pseudoaneurysm. Treatment consisted of a partial aneurysmectomy preserving the healthy medial wall of the jugular vein. An autologous saphenous vein patch technique was used to reconstruct the vein with the largest possible lumen to minimise thrombus formation. Anticoagulants, antimicrobial and anti-inflammatory drugs were administered pre- and post-operatively. Follow-up ultrasound examinations initially showed local narrowing of the graft. At 2 and 8 months post-operatively, no external deformation of the jugular vein was visible and ultrasonography revealed patent blood flow with only a slight difference in lumen diameter. The patch venoplasty proved a viable surgical technique for jugular vein reconstruction resulting in a sufficient lumen, no thrombus formation and a good cosmetic and functional outcome.  相似文献

Sixty cows were studied over 3 days, to determine the effect of site of sampling, handling stress and the tranquilliser xylazine hydrochloride, on blood inorganic phosphorus (P). Jugular venous and coccygeal mixed arterial and venous blood were compared. Saliva was also analysed for P. Buffered and unbuffered trichloracetic acid (TCA) were compared as deproteinising agents. Tail blood P was consistently 12% (range 7-17%) higher than that from the jugular vein (P less than 0.01). This difference decreased when jugular blood was sampled first but increased following administration of the tranquilliser. The drug significantly (P less than 0.01) increased the concentration of P in the saliva on 2 out of 3 days. The possible role of salivary P in affecting jugular P values, is discussed. The buffered TCA analyses gave consistently lower P readings than the unbuffered analyses. The magnitude of this effect was time dependent, being greater when the samples were left for 4 days (P less than 0.05) than when analysed the next day. There was a very strong correlation between plasma and whole blood P. It was concluded that the tissue of choice in phosphorus studies is tail blood (plasma or serum) treated with buffered TCA.  相似文献

Effect of heat stress on early embryonic development in the beef cow   总被引:6,自引:0,他引:6  
Hereford and Hereford X Angus cows (n = 31) were utilized to determine the effects of heat stress on early embryonic development and survival. After acclimation to handling, cows were cannulated via the jugular vein on d 7 and assigned to either a control (C) chamber environment of 22 C, 35% relative humidity (RH) or one of two heat stress treatments. Ambient temperature was maintained at 37 C for 12 h followed by a decrease to 33 C for the remainder of the day in both treatment groups. Relative humidity was maintained at 27% in treatment 1 (TRT 1) and 38% in treatment 2 (TRT 2). On d 8 to 16, daily measurements of respiration rate (RES), rectal temperature (REC) and water intake were taken along with samples of blood, which were analyzed for hematocrit (HEM) and plasma concentration of protein (PP), progesterone (P4), estradiol-17 beta (E2), thyroxine (T4) and glucose (GLU). The uterus was recovered and flushed with saline on d 17 to recover the conceptus and uterine contents. Conceptus (if present) and corpus luteum (CL) wet weight were determined. Cows subjected to TRT 2 had increased RES and REC (P less than .01), while HEM was decreased (P less than .05) compared with C cows. Plasma T4 concentration was decreased (P less than .10) in TRT 2 compared with TRT 1 and cows, while P4 concentration were not significantly different. Catheters were surgically implanted in the carotid artery, jugular vein, and middle uterine vein of 8 cows at 245 days of gestation. Four cows were given 4 mg of estrone/hour via continuous jugular infusion from 0800 hours on day 246 of gestation through 0800 hours on day 250 of gestation; the remaining 4 cows (controls) were given the vehicle for estrone at 10 ml/hour for the same period. Blood samples were collected from the carotid artery every 3 hours during the infusion. Samples were collected hourly from the middle uterine vein from 0 through 8, 54 through 66, and 112 through 120 hours of the infusion periods. After completion of the infusion, corpora lutea were enucleated and blood samples were collected from the carotid artery and uterine vein at hourly intervals for an additional 8 hours. Dispersed cell preparations of the corpora lutea were incubated with and without luteinizing hormone (LH) or dibutyryl cyclic adenosine monophosphate (dbcAMP). Circulating concentrations of unconjugated and conjugated estrone and estradiol-17 beta were higher (P less than 0.05) in the group infused with estrone than in the vehicle-infused group. Mean and base-line concentrations of F series prostaglandins (PGF) for each blood collection period tended to increase (P less than 0.10) during infusion with estrone, but not during infusion with vehicle. After luteectomy, mean and base-line concentrations of PGF also tended (P less than 0.10) to be greater in the estrone-infused cows than in the control cows, but a surge in PGF concentrations due to removal of the ovarian source of progesterone did not develop.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

A cannulation technique was developed to collect blood samples from the ovarian vein of mares over an extended period. Ovarian venous cannulae placed in 4 mares remained patent for a mean (+/- SEM) duration of 36.8 (+/- 6.2) days. During mid-diestrus, concentrations of progesterone in the ovarian vein ipsilateral to the corpus luteum (1,663.8 +/- 238.8 ng/ml) were significantly (P less than 0.001) higher than concentrations measured in paired samples from the jugular vein (6.1 +/- 0.3 ng/ml). Concentration of estradiol in the ovarian vein ranged from a mean of 1,053.2 +/- 303.1 pg/ml during diestrus to 3,353.8 +/- 1,052.8 pg/ml during estrus, whereas values for 74% of samples collected concurrently from the jugular vein were near or below the sensitivity of the assay (10 pg/ml). Results of the study indicate that patent long-term ovarian vein cannulation can be achieved in mares. The cannulation technique provides access to important fundamental information on equine reproductive endocrinology, which to our knowledge, has not been available.  相似文献

Three independent experiments were conducted with cyclic gilts kept under different conditions (2 x 12 on platforms and 100 from production unit), i.e. hormonally unaffected or under the usual conditions of synchronised oestrus (100 mg Suisynchron per animal and die over 20 days followed by 1,000 I.U. PMS and again followed, three days later, by 250 I.U. HCG). Blood samples were repeatedly drawn from the jugular vein and brachiocephalic vein during phases of treatment and oestrus. Acid and alkaline phosphatases were determined immediately in the plasms, and the results were calculated by variance analysis. Synchronisation, using Suisynchron, proved conducive to lowering with significance (P less than 0.01) the levels of acid and alkaline phosphatases. All changes in phosphatase values were not in full conformity within the experiments during the other phases of treatment. Phosphatase levels were not affected by discontinuation of Suisynchron nor by administration of Gn hormone. The levels of acid and alkaline phosphatases in biotechnically treated animals appeared to be lower than those in animals with spontaneous oestrus at the time of full oestrus. While the findings were of period-specific significance (P less than 0.01), the results obtained from the individual animal failed to provide specific information, since gaps were too wide between animals and between animals, on the one hand, and periods, on the other (P less than 0.01).  相似文献

Cranial superficial epigastric (milk vein) and jugular venous blood pressure were measured in 8 cows with udder edema and in 3 normal control cows at least 2 weeks before parturition, at parturition, and 2 weeks after parturition. Cows with udder edema had a significant mean increase (P less than or equal to 0.05) in cranial superficial epigastric venous pressure at parturition when compared with pressures 2 weeks prepartum and 2 weeks postpartum. Unaffected control cows had an insignificant increase in cranial superficial epigastric venous pressure during these 3 periods of measurement. There was an insignificant increase in jugular venous blood pressure during the 3 periods of measurement in affected and control cows. A correlation was observed between mammary blood flow and cranial superficial epigastric blood pressure at parturition (r = -0.659, P less than or equal to 0.05).  相似文献

This study reviewed confirmed cases of concurrent chylothorax and cranial vena caval (CrVC) thrombosis in dogs and cats, and determined predisposing factors for the development of chylothorax associated with CrVC thrombosis. The extent and location of the thrombus, the treatment regime, and the outcome are described. In all 4 cases, implantation of a jugular device was a predisposing factor to thrombosis of the CrVC, and there was extensive thrombosis of the CrVC extending from at least 1 jugular vein to just cranial to the heart. Chylothorax resolved in 3 of the 4 cases after medical and/or surgical intervention. The development of chylothorax concurrently with thrombosis of the CrVC in dogs and cats is likely dependent on the extent and location of the thrombus. Veterinary patients with indwelling jugular devices that develop acute respiratory signs should be assessed for chylothorax associated with thrombosis of the CrVC.  相似文献

This investigation characterized serum concentrations of luteinizing hormone (LH), estradiol-17 beta (E2), progesterone (P4) and cortisol (C) in anestrous sows. Twenty-two sows that had not returned to estrus within 45 days after weaning (anestrous sows), and ten sows that had returned to estrus within seven days following weaning (cyclic sows) were nonsurgically fitted with indwelling jugular vein cannulae. Blood samples were collected at 6 h intervals for seven days and at 15 min intervals for 8 h on the fifth day after cannulation. Serum LH concentrations were determined in all samples, while C, E2 and P4 levels were quantitated in serum collected at 6 h intervals. Serum P4 concentrations in anestrous sows were consistently less than 0.5 ng/mL, and E2 levels ranged from 10 to 19 pg/mL. Concentrations of LH remained less than 1.0 ng/mL in anestrous sows, whereas a preovulatory LH surge was observed in five of ten cyclic sows. There was a circadian rhythm in mean C levels with C peaks occurring at 0600 or 2400 h and nadir levels observed at 1200 and 1800 h. Few differences in C levels were detected between anestrous and cyclic sows. It was evident that anestrous sows did not exhibit cyclic or predictable variations in steroid hormone concentrations. Unfortunately, the results of this study failed to elucidate the endocrine pathogenesis of the anestrous sow.  相似文献

The secretion of ovine lymphocyte suppressor factor from jugular vein (JV), uterine vein (UV), and curetted hemopoietic uterine luminal (UL) mononuclear cells was evaluated on d 14 of the cycle, following ovariectomy (OVX) and after 14 d of progesterone injections (OVX + P4, 1 mg/kg BW). Mononuclear cells (predominantly lymphocytes) were harvested by Ficoll-Paque and placed into culture (2.5 x 10(6).mL-1.well-1 in RPMI-1640). Cellular supernatants were obtained at 72 h and volumes (5 to 50 microL) were tested for suppression of phytohemagglutinin (PHA [.08 microgram l)-treated peripheral blood lymphocytes (1.0 x 10(5)). In a concurrent experiment, PHA-treated JV, UV, and UL cells (1.0 x 10(5)) were cultured singly and JV cells (1.0 x 10(5)) were also cocultured with each of 1.0 x 10(5), 5.0 x 10(4), and 2.5 x 10(4) UV and UL cells. The incorporation of thymidine into DNA was quantified at 60 h. For the cellular supernatant experiment, thymidine incorporation was affected by reproductive phase (P less than .036), lymphocyte source (P less than .0001), and phase x source (P less than .004). For UL cells, the degree of suppressor activity follows: d 14 greater than OVX greater than OVX+P4 (P less than .05). The UL supernatant from OVX+P4-treated ewes and supernatants of JV and UV cells, irrespective of reproductive phase, lacked suppressor activity. Sephacryl S-200 chromatography revealed that UL supernatant from d-14 ewes contained a greater than or equal to 248,000 molecular weight suppressor macromolecule. Holstein heifers (n = 21) were balanced across sires and assigned to three service-sire-breed groups in which heifers were inseminated artificially to either purebred Angus (n = 7), Holstein (n = 7) or Brahman (n = 7) bulls. Semen from four bulls was used for each service sire-breed group. Blood samples were collected from a jugular vein thrice weekly from d 160 to 265 of pregnancy, daily thereafter until 15 d postpartum, and then thrice weekly until d 60 postpartum. Concentrations of progesterone, estrone, estradiol, and estrone sulfate from 23 d prepartum to parturition, and of 15-keto-13,14-dihydro-prostaglandin F2 alpha (PGFM) from 2 d prepartum to d 15 postpartum were measured by radioimmunoassay. Heifers within the Brahman-service-sire group had longer gestations (P less than .05) than those of Holstein- or Angus-service-sire groups (285.0 vs 278.7), 279.0 d). Calf birth weight was lower (P less than .05) in Angus- than Holstein- and Brahman-service-sire groups (30.6 vs 36.1, 43.4 kg). Daily trends of prepartum maternal progesterone concentrations were approximately 1 ng/ml lower (P less than .01) in Angus- than Holstein- or Brahman-service-sire groups until luteolysis occurred. Heifers bearing crossbred Angus conceptuses had lower daily trends of prepartum estrogens concentrations (P less than .01), whereas heifers of the Holstein- and, even more dramatically, of the Brahman-service-sire groups had a higher magnitude and greater rise of plasma estrogens concentrations between d -10 and -1 prepartum (less than .01). Postpartum mean concentrations (P less than .05) and response curves of PGFM were lower (P less than .01) in the Angus- than in the Holstein- or Brahman-service-sire groups. Calf birth weights were correlated with least-squares means for maternal concentrations of prepartum estrone (r = .57), estradiol (r = .59) and estrone sulfate (r = .64) and postpartum maternal concentrations of PGFM (r = .56). Functional responses of the conceptus (e.g., estrogens) and maternal units (e.g., progesterone and PGFM) were influenced by conceptus genotype during the periparturient period.  相似文献

A study was conducted to determine the effect of social crowding on the development of endocrine organs involved in the reproductive process in growing and finishing gilts. One hundred four littermate gilts weighing approximately 30 kg were randomly assigned to treatment groups of either 8 or 16 pigs/pen. Gilts raised in pens of eight were allotted 1.06 m2 per pig from 30 to 65 kg and 1.25 m2 from 65 to 100 kg; whereas, one-half the area was provided for gilts raised in pens of 16. Littermate gilts raised in pens of 8 and 16 were slaughtered on the same day when the average weight of pigs reached 100 kg. At slaughter, the brain, adrenals, pituitary, uterus and ovaries were removed and weighted. The brain was lyophilyzed and reweighed to yield dry brain weight. Gilts raised in the low-density groups had heavier adrenal (P less than .01), pituitary (P less than .08), brain (P less than .11), dry-brain (P less than .02), uterine (P less than .09) and ovarian (P less than .07) tissues. Furthermore, significantly more gilts in the low-density, compared with the high-density groups, had ovulated at slaughter. In a second study, eight littermate gilts raised in pens of 8 or 16 and weighing approximately 100 kg were moved to individual pens and fitted with a jugular vein catheter. Following blood sample collection at 15-min intervals for 2 h, 200 IU of adrenocorticotropin (ACTH) were infused into the cannula. Blood samples were collected at 15-min intervals for an additional 2 h. OBJECTIVE: To quantify glutamine use in viscera drained by the portal vein in neonatal calves and to assess the relative nutritional importance of glutamine, glucose, and acetate for enterocytes. ANIMALS: 5 healthy neonatal calves. PROCEDURE: A femoral artery, jugular vein, and the portal vein were surgically cannulated in each calf. Blood flow in the portal vein was measured by use of an ultrasonographic transit-time flow probe. A series of solutions was infused on 4 days for each calf. On the infusion days, acetate, glucose, glutamine, and saline (0.9% NaCl; control) solutions were administered IV during 1-hour periods via the jugular vein. Venous and arterial blood samples were collected during the last 15 minutes of each 1-hour infusion. RESULTS: Uptake of glutamine and glucose by viscera drained by the portal vein was 0.3+/-1.1 and 1.9+/-3.1 micromol/kg0.75/min, respectively, during saline infusion. During acetate, glucose, and saline infusions, glucose was a greater source of energy for the intestines than was glutamine. However, during glutamine infusion, uptake of glutamine by viscera drained by the portal vein increased significantly (29.9+/-11.2 micromol/kg0.75/min), which was associated with an increase in ammonia production (7.0+/-0.5 micromol/kg0.75/min). Toxicosis was not associated with IV administration of glutamine. CONCLUSION: Glutamine infusion resulted in an increase in glutamine uptake by viscera drained by the portal vein, which was associated with an increase in ammonia production and a slight increase in oxygen consumption. CLINICAL RELEVANCE: These solutions may be used to develop treatments that enhance healing of intestines of diarrheic calves.  相似文献

Udder oedema was studied in eight Jersey cows with rectovaginal constriction (RVC) and three normal Jersey cows. A complete physical examination was performed at least two weeks before parturition, at parturition and two weeks after parturition. Jersey cows with RVC developed udder oedema as early as 14 days before parturition, and condition persisted for three to 10 days following parturition. Mammary blood flow and cranial superficial epigastric venous blood pressure were measured in three Jersey cows with RVC with udder oedema and three normal Jersey cows. The former with udder oedema had a significant decrease in mammary blood flow at parturition compared to the normal Jersey cows (P less than 0.05); there was no significant difference in mammary blood flow between Jersey cows with RVC and normal Jersey cows two weeks before parturition (P greater than 0.05). A significant mean increase in cranial superficial epigastric venous blood pressure occurred at parturition when compared to two weeks before parturition in Jersey cows with RVC (P less than 0.05), but normal Jersey cows had no significant increase in cranial superficial epigastric vein pressure during these two periods of measurement (P greater than 0.05). There was no significant increase in jugular vein blood pressure during the two periods of measurement in Jersey cows with RVC or normal Jersey cows (P greater than 0.05). There was a significant negative correlation between mammary blood flow and cranial superficial epigastric pressure at parturition (r = 0.733, P less than 0.05).  相似文献

The present study was designed to examine the usefulness of cannulation of the caudal caval vein through the femoral vein for the measurement of hormone concentrations in the reproductive tract in the pig. The experiment was performed on sexually pubertal gilts (Polish Large White x Polish Landrace) of a similar age (7-8 months) and body mass (100-110 kg) after two controlled subsequent estrous cycles. Six gilts in the luteal phase (10th day) of the estrous cycle were used in this experiment. The animals were subjected to a surgical procedure which included:--premedication (Combelen, i.m. 1 ml/10 kg of body mass) and than after 20-30 min general anaesthesia (Vetbutal, i.v., dose 30-40 ml) according to body mass and the symptoms observed,--insertion of cannulas (o.d. 2.2 and i.d. 1.8 mm)--one into the jugular vein and the other into the caudal caval vein through the femoral vein. In several gilts the cannulas were inserted into the caudal caval vein to a depth of 14, 18, 20, 23, 25 and 30 cm from the femoral ring. The concentrations of progesterone (P4) and testosterone (T) were analysed in samples of blood plasma from the jugular and caudal caval veins by radioimmunoassay (RIA). The largest differences in hormone concentrations between the jugular and caudal caval veins were ascertained when a cannula was inserted into the caudal caval vein to a depth of 20 cm from the femoral ring. In other cases the differences were less prominent, or no differences were observed (e.g. 14 cm for progesterone and testosterone or 18 cm for testosterone).  相似文献

This study determined if the insulin and glucose responses to glucose infusion in obese (n = 4) and lean (n = 4) Holstein heifers were affected by stage of the estrous cycle. Glucose (.35 g/kg) was infused within 2 min into the jugular veins of heifers during diestrus (d 15) and at the subsequent estrus (d 0). Concentrations of insulin and glucose were determined in jugular venous serum obtained from blood samples collected at 60, 45, 30, 15 and 1 min before and at 2.5, 5, 10, 15, 20, 30, 40, 50, 60, 80, 100, 120, 140, 160, 180, 210 and 240 min after glucose. Mean (+/- SE) pretreatment concentrations of glucose (mg/100 ml) in obese (68 +/- 1.9) and lean (71 +/- 2.5) heifers were unaffected by body condition and stage of the cycle. Mean (+/- SE) pretreatment concentrations of insulin (microU/ml) were unaffected by stage of the cycle but were higher (P less than .05) in obese (33 +/- 3.6) than in lean (18 +/- 2.7) heifers. Body condition affected the insulin response with greater absolute concentrations (P less than .01) and total (P less than .005) response areas of insulin in obese than in lean heifers. OBJECTIVE: To characterize the variation in plasma lactate concentration among samples from commonly used blood sampling sites in conscious, healthy dogs. ANIMALS: 60 healthy dogs. PROCEDURE: Cross-sectional study using a replicated Latin square design. Each dog was assigned to 1 of 6 groups (n = 10) representing all possible orders for 3 sites (cephalic vein, jugular vein, and femoral artery) used to obtain blood. Samples were analyzed immediately, by use of direct amperometry for pH, PO2, Pco2, glucose, and lactate concentration. RESULTS: Significant differences in plasma lactate concentrations were detected among blood samples from the cephalic vein (highest), femoral artery, and jugular vein (lowest). Mean plasma lactate concentration in the first sample obtained, irrespective of sampling site, was lower than in subsequent samples. Covariation was identified among plasma lactate concentration, pH, and PCO2, but correlation coefficients were low. CONCLUSIONS AND CLINICAL RELEVANCE: Plasma lactate concentrations differed among blood samples from various sites. A reference range for plasma lactate concentration was 0.3 to 2.5 mmol/L. Differences in plasma lactate concentrations among samples from various sites and with repeated sampling, in healthy dogs, are small. Use of the reference range may facilitate the clinical use of plasma lactate concentration in dogs.  相似文献
