The effects of fruit and vegetable extract (Oxxynea) on plasma cholesterol, early atherosclerosis, cardiac production of superoxide anion, and NAD(P)H oxidase expression were studied in an animal model of atherosclerosis. Thirty six hamsters were divided into two groups of 18 and fed an atherogenic diet for 12 weeks. They received by gavage either water or Oxxynea in water at a human dose equivalent of 10 fruits and vegetables per day. Oxxynea lowered plasma cholesterol and non-HDL cholesterol, but not HDL-cholesterol, and increased plasma antioxidant capacity. It also strongly reduced the area of aortic fatty streak deposition by 77%, cardiac production of superoxide anion by 45%, and p22phox subunit of NAD(P)H oxidase expression by 59%. These findings support the view that chronic consumption of antioxidants supplied by fruits and vegetables has potential beneficial effects with respect to the development of atherosclerosis. The underlying mechanism is related mainly to inhibiting pro-oxidant factors and improving the serum lipid profile.  相似文献

Both selenium and allophycocyanin (APC) have been reported to show novel antioxidant activities. In this study, a fast protein liquid chromatographic method for purification of selenium-containing allophycocyanin (Se-APC) from selenium-enriched Spirulina platensis and the protective effect of Se-APC on 2,2'-azobis(2-amidinopropane) dihydrochloride (AAPH)-induced oxidative stress have been described. After fractionation by ammonium sulfate precipitation, and separation by DEAE-Sepharose ion-exchange and Sephacryl S-300 size exclusion chromatography, Se-APC with purity ratio (A652/A280) of 5.30 and Se concentration of 343.02 μg g(-1) protein was obtained. Se-APC exhibited stronger antioxidant activity than APC by scavenging ABTS (2,2'-azinobis-3-ethylbenzothiazolin-6-sulfonic acid) and AAPH free radicals. The oxidative hemolysis and morphological changes induced by AAPH in human erythrocytes were effectively reversed by coincubation with Se-APC. Lipid oxidation induced by the pro-oxidant agent cupric chloride in human plasma, as evaluated by formation of conjugated diene, was blocked by Se-APC. The accumulation of malondialdehyde, loss of reduced glutathione, and increase in enzyme activities of glutathione peroxidase and reductase induced by AAPH in human erythrocytes were effectively suppressed by Se-APC. Furthermore, Se-APC significantly prevented AAPH-induced intracellular reactive oxygen species (ROS) generation. Taken together, our results suggest that Se-APC demonstrates application potential in treatment of diseases in which excess production of ROS acts as a casual or contributory factor.  相似文献

Plant studies have been carried out to identify the nature and extent of the formation of adducts with DNA bases when treated with pesticide formulations. DNA extracted from crop plants after treatment with pesticide formulations has yielded evidence of adduct formation. The extent of DNA modification has been established by (32)P postlabeling studies. The radiochromatograms from (32)P postlabeling of isolated plant DNA from grapes, bush beans, soybeans, pumpkins, and cucumbers show elevated adduct levels in treated vegetable plants as compared with untreated controls. A number of different adduct spots appear, likely indicating adduct formation with pesticide molecules or their metabolites. The DNA adducts from hexenal and 4-hydroxy-2-nonenal were clearly observed, indicating oxidative stress and lipid peroxidation in the plant.  相似文献

Two hydroponic culture experiments were conducted to investigate cadmium (Cd)‐induced oxidative stress in winter wheat (Triticum aestivum L.) seedlings and the effects of L‐Galactono‐1, 4‐lactone (GalL), the biosynthetic precursor of the antioxidant ascorbate (AsA), on the oxidative stress induced by Cd. In experiment 1, with application of Cd (0, 10, 25, 50 µM) in nutrient solution, hydrogen peroxide (H2O2) and malondialdehyde (MDA) levels as well as membrane permeability in both shoots and roots were significantly increased, indicating Cd‐induced oxidative stress and lipid peroxidation as well as plasma‐membrane damage in the plants. In experiment 2, H2O2 levels in plants exposed to Cd were significantly reduced by the addition of GalL (25 mM), associated with increased activities of peroxidase (POD), indicating that GalL alleviated the oxidative stress induced by Cd. Unexpectedly, however, the MDA levels were not reduced by the addition of GalL. Does Cd also induce lipid peroxidation directly besides via formation of reactive oxygen species (ROS)? The effects of continuous application of cattle manure on the quantity and quality of soil organic matter (OM) were investigated in an Ando soil (Melanudand). Surface soil samples were periodically taken from NPK and NPK+ manure (80, 160, and 320 Mg ha-1 y-1) plots over a period of 20 y. Particulate (>53 µm) and mineral-associated <53 µm) OM fractions were separated from the soil samples by sieving after:mechanical dispersion. For the NPK treatment, both the organic C and total N concentrations of the whole soil continued to decrease, the reduction reaching about 10% after 20 y. Manure application at the rate of 80 Mg ha-1 y-1 did not lead to an increase in the amounts of organic C and total N in the whole soil. In contrast, both the organic C and total N concentrations increased by the application of 160 and 320 Mg ha-1 y-1 manure. Manure application at the rate of 320 Mg ha-1 y-1 increased the organic C concentration by 30% and total N concentration by 48% after 20 y. The decrease in the organic C and total N concentrations in soil with NPK fertilization was attributed to a decrease in the amount of mineralassociated OM. Manure application increased significantly the amount of particulate OM, while it did not affect the amount of mineral-associated OM. It also resulted in a decrease in the C/N ratio of soil OM, especially of the particulate OM. The analysis of humic acids showed that manure application induced a decrease in the degree of humification and an accumulation of high molecular weight components. The quantitative'and qualitative changes of OM in the Ando soil upon manure application were mainly due to the accumulation of manure-derived particulate OM.  相似文献
