Improvements in bull reproductive performance are necessary to optimize the efficiency of cattle production. Female fertility has been enhanced through assisted reproductive technologies as well as genetic selection; however, improving beef bull fertility has been largely ignored. Phenotypes routinely collected at bull semen collection facilities are believed to affect fertility and provide the phenotypes necessary for a genetic evaluation. The first objective of this study was to determine the significant fixed effects for modeling beef bull fertility using data from bull semen collection facilities. The second objective was to estimate variance components, heritabilities, repeatabilities, and correlations between beef bull semen attributes. Beef bull fertility phenotypes including volume (VOL), concentration (CONC), number of spermatozoa (NSP), initial motility (IMot), post-thaw motility (PTMot), 3-h post-thaw motility (3HRPTMot), percentage of normal spermatozoa (%NORM), primary abnormalities (PRIM), and secondary abnormalities (SEC) were obtained from two bull semen collection facilities. A total of 1,819 Angus bulls with 50,624 collection records were analyzed. Of the fixed class and covariate effects tested, the significant class effects were collection location and collection day within year and the significant covariate effects included age at collection, days since previous collection, and cumulative comprehensive climate index (CCI). For this study, the CCI was calculated for a 75-d period including the 61-d spermatogenesis cycle and 14-d epididymal transit time. The 75 d prior to collection accounted for the environmental stress a bull may have experienced over the course of development of the spermatozoa, which was more significant than the CCI calculated for collection day or spermatogenesis start date. Pre-thaw beef bull semen traits had low heritability estimates of 0.11 ± 0.02 (VOL), 0.09 ± 0.02 (CONC), 0.08 ± 0.02 (NSP), and 0.12 ± 0.03 (IMot). Heritabilities of post-thaw beef bull semen attributes were more variable at 0.10 ± 0.02 (PTMot), 0.05 ± 0.04 (3HRPTMot), 0.10 ± 0.04 (%NORM), 0.03 ± 0.03 (PRIM), and 0.18 ± 0.04 (SEC). Correlations of breeding values for these traits with scrotal circumference (SC) expected progeny difference (EPD) are low. The low to moderate heritability estimates indicate that genetic improvement can be made in beef bull semen quality traits if new tools are developed to augment the scrotal circumference EPD that are currently available within the industry.  相似文献

The objective of this experiment was to investigate the relationship between residual feed intake (RFI) and parameters associated with reproductive efficiency in growing beef heifers. One hundred and seventy-nine replacement beef heifers (Bos taurus and Bos indicus-influenced) were enrolled in a retrospective cohort design. Heifers were assigned to a 70-d feed efficiency test, and results were utilized to retrospectively classify heifers into low (n = 51), medium (n = 66), or high (n = 62) RFI groups based on their individual RFI values. Blood samples were collected weekly throughout the feed efficiency test and breeding season, which were analyzed for plasma concentration of progesterone to determine age at puberty (PUB). By design, residual feed intake differed among RFI groups where high (1.21 ± 0.08 kg/d) had greater (P < 0.01) RFI than medium (0.00 ± 0.08 kg/d) and low RFI groups (–1.49 ± 0.09 kg/d), and medium had greater (P < 0.01) RFI than low RFI heifers. Yet, no differences were observed between breed types for RFI (P = 0.53). Average daily gain (ADG) differed between breed types where Bos taurus heifers had greater ADG (P = 0.02) than B. indicus-influenced heifers. Furthermore, ADG tended (P = 0.08) to differ among RFI groups, where medium RFI heifers tended to have reduced ADG compared with low and high RFI heifers. No differences were determined between breed types for gain-to-feed ratio (G:F; P = 0.20; however, G:F differed among RFI groups and was greater (P < 0.01) in low vs. high, and low vs. medium RFI heifers. The percentage of cycling heifers by the start of the breeding season differed (P = 0.03) by RFI group, where a greater percentage of low RFI heifers were cycling compared with high RFI heifers. A difference was determined on PUB between breed types (P = 0.03), where Bos taurus (393.40 ± 4.64 d) heifers had a reduced PUB compared with B. indicus-influenced (406.90 ± 5.07 d) heifers. In addition, PUB differed by RFI group where low (385.96 ± 6.20 d; P < 0.01) and medium (398.47 ± 5.47 d; P = 0.02) RFI heifers had a reduced PUB compared with High RFI heifers (416.03 ± 5.58 d). In conclusion, more feed efficient heifers attained PUB earlier than less feed efficient heifers. Therefore, selection of heifers for feed efficiency may positively impact reproductive performance of replacement beef heifers.  相似文献

Immunoglobulin (Ig) G and natural antibody (NAb) IgM are passively transferred to the neonatal calf through bovine colostrum. Maternal IgG provides pathogen- or vaccine-specific protection and comprises about 85% of colostral Ig. NAb-IgM is less abundant but provides broad and nonspecific reactivity, potentially contributing to protection against the dissemination of pathogens in the blood (septicemia) in a calf’s first days of life. In the dairy and beef industries, failure of passive transfer (FPT) of colostral Ig (serum total protein [STP] <5.2 g/dL) is still a common concern. The objectives of this study were to: (1) compare colostral IgG concentrations and NAb-IgM titers between dairy and beef cows; (2) assess the effect of beef breed on colostral IgG; (3) compare passive transfer of colostral Ig in dairy and beef calves; and (4) estimate the heritability of colostral IgG and NAb-IgM. Colostrum was collected from Holstein dairy (n = 282) and crossbred beef (n = 168) cows at the University of Guelph dairy and beef research centers. Colostral IgG was quantified by radial immunodiffusion and NAb-IgM was quantified by an enzyme-linked immunosorbent assay. In dairy (n = 308) and beef (n = 169) calves, STP was estimated by digital refractometry. Beef cows had significantly greater colostral IgG (146.5 ± 9.5 standard error of the mean [SEM] g/L) than dairy cows (92.4 ± 5.2 g/L, P <0.01). Beef cows with a higher proportion of Angus ancestry had significantly lower colostral IgG (125.5 ± 5.8 g/L) than cows grouped as “Other” (142.5 ± 4.9 g/L, P = 0.02). Using the FPT cutoff, 13% of dairy and 16% of beef calves had FPT; still, beef calves had a significantly larger proportion with excellent passive transfer (STP ≥6.2 g/dL, P <0.01). The heritability of colostral IgG was 0.04 (±0.14) in dairy and 0.14 (±0.32) in beef. Colostral NAb-IgM titers in dairy (12.12 ± 0.22, log2 [reciprocal of titer]) and beef cows (12.03 ± 0.19) did not differ significantly (P = 0.71). The range of NAb-IgM titers was 9.18–14.60, equivalent to a 42-fold range in antibody concentration. The heritability of colostral NAb was 0.24 (±0.16) in dairy and 0.11 (±0.19) in beef cows. This study is the first to compare colostral NAb-IgM between dairy and beef cows. Based on the range in NAb-IgM titers and the heritability, selective breeding may improve colostrum quality and protection for neonatal calves in the early days of life.  相似文献

It is of interest to evaluate crossbred pigs for hot carcass weight (HCW) and birth weight (BW); however, obtaining a HCW record is dependent on livability (LIV) and retained tag (RT). The purpose of this study is to analyze how HCW evaluations are affected when herd removal and missing identification are included in the model and examine if accounting for the reasons for missing traits improves the accuracy of predicting breeding values. Pedigree information was available for 1,965,077 purebred and crossbred animals. Records for 503,716 commercial three-way crossbred terminal animals from 2014 to 2019 were provided by Smithfield Premium Genetics. Two pedigree-based models were compared; model 1 (M1) was a threshold-linear model with all four traits (BW, HCW, RT, and LIV), and model 2 (M2) was a linear model including only BW and HCW. The fixed effects used in the model were contemporary group, sex, age at harvest (for HCW only), and dam parity. The random effects included direct additive genetic and random litter effects. Accuracy, dispersion, bias, and Pearson correlations were estimated using the linear regression method. The heritabilities were 0.11, 0.07, 0.02, and 0.04 for BW, HCW, RT, and LIV, respectively, with standard errors less than 0.01. No difference was observed in heritabilities or accuracies for BW and HCW between M1 and M2. Accuracies were 0.33, 0.37, 0.19, and 0.23 for BW, HCW, RT, and LIV, respectively. The genetic correlation between BW and RT was 0.34 ± 0.03, and between BW and LIV was 0.56 ± 0.03. Similarly, the genetic correlation between HCW and RT was 0.26 ± 0.04, and between HCW and LIV was 0.09 ± 0.05, respectively. The positive and moderate genetic correlations between BW and other traits imply a heavier BW resulted in a higher probability of surviving to harvest. Genetic correlations between HCW and other traits were lower due to the large quantity of missing records. Despite the heritable and correlated aspects of RT and LIV, results imply no major differences between M1 and M2; hence, it is unnecessary to include these traits in classical models for BW and HCW.  相似文献

The development of technologies that promote environmental stewardship while maintaining or improving the efficiency of food animal production is essential to the sustainability of producing a food supply to meet the demands of a growing population. As such, Elanco (Greenfield, IN) pursued an environmental indication for a selective β-modulator (lubabegron; LUB). LUB was recently approved by the United States Food and Drug Administration (FDA) to be fed to feedlot cattle during the last 14 to 91 d of the feeding period for reductions in gas emissions/kg of unshrunk final BW and HCW. A 4 × 2 factorial arrangement of treatments was used with the factors of dose (0.0, 1.38, 5.5, or 22.0 mg·kg−1 DM basis) and sex (steers or heifers). Three 91-d cycles were conducted (112 cattle/cycle) with each dose × sex combination being represented by a single cattle pen enclosure (CPE; 14 cattle/CPE) resulting in a total of 168 steers and 168 heifers (n = 6 replicates/dose). There were no interactions observed between dose and sex for any variable measured in the study (P ≥ 0.063). Five gases were evaluated for all pens based on CPE concentrations relative to ambient air: NH3, CH4, N2O, H2S, and CO2. Cumulative NH3 gas emissions were reduced by feeding cattle 5.5 and 22.0 mg·kg−1 LUB (P ≤ 0.023) and tended (P = 0.076) to be lower for the cattle fed 1.38 mg·kg−1 LUB compared with the negative controls (CON). The cumulative NH3 gas emission reductions of 960 to 1032 g, coupled with HCW increases (P ≤ 0.019) of 15 to 16 kg for all LUB doses vs. CON, led to reductions in NH3 gas emissions/kg HCW for all three LUB treatments (P ≤ 0.004). Similar to HCW, reductions in NH3 gas emissions/kg of unshrunk final BW were observed for all LUB doses (P ≤ 0.009) and were attributable to both decreases in NH3 gas emissions and numerical increases in BW. Dose had no effect on cumulative emissions or emissions standardized by BW or HCW for the other four gases (P ≥ 0.268). LUB is a novel tool to reduce emissions of NH3 gas per kilogram of unshrunk live BW and hot carcass weight.  相似文献

In the pig industry, purebred animals are raised in nucleus herds and selected to produce crossbred progeny to perform in commercial environments. Crossbred and purebred performances are different, correlated traits. All purebreds in a pen have their performance assessed together at the end of a performance test. However, only selected crossbreds are removed (based on visual inspection) and measured at different times creating many small contemporary groups (CGs). This may reduce estimated breeding value (EBV) prediction accuracies. Considering this sequential recording of crossbreds, the objective was to investigate the impact of different CG definitions on genetic parameters and EBV prediction accuracy for crossbred traits. Growth rate (GP) and ultrasound backfat (BFP) records were available for purebreds. Lifetime growth (GX) and backfat (BFX) were recorded on crossbreds. Different CGs were tested: CG_all included farm, sex, birth year, and birth week; CG_week added slaughter week; and CG_day used slaughter day instead of week. Data of 124,709 crossbreds were used. The purebred phenotypes (62,274 animals) included three generations of purebred ancestors of these crossbreds and their CG mates. Variance components for four-trait models with different CG definitions were estimated with average information restricted maximum likelihood. Purebred traits’ variance components remained stable across CG definitions and varied slightly for BFX. Additive genetic variances (and heritabilities) for GX fluctuated more: 812 ± 36 (0.28 ± 0.01), 257 ± 15 (0.17 ± 0.01), and 204 ± 13 (0.15 ± 0.01) for CG_all, CG_week, and CG_day, respectively. Age at slaughter (AAS) and hot carcass weight (HCW) adjusted for age were investigated as alternatives for GX. Both have potential for selection but lower heritabilities compared with GX: 0.21 ± 0.01 (0.18 ± 0.01), 0.16 ± 0.02 (0.16 + 0.01), and 0.10 ± 0.01 (0.14 ± 0.01) for AAS (HCW) using CG_all, CG_week, and CG_day, respectively. The predictive ability, linear regression (LR) accuracy, bias, and dispersion of crossbred traits in crossbreds favored CG_day, but correlations with unadjusted phenotypes favored CG_all. In purebreds, CG_all showed the best LR accuracy, while showing small relative differences in bias and dispersion. Different CG scenarios showed no relevant impact on BFX EBV. This study shows that different CG definitions may affect evaluation stability and animal ranking. Results suggest that ignoring slaughter dates in CG is more appropriate for estimating crossbred trait EBV for purebred animals.  相似文献

The details of a small study on the volume of colostrum and colostral gammaglobulins produced by beef cows at the first milking immediately after calving and the serum concentrations of passively acquired colostral gammaglobulins attained by beef calves are reported. The mean volume of colostrum and colostral gammaglobulins produced were 2990 ± 2100 (SD) mL and 314.75 ± 211.8 (SD) g respectively. The mean serum concentrations of passively acquired colostral gammaglobulins were 29.9 ± 10.1 (SD) ZST units (n = 40, Ranch A) and 19.2 ± 7.5 (SD) ZST units (n = 42, Ranch B). The main reason for this difference appeared to be nutritional; the cows on ranch B being in much poorer condition and almost certainly producing less colostrum than those on ranch A.  相似文献

An experiment was designed to determine the best practical method of thawing which could be applied to all semen processed by Canadian A.I. centers. Semen in Tris, Triladyl, whole milk, or Fresh Plus Extender packaged in 1 mL ampules or 0.5 mL French straws was used. Semen was thawed in water at 35°C, 20°C, or 5°C, or in a shirt pocket for 5, 5, 10, or 10 minutes, respectively. Post-thaw viability of sperm cells was assessed by determining the percentage of progressively motile cells, rate of progression, and percentage of intact acrosomes, at 0 hours and after 2 hours of incubation at 37°C. Post-thaw viability was significantly higher (P < 0.05) for the 35°C thaw than for any of the other thaw methods regardless of extender or packaging. Our data indicate that when AI center recommendations for thawing semen are unknown, semen in ampules and straws should be thawed in a 35°C water bath and maintained at that temperature until it can be inseminated.  相似文献

The effects of birth weight (BiW; low BiW [LBW] vs. high BiW [HBW]) and estimated breeding value (EBV) for protein deposition (low EBV [LBV] vs. high EBV [HBV]) on N retention, N efficiency, and concentrations of metabolites in plasma and urine related to N efficiency in growing pigs were studied. At an age of 14 wk, 10 LBW–LBV (BiW: 1.07 ± 0.09 [SD] kg; EBV: −2.52 ± 3.97 g/d, compared with an average crossbred pig with a protein deposition of 165 g/d), 10 LBW–HBV (BiW: 1.02 ± 0.13 kg; EBV: 10.47 ± 4.26 g/d), 10 HBW–LBV (BiW: 1.80 ± 0.13 kg; EBV: −2.15 ± 2.28 g/d), and 10 HBW–HBV (BiW: 1.80 ± 0.15 kg; EBV: 11.18 ± 3.68 g/d) male growing pigs were allotted to the experiment. The pigs were individually housed in metabolism cages and were subjected to an N balance study in two sequential periods of 5 d, after an 11-d dietary adaptation period. Pigs were assigned to a protein adequate (A) or protein restricted (R, 70% of A) regime in a change-over design. Pigs were fed 2.8 times the energy requirements for maintenance. Nontargeted metabolomics analyses were performed in urine and blood plasma samples. The N retention (in g/d) was higher in the HBW than in the LBW pigs (P < 0.001). The N retention (in g/[kg metabolic body weight (BW0.75) · d]) and N efficiency, however, were not affected by the BiW of the pigs. The N retention (P = 0.04) and N efficiency (P = 0.04) were higher in HBV than in LVB pigs on the A regime but were not affected by EBV in pigs on the R regime. Restricting the dietary protein supply with 30% decreased the N retention (P < 0.001) but increased the N efficiency (P = 0.003). Nontargeted metabolomics showed that a hexose, free amino acids (AA), and lysophosphatidylcholines were the most important metabolites in plasma for the discrimination between HBV and LBV pigs, whereas metabolites of microbial origin contributed to the discrimination between HBV and LBV pigs in urine. This study shows that BiW does not affect N efficiency in the later life of pigs. Nitrogen efficiency and N retention were higher in HBV than in LBV pigs on the A regime but similar in HBV and LBV pigs on the R regime. In precision feeding concepts aiming to further optimize protein and AA efficiency in pigs, the variation in EBV for protein deposition of pigs should be considered as a factor determining N retention, growth performance, and N efficiency.  相似文献

Understanding the utilization of feed energy is essential for precision feeding in beef cattle production. We aimed to assess whether predicting the metabolizable energy (ME) to digestible energy (DE) ratio (MDR), rather than a prediction of ME with DE, is feasible and to develop a model equation to predict MDR in beef cattle. We constructed a literature database based on published data. A meta-analysis was conducted with 306 means from 69 studies containing both dietary DE and ME concentrations measured by calorimetry to test whether exclusion of the y-intercept is adequate in the linear relationship between DE and ME. A random coefficient model with study as the random variable was used to develop equations to predict MDR in growing and finishing beef cattle. Routinely measured or calculated variables in the field (body weight, age, daily gain, intake, and dietary nutrient components) were chosen as explanatory variables. The developed equations were evaluated with other published equations. The no-intercept linear equation was found to represent the relationship between DE and ME more appropriately than the equation with a y-intercept. The y-intercept (−0.025 ± 0.0525) was not different from 0 (P = 0.638), and Akaike and Bayesian information criteria of the no-intercept model were smaller than those with the y-intercept. Within our growing and finishing cattle data, the animal’s physiological stage was not a significant variable affecting MDR after accounting for the study effect (P = 0.213). The mean (±SE) of MDR was 0.849 (±0.0063). The best equation for predicting MDR (n = 106 from 28 studies) was 0.9410 ( ± 0.02160) +0.0042 ( ± 0.00186) × DMI (kg) – 0.0017 ( ± 0.00024) × NDF(% DM) – 0.0022 ( ± 0.00084) × CP(% DM). We also presented a model with a positive coefficient for the ether extract (n = 80 from 22 studies). When using these equations, the observed ME was predicted with high precision (R2 = 0.92). The model accuracy was also high, as shown by the high concordance correlation coefficient (>0.95) and small root mean square error of prediction (RMSEP), <5% of the observed mean. Moreover, a significant portion of the RMSEP was due to random bias (> 93%), without mean or slope bias (P > 0.05). We concluded that dietary ME in beef cattle could be accurately estimated from dietary DE and its conversion factor, MDR, predicted by the dry matter intake and concentration of several dietary nutrients, using the 2 equations developed in this study.  相似文献

Alkaline phosphatase activity was recorded in forty ejaculates of the sperm rich fraction of boar semen as 9,790 ± 5,250 Klein-Babson-Read units per 100 ml. of seminal plasma. Acid phosphatase activity in the same ejaculates was 681 ± 304 Babson-Read units per 100 ml. of seminal plasma. No alkaline phosphatase activity was detected in the seminal plasma of vasectomized boars.

The pH of the sperm rich fractions was 7.69 ± 0.33 and the osmotic pressure was 313.56 ± 7.98 milliosmols.

Two experiments were designed to test the hypothesis that induction of parturition in the cow would be more predictable with the simultaneous use of a combination of cloprostenol and dexamethasone than with either hormone used alone.

In experiment I all 19 beef cows treated with 500 μg cloprostenol and 25 mg dexamethasone in combination calved within 72 hours whereas dexamethasone (n = 19) or cloprostenol (n = 16) treatments alone each resulted in two induction failures. In those cows successfully induced, the mean interval from treatment to birth was 34.6 ± 1.4 hours for the cloprostenol plus dexamethasone group, 43.3 ± 2.4 hours for the dexamethasone group and 44.9 ± 2.1 hours for the cloprostenol group. Control cows (n = 15) did not calve during the first 72 hours after treatment with saline. The incidence of retained placenta ranged from 19 to 53% in induced groups whereas placentae were not retained by cows in the control group.

In experiment II all 30 beef cows in the cloprostenol plus dexamethasone group calved within the 72 hour limit, with a mean interval of 39.1 ± 1.0 hours. Twenty-six of 31 cows calved within 72 hours with a mean interval of 51.9 ± 3.4 hours after a single injection of cloprostenol and 29 of 33 cows calved within 72 hours with a mean interval of 52.6 ± 3.3 hours after two injections of cloprostenol, 12 hours apart. Five of 34 control cows calved within 72 hours of time of treatment. The incidence of retained placenta was again high in induced cows. Results indicate that the simultaneous administration of cloprostenol and dexamethasone does constitute a safe, reliable and effective method of inducing parturition in the cow.

Serum samples were obtained from 26 beef cows with uterine prolapse and from 15 with minor dystocia (controls). The serum of animals with uterine prolapse had significantly lower calcium concentration (mean ± S.D. = 8,22 ± 0,69 mg/dL, P≤0,01), higher phosphorus concentration (mean ± S.D. = 4,78 ± 1,75 mg/dL, P≤0,05) and lower calcium to phosphorus ratios (mean ± S.D. = 1,99 ± 0,88, P≤0,01) than for the control animals (means ± S.D. = 8,91 ± 0,75 mg/dL, 3,54 ± 1,41 mg/dL and 2,99 ± 1,41 respectively). Mild hypocalcemia (6,9 mg/dL-7,9 mg/dL) was present in 11 (42,3%) of the cows with prolapse as compared to only one (6,7%) of the controls. Hypophosphatemia was present in 11 (42,3%) of the animals with prolapse and in ten (66,7%) of the controls.

Eighteen (69,2%) of the animals with prolapse were alert and ambulatory when treated and 15 (57,7%) were known to have required help to deliver the calf. Of the cattle group with uterine prolapse, 14 (53,8%) were two years old, six (23,1%) were three years old, and six (23,1%) were four years of age or older.

It was concluded that mild hypocalcemia and some degree of dystocia were associated with the uterine prolapses. The phosphorus results were equivocal but the high incidence of hypophosphatemia may reflect a phosphorus deficient diet.

Background

The provision of feed is a major cost in beef production. Therefore, the improvement of feed efficiency is warranted. The direct assessment of feed efficiency has limitations and alternatives are needed. Small intestine micro-architecture is associated with function and may be related to feed efficiency. The objective was to verify the potential histomorphological differences in the small intestine of animals with divergent feed efficiency.

Methods

From a population of 45 feedlot steers, 12 were selected with low-RFI (superior feed efficiency) and 12 with high-RFI (inferior feed efficiency) at the end of the finishing period. The animals were processed at 13.79 ± 1.21 months of age. Within 1.5 h of slaughter the gastrointestinal tract was collected and segments from duodenum and ileum were harvested. Tissue fragments were processed, sectioned and stained with hematoxylin and eosin. Photomicroscopy images were taken under 1000x magnification. For each animal 100 intestinal crypts were imaged, in a cross section view, from each of the two intestinal segments. Images were analyzed using the software ImageJ®. The measurements taken were: crypt area, crypt perimeter, crypt lumen area, nuclei number and the cell size was indirectly calculated. Data were analyzed using general linear model and correlation procedures of SAS®.

Results

Efficient beef steers (low-RFI) have a greater cellularity (indicated by nuclei number) in the small intestinal crypts, both in duodenum and ileum, than less efficient beef steers (high-RFI) (P < 0.05). The mean values for the nuclei number of the low-RFI and high-RFI groups were 33.16 and 30.30 in the duodenum and 37.21 and 33.65 in the ileum, respectively. The average size of the cells did not differ between feed efficiency groups in both segments (P ≥ 0.10). A trend was observed (P ≤ 0.10) for greater crypt area and crypt perimeter in the ileum for cattle with improved feed efficiency.

Conclusion

Improved feed efficiency is associated with greater cellularity and no differences on average cell size in the crypts of the small intestine in the bovine. These observations are likely to lead to an increase in the energy demand by the small intestine regardless of the more desirable feed efficiency.  相似文献

Artificial insemination (AI) can help to avoid inbreeding and genetic degeneration for sustaining genetically healthy populations of endangered species in captivity. Collection of a sufficient quantity of viable sperm is an essential first step in the AI process. In the present study, we examined the effects of frequent electroejaculation on semen characteristics in a Siberian tiger. We collected semen in all 17 trials during 6 breeding seasons (6 years). The mean number of sperm and the percentage of motile sperm were 294.3 ± 250.2×106/ejaculate and 82.4 ± 11.4%, respectively. The number of motile sperm tended to increase during frequent electroejaculation in the same breeding season. Semen collection by electroejaculation can be performed effectively up to the fourth sequential ejaculate, which contained the most sperm in the study. In conclusion, frequent collection of sperm by electroejaculation from tigers may be effective for collection of a large number of motile sperm.  相似文献

In the Japanese macaque, semen has been collected by electro-ejaculation (EE), using the higher voltage stimuli compared to other species including genus Macaca. Semen coagulates immediately after ejaculation, which makes difficult to produce high-quality semen for artificial insemination. Recently, semen collection using urethral catheterization (UC) has been reported in carnivore and this technique may allow semen collection without coagulation in a less invasive manner. Further, the temporal preservation temperature and cooling rate of semen during cryopreservation affect post thawing sperm quality. In this study, to improve semen quality and quantity, as well as the animal welfare, semen collection was performed by EE with high (5–15 V) or low (3–6 V) voltage, UC and a combination of the two (EE-UC). It has been suggested that a high voltage is necessary for semen collection, but 10 V stimulation was effective enough and 15 V is for additional sperm collection. Also, liquid semen was collected by EE-UC and this could increase the total number of sperm. Further, to improve the post thawing sperm motility, semen was kept at four temperatures (4, 15, 25 and 37°C) for 60 min, and processed with two cooling procedures (slow cooling before second dilution and fast cooling after second dilution). Holding semen at 25°C and fast cooling after the second dilution maintained progressive motile sperm rate. The present results will contribute to the improvement of semen collection and animal welfare of Japanese macaques.  相似文献

The first (1 to 3) sperm-rich fractions of the ejaculate were collected from 4 stallions using an open-ended vagina. The volume of the collected fractions was 12 ± 8 ml with a density of 475 ± 200 million spermatozoa/ml. Before freezing, the semen was diluted with a skim-milk based extender 1:1 to 1: 8 (volume of semen: volume of extender), depending on the initial sperm concentration to achieve a final concentration of 100 million/ml. The total number of spermatozoa in an insemination dose ranged from 0.7 to 1 billion spermatozoa. Within 12 h after ovulation, 48 mares were inseminated in 70 cycles. The total single-cycle pregnancy rate at day 21 was 24%, but varied from 10% to 33% per cycle among the stallions.  相似文献

Animal manure can be a source of antibiotic-resistant genes (ARGs) and pharmaceutical residues; however, few studies have evaluated the presence of ARG in pasture-raised animal production systems. The objective of this study was to examine changes in microbiome diversity and the presence of antibiotic residues (ABRs) on three farms that contained a diverse range of animal species: pasture-raised poultry (broiler and layer), swine, and beef cattle. Total bacterial communities were determined using 16S rRNA microbiome analysis, while specific ARGs (sulfonamide [Sul; Sul1] and tetracycline [Tet; TetA]) were enumerated by qPCR (real-time PCR). Results indicated that the ARG abundances (Sul1 [P < 0.05] and TetA [P < 0.001]) were higher in layer hen manures (16.5 × 10−4 and 1.4 × 10−4 µg kg−1, respectively) followed by broiler chickens (2.9 × 10−4 and 1.7 × 10−4 µg kg−1, respectively), swine (0.22 × 10−4 and 0.20 × 10−4 µg kg−1, respectively) and beef cattle (0.19 × 10−4 and 0.02 × 10−4 µg kg−1, respectively). Average fecal TetA ABR tended to be greater (P = 0.09) for broiler chickens (11.4 µg kg−1) than for other animal species (1.8 to 0.06 µg kg−1), while chlortetracycline, lincomycin, and oxytetracycline ABRs were similar among animal species. Furthermore, fecal microbial richness and abundances differed significantly (P < 0.01) both among farms and specific species of animal. This study indicated that the microbial diversity, ABR, ARG concentrations, and types in feces varied from farm-to-farm and from animal species-to-animal species. Future studies are necessary to perform detailed investigations of the horizontal transfer mechanism of antibiotic-resistant microorganisms (ARMs) and ARG.  相似文献

To test the hypothesis that epidural administration of lidocaine, xylazine or xylazine plus hyaluronidase provides reduced pain and stress during electroejaculation in bulls, eight 30-month-old Nellore bulls received saline solution (control), 2% lidocaine, 2% xylazine or 2% xylazine plus hyaluronidase injected into the first intercoccygeal (Co1–Co2) epidural space in randomized order. Heart rate, respiratory rate, mean arterial pressure, analgesia, animal behavior and motor blockade were evaluated before treatment and at predetermined intervals during and after treatment. Pain and stress were scored subjectively, and semen quality was evaluated. The onset of anesthetic action was significantly faster with lidocaine (3.0 ± 1.2 min) than with xylazine or xylazine plus hyaluronidase (8.9 ± 1.5 and 5.5 ± 2.6 min, P=0.021 and P=0.012, respectively), and the onset of anesthesia with xylazine plus hyaluronidase was significantly faster than that with xylazine alone (P=0.032). Treatment with xylazine or xylazine plus hyaluronidase resulted in less discomfort than treatment with lidocaine, as indicated by animal behavior. Changes in heart rate, respiratory rate and arterial pressure were within acceptable limits. Penile protrusion and semen emission occurred in all animals during all four treatments. Our results suggest that xylazine plus hyaluronidase reduced discomfort during electroejaculation more effectively than xylazine or lidocaine alone. Further experiments are necessary to determine whether electroejaculation with xylazine plus hyaluronidase is feasible for obtaining semen from Nellore bulls unaccustomed to being handled or restrained.  相似文献

Two experiments were performed to develop prediction equations of saleable beef and to validate the prediction equations. In Exp. 1, 50 beef cattle were finished to typical slaughter weights, and multiple linear regression equations were developed to predict kilograms of trimmed boneless, retail product of live cattle, and hot and cold carcasses. A four-terminal bioelectrical impedance analyzer (BIA) was used to measure resistance (Rs) and reactance (Xc) on each animal and processed carcass. The IMPS cuts plus trim were weighed and recorded. Distance between detector terminals (Lg) and carcass temperature (Tp) at time of BIA readings were recorded. Other variables included live weight (BW), hot carcass weight (HCW), cold carcass weight (CCW), and volume (Lg2/Rs). Regression equations for predicting kilograms of saleable product were [11.87 + (.409 x BW) - (.335 x Lg) + (.0518 x volume)] for live (R2 = .80); [-58.83 + (.589 x HCW) - (.846 x Rs) + (1.152 x Xc) + (.142 x Lg) + (2.608 x Tp)] for hot carcass (R2 = .95); and [32.15 + (.633 x CCW) + (.33 x Xc) - (.83 x Lg) + (.677 x volume)] for cold carcass (R2 = .93). In Exp. 2, 27 beef cattle were finished in a manner similar to Exp. 1, and the prediction equations from Exp. 1 were used to predict the saleable product of these animals. The Pearson correlations between actual saleable product and the predictions based on live and cold carcass data were .91 and .95, respectively. The Spearman and Kendall rank correlations were .95 and .83, respectively, for the cold carcass data. These results provide a practical application of bioelectrical impedance for market-based pricing. They complement previous studies that assessed fat-free mass.  相似文献
