Twelve dogs with lymphosarcoma and hypercalcemia were treated over a period of 36 months. Signs and laboratory findings were referable to hypercalcemia and azotemia. All dogs were staged, classified histologically, and given cytoreductive chemotherapy, using 5 drugs (vincristine sulfate, cytosine arabinoside, cyclophosphamide, L-asparaginase and prednisone). For azotemia, symptomatic therapy (0.9% NaCl solution and furosemide) was given. Seven dogs responded completely, with marked reduction of lymphadenopathy and return of serum calcium concentration to normal. Median duration of remission in this group was 48 days (range, 14 to 93), and median survival time was 112 days (range, 85 to 153). Five nonresponding dogs had less than 50% reduction in measurable tumor mass, although serum calcium concentration returned to normal. The median survival time for this group was 34 days (range, 23 to 68). Two of the nonresponders died from sepsis and another from disseminated intravascular coagulation. Response to therapy did not appear to be influenced by age, breed, sex, initial calcium concentration, degree of azotemia, or histologic classification.  相似文献

Renal function was investigated in 12 dogs with lymphosarcoma and accompanying hypercalcaemia. There was no age or breed predilection. Renal function was evaluated using the endogenous clearance of creatinine to estimate glomerular filtration rate and urinary and fractional excretion of endogenous substances. Standard serum biochemical determinations and urinalyses were also employed. Serum calcium concentrations were high (>2·99 mmol/litre) in ail dogs, while serum phosphorus concentrations approximated or were below those of normocal-caemic dogs in most cases. Hypercalcaemic nephropathy and impaired renal function were evident in all 12 dogs studied. Ten of the dogs (83 per cent) were isosthenuria and 11 (92 per cent) were hypercalciuric. Endogenous creatinine clearance was reduced by more than 50 per cent in all dogs examined. Urinary and fractional excretions of calcium, phosphorus, sodium, chloride and potassium were increased over reference values. The observed alterations in renal function could be attributed to the direct effects of a high concentration of intrarenal calcium on renal tubular function and haemodynamics and to the presence of a renotropic factor that interacted specifically with the parathyroid hormone receptor in the kidney.  相似文献

Canine appendicular osteosarcoma (OSA) is a commonly diagnosed cancer that is capable of inducing pathologic bone remodeling. Investigating surrogate indices of bone metabolism may contribute to the diagnostic and therapeutic management of bone malignancies in companion animals. This study evaluated the excretion of N-terminal telopeptide (NTx), a marker of bone resorption that is detected in urine. Sixty-three dogs with appendicular OSA were compared with 29 age-matched healthy dogs. Dogs with appendicular OSA had significantly higher baseline urine NTx excretion than healthy controls (P < .0001). In 17 dogs with OSA treated with either amputation or standardized palliative therapies, significant reductions in urine NTx excretion were observed, suggesting that excessive bone resorption in dogs with OSA may be linked with focal skeletal osteolysis or its consequences. To identify any relationship between indicators of pathologic bone turnover, baseline urine NTx excretion was correlated with serum bone alkaline phosphatase (bALP) or radiographic tumor lengths at diagnosis. No significant correlations were identified between baseline urine NTx excretion and either bALP or tumor length. The findings from this study suggest that high urinary NTx excretion may support the diagnosis of focal skeletal osteolysis in dogs, and reductions in urine NTx excretion after treatment may reflect elimination or minimization of pathologic bone resorption.  相似文献

Subsequent to a possum (Trichosurus vulpecula) poisoning operation purportedly using 1% phosphorus baits, six dogs with access to poisoned possums died of phosphorus poisoning. Two dogs survived, following treatment with oral copper sulphate and parenteral vitamin K. Clinical signs included depression, jaundice, vomiting and bloody diarrhoea. Post-mortem lesions included large areas of subcutaneous, interstitial and intermuscular haemorrhage, subserosal haemorrhage and liver degeneration. Free phosphorus was detected in the ingesta of three of the dogs which died up to 7 days after the last of the poison was laid. Analysis of two batches of baits used showed P levels of 1.17% and 1.24%.  相似文献

Three experiments were conducted to compare the excretion of water-soluble phosphorus (P) of starter, grower, and finisher pigs fed corn-soybean meal-based P-deficient basal diet containing no added inorganic P (B), P-adequate diet (the basal diet with added dicalcium phosphate; B + P), the basal diet plus 500 units of microbial phytase/kg (B + 500), or the basal diet plus 1000 units of microbial phytase/kg (B + 1000). There were 6 barrows per diet, in a randomized complete block design, with an average initial body weight of 10.4, 20.0, or 51.1 kg for each of starter, grower, and finisher pigs in the three phosphorus balance experiments, respectively. In the experiments, the addition of dicalcium phosphate or phytase to the basal diet increased ( P  < 0.05) the digestibility and retention of P, and there were linear reductions ( P  < 0.05) in water-soluble P excretion as a result of supplementing the basal diet with phytase. In the starter pig experiment, phytase addition at 500 or 1000 units/kg reduced ( P  < 0.05) water-soluble P excretion by 28 or 42%, respectively when compared with the B + P diet. In the grower pig experiment, adding phytase at 500 or 1000 units/kg reduced ( P  < 0.05) water-soluble P excretion by 24 or 34%, respectively when compared with the B + P diet. The use of phytase at 500 or 1000 units/kg reduced ( P  < 0.05) water-soluble P excretion by 11 or 30%, respectively in the finisher pig experiment. The proportion of water-soluble phosphorus in total phosphorus was not affected by dietary treatment in any of the three experiments. In conclusion, adding phytase at 1000 units/kg to a corn-soybean meal-based P-deficient diet basal diet containing no added inorganic P compared with B + P diet reduced the daily excretion of water-soluble P in starter, grower, and finisher pigs by 42, 34, and 30%, respectively.  相似文献

Background

Phosphorus (P) supplementation is costly and can result in excess P excretion. This study investigated the effects of reducing dietary P on milk production and P excretion in dairy cows over a full lactation.

Method

Forty-five multiparous Holstein dairy cows were divided into 15 blocks according to expected calving date and previous milk yield, and assigned randomly to one of the three dietary treatments: 0.37, 0.47, and 0.57% P (DM basis); these P levels represent the NRC recommendations, Chinese recommendations, and the amount of dietary P commonly fed by Chinese dairy farmers, respectively. Average daily feed intake was calculated from monthly data on feed offered and refused. Milk yields of individual cows were recorded weekly, and milk samples were taken for analysis of protein, fat, solids-not-fat, lactose, and somatic cell count. Blood samples were collected on days −6, −3, 0, 3, 6 relative to calving, and then monthly throughout lactation, and analyzed for P and Ca concentrations. Spot samples of feces and urine were collected for 3 consecutive d during weeks 12, 24, and 36, and P concentrations were analyzed. Reproduction and health data were recorded.

Results

Dietary P did not affect dry matter intake or milk yield (P > 0.10). Milk fat content was slightly higher in cows fed 0.37% P than in cows fed 0.47% P (P = 0.05). Serum concentrations of P and Ca did not reflect dietary P content (P > 0.10). Fecal and urinary P both declined linearly (P < 0.05) as dietary P decreased from 0.57 to 0.37%. Fecal P content was 25% less when dietary P was 0.37% compared to 0.57%. Health events and reproductive performance were not associated with dietary P content (P > 0.05).

Conclusions

Lowering dietary P from 0.57 to 0.37% did not negatively affect milk production, but did significantly reduce P excretion into environment.  相似文献

The urinary enzyme markers of renal damage, alkaline phosphatase (AP), gamma-glutamyl transferase (GGT) and N-acetyl-beta-glucosaminidase (NAG), glomerular filtration rate (GFR) and renal biopsies were studied to evaluate renal status in dogs with pyometra. After ovariohysterectomy, urinary enzymes were measured daily for 12 days in 55 dogs, and again at a later follow-up visit. Thirteen dogs had high levels of at least one enzyme at initial presentation. Seventeen dogs had a transient increase in urinary enzyme values between one and five days after surgery. Enzyme values usually declined to low activities within 12 post-operative days. Renal biopsies demonstrated tubular abnormalities in many dogs. Mean GFR was 2.4 and 2.0 ml min(-1) kg(-1), respectively on day 1 post-operatively and at the follow-up visit 1-4 months later. High urinary enzyme values often reflected extensive lesions in renal proximal tubular cells and sometimes reduced GFR.  相似文献

Four diets were formulated to contain: 16% protein and 0.4% phosphorus--diet 1; 16% protein and 1.4% phosphorus--diet 2; 32% protein and 0.4% phosphorus--diet 3; and 32% protein and 1.4% phosphorus--diet 4. Forty-eight dogs were fed diet 1 for 3 months after surgical reduction of renal mass, then were allotted to 4 groups of 12 dogs each, with equal mean values for glomerular filtration rate (GFR). Dog of groups 1-4 were fed diets 1-4, respectively, for 24 months. Data collected from the dogs during and at termination of the study were analyzed statistically for effects of dietary protein, phosphorus (P), time, and interactions between these factors. During the 24 months of study, 24 dogs developed uremia and were euthanatized for necropsy. Necropsy also was performed on the remaining 24 dogs after they were euthanatized at the end of the study. Dog survival was significantly enhanced by 0.4% P diets (vs 1.4% P diets), but survival was not significantly influenced by amount of dietary protein. The 0.4% P diets (vs 1.4% P diets) significantly increased the period that GFR remained stable before it decreased, but dietary protein did not have significant effect. Significant blood biochemical changes attributed to P, protein, and time were identified during the study. Terminally, plasma parathyroid hormone concentration was significantly increased from prediet values in all groups of dogs. Urine protein excretion was not significantly affected by dietary amount of either protein or P, when measured by either timed urine collection or urine protein-to-creatinine ratio.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

A balance study was performed in order to quantify the effect of continuously increased phosphorus (P) intake on faecal and urinary P excretion. The aim was to quantify the level of intake where regulatory P excretion becomes relevant for comparative digestibility measurements on P, and when the pig adapts its urinary P excretion to increased P intake. Phosphorus intake of growing pigs was continuously increased on a daily basis starting at a marginal level and P excretion via faeces and urine was continuously followed for 92 days. Two semi-synthetic diets were prepared with different proportions of Na2HPO4 resulting in 2.4 (diet 1) and 6.3 (diet 2) g P/kg DM. Concentration of Ca was adapted to achieve a Ca supply approximately 3.1 fold the digestible P supply. Six castrated male crossbred pigs (31 kg BW) were kept individually in metabolism crates after they had undergone a 14 d P depletion period during which they were fed diet 1 solely. Pigs received 1.04kg of diet 1 per day throughout the experiment, and each day the amount of feed and P supplied to pigs from diet 2 was increased by 12 g and 69 mg, respectively. ME supply was approximately 2.4 fold maintenance and average daily BW gain of pigs during the entire experiment was 690 +/- 30 g. While intake increased linearly, faecal excretion of P and Ca increased non-linearly and could be best described by third order polynomial functions. The proportion of ingested P not excreted via faeces followed a quadratic type of curve with a maximum of 81% at 25 days on experiment and P intake of 4.0 g/d. Thereafter, the proportion decreased continuously. The digestibility of P from diet 2, determined by the slope ratio technique, was constant and not affected by P intake up to a P intake of 5 g/d. Renal P excretion did not exceed inevitable losses until day 60 and increased exponentially thereafter when body P reserves were restored. It is concluded, that an adaptation to surplus P supply occurred earlier on the intestinal than on the renal level. While faecal P excretion appeared regulated depending on the actual requirement for P retention, the regulation via urine depended on the P status of the pig. Once the renal P excretion of growing pigs exceeds a level of 25 mg/d, intake of digestible P cannot be regarded sufficiently low to measure P digestibility as a capacity of the feedstuff.  相似文献

Twenty-four dogs with induced, severe chronic renal failure were allotted to 2 groups of 12 each. Group-A dogs were fed a 0.4% phosphorus (P)/0.6% calcium, 32% protein diet, and group-B dogs were fed a 1.4% P/1.9% calcium, 32% protein diet. Dogs were studied over 24 months to determine clinical status, survival, blood biochemical alterations, glomerular filtration rate (GFR), urinary excretion of P and protein, renal morphologic changes, and renal tissue concentrations of calcium, P, and magnesium. Group-A dogs developed statistically significant differences from group-B dogs in several blood biochemical values (PCV and total solids, calcium, P, potassium, sodium, chloride, total CO2 (TCO2), anion gap, and parathyroid hormone concentrations) and in urinary P excretion. Mean (+/- SEM) GFR values in group-A and group-B dogs were nearly identical when diets were initiated (group A = 0.73 +/- 0.05 ml/min/kg of body weight; group B = 0.72 +/- 0.08 ml/min/kg), but significantly (P = 0.0346) lower GFR developed in group-B than in group-A dogs over time. At 24 months, GFR in survivors was 0.83 +/- 0.08 and 0.63 +/- 0.15 ml/min/kg for dogs of groups A and B, respectively. Other measurements favored the hypothesis that P/calcium restriction was beneficial, but values failed to reach statistical significance. Survival was greater at 24 months in group-A than in group-B (7 vs 5) dogs, and renal tissue concentrations of calcium and P were higher in group-B than in group-A dogs. Differences were not detected between groups in urinary excretion of protein and in the type or severity of renal lesions.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献
