One hundred and forty-seven Chinese barley varieties maintained at the Gene Bank of the National Barley Improvement Centre, Zhejiang, and 84 progenies from these varieties were tested at the seedling stage for their reaction to 32 selected pathotypes of Blumeria graminis f. sp. hordei. Eighteen resistance spectra were identified comprising single or combined resistances from eight known (Ml(Bw), Ml(Ch), Mla7, Mla8, Mla9, Mla13, MlaRu4 and Mlg) and six unknown resistance genes. The most frequent gene was Ml(Bw), which was found in 69 varieties and previously detected in only a few European winter barley varieties. The genes Mla8 and Ml(Ch) were also often present, but other resistance genes were rare. After inoculation, considerably fewer pathogen colonies were observed in ‘Aiganqi’ and one line of ‘Tong5’. Twenty varieties were composed of lines with different resistance genes. China is likely to be a region of origin of the genes Ml(Bw), Mla7, as well as three unknown genes found in original landraces and perhaps another three unknown genes detected in cultivars bred in China. The resistances of varieties from the Zhejiang province and those originating from 11 other Chinese provinces were quite different. Unfortunately, none of the varieties are promising sources of resistance to powdery mildew and China does not seem to be a region suitable for identifying such sources.  相似文献

Temporal variation of diversity in Italian durum wheat germplasm   总被引:1,自引:0,他引:1  
The aim of this work is to analyse the temporal change of genetic diversity in Italian durum wheat germplasm. The germplasm deployed in this study (158 accessions), belonging to 5 different historical classes, was characterised for its microsatellite and gliadin markers. The level of genetic diversity (He), based on gliadin and SSR markers results – on average – greater in indigenous landraces present in Italy before 1915, with the exception of pure line material which had been selected from landraces (showing highest level of heterozigosity for gliadin markers). Genotypes obtained from crosses or mutagenesis (referring to the 1950–1960 period) along with those resulting from crosses between CIMMYT lines and old materials (1970s and beyond) were also genetically more diverse. Forty-nine percent of indigenous landraces were genetically heterogeneous. Nine out of 53 landrace accessions were able to capture 4 different SSR private alleles. It is speculated that the reduction of allele richness is an indicator of the genetic erosion of the pre-breeding germplasm and it is pointed out that the implementation of appropriate methods of genetic conservation of this germplasm is a priority for breeding and food safety.  相似文献

In 1999, natural populations of Dactylis glomerata L., Festuca arundinacea Schreb., Phalaris tuberosa L. and the macrotherm species Brachypodium rupestre (Host) R. et S., were collected as seed in Southern Italy (Apulia, Basilicata and Campania regions) to evaluate the potential as forage of native germplasm under dry conditions, using available commercial cultivars as controls. The persistence of accessions, biometric, phenologic, productive and qualitative parameters of forage have been studied for a 3-year period (2000–2002) in a typical Mediterranean environment. D. glomerata is widely present in the studied area with two subspecies (subsp. glomerata L., subsp. hispanica (Roth) Nyman). The research pointed out significant differences in plant size, earliness and single plant dry matter (DM) production. All the natural ecotypes belonging to this species showed higher persistence than the control cultivars. Some natural ecotypes appeared to have potential for improving DM yield (subsp. glomerata ecotypes) and reducing neutral detergent fibre (NDF) content (subsp. hispanica ecotypes). Natural ecotypes belonging to F. arundinacea, P. tuberosa, and B. rupestre are sporadically present in the studied area. Among them, two ecotypes of P. tuberosa, showing higher winter growth and earliness compared to cv. ‘Holdfast’, seem to be interesting for a future breeding programme.  相似文献

Although common bean (Phaseolus vulgaris L.) is cultivated throughout India, the Himalayas hold largest diversity of bean germplasm. No studies on characterization of phaseolin types on this germplasm have been conducted earlier. In order to determine whether the common bean cultivars collected from various areas in the Northern Himalayas represent introductions from the Central American and Andean domestication centers or are local domesticates, we have analyzed the electrophoretic variation (SDS-PAGE) of phaseolin types in several bean accessions. A few species of Vigna were also included in this study to determine whether phaseolin (vignin in Vigna) patterns can be used to resolve the Phaseolus–Vigna complex. The present investigation on phaseolin (globulin) patterns of Phaseolus vulgaris and Vigna spp. clearly shows much variability in globulin patterns. Three new types of phaseolin patterns were recorded. An attempt to resolve phylogenetic problems in this complex was made using the phaseolin data.  相似文献

Lentil anthracnose (Colletotrichum truncatum (Schwein.) Andrus et W.D. Moore is a potential threat in many lentil (Lens culinaris Medik.) production regions of North America. In the lentil germplasm maintained in Germany and North America, 16 lines were reported to have resistance to race Ct1, but none has resistance reported to race Ct0. The objective of this study was to examine accessions of wild Lens species for their resistance to races Ct1 and Ct0 of lentil anthracnose. Five hundred and seventy-four wild accessions of six species and control lines were screened in two replications under both field and greenhouse conditions using a 1–9 scoring scale (1, highly resistant; 2–3, resistant; 4–5, moderately resistant; 6–7, susceptible; and 8–9, highly susceptible). Indianhead and PI 320937 were resistant while Eston and Pardina were susceptible to race Ct1 as expected. However, none of the check lines were resistant to race Ct0. Among the six Lens wild species tested, accessions of Lens ervoides (Brign.) Grande had the highest level of resistance, 3–5 to race Ct1 and Ct0 followed by L. lamottei Czefr. in the field and greenhouse. Lens orientalis (Boiss.), L. odemensis L., L. nigricans (M. Bieb.) Godron and L. tomentosus L. were highly susceptible, 8–9 to race Ct0 in the greenhouse. The highest frequency of resistance, especially in L. ervoides (Brign.) Grande, was found in accessions originating from Syria and Turkey. The usefulness of these L. ervoides (Brign.) Grande accessions as sources of resistance to the more virulent race of anthracnose in a lentil breeding program is discussed.  相似文献

Lupinus angustifolius L. is a Mediterranean species, domesticated in the 20th century, representing an important grain legume crop in Australia and other countries. This work is focused on the collection of wild germplasm and on the characterisation of morphological and molecular diversity of germplasm accessions. It reports the collection of 81 wild L. angustifolius accessions from the South and Centre of Portugal, available at the ‘Instituto Superior de Agronomia Gene Bank’, with subsequent morphological and molecular characterisation of a selection of these and other accessions. A multivariate analysis of morphological traits on 88 L. angustifolius accessions (including 59 wild Portuguese accessions, 15 cultivars and 14 breeding lines) showed a cline of variation on wild germplasm, with plants from Southern Portugal characterised by earlier flowering, higher vegetative development and larger seeds. AFLP and ISSR molecular markers grouped modern cultivars as sub-clusters within the wider diversity of wild germplasm, revealing the narrow pool of genetic diversity on which domesticated accessions are based. The importance of preserving, characterising and using wild genetic resources for L. angustifolius crop improvement is outlined by the results obtained.  相似文献

The opium latex of 1470 individual plant samples of 98 germplasm lines were screened for five major economic alkaloids. The alkaloids profile showed that morphine content ranged from 9.20–20.86% with an average of 15.00 ± 0.267. Codeine ranged from 1.69–6.48% with arithmetic mean of 3.35 ± 0.091, thebaine from 0.52–7.95% with an average of 2.27 ± 0.116. Narcotine ranged from 8.79–17.97% with an average 8.79 ± 0.266 and papaverine content ranged from 0.00–6.07% with an average 1.18 ± 0.167 in over all the germplasm lines. The content of different alkaloids categorized into class interval which exhibited that maximum number of plants and accessions for morphine content fall in group of 10–15% followed by 15–20%, for codeine content in group of 2–4% followed by 4–6%, for thebaine content in 1–2% followed by 2–4%, for narcotine in 5–10% followed by 10–15% and for papaverine content 0–2%. 24 germplasm lines had morphine content above 16.0%. Similarly, 37 germplasm lines for codeine and 13 for thebaine showed consistently high content, i.e., above 3%. 33 germplasm lines had narcotine content above 9% and 20 lines exhibited papaverine content above 2%. Line BR 316 was found unique having thebaine content 6.27–9.33%. The probable reasons of increase or decrease of different alkaloids based on their biosynthetic pathway has been discussed in detail.  相似文献

Knowledge of the effects of farmer practices on population genetic parameters of peach palm (Bactris gasipaes Kunth) is relevant to the improvement and conservation of the palm’s genetic resources. Microsatellite markers were used to assess genetic diversity and population structure of peach palm in swidden-fallow agroforestry systems in northeastern Peru. The study covered eight communities, comprising two study areas 160 km apart – one occupied by indigenous Amerindians and the other by mixed race campesinos. Simultaneous analysis of an ex situ peach palm germplasm collection provided a means to compare population genetic parameters. Farmers who were surveyed on seed selection practices for peach palm reported that an average of only four palms (4.3 for campesino and 1.5 for indigenous populations) were used to provide seed for the establishment of the forest gardens sampled. As expected, inbreeding coefficients observed within communities were relatively high (f = 0.105 − 0.210), however, observed heterozygosities within communities were also high (0.625–0.741). A metapopulation approach was used to describe migration within and among regions, implying a hierarchical structure of gene flow which maintains relatively high levels of genetic diversity. Seed migration was found to occur over longer distances (≤600 km) and at a higher frequency (46% of palms sampled) in the indigenous study area, and a proportionally greater number of alleles was found (49 vs. 43 over three loci) with twice as many private alleles occurring only in the indigenous populations. The farmers’ practice of preserving remnant palms through successive swidden generations may have contributed to the maintenance of alleles by reducing the severity of founder effects. Although the campesino study area exhibited a significant (20% of the variation; p < 0.01) isolation-by-distance relationship across 35 km distance, in general, both study populations had relatively limited genetic structure (θ = 0.012–0.03), which is believed to have resulted from the exchange of seeds over long distances and periods of time.  相似文献

Phenotypic diversity was assessed for quantitative and qualitative traits in a salt-tolerant subset of the international safflower (Carthamus tinctorius L.) germplasm collection originating from 11 countries in three regions (Central Asia, Southwest Asia and Africa) of the Middle East. Phenotypically, the germplasm, among and within regions, was highly variable, especially for rosette- and yield-related traits. Frequency of desirable variants of seven agronomically important traits ranged from 14% for long rosette period to 50% for no or few spines. Level of population differentiation was high for number of capitula per plant (30%), whereas most traits partitioned their diversity (82–87%) within populations. Region-specific nonrandom associations among sets of qualitative traits and the existence of broad morphological and phenotypic diversity in this germplasm were supported by the large number of log-linear models needed to describe qualitative trait associations, the high number of principal components needed to account for total variability, and the low discriminatory power of phenotypic traits among germplasm from regions and countries in the Middle East. These results suggest that adaptation of the species to the wide spatial and temporal variation in the Middle East resulted in a multitude of ecotypes and in enormous amount of local variation. A multivariate selection criterion for high biological and seed yield, long rosette period and no or few spines identified five accessions from Southwest Asia that can be introduced into subsistence farming systems as a multipurpose crop under saline agriculture.  相似文献

Wild soybean (Glycine soja), the progenitor of soybeans, has become an important germplasm source for soybean breeding. The Chinese G. soja germplasm collection includes two subcollections, one typical wild soybean (G. soja) and the other ‘big seed type’ (semi-wild soybean, G. gracilis). In order to understand the nature of this big seed type subcollection, a considerable number of 1,185 accessions were investigated focusing on phenotypes, compositional structure and genetic diversity of this subcollection. The results showed that there were 617 combined morphotypes formed by six major morphological traits. The morphotype number and diversity gradually decreased among four sections from small to large 100-seed weight sizes. The big seed subcollection accumulated distinctly high frequencies for white flower, grey pubescence, seed non-bloom and four seed coat colours (green, yellow, brown and black), differing from the typical G. soja. Between the two subcollections, only the compositional structures of leaf shapes were almost in agreement. A rare strip-shaped leaf only appeared in typical G. soja and small-seeded grade (100-seed wt) of the big seed type. Seed non-bloom trait showed that the increased frequencies were concomitant with the augmenting of 100-seed weight. The data suggested that white flower accumulated rapidly in the big seed type, followed by seed non-bloom, while the slowest rate of accumulation was grey pubecence, and that the ‘large-seeded type (2.51–3 g 100-seed wt)’ of typical G. soja and the ‘smallest-seeded type (3.01–3.5 g 100-seed wt)’ of the big seed type could have a closer genetic relationship. The morphotype number and genetic diversity showed reduced values with increased 100-seed weights. The geographical distribution of genetic diversity indicated two tendencies: decreased from north to south and also from east to west. Our results suggested that Northeast China was the first centre of morphological diversity for the big seed type, and the North was the secondary centre in China.  相似文献

Landraces of rice (Oryza sativa L.) are valuable sources of genetic variation that have been lost in advanced cultivars. Seeds of a rice landrace stored for almost 100 years were found on Sado Island in Niigata prefecture, Japan. This report aims to present basic data on the genetic variation of this landrace, which was known as ‘Echizen’. Five samples of ‘Echizen’, consisting of two old samples, one sample maintained on farm, and two lines regenerated from old seeds were compared with other advanced cultivars and landraces using 19 microsatellite markers. Among the five samples of Echizen, the two stored samples showed greater diversity than the other samples. Cluster analysis based on the UPGMA method also showed that old Echizen was a diverse landrace that could cover the genetic diversity of most Japanese rice cultivars.  相似文献

The Hessian fly, Mayetiola destructor (Say), is a major pest of wheat in North Africa, southern Europe, North America, and northern Kazakhstan. It is believed this pest (like wheat) originated in West Asia. The Syrian Hessian fly biotype has been found to be the most virulent worldwide, and has been used at the International Center for Agricultural Research in the Dry Areas (ICARDA) for screening wheat and its wild relatives to identify new sources of resistance. The screening was conducted in an insect rearing room set at 20°C and 70% RH using a Hessian fly population collected from Lattakia region, Syria. The experimental design was a randomized complete block with four replications. ‘Nasma’ (bread wheat) and ‘Cando’ (durum wheat) were used as susceptible and resistant checks, respectively. A total of 623 lines/accessions of wheat and its wild relatives (Aegilops and Triticum) were evaluated. Twenty-nine Aegilops accessions and four synthetic derived bread wheat lines were found resistant. The presence of dead first instars confirmed the resistance reaction and also showed that antibiosis is the major mechanism of resistance in these materials. These sources of resistance are used in ICARDA’s wheat breeding programs for the development of Hessian fly-resistant germplasm/varieties.  相似文献

To achieve higher yields and better soil quality under rice–legume–rice (RLR) rotation in a rainfed production system, we formulated integrated nutrient management (INM) comprised of Azospirillum (Azo), Rhizobium (Rh), and phosphate-solubilizing bacteria (PSB) with phosphate rock (PR), compost, and muriate of potash (MOP). Performance of bacterial bioinoculants was evaluated by determining grain yield, nitrogenase activity, uptake and balance of N, P, and Zn, changes in water stability and distribution of soil aggregates, soil organic C and pH, fungal/bacterial biomass C ratio, casting activities of earthworms, and bacterial community composition using denaturing gradient gel electrophoresis (DGGE) fingerprinting. The performance comparison was made against the prevailing farmers’ nutrient management practices [N/P2O5/K2O at 40:20:20 kg ha−1 for rice and 20:30:20 kg ha−1 for legume as urea/single super-phosphate/MOP (urea/SSP/MOP)]. Cumulative grain yields of crops increased by 7–16% per RLR rotation and removal of N and P by six crops of 2 years rotation increased significantly (P < 0.05) in bacterial bioinoculants-based INM plots over that in compost alone or urea/SSP/MOP plots. Apparent loss of soil total N and P at 0–15 cm soil depth was minimum and apparent N gain at 15–30 cm depth was maximum in Azo/Rh plus PSB dual INM plots. Zinc uptake by rice crop and diethylenetriaminepentaacetate-extractable Zn content in soil increased significantly (P < 0.05) in bacterial bioinoculants-based INM plots compared to other nutrient management plots. Total organic C content in soil declined at 0–15 cm depth and increased at 15–30 cm depth in all nutrient management plots after a 2-year crop cycle; however, bacterial bioinoculants-based INM plots showed minimum loss and maximum gain of total organic C content in the corresponding soil depths. Water-stable aggregation and distribution of soil aggregates in 53–250- and 250–2,000 μm classes increased significantly (P < 0.05) in bacterial bioinoculants-based INM plots compared to other nutrient management plots. Fungal/bacterial biomass C ratio seems to be a more reliable indicator of C and N dynamics in acidic soils than total microbial biomass C. Compost alone or Azo/Rh plus PSB dual INM plots showed significantly (P < 0.05) higher numbers of earthworms’ casts compared to urea/SSP/MOP alone and bacterial bioinoculants with urea or SSP-applied plots. Hierarchical cluster analysis based on similarity matrix of DGGE profiles revealed changes in bacterial community composition in soils due to differences in nutrient management, and these changes were seen to occur according to the states of C and N dynamics in acidic soil under RLR rotation.  相似文献

The difficulty and problems encountered in the study of cultivated plants, in general and sugarcane, in particular has been indicated. In order to understand these problems, a brief review on the taxonomy of Saccharum and closely related taxa, namely, Erianthus, Sclerostachya, Narenga and Miscanthus (generally known as ‘Saccharum complex’) has been given. A short account on the important morphological features that are specific to sugarcane has also been stated as classification is commonly based on such morphological characters. A note has been added on the chromosome number, origin, and distribution of the species of ‘Saccharum complex’ members. Taxonomic keys have been devised for identification of the genera of Saccharinae and for the species of Saccharum and Erianthus occurring in India. A new combination, Sclerostachya fallax (Balansa) Amalraj et Balasundaram, has also been proposed.  相似文献

Information on genetic diversity and genetic relationships among genotypes of Brassica carinata is currently limited. The objectives of this study were to evaluate patterns and levels of genetic diversity in B. carinata based on amplified fragment length polymorphisms (AFLP) as compared with Brassica juncea and Brassica nigra, and to evaluate agronomic and seed quality data for plants grown in the field in western Canada. A total of 296 AFLP bands were generated from four primer pair combinations and scored for presence/absence in 66, 20 and 7 accessions of B. carinata, B. juncea and B. nigra, respectively. B. carinata was less genetically diverse than the other two species. Differences in diversity were evident in the proportion of polymorphic loci within each species: 23, 35 and 50% for B. carinata, B. nigra and B. juncea, respectively. Pair-wise similarity measures based on the Jaccard coefficient were highest among accessions of B. carinata and showed the narrowest range: 0.911 (0.810–0.981) compared to B. nigra: 0.569 (0.438–0.660) and B. juncea: 0.715 (0.345–0.951). AFLP-based genetic distance information can be used by plant breeders to select diverse genotypes. AFLPs are also useful for fingerprinting cultivars and two primer pair combinations were sufficient to uniquely identify all the accessions of B. carinata. More variation among accessions was identified in the agronomic trial than had previously been described in studies of B. carinata in western Canada, but the data were too limited to draw conclusions regarding specific accessions. Overall, the findings were in agreement with other published work describing the favourable agronomic potential of this species.  相似文献

Artemisia pallens Bess. is a low volume and high value essential oil plant used in perfumery, cosmetic and flavouring industries. On account of the failure of conventional procedures to induce variability in species, mutation techniques have been tried in our experiments. Dry and viable seeds (moisture content 8%) of homozygous pure breeding lines were subjected to 150–500 Gy doses of gamma rays and 0.01–0.1% ethyl methane sulphonate (EMS) for 8 h. Desirable qualitative mutants were recovered from segregating M2 generation (4,283 plants scored) raised as single plant progenies. The spectrum of morphological mutants included late and early flowering types; bushy and high yielding types; tall and more capitula-producing types and high oil and high davanone yielding types. These were raised through M3 families to evaluate stability and transmission of mutant characters. As such out of 15 different types selected in M2, only 11 types bred true to their characteristic variability. Based on their performance, the mutants were characterised depending upon their distinguishing features. Davanone, the main component of oil showed the maximum increase (64.22% against 54.64% in control) in mutant ‘S–5’ recovered from exposure with 250 Gy γ-rays. Mutant ‘E-6’ was economically most viable having increased oil biosynthesis (0.36% against 0.22% in control) and hence yields higher oil per unit area than the parental control (isolated from 0.05% EMS treatment).  相似文献

Vigna vexillata is considered as a pantropical distributed wild species closely related to the cowpea (Vigna uniculata) and adapted to infertile soils. The species is occasionally used for its storage roots as well as forage and erosion control plant. The objective of this study was to pursue personal communications that V. vexillata has be transformed into a cultivar in Indonesia. Seven Indonesian islands were visited (Java, Bali, Sumba, Flores, Timor, Kalimantan and Sulawesi). Wild V. vexillata was found in Java, Bali, Sumba, Flores and Timor, occasionally used as wild ‘forest food’, and cultivated V. vexillata was found in Bali and Timor, Seven cultivars were collected and two of these were made available for the National Botanic Garden of Belgium. The cultivars were primarily cultivated for their storage roots, propagated by seeds, required no scarified seeds for good germination and formed non-dehiscent pods. On-farm root yields of 18–30 t ha−1 and seed yields of 0.7–1.2 t ha−1 were estimated. A brief discussion about the common names of wild V. vexillata is given and it is proposed to use the name ‘tuber cowpea’ for V. vexillata accessions which are cultivated for their storage roots. The material may be of interest for regions, where the growing season is too short or the rainfall too low for sweet potato and cassava as well as to incorporate cultivar characteristics into wild V. vexillata accessions which are used for their storage roots.  相似文献

Information regarding the amount of genetic diversity is necessary to enhance the effectiveness of breeding programs and germplasm conservation efforts. Genetic variation between 21 switchgrass genotypes randomly selected from two lowland (‘Alamo’ and ‘Kanlow’) and one upland (‘Summer’) synthetic cultivars were estimated using restriction fragment length polymorphism (RFLP) markers. Comparison of 85 RFLP loci revealed 92% polymorphism between at least two genotypes from the upland and lowland ecotypes. Within ecotypes, the upland genotypes showed higher polymorphism than lowland genotypes (64% vs. 56%). ‘Kanlow’ had a lower percent of polymorphic loci than ‘Alamo’ (52% vs. 60%). Jaccard distances revealed higher genetic diversity between upland and lowland ecotypes than between genotypes within each ecotype. Hierarchical cluster analysis using Ward's minimum variance grouped the genotypes into two major clusters, one representing the upland group and the other the lowland group. Phylogenetic analysis of chloroplast non-coding region trnL (UAA) intron sequences from 34 switchgrass accessions (6 upland cultivars, 2 lowland cultivars, and 26 accessions of unknown affiliation) produced a neighbor-joining dendrogram comprised of two major clusters with 99% bootstrap support. All accessions grouped in the same cluster with the lowland cultivars (‘Alamo’ and ‘Kanlow’) had a deletion of 49 nucleotides. Phenotypic identification of greenhouse-grown plants showed that all accessions with the deletion are of the lowland type. The deletion in trnL (UAA) sequences appears to be specific to lowland accessions and should be useful as a DNA marker for the classification of upland and lowland germplasm.  相似文献
