Thirty-three cultivars and advanced lines originated from China, Mexico, and Australia were sown in four environments in Chinese spring wheat regions to investigate the association between gluten protein fractions determined by reversed-phase high-performance liquid chromatography (RP-HPLC), and dough properties and northern-style Chinese steamed bread (CSB) quality. The genotypes were divided into two groups of 10 and 23 entries with and without the 1B/1R translocation, respectively. 1B/1R translocation lines had significantly high amounts of ω  -gliadins, and low levels of glutenin and low molecular weight glutenin subunits (LMW-GS), but no significant difference in dough properties and CSB quality from non-translocation lines. The association between protein fractions and dough properties, and CSB quality largely depended upon the presence of 1B/1R translocation. Gliadin contributed more in quantity to flour protein content (FPC) than glutenin, while glutenin and its fractions contributed more to dough strength and CSB quality. Among non-translocation lines, moderate to high correlation coefficients between quantified glutenin and its fractions, and farinograph development time (DT, r=0.85r=0.85–0.92) and stability (ST, r=0.81r=0.81–0.93), extensograph maximum resistance (Rmax, r=0.90r=0.90–0.93), CSB stress relaxation (SR, r=0.55r=0.55–0.61) and CSB score (r=0.56r=0.56–0.62), were observed. Gliadin:glutenin ratios showed significant and negative associations with dough properties and CSB quality. Correlation coefficients between gliadin:glutenin, gliadin:HMW-GS, gliadin:LMW-GS ratios, and CSB score were −0.79, −0.73, and −0.79 among non-translocation lines, respectively. HMW-GS and LMW-GS, x-type HMW-GS and y-type HMW-GS contributed similarly to dough properties and CSB quality for non-translocation lines. Weak correlations between protein fractions and dough properties, and CSB quality were observed among translocation lines. This information should be useful for improvement of dough properties and CSB quality.  相似文献

Although significant correlations exist for glutenin macropolymer (GMP) quantity and rheological properties/bread making quality of dough, little information about these links is available. The relationship between GMP contents measured by UV absorption method/RP-HPLC and dough viscoelastic properties determined by TA-XT2i texturometer from three wheat varieties (Xiaoyan6, Yumai56 and Zhengnong8805) during mixing was investigated. GMP contents of doughs decrease significantly (P<0.05) during mixing. During the initial mixing stage, amounts of the HMW-GS and LMW-GS and GMP decrease significantly (P<0.05). Their contents begin to increase beyond peak dough development time (DDT). This indicates that during further mixing after peak DDT some glutenin subunits are incorporated into GMP by repolymerization. The HMW/LMW-GS ratio has a significant effect on load-deformation properties (area, resistance and extensibility) of dough. The varieties behaved differently in relation to the contribution of their HMW-LMW-GS ratio to the rheological properties.  相似文献

The extensigraph is particularly useful in characterizing dough viscoelastic properties; however, testing throughput for standard method is low due to the prerequisite for farinograph water absorption, long dough resting and milling to prepare large amounts of flour. Therefore, a rapid extensigraph method was developed that reduced sample size (165 g wheat) for milling and more than tripled throughput. Wheat is milled in Quadrumat Junior mill with a modified sieving system. The resulting flour (100 g) was mixed with a pin mixer at constant water absorption to allow the evaluation of wheat genotypes at the absorption level they are expected to perform. Dough was subsequently stretched by an extensigraph after 15 min of floor time and 30 min resting. Strong correlations for extensigram Rmax (r > 0.93), extensibility (r > 0.64) and area (r > 0.88) were found for the proposed method compared to the standard method. Mixing parameters (time and energy) obtained during dough preparation provided further information about dough strength and mixing requirement. By significantly reducing sample size requirement and increasing testing throughput, this rapid extensigraph method can be widely adopted in milling and baking industry and meets the need for a fast evaluation of dough strength in breeding trials.  相似文献

The objective of this research was to develop a methodology to describe the movement of the coffee berry borer Hypothenemus hampei (Coleoptera: Curculionidae) in the field through: (i) the evaluation of allele variation of a microsatellite marker on polymorphic Colombian H. hampei populations; (ii) the invention of a device for releasing H. hampei adults; (iii) the standardization of a release-recapture technique for H. hampei populations; (iv) the estimation of the flight distance of the insect; and (v) the calculation of a mathematical expression that describes the movement of H. hampei in space over time. The results indicated that: (i) the microsatellite molecular marker HHK.1.6 was exclusively present in a population from Guapotá-Santander, was dominant and allows the evaluation of H. hampei movement for several generations; (ii) a device that released 88.8% of H. hampei adults in 2 s was designed; (iii) this device was used as H. hampei populations containing HHK.1.6 marker release strategy, and coffee seeds as recapture strategy; (iv) it was estimated that H. hampei adults flew as far as 65 m, however, 90% were recovered in a radius of <40 m. Finally, (v) the mathematical expression that described the movement of H. hampei in space over time was Y^=αβXi, being Y^ the average number of borer beetles recaptured per tree, and x the distance in meters. This method will allow to determine the movement of H. hampei from different environmental and ecological scenarios.  相似文献

Three large deformation rheological tests, the Kieffer dough extensibility system, the D/R dough inflation system and the 2 g mixograph test, were carried out on doughs made from a large number of winter wheat lines and cultivars grown in Poland. These lines and cultivars represented a broad spread in baking performance in order to assess their suitability as predictors of baking volume. The parameters most closely associated with baking volume were strain hardening index, bubble failure strain, and mixograph bandwidth at 10 min. Simple correlations with baking volume indicate that bubble failure strain and strain hardening index give the highest correlations, whilst the use of best subsets regression, which selects the best combination of parameters, gave increased correlations with R2=0.865 for dough inflation parameters, R2=0.842 for Kieffer parameters and R2=0.760 for mixograph parameters.  相似文献

Frozen bread dough: Effects of freezing storage and dough improvers   总被引:1,自引:0,他引:1  
This review focuses on the effects of freezing storage on the microstructure and baking performance of frozen doughs, and provides an overview of the activities of dough improvers, including emulsifiers, hydrocolloids and other improvers used in frozen dough applications. The overall quality of bread baked from frozen dough deteriorates as the storage of the dough at sub-zero temperatures increases due to several factors which are discussed. Lipid-related emulsifiers such as diacetyl tartaric acid esters of mono and diglycerides and sucrose esters employed as anti-staling agents, dough modifiers, shortening sparing agents, and as improvers for the production of high-protein bread have also been employed in frozen doughs. Hydrocolloids are gaining importance in the baking industry as dough improvers due to their ability to induce structural changes in the main components of wheat flour systems during breadmaking steps and bread storage Their effects in frozen doughs is discussed. Other dough improvers, such as ascorbic acid, honey and green tea extract, are also reviewed in the context of frozen doughs.  相似文献

Organic-inorganic hybrid composites consisting of poly(vinylidene fluoride) (PVDF) and SiO2 were prepared through a sol-gel process and the crystallization behavior of PVDF in the presence of SiO2 networks was investigated by spectroscopic, thermal and x-ray diffraction measurements. The hybrid composites obtained were relatively transparent, and brittleness increased with increasing content of tetraethoxysilane (TEOS). It was regarded from FT-IR and DSC thermal analyses that at least a certain interaction existed between PVDF molecules and the SiO2 networks. X-ray diffraction measurements showed that all of the hybrid samples had a crystal structure of PVDFγ-phase. Fresh gel prepared from the sol-gel reaction showed a very weak x-ray diffraction peak near 2θ=21° due to PVDF crystallization, and intensity increased gradually with time after gelation. The crystallization behavior of PVDF was strongly affected by the amount of SiO2 networks. That is, SiO2 content directly influenced preference and disturbance for crystallization. In polymer-rich hybrids, SiO2 networks had a favorable effect on the extent of PVDF crystallization. In particular, the maximum percent crystallinity of PVDF occurred at the content of 3.7 wt% SiO2 and was higher than that of pure PVDF. However, beyond about 10 wt% SiO2, the crystallization of PVDF was strongly confined.  相似文献

Antioxidants are the chemical substances that reduce or prevent oxidation. The present study aimed to assess in vitro and ex vivo antioxidant activities of four acetonic extracts Tunisian halophytes (Suaeda fruticosa, Suaeda pruinosa, Suaeda mollis and Suaeda maritima). Various experimental models were used for characterization of antioxidant activities of shoot extracts. Eventually, the promising specie was subjected to phenolic identification using RP-HPLC. The analyzed shoot extracts exhibited that antioxidant activities varied considerably as function of species. The highest DPPH scavenging ability was found in S. mollis with the lowest IC50 value (2.5 μg/ml), followed by S. pruinosa, S. fruticosa and S. maritima. The same tendency was observed with ferric reducing power. Concerning β-carotene bleaching assays and total antioxidant activity, results showed that S. fruticosa exhibited the highest antioxidant ability against the inhibition of β-carotene bleaching, and a better total antioxidant capacity. Moreover antioxidant capacities using ORAC method and a cell based-assay showed that S. mollis, S. fruticosa, and S. pruinosa exhibit statistically similar antioxidant activity. The identification of phenolic compounds in S. mollis extract using RP-HPLC revealed that 2,5-dihydroxybenzoic acid and rutin hydrate were the major molecules. These results suggested that Suaeda species showed a variability of their antioxidant activities.  相似文献

为了解 Sec-1位点缺失对1BL/1RS易位系品质的影响,用 Sec-1位点缺失突变体与郑麦7698多次回交,系谱法辅助分子标记多代选择后得到 Sec-1位点缺失的1BL/1RS易位系(简称 Sec-1位点缺失易位系)和正常1BL/1RS易位系(简称正常易位系),并进行品质性状、抗病性及农艺性状鉴定。结果表明,蛋白质含量、湿面筋含量、干面筋含量和中峰值高度在 Sec-1位点缺失易位系和正常易位系之间差异不显著,而 Sec-1位点缺失易位系的面筋指数、面团形成时间、稳定时间、粉质质量指数、和面时间、中峰值宽度和8 min尾宽显著高于正常易位系。正常易位系的吸水率和衰落角显著大于 Sec-1位点缺失易位系。 Sec-1位点缺失易位系的条锈病抗性和白粉病抗性均低于正常易位系,但二者之间条锈病的病情指数差异不显著,而白粉病的病情指数差异达显著水平。正常易位系与 Sec-1位点缺失易位系间的千粒重、穗粒数、产量水平差异均不显著。综上所述, Sec-1位点缺失对小麦籽粒品质的正向调控作用显著,而对农艺性状的负面效应不显著, Sec-1位点缺失突变体是研究和改良小麦籽粒品质的优良遗传材料。  相似文献

Because CO2 is needed for plant photosynthesis, the increase in atmospheric CO2 concentration ([CO2]) has the potential to enhance the growth and yield of rice (Oryza sativa L.), but little is known regarding the impact of elevated [CO2] on grain quality of rice, especially under different N availability. In order to investigate the interactive effects of [CO2] and N supply on rice quality, we conducted a free-air CO2 enrichment (FACE) experiment at Wuxi, Jiangsu, China, in 2001–2003. A long-duration rice japonica with large panicle (cv. Wuxiangging 14) was grown at ambient or elevated (ca. 200 μmol mol−1 above ambient) [CO2] under three levels of N: low (LN, 15 g N m2), medium (MN, 25 g N m2) and high N (HN, 35 g N m2 (2002, 2003)). The MN level was similar to that recommended to local farmers. FACE significant increased rough (+12.8%), brown (+13.2%) and milled rice yield (+10.7%), while markedly reducing head rice yield (−13.3%); FACE caused serious deterioration of processing suitability (milled rice percentage −2.0%; head rice percentage −23.5%) and appearance quality (chalky grain percentage +16.9%; chalkiness degree +28.3%) drastically; the nutritive value of grains was also negatively influenced by FACE due to a reduction in protein (−6.0%) and Cu content (−20.0%) in milled rice. By contrast, FACE resulted in better eating/cooking quality (amylose content −3.8%; peak viscosity +4.5%, breakdown +2.9%, setback −27.5%). These changes in grain quality revealed that hardness of grain decreased with elevated [CO2] while cohesiveness and resilience increased when cooked. Overall, N supply had significant influence on rice yield with maximum value occurring at MN, whereas grain quality was less responsive to the N supply, showing trends of better appearance and eating/cooking quality for LN or MN-crops as compared with HN-crops. For most cases, no [CO2] × N interaction was detected for yield and quality parameters. These data suggested that the current recommended rates of N fertilization for rice production should not be modified under projected future [CO2] levels, at least for the similar conditions of this experiment.  相似文献

Dust has been widely recognised as an important source of nutrients in the marine environment and as a vector for transporting pathogenic microorganisms. Disturbingly, in the wake of a dust storm event along the eastern Australian coast line in 2009, the Continuous Plankton Recorder collected masses of fungal spores and mycelia (~150,000 spores/m3) forming a floating raft that covered a coastal area equivalent to 25 times the surface of England. Cultured A. sydowii strains exhibited varying metabolite profiles, but all produced sydonic acid, a chemotaxonomic marker for A. sydowii. The Australian marine fungal strains share major metabolites and display comparable metabolic diversity to Australian terrestrial strains and to strains pathogenic to Caribbean coral. Secondary colonisation of the rafts by other fungi, including strains of Cladosporium, Penicillium and other Aspergillus species with distinct secondary metabolite profiles, was also encountered. Our bioassays revealed that the dust-derived marine fungal extracts and known A. sydowii metabolites such as sydowic acid, sydowinol and sydowinin A adversely affect photophysiological performance (Fv/Fm) of the coral reef dinoflagellate endosymbiont Symbiodinium. Different Symbiodinium clades exhibited varying sensitivities, mimicking sensitivity to coral bleaching phenomena. The detection of such large amounts of A. sydowii following this dust storm event has potential implications for the health of coral environments such as the Great Barrier Reef.  相似文献

The bioactive materials in brown seaweeds hold great interest for developing new drugs and healthy foods. The oil content in brown seaweeds (Saccharina japonica and Sargassum horneri) was extracted by using environmentally friendly supercritical CO2 (SC-CO2) with ethanol as a co-solvent in a semi-batch flow extraction process and compared the results with a conventional extraction process using hexane, ethanol, and acetone mixed with methanol (1:1, v/v). The SC-CO2 method was used at a temperature of 45 °C and pressure of 250 bar. The flow rate of CO2 (27 g/min) was constant for the entire extraction period of 2 h. The obtained oil from the brown seaweeds was analyzed to determine their valuable compounds such as fatty acids, phenolic compounds, fucoxanthin and biological properties including antioxidant, antimicrobial, and antihypertension effects. The amounts of fucoxanthin extracted from the SC-CO2 oils of S. japonica and S. horneri were 0.41 ± 0.05 and 0.77 ± 0.07 mg/g, respectively. High antihypertensive activity was detected when using mixed acetone and methanol, whereas the phenolic content and antioxidant property were higher in the oil extracted by SC-CO2. The acetone–methanol mix extracts exhibited better antimicrobial activities than those obtained by other means. Thus, the SC-CO2 extraction process appears to be a good method for obtaining valuable compounds from both brown seaweeds, and showed stronger biological activity than that obtained by the conventional extraction process.  相似文献

Hybrid indica rice (Oryza sativa L.) cultivars play an important role in rice production system due to its heterosis, resistance to environmental stress, large panicle and high yield potential. However, no attention has been given to its yield responses to rising atmospheric [CO2] in conjunction with nitrogen (N) availability. Therefore we conducted a free air CO2 enrichment (FACE) experiment at Yangzhou, Jiangsu, China (119°42′0′′E, 32°35′5′′N), in 2004–2006. A three-line hybrid indica rice cv. Shanyou 63 was grown at ambient and elevated (ca. 570 μmol mol−1) [CO2] under two levels of supplemental N (12.5 g Nm−2 and 25 g Nm−2). Elevated [CO2] had no effect on phenology, but substantially enhanced grain yield (+34%). The magnitude of yield response to [CO2] was independent of N fertilization, but varied among different years. On average, elevated [CO2] increased the panicle number per square meter by 10%, due to an increase in maximum tiller number under enrich [CO2], while productive tiller ratio remained unaffected. Spikelet number per panicle also showed an average increase of 10% due to elevated [CO2], which was supported by increased plant height and stem dry weight per tiller. Meanwhile, elevated [CO2] caused a significant enhancement in both filled spikelet percentage (+5%) and individual grain weight (+4%). Compared with the two prior FACE studies on rice, hybrid indica rice cultivar appears to profit much more from elevated [CO2] than japonica rice cultivar (ca. +13%), not only due to its stronger sink generation, but also enhanced capacity to utilize the carbon sources in a high [CO2] environment. The above data has significant implication with respect to N strategies and cultivar selection under projected future [CO2] levels.  相似文献

In our ongoing search for bioactive substances from marine organisms, novel alkaloids have been isolated. Pinnatoxins and pinnamine, potent shellfish poisons, were purified from the Okinawan bivalve Pinna muricata. Pinnatoxins activate Ca2+ channels. Halichlorine was isolated from the marine sponge Halichondria okadai. This compound inhibits the induction of VCAM-1. Drugs that block VCAM-1 may be useful for treating coronary artery diseases, angina, and noncardiovascular inflammatory diseases. Pinnaic acids, which are cPLA2 inhibitors, were also obtained from P. muricata. Interestingly, the structures of pinnaic acids are closely related to that of halichlorine. Norzoanthamine hydrochloride, isolated from the colonial zoanthid Zoanthus sp., suppresses decreases in bone weight and strength in ovariectomized mice, and could be a good candidate for an osteoporotic drug. Ircinamine, purified from the marine sponge Ircinia sp., has a reactive thioester. Aburatubolactams, inhibitors of superoxide anion generation, were isolated from Streptomyces sp. This article covers the bioactive marine alkaloids that have been recently isolated by this research group.  相似文献

Final kernel number in the uppermost ear of temperate maize (Zea mays L.) hybrids is smaller than the potential represented by the number of florets differentiated in this ear, and than the number of silks exposed from it (i.e., kernel set <1). This trend increases when stressful conditions affect plant growth immediately before (GS1) or during (GS2) silking, but the magnitude of change has not been documented for heat stress effects and hybrids of tropical background. In this work we evaluated mentioned traits in field experiments (Exp1 and Exp2), including (i) two temperature regimes, control and heated during daytime hours (ca. 33-40 °C at ear level), (ii) two 15-d periods during GS1 and GS2, and (iii) three hybrids (Te: temperate; Tr: tropical; TeTr: Te × Tr). We also measured crop anthesis and silking dynamics, silk exposure of individual plants, and the anthesis-silking interval (ASI). Three sources of kernel loss were identified: decreased floret differentiation, pollination failure, and kernel abortion. Heating affected all surveyed traits, but negative effects on flowering dynamics were larger (i) for anthesis than for silking with the concomitant decrease in ASI, and (ii) for GS1 than for GS2. Heat also caused a decrease in the number of (i) florets only when performed during GS1 (−15.5% in Exp1 and −9.1% in Exp2), and only among Te and TeTr hybrids, (ii) exposed silks of all GS × Hybrid combinations, and (iii) harvestable kernels (mean of −51.8% in GS1 and −74.5% in GS2). Kernel abortion explained 95% of the variation in final kernel numbers (P < 0.001), and negative heat effects were larger on this loss (38.6%) than on other losses (≤11.3%). The tropical genetic background conferred an enhanced capacity for enduring most negative effects of heating.  相似文献

There has been much interest in artemisinin owing to its excellent activity against malaria, an infectious disease threatening the tropical world. However, the low artemisinin content (0.01-0.8%, DW) in Artemisia annua, which is the only commercial source of artemisinin, makes artemisinin expensive to produce and not yet available on a global scale. Here we show that foliar application of 100 mg l−1 chitosan improved artemisinin biosynthesis in A. annua. The content of dihydroartemisinic acid and artemisinin in chitosan-treated leaves increased by 72% and 53% compared with control values, respectively. Chitosan induced the expression of ADS and DBR2, which could explain the increase in level of artemisinic metabolites. After chitosan treatment, the amounts of hydrogen peroxide (H2O2) and superoxide anion (O2) in leaves of A. annua were 1.4 and 3.0 times higher than those of the control, respectively. Accumulation of reactive oxygen species (ROS) probably accelerated the conversion of dihydroartemisinic acid to artemisinin. Foliar application of 100 mg l−1 chitosan had no harmful effect on A. annua growth. The simple method described here could be an effective method to improve artemisinin production in A. annua field cultivation.  相似文献

Plant responses to water deficit need to be monitored for producing a profitable crop as water deficit is a major constraint on crop yield. The objective of this study was to evaluate physiological responses of cotton (Gossypium hirsutum) to various environmental conditions under limited water availability using commercially available varieties grown in South Texas. Soil moisture and variables of leaf gas exchange were measured to monitor water deficit for various varieties under different irrigation treatments. Lint yield and growth variables were also measured and correlations among growth parameters of interest were investigated. Significant differences were found in soil moisture, leaf net assimilation (An), stomatal conductance (g), transpiration rate (Tr), and instantaneous water use efficiency (WUEi) among irrigation treatments in 2006 while no significant differences were found in these parameters in 2007. Some leaf gas exchange parameters, e.g., Tr, and leaf temperature (TL) have strong correlations with An and g. An and WUE were increased by 30–35% and 30–40%, respectively, at 600 μmol (CO2) m−2 s−1 in comparison with 400 μmol (CO2) m−2 s−1. Lint yield was strongly correlated with g, Tr, WUE, and soil moisture at 60 cm depth. Relative An, Tr, and TL started to decrease from FTSW 0.3 at 60 cm and FTSW 0.2 at 40 cm. The results demonstrate that plant water status under limited irrigation management can be qualitatively monitored using the measures of soil moisture as well as leaf gas exchange, which in turn can be useful for describing yield reduction due to water deficit. We found that using normalized An, Tr, and TL is feasible to quantify plant water deficit.  相似文献
