根据已发表的犬瘟热病毒(Canine distemper virus,CDV)Onderstepoort株序列设计1对引物,RT-PCR法扩增出约1 000 bp的融合蛋白基因片段,通过T-A克隆技术,将PCR产物克隆至pMD18-T载体。序列分析表明CDV蓝狐分离株F蛋白基因片段由1 044 bp组成,编码348个氨基酸,与其它CDV25259株、2544-Han95株、A75-17株、DOGDK91C株、5804P株、ONP株、PDV-2株核苷酸序列同源性分别为94.5%、94.1%、94.7%、94.2%、94.2%、97.6%和94.9%;氨基酸序列的同源性分别为98.6%、97.1%、98.0%、98.0%、98.3%、97.7%和98.6%;与其它CDV毒株相比,蓝狐分离株存在核苷酸缺失突变(1 030位~1 038位)和氨基酸缺失突变(344位~348位)。  相似文献

The C-terminal part of the nucleocapsid protein gene of 13 canine distemper virus (CDV) isolates from Thailand, were analyzed. The nucleotide sequences were assigned to two clusters; cluster A exhibited a high degree of homology with the vaccine strain Onderstepoort, 99.10 and 97.61%, respectively, in the two isolates examined. Cluster B appeared closely related to virulent strains registered in the GeneBank database and to the virulent reference strain (A75/17); a total of 11 samples were analyzed, with 94.63-99.10% homology at the same position. The deduced amino acid sequences correlated with the two-nucleotide sequence clusters. However, there was no association among the CDV groups with histories of vaccination, sex, ages, clinical findings and evidence of viral antigen in tissues.  相似文献

Antiserum against canine distemper virus (CDV) was raised in pigs by intranasal inoculation with CDV strains CND65 and ROCKBORN. Immunoglobulin fractions were conjugated with horseradish peroxidase. Peroxidase-conjugated anti-CDV immunoglobulin preparations were used for the detection and titration of CDV, seal-derived (phocine) distemper virus (PDV) and rinderpest virus (RPV) in Vero cell cultures. For the detection and titration of corresponding neutralizing antibodies a direct neutralizing peroxidase-linked antibody (NPLA) assay was established. The results were compared with those obtained with the conventional microtitre neutralization test (MNT) based on CPE reading. In addition the sensitivity of an indirect peroxidase-linked antibody (PLA) assay was tested in parallel with that of the NPLA assay using sera obtained from CDV-immunized pigs.  相似文献

利用Vero-DST细胞从死亡犬、狐狸肺、脾内分离到4株犬瘟热病毒-CDV-TM-CC、CDV-Dog-SCh、CDV-Fox-SY、CDV-Fox-WF,在Vero-DST细胞上传5代没有细胞病变,5代细胞培养物经电镜、间接免疫荧光及PCR鉴定为阳性。与本实验室保存的CDV-Monkey-BJ进行基因测序,并对与致病相关的F蛋白、V蛋白进行分析,F蛋白分析5株毒具有6个潜在N-末端糖基化位点,与强毒株同源性在92%~99.7%,与疫苗株不高于93.3%,在F1亚基内有7个特有氨基酸位点,这可能与其对灵长类致病有关。V蛋白分析表明,其与强毒株同源性在94%~99.7%之间,而与疫苗株不高于93.3%,CDV-Monkey-BJ C272R突变使其Zn结合能力丧失。试验结果显示,所有分离株均为强毒株,不同宿主毒株序列存在差异,值得进一步研究。  相似文献

During the fatal seal epizootics in the North and Baltic Seas in summer 1988 a virus was isolated which was shown to be the causal agent. It was subsequently classified as morbillivirus by neutralization assays, reaction with monoclonal antibodies and nucleic acid hybridization studies. The virus (tentatively called Phocine Distemper Virus, PDV) is difficult to grow in culture making rapid diagnosis difficult. We have used the Polymerase Chain Reaction (PCR) as an alternative and fast method to detect the presence of virus-specific nucleic acid and we describe here the amplification of cell culture derived PDV RNA in a "one-tube" reaction using heterologous (Rinderpest Virus cDNA derived) F gene primers. The resulting 370 bp DNA fragment was shown to be morbillivirus derived by Southern blot hybridization using cloned RPV F gene as probe.  相似文献

Recently the isolation and characterisation of a morbillivirus which caused high mortality in common seals (Phoca vitulina) in 1988 have been reported. Because of the clinical and pathological similarity of the disease in seals to that of distemper in dogs, the name phocine distemper virus (PDV) has been proposed. There are marked differences in the virus-induced proteins of PDV compared to other morbilliviruses and the humoral immune response of moribund and dead seals to PDV was restricted to some of the internal antigens of PDV, similar to the response described earlier for canine distemper virus infection in dogs.  相似文献

应用RT-PCR的方法从2013-2015年来自我国部分地区20份犬、貂、狐和貉源犬瘟热阳性病料中克隆得到犬瘟热病毒(CDV)H基因。序列分析结果表明,20株CDV的H基因核苷酸与推导氨基酸序列的同源性分别为:90.6%~100%和90.5%~100%;相应地,与CDV3、Onderstepoort等传统疫苗株相比,其同源性分为90.4%~99.9%和89.0%~99.8%。基于CDV H基因推导氨基酸序列构建系统进化树的结果显示,除TS1510属于Amecica-1型外,其余全部为Asia-1型。表明Asia-1型犬瘟热病毒仍为目前一些地区流行基因型,弱毒感染动物也可使之产生典型犬瘟热症状。  相似文献

以250MPa的静水压力对犬瘟热病毒(CDV)处理30min之后,CDV被灭活,由其制成的灭活疫苗诱导犬产生中和抗体的能力明显优于传统的福尔马林灭活疫苗,免疫保护率达90%。电镜观察显示,经不同程度静水压力处理后的CDV粒子均发生变形,表面呈现凸起,但CDV H基因部分片段的RT-PCR扩增结果揭示,高达510MPa的静水压力作用30min也未能使其基因片段受到破坏。  相似文献

Phocine distemper virus (PDV) caused thousands of deaths among harbor seals (Phoca vitulina) from the North Sea in 1988 and 2002. To examine the effects of different factors on the pathology of phocine distemper, we performed necropsies and laboratory analyses on 369 harbor seals that stranded along the Dutch coast during the 2002 PDV epidemic. Diagnostic tests for morbillivirus infection indicated a differential temporal presence of morbillivirus in lung and brain. Seals of 3 years or older were significantly more often IgG positive than younger seals. The most frequent lesions in PDV cases were bronchopneumonia, broncho-interstitial pneumonia, and interstitial emphysema. Extra-thoracic emphysema was rare in <1-year-olds compared with older seals, even though severe pneumonia was more common. PDV cases generally had empty stomachs and less blubber than by-caught seals from before the epidemic. In PDV cases involving older animals, lung, kidney, and adrenal weights were significantly increased. Bordetella bronchiseptica was isolated from lungs in two thirds of the PDV cases examined. Our results indicate that brain should be included among the tissues tested for PDV by RT-PCR; that either phocine distemper has a longer duration in older seals or that there are age-related differences in immunity and organ development; that dehydration could play a role in the course and outcome of phocine distemper; and that bacterial coinfections in lungs are more frequent in PDV cases than gross lesions suggest. These results illustrate how quantitative analysis of pathology data from such epidemics can improve understanding of the causative disease.  相似文献

Canine distemper virus (CDV) antigen was detected in the serum of dogs by an ELISA and the results of this assay were compared with an anti-CDV immunoglobulin M (IgM) antibody test. In paired sera from 26 naturally infected dogs, the antigen-positive rate was 26.9 per cent at the first examination and 11.5 per cent at the second examination two to three weeks later. The antigen was detected in three of the 10 dogs which were negative for anti-CDV IgM antibody at the first examination. It could also be detected in the serum of between eight and two of 40 specific pathogen-free dogs vaccinated against CDV, for up to four weeks after they were vaccinated.  相似文献

Austrian field cases of canine distemper (14 dogs, one badger [Meles meles] and one stone marten [Martes foina]) from 2002 to 2007 were investigated and the case histories were summarised briefly. Phylogenetic analysis of fusion (F) and haemagglutinin (H) gene sequences revealed different canine distemper virus (CDV) lineages circulating in Austria. The majority of CDV strains detected from 2002 to 2004 were well embedded in the European lineage. One Austrian canine sample detected in 2003, with a high similarity to Hungarian sequences from 2005 to 2006, could be assigned to the Arctic group (phocine distemper virus type 2-like). The two canine sequences from 2007 formed a clearly distinct group flanked by sequences detected previously in China and the USA on an intermediate position between the European wildlife and the Asia-1 cluster. The Austrian wildlife strains (2006 and 2007) could be assigned to the European wildlife group and were most closely related to, yet clearly different from, the 2007 canine samples. To elucidate the epidemiological role of Austrian wildlife in the transmission of the disease to dogs and vice versa, H protein residues related to receptor and host specificity (residues 530 and 549) were analysed. All samples showed the amino acids expected for their host of origin, with the exception of a canine sequence from 2007, which had an intermediate position between wildlife and canine viral strains. In the period investigated, canine strains circulating in Austria could be assigned to four different lineages reflecting both a high diversity and probably different origins of virus introduction to Austria in different years.  相似文献

Forty-two cases of canine pneumonia were examined for the presence of canine distemper virus. For that purpose canine distemper virus inclusion bodies were located. The histopathological lesions were related to the presence of canine distemper antigen, as demonstrated with an immunoperoxidase technique. This technique was more sensitive for detecting canine distemper infection in lung tissue than was the study of inclusion bodies. Attention was also paid to combined infection with canine adenovirus and Bordetella bronchiseptica.  相似文献

Canine distemper virus causes a multi systemic disease in dogs often with severe neurological signs. These signs are the result of viral replication in neurons and glial cells leading to grey matter lesions and demyelination. Inflammation leads to further destruction of the tissue. As extraneural signs are often lacking and only one localisation may be found on neurological examination, distemper may be difficult to diagnose. Myoclonus is almost pathognomonic for this disease but occurs in less than half of the cases. The inflammation of the central nervous system that occurs during the chronic stage of the disease can be detected on examination of the cerebrospinal fluid, in particular by determination of the IgG index. Viral antigen can be demonstrated in cerebrospinal fluid cells by fluorescent antibody techniques. The prognosis of nervous distemper is generally poor although dogs can recover from this disease. Treatment is largely supportive and symptomatic. The importance of regular vaccination is stressed.  相似文献
