In the present report, we describe targeted next-generation sequencing of the EDA gene of a male poodle with a clinical and histopathological diagnosis of X-linked hypohidrotic ectodermal dysplasia. The result was compared with the reference sequence and with the result of the sequencing of a normal dog's EDA gene. No point variant, small deletion or insertion were found in the exons and splice sites, but a transition and a transversion were found in the intron 6 and 3′ UTR, respectively. The cause of the dysplasia of the affected dog in this study is neither a point variant nor a small deletion or insertion in the exons and splice sites of the EDA gene. Therefore, patients with phenotype of XLHED may have other types of variants in the EDA gene or variants in other genes of the EDA signalling pathway.  相似文献

IgA immune responses in the respiratory tract of pigs   总被引:1,自引:0,他引:1  
Experiments were conducted in a group of pigs to determine the ontogeny of antigen specific IgA in the trachea. The results showed that following intraperitoneal immunisation with ovalbumin (OVA) there were antigen specific IgG and IgA responses in both serum and respiratory tract secretion (RTS). After intratracheal challenge the IgA response in RTS was greater than that in serum as judged by the IgA/IgG ELISA ratios. Furthermore, following intratracheal challenge the IgA/IgG RTS ratio remained elevated for about 30 days. At slaughter, the anti-OVA containing-cell (AOCC) response and the corresponding immunoglobulin class were assessed using double fluorochrome labelling techniques. Pigs challenged intratracheally on four occasions between 11 and 67 days after intraperitoneal immunisation had a population of AOCC in the trachea which was 49 +/- 15 per cent IgA. In contrast, groups of pigs intraperitoneally immunised but challenged intratracheally on fewer occasions had significantly lower proportions of AOCC that were IgA. In conjunction with previous findings these results suggest that following intraperitoneal immunisation regular antigenic challenge of the trachea over an extended period leads to an increase in the proportion of AOCC which are IgA. These results are discussed in terms of the mechanisms involved in protecting the respiratory tract.  相似文献

This review presents the clinical, dermato-histopathological and genetic features of canine X-linked ectodermal dysplasia in previously reported cases and in three new spontaneous cases. The condition is compared with anhidrotic ectodermal dysplasia in humans and, based on current genetic concepts, we suggest that the two conditions are caused by the same gene and, consequently, represent a single pathological entity that affects both humans and dogs.  相似文献

Immune responses to mycoplasma infections of the respiratory tract   总被引:1,自引:0,他引:1  
Mycoplasmas are capable of causing respiratory disease in a number of species of animals. The pathogenicity of the mycoplasma species ranges from those that cause major disease outbreaks and economic loss to what might be considered the more highly evolved and successful parasites at the other end of the spectrum that survive for long periods in the host without being recognised and evicted. This prolonged colonisation of mucous membranes which is typical of many mycoplasmas is related to certain unique features of the mycoplasma and its interaction with the hosts immune system. An initial step in infection is the attachment of the mycoplasma to the epithelial lining of the respiratory tract. Lack of cell wall confers plasticity and may engender the intimate association of mycoplasma and host cell that has been noted. This in turn may favour persistence of the extracellular parasite. Before the specific immune response is produced avoidance of the non-specific immune mechanisms would clearly aid survival. Both passive (capsules) and active (toxic effects) mechanisms of avoiding phagocytosis have been proposed. Both humoral and cell mediated responses are generated by mycoplasma infections. The serum antibody response follows the usual course IgM, G and A. The indications of cell mediated immunity that have been reported include; delayed type hypersensitivity reactions, lymphocyte transformation responses and inhibition of macrophage migration. The concept that the pathological lesions are in a large part due to host reactivity is well accepted. The lung lesions may contain infiltrating and dividing lymphocytes some of which are producing specific antibody. Evidence for the lung lesion in some animals being partly due to the hosts cell mediated response has also been produced. The local immune response appears to be of greater relevance to immunity to infection than the systemic response, in general the association between local antibody and immunity is much better than for serum antibody. Of particular note is the high contribution of local IgG production, particularly in the lower respiratory tract. Attempts are now being made to use this increased understanding to produce effective killed vaccines that produce immune responses in the lung. Such studies will hopefully lead to the development of 'killed' vaccines that are effective. It can be urged that mycoplasmas would be less pathogenic if they did not produce an inflammatory response and some species have been shown to have an immunosuppressive effect. A 3-year-old Thoroughbred filly had a pharyngeal abscess causing obstruction of the upper respiratory tract. Diagnosis was made by a combination of endoscopy, biopsy, cytology, and radiography. Treatment with antimicrobials and analgesics resulted in successful resolution of the abscess.  相似文献

This paper reports on a study of the aetiology of calf pneumonia and the clinical efficacy of florfenicol, a new antibiotic in Turkey. Twenty-seven weaned and unweaned calves (13 males and 14 females) between 1 and 16 months of age brought to the clinics of Sel?uk University, Faculty of Veterinary Science. Broncho-alveolar lavage (BAL) fluid samples were taken from the animals diagnosed to have upper respiratory tract infection associated with bronchitis (N=2), bronchitis (N=5), bronchopneumonia (N=4), pneumonia (N=3), pleuropneumonia (N=11), bronchopneumonia plus pulmonary oedema (N=2) based on the results of the clinical and laboratory examinations. Then microbiological isolation and antibiotic culturing were performed. The animals were treated with 1 ml/15 kg (20 mg/kg) florfenicol (Nuflor, DIF) twice within 48 hours via intramuscular injection. At the end of the treatment, 23 of the weaned and unweaned calves were completely healed, 1 calf had died and 3 calves showed no healing. The results of BAL samples and microbiological examinations of the 3 calves that did not respond to the treatment indicated that these cases were affected by mixed infections of yeasts, fungi, and bacteria. Widespread pleuropneumonia was observed. According to the results of the microbiological examination of the BAL samples, Mannheimia (Pasteurella) haemolytica had the highest isolation rate (25%) compared with the other isolated bacteria, namely, Klebsiella pneumonia (20%), Actinomyces pyogenes (15%), beta-hemolytic streptococci. (10%), Staphylococcus spp. (5%), and E. coli (5%). The study also revealed fungi [Penicillum spp. (5%) and Aspergillus spp. (5%)] and two calves (10%) had a yeast infection.. We conclude that florfenicol has a high bacteriological and clinical efficacy (100% and 96% respectively) in the treatment of calf respiratory tract diseases.  相似文献

1. Riboflavin‐deficient practical and semi‐purified diets were fed to broiler breeder hens.

2. Yolk and albumen riboflavin concentrations were depressed by the deficient diets and cases of curled toes were seen in the chicks.

3. The few instances of defective down did not resemble cases seen in field outbreaks of a condition involving depressed hatchability and a defective down syndrome.

4. It is concluded from observations in this experiment and in the field outbreaks that there is a condition in breeding chickens causing depressed hatchability and defective down that is not attributable to riboflavin deficiency.  相似文献

Local immunity is the first defence mechanism against viral respiratory infections in young calves. The great variety of different viruses which may infect the very young animal makes it necessary to develop effective local defences early in life. The best approach appears to be to increase the subjects' defences by using immunoadjuvants and interferon inducers. With this end in view we should increase our knowledge of the immune system of the calf respiratory tract, particularly with respect to specific immunoglobulins, and cells which mediate the immune response.
Kurzfassung Die örtliche Immunität ist der erste Abwehrmechanismus gegen virale Erkrankungen der Atemwege beim jungen Kalb. Da sich diese sehr jungen Tiere mit den unterschiedlichsten Viren infizieren können, ist ein frühzeitiger Aufbau einer wirksamen örtlichen Abwehr unerlässlich; dieses Problem liesse sich vielleicht durch eine Steigerung der Abwehrkräfte bei den Tieren mit Immunitatsadjuvanten und Induktoren von Interferonbildung bewältigen. Die bisher vorliegenden Angaben über das Immunsystem der Atemwege beim Kalb (spezifische Immunglobuline, Immunzellen) müssen zu diesem Zweck noch vertieft werden.

Resume L'immunité locale est le premier mécanisme de défense contre les affections réspiratoires virales du jeune veau. La grande diversité des virus pouvant infecter ces sujets très jeunes rend indispensable l'établissement précoce de défenses locales éfficaces; l'augmentation des défenses des sujets par l'emploi d'adjuvants de l'immunite et d'inducteurs d'interféron semble être le moyen d'aborder ce problème. Les données que nous possédons sur le système immunitaire de l'appareil réspiratoire du veau (immunoglobulines specifiques, cellules effectrices de l'immunité) doivent, dans ce but, être approfondies.

Riassunto L'immunita locale e il primo meccanismo di difesa del giovane vitello contro le affezioni virali delle vie respiratorie. La grande diversità dei virus in grado di infettare soggetti molto giovani rende necessaria l'instaurazione precoce di difese locali efficaci; la soluzione migliore dovrebbe consistere nel potenziamento delle difese dei soggetti mediante adiuvanti dell'immunità e induttori di interferon. A tal fine e necessario approfondire i dati in nostro possesso sul sistema immunitario dell'apparato respiratorio del vitello (Immunoglobuline specifiche, immunociti).

BACKGROUND: Reticulocyte indices, especially reticulocyte hemoglobin content (CH retic), have shown promise as markers of iron deficiency (ID), but there have been no prospective investigations of reticulocyte indices in experimental models of ID. OBJECTIVES: The objective of this study was to compare reticulocyte indices with conventional hematologic and biochemical indices as markers of ID in dogs. METHODS: Iron deficiency was induced in 7 dogs by feeding an iron-deficient diet, and corrected by restoring dietary iron and by giving iron parenterally. Blood samples were collected at weekly intervals. Results of hematologic and biochemical tests were compared using t-tests and receiver operator characteristic (ROC) curve analysis. RESULTS: Comparing mean values on days 0 and 35, by which time hemoglobin concentration decreased to 90% of baseline in all dogs, % Macro retic, % Hypo retic, % Low CH retic, and % High CH retic differed by greater than 3-fold, whereas no conventional hematologic or biochemical indices differed by as much as 2-fold. Comparing conventional hematologic and reticulocyte indices by ROC curve analysis using 4 different biochemical diagnostic criteria of ID, CH retic, % Hypo retic, % Low CH retic, and % High CH retic had higher areas under the curve (AUC) than either MCV conv or MCHC conv according to all 4 criteria, and % Macro retic and MCV retic had higher AUC values according to 3 of 4 criteria (differences were not always statistically significant). CONCLUSIONS: Results of this study support the value of reticulocyte indices in the diagnosis and monitoring of ID in dogs. Species similarities in the pathophysiology and hematologic manifestations of ID suggest these findings also may be relevant to ID in people.  相似文献

An investigation was made of 3 serological tests (virus neutralization, complement fixation and indirect immunofluorescence), which are applicable to epidemiological studies of infections by Equid herpesvirus 1 (EHV-1). Sera from gnotobiotic foals inoculated intranasally with various strains of EHV-1 were unable in some cases to neutralize heterologous strains and these results were not consistent with the existence of clearly-defined subtypes of EHV-1, as previously proposed. The cross-reactions in complement-fixation tests paralleled those with neutralization but immunofluorescence tests were found to be both more sensitive and more broadly reactive than the other two. Complement-fixing antibodies declined more rapidly following experimental infection than did those measured by neutralization or immunofluorescence. The results are discussed in relation to the diagnosis of EHV-1 infection and the significance they may have for the epidemiology of this disease.  相似文献

The etiological participation of different viruses in the pneumonia complex of cattle is undisputed. The significance of the individual viruses in the genesis of disease symptoms is still mostly unclear. Since the pneumonia complex of cattle is a multifactorial syndrome, vaccination programs as the only measure of prophylaxis in most cases do not show the desired effect. Against those viral agents causing self-standing diseases of the respiratory tract, specific vaccinations are applicable with success. Prerequisite for their effectiveness, however, is the exact etiological clarification of the cause of disease or of the existing infections respectively in the herd. The frequency of possible viral agents in infections of the respiratory tract in cattle from herds of the province of Schleswig-Holstein found in routine diagnostic work during the last four years is presented in addition.  相似文献

Infectious respiratory disease in dogs is a constant challenge because of the involvement of several pathogens and environmental factors. Canine respiratory coronavirus (CRCoV) is a new coronavirus of dogs, which is widespread in North America, Japan, and several European countries. CRCoV has been associated with respiratory disease, particularly in kenneled dog populations. The virus is genetically and antigenically distinct from enteric canine coronavirus; therefore, specific tests are required for diagnosis.  相似文献
