In two grazing experiments carried out in 1982 and 1983 the prophylactic effect on gastro-intestinal helminthiasis of a single ivermectin treatment of calves 3 weeks after turnout was studied in animals which were turned out early on contaminated pasture and in calves which were turned out late on mown pasture. A single ivermectin treatment 3 weeks after early turnout did not prevent heavy helminth infections. However, such single treatment of calves 3 weeks after a late turnout on mown pasture appears to be a promising way of preventing heavy infections. Late turnout on mown pasture without anthelmintic treatment was not enough to prevent heavy infections. Ivermectin treatment was more effective against Ostertagia spp. than against Cooperia spp. There was no effect on the faecal egg output of Nematodirus helvetianus. Predominantly due to the dry summer, infections were lighter in 1983 than in 1982. These lower infections, and possibly also a delay in the onset of inhibition and differences in helminth strains, may explain the lower proportion of inhibited development in Ostertagia spp. in 1983 than in the comparable groups in 1982.  相似文献

An investigation of sources of helminth infection was carried out on three flocks: a traditional permanent pasture flock (A), one operating a 'clean' grazing system (B), and an East of Scotland College flock (C) which had operated a clean grazing system for nine years. Ewe and lamb worm egg output and pasture larval levels were recorded and tracer lambs were grazed during July and August on each farm. Considerable contamination was present on farm B fields compared with farms A and C, which resulted in higher worm burdens in late summer in farm B tracer lambs and lower weight gains. The main source of this infection was thought to be ewe periparturient egg output, as 21 per cent of ewes had positive worm egg counts over the lambing period. Differences in management practices between farms B and C, eg, earlier stocking of farm B fields, were also considered contributory factors.  相似文献

The efficacy of the Paratect Flex-Bolus for the control of parasitic gastroenteritis in calves was evaluated in a field experiment in the Swiss midland region. The bolus was administered to 9 first year grazing calves at 4 to 5 months of age before turnout on June 26 while 9 calves remained as untreated controls. Both groups were rotated between 8 paddocks that had been pregrazed by older cattle in spring. For a period of 12 weeks the faecal egg output of the treated calves was reduced significantly (p < 0.05) compared to the controls, whereas no significant differences were observed in the mean serum pepsinogen values of both groups. At the end of the experiment (November 14) the bolus-treated calves showed a 4 kg weight gain advantage over the controls which was not significant. The mild infection levels in both groups were probably due to the low pasture contamination with infective larvae throughout the season which most likely resulted from the late turnout of the calves. An outbreak of dictyocaulosis was observed in both groups in October and confirmed that the Paratect Flex-Bolus provides insufficient protection against this infection.  相似文献

Two groups of second grazing season cattle, which had been treated with an ivermectin-sustained-release bolus (ISRB) in their first grazing season, were monitored during consecutive years (1995 and 1996) on the same second grazing season pasture. In the preceding year (1994), this pasture had been grazed by yearlings that had not received chemoprophylaxis in their first grazing season. The objectives of the study were, firstly, to investigate whether the chemoprophylactic-treated yearlings were less resistant to gastrointestinal nematodes upon subsequent exposure, and hence excreted more strongyle eggs compared to the control yearlings; secondly, whether an increased susceptibility of the previously treated animals resulted in a yearly increase of the pasture infestation on the second grazing season pasture; and finally, whether this affected the second year weight gain of the animals. In 1996, the yearlings that had been chemoprophylactic-treated in 1995 excreted higher numbers of nematode eggs, compared to the previously ‘untreated' yearlings. In addition, the proportion of Cooperia larvae was markedly higher in the faecal cultures from the chemoprophylactic treated-animals, suggesting a negative effect of preventive treatment with an ISRB on the acquired resistance of the animals. However, there was no evidence that the slightly higher egg output in the previously treated yearlings had an effect on the larval contamination of the second grazing season pasture. A significant yearly decrease in the second season average daily weight gains was observed, but it could not be inferred from the results of the parasitological parameters that the differences in second year growth were caused by different levels of resilience between chemoprophylactic-treated and -untreated animals. As the study covered three consecutive second grazing seasons, an effect of differences between years (e.g. in weather conditions or grass growth) on the results cannot be excluded.  相似文献

The residual effect of treatment with ivermectin after experimental reinfection in calves was tested. Twenty-four calves were divided into 6 groups of 4 calves each. All calves received a primary infection of 50,000 larvae of both Ostertagia ostertagi and Cooperia oncophora and 1000 Dictyocaulus viviparus larvae. Calves of group 1 remained untreated, and all other calves were treated 21 days after primary infection (0.2 mg/kg injected subcutaneously). Calves of groups 1 and 2 were slaughtered 7 days later. Calves of groups 3-6 were reinfected with the same number of larvae 3 days, 1, 3 and 6 weeks after treatment respectively. Slaughter was 21 days after reinfection. Based on post-mortem worm counts the efficacy of ivermectin after primary infection was 99.7% for O. ostertagi, 95.1% for C. oncophora and 100% for D. viviparus. A residual effect was present for at least one week, but could not be observed 3 weeks after treatment.  相似文献

Two groups of three ponies were used to study the effect of three ivermectin or pyrantel treatments given at intervals of 5 weeks at the beginning of the grazing season. Although each pyrantel treatment resulted in a greater than 95% reduction in faecal egg counts during the first 3 weeks, high pasture larval counts were seen from the beginning of August onwards and substantial cyathostomine burdens were found at necropsy in December. The ivermectin treatments resulted in an even more pronounced reduction in faecal egg output, and the pasture larval counts and cyathostomine burdens at necropsy were considerably lower than in the pyrantel group. The proportion of inhibited early L3 of the cyathostomines was lower in the ivermectin than in the pyrantel group. Faecal egg output of the large strongyles was completely suppressed in the ivermectin group. Nevertheless, Strongylus vulgaris larvae were found in the arteries of all three ponies, possibly as a result of overwintering of infective larvae on pasture. In the pyrantel group, the egg output of Strongylus edentatus and, to a much lesser extent, Strongylus vulgaris, was not completely suppressed.  相似文献

The effects of anthelmintics treatments in controlling gastrointestinal nematodes in breeding ewes in a semi-arid area of Kenya were determined. The study carried out during two breeding seasons, between June 2000 and December 2001 where albendazole was administered to groups of ewes, 2 weeks before mating, 3 weeks to lambing and mid lactation indicated significantly lower nematode egg counts in treated than untreated groups of ewes. In the first breeding season, reduced rainfall resulted in pasture scarcity and weight loss in both groups of ewes through out the gestation period, but losses were higher for the untreated group. In the second season, both groups of ewes showed a steady increase in weight gain during the gestation period and post-partum, but weight gains were higher in the treated group. In lambs, weight gains at 6 weeks were higher for treated ewes than control groups, in both breeding seasons. The results of this trial indicate that anthelmintic treatments in breeding ewes in the study area are beneficial in reducing gastrointestinal nematode infections and improving performance of the ewes and their lambs. In addition to the treatments, breeding ewes should be given feed supplementation particularly during periods of pasture scarcity.  相似文献

This study investigated the effect of food or water deprivation on the pharmacokinetics of paracetamol in 30 Holstein-Friesian preruminant calves (10 controls, 10 food withheld and 10 water-deprived) aged 24-25 days. Control calves were given paracetamol at 24-25 days and again at 28-29 days of age. In the food withheld and water-deprived calves paracetamol studies were performed before and after 4 days of food or water deprivation. In the control group there were no significant differences in pharmacokinetic parameters for paracetamol in 24-25 and 28-29-day-old calves. Witholding food for 4 days was associated with an increase in the mean residence time (MRT) of paracetamol (P < 0.01). When food was withheld total body clearance (ClB) of paracetamol was significantly decreased (P < 0.05). The volume of distribution (Vss) was not significantly altered. Similarly, water deprivation was associated with a significant increase in MRT and significant decrease in ClB of paracetamol (P < 0.01). The Vss was not significantly altered. Food or water deprivation also influenced the formation of major metabolites (glucuronide and sulphate) of paracetamol. It is concluded that food or water deprivation may impair the elimination drugs that undergo metabolism by UDP-glucuronyltransferase and sulphotransferase in cattle.  相似文献

Paranatal transmission of Toxocara canis infection could be prevented in pups if an effective drug were administered to pregnant bitches. This drug also could eliminate the larvae in dogs that have been experimentally infected repeatedly to produce protective immunity. For these reasons, we assayed the effect of 2 doses of levamisole hydrochloride or ivermectin on T canis larvae. Mice (5 groups) were infected with 1,000 infective T canis larvae and then treated with 2 different dosages of levamisole hydrochloride (6 mg/kg or 12 mg/kg, given subcutaneously), 2 different dosages of ivermectin (0.2 mg/kg or 0.4 mg/kg, given IM) or 0.15M NaCl (given subcutaneously) once a day from days 15 to 28 of infection. On day 33 of infection, the parasites in liver, lungs, brain, and carcass were obtained and compared between groups. The smaller dosage of levamisole hydrochloride (6 mg/kg) significantly decreased only carcass parasitism to 17% of that in the controls, but did not affect significantly the total parasite load. The larger dosage of levamisole hydrochloride (12 mg/kg) decreased the infection in all organs, but particularly in carcass and brain; total parasitism was only 36% of that in the controls. The smaller dosage of ivermectin (0.2 mg/kg) significantly increased the number of larvae in the lungs to 550% of that in the controls, but it did not significantly affect the total parasite load. The larger dosage of ivermectin (0.4 mg/kg) significantly decreased only brain parasitism, but liver and total parasitism were decreased to 40% and 57%, respectively, compared with that in the controls.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

The aim of this study was to determine the effect of short-term starvation or water deprivation on the pharmacokinetics of caffeine in calves. The experiment was carried out on 30 Holstein-Friesian calves (10 calves in a control group, 10 calves in a 'starved' group and 10 calves in a 'water-deprived' group) aged 24-25 days. Control group calves were given caffeine at 24-25 and 28-29 days of age. In the experimental groups caffeine studies were performed before and after 4 days of starvation or water deprivation.In the control group no significant differences between the pharmacokinetic parameters of caffeine in 24-25 and 28-29 day-old calves were observed. Starvation for 4 days was associated with an increase in the mean residence time (MRT) of caffeine in each subject. The increase was statistically significant (P < 0.01). After starvation the total plasma clearance of caffeine decreased (about 20 per cent). The decrease was statistically significant (P < 0.05). The volume of caffeine distribution (V(ss)) was slightly but not significantly increased. Similarly, water deprivation was associated with significant increase in the mean residence time of caffeine in each subject. The total plasma clearance of caffeine decreased (about 30 per cent). This decrease was statistically significant (P < 0.01). The volume of caffeine distribution was slightly but not significantly decreased.Results obtained in this study indicate that short-term (for 4 days) starvation or water deprivation leads to a general inhibition of hepatic P450 enzymatic system and may impair the elimination of drugs that undergo metabolism by these enzymes.  相似文献

Two controlled tests were conducted in 1981 and 1982 in dairy calves on the University of Kentucky research farm to determine activity of the bolus formulation of levamisole given at the dose rate of 8 mg/kg against naturally occurring infections of Ostertagia ostertagi. Removal efficacies of mature O ostertagi were 98% in the 1981 test (3 treated and 3 nontreated calves) and 94% in the 1982 test (7 treated and 8 nontreated calves). Against immature Ostertagia spp, removal efficacies were 100% and 65% for the 1st and 2nd tests, respectively. The calves were grazed on the same pasture as dairy calves in previous controlled tests in 1979 and 1980 where activity of levamisole against mature O ostertagi (data recently published) was much less than in the present tests. It does not appear that the poor performance in the early tests can be attributed to the drug resistance phenomenon. Data on overwinter survival (119 days) of free-living stages of gastrointestinal parasites on pasture were derived from the nontreated calves in the 1982 controlled test. The calves, raised helminth-free, were placed on the pasture on Apr 5, 1982. Helminths recovered at necropsy of the calves, besides O ostertagi, included Trichostrongylus axei, Nematodirus helvetianus, Nematodirus spp, Cooperia oncophora, Trichuris spp, and Moniezia spp. The lung-worm, Dictyocaulus viviparus, previously found in cattle on the farm, was not found in these calves.  相似文献

Forty crossbred steers (Brahman x English) were categorized into two groups: 1) early weaned (EW; n = 20); and 2) normal weaned (NW; n = 20). Calves were 89 and 300 d of age at the time of EW and NW, respectively; SEM = 4.4. Early-weaned calves were kept on-site (University of Florida, Ona), provided supplement (1% of BW), and grazed on annual and perennial pastures until NW. At the time of normal weaning, all calves were loaded on a commercial livestock trailer and transported to the North Carolina State University Research Feedlot in Butner (approximately 1,200 km). Upon arrival, calves were stratified by BW and randomly allotted to four pens per weaning age treatment. Individual calf BW and blood samples were collected at the time of normal weaning, on arrival at the feedlot (d 1; 24 h following weaning), and on d 3, 7, 14, 21, and 28 of the receiving period. Individual BW was collected at the start and end of the growing and finishing periods, and feed intake by pen was measured daily. As an estimate of stress during the receiving period, plasma was collected and analyzed for the acute-phase proteins, haptoglobin and ceruloplasmin. Early-weaned calves were lighter (P = 0.03) at normal weaning than NW calves (221 vs. 269 kg; SEM = 10.6). By d 28, EW calves tended (P = 0.12) to be lighter than NW calves (242 vs. 282 kg, respectively). Gain:feed was improved for EW compared with NW calves during both the receiving (G:F = 0.157 vs. 0.081) and growing (0.159 vs. 0.136) periods. There tended (P < 0.10) to be weaning age x day interactions for each acute-phase protein. Ceruloplasmin concentrations increased in NW, but not EW calves, and peaked on d 7 (27.6 and 34.2 mg/100 mL for EW and NW calves, respectively; P < 0.05). Haptoglobin concentrations increased in both groups and were greatest (P < 0.05) in NW calves on d 3 (7.63 vs. 14.86 mg of haptoglobin/hemoglobin complexing/100 mL). No differences in ADG or G:F were detected during the finishing phase; however, overall G:F was improved (P = 0.03) for EW vs. NW calves (0.155 vs. 0.136). Carcass measures, including backfat thickness, USDA yield grade, marbling score, and LM area, did not differ between treatments. These data imply that EW calves, which are maintained onsite before shipping, may be more tolerant to the stressors associated with transportation and feed yard entry. Early weaned calves, managed within the system described in this study, may have improved G:F.  相似文献

SUMMARY A total of 498 dairy cows in 5 predominantly pasture-fed herds were allocated to pairs. One cow in each pair was treated with a single dose of ivermectin during the dry period. Treated and untreated cows were managed as a single group throughout the trial. Most cows calved between 45 and 115 days after treatment. When data from all herds were pooled, treated cows produced an extra 74 L of milk over the first 100 days of the subsequent lactation (95% confidence interval 20 to 128). Means were greater among treated groups relative to untreated groups in all 5 herds. However, when analysed individually, differences were statistically significant (P < 0.05) in 1 herd only. Over the complete lactation, mean milk volume for treated cows was 86 L greater than for untreated cows (95% confidence interval of difference -57 to 229; P = 0.24). Untreated cows produced 2473 L and 5883 L for the first 100 days of lactation and for the complete lactation, respectively. Milk production responses to treatment did not vary significantly with parity, body condition score, previous production index, calving date category or with plasma pepsinogen concentration or faecal egg count at the time of treatment. Faecal egg counts and plasma pepsinogen concentrations were low at the start of the study. The interval from calving to conception was 4.8 days less in treated cows (95% confidence interval 1.2 to 8.2) relative to untreated cows when data from all 5 herds were pooled. Differences within individual herds were not statistically significant.  相似文献

Orally administered ivermectinawas 100% effective against experimentally induced 11-day-old infections of Parascaris equorum (L3) in 2 trials. In 4 additional trials there was 98% efficacy of ivermectin treatment against induced 28-day-old infections of P. equorum (L4). In 2 trials with natural infections of ascarids, invermectin paste had 93% efficacy against immature P. equorum and 100% efficacy against the adult. We evaluated the comparative plasma and abomasal fluid disposition kinetics of albendazole (ABZ) and its metabolites in calves either grazing on pasture or fed a grain-based concentrate diet. Six male Holstein calves (weight 180 to 200 kg) were allowed to graze on lush pasture for three weeks before intraruminal administration of ABZ at 10 mg kg-1(pasture group). After a three-week wash-out period, the same animals were housed and fed on a grain-based concentrate diet for three weeks prior to receiving the same ABZ treatment (concentrate group). Jugular blood and abomasal fluid samples were collected over 120 hours post-treatment. Plasma and abomasal fluid samples were analysed by high performance liquid chromatography (HPLC). The digesta transit time was measured using cobalt (Co) as a fluid marker; abomasal fluid and faecal samples were collected and Co concentrations measured by atomic absorption spectrophotometry. Complementary studies of the in vitro dissolution of ABZ particles at different pH values were also conducted. The pH of abomasal fluid collected from animals kept under both feeding conditions was registered. Increased concentrations of ABZ sulphoxide (ABZSO) and sulphone (ABZSO2) in plasma, resulting in significantly higher Cmax and area under the curve (AUC) values for both metabolites, were obtained in calves fed on the concentrate diet compared to those grazing on pasture. Enhanced abomasal fluid levels of ABZ and ABZSO were observed in concentrate-fed calves. The mean retention time of the digestive fluid marker in the gastrointestinal (GI) tract was significantly longer in the animals fed the grain-based diet. The in vitro dissolution of ABZ at a pH value equivalent to that obtained in the abomasum of the concentrate-fed calves (1.75) was significantly greater than that obtained at the pH registered in pasture-fed animals (2.00). The characterisation of the kinetic/metabolic behaviours and the resultant efficacy of antiparasitic drugs in animals reared under different management conditions may be relevant in increasing parasite control in livestock.  相似文献

The concentration of CLA in adipose tissue can be increased in ruminants by feeding pasture and extruded soybeans. The objective of this study was to evaluate maternal supplementation of raw (RS) or extruded (ES) soybeans on the concentrations of CLA in milk fat of cows and s.c. adipose tissue of suckling calves. Thirty-two spring-calving cows (BW 624 +/- 76 kg; BCS 3.5 +/- 0.4; mean +/- SD) and calves (BW 127 +/- 15 kg) were separated into 2 groups. Cows were distributed to have 8 calves of each sex in both groups. When animals were turned out to pasture, dams received 2 kg/d of either RS or ES. Dietary treatments had no effect on average milk intake (P = 0.22) and pasture forage intake (P = 0.13) for calves over the course of the grazing season. As a result, no effect of treatments was observed on ADG (P = 0.26). At weaning, milk fat content of CLA reached 15.4 and 24.2 mg/g of total fatty acids for cows fed RS and ES, respectively (P = 0.02). The CLA concentrations in adipose tissue were 16.9 and 25.0 mg/g of total fatty acids for calves suckling dams fed RS and ES, respectively (P < 0.001). Overall, results demonstrated supplementing grazing cows with ES increased CLA content in milk and adipose tissue of suckling calves.  相似文献

A study was carried out to evaluate the effects of strategic early-season treatments with doramectin on first-season grazing calves exposed to trichostronglylid infection on a naturally contaminated pasture. Two groups of cross-bred Limousine/Red Danish calves were turned out in early May on two plots that were similar with respect to size and herbage infectivity. They grazed separately until housing in early October. One of these groups was given doramectin at turnout and 10 weeks later, while the other group served as untreated controls. The results showed that the treatments significantly reduced trichostrongylid loads throughout the season, as evidenced by significant reductions in both their Ostertagia ostertagi burdens and serum pepsinogen levels compared with the controls. Furthermore, the results of herbage larval counts and post-mortem worm counts in tracer animals demonstrated that the treatment had successfully suppressed herbage infectivity on the treated plot.  相似文献
