一株具有杀松材线虫活性细菌的筛选和鉴定   总被引:1,自引:0,他引:1  
为了筛选对松材线虫(Burrmphezen曲“5xylophilus)具有较高杀线活性的细菌,作者从腌制芥菜的表面分离出了一株对松材线虫具有较高杀灭活性的细菌,命名LZ~01。经形态学观察、生理生化分析及对该菌16SrDNA序列分析,将该茵鉴定为BacilluscereusLZ-01。利用该菌培养滤液对松材线虫的杀灭活性进行了检测,结果表明:处理10h时松材线虫的校正死亡率可达100%。  相似文献

The gene coding for green fluorescent protein (GFP) from the jellyfish Aequorea victoria was successfully used as a vital marker for the transformation of three woody plant species, black spruce (Picea mariana (Mill.) BSP), white pine (Pinus strobus L.) and poplar (Populus spp.). The gfp gene and the gene conferring resistance to kanamycin (nptII) were introduced by microprojectile bombardment or Agrobacterium tumefaciens-mediated technology. Screening by fluorescence microscopy of the transformed plant material, under the selection of kanamycin, identified five to eight cell lines from each tree species that clearly expressed GFP. Expression of GFP was observed in somatic embryonal cells of the coniferous species and in stem sections of poplar. For all species, GFP transgene expression was stable over multiple subcultures. Stable integration of the gfp gene into plant genomes was confirmed by Southern hybridization or polymerase chain reaction (PCR) analysis. We conclude that GFP can be used as a vital marker and reporter protein in transformation experiments with gymnosperms and deciduous trees.  相似文献

采集人工种植2年的铁皮石斛,将根、茎和叶研磨为粉末,采用NA培养基分离纯化其内生细菌,酶切去除冗余,利用16S rRNA基因序列同源性分析,对铁皮石斛根、茎和叶中分离得到的49株内生细菌进行分离鉴定。结果表明,获得的49株内生细菌在根、茎、叶中分别有24株、15株和10株,分别属于芽孢杆菌属的5个种、假单孢杆菌属的1个种和沙雷氏菌属的1个种。分离出的内生细菌表现出一定程度的细菌区系相似性,也表现出细菌区系的器官特异性。铁皮石斛内生细菌在石斛根部分布的属、种数量明显比茎、叶多,土壤可能是铁皮石斛内生细菌的主要来源;铁皮石斛不同部位内生细菌群落结构存在一定差异,群落种数根>茎>叶。研究获得荧光假单胞杆菌20株,占总内生细菌数量的40.82%,判断其可能为优势菌株。  相似文献

The electrolyte conductivity and activity of dehydrogenase of bare-root seedlings of both Chinese fir (Cunninghamia lanceolata (Lamb.)Hook.)and Masson pine (Pinus massoniana Lamb.) under freezing and desiccation treatments were studied.The results showed that needle electrolyte conductivity of both speices increase significantly after freezing treatment and there are no significant differences in needle electrolyte conductivity between the two species.The dehydrogenase activity(ARD) of fine roots of both Chinese fir and Masson pine was negatively correlated with increasing freezing and desiccation.The results suggest that both electrolyte conductivity and dehydrogenase activity could be used as quick indicators of Chinese fir and Masson pine bare-root seedling quality.  相似文献

Organic acid exudation from the roots of Chinese fir (Cunninghamia lanceolata) and Masson pine (Pinus massoniana) seedlings under low phosphorus stress was studied using the solution culture method. The results revealed that organic acid exudation from the roots of Chinese fir and Masson pine seedlings under low phosphorus stress increased. Compared with P3 (KH2PO4, 0.5 mmol/L), the average organic acid exudation from the root of Masson pine seedlings under P0 (KH2PO4, 0 mmol/L), P1 (KH2PO4, 0.03 mmol/L) and P2 (KH2PO4, 0.09 mmol/L) increased by 328.6%, 267.9% and 126.4% respectively. Masson pine from Zhejiang Province in China had the highest organic acid exudation. Under low phosphorus stress, the increase in organic acid exudation from the different provinces of Chinese fir and Masson pine varied. Masson pine from Zhejiang Province mainly increased oxalic acid, tartaric acid, citric acid and malic acid exudation, that from Guangxi Province mainly increased oxalic acid and tartaric acid exudation, and that from Guizhou Province, China mainly increased oxalic acid, tartaric acid and malic acid exudation. We examined effects of nutrient availability and changing root zone temperature (RZT) on growth, gas exchange and plasma membrane H(+)-ATPase (PM-ATPase) activity of roots of 1-year-old Scots pine (Pinus sylvestris L.) seedlings during spring flushing. The 6-week growth-chamber experiment was carried out in hydroponic cultures that supplied the seedlings with low (0.5 mM N) or high (3 mM N) nutrient concentration and two rates of increase in RZT were simulated: slow warming (SW-treatment) and fast warming (FW-treatment). Air temperature, humidity, and light conditions were similar in all treatments. Growth of roots and shoots was retarded at low RZT, and fresh mass increment of roots was closely correlated with RZT sum. High nutrient availability increased nitrogen concentrations of needles and stems, but only at RZTs >13 degrees C. Low RZT and low availability of nutrients suppressed gas exchange of the seedlings. Real PM-ATPase activity was highly dependent on RZT. At high RZTs, real PM-ATPase activity was affected by nutrient availability but this effect was related to root growth. We conclude that, under conditions of high nutrient availability, Scots pine seedlings can compensate for the suppressive effects of long-term exposure to low RZT by rapidly accelerating growth, gas exchange and root metabolism, but only when RZT has increased above a threshold value, which was 13 degrees C in this study.  相似文献

Reported in this paper are our findings of the study on changes in species diversity of different Masson pine communities after the invasion of Bursaphelenchus xylophilus (Steiner and Buhren) Nickle and subsequent and different removal disturbance management approaches. Based on the results, the species diversity indices for the arbor layers of the different communities can be arranged in the following order (from high to low): 1) broad-leaved stand after the removal of all infected pine trees in the pure pine stand in Fuyang; 2) lightly infected Masson pine and Schima superba mixed stand in Fuyang; 3) uninfected stand mixture of Masson pine and Castanopsis fargesii as a control; 4) lightly infested pure Masson pine stand in Fuyang; 5) Quercus variables stand formed following the selective removal of infected pine trees from a mixed Masson pine and Q. variables stand in Zhoushan Islands; 6) pure young Masson pine stand formed following the removal of all infected pine trees from a pure Masson pine stand; 7) pure Liquidambar formosana stand after the removal of infected pine trees from a pure pine stand in Zhoushan Islands; 8) a mixed stand consisting of Pinus thunbergii and the Masson pine in Zhoushan Island; and 9) moderately infected Masson pine stand in Zhoushan Islands. All the three diversity indices (R 0 = richness, H′ = Shannon-Wiener index, and E = evenness) for the shrub layer did not show any significant differences among the various communities except for the pure pine stand in Zhoushan Island, which had the lowest diversity. The three indices for the herb layer of the pure young Masson pine, Q. variables stand, and L. formosana stand were higher than that of other stands. The integrated analysis showed that the Masson pine forest in different geographical situations and extent of damage had distinct disparity, just as in different disturbance degrees and restoring manners. We created the “index of disturbing intensity of stump and fallen woods” (IDISFW) to represent the degree of disturbance of tree removal to plant diversity. We found that the relationship between the species diversity index and the IDISFW shows both a lower and higher degree of removal disturbance that follows the “mid-altitude bulge” theory. Specifically, both excessive and insufficient removal of infected trees will cause a decline of plant species diversity to a certain degree. 从盘龙参[Spiranthes sinensis(Pers)Ames]的根、茎、叶中共分离得到49株内生真菌,经形态观察,鉴定为3目、4科、9属。其中曲霉属、镰刀孢属、丝核菌属为优势种群,分别占已分离菌株数的16.3%、14.3%和14.3%。不同组织部位所分离得到的内生真菌在种群及数量上都存在差异:根中的优势属为镰刀孢属,占到了根中分离菌种数的23.5%;叶中的优势属是链格孢属,占到叶中分离菌种数的26.3%;茎中的优势属为长蠕孢属,占到叶中分离菌种数的23.1%。表明盘龙参内生真菌的分布具有一定的组织专一性。  相似文献

The main aim of this study was to assess critically the effects of binucleate Rhizoctonia (BnR) inoculation on Scots pine (Pinus sylvestris L.) seedling and hypocotyl root architecture and early plant growth in tree seedling nursery soil, Sphagnum peat and forest humus. The BnR isolates (251, 266, 268, 269) stimulated early seedling growth in the nitrogen-limited nursery soil 86 days postinoculation (p.i.). These seedlings exhibited significantly higher root length and reduced root width, although percentage root infection levels were <6%. At a harvest 240 days p.i., no significant plant and root growth differences were identified, although short root numbers were significantly increased. BnR infection detected in roots was characterized by the presence of intercellular fungal hyphae and subtending intracellular monilioid fungal cells located in outer cortical cells of long roots. Similar endophytic infection morphology was detected in adventitious roots generated in prerooted hypocotyl cuttings [Kaparakis, G. & Sen, R. (2006). Scandinavian Journal of Forest Research, 21] exposed to all BnR isolates in nursery peat and forest humus, although isolate-specific cutting growth was detected. In conclusion, these BnR isolates, known to be genetically related to orchid mycorrhizal Ceratorhiza spp., are non-pathogenic and have the capacity to stimulate significantly early Scots pine root development in nursery soils and forest humus.  相似文献

采用绿色荧光蛋白基因标记技术,研究了假单胞茵(Pseudomonassp.)YT3的标记茵YT3-gfP在土壤中的定殖能力。结果表明,gfP基因可以在YT3中稳定表达,且质粒在无选择压力的情况下可以随YT3稳定遗传。YT3主要定殖于樱桃根际土壤中,根际土壤中YT3的数量是非根际土壤中的8.75~28.77倍。YT3在樱桃根际土壤中经历一个快速下降的过程后(0~20d),在第20~40d数量有所上升,第40~80d数量保持平稳,第80~120d缓慢下降。以上分析认为,YT3能够成功进行绿色荧光蛋白标记,主要是定殖在樱桃的根际土壤中,且能长期稳定的定殖。  相似文献

In order to understand the impact of formaldehyde/urea (F/U) mole ratio on penetration characteristics of urea–formaldehyde (UF) resin into softwood tissues, a quantitative measurement of UF resin penetration into radiata pine (Pinus radiata) tissues from the bond-line was undertaken. Four different F/U mole ratios (1.6, 1.4, 1.2, and 1.0) of UF resins with different viscosities and two levels of hardener (NH4Cl) for two extreme F/U mole ratios (1.6 and 1.0) were studied. Firstly, field emission scanning electron microscope and confocal laser scanning microscopy were used to localize UF resins in the bond-line for the qualitative evaluation of resin penetration. Then light microscopy was employed to quantitatively measure the resin penetration and bond-line thickness. A decrease in the F/U mole ratio of UF resin that proportionately decreased the resin viscosity resulted in an increase in the average resin penetration and a decrease in the bond-line thickness. Higher hardener level provided a greater resin penetration with all F/U mole ratio UF resins. These results demonstrated that F/U mole ratio had an impact on the penetration and bond-line thickness of UF resins, owing to differences in the reactivity of resins, with higher F/U mole ratio resins being more reactive.  相似文献

In the province of Québec, Canada, the majority of planted jack pine (Pinus banksiana Lamb.) seedlings are produced in rigid wall containers. More than 95% of them exhibit deformations of the root system which may induce stem instability. Studies of the root architecture of planted jack pine have been limited to a 30 cm radius from the stem, as barely any studies have been devoted to naturally regenerated stands. Moreover, only a few researches have focused on temporal evolution of root systems. The aim of the present study was to characterize the architectural, spatial, and temporal development of jack pine roots in natural and planted stands. Study sites were located in the continuous boreal forest of Quebec. The plantation was done in 1987, so that the trees were 15 years old at the time of sampling. Trees from natural stand had regenerated after a fire in 1983 and were 13–16 years old. The root systems of 14 jack pine trees per site were manually excavated up to a <5 mm diameter, without regard to their distance from the stem. The number, length, diameter, and the spatial and temporal development of roots were analyzed according to three scales of root architecture: the root system, axes, and segments. Overall, the numbers and lengths of roots were higher with planted pines. However, naturally regenerated trees displayed a better distribution of their roots around the stem and at depth, combined with more rapid length growth during the first years. In natural stands, all the trees had a taproot and 30% of the main roots originated at a depth of more than 20 cm, and they are regularly distributed around the stems. Planted trees did not present a taproot and 97% of the main roots originated in the first 20 cm beneath the soil surface. Moreover, 50% of root length was located in one-third of the area surrounding the stems, an area that corresponded to the furrow. Finally, the annual development of lateral roots in planted stand displayed a 5-year delay when compared with natural stand, which also affected maximum growth length and development of the branching pattern. Root distribution and temporal development are known to play a major role in the stability of aerial parts. Seedling production methods, container type, site preparation and planting techniques need to be examined in greater detail in order to assess their effect throughout the development of the root system. It is necessary to compare different sylvicultural practices and with natural/planted stands to gain a clearer understanding of this problem.  相似文献

1.5年生马尾松植株接种松材线虫后,苯丙氨酸解氨酶(PAL)、过氧化物酶(POD)、多酚氧化酶(PPO)的活性水平均高于对照,活性高峰通常出现在症状表现前夕,并在植株发病后下降.叶内PAL和POD活性高峰的出现晚于茎,与叶部病害发展一致.茎中酚类物质的含量随着线虫侵染时间的延长逐渐增加,至发病时达到高峰.酚类物质含量的变化与PAL、POD、PPO 3种酶活性的变化趋势基本一致.松材线虫的侵染明显影响寄主植物的苯丙烷类代谢,由此可知,松材线虫病的发生发展与松材线虫诱导感病寄主苯丙烷类代谢的异常变化有关.  相似文献

Individual ponderosa pine (Pinus ponderosa Dougl. ex Laws.) seedlings were grown in mesocosms with three densities of blue wild-rye grass (Elymus glaucus Buckl.) (equivalent to 0, 32 or 88 plants m-2) to determine if the presence of a natural competitor alters the response of ponderosa pine seedlings to ozone. After 3 years of ozone exposure, grass presence reduced total ponderosa pine dry mass by nearly 50%, whereas ozone alone had no significant effect on ponderosa pine growth. The combination of ozone and grass further reduced needle, stem and branch dry mass significantly below that induced by grass competition alone. Root:shoot ratios increased in response to the combined grass and ozone treatments. Grass competition significantly reduced soluble sugar concentrations in all ponderosa pine tissue components examined. Starch concentrations were highly variable but did not differ significantly between treatments. Ozone significantly reduced soluble sugar concentrations in fine roots and stems. In the absence of grass, ozone-treated seedlings tended to have higher tissue N concentrations than controls. In the presence of grass, ozone-treated seedlings had lower N concentrations than controls, resulting in a significant interaction between these two stresses in 1- and 2-year-old needles. Needle C:N ratios decreased in response to grass competition, as a result of increased N concentration and no change in C concentration. The opposite response was observed in ozone-treated seedlings as a result of decreased N concentrations, indicating that ozone-treated seedlings were unable to take up or retain as much nitrogen when grown in the presence of grass. We conclude that ponderosa pine seedlings are more susceptible to ozone when grown in competition with blue wild-rye grass.  相似文献
