Adsorption of hesperidin from aqueous solutions on styrene-divinylbenzene (SDVB) and acrylic resins was investigated at different pH values (1.5-5.5) and temperatures (10-40 degrees C). Adsorption was not affected by pH variation, whereas it increased on increasing temperature for the SDVB resins and remained substantially unchanged for the acrylic ones. The different behavior of the two types of resins was ascribed to the different wetting, because of their hydrophobic or partially hydrophilic properties, respectively. The equilibrium data at 20 degrees C were determined on 13 commercial resins displaying a wide range of surface areas (S(A), 330-1200 m(2)/g) and pore radii (P(R), 20-260 A). Data were well fitted to the Freundlich isotherm, and its parameters were used to compare the adsorption capacity of different resins. The most effective resin is a SDVB copolymer with the largest S(A) (1200 m(2)/g) and an intermediate P(R) (90 A). The Freundlich constants (K(F)) were rationalized in terms of a two parameter equation, including S(A) and P(R) as independent variables. The adsorption constant increased on increasing both S(A) and P(R) for the resins having P(R) = 90 A; beyond this limit, the contribution of S(A) was predominant and that of P(R) was slightly negative. 相似文献

Caffeic acid o-quinone (CQ) was prepared by oxidation of caffeic acid with o-chloranil in organic media. The reaction between the purified CQ and cyanidin 3-glucoside (Cy 3-glc, o-diphenolic anthocyanin) was monitored by HPLC, and quantitative analyses were performed to establish the stoichiometry of the reaction. The results indicate that Cy 3-glc is degraded by a coupled oxidation mechanism with integration of CQ into the degradation products. The ratio of degraded Cy 3-glc to CQ incorporated into the condensation products was approximately 2.0. No brown products could be detected, only a slight orange color. Moreover, the addition of purified polyphenol oxidase to the slightly colored media resulted in the disappearance of the caffeic acid formed from the reaction of coupled oxidation (Cy 3-glc/CQ) and the formation of brown polymers. The degradation products were isolated by gel filtration on Sephadex G-25. The UV-vis spectra and chemical analysis (acidic hydrolysis) of the degradation products suggest that they resulted from the condensation of caffeic acid and Cy 3-glc. At present, little is known about the mechanisms responsible for intestinal absorption of anthocyanins (ACNs). For example, it has not yet been established if ACNs are absorbed through an active transport mechanism, such as the sodium-dependent glucose transporter (SGLT1), or by passive diffusion. Previously, we found that the absorption of ACNs differs between regions of the digestive tract and is maximal in the jejunum, suggesting that an active transport mechanism is involved. In the present study, we examined the effect of d-glucose (main substrate of SGLT1), phloridzin (inhibitor of SGLT1), and quercetin-3-glucose (Q3G, a flavonol) on the absorption of cyanidin-3-glucoside (C3G; approximately 5 micromol/L) by mouse jejunum mounted in Ussing chambers. We found that the presence of either D-glucose (10, 20, and 40 mmol/L) or phloridzin (50, 100, and 200 micromol/L) resulted in a small but insignificant inhibition of C3G disappearance from the mucosal solution (decrease of disappearance with glucose, 33%; with phloridzin, 18%; NS). However, when the flavonol Q3G (50 micromol/L) was added to the mucosal solution together with the C3G, the disappearance of C3G was significantly decreased (74%; p < 0.001), and Q3G disappeared instead. In addition, we found phloretin and quercetin, the aglycones of phloridzin and Q3G, respectively, present in the mucosal solution and tissue extracts, indicating hydrolysis of these compounds by the enterocytes of the jejunum. In contrast, the aglycone cyanidin was not detected at all. Our results show that in the mouse small intestine, ACN absorption is not solely dependent on the activity of the SGLT1 transporter, as d-glucose and phloridzin had only a slight effect on uptake. Q3G, however, clearly inhibited C3G disappearance. These results suggest that there might be a competitive inhibition between C3G and Q3G absorption. It is possible that an absorption mechanism other than the SGLT1 is involved, which has a structural preference toward flavonols. 相似文献

Malvidin-3-glucoside has been labeled by enzymatic synthesis in a single-step experiment. Catechol-O-methyl transferase catalyzed the B-ring methylation of petunidin-3-glucoside, and S-Adenosyl-L-[methyl-(3)H] methionine was the methyl donor. Solid phase extraction and preparative high-performance liquid chromatography were necessary to separate [3'-O-methyl-(3)H]malvidin-3-glucoside from an isomer and the starting material. The specific activity was 2.2 Ci mmol(-1), and the yield of incorporation was 1.1%. A possible application of the labeled material is the study of anthocyanin reactions in complex mixtures such as red wine where products are difficult to isolate and analyze. 相似文献

For better understanding of the physiological function of anthocyanins, the absorption and metabolism of cyanidin 3-O-beta-D-glucopyranoside (Cy3G), which is one of the major anthocyanins in colored food materials, were precisely investigated. Combining two modalities newly developed, that is, highly sensitive semi-micro-HPLC and vein cannulation, Cy3G and its four major metabolites (M1-M4) were detected in the blood plasma of rats after oral administration of Cy3G (100 mg/kg of body mass). The plasma concentration of Cy3G reached its maximum at 15 min after the ingestion. Metabolite 2 (M2) and metabolite 3 (M3) showed their maximum plasma levels at 15 and 30 min, respectively, whereas metabolite 1 (M1) and metabolite 4 (M4) showed their maximum levels at 60 and 120 min, respectively. The maximum plasma concentrations of the four metabolites were in the following order: M3 (21 nM) > M4 (20 nM) > M1 (8.5 nM) > M2 (5 nM). When Cy3G was directly injected into the neck vein, only M2 and M3 were detected in the plasma, indicating that both M1 and M4 were produced during absorption from the gastrointestinal tract. Tandem MS analysis of the metabolites showed that M2 and M3 were monomethylated Cy3G, while M1 and M4 were glucuronides of Cy and methylated Cy, respectively. M3 was assigned as peonidin 3-O-beta-D-glucopyranoside (Pn3G) from the comparison of the retention time of authentic Pn3G. 相似文献

Anthocyanins have been suggested to improve visual functions. This study examined the effect of four anthocyanins in black currant fruits on the regeneration of rhodopsin using frog rod outer segment (ROS) membranes. Cyanidin 3-glycosides, glucoside and rutinoside, stimulated the regeneration, but the corresponding delphinidins showed no significant effect. The formation of a regeneration intermediate was suggested to be accelerated by cyanidin 3-rutinoside. Their effects on the cGMP-phosphodiesterase activity in the ROS membranes were also investigated but found to be negligible. It was concluded that the major effect of anthocyanins in rod photoreceptors is on the regeneration of rhodopsin. 相似文献

The effectiveness of seven phenolic compounds (catechin, epicatechin, procyanidin B2, caffeic acid, p-coumaric acid, myricitrin, and quercitrin) as copigments of malvidin 3-glucoside, the major anthocyanin in red wines from Vitis vinifera, using a copigment/pigment molar ratio of 1:1 was assayed in model wine solutions under the same conditions (pH=3.6, 12% ethanol). The stability of the copigment-pigment complexes formed was studied during a storage period of 60 days at 25 degrees C. Tristimulus colorimetry was applied for color characterization of the copigmentation process, and HPLC-DAD-MS was used to monitor changes in the composition of the samples. Copigmentation has been found to occur in all cases despite the low copigment/pigment molar ratio used, although the effect was different depending on the compound. Flavan-3-ols appeared as the less effective copigments, procyanidin B2 being even worse than monomeric flavanols, whereas flavonols behaved as the best ones. These latter copigments also induced the most statistically significant bathochromic shift in lambdamax. In the colorimetric analysis, it was observed that the lightness L* of the copigmented solutions increased with time, chroma C*ab decreased, and the hue hab increased. The copigments that produced a greater increase in the hue angle were the monomeric flavan-3-ols, which therefore showed to be the least protective cofactors against browning of the anthocyanin solution during the storage. With the time of storage, the formation of new pigments was observed in the solutions containing flavanols (xanthylium structures) and hydroxycinnamic acids (pyranoanthocyanins), which explains some of the color modifications produced in these solutions. Another relevant observation was that the stability of the anthocyanin was not much improved by most of the assayed copigments, since quite similar degradation rates were observed in the presence and absence of those cofactors. 相似文献

Anthocyanin constituents in black raspberries (Rubus occidentalis L.) were investigated by HPLC-DAD, and their involvement as potent, significant antioxidants in black raspberries was demonstrated by three common antioxidant assays (FRAP, DPPH, ABTS) in this study. Five anthocyanins were present in black raspberries: cyanidin 3-sambubioside, cyanidin 3-glucoside, cyanidin 3-xylosylrutinoside, cyanidin 3-rutinoside, and pelargonidin 3-rutinoside. Their identities and structures, with particular emphasis on cyanidin 3-xylosylrutinoside, were confirmed by NMR spectroscopy. Two of these anthocyanins, cyanidin 3-rutinoside and cyanidin 3-xylosylrutinoside, predominated, comprising 24-40 and 49-58%, respectively, of the total anthocyanins in black raspberries. On the basis of both potency and concentration, cyanidin 3-rutinoside and cyanidin 3-xylosylrutinoside were found to be the significant contributors to the antioxidant systems of black raspberries. These findings indicate that these two anthocyanin compounds may function as the primary phenolic antioxidants in black raspberries. These two compounds exhibit potential biological activities that may be exploited in conjunction with other naturally occurring bioactive compounds in black raspberry fruit-based products used in clinical trials for the treatment of various types of cancer. 相似文献

The reaction between (+)-catechin and cyanidin 3-O-glucoside was investigated in the presence of furfural and 5-(hydroxymethyl)furfural using LC/DAD and LC/MS analysis, and the obtained results were compared with those recorded with malvidin 3-O-glucoside. The appearance of colorless and red and yellow compounds was observed showing that the two polyphenols competed in the condensation process with a predominant formation of the reddish adducts. The colored compounds formed in the case of cyanidin 3-O-glucoside seemed to be more stable than those formed when the reaction was conducted with malvidin 3-O-glucoside. The detection of these reddish and yellowish compounds constitutes a new support for the contribution of this kind of reaction in the color evolution of fruit-derived beverages. In addition, other unidentified compounds were also detected, showing the occurrence of other interaction pathways in addition to the polymerization process yielding oligomeric bridged derivatives and opening perspectives of further investigations of these model solutions. 相似文献

We previously determined that five rather hydrophobic metabolites appeared in blood plasma after oral administration of cyanidin 3-O-beta-D-glucopyranoside, but a group of hydrophilic metabolites still remained unidentified. In the present study, 12 hydrophilic metabolites found were collected from urine and plasma samples by high-performance liquid chromatography (HPLC) and then analyzed by tandem MS spectrometry. From the MS spectra, four metabolites out of 12 were assigned as glucuronides of cyanidin 3-O-beta-D-glucopyranoside and six out of 12 were glucuronides of the primary metabolites of cyanidin 3-O-beta-D-glucopyranoside (O-methyl cyanidin 3-O-beta-D-glucopyranoside). Extended glucuronides of cyanidin 3-O-beta-D-glucopyranoside and O-methyl cyanidin 3-O-beta-D-glucopyranoside showed their maximum plasma concentrations at 15 and 60 min (or 30 min) after oral administration, respectively. Their maximum plasma concentrations ranged from 15 to 70 nM. From the profile of urinary-excreted anthocyanins after intravenous administration, it was deduced that extended glucuronides of cyanidin 3-O-beta-D-glucopyranoside and O-methyl cyanidin 3-O-beta-D-glucopyranoside were mainly produced in the liver rather than by intestinal flora. The area under the plasma concentration curve was 0.25 micromol min/L for extended glucuronides of cyanidin 3-O-beta-D-glucopyranoside and 0.14 micromol min/L for O-methyl cyanidin 3-O-beta-D-glucopyranoside, respectively, when evaluated as cyanidin 3-O-beta-D-glucopyranoside equivalent, indicating that extended glucuronidation is a critical pathway in cyanidin 3-O-beta-D-glucopyranoside metabolism in rats. 相似文献

Hydroxyl radicals (.OH) seem to have an important role in the oxidation of wine constituents and the production of important electrophilic aldehydes and ketones. In this experiment, glyceraldehyde, a .OH oxidation product of glycerol, recently described in wine, reacts with (+)-catechin, (-)-epicatechin, and malvidin-3-glucoside (Mv3gl), in model solutions, yielding new condensed phenolic compounds. The adduct compounds formed were separated by means of reversed phase liquid chromatography and detected and characterized using UV-vis and electrospray ionization mass spectrometry. Flavanol-flavanol and anthocyanin-flavanol adducts linked with glyceraldehyde yielded compounds with m/z ratios for their main ions, in positive ion mode, of 653.2 for the (+)-catechin dimer or the (-)-epicatechin dimer and 855.5 for Mv3gl/(+)-catechin or Mv3gl/(-)-epicatechin dimers. The possible occurrence of these compounds in wine is suggested, and the potential role of these and related reactions in wine aging is discussed. 相似文献

为了选择紫甘薯色素粗提液纯化性能较好的树脂,采用AB-8型、S-8型、NKA-II型和NKA-9型4种大孔树脂对紫甘薯色素进行了吸附与解吸试验,研究了大孔树脂对紫甘薯色素的静态吸附动力学曲线、吸附等温曲线及解吸特性。结果表明:在4种树脂中,AB-8型树脂吸附平衡的速率常数为最大,对紫甘薯色素的吸附量和解吸量分别是S-8、NKA-II及NKA-9的119%,139%,118%和105%,116%,118%,因而AB-8型大孔树脂是用于纯化紫甘薯色素较为合适的吸附剂,解吸时宜选用60%乙醇溶液。 相似文献

The efficient purification method of high-purity flavonoids from Hippophae rhamnoides L. (sea buckthorn) is reported. A novel room temperature ionic liquid-based macroporous adsorption resin (MAR), N-methylimidazole/MARs (Mim/MARs), was prepared on the basis of the Friedel-Crafts-catalyzed and surface-modified technique. The material exhibited favorable characteristics for adsorption application, including high pore volume (1.90 cm(3)/g, 3 times as big as the optimal commercial adsorbent), good pore structure (type IV isotherm with an H1 hysteresis loop, the most favorable structure for adsorption purposes), narrow particle size and pore size distribution (1.2 mm with a standard deviation of 0.106 mm), and excellent chemical stability. This paper also presents the first experimental evidence that the functional groups of the modified materials and composite action of certain molecular interactions between the adsorbent and flavonoids affected the adsorption process. Moreover, a new sphere-size adsorption kinetics model in which the adsorption process contained three or more compartments and detailed parameters of sphere size was developed according to the multicompartment kinetics model and Karichhoff's theory by investigating the regression of the experimental results. The conclusion that the first compartment of the adsorption process onto Mim/MARs mainly occurred on spheres larger than 0.83 mm and the second and third ones mainly occurred on spheres of 0.46-0.83 and 0.22-0.46 mm, respectively, was drawn from this new sphere-size adsorption kinetics model. 相似文献

Fusarium toxins, Alternaria toxins, and ergot alkaloids represent common groups of mycotoxins that can be found in cereals grown under temperate climatic conditions. Because most of them are chemically and thermally stable, these toxic fungal secondary metabolites might be transferred from grains into the final products. To get information on the commensurate contamination of various cereal-based products collected from the Czech retail market in 2010, the occurrence of "traditional" mycotoxins such as groups of A and B trichothecenes and zearalenone, less routinely determined Alternaria toxins (alternariol, alternariol monomethyl ether and altenuene), ergot alkaloids (ergosine, ergocryptine, ergocristine, and ergocornine) and "emerging" mycotoxins (enniatins A, A1, B, and B1 and beauvericin) were monitored. In a total 116 samples derived from white flour and mixed flour, breakfast cereals, snacks, and flour, only trichothecenes A and B and enniatins were found. Deoxynivalenol was detected in 75% of samples with concentrations ranging from 13 to 594 μg/kg, but its masked form, deoxynivalenol-3-β-d-glucoside, has an even higher incidence of 80% of samples, and concentrations ranging between 5 and 72 μg/kg were detected. Nivalenol was found only in three samples at levels of 30 μg/kg. For enniatins, all of the samples investigated were contaminated with at least one of four target enniatins. Enniatin A was detected in 97% of samples (concentration range of 20-2532 μg/kg) followed by enniatin B with an incidence in 91% of the samples (concentration range of 13-941 μg/kg) and enniatin B1 with an incidence of 80% in the samples tested (concentration range of 8-785 μg/kg). Increased oxidative stress and apoptosis were detected in atherosclerotic lesions. Oxidized low-density lipoprotein (oLDL) may induce oxidative stress and apoptosis via multiple pathways in vascular endothelial cells (EC). Delphinidin-3-glucoside (D3G), an anthocyanidin glycan enriched in dark-skin berries, may neutralize those effects of oLDL in EC. The present study demonstrated that oLDL increased the generation of intracellular NADPH-dependent superoxide and impaired redox status in cultured porcine aortic EC (PAEC). The activities of mitochondrial respiratory chain complex I-IV and the contents of NADH dehydrogenase (ND)1, ND6 (complex I enzyme subunits), or cytochrome b (complex III enzyme subunit) were significantly reduced in PAEC treated with oLDL compared to controls. Treatment with oLDL significantly increased the abundances of NADPH oxidase (NOX)2, NOX4, and p22phox in PAEC. oLDL reduced cell viability and the protein content of B-cell lymphoma (Bcl)-2, but increased the content of caspase 3 in PAEC. Co-treatment with D3G prevented oLDL-induced increases in intracellular superoxide or in the protein content of NOX2, NOX4, p22phox, or caspase 3, inhibited the impairment of redox statues or cell viability, and prevented the attenuation of mitochondrial enzyme activities and the reductions of Bcl-2, ND1, or cytochrome b contents in PAEC. The findings suggest that oLDL induced oxidative stress and apoptosis in EC, which was associated with the activation of NOX, the impairment of mitochondrial respiration chain enzymes, and the disorder of key regulators for apoptosis. D3G neutralized the harmful effects of oLDL on oxidative stress, mitochondrial dysfunction, and apoptosis in cultured vascular EC. 相似文献

The co-occurrence of the major Fusarium mycotoxin deoxynivalenol (DON) and its conjugate deoxynivalenol-3-glucoside (DON-3-Glc) has been documented in infected wheat. This study reports on the fate of this masked DON within milling and baking technologies for the first time and compares its levels with those of the free parent toxin. The fractionation of DON-3-Glc and DON in milling fractions was similar, tested white flours contained only approximately 60% of their content in unprocessed wheat grains. No substantial changes of both target analytes occurred during the dough preparation process, i.e. kneading, fermentation, and proofing. However, when bakery improvers enzymes mixtures were employed as a dough ingredient, a distinct increase up to 145% of conjugated DON-3-Glc occurred in fermented dough. Some decrease of both DON-3-Glc and DON (10 and 13%, respectively, compared to fermented dough) took place during baking. Thermal degradation products of DON, namely norDON A, B, C, D, and DON-lactone were detected in roasted wheat samples and baked bread samples by means of UPLC-Orbitrap MS. Moreover, thermal degradation products derived from DON-3-Glc were detected and tentatively identified in heat-treated contaminated wheat and bread based on accurate mass measurement performed under the ultrahigh mass resolving power. These products, originating from DON-3-Glc through de-epoxidation and other structural changes in the seskviterpene cycle, were named norDON-3-Glc A, B, C, D, and DON-3-Glc-lactone analogically to DON degradation products. The degradation mechanism of cyanidin 3-rutinoside in the presence of (-)-epicatechin and litchi pericarp polyphenol oxidase (PPO) was investigated using several model systems. The enzymically generated (-)-epicatechin o-quinone could induce cyanidin 3-rutinoside degradation. The results obtained in this study allowed us to propose a pathway for cyanidin 3-rutinoside degradation in the presence of (-)-epicatechin and litchi pericarp PPO. First, enzymatic oxidation of (-)-epicatechin produced the corresponding o-quinone, and then cyanidin 3-rutinoside and (-)-epicatechin competed for (-)-epicatechin o-quinone, resulting in degradation of cyanidin 3-rutinoside and regeneration of (-)-epicatechin. Moreover, the results of kinetic studies indicated this competition was influenced by both (-)-epicatechin concentration and cyanidin 3-rutinoside concentration in the model system. 相似文献

The bioactive anthocyanins present in tart cherries, Prunus cerasus L. (Rosaceae) cv. Balaton, are cyanidin 3-glucosylrutinoside (1), cyanidin 3-rutinoside (2), and cyanidin 3-glucoside (3). Cyanidin (4) is the major anthocyanidin in tart cherries. In our continued evaluation of the in vivo and in vitro efficacy of these anthocyanins to prevent inflammation and colon cancer, we have added these compounds to McCoy's 5A medium in an effort to identify their degradation products during in vitro cell culture studies. This resulted in the isolation and characterization of protocatechuic acid (5), the predominant degradation product. In addition, 2,4-dihydroxybenzoic acid (6) and 2,4,6-trihydroxybenzoic acid (7) were identified as degradation products. However, these degradation products were not quantified. Compounds 5-7 were also identified as degradation products when anthocyanins were subjected to varying pH and thermal conditions. In cyclooxygenase (COX)-I and -II enzyme inhibitory assays, compounds 5-7 did not show significant activities when compared to the NSAIDs Naproxen, Celebrex, and Vioxx, or Ibuprofen, at 50 microM concentrations. However, at a test concentration of 50 microM, the antioxidant activity of protocatechuic acid (5) was comparable to those of the commercial antioxidants tert-butylhydroquinone (TBHQ), butylated hydroxytoluene (BHT), and butylated hydroxyanisole (BHA), and superior to that of vitamin E at 10 microM concentrations. 相似文献

The adsorption characteristics of cadmium on bed sediments ofriver Hindon in western Uttar Pradesh (India) have been studied.The effect of various operating variables, viz., initial concentration, solution pH, sediment dose, contact time, particlesize and temperature has been studied. The optimum equilibrationtime was found to be 60 min, which was independent of initialconcentration of cadmium ions. The adsorption curves were smoothand continuous leading to saturation, suggesting the possible monolayer coverage of cadmium ions on the surface of the adsorbent. The adsorption of cadmium increased with an increasein pH. Furthermore the adsorption of cadmium increased with increasing adsorbent concentration and decreased with adsorbentparticle size. The important geochemical phases, iron and manganese oxide, support the adsorption of cadmium ions. The adsorption data were analysed using the Langmuir and Freundlichadsorption models to determine the mechanistic parameters relatedto the adsorption process. Thermodynamic parameters, viz., freeenergy change, enthalpy change and entropy change, were alsodetermined. The negative values of free energy change (ΔG°) indicated the spontaneous nature ofthe adsorption and positive values of enthalpy change (ΔG°) suggested the endothermic nature ofthe adsorption process. The intraparticle diffusion of cadmium through pores in the adsorbent was found be the main rate-limiting step. 相似文献
