Three hundred and twenty 30-week old White Leghorn cockerels were housed in individual cages and distributed in a completely randomized factorial design of 5 x 3, with five oil sources (sunflower, soybean, canola, linseed and fish/soybean) and three levels of antioxidant (30, 200 and 400 mg of vitamin E/kg). The aim of this study was to evaluate the effect of the ratio of omega6: omega3 fatty acids by the inclusion of different oil sources and of dietary supplementation with vitamin E on the reproductive performance of cockerels. The use of the fish/soybean combination determined the lowest total antioxidant status of the semen. However, the addition of vitamin E to the fish/soybean-oil-based diet resulted in a linear increase in semen volume, motility and sperm vigour in the 38th week and again in the 52nd week for motility and for sperm vigour and fertility rate in the periods from 50-53 and 41-53 weeks of age. The use of canola oil in the diet resulted in the highest fertility rate during 50-53 and 41-53 weeks of life. Animals receiving the soybean oil based diet showed the smallest fertility rate in the range from 50-53 weeks of age and concomitantly the highest level of cholesterol in the spermatozoa in the range from 47-51 weeks. An interaction between the vitamin E level and soybean oil was verified by the linear increase in motility and sperm vigour at 38 weeks of age. Later, the contrary was shown by a linear reduction in fertility in the periods from 44-46, 47-49 and 41-53 weeks of age. Cockerels that had been fed on the sunflower-oil-based diet showed the highest content of saturated fatty acids in the spermatozoa from 48-51 weeks. An interaction effect was observed between the vitamin E level and sunflower oil shown by a linear increase in the content of saturated fatty acids in the spermatozoa and a linear reduction in the C18: 1omega9, C18 :2 omega6 and PUFA (C18 : 2omega6 + C20 : 4omega6) contents in the spermatozoa at 48-51 weeks and in sperm volume at 52 weeks of age.  相似文献

1. One-day-old Taiwan Native Breeder female chicks were fed on maize/soybean growing diets without supplemental vitamin E from hatch to 17 weeks of age. After 17 weeks the birds (n = 300) were randomly assigned to 5 dietary treatments and fed on maize/soybean laying diets supplemented with 0, 40, 80, 120 and 160 mg/kg of vitamin E (dl-alpha-tocopherol acetate), respectively, until 46 weeks of age. The variates measured included: age at first egg, feed consumption (FC), feed efficiency (FE), egg production (EP), egg weight (EW), egg specific gravity (ESG), eggshell strength (ESS), fertility and hatchability. 2. The addition of 120mg/kg of vitamin E lowered the first EW (P<0.05); however, there was no significant difference in the age or body weight (BW) of pullets at first egg or mortality rate to 46 weeks of age among the treatments. FE and egg mass were improved (P<0.05) in pullets fed 80 mg/kg of supplemental vitamin E. A significant increase in EP was observed after peak EP in pullets given 80 mg/kg of supplemental vitamin E. However, this favourable effect decreased as supplemental vitamin E exceeded 80 mg/kg. 3. From 17 to 46 weeks of age, egg quality (ESG and ESS) decreased with age. However, there was no correlation between age and fertility or hatchability during the experimental period, suggesting that egg quality is more age-sensitive than reproductive performance for breeder pullets. 4. Compared with the control, fertility and hatchability of all eggs set for the treatment with 80 mg/kg supplemental vitamin E increased by 7.7 and 13.4%, respectively. There was no difference in the hatchability of fertile eggs. 5. These results suggest that using supplemental vitamin E during the laying period can improve the reproductive performance of breeder pullets. The addition of 80 mg/kg of vitamin E obtained the best performance in EP, egg mass, FE (feed/egg), hatchability and fertility.  相似文献

1. We examined the effects of supplemental dietary vitamin E (Vit E) on semen quality and antioxidative status in male domestic fowls exposed to oxidative stress induced by synthetic glucocorticoid, dexamethasone (DEX) injection. 2. Thirty-six Egyptian local cross males, 42 weeks old, were housed individually in cages in an open-sided building under 16 h light:8 h dark and were provided with commercial feed and water ad libitum. Birds were divided into 4 groups: DEX (4 mg/bird/d), Vit E (200 mg/kg diet), DEX + Vit E (4 mg/bird/d + 200 mg/kg diet, respectively) and control, n = 9. All treatments lasted for 7 continuous days. 3. Oxidative stress induced by injection of DEX (4 mg/bird/d) resulted in decreased sperm count and motility correlated with an increased percentage of dead sperms. Vit E (200 mg/kg diet) enhanced sperm count and viability when supplemented to stress-induced birds, compared to DEX treatment alone. 4. In seminal plasma, low calcium concentration, high lipid peroxidation and reduced activity of glutathione peroxidase were associated with the oxidative stress. Vit E reduced lipid peroxidation in the seminal plasma. 5. In conclusion, excessive supplemental dietary Vit E improved semen quality when cockerels were subjected to stress conditions. It increased both sperm count and motility, reduced the percentage of dead sperm and enhanced the antioxidative status of seminal plasma.  相似文献

Three experiments evaluated the effects of dietary Se and vitamin E on the ultrastructure of spermatozoa, ATP concentration of spermatozoa, and the effects of adding sodium selenite to semen extenders on subsequent sperm motility. The experiment was a 2 x 2 arrangement of treatments in a randomized complete block design. A total of 10 mature boars were fed from weaning to 18 mo of age diets fortified with two levels of supplemental Se (0 or .5 ppm) or vitamin E (0 or 220 IU/kg diet). The nonfortified diets contained .06 ppm Se and 4.4 IU vitamin E/kg. In Exp. 1, the spermatozoa from all boars were examined by electron microscopy. Vitamin E had no effect on structural abnormalities in the spermatozoa. When the low-Se diet was fed the acrosome or nuclei of the spermatozoa was unaffected, but the mitochondria in the tail midpiece were more oval with wider gaps between organelles. The plasma membrane connection to the tail midpiece was not tightly bound as when boars were fed Se. Immature spermatozoa with cytoplasmic droplets were more numerous when boars were fed the low-Se diet, but the occurrence of midpiece abnormalities occurred in boars fed diets with or without Se or vitamin E. Our results suggest that Se may enhance spermatozoa maturation in the epididymis and may reduce the number of sperm with cytoplasmic droplets. In Exp. 2, the concentration of ATP in the spermatozoa was evaluated in the semen of all treatment boars. When the low-Se diet was fed, ATP concentration was lower (P < .01), whereas vitamin E had no effect on ATP concentration. Experiment 3 investigated the effect of diluting boar semen with a semen extender with sodium selenite added at 0, .3, .6, or .9 ppm Se. Three ejaculates from each boar were used to evaluate these effects on sperm motility to 48 h after dilution. Sperm motility declined (P < .01) when Se was added to the extender, and this decline was exacerbated as the concentration of added Se increased (P < .01). The added Se was demonstrated to be tightly adhered to the spermatozoa. Overall, these results suggest that low Se-diets fed to boars resulted in abnormal spermatozoal mitochondria, a lower ATP concentration in the spermatozoa, and a loose apposition of the plasma membrane to the helical coil of the tail midpiece, but no effect from inadequate vitamin E was demonstrated. Adding sodium selenite to the semen extender reduced sperm cell motility.  相似文献

1. The effects of selenium and vitamin E supplementation on some immune parameters were investigated in broilers. 2. Broiler chicks were fed on maize-soybean diets with different concentrations of vitamin E (0-200 mg/kg) and selenium (0-0.2 mg/kg diet) either alone or in combinations from 1 to 42 d of age. 3. Chicks were immunised against Newcastle disease virus (NDV) vaccine at 21 d of age and haemagglutination inhibition (HI) titres were determined after 10 d. 4. Chicks receiving supplements of 200 mg vitamin E/kg and 0.2 mg selenium/kg produced significantly higher HI antibody titres. This was associated with an increased serum concentration of total immunoglobulins and circulatory immune complexes. 5. The chicks given 200 mg vitamin E/kg and 0.2 mg selenium/kg had significantly heavier spleen and bursa. 6. These results suggested that vitamin E and selenium have synergistic effects on immune responses.  相似文献

1. The effects of supplemental dietary vitamin E and organic selenium (Se), and their combination, on improving semen quality characteristics and antioxidative status were investigated in cockerels exposed to high ambient temperature.

2. A total of 36 Egyptian local cross males, 40 weeks old, were housed individually in cages in an open-sided building (average daily temperature ranged from 33 to 36°C and relative humidity from 60 to 70%). Birds were divided randomly into 4 experimental treatments (n?=?9 each): (1) control (basal diet without any supplementation with vitamin E or Se); (2) vitamin E (basal diet +200?mg α-tocopherol acetate/kg diet); (3) Se (basal diet +0·3?mg organic Se/kg diet); and (4) vitamin E?+?Se (basal diet +200?mg α-tocopherol acetate/kg diet +0·3?mg organic Se/kg diet).

3. Under heat stress conditions, inclusion of vitamin E and/or organic Se in the diets enhanced the semen quality traits, including the spermatozoa count and motility, and reduced the percentage of dead spermatozoa.

4. A combination of 200?mg/kg vitamin E with 0·3?mg/kg organic Se reduced the thiobarbituric acid reactive substance concentration in seminal plasma samples to about 28% of the controls; and also enhanced the seminal plasma glutathione peroxidase activity by two-fold compared with controls.

5. It was concluded that dietary vitamin E in combination with organic Se has a synergistic effect in minimising lipid peroxidation and improving the antioxidative status in seminal plasma of the domestic fowl, which probably translated into enhanced spermatozoa count, motility and reduced percentage of dead spermatozoa under heat stress conditions.  相似文献

1. The effects of different dietary concentrations of vitamin E (alpha-tocopherol acetate) were investigated on laying hens exposed to chronic heat stress at 32 degrees C from 26 to 30 weeks of age. 2. Diets containing 5 dietary concentrations of vitamin E (a control diet containing 10 mg alpha-tocopherol/kg or this diet supplemented to contain 125, 250, 375 and 500 mg alpha-tocopherol/kg) were fed to 335 birds. Half of the birds received the supplemented diets for only 4 weeks before the heat stress period (short supplementation duration, SSD) and were fed on the control diet for a further 12 weeks. The remaining birds were fed on the supplemented diets throughout the experiment, 4 weeks before, 4 weeks during and 8 weeks after the heat stress period (long supplementation duration, LSD). 3. Egg production was significantly higher during (80-6 vs 68.9%, P<0.02) and after (75.3 vs 62.7%, P<0.02) the period of stress in the LSD group fed on the diet containing 250 mg vitamin E/kg compared with the group fed on the control diet. LSD birds given 375 and 500 mg vitamin E/kg also had higher egg production than control birds during heat stress but the differences failed to reach significance (74.6 vs 68.9% and 77.1 vs 68.9% respectively). In the SSD groups, mean egg production of the birds given the diets supplemented with 125 mg vitamin E/kg or more was significantly different from the control group after heat stress (70.3 vs 62.7%, P<0.05). Egg weight and food intake were similar in all the groups. 4. Plasma and liver vitamin E concentrations were proportional to the vitamin E intake before the stress period, dropped during heat stress in the SSD groups but were maintained at concentrations closer to those observed before heat stress in the LSD groups. 5. It is concluded that a dietary supplement of 250 mg vitamin E/kg provided before, during and after heat stress is optimum for alleviating, at least in part, the adverse effects of chronic heat stress in laying hens.  相似文献

Semen characteristics in boars fed organic or inorganic sources of Se were assessed in 3 experiments. Crossbred boars were randomly assigned at weaning to 1 of 3 dietary treatments: I) basal diets with no supplemental Se (control), II) basal diets with 0.3 mg/kg of supplemental Se from an organic source (Sel-Plex, Alltech Inc., Nicholasville, KY), and III) basal diets supplemented with 0.3 mg/kg of supplemental Se from sodium selenite (Premium Selenium 270, North American Nutrition Co. Inc., Lewisburg, OH). For Exp. 1, semen was collected from boars (n = 10/dietary treatment) on 5 consecutive days at 15 mo of age. Effects of treatment × day were detected for the proportions of progressively motile (P = 0.02) and rapidly moving (P = 0.03) spermatozoa, and measures of sperm velocity, including path velocity of the smoothed cell path (P = 0.05) and average velocity measured in a straight line from the beginning to the end of the track (P = 0.05). Negative effects of day of semen collection on sperm motility were least pronounced in boars fed Sel-Plex. Experiment 2 was conducted when boars were 17 mo of age, and semen was collected (n = 10 boars/dietary treatment), diluted in commercially available extenders, and stored at 18°C for 9 d. Effects of treatment × day were detected for percentages of motile (P = 0.01) and static (P = 0.01) spermatozoa, amplitude of lateral head displacement (P = 0.02), frequency with which the sperm track crossed the sperm path (P = 0.04), straightness (P = 0.01), and average size of all sperm heads (P = 0.03). In general, sperm cells from boars fed Sel-Plex were better able to maintain motility during liquid storage compared with boars fed sodium selenite. For Exp. 3, semen was collected from boars (n = 6/dietary treatment) at 23 mo of age, and spermatozoa were evaluated at d 1 and 8 after semen collection using in vitro fertilization procedures. There was a tendency for an effect (P = 0.11) of dietary treatment on fertilization rate with Sel-Plex-fed boars having the greatest value (70.7%). The results of this study suggest that there are positive effects of dietary supplementation with Sel-Plex on boar semen characteristics and that organic Se supplementation may help ameliorate the negative effects of semen storage on characteristics of sperm motility.  相似文献

The effect of dietary vitamin E, selenium (Se) and their different combinations on body weight gain, food consumption, food conversion efficiency, leukocyte migration inhibition and antibody production was determined in broilers. 2. Chicks were fed on maize-soya bean based diets with concentrations of supplemental vitamin E varying from 0 to 300 IU/kg and selenium concentrations varying from 0 to 1 mg/kg either alone or in combination from 1 to 42 d of age. 3. The chicks were immunised for Newcastle Disease Virus (NDV) vaccine at 21 d. Per cent leukocyte migration inhibition (LMI) was studied on 42 d. Antibodies to NDV in serum were determined at 10 and 21 d post immunisation (PI). 4. Chicks receiving Se, 1 mg/kg and vitamin E 300 IU/kg had significantly higher cellular immune responses in terms of per cent LMI. 5. Maximum body weight gain and best efficiency of food utilisation were obtained in chicks fed diets containing 0.50 mg/kg Se and 300 IU/kg vitamin E. 6. Significantly higher antibody titres (HI and ELISA) at 10 d PI were attributed to 0.06 mg/kg and 150 IU/kg Se and vitamin E, respectively. 7. These data suggest that optimum growth and immune response may be achieved at supplemental level of Se of 0.06 mg/kg and vitamin E at 150 IU/kg. Two experiments were conducted to evaluate alpha-tocopherol accumulation in muscle of lambs finished on pasture or concentrates. The objective for Exp. 1 was to compare accumulation of alpha-tocopherol in the longissimus muscle of pasture-fed lambs to that of lambs fed three concentrations (15, 150, and 300 IU/kg of DM) of supplemental vitamin E (all rac alpha-tocopheryl acetate) in all-concentrate diets. The objective in Exp. 2 was to investigate the effect of duration of supplemental vitamin E feeding on alpha-tocopherol content and color change during display case storage of lamb muscle. Treatments evaluated in Exp. 2 were: 15 IU of supplemental vitamin E/kg DM fed to finish; 15 IU/kg followed by 300 IU/kg of DM during the last 21 d; and 15 IU/kg DM until 7 d prior to finish, then 300 IU/kg DM. In Exp. 1, alpha-tocopherol concentration of rotational grazed alfalfa and perennial ryegrass averaged 137 and 169 mg/kg of DM. Vitamin E treatments for lambs fed concentrate diets did not affect ADG (P > 0.15), but ADG was greater (P < 0.01) for concentrate-fed lambs than for grazing lambs. For the concentrate-fed lambs, alpha-tocopherol in longissimus muscle increased quadratically (P < 0.05) as dietary concentrations of vitamin E increased. Predicted maximum alpha-tocopherol concentration in muscle occurred at about 400 IU/kg of diet DM. Longissimus muscle from lambs grazing alfalfa or ryegrass had similar (P > 0.50) alpha-tocopherol concentrations, and those concentrations were similar to values obtained when the concentrate diet supplemented with 150 IU of vitamin E/kg was fed. In Exp. 2, no differences (P > 0.10) in ADG were observed. Concentrations of longissimus alpha-tocopherol were highest when 300 IU supplemental vitamin E was fed for 21 d prior to slaughter. During a 6-d display period, semimembranosus steaks from lambs fed 300 IU of supplemental vitamin E/kg for either 7 or 21 d had higher a* and b* color readings than steaks from lambs fed 15 IU/kg of supplemental vitamin E. Increased consumption of vitamin E either via pasture or supplementation results in higher alpha-tocopherol concentrations in meat.  相似文献

1. Laying performance, egg quality, fertility and hatchability, and fat deposition in liver and abdomen were recorded in broiler breeders (29 to 48 weeks of age) fed on diets containing perarl millet (Pennisetum typhoides) (PM), broken rice (BR) or yellow maize (YM) (600 g/kg diet). Constant ratios of metabolisable energy (ME) to other nutrients were maintained in all the diets. Food grade choline chloride (50%) was added to the diets at 3 concentrations (0, 760 and 1,520 mg/kg). Each diet was offered to 3 replicate groups of 15 birds (12 hens and 3 cockerels), maintained in deep litter pens, to provide 1.46 MJ ME/bird/d. 2. Neither the source of energy nor dietary choline content had any influence on hen-d egg production, fertility or hatchability. Food efficiency and egg weight were significantly reduced in BR-fed groups compared to those fed on the other energy sources. 3. The efficiency of energy and protein utilisation increased and liver fat content was decreased significantly by dietary choline supplementation. 4. Haugh unit score, egg shell weight, liver weight and intestinal weight were not influenced by either supplementary choline or the source of energy. However, the yolk colour index was significantly reduced in PM- or BR-fed groups compared to those fed on the maize-based diet. 5. Deposition of abdominal fat was significantly greater in BR-fed birds compared to those fed on the YM-based diet, while liver fat content was significantly greater in the birds fed on the PM-or BR-based diets than those based on YM. Although supplementation of the diet with choline had no influence on abdominal fat deposition, liver fat content was significantly reduced in birds given diet containing 760 mg supplemental choline/kg diet. 6. The present study indicates that PM or BR can be used as principal energy sources in place of YM in broiler breeder diets without affecting egg production, fertility or hatchability. Liver fat content can be reduced by adding choline at 760 mg/kg to diets based on different energy sources.  相似文献

1. The effect of high concentrations of vitamin E (alpha-tocopheryl acetate) fed during various age periods on the performance and the oxidative stability (thiobarbituric acid [TBA] values) of the drumstick meat of 7-week-old broiler chicks was determined. The basal diets (for the age periods 0 to 3, 3 to 6 and 6 to 7 weeks) contained 60 g soyabean oil and 24 mg vitamin E/kg. The following five treatments were evaluated: (1) the basal diets from 0 to 7 weeks of age (control); (2) vitamin E, 100 mg/kg diet from 0 to 7 weeks of age; (3) vitamin E, 150 mg/kg diet from 0 to 3 weeks of age; (4) vitamin E, 150 mg/kg diet from 0 to 3 weeks of age and 100 mg/kg diet from 6 to 7 weeks of age; (5) vitamin E, 100 mg/kg diet from 5 to 7 weeks of age. 2. Food intake, weight gain and food efficiency were not significantly (P greater than 0.05) affected by the vitamin E treatments. 3. Plasma alpha-tocopherol (AT) concentrations in treatments 2, 4 and 5 were similar, and markedly higher than those in treatments 1 and 3, while those of treatment 3 were significantly (P less than 0.001) higher than those of treatment 1. Plasma retinol concentrations were not significantly (P greater than 0.05) affected by the vitamin E treatments. 4. TBA values of the meat were very low and not significantly affected by the vitamin E treatments. However, after incubation the TBA values were highly significantly (P less than 0.01) negatively correlated with the amount of vitamin E consumed during the experiment. The stability of meat of birds fed the various combinations of vitamin E (treatments 3, 4 and 5) was significantly (P less than 0.05) higher than that of birds which did not receive additional vitamin E (treatment 1), but it was significantly (P less than 0.001) lower than that of birds which received vitamin E continuously (treatment 2). 5. It is concluded that a high concentration of vitamin E fed during 0 to 3 weeks of age may significantly improve AT status of the broiler chick up to 7 weeks of age.  相似文献

本试验旨在研究饲粮中维生素K3添加水平对五龙鹅生长性能、屠宰性能及养分表观利用率的影响。试验分为2个阶段。1~4周龄阶段,选用1日龄五龙鹅360只,随机分为6个组,每个组6个重复,每个重复10只鹅。Ⅰ组为对照组,饲喂基础饲粮(维生素K3含量1.23 mg/kg),Ⅱ~Ⅵ组在基础饲粮中分别添加1、2、4、8、16 mg/kg的维生素K3。5~16周龄阶段,选用28日龄五龙鹅324只,随机分为6个组,每个组6个重复,每个重复9只鹅。A组为对照组,饲喂基础饲粮(维生素K3含量1.18 mg/kg),B~F组在基础饲粮中分别添加2、4、8、16、32 mg/kg的维生素K3。试验期16周。结果表明:1)经回归分析得出,1~4周龄阶段五龙鹅饲粮中添加4.81 mg/kg维生素K3,平均日增重最大;5~16周龄阶段饲粮中添加11.59 mg/kg维生素K3,平均日增重最大。2)与对照组相比,1~4周龄阶段,饲粮中添加4 mg/kg维生素K3能显著或极显著提高鹅的全净膛率和腹脂率(P0.05或P0.01);5~16周龄阶段,饲粮中添加8 mg/kg维生素K3能显著或极显著提高屠宰率、半净膛率、全净膛率和腿肌率(P0.05或P0.01)。3)与对照组相比,5~16周龄阶段,饲粮中添加8 mg/kg维生素K3能极显著提高钙的表观利用率(P0.01),显著提高干物质、粗蛋白质、粗脂肪、中性洗涤纤维、酸性洗涤纤维和磷的表观利用率(P0.05)。由此可见,本试验条件下,1~4周龄和5~16周龄五龙鹅饲粮中维生素K3适宜添加水平分别为4.81和11.59 mg/kg。  相似文献

Our aim was to determine the effect of n-3 (2%, wt/wt, fish oil rich diet) and n-6 (2%, wt/wt, evening primrose oil rich diet) fatty acid dietary supplementation and their combination with two concentrations of vitamin E (40 vs 200 mg/kg) on semen variables and on fatty acid and vitamin E profiles of spermatozoa in broiler breeders at 32, 42 and 52 weeks of age. The inclusion of fish oil in the cockerel diets increased the docosahexaenoic acid proportion in the sperm phospholipid fraction, which was almost threefold higher compared to the other two groups irrespective of vitamin E supplementation. In contrast, an increase in the proportion of total n-6 polyunsaturates, mainly 22:4n-6, was observed in the evening primrose oil group compared to the control only when the dietary content of vitamin E was increased to 200 mg/kg. Sperm concentration was decreased in the fish and evening primrose oil groups if vitamin E was 40 mg/kg, but such an effect was prevented in the fish, not the evening primrose oil group, by increasing the vitamin E to 200 mg. The proportion of motile spermatozoa was improved by the increased supplementation of vitamin E in all oil treatments.  相似文献

The objective of this study was to evaluate the effects of vitamin C on growth and quality of semen from Oreochromis niloticus breeders. One hundred and sixty males were fed with different levels of vitamin C (0, 261, 599 and 942 mg/kg diet). The higher weight values were recorded for 599 (166 g) and 942 (175 g) mg of vitamin C/kg diet. Sperm motility, vigour and concentration were higher with 599 and 942 mg of vitamin C/kg diet. The semen volume, gonadosomatic index and plasma protein data from the last week showed a direct relationship with increasing levels of vitamin C. No changes were observed in the hepatosomatic index and blood glucose. The haematocrit and erythrocyte showed higher values estimated by equations derived at 850 and 638 mg vitamin C/kg diet, respectively. The leucocytes were inversely proportional to the increasing levels of vitamin C. After 100 days of feeding, animals fed the diet containing 942 mg vitamin C/kg diet had higher sperm motility, linearity, curvilinear velocity, straight line velocity and average path velocity (p < .05). Higher values of beat cross‐frequency were observed in broodfish fed diets containing 942 and 599 mg vitamin C/kg. The different vitamin C levels did not cause differences in straightness, lateral head displacement and sperm morphology. 1. It was previously found that cockerels vaccinated with live attenuated avian infectious bronchitis virus (AIBV) have decreased serum testosterone concentrations, epididymal stones and reduced fertility. The objectives of this study were twofold: to determine if reduced fertility following vaccination with live attenuated virus was the result of reduced sperm concentration or reduced sperm quality and to determine if vaccination with a killed strain of virus caused a similar reduction in sperm function in vivo. 2. Specific-pathogen-free Single Comb White Leghorn cockerels were divided into three treatment groups: no vaccination (NONVAC), vaccination with killed AIBV virus (KVAC) or vaccination with live attenuated AIBV virus (LVAC). Semen was collected daily from 17 to 27 weeks of age, and semen quality was assessed frequently by analysing sperm concentration, viability, motility, and ability to reach and interact with the ovum in vivo. Blood plasma was assayed for testosterone concentration. 3. Differences in sperm analysis among treatment groups were limited. Sperm viability was increased in NONVAC during week 20 which then decreased in week 22 when compared to vaccinated cockerels. Acrosome damage was increased in vaccinated cockerels in week 22, and decreased in weeks 25 and 27 when compared to controls, which correlate to the period of epididymal stone development. Plasma testosterone concentrations and sperm concentrations among treatment groups were different only at 16 and 19 weeks of age, respectively. There were no differences across treatment groups in sperm mobility through Accudenz or in numbers of sperm holes in perivitelline membranes of eggs following insemination with semen from 27-week-old cockerels. No differences were observed in viability or acrosome integrity between cockerels with and without epididymal stones within treatment groups. 4. In conclusion, pre-pubertal vaccination against AIBV and subsequent epididymal stone formation had a limited effect on sperm concentration, sperm quality and plasma testosterone concentrations. Vaccination with killed AIBV vaccine did not diminish effects on sperm function in vivo.  相似文献

1. Three experiments were conducted using a low temperature model to induce pulmonary hypertension (PH) and ascites in broiler chickens. Diets containing 25 g or 50 g flax oil/kg food and control diets with an equivalent amount of animal/vegetable (A/V) blend oil, with and without supplemental antioxidants (vitamin C and vitamin E) were used. The amount of PH was assessed by the ratio of right ventricle weight to total ventricle weight (RV/TV ratio). Birds were considered to suffer from pulmonary hypertension syndrome (PHS) if the RV/TV ratio was greater than 0.299. 2. In experiment 1, the test diets contained 50 g oil/kg food and were given during the grower period only. Birds fed on the flax oil diet tended to have a lower incidence of PHS, ascites and lower RV/TV ratios than birds fed on the control diet. However, when the flax oil diet was supplemented with antioxidants, the incidence of ascites, PHS, haematocrit and whole blood and plasma viscosity increased compared with birds fed on the flax oil diet without antioxidants. These effects were not seen in experiment 2, when the test diets containing 30 g oil/kg food (25 g flax oil plus 5 g A/V blend oil/kg food compared to 30 g A/V blend oil/kg food) were given during the grower period. However, in experiment 3, when the test diets containing 30 g oil/kg food were given from day 1 to week 8, birds fed on the control diet supplemented with antioxidants had a higher incidence of PHS than those fed on the control diet alone. 3. In all 3 experiments, there was no significant effect of dietary fat source or supplemental antioxidants on total food intake or food conversion. 4. We conclude that diets containing 50 g flax oil/kg food tend to reduce the incidence of PHS and ascites in broilers using a low temperature model but the results were not statistically significant. In some cases, supplementing diets with a combination of vitamin E and vitamin C increased the incidence of ascites and PHS.  相似文献

An experiment involving a total of 61 crossbred boars evaluated the effects of dietary Se and vitamin E on spermatogenic development at various stages of sexual development and the prostaglandin F2alpha (PGF2alpha) content in the seminal vesicle and prostate glands at 18 mo of age. The experiment from 5.4 to 9 mo of age was conducted as a 2 x 2 factorial in a randomized complete block design. Dietary Se at 0 or .5 ppm was the first factor and vitamin E at 0 or 220 IU/kg diet was the second. From 9 to 18 mo of age, a group of sexually active and inactive boars was a third factor. Treatment diets were fed from weaning (28 d of age) to the end of the experiment. Three boars per treatment group at 5.4 (105 kg BW), 6.2 (130 kg BW), and 9.0 (150 kg BW) mo of age were killed and the testes collected. From 9 to 18 mo of age, three boars from each dietary treatment group were used for semen collection, and another set of three to four boars from each treatment group remained sexually inactive. At 18 mo, both sets of boars were killed and their testes, prostates, and seminal vesicles were collected. The testis at each age was evaluated for sperm reserve numbers and germ and Sertoli cell populations. At 5.4 or 6.2 mo of age, testicular sperm reserves were not affected by dietary Se (P > .15), at 9.0 mo of age there was a trend for a higher (P < .10) number of sperm reserves, and by 18 mo of age the Se-fed boars had higher (P < .01) numbers of sperm reserves. Vitamin E had no effect (P > .15) on testicular sperm reserves at any age period. Boars fed dietary Se had a greater number of Sertoli cells (P < .01) and round spermatids (P < .01) at 6.2 mo of age, but by 18 mo of age the boars fed Se had more Sertoli cells (P < .05), more secondary spermatocytes (P < .01), and more round spermatids (P < .05). Vitamin E did not affect Sertoli or germ cell populations at the various ages. Boars at 18 mo of age had lower PGF2alpha concentrations in the prostate (P < .05) and seminal vesicles (P < .01) when vitamin E was fed, whereas Se had no effect. Sexually active boars had lower PGF2alpha concentrations in the seminal vesicles (P < .01) than sexually inactive boars, but there was no effect (P > .15) of sexual activity on the number of Sertoli cells, primary or secondary spermatocytes, or round spermatids. Our results indicate that Se has a role in establishing the number of boar spermatozoal reserves and Sertoli cells, whereas supplemental vitamin E did not affect these criteria.  相似文献

1. Two experiments were conducted to examine the effect of feeding higher supplemental vitamin E (VE) concentrations on male reproductive variables, fertility and hatchability of quails. 2. In experiment 1, sixty 5-week-old male quails, reared in individual cages, were fed male breeder diets supplemented with 0, 75, 150, 225 or 300 IU alpha-tocopherol acetate/kg. At 10 weeks, observations on cloacal gland size, foam production, testes weight and blood samples for testosterone estimation were taken. 3. In experiment 2, 50 male and 50 female cage-reared quails (5 weeks old) were fed male breeder and layer rations, respectively, supplemented with 0, 75, 150, 225 or 300 IU/kg. At 13 weeks, 9 different mating pairs (5 pairs/group) were formed. Group 1 contained one control male and one control female in each of 5 pens. In groups 2 to 5, control males were paired with females on supplements of 75, 150, 225 or 300 IU/kg. In groups 6 to 9, control females were paired with males on supplements of 75, 150, 225 and 300 IU/kg. During subsequent adaptation and egg collection periods, each of 10 d, control layer ration was fed to all groups. All the eggs laid during the 10-d egg collection period were incubated artificially to estimate fertility and hatchability. The trial was repeated at 15 weeks. 4. Adult male quails receiving moderate supplemental VE (75 and 150 IU/kg) had a higher cloacal gland index, quantity of foam secretion, testicular weight and plasma testosterone than quails fed on either VE-deficient or more highly supplemented diets (225 and 300 IU alpha-tocopherol acetate/kg). 5. It was concluded that a supplement of 75 IU VE/kg in maize/soybean diet could provide the best reproductive performance of male breeder quails. Cloacal gland index, quantity of foam secretion, testicular weight and plasma testosterone can serve as indirect indicators of testicular activity and fertilising ability in quails. Supplemental VE did not affect the fertility and hatchability of male and female Japanese quails.  相似文献
