The effect of current 48-hour infusion of estradiol benzoate (EB) on the dynamics of the concentration of progesterone (P) and thyroxine (T4) was studied in four heifers in the luteal stage of their cycle. The parameters were evaluated in three intervals: infusion (A), post-infusion (B) and "uninfluenced" interval (C). In the course of infusion the concentrations of P in three animals had a markedly descending tendency. The P concentration increase in the 36th hour of infusion was maintained also during the post-infusion interval. The decrease in P in the third, "uninfluenced", interval was replaced by a new rise towards the end. One heifer showed an increase in P concentrations at the beginning of EB infusion. A decrease was recorded towards the end of the interval. The concentration decreased in the post-infusion period. A comparatively high concentration of P in the "uninfluenced" interval was again replaced by a drop towards the end in the 286th hour. The concentration of T4 in three heifers had a descending tendency in the infusion and post-infusion intervals. The decrease in the infusion interval was more pronounced. In the "uninfluenced" interval the T4 concentrations increased step by step. In one heifer the concentration of T4 slightly increased during infusion. A decrease was recorded in the post-infusion and "uninfluenced" interval. Rectal examination in the 172nd hour revealed the original corpus luteum in three animals and the production of the corpus luteum in the same ovary was found in this period in one heifer. In the 88th hour this animal showed clinical manifestations of oestrus. On the basis of the dynamics of the studied hormones and clinical studies, the thyroid hormones can be regarded as an important secondary factor involved in the permanent cyclic changes in reproduction process.  相似文献

The objective of the present study was to evaluate estrus synchronization and conception rate after progesterone releasing intravaginal device (PRID) treatment from the early luteal phase in the presence or absence of estradiol benzoate (EB) in heifers. Heifers (n=11) were assigned randomly to two treatments; insertion of a PRID containing 1.55 g progesterone with a capsule attached including 10 mg EB (P+EB; n=6) and the PRID withdrawn the EB capsule (P-EB; n=5). The PRID was inserted into the vagina on Day 2 of the estrous cycle (Day 0 was the day of ovulation) and was left for 12 days. The proportion of heifers exhibiting standing estrus within 3 days after PRID removal was 83.3% (5/6) for the P+EB group, and 80.0% (4/5) for the P-EB group, respectively. Conception rate by artificial insemination on synchronized estrus was 80.0% (4/5) in the P+EB group, and 100% (4/4) in the P-EB treatment group, respectively. These results suggest that a PRID treatment from 2 days after ovulation for 12 days in the presence or absence of EB has an effect on the synchronization of estrus and produces a beneficial conception rate in heifers.  相似文献

The effect of glucocorticoids on gonadal steroid and gonadotropic hormone concentrations and subsequent follicular activity in cows undergoing normal estrous cycles was evaluated by administration of dexamethasone (DXM) during the middle of the luteal phase. Seven cows were given physiologic saline solution twice daily from day 13 to day 17 of the estrous cycle (control experiment). During the next estrous cycle, cows were administered DXM (2 mg, IM) twice daily on days 13 through 17. Plasma specimens obtained twice daily throughout the control and DXM-treatment cycles were assayed for progesterone and estradiol concentrations. The appearance of estrus after DXM treatment was delayed until days 23 to 25 in 3 cows and was not seen by day 35 in the other 4 cows, compared with mean (+/- SD) cycle length of 22.4 +/- 3.2 in cows during the control experiment. Progesterone concentration remained significantly (P less than 0.01) high on days 19 to 23, whereas estradiol values failed to increase (P less than 0.05) on days 19 and 20 after treatment with DXM. Blood samples were obtained at 15-minute intervals for 12 hours to compare (by analysis of covariance) the effect of DXM treatment on plasma hormone concentrations on day 15 of each cycle with those of day 10. Compared with values during the control experiment, a significant (P less than 0.05) decrease was observed in the size of the pulses of luteinizing hormone (LH) and estradiol, although the number of pulses of each hormone per 12 hours was not affected when cows were given DXM. The objective of the present study was to compare the concentrations of 17 beta-estradiol, progesterone, cyclic adenosine monophosphate and cyclic quinosine monophosphate in the largest follicles of cows that persist for seven days after insemination following the preceding synchronization of oestrus and superovulation and in follicles of the luteal phase of cycle (5th-10th days). Animals included in the experiment were selected on the basis of rectal examination. Synchronization of oestrus was achieved in 24 crossbreds of Slovak Pied x Lowland Black Pied breeds (SS x Nc) using two doses of cloprostenol of Czechoslovak provenience Oestrophan Spofa, 500 micrograms in each, within 11 days. Serum gonadotrophin at the amount of 2500 I. U. was administered forty-eight hours before administration of the second dose PGF2 alpha. Experimental animals were inseminated after 72 hours. On the 7th day after mating the cows were killed at a slaughterhouse. Evaluated were only the ovaries of the 14 cows in which the persistent large follicles occurred. Ovaries of the 13 control cows in the luteal phase between the 5th-10th days were obtained at the slaughterhouse by the method after Ireland et al. (1980). Correct determination of the phase of sexual cycle was substantiated by determination of progesterone concentrations in blood serum. Follicular fluid was obtained from the largest follicles by aspiration and centrifuged in a cooled centrifuge at 3000 G. The concentrations of 17 beta-estradiol and progesterone in follicular fluid were determined using kits from URVJT at Kosice, designated RIA-test-ESTRA (SI-125-9) or RIA-test-Prog (SI-125-6).2+ persistent follicles (9.15 +/- 5.47 nmol.l-1).(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Data are presented on the effects of a short-term progesterone treatment for oestrous cycle control in cattle. Progesterone was administered by intravaginal sponge pessaries inserted for a 10-day period. Progesterone pessaries alone did not affect oestrous cycle length or corpus luteum function at either early (day 2) or midluteal (day 12) cycle stages. However, when progesterone (250 mg) and oestradiol benzoate (7-5 mg) were given intramuscularly on the day of pessary insertion corpus luteum development was inhibited in animals treated at day 2 and was regressed in animals at day 12. This combined oestrogen-progesterone treatment efficiently controlled oestrous cycle length.  相似文献

Radioimmunological investigation (RIA) of the level of pituitary FSH in the peripheral blood of cows after parturition demonstrated that this level increased successively. The FSH value of 32.93 +/- 3.71 ng per 1 ml, recorded the fourth to sixth day post partum, increased to as much as 57.78 +/- 40.98 ng per 1 ml 40 days after parturition. The LH level changed only slightly over the same period (from 1.12 +/- 0.21 ng per ml to 1.72 +/- 1.15 ng per ml). The concentration of progesterone in the blood of the cows was about 0.40 ng per ml during the first 15 days after parturition, but from the 25th day post partum it trebled, on an average (1.53 +/- 1.19 - 1.59 +/- 1.25 ng per ml). The response of the adenohypophysis of the cows to the administration of 200 micrograms of synthetic gonadoreline (spec. Dirigestran inj. Spofa) increased with increasing length of time from parturition. FSH concentration increased less markedly during the first 28 days p.p. and this increase was not uniform in time (the average increase was 1.5 to three times); in the later period the highest increase 300-500% was recorded, as a rule, 120 minutes after GnRH administration. The situation was similar in the increase in LH concentration in peripheral blood, but after the 26th day post partum the average increase in LH accounted for 500 to 600%. The concentration of progesterone in peripheral blood increased by more than 300%, on an average, from the 25th day after parturition. This testifies to the first post partum ovulation and to the formation of a new yellow body in the majority of cows under study.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

This study was designed to test the effects of progesterone or GnRH treatment on day 5 post-AI on fertility and luteal function in dairy cows and heifers. Five days after AI, 32 animals were randomly assigned to a control, intravaginal progesterone for 14 days progesterone releasing intravaginal device (PRID) or GnRH treatment group. On days 5, 7, 12, 14, 17 and 19 post-AI, each animal underwent colour Doppler ultrasonography of the corpus luteum and blood samples were collected for cell counts and plasma progesterone determination. Through general linear model repeated measures analysis of variance, significant effects were observed of treatment, parity, inseminating bull, reduced vascularization of the CL and pregnancy on plasma progesterone concentrations, whereas mean plasma progesterone and time luteal phase day, and treatment and plasma progesterone concentration on day 5 post-AI were found to, respectively, affect neutrophil and lymphocyte counts throughout the luteal phase. Moreover, two binary logistic regression analyses were performed. Based on the odds ratio, the likelihood of pregnancy by days 26-32 post-AI was 23.4 times higher in animals with high mean progesterone levels throughout the study period, compared with animals with low mean progesterone. The likelihood of reduced CL vascularization was 14 times higher in animals treated with PRID, compared with control and GnRH-treated animals. In conclusion, our results indicate that treatment on day 5 post-AI with PRID reduced subsequent CL vascularization, whereas GnRH treatment increased plasma progesterone concentrations on day 12 post-AI, although an effect was identified of the inseminating bull on plasma progesterone levels. Pregnant animals showed higher mean plasma progesterone concentrations than in nonpregnant ones and heifers higher than in lactating cows, whereas blood cell counts differed depending on the treatment and on the mean plasma progesterone concentration on day 5 post-AI.  相似文献

本实验采用放射免疫分析法(RIA)测定了11头奶牛一个发情周期里毛、唾液、乳汁中孕酮(P_4)含量。经产牛(n=8)的毛和唾液中P_4水平变化与乳汁中P_4水平变化一致。青年牛(n=3)的毛和睡液中的P_4水平变化一致,并分别与经产牛毛和唾液中P_4水平变化一致。7头配种后的经产牛和5头配种后的青年牛分别取20和23天的样品进行妊娠诊断,与配种后60天的直检结果相对照,依据经产牛毛、乳P_4水平进行妊娠诊断的阳性准确率分别是85.7%(6/7)和100%(7/7),依据青年牛毛P_4水平进行妊娠诊断的阳性准确率为100%(5/5)。  相似文献

The objective of this study was to determine the effect of exogenous progesterone administration at ovulation and during the early development of the CL, on its future sensitivity to a single administration of PGF2a in mares and cows. Horse Retrospective reproductive data from an equine clinic in the UK during three breeding seasons were used. Mares were divided into: control group, cycles with single ovulations; double ovulation group cycles with asynchronous double ovulations; and PRID group: cycles with single ovulations and treatment with intravaginal progesterone device (CIDR) immediately after the ovulation. All mares were treated with d‐cloprostenol (PGF) at either: (i) 88 hr; (ii) 96 hr; (iii) 104 hr; or (iv) 112 hr after the last ovulation. Cattle A total of nine non‐lactating Holstein cows were used. All cows were administered PGF14 d apart and allocated to one of two groups control group GnRH was administered 56 hr after the second PGF administration. CIDR group CIDR was inserted at the same time of GnRH administration. All cows were administered PGF at 120 hr post‐ovulation. The complete luteolysis rate of mares with double ovulation (66.7%) and those treated with exogenous progesterone (68.4%) was significantly higher than the rate of mares with single ovulation (35.6%) at 104 hr. In the cow, however, the treatment with CIDR did not increase the luteolytic response in cows treated at 120 hr post‐ovulation. In conclusion, the degree of complete luteolysis can be influenced by increasing the concentration of progesterone during the early luteal development in mares.  相似文献

Farm trials were carried out to determine if cows and heifers could be inseminated on a fixed time basis following a 12-day treatment with progesterone coils and an injection of 5 mg oestradiol benzoate and 200 mg progesterone at the start of treatment. The retention rate of the coils was significantly higher (P less than 0.05) when a 5.5 cm diameter was used compared with a diameter of 7.0 cm. Calving rate was similar in treated cows bred at a detected oestrus, at 56 + 74 hours after treatment or at 56 hours after treatment and injection of 100 microgram gonadotrophin releasing hormone 20 hours previously and in control cows bred at oestrus. Fertility to the first repeat oestrus was also similar in treated and control cows. Significantly (P less than 0.05) more synchronised cows calved following fixed time AI compared with the calving rate in control cows inseminated for a 24-day experimental period. In beef suckler cows, calved at least 50 days, and dairy heifers weighing over 280 kg, calving rate was similar in treated animals bred at 56 and 74 hours after treatment compared to calving rate in control animals bred at oestrus.  相似文献

Stanniocalcin-1 is a hormone that possesses both paracrine and endocrine functions and has recently been identified in mammals. While paracrine functions have been determined for several organs, the role of circulating stanniocalcin-1 in cattle is still unclear but, observations in mice and cows suggest that stanniocalcin-1 plays a role in both gestation and lactation. The changes in serum stanniocalcin-1 content in different physiological states have never been evaluated in ruminants. We measured stanniocalcin-1 levels in sera from cattle ranging in age from post-weaned calves to 17-month-old heifers and in sera from cows during lactation and pregnancy. Our results indicate that the blood concentration of stanniocalcin-1 is increased by pregnancy, but not by lactation. The highest levels of stanniocalcin-1 were found in young calves and during the non-lactating period preceding calving. This suggests that stanniocalcin-1 is important for gestation and the preparation of the mammary gland for lactation.  相似文献

In Trial 1, 26 heifers were allotted randomly to a control group or one of four groups to receive a single injection of 3,000 IU hCG on d 1, 4, 7 or 10 of an estrous cycle. Heifers next completed a nontreated cycle, and at their third estrus were reallotted to one of the five groups described previously. Estrous cycle length was extended in cycle 1 but was not altered during the nontreated cycle or in cycle 3. Administration of hCG on d 4 or 7 increased (P less than .05) mean serum progesterone (P4) over the first 16 d of the estrous cycle by .9 and .8 ng/ml, respectively. In Trial 2, 23 heifers were allotted randomly to one of two groups to receive a placebo or a single injection of 3,000 IU hCG on d 4 of an estrous cycle. Heifers were inseminated artificially at subsequent estrus. On d 4 postbreeding, treatments were repeated. Administration of hCG on d 4 of the prebreeding estrous cycle increased (P less than .05) mean P4 over the first 16 d by .9 ng/ml, whereas mean P4 over the first 16 d postbreeding was not affected by a second injection of hCG on d 4 postbreeding. Administration of hCG increased (P less than .05) first-service pregnancy rate (92 vs 55%). In conclusion, progesterone concentrations were enhanced by hCG given on d 4 or 7 of the estrous cycle, and pregnancy rate was increased when hCG was administered both on d 4 of the prebreeding cycle and d 4 postmating.  相似文献

Laboratory trials were conducted to study the effect of various concentrations of cloprostenol on the motility and morphological changes of the acrosomes of boar spermatozoa. As found, a cloprostenol concentration of 250 ng per ml of semen to 2500 ng per ml of diluted semen has no adverse effect on the motility of spermatozoa and on the morphological changes of their acrosomes. The concentration of 5000 ng of cloprostenol (in the Oestrophan Spofa product) per 1 ml of diluted semen negatively influences the motility of spermatozoa. An insemination dose of 100 ml of diluted sperm treated with 500 ng of cloprostenol was used for the artificial insemination of 152 sows; 166 sows of the same farm inseminated with untreated semen were used as controls. No gilts were included in the trial. Out of the 152 test sows, 113 conceived after the first insemination, i.e. 74.35%, and the average litter size was 10.04 piglets. In the control group, 125 sows delivered their litters, i.e. 75.30% of the total number, the average litter size being 9.96 piglets. Comparing the reproduction parameters of the experimental and control groups it can be said that the treatment of an insemination dose with 500 ng of cloprostenol immediately before insemination had no influence on the pregnancy rate and on the size of litters.  相似文献

Histological and morphometrical examinations were made to investigate the explants of yellow bodies in four gravid cows slaughtered the seventh day after PMSG superovulation and after artificial insemination. The medium of luteal tissue explants was added the test substance 15-glycal cloprostenol at doses of 2.5; 25 and 250 micrograms per ml of the medium. Parallelly cloprostenol in the Oestrophan inj. Spofa preparation was investigated at a dose of 25 micrograms per ml of the medium. The action of the medium lasted 24 hours at temperatures of 37 and 24 degrees C. Besides an overall histological examination the following morphometric indicators were studied: total counts of luteal cells per 1 mm2 area, average area of luteal cells and their nuclei and determination of the nucleocytoplasmic ratio of these cells, parenchymal area (area of luteal cells and stroma) of the luteal tissue per 1 mm2 area. The examinations have indicated that 15-glycal cloprostenol at a dose of 25 micrograms per ml of the medium has stimulating, luteotropic effects on the luteal tissue. The effects were manifested by luteal tissue hyperplasia and the largest area of luteal tissue parenchyma. On the other hand the effects of a tenfold higher dose (250 micrograms per ml of the medium) of the same substance were luteolytic similarly like those of parallelly tested cloprostenol.  相似文献
