Two adult female rock hyraxes (Procavia capensis) at the Dallas Zoo were confirmed with spontaneous diabetes mellitus from 1997-2000, whereas a third animal with a similar clinical presentation never became hyperglycemic. The pancreas in all three animals showed pancreatic islet fibrosis (PIF). Retrospective examination of medical records for rock hyraxes acquired by this collection or born into it from 1991-2002 identified eight more animals affected with PIE All affected animals, including three males and eight females, were 1-7 yr of age and presented either with vague clinical signs of soft feces and rough hair coat or were acutely moribund or dead. Clinical pathology data was available for seven of the animals before onset of overt clinical signs and revealed inappropriate hyperglycemia in six, as well as elevated serum concentrations of creatine phosphokinase, amylase, and lipase in all seven animals. Pedigree evaluation did not support a familial pattern for PIE Review of the histopathology findings from nine other zoologic collections with rock hyrax deaths during the study period identified six institutions with 12 additional cases genetically unrelated to the incident collection. Histopathology and viral serology did not support an infectious cause. Analysis of serum anti-islet and anti-insulin antibodies did not suggest autoimmune disease, and none of the animals had known exposure to toxic substances. Limited nutritional analyses did not support a nutritional basis for the condition, and the cause for PIF remains unknown.  相似文献

Normal serum biochemistry values are frequently obtained from studies of captive sedentary (zoo) or free-ranging (wild) animals. It is frequently assumed that values obtained from these two populations are directly referable to each other. We tested this assumption using 20 captive gray wolves (Canis lupus) in Minnesota, USA, and 11 free-ranging gray wolves in Alaska, USA. Free-ranging wolves had significantly (P < 0.05) lower sodium, chloride, and creatinine concentrations and significantly higher potassium and blood urea nitrogen (BUN) concentrations; BUN to creatinine ratios; and alanine aminotransferase, aspartate aminotransferase, and creatine kinase activities relative to captive wolves. Corticosteroid-induced alkaline phosphatase activity (a marker of stress in domestic dogs) was detected in 3 of 11 free-ranging wolves and in 0 of 20 captive wolves (P = 0.037). This study provides clear evidence that serum biochemical differences can exist between captive and free-ranging populations of one species. Accordingly, evaluation of the health status of an animal should incorporate an understanding of the potential confounding effect that nutrition, activity level, and environmental stress could have on the factor(s) being measured.  相似文献

The aim of this study was to investigate the form and structure of the different parts of the gastrointestinal tract of the hyraxes and to ascertain whether some parts show typical genus-dependent differences in one or the other of the three hyrax genera. We also tried tO homologize particularly the ascending colon of the hyraxes with its unpaired and paired diverticula with that of the domestic mammals. A Mycobacterium tuberculosis complex organism was isolated from a zoo resident rock hyrax (Procavia capensis) imported into Canada from South Africa. The strain was identified biochemically as Mycobacterium microti. The spoligotype pattern obtained for this isolate was found to be rare. This represents the first report of isolation and spoligotyping of M. microti in North America.  相似文献

The differences between the capture stress responses of captive and free-ranging roe deer (Capreolus capreolus) and their modulation by acepromazine, during a period of three hours' physical restraint after capture in drive-nets, were examined in 16 free-ranging and 16 captive roe deer. Eight of the free-ranging and eight of the captive animals received acepromazine intramuscularly, and the other eight free-ranging and eight captive deer received the same volume of saline. Heart rate, body temperature and haematological and serum biochemical parameters were analysed. In the groups treated with acepromazine, the heart rate stabilised sooner, and the red blood cell (RBC) count, haemoglobin concentration, packed-cell volume, the serum activities of creatine kinase (CK), aspartate aminotransferase (AST), alanine aminotransferase (ALT) and lactate dehydrogenase (LDH) and the concentrations of creatinine and lactate were significantly lower, and serum glucose started to decrease earlier, than in the untreated groups. Serum potassium levels decreased over time only in the untreated groups. The body temperature stabilised earlier, and the RBC count, haemoglobin concentration, serum CK, AST, ALT and LDH activities, and serum creatinine, lactate, cholesterol and glucose concentrations were significantly lower in the free-ranging roe deer than in the captive deer.  相似文献

OBJECTIVE: To compare physiologic, hematologic, and selected serum and plasma biochemical variables obtained from horses competing in 48-, 83-, or 159-km endurance rides before competition and at the same cumulative distance points. ANIMALS: 83 horses. PROCEDURE: Weight and rectal temperature measurements and blood samples were obtained from horses before, during, and after 1 of 3 rides conducted on the same day. Plasma protein (PP), lactate, WBC, serum electrolyte, and calcium concentrations; PCV; and creatine kinase (CK) activity were determined. Assessments were made to determine whether any differences among groups, with respect to total distance competed, could be explained by differences in lap speed or conditioning and to investigate the effect of time in transit or on-site prior to competition on results of blood analyses or competition outcome. RESULTS: Horses in the 159-km distance group had the lowest preride serum sodium, chloride, bicarbonate, and calcium concentrations. As hours in transit increased, preride PP concentration was significantly greater; serum sodium, chloride, and bicarbonate concentrations were lower; CK activity at 159 km was greater; and horses were more likely to be eliminated. The preride sodium was significantly greater in horses that completed the ride, compared with those eliminated. CONCLUSIONS AND CLINICAL RELEVANCE: Among distance groups, distance ridden, speed, level of fitness, and years of experience of horses had little effect on the variables examined. Electrolyte and water supplementation and earlier arrival at the event may be beneficial for horses that are transported long distances to endurance competition.  相似文献

Serum chemistry analyses were compared between captive and free-ranging giraffes (Giraffa camelopardalis) in an attempt to better understand some of the medical issues seen with captive giraffes. Illnesses, including peracute mortality, energy malnutrition, pancreatic disease, urolithiasis, hoof disease, and severe intestinal parasitism, may be related to zoo nutrition and management issues. Serum samples were collected from 20 captive giraffes at 10 United States institutions. Thirteen of the captive animal samples were collected from animals trained for blood collection; seven were banked samples obtained from a previous serum collection. These samples were compared with serum samples collected from 24 free-ranging giraffes in South Africa. Differences between captive and free-ranging giraffes, males and females, and adults and subadults were analyzed by using a 2 x 2 x 2 factorial and Fisher's least significant difference for mean separation; when necessary variables were ranked and analyzed via analysis of variance. Potassium and bilirubin concentrations and alanine aminotransferase (ALT) activities were different between captive and free-ranging giraffes, but all fell within normal bovid reference ranges. The average glucose concentration was significantly elevated in free-ranging giraffes (161 mg/dl) compared with captive giraffes (113 mg/dl). All giraffes in this study had glucose concentrations higher than bovine (42-75 mg/ dl) and caprine (48-76 mg/dl) reference ranges. Differences were also seen in lipase, chloride, and magnesium though these findings are likely not clinically significant. There were no differences detected between sexes. Adults had higher concentrations of potassium, total protein, globulins, and chloride and higher gamma glutamyltransferase activities, whereas subadults had higher concentrations of phosphorus. Within the captive group, nonimmobilized animals had higher concentrations of total protein and globulins. Captive giraffe diets need further investigation to determine if the differences seen in this study, especially glucose and bilirubin concentrations and ALT activities, may result in some health problems often seen in captive giraffes.  相似文献

Blood from 31 healthy, free-ranging golden jackals held in captivity for seven days was collected while they were anaesthetised. Haematological and serum biochemical measurements were analysed and the 95 per cent confidence interval for each variable was compared with the reference value for domestic dogs. The measurements of their red blood cells were within the reference interval for dogs, but the jackals had higher white blood cell counts and eosinophil counts than dogs. The male jackals had a higher haematocrit, red blood cell count, mean corpuscular volume and mean corpuscular haemoglobin concentration, and a lower red blood cell distribution width than the female jackals. High activities of muscle enzymes were detected in many of the jackals, in several of which the activity of creatine kinase exceeded 5000 U/l; these were considered abnormal.  相似文献

BACKGROUND: Diseases and abnormal physiologic conditions can alter the concentrations of enzymes, metabolites, minerals, and hormones in the blood of animals. The ringed seal (Pusa hispida) has been selected as a key species for environmental monitoring, but information on disease and health parameters for this species is scarce. OBJECTIVES: The aim of the study reported here was to obtain serum chemistry reference intervals for free-ranging ringed seals in Svalbard, and then to evaluate serum chemistry values in relation to age, body condition, and sex. METHODS: Blood samples were collected after death from ringed seals in Wijdefjorden and Billefjorden, Svalbard (2002-2003; n = 75). Serum was analyzed for 24 selected serum chemistry parameters (enzymes, protein, metabolites, minerals, and cortisol). RESULTS: Compared with younger or older animals, seals between 7 and 16 years of age had larger variations in the activities of alanine transaminase, aspartate transaminase, lactate dehydrogenase, and creatine kinase (CK). Animals classified as having low body condition status had more variation in the serum activity of these enzymes, compared with that in animals with higher condition scores. Serum cortisol concentration was higher in young animals (1-5 years) than in older animals. Serum CK activity was higher in males than in females. CONCLUSION: The data reported here may be useful in monitoring the health of ringed seals and for tracking the impact of environmental changes in the Arctic.  相似文献

Physiologic variables during anesthesia with medetomidine-zolazepam-tiletamine were evaluated in 52 free-ranging brown bears (Ursus arctos) darted from a helicopter and in six captive brown bears darted at a zoo. During anesthesia, rectal temperature, respiratory rate, heart rate, and pulse oximetry derived hemoglobin oxygen saturation were recorded. Arterial blood samples were collected and immediately analyzed for evaluation of pulmonary gas exchange, acid-base status, and selected hematologic and plasma variables. At the end of anesthesia, atipamezole was administered intramuscularly at five times the medetomidine dose. Capture-induced hyperthermia and lactic acidemia were documented in free-ranging bears. Hypoxemia during anesthesia was documented in both free-ranging and captive bears. In free-ranging bears, rectal temperature, heart rate, lactate, hematocrit, and hemoglobin decreased significantly during anesthesia, whereas partial pressure of arterial carbon dioxide, pH, potassium, and glucose increased. Yearlings had a significantly higher heart rate, pH, base excess, bicarbonate, and glucose, and had a significantly lower rectal temperature, sodium, hematocrit, and hemoglobin when compared with subadult and adult brown bears. In conclusion, alterations in pulmonary gas exchange and acid-base status in brown bears during anesthesia with medetomidine-zolazepam-tiletamine with the doses and capture methods used in this study were identified. Oxygen supplementation is recommended to counteract hypoxemia during anesthesia.  相似文献

BACKGROUND: To the authors' knowledge, on the basis of sample type, storage condition, or hemolysis, differences in serum and plasma biochemical values have not been evaluated in orange-winged Amazon parrots (Amazona amazonica). OBJECTIVES: The purpose of this study was to compare values for biochemical analytes in serum vs plasma, fresh vs frozen plasma, and nonhemolyzed vs hemolyzed samples in orange-winged Amazon parrots. We also compared differences in serum and plasma yield from whole-blood aliquots. METHODS: Fifteen biochemical analytes were evaluated in paired serum and plasma, fresh and frozen plasma, nonhemolyzed and hemolyzed serum and plasma samples from orange-winged Amazon parrots (n = 10) using a wet reagent analyzer. Hemolysis was assessed qualitatively (visually) and quantitatively (hemoglobin [Hgb] measured spectrophotometrically). Serum and plasma yields from 500-microl whole-blood aliquots were determined from centrifuged samples. RESULTS: Analyte values significantly differed among sample groups, but were still within published reference intervals, with the exception of increases in potassium concentration in markedly hemolyzed serum and plasma samples. Clinically important changes in hemolyzed serum and plasma samples included increases in potassium, phosphorus, and albumin concentrations and lactate dehydrogenase activity. The degree of hemolysis assigned qualitatively did not correlate with quantitative Hgb concentration. A significantly greater yield of plasma (288 +/- 13 microL) than serum (241 +/- 44 microL) was obtained. CONCLUSIONS: Significant differences may occur in different sample types, however, only changes in potassium, phosphorus, albumin, and lactate dehydrogenase values in hemolyzed samples were considered clinically relevant. Lack of agreement between qualitative and quantitative Hgb concentration indicates the unreliability of visual estimation. Based on higher sample yield, and lack of clinically relevant differences from serum, plasma is a better sample choice for clinical chemistry analysis in birds.  相似文献

Huddling is a behavioural energy conservation mechanism that is widely used by many small endotherms at low ambient temperatures. Huddling has many benefits, including decreasing the metabolic cost of maintaining body temperature (Tb), reducing the amount of heat lost to the environment, and increasing the local temperature of the nest. To test the effects of huddling on Tb, 10 rock hyrax, Procavia capensis, were housed in outside cages in four groups, varying from one to four individuals. iButtons® were surgically implanted into each rock hyrax to record their Tb every 15 min from August to November. Despite considerable variations in ambient temperatures, the rock hyrax were found to display some degree of heterothermy by varying their mean Tb from 36.70 to 37.72°C (n = 10) but not allowing it to drop below 33.60°C or rise above 39.67°C. Contrary to what was predicted, rock hyrax did not display any significant effects of huddling on Tb, irrespective of group size.  相似文献

BACKGROUND: The radiated tortoise (Geochelone radiata) is a critically endangered species in its native land, the southern portion of the island of Madagascar. Captive breeding programs have generated data on the breeding behavior and ecology of G radiata; however, hematologic and biochemical data also are critically important in managing populations. OBJECTIVE: The purpose of this study was to evaluate sex and seasonal effects on hematologic and biochemical data from captive radiated tortoises. METHODS: Whole blood was collected in January and August 2001 from 18 radiated tortoises (10 male, 8 female) housed at the Wildlife Survival Center on St Catherines Island, Georgia, as part of a routine health assessment. Routine hematologic and plasma biochemical analyses and electrophoresis were done using standard methods. Data from male and female tortoises were compared within and between seasons using 2-way ANOVA. RESULTS: RBC and HCT values were significantly higher in summer than in winter and were higher in males than in females. Total protein concentration did not differ significantly between males and females; however, female tortoises had significantly higher concentrations of alpha1- and beta-globulins in winter and summer compared to males. Male tortoises had significantly higher sodium and uric acid concentrations and LDH activity during winter, and higher urea concentration and LDH and CK activities in summer, compared with females. Female tortoises had significantly higher triglyceride and phosphorus concentrations in winter, and higher phosphorus, cholesterol, and triglyceride concentrations in summer, compared with males. CONCLUSION: Sex and seasonal differences in hematologic and biochemical values for radiated tortoises likely reflect vitellogenesis and egg production in females, and altered hydration status and activity in summer. Data from the tortoises in this study will be useful for the seasonal health assessment of this species.  相似文献

To investigate the possible role of common brushtail possums (Trichosurus vulpecula) in the transmission of Toxoplasma gondii within a zoo environment, a serological survey of a free-ranging population resident within Taronga Zoo, Sydney, Australia was undertaken using the modified agglutination test (MAT). For comparison, the seroprevalence of T. gondii antibodies was also assessed in a possum population inhabiting a felid-free, non-urban woodland habitat. Six of 126 possums (4.8%) from the zoo population had antibodies to T. gondii with a MAT titre of 25 or higher, while in contrast, all of the 17 possums from woodland were seronegative. These observations suggest that possums were at a higher risk of exposure to the parasite as a consequence of co-existing with domestic, stray and captive felids associated with urbanisation. Screening of captive felids at the zoo indicated 16 of 23 individuals (67%) and all 6 species were seropositive for T. gondii, implicating them as a possible source of the parasite within the zoo setting. In addition captive, non-felid carnivores including the chimpanzee (Pan troglodytes), saltwater crocodile (Crocodylus porosus), dingo (Canis lupis) and leopard seal (Hydrurga leptonyx) were tested for the presence of T. gondii antibodies as these species predate and are a leading cause of death amongst zoo possums. In total, 5 of 23 individuals (22%) were seropositive, representing 2 of the 4 carnivorous species; the dingo and chimpanzee. These data suggest that carnivory was not a highly efficient pathway for the transmission of T. gondii and the free-ranging possum population posed minimal threat to the health of zoo animals.  相似文献

Identification of the sex of aquatic mammals such as the hippopotamus (Hippopotamus amphibius) is difficult in the field. We have developed a PCR-based method for sex identification in the hippopotamus. This method amplifies a short fragment of the ZFX and ZFY genes from the X and Y chromosomes. The PCR products are then digested with the restriction endonuclease HaeIII. The method was verified using tissue samples taken from six captive animals in zoos. We then collected 54 tissue samples from free-ranging hippos in Kruger National Park, South Africa, using a biopsy dart and cross-bow. The PCR method correctly identified the sex of all individuals for whom the sex was known (either zoo animals, or those exhibiting clear behaviours or morphologies in the field). Field identification of the sex of other individuals was prone to error (6 of 22 misidentified, 27 %).  相似文献

Pancreatic islet fibrosis with varying degrees of islet cell hyperplasia or islet effacement was diagnosed histologically in 19 rock hyraxes (Procavia capensis) from seven zoological parks. Some, but not all, affected hyraxes were from a common lineage. The condition was associated with apparent hyperglycemia in seven and diabetes mellitus in two. Immunohistochemistry revealed hyperplasia of beta, alpha, and delta cells proportional to the degree of the fibrosis. Electron microscopy revealed collagen deposition and fibroplasia within and around the islets. Special stains and electron microscopy were negative for the presence of amyloid. Beta cell depletion was never identified. The condition has morphologic features that resemble islet fibrosis of human infants born to diabetic mothers.  相似文献

Blood samples were obtained from 20 bison (Bison bison) from a ranch in northern lower Michigan, as well as from 20 free-ranging bison of the same sex and similar age from the Badlands National Park in South Dakota. Hematologic and serum biochemical values were determined. The values were comparable in both groups, except for those for BUN, aspartate transaminase, and phosphorus, which were significantly (P less than 0.001) higher in the ranched bison than in the free-ranging bison. These differences were attributed to nutritional effects. Impact of age on blood characteristics was assessed in the ranched bison only by comparing values from calves weighing less than 185 kg with those from bison weighing more than 185 kg. Calves had significantly (P less than 0.001) higher values for phosphorus and RBC counts and lower total protein values than adults. Adult bison had higher eosinophil and neutrophil counts with lower numbers of lymphocytes, suggestive of a stress leukogram, whereas calves had the typical bovine neutrophil:lymphocyte ratio.  相似文献

Semen was opportunistically collected from a free-ranging, 10-year-old, 275 cm (total length) Florida manatee (Trichechus manatus latirostris) during rehabilitation treatments. Ultrastructure of the spermatozoa was examined by scanning and transmission electron microscopy and differed slightly from that described for other mammals. Comparisons to the manatee's closest phylogenetic relatives, the elephant and hyrax, were made. The manatee spermatozoa had a similar acrosome but a distinct annulus and lacked the dense bodies observed in the neck of the elephant spermatozoa. Additionally, manatee spermatozoa lacked the lateral vacuoles observed in the nuclear chromatin from of the hyrax spermatozoa. These data add to our understanding of manatees and allow for comparative studies with other species that may be useful in phylogenetic and reproductive studies.  相似文献

This retrospective case series describes clinicopathological data and outcome of hospitalised atypical myopathy (AM) cases in the South-East of England. The study aimed to describe the frequency of metabolic abnormalities (hyperglycaemia, hyperlactataemia, hypertriglyceridaemia) and outcome in AM cases in the South-East of England and test the hypothesis that serum creatine kinase (CK) activity and blood glucose, lactate and triglyceride concentrations are associated with outcome. Medical records (2011–2017) from three referral hospitals were reviewed for cases with a clinical diagnosis of AM. A previously described algorithm was applied and cases were included if a diagnosis of AM was considered highly likely. In cases admitted after 2013 known or possible exposure to sycamore trees was also required for inclusion. Sixty-four animals were included, 44% (28/64) survived. Hyperglycaemia, hyperlactataemia and hypertriglyceridaemia were present in 76%, 89% and 92% of horses on admission, respectively. Survivors had lower blood lactate concentrations (survivors: median 3.5 mmol/L; range 0.5–10.4 mmol/L vs. nonsurvivors: median 7.3 mmol/L; range 2.5–16.5 mmol/L; P = 0.011) and serum CK activities (survivors: median 38,369 U/L; range 7024–570,498 U/L vs. nonsurvivors: median 172,687 U/L; range 2036–570,953 U/L; P = 0.027) on admission when compared to nonsurvivors. Increasing CK activity (P = 0.008) and triglyceride concentrations (P = 0.038) during hospitalisation were associated with nonsurvival. More nonsurvivors required sedation (18/29; 62.1% vs. 4/22; 18.2%; P = 0.002). The prognosis for hospitalised horses with AM is guarded and outcome in this population was associated with admission CK activity and lactate concentrations, and increasing CK activity and triglyceride concentrations and need for sedation during hospitalisation.  相似文献

The aim of this study was to investigate whether marcha exercise results in changes in serum hematologic and biochemical variables and whether this condition differs in male and female horses. Thirty-five Mangalarga Marchador horses, 18 males and 17 females were included in the study. Blood samples were obtained before and immediately after a marcha competition. Samples were used for measurement of hematocrit (HCT), hemoglobin concentration (HGB), red blood cell (RBC), total and differential white blood cell (WBC), total protein (TP), albumin (Alb), aspartate aminotransferase (AST), gamma glutamyltransferase (GGT), creatine kinase (CK), alkaline phosphatase (ALP), lactate dehydrogenase (LDH), creatinine (Crea), and urea (BUN) levels. Data were submitted to analysis of variance using the SAS statistical program, and means were compared by the Tukey test (P < .05). In both males and females, RBC, HCT, AST, and GGT activity increased in response to exercise, but there were no significant differences between genders. In females, significant increases in TP, BUN, Crea, CK, and LDH levels were detected after the marcha gait, but no such changes were detected in males. Total WBC concentrations increased in response to the exercise in both, males and females; however, there were no differences between genders. Plasma HGB, Alb, and ALP levels after marcha gait did not differ in both genders. Marcha exercise–induced significant hematologic and biochemical changes in serum of Mangalarga Marchador horses, independently of gender. However, females presented more blood changes after marcha gait. Gender differences should be taken into consideration in experiments with athletic horses.  相似文献
