Sixteen crossbred sows (Yorkshire x Duroc) were used to determine the effect of recombinantly derived porcine somatotropin (pST) on lactational performance and the pattern of plasma metabolites and growth rate of nursing pigs. Daily s.c. injections of either pST (8.22 mg.sow-1.d-1) or excipient were administered at 1000 on d 12 through d 29 of lactation. Jugular cannulas were inserted in three sows/treatment and hourly blood samples were collected on d 11 to 13 and d 28 to 29 of lactation to determine the effect of treatment on plasma concentrations of somatotropin, glucose and nonesterified fatty acids in plasma. Milk production and weight of nursing pigs were determined pretreatment (d 9 and 10) and on d 16, 22 and 28. Milk production of sows receiving pST progressively increased above that of control sows and was 22% greater on d 28. Milk composition was not affected by treatment with pST (P greater than .10), so that the increase in yields of milk fat, lactose and solids paralleled the increases in milk yield. Total milk protein yield tended to be higher in sows receiving pST, but protein yield was greater (P less than .10) only on d 28. Pigs suckling sows treated with pST weighed .34 kg more at the end of the lactation period (P less than .05). Sows receiving pST consumed less feed (P less than .05) during the treatment period, and, as a result, lost more weight (P less than .10) and backfat (P less than .05) than control sows. Average concentrations of plasma somatotropin were elevated approximately 2.5-fold above baseline levels by exogenous pST. No acute alterations in plasma glucose or nonesterified fatty acids were observed in response to pST treatment, however, sows receiving pST had a chronic elevation of plasma glucose on d 29 of lactation.  相似文献

Two experiments were conducted to evaluate whether administration of recombinant porcine somatotropin (pST) to sows (Hampshire-Yorkshire) enhanced lactational performance. In Exp. 1, sows (n = 84) were fed a corn-soybean meal diet (17.8% CP), or a similar diet with 8% added fat, from d 108 of gestation to d 28 of lactation. Half of the sows fed each diet were injected with 6 mg/d of pST from d 108 of gestation to d 24 of lactation. Diets were fed at 2.27 kg/d from d 108 of gestation until farrowing and then were self-fed during lactation. By d 3 of lactation, litter size was standardized at 8 to 10 pigs per litter. Treating sows with pST resulted in a 10-fold increase (P less than .001) in serum somatotropin at 4 h postinjection. Serum glucose was increased (P less than .01) and serum triglycerides, creatinine, and urea N were decreased (P less than .01) by pST. During the summer, apparent heat stress occurred in pST-treated sows, resulting in 14 deaths. Most (10) of the deaths occurred just before, during, or shortly after farrowing. Fewer (P less than .08) deaths occurred when pST-treated sows were fed the diet with added fat. Sows treated with pST consumed less feed (P less than .10) and lost more backfat (P less than .10) during lactation than controls. Increasing the dietary fat did not prevent these changes. Weaning weights of pigs and milk yield of sows (estimated by deuterium oxide dilution) were not affected by pST treatment. In Exp. 2, sows (n = 42) were injected weekly with 0 or 70 mg of pST on d 3, 10, 17, and 24 of lactation. Litters were standardized by d 3 at 8 to 10 pigs, and sows were fed the same control (low fat) diet as in Exp. 1. Sows treated with pST consumed less feed and lost more weight and backfat during lactation than untreated sows. Litter size, average pig weaning weights, and milk yield were not influenced by pST treatment. These data indicate that a 6-mg daily injection of pST from 6 d prepartum to d 24 of lactation or a 70-mg weekly injection of pST from 3 d postpartum to d 24 of lactation does not increase milk production in lactating sows.  相似文献

The introduction of specific genes into the genome of farm animals and its stable incorporation into the germ line has been a major technological advance in agriculture. Transgenic technology provides a method to rapidly introduce "new" genes into cattle, swine, sheep, and goats without crossbreeding. It is a more extreme methodology, but in essence, not really different from crossbreeding or genetic selection in its result. Methods to produce transgenic animals have been available for more than 20 yr, yet recently lines of transgenic livestock have been developed that have the potential to improve animal agriculture and benefit producers and/or consumers. There are a number of methods that can be used to produce transgenic animals. However, the primary method to date has been the microinjection of genes into the pronuclei of zygotes. This method is one of an array of rapidly developing transgenic methodologies. Another method that has enjoyed recent success is that of nuclear transfer or "cloning." The use of this technique to produce transgenic livestock will profoundly affect the use of transgenic technology in livestock production. Cell-based, nuclear transfer or cloning strategies have several distinct advantages for use in the production of transgenic livestock that cannot be attained using pronuclear injection of DNA. Practical applications of transgenesis in livestock production include enhanced prolificacy and reproductive performance, increased feed utilization and growth rate, improved carcass composition, improved milk production and/or composition, and increased disease resistance. One practical application of transgenics in swine production is to improve milk production and/or composition. To address the problem of low milk production, transgenic swine over-expressing the milk protein bovine alpha-lactalbumin were developed and characterized. The outcomes assessed were milk composition, milk yield, and piglet growth. Our results indicate that transgenic overexpression of milk proteins may provide a means to improve swine lactation performance.  相似文献

Classic studies in rodents conducted in the 1950s showed that growth hormone (GH) is essential for mammary development both in the pubertal phase and during pregnancy. Since then, a considerable number of experiments have been carried out in ruminants to investigate the role of GH for regulation of normal mammary development and to examine the possibility of enhancing mammary growth by administration of GH. The available evidence demonstrates that GH treatment stimulates mammary growth before puberty, but the data do not convincingly support the idea that the effect is translated into increased milk yield. GH treatment during late pregnancy seems to stimulate both mammary growth and milk yield during lactation. The limited data concerning the effect of GH on mammary growth during lactation indicate that mammary growth is unaffected by GH treatment in early lactation, whereas GH seems to increase the amount of mammary parenchyma in mid-lactation. The mechanism of action of GH remains a puzzle, but the effect of exogenous GH most likely involves insulin-like growth factor-I (IGF-I). Full understanding of the role of endogenous GH for regulation of normal mammary development requires more knowledge about the interaction between GH and IGF-I and the interplay between the GH-IGF-I axis and locally produced factors, including receptors, binding proteins, and growth factors.  相似文献

The effects of long-term administration of recombinant bovine somatotropin (rbST) on glucose turnover and the utilization of glucose in the mammary gland using a continuous infusion of [3-3H]glucose and [U-14C]glucose in lactating crossbred Holstein Friesian(HF) cattle were investigated. Glucose turnover of rbST-treated animals was significantly higher than those of control animals ( P  < 0.05) in mid lactation, while plasma glucose concentrations were not affected. The utilization of glucose of non-mammary tissues of rbST-treated animals significantly increased ( P  < 0.05) as lactation advanced. The glucose taken up by the mammary gland in early lactation increased flux through the lactose synthesis and the pentose cycle pathway with significant increases in NADPH formation for fatty acid synthesis during rbST administration. The utilization of glucose carbon incorporation into milk appeared to increase in milk citrate and milk triacylglycerol but not for milk lactose as lactation advances in rbST-treated animals. The stimulant effect for milk yield by rbST treatment was transiently and significantly increased in early lactation and was decreased in late lactation even though there was a high level of udder blood flow. These findings demonstrate that the regulation of biosynthetic capacity within the mammary gland would be influenced more by local than by systemic factors. The proportion of glucose would be metabolized less for lactose synthesis, but metabolized more via the Embden-Meyerhof pathway and the tricarboxylic acid cycle as lactation advances.  相似文献

本研究收集了3个千头以上奶牛场共8872条相关信息,采用多因素方差分析法分析了不同产犊季节、胎次和牛场对泌乳天数、305d校正奶量、305d脂肪产量、305d蛋白产量和全泌乳期产奶量5个泌乳性能指标的影响。结果表明,不同产犊季节、胎次和牛场都极显著影响奶牛的该5项泌乳性能(P〈0.01)。夏季和秋季产犊奶牛的泌乳性能较为理想。夏季产犊奶牛的泌乳天数和全泌乳期产奶量最高,其他三个泌乳性能指标都位居第二;秋季产犊奶牛的305d校正奶量、脂肪产量和蛋白产量都显著高于其他三个季节(P〈0.05),泌乳天数和全泌乳期产奶量也仅次于夏季。头胎牛的各项泌乳性能都显著高于其他胎次的奶牛(P〈0.05),其次为2胎产犊奶牛。随着胎次的增加,泌乳性能的各项指标都有不同程度的降低。不同的牛场极显著地影响奶牛的泌乳性能(P〈0.01)。  相似文献

Twenty-four barrows were used to investigate the effects of exogenous porcine somatotropin (pST) administration (0 and 100 micrograms.kg-1.d-1) between 30 and 60 kg on longissimus muscle morphology and meat tenderness of pigs grown to 90 kg. Administration of pST was by daily i.m. injection. Pigs were fed a fortified diet in restricted amounts between 30 and 60 kg and had ad libitum access to this diet from 60 to 90 kg. Excipient pigs had fewer alpha R fibers and more alpha W fibers than pST-treated pigs (P less than .05). Administration of pST increased (P less than .05) muscle fiber area for all three fiber types at both the 60 kg (34%) and 90 kg (29%) slaughter weights. Furthermore, pST administration increased shear-force (30% at 60 kg; 19% at 90 kg) of the longissimus muscle, indicating that pST administration reduced meat tenderness. All the pST-treated pigs exhibited the "giant fiber syndrome" and a high proportion (62%) of pST-treated pigs exhibited pale, soft, exudative muscle. Whether the giant fiber anomaly resulted from increased muscle protein accretion rate or fiber degeneration is unknown. Results indicated that the stimulatory effects of pST on muscle growth are sustained following cessation of hormone treatment. However, the incidence of pale, soft, exudative muscle in 62% of the pST-treated pigs indicates that pST potentially has undesirable side effects.  相似文献

Experiments were performed to examine the effects of long‐term treatment with recombinant bovine somatotropin (rbST) on milk plasmin–plasminogen and milk composition by one injection of 500 mg of rbST in every 14 days throughout lactation in crossbred Holstein cattle. The animals receiving rbST gave a greater milk yield and rate of blood flow to the udder during early lactation than the control group. The milk lactose concentration remained constant while the milk protein concentration increased as lactation advanced in both groups. The level of milk fat in rbST‐treated animals was significantly greater than in controls in the early lactation period (P < 0.05). The milk sodium and chloride concentrations of the rbST‐treated animals significantly decreased in early lactation as compared with the control animals. The sodium : potassium ratio of the rbST‐treated animals was significantly lower than those of control animals in the early lactation (P < 0.05) and it markedly increased in late lactation. As lactation advanced, the concentration of plasmin in the milk gradually increased, while the milk plasminogen concentration significantly increased in both groups. The plasminogen : plasmin ratio decreased in the control animals while it increased in the rbST‐treated animals as lactation advanced. These findings demonstrate that rbST is involved the activity of the plasmin–plasminogen system but is not involved in maintaining tissue integrity in the mammary gland during late lactation in crossbred dairy cattle.  相似文献

分布于红原及其毗邻地区的牦牛具有较优良的产奶性能和乳质特性。该地区牦牛184天产奶量为211 30kg;乳脂率6 82%,乳蛋白5 60%,乳糖5 20%,均高于黑白花奶牛。暖季适当补饲可提高泌乳黑犏牛日产奶量0 77kg;冷季半舍饲+补饲可使泌乳黑犏牛全泌乳期产量平均提高1096kg,延长泌乳期109~137天。畜群结构调控和常规繁殖技术,可显著提高适龄母牛的群体比率和繁殖率,从而提高群体产奶水平。应用现代生物技术,有望解决牦牛种间杂交过程中存在的一些问题,如提高改良犏牛的繁殖成活率,扩大改良规模、实现改良犏牛繁殖改良犏牛等,可提高牦牛改良的经济效益。  相似文献

选用22d断奶SD大鼠高频饮用牛奶或葡萄糖40d,分别记录各组动物的阴道开张时间和阴道黏液、每周日增重、卵巢及子宫所占体重百分比、血糖水平,收集卵巢进行组织学处理以观察卵泡发育情况,探讨高频饮用牛奶和葡萄糖对雌性大鼠初情期的影响。结果显示,奶组和糖组大鼠较对照组阴道开张时间来得早,预示初情期可能提前,而且,奶组和糖组大鼠的卵巢占体重百分比较对照组高(P<0.05)。牛奶组血糖水平比对照组高,但糖组与对照组血糖水平差异不显著。通过对卵巢进行石蜡切片、HE染色及卵泡计数,结果显示:奶组的闭锁卵泡、每张切片卵泡总平均数都比对照组多(P<0.05),而葡萄糖组的黄体数比对照组少(P<0.05)。试验表明高频饮用牛奶和葡萄糖可对性成熟前雌性大鼠的生殖机能产生一定影响。  相似文献

To examine the effects of exogenous porcine (p) ST on measures of stress and immune function in weaned pigs with or without transport, pigs (20 +/- 1 d of age) received daily injections of pST (0.5 mg/kg; n = 16) or saline (n = 16) for 5 d. On d 5, a blood sample was collected immediately before injection. At 4 h postinjection, pigs were weighed, sampled for blood, injected with di-nitrophenyl-conjugated keyhole limpet hemocyanin, and weaned. One half of the pigs in each group were transported for 3 h before placement in the nursery. Pigs were weighed, and blood was collected on 1, 7, and 14 d postweaning. Statistical significance was set at P < 0.05. Serum IGF-I concentrations were increased by pST and decreased by weaning, but not affected by transport. The free cortisol index was elevated in all pigs 1 d postweaning, although less in transported versus nontransported pigs. By 7 d postweaning, the free cortisol index returned to prewean values. Serum concentrations of immunoglobulin (Ig) G increased in all pigs by 14 d postweaning, but were not affected by pST or transport. Serum IgM concentrations were elevated at 7 and 14 d postweaning. Before weaning and again 1 d postweaning, pigs treated with pST had greater concentrations of IgM than did control animals. Circulating neutrophils increased in pST-treated pigs 4 h after the final pST injection. Improved immune function in weaned pigs by pST may lead to greater health and growth in a commercial setting.  相似文献

为探索奶牛泌乳早期乳中尿素氮含量对产后繁殖性能的影响,本研究选取海丰奶牛场的头胎荷斯坦牛1932头,于产犊后30d内采集乳样,测定乳中尿素氮含量,并收集试验牛只产后繁殖资料,用多因素方差分析法分析乳中尿素氮对产后繁殖性能的影响。结果表明,试验牛只产犊后1个月内乳中尿素氮含量为9.22±2.78mg/dL;乳中尿素氮含量对个体情期受胎率有显著影响(P〈0.05),而对产后首次配种天数有极显著影响(P〈0.01),乳中尿素氮含量大于15mg/dL的奶牛产后第一次配种天数显著高于低于此数值的奶牛,且个体情期受胎率显著低于此数值低的奶牛。同时乳中尿素氮含量与个体情期受胎率呈极显著负相关(P〈0.01)。据此认为,作为反映能氮平衡的关键指标,泌乳早期乳中尿素氮含量可作为预测奶牛产后繁殖性能的指标加以利用。  相似文献

This study was conducted in South Wollo Zone of Amhara Regional State, Ethiopia, to assess the milk yield and reproductive performance of indigenous and crossbred cattle under smallholder management conditions. Questionnaire survey was used to collect retrospective data on the performance of dairy cattle in 186 households. Thirty two postpartum cows (16 indigenous and 16 crossbred) were selected purposively and monitored for five months and milk samples were collected twice per week from each cow to determine postpartum ovarian activity using ELISA techniques. Daily milk yields at all stages of lactation were significantly (P < 0.01) higher in crossbred than indigenous cows. Crossbred cows had significantly (P < 0.01) longer lactation length and shorter age at first calving, while calving interval was significantly shorter in cows that gave previous calves during the wet season than the dry season. Only six cows (18.8%) showed heat signs on an average of 136 days postpartum of which one was a crossbred and five were indigenous. While, 11 cows (34.3%) resumed ovarian activity until 150th day postpartum among which three were crossbred and eight were indigenous.  相似文献

The ontogeny of the somatotropin/insulin-like growth factor system was examined in well-fed pigs under basal conditions and during a short-term challenge of porcine ST (pST). The study was conducted with two replicates of eight castrate male pigs from 3.8 kg BW (10 d of age) to 92 kg BW (129 d of age). Pigs were reared individually with ad libitum access to milk replacer through 23 d of age. Thereafter, pigs were fed a corn, soybean meal, and dry skim milk diet formulated to exceed nutrient requirements by approximately 30%. Pigs were randomly assigned to receive daily i.m. injections of either 0 (buffer) or 120 microg/kg BW of pST for a duration of 4 d starting at 10, 19, 33, 43, 63, 83, and 125 d of age. Blood was collected via jugular venipuncture on d 0 and 4 of the challenge. Circulating levels of IGF-I were not dramatically affected by age, but levels of IGF-II were low from 10 to 19 d of age and then increased through later stages of growth. Circulating concentrations of IGF binding protein (BP)-3 increased with age (P < .05), but levels of IGFBP-2, a 30-kDa IGFBP, and IGFBP-4 were unchanged (P > .10). The pST challenge reduced plasma urea nitrogen at all ages, but the magnitude of the response was less in younger pigs compared with the maximum response in pigs greater than 30 kg BW (63 d of age). The IGF-I response to the pST challenge also increased from approximately 30% in young pigs to a threefold increase in older pigs. Regardless of age, concentrations of IGF-II were minimally affected by the pST challenge. Circulating levels of IGFBP-3 increased and IGFBP-2 levels decreased in response to the pST challenge, and the magnitude increased with age. The high nutritional status of pigs in the early phases of growth diminished the postnatal changes in IGF-I and IGFBP-2, but not IGF-II or IGFBP-3. Overall, data demonstrate a developmental regulation of the ST/IGF system, with pST challenges altering circulating concentrations of IGF-I, IGFBP-3, and IGFBP-2 coincident with changes in amino acid metabolism.  相似文献

Thirty-seven pigs with an initial live weight of 60 kg were used to investigate the effects of daily exogenous porcine somatotropin (pST) administration at two dose levels (0 and 100 micrograms.kg-1.d-1) for a 31-d period on muscle fiber characteristics and meat tenderness of boars, gilts and barrows. Excipient boars and gilts had more alpha W and fewer alpha R fibers than did those receiving pST. The percentage of muscle fiber type for barrows was not affected by pST treatment. The administration of pST resulted in an increase in muscle fiber size for all three fiber types in all three sexes, but these changes were of greater magnitude in barrows (31.8%) and gilts (27.8%) than in boars (9.3%). Somatotropin negated the intrinsic sex effect differences in fiber area of the pigs. There was no difference in tenderness among excipient boars, barrows and gilts; however, with the inclusion of pST, shear force decreased in boars and gilts and increased in barrows. A high proportion of the pST-treated pigs contained giant fibers in the longissimus muscle. Furthermore, a small proportion of the pST-treated pigs exhibited pale, soft, exudative muscle. Whether the giant fiber anomalies occurred through increased muscle activity or from fibers undergoing degenerative changes was not determined.  相似文献

The maintenance energy required to sustain the cow herd is a major cost of beef production. This work proposes modifying parameter estimates for a population-specific lactation curve with genetic evaluations for the maternal genetic effect on calf gain from birth to weaning to provide inputs for a commonly used prediction of energy requirement. Daily milk production (y) was modeled as a function of stage of lactation (T, d) using the function y = AT(B)exp(-CT) modified to incorporate effects of genetic evaluation for the maternal effect on calf gain from birth to weaning and age of dam. A 1-kg increase in predicted maternal breeding value for calf gain from birth to weaning from within-herd genetic evaluation increased the lactation curve parameter A by 10.3+/-4.6% and reduced the B parameter by 1.0+/-0.6%. Similarly, a 1-kg increase in maternal breeding value for gain from birth to weaning from national cattle evaluation increased the A parameter by 1.7+/-0.2%. Corresponding estimates of peak milk yield and time of peak lactation were derived for individual animals from their genetic evaluation. Additional inputs for predicting maintenance energy requirements were derived from genetic evaluations for birth weight and mature size. The methodology is demonstrated using genetic evaluations of sires from the Miles City Line 1 Hereford population. Further refinement and application of this methodology may facilitate characterization of beef cattle seedstock for their potential genetic contributions to profitability.  相似文献

During three years (1982 to 1984) data were collected concerning diseases and reasons for culling in 612 dairy farms with Brown Swiss, Simmental and Black and White cattle. The frequency of culling increases in all three breeds with increasing age, from 20% at the first lactation up to 33% from the 4th lactation on. The main reasons for disposal are insufficient production, reproductive problems or udder diseases. After treatment for reproductive problems, mastitis or acetonemia, the risk for culling within the same lactation period is increased. Reproductive problems (no heat symptoms, repeat breeder, ovarian cysts) and acetonemia increase with increasing milk production in all three breeds. In cows with high 100-day-performance they are also more frequent. The occurrence of acute mastitis shows no general trend with increasing milk production or with high 100-day-performance. No systematic relationship is found between reproductive disorders or acute mastitis and persistency of milk production. On the other hand, acetonemia is somewhat more frequent in cows showing higher persistency.  相似文献

A monoclonal antibody (mAb), PS-7.6, to porcine somatotropin (pST) significantly enhanced the growth responses to pST injections in hypophysectomized (hypox) rats but could not be tested in pigs because of the large quantity of antibody required for a growth trial. Because pST inhibits the hypoglycemic effects of insulin, an insulin tolerance test procedure was established to measure pST activity in jugular-catheterized pigs. Doses of 0, 30, 100, and 300 μg/kg per day of pST were split and administered subcutaneously (sc) in equal portions twice daily for 2 d. After a 17-hr fast, plasma samples were obtained at 10-min intervals for 30 min before an intravenous injection of insulin (0.08 IU/kg) and then for an additional 50 min. Because pST increased fasting plasma glucose concentrations, preinsulin glucose values were used as a covariate to adjust the postinsulin concentrations. pST caused a dose-dependent increase in resistance to the insulin injection in these pigs. The areas under the curves (AUC) for plasma glucose were 22.l, 29.0, 39.0, and 47.2 mg/dl per min for the 0, 30, 100, and 300 μg/kg pST doses, respectively. Because different doses of pST could be detected, the PS-7.6 enhancement of pST treatment was evaluated. In the first experiment, five pigs/group each received sc injections of either vehicle, pST (75 μg/kg; 3.0 mg/d), pST (75 μg/kg) + PS-7.6 at 3.75 mg/kg, or pST (75 μg/kg) + PS-7.6 at 15 mg/kg for 2 d before the insulin test. The pST and PS-7.6 were combined and incubated for at least 1 hr at room temperature before being injected. The injection of pST alone did not significantly change insulin tolerance activity (23.1 vs. 21.1, AUC, but insulin resistance was enhanced when this dose of pST also included PS-7.6 (27.4 and 29.5, AUC, respectively; P < 0.05). In a second experiment, the effects of PS-7.6 and PS-4.2, a mAb that did not potentiate the pST-stimulated growth of hypox rats, were compared. The five pigs/treatment received either vehicle, pST (75 μg/kg), pST (75 μg/kg) + PS-7.6 (3.75 mg/kg), or pST (75 μg/kg) + PS-4.2 (3.75 mg/kg) for 2 d. The administration of pST increased the resistance to insulin (26.7 vs. 18.8, AUC; P < 0.01), which was markedly potentiated by PS-7.6 (54.3, AUC, P < 0.001) but not affected by PS-4.2 (27.6 AUC. The injection of PS-7.6 at 7.5 mg/kg without exogenous pST did not alter the sensitivity to insulin. These results indicate that PS-7.6, but not PS-4.2, enhanced the insulin antagonistic activity of pST in swine, suggesting that an enhancement of pST-stimulated growth would also occur in PS-7.6-treated pigs.  相似文献
