为阐明金属硫蛋白在铜缺乏奶牛肝脏中的代谢特征 ,选择铜缺乏区有明显临床症状的奶牛 6头 ,采集肝脏样品后用差速离心法分离肝脏金属硫蛋白 ,以Sephadex G75进行层析纯化 ,测定各部分洗脱液中金属元素铜、锌、铁、镁的含量。将 6头健康奶牛的肝脏样品作为对照。试验结果表明 ,发病奶牛肝细胞浆Sephadex G75柱层析洗脱液中铜、铁含量峰值显著高于健康奶牛 ,且在后面部分层析液中出现小峰 ;发病奶牛肝细胞浆金属硫蛋白 (MT)层析液锌、镁含量曲线峰值宽阔且高于对照组 ,第 2峰形宽大且有前移现象。结论 :铜缺乏能够改变奶牛肝脏中金属硫蛋白的代谢 相似文献

Copper and zinc are frequently added at high concentrations to pig diets as growth promoters. Livestock grazing pasture contaminated with pig slurry may, therefore, be at risk from excessive intake of these elements. High liver copper concentrations have been detected in cattle from the agricultural region of Galicia (NW Spain), especially where there is intensive pig farming. The aim of this study was to evaluate whether pig farming does affect accumulation of copper and zinc in cattle in Galicia. Hepatic copper and zinc concentrations in calves were elevated in areas with naturally high levels of these elements in the soil. The densities of young pigs (piglets and growing-finishing pigs), but not reproductive sows, also influenced copper accumulation in calves. Liver copper levels in calves were significantly and positively related to the density of young pigs in the region. In areas with the highest pig densities, more than 20% of the cattle analysed had hepatic copper concentrations that exceeded the potentially toxic concentration of 150 mg/kg fresh weight. There was no evidence that zinc accumulation in calves was affected by pig density. 相似文献

The increasing frequency of copper (Cu) toxicosis episodes in cattle in recent years, mainly associated with excess Cu supplementation, underscores the need to identify animals in the clinically silent phase of hepatic Cu accumulation. The aim of the study reported here was to evaluate the suitability of various blood parameters as potential early markers of hepatic Cu accumulation in cattle. Paired liver and blood samples from 70 calves aged 6 to 10 months were obtained at slaughter in a region in northwestern Spain where animals usually have hepatic Cu concentration higher than safe values. Neither serum Cu concentration nor ceruloplasmin (CP) concentration, the 2 parameters most commonly used for diagnosis of Cu deficiency, were significantly associated with hepatic Cu concentration. However, whole-blood Cu concentration had a slight but significant correlation with hepatic Cu concentration (r = 0.269, P = 0.026). The use of calculated blood parameters, such as the serum or whole-blood non-CP Cu fraction, or the CP-to-serum Cu ratio, increased the correlation with the hepatic Cu concentration (r = 0.393, P = 0.001, in the case of whole-blood, non-CP Cu concentration), but the strength of the association remained insufficient for accurate prediction of hepatic Cu values. Likewise, hepatic enzyme (aspartate transaminase and gamma-glutamyltransferase) activities were significantly or nearly correlated significantly with hepatic Cu concentration, but the strength of the association remained too low for accurate prediction. It is concluded that direct analysis of Cu concentration in liver biopsy specimens--preferably postmortem liver samples obtained at slaughter--is the best technique currently available for detecting chronic subclinical Cu accumulation in cattle at risk for supraoptimal Cu exposure. 相似文献

Clinical cases of both Zn and Cu deficiencies are reported in a cattle farm in Kordofan Region of the Sudan after drought. The animals showed general weakness, stunted growth, infertility, parakeratosis and achromotrichia. There was macrocytic hypochromic anaemia and low Cu and Zn concentrations in sera. The condition was more prevalent in zebu-Friesian crosses than the local breeds. Drought and marginal or low Cu and Zn content in pasture may be the predisposing factors. 相似文献

A one-year-old, neutered female Skye terrier presented with anorexia, vomiting, seizures and ascites. Portal venography demonstrated the presence of multiple acquired portosystemic shunts. Hepatic biopsy confirmed the presence of copper accumulation and fibrosis. Treatment included ursodeoxycholic acid therapy, colchicine and oral zinc. To the authors' knowledge, this is the first case report detailing successful management of Skye terrier hepatopathy. 相似文献

The efficacy of a molybdate formulation and a zinc oxide bolus as prophylactic agents for enzootic icterus was evaluated in sheep. Before copper loading, liver biopsies were performed on 12 male, 6-month-old, Mutton Merino sheep to determine hepatic copper (Cu) and zinc (Zn) concentrations. The animals were restrictively randomised according to liver copper concentrations to 3 treatment groups (n = 4) to achieve similar mean liver copper concentrations per group. All sheep received 4 ml/kg of a 0.5 % aqueous solution of CuSO4 5H2O intraruminally 7 days per week for 10 weeks. On Day 0 the sheep in the Mo-group were injected subcutaneously with 42 mg molybdenum (Mo) contained in a commercial molybdate formulation. The animals in the Zn-group each received a zinc oxide bolus, containing 43 g zinc oxide, via a rumen cannula. Treatment was repeated on Day 42. Four animals served as untreated controls. Urinary copper excretion, plasma copper concentration, haematocrit and glutamate dehydrogenase (GLDH) activity were determined throughout the trial. The animals were sacrificed after 10 weeks and liver samples were submitted for histopathological examination. Liver and kidney copper and zinc concentrations were determined. Neither the molybdate treatment nor the zinc oxide boluses prevented hepatic copper accumulation. The urinary copper excretion, plasma copper concentration, haematocrit and GLDH activity were not significantly different (P > 0.05) from the controls. 相似文献

The intracellular distribution of copper (Cu) and zinc (Zn) in the livers of normal and moderately Cu-exposed cattle was investigated with the aim of improving understanding of the pathophysiology of Cu accumulation in cattle. At total liver Cu concentrations within the generally accepted normal range (25-100 mg/kg fresh weight) the large-granule fraction was the main subcellular compartment for Cu accumulation, followed by the cytosol and the nucleus, whereas in the microsomal fraction Cu accumulation was very low. With increasing Cu exposure, the capacity of the large-granule fraction to accumulate Cu decreased, proceeding towards a plateau (estimated at about 80 mg/kg at a projected total liver Cu concentrations of 450 mg/kg), accompanied by progressively greater Cu accumulation in the nucleus and cytosol. Total liver Cu concentration had little influence on subcellular Zn distribution, with hepatic Cu concentrations being only moderately correlated with Zn concentration in the large-granule fraction. There was a strong association between the proportion of total Cu bound to metallothionein (MT) and the proportion of total Zn in the large-granule fraction, suggesting that Zn displaced from MT is taken up by the lysosomes for excretion. This pattern of Cu accumulation, as in sheep, may be due to the limited capacity for metallothionein binding of Cu and excretion in bile. 相似文献

CASE HISTORY: A 400-cow dairy unit in the Waikato suffered a severe outbreak of facial eczema (FE) despite consistent zinc supplementation and significantly elevated serum zinc concentrations. CLINICAL FINDINGS: FE prevention had begun in mid-January 2006, via zinc sulphate supplementation in the water. Photosensitisation was reported on 06 April, contemporaneous with a marked increase in the number of Pithomyces chartarum spores in pasture samples. Within 10 days of this first clinical case, 100 affected cows had been dried off, eight of which subsequently died or were culled. Blood samples were collected from ten affected cows; all had serum zinc concentrations >17 micromol/L, and eight had gamma-glutamyl transferase (GGT) activities >200 U/ml, indicating that the photosensitisation was secondary to liver damage, i.e. probably FE. Further investigation identified that this herd had been receiving excess copper supplementation; 4/6 culled cows had liver copper concentrations within or above the marginal range for copper toxicity. DIAGNOSIS: Severe FE despite zinc supplementation that increased serum zinc concentrations above recommended levels. CLINICAL RELEVANCE: As zinc supplementation significantly reduces apparent copper status, copper supplementation is often used to counteract this. Previous excess copper intake may reduce the efficacy of zinc in preventing FE, thus copper intake should be assessed prior to the start of zinc supplementation. 相似文献

The aim of the study was to determine the effect of weaning and the effect of increasing dietary zinc concentrations on the zinc and copper status of weaned piglets (study 1) and to study the effect of high concentrations of dietary zinc and/or copper on zinc and copper status of weaned piglets (study 2). Study 1 included 54 piglets (six litters of nine piglets). One piglet from every litter was killed 1 day before weaning. The remaining 48 piglets were allocated at weaning (28 days) to four dietary zinc treatments (100, 250, 1000 or 2500 ppm) and subsequently killed 1-2, 5-6 or 14-15 days after weaning. Study 2 included 48 piglets (six litters of eight piglets) allocated to four dietary treatments, consisting of low or high dietary zinc (100 or 2500 ppm) in combination with low or high dietary copper (20 or 175 ppm). All piglets in study 2 were killed 5-7 days after weaning. In both studies, the trace mineral status was assessed by zinc and copper concentrations and alkaline phosphatase (AP) activity in plasma and mucosal tissue. In study 2, lymphocyte metallothionein (MT) mRNA and intestinal mucosa MT mRNA concentrations were included as zinc status markers. The results showed that the zinc status, measured as zinc in plasma and mucosa, was not affected by weaning of the piglets. Plasma copper concentrations decreased during the first 2 weeks after weaning. High dietary copper concentrations did not affect the concentration of copper in plasma, but increased the concentration of copper in mucosa and the concentration of zinc in plasma. The dietary zinc treatments increased the zinc concentration in plasma as well as the zinc and MT mRNA concentration in mucosa. Lymphocyte MT mRNA concentrations did not reflect the differences in dietary zinc supplementation. 相似文献

1. The effect of intravenously injected zinc (Zn) on tissue Zn concentrations and pancreas metallothionein (MT) gene expression in broilers was investigated to detect differences in the tissue utilisation of Zn from different Zn sources. 2. A total of 432 male chickens were randomly allotted on d 22 post-hatch to one of nine treatments in a completely randomised design. Chickens were injected with either a 0.9% (w/v) NaCl solution (control) or a saline solution supplemented with Zn sulphate or one of three organic Zn chelates with weak (Zn-AA W), moderate (Zn-Pro M) or strong (Zn-Pro S) chelation strengths at two injected Zn dosages calculated according to two Zn absorbability levels (6 and 12%). 3. Bone and pancreas Zn concentrations, pancreas MT mRNA levels and MT concentrations increased on d 6 and 12 after Zn injections as the injected Zn dosages increased. Chickens injected with the Zn-Pro S had lower bone Zn concentration than those injected with the Zn-Pro M or Zn-AA W on d 6 after injections. However, no differences among Zn sources were observed in bone Zn concentration on d 12 after injections, pancreas Zn concentrations, pancreas MT mRNA levels and MT concentrations on both d 6 and d 12 after injections. 4. It was concluded that the injected Zn-Pro S was the least favourable for bone Zn utilisation of broilers. The pancreas Zn concentration and pancreas MT gene expressions might not be sensitive enough to detect differences in the tissue utilisation of injected Zn in broilers between organic and inorganic Zn sources or among organic Zn sources. 相似文献

The effect of copper on intact cattle erythrocytes was investigated in vitro. When treated with copper, a decrease in the GSH content, accumulation of copper in the cell, loss of potassium and gain of sodium, cross-linking of membrane proteins, and echinocytic transformation were observed. All of these phenomena seem to be caused by a potent oxidant action of copper. These cytotoxic effects of copper were markedly inhibited by the addition of bovine serum albumin in the incubation medium. These results may help to understand the mechanism of hemolysis associated with copper poisoning in vivo. 相似文献

In sheep the effects of Zn‐supplementation on the Cu‐accumulation in the liver were studied. At a relatively high Cu‐level in the feed, dietary Zn reduced the hepatic Cu‐level in the Texel breed and in the crossbreed Texel x Friesian milksheep, but not in the Friesian milksheep breed. On account of this result it can be concluded that in sheep the mutual interaction between Cu and Zn may be dependent on genetic factors. 相似文献

Objective – To review zinc physiology and pathophysiology and the importance of zinc toxicity and deficiency in veterinary patients. Data Sources – A review of human and veterinary medical literature. Human Data Synthesis – There is a significant amount of original research in humans and animals on the role of zinc in multiple organ systems. There is also significant data available on human patients with zinc abnormalities. Veterinary Data Synthesis – Zinc deficiency has been studied in dogs with genetic disease and dietary deficiency leading to dermatological disease and immune deficiency. Zinc toxicity has been described after ingestion of metallic foreign bodies containing zinc. Conclusions – Historically, the role of zinc in health and disease has been studied through patients with toxicity or severe deficiency with obvious clinical signs. As the ubiquitous contribution of zinc to structure and function in biological systems was discovered, clinically significant but subtle deficiency states have been revealed. In human medicine, mild zinc deficiencies are currently thought to cause chronic metabolic derangement leading to or exacerbating immune deficiency, gastrointestinal problems, endocrine disorders, neurologic dysfunction, cancer, accelerated aging, degenerative disease, and more. Determining the causal relationships between mild zinc deficiency and concurrent disease is complicated by the lack of sensitive or specific tests for zinc deficiency. The prevalence of zinc deficiency and its contribution to disease in veterinary patients is not well known. Continued research is warranted to develop more sensitive and specific tests to assess zinc status, to determine which patients are at risk for deficiency, and to optimize supplementation in health and disease. 相似文献
