ABSTRACT Controlled environment experiments were conducted to study the effects of inoculum density, temperature, and their interaction on germination of Puccinia allii urediniospores and infection of leek leaves. Percent germination of P. allii urediniospores and percent branching of germ tubes increased with 3 density of urediniospores and approached a plateau for densities above approximately 20 spores cm(-2) of leaf area. Percent germination was highest at 12 to 21 degrees C, a wide-range temperature optimum. Branching occurred at temperatures ranging from 5 to 25 degrees C, but there were few germ tubes branching at 25 degrees C. P. allii successfully infected leek leaves at temperatures ranging from 7 to 22 degrees C. The number of pustules produced increased with urediniospore density on leek leaves. At low spore densities, pustule production was little affected by temperature; at higher spore densities, pustule production was greatest between 9 to 11 degrees C, and numbers of pustules decreased greatly with temperature increasing above this optimum. Latent period was affected by temperature, with latent period being shortet between 19 and 22 degrees C, and latent period increasing when temperature decreased. Latent periods became approximately 1.8 days shorter for every 10-fold increase in spore density. The rate of pustule production increased with increasing spore density on leaves and was greatest between 11 to 14 degrees C. Computer simulation of leek rust progress based on the found relationships suggested that at optimal temperatures the development of leek rust epidemics may be little affected by initial spore density and density caused by each pustule, but that at sub- and supra-optimal temperatures the development is greatly affected by these variables.  相似文献

Penicillium expansum is one of the main postharvest pathogens of apples in Israel. Heating apple fruit inoculated with P. expansum for 96 h at 38°C completely inhibited decay development. Fruit held for 24 h at 42°C or 12 h at 46°C had significantly reduced decay after an additional 14 days incubation at 20°C, compared with unheated inoculated control fruit. Mycelial growth and percentage spore germination in vitro were inversely proportional to length of time of exposure to various temperatures. The ET50 for spore germination was 42, 34 and 20 h at 38, 42 and 46°C, respectively, while the ET50 for mycelial growth was 48, 44 and 36 h at those temperatures. When Penicillium spores were incubated on crude extract prepared from the peel of apple fruits held 4 days at 38°C, germ tube elongation was significantly reduced, while the walls of the tubes were thicker, compared with germ tubes from spores incubated on crude extract prepared from peel of non-heated fruit. The evidence presented here supports the hypothesis that the effect of heating on the decay of apples caused by P. expansum is not only the result of direct inhibition of fungal germination and growth by high temperature, but is also partly due to the formation of an inhibitory substance in the heated peel.  相似文献

Celery (Apium graveolens L.) plants were inoculated with 3-µl droplets of a pycnidiospore suspension ofSeptoria apiicola Speg., and then covered with polyethylene bags and kept at an ambient temperature of 23±2°C. Under these conditions disease expression was related to incubation time up to 8 days, and to spore concentrations up to 500/3 µl. An inoculum concentration as low as four viable spores per 3 µl caused 26% as many lesions as did 500 spores/3 µl. In plants not covered, or covered with polyethylene bags for only 1 day, no disease symptoms appeared regardless of the spore concentration applied to the plants. Disease symptoms on inoculated leaves appeared on 10-50-day-old but not on 50-90-day-old leaves. The pathogenicity of 16 isolates collected from two different regions of Israel, and of eight replicates of an isolate used in previous pathogenicity tests, was similar.  相似文献

Effects of environmental factors on the germination and seedling emergence of glyphosate‐resistant (R) and ‐susceptible (S) biotypes of Eleusine indica (L.) Gaertn. were examined under laboratory and greenhouse conditions. The R biotype exhibited a higher germination percentage compared with the S biotype at constant temperatures of 20 and 35°C under dark conditions, and alternating temperatures of 30/25°C, and 35/25°C during a 12 h photo period. For both biotypes, germination was optimal at alternating temperatures of 30/20°C and 35/20°C. However, there was no significant difference (P > 0.05) in the germination between the R and S biotypes at these temperature regimes. The germination of both biotypes was inhibited by osmotic stress imposed by a water potential of ?0.80 MPa. When the moisture stress was released and the seeds were subsequently transferred to distilled water, the germination was enhanced to approximately 90% and 16% for the R and S biotype seeds, respectively. Higher emergence rates were obtained in shallow seed depths (0 or 2 cm) compared to deep depths. Emergence percentage of the R biotype was higher than that of the S biotype at 0 cm and 2 cm depths. The maximum emergence percentage of the R biotype was higher than that of S biotype when seeds were sown on the surface of either loamy or clay loam soil taken from three different sites.  相似文献

Uromyces transversalis is an autoecious microcyclic rust mainly infecting Gladiolus spp. The pathogen is considered of plant quarantine importance in Europe and the USA. In 2006, the pathogen was found for the first time in the USA in several commercial nurseries in Florida and California. The US Department of Agriculture (USDA) initiated an eradication programme that recommended the immediate removal and destruction of infected plants followed by a host-free period, use of a fungicide treatment schedule, and equipment decontamination. In support of this plan, a study was conducted to determine how long urediniospores of U. transversalis would continue to germinate at temperatures of 2.8, 15.0, 18.8 and 25.0°C under controlled relative humidities (RH) of 11, 23, 43, 75, 93 and 100%. Choice of temperature and humidity parameters were mostly based on historical multi-year climate data from areas where the disease was detected in California and Florida. Analysis of variance (ANOVA) indicated no significant effect of RH on urediniospore germination but a highly significant effect of temperature. No germinating urediniospores were detected after 79 days for any treatment, but the 15°C treatment was more likely to be the result of germination independent of any low or high temperature-induced spore quiescence. Thus, lack of germination after 79 days was probably a good indicator of the lack of viable spores after this time for the 15°C treatment.  相似文献

研究了10种杀真菌剂在田间使用浓度的中间浓度及其10倍稀释液下,对昆虫病原真菌块耳霉孢子萌发的影响。结果表明各杀菌剂对块耳霉孢子萌发有不同程度的抑制,浓度愈高,抑制作用愈大。多菌灵、普力克对孢子的抑制作用最小,与块耳霉表现出良好的相容性,是与块耳霉实施菌药合用的候选药剂。施保功、大生、达科宁3种杀菌剂对块耳霉孢子的杀伤力最大,在两种浓度下孢子萌发率均为0。施佳乐、速克灵和敌力脱在低浓度下,与块耳霉的相容性良好,处理后24~48 h与对照无显著性差异;但在中间浓度水平下,处理后48 h孢子萌发率分别为12.45%,0,0。其余药剂随处理时间和浓度不同,对块耳霉孢子萌发有不同程度的抑制。  相似文献

To elucidate the mechanism of clubroot suppression under neutral soil pH, a highly reproducible germination assay system under soil culture conditions was designed based on the hypothesis that germinated spores of Plasmodiophora brassicae could be identified by the absence of a nucleus (i.e. having released a zoospore to infect a root hair of the host plant). Brassica rapa var. perviridis seedlings were inoculated with a spore suspension of P. brassicae at a rate of 2·0 × 106 spores g−1 soil and grown in a growth chamber for 7 days. The spores were recovered from rhizosphere and non-rhizosphere soils and stained with both Fluorescent Brightener 28 (cell-wall-specific) and SYTO 82 orange fluorescent nucleic-acid stain (nucleus-specific stain). Total numbers of spores were counted under UV-excitation, and spores with a nucleus that fluoresced orange under G-excitation were counted. The significant increase in the percentage of spores without a nucleus (germinated spores) in the rhizosphere after 7 days' cultivation and the correlation with root-hair infections validated the assay system. Applications of calcium-rich compost or calcium carbonate to neutralize the soil significantly reduced the percentage of germinated spores in the rhizosphere, as well as the number of root-hair infections. The present study provides direct evidence that the inhibition of spore germination is the primary cause of disease suppression under neutral soil pH.  相似文献

The effect of environmental factors on the development of each stage ofPeronospora destructor (Berk.) Caspary on onions is reviewed. For sporulation to take place, a period of light must precede the period of darkness and high humidity in which spores are formed. Spores are discharged when the relative humidity (RH) is increasing or decreasing, and over a wide range of temperatures. Their discharge is triggered by exposure to red-infrared radiation and by vibration of the leaf. Dissemination of spores follows a daily periodic cycle and spores can be blown by wind over long distances. Duration of spore survival depends on temperature, RH and, especially, the absence of strong radiation. The rate of spore germination is highest at 10°C and declines with the rise in temperature. Germ tubes develop in liquid water, and a continuous period of wetness is required for infection to be completed. Systemic infection is common in cooler climates, where necks of onion bulbs are slow to dry. The principal sources of downy mildew infection by wind-borne spores are systemically infected propagation material, onion volunteer plants, and neighboring older crops.  相似文献

The monocyclic phase of Stemphylium vesicarium is part of its life cycle and a possible factor for forecasting and the integrated control of purple spot on asparagus. The purpose of the study was to model the flight, germination and germ tube growth of ascospores as basis for the development of a forecasting system. During 2014–2016, the flight period was determined by spore traps. The ascospores flew between March and early July, but most were released in early May. The cumulative percentage of trapped ascospores depending on the daily summed temperature (base 5 °C) on rainy days starting from February 1st was described best by a Chapman Richards function. The germination and germ tube length of ascospores depending on leaf wetness duration and temperature were investigated in laboratory trials. Ascospores germinated rapidly in a wide temperature range. The fitted Chapman Richards function with a temperature-dependent capacity and rate described germination adequately with a calculated optimal temperature of 31.0 °C. The germ tube length was modelled by a combined generalised beta-linear function and it was optimal at 30.4 °C with a narrow temperature range of 25–35 °C for values close to the optimum length. Therefore, the infection process is restricted more severely by the germ tube length than by germination. The ascospore flight is often finished before the end of the harvest, so fungicide treatments during the monocyclic phase might be ineffective in many production sites in Germany. The situation could be different for shorter harvest periods and in non-harvested young plants.  相似文献

Darkness suppresses ascospore release in Venturia inaequalis, but the impact of light levels during the extended twilight and dusk that typify Nordic spring conditions is poorly understood. Volumetric spore traps were operated at two different locations in Norway over several years. During the season of asocspore release (approximately 1 April to 30 June), on 25 occasions when rain started during night (after 23:00 h and before 04:00 h) and leaves remained wet until at least midnight the following day, the cumulative percentage of spores trapped at sunrise did not exceed 1%, irrespective of temperature. Three hours after sunrise, cumulative ascospore release reached 0.8%, 3.0%, and 8.1% at temperatures of 0 to 5°C, 5 to 10°C, and >10°C, respectively, and 50% release occurred at 11, 9, and 8 h after sunrise. Additional field and laboratory studies indicated that the protracted dawn and dusk of Nordic latitudes, either alone or in combination with low temperatures, does not substantially alter previously reported patterns of ascospore release.  相似文献

Freshly harvested seeds of Poa annua L. collected in south Louisiana were stored in moist soil at seven temperatures between 5°C and 35°C. At monthly intervals, seed lots were removed and germinated at each of the seven temperatures. Seed were dormant for at least 1 month at all test temperatures. Seeds stored for 2 months at 30 and 35°C showed conditional dormancy; there was 100% germination at 10 or 15°C, and poorer germination at 5 or 20°C. Seeds started to lose viability after 2 months at 35°C and were dead after 7 months. In seeds stored at 10–30°C, there were increased percentages and a wider range of germination temperatures as storage time or storage temperatures increased. Seeds stored at 10°C remained dormant for 9 months, but by 12 months of storage the seeds germinated only at 5 or 10°C. Nearly all seeds stored at the same temperatures in air dry soil remained dormant for 6 months, regardless of storage temperature. These results differ from other reports of low temperatures breaking seed dormancy in Poa annua L. and suggest an adaptation to subtropical climates.  相似文献

 利用碱抽提法从串珠镰刀菌(Fusarium moniliforme)菌丝细胞壁得到多糖粗提物。该多糖提取物可以激活寄生于串珠镰刀菌上的活体寄生真菌纤细齿梗孢(Olpitrichum tenellum)的孢子萌发,多糖浓度为0.2 mg/m L时,萌发率达80.5%。纤细齿梗孢的孢子仅在存在该多糖提取物的条件下才能萌发,在马铃薯-葡萄糖或葡萄糖-酵母液体培养基上不能萌发,一些糖、氨基酸、无机盐、维生素不能促使其萌发,光也不影响萌发。纤细齿梗孢孢子萌发最适p H是7.0,最适温度25℃。结果表明,串珠镰刀菌菌丝细胞壁多糖可能在激活纤细齿梗孢孢子萌发中起重要作用。  相似文献

Thermal requirements for the germination of Amaranthus quitensis, a common annual weed in Argentina, were studied. In addition, temporal changes in dormancy from seeds produced at different times during the growing season were examined. For this second objective, thermal and light requirements for germination were tested in seeds buried at different depths, with or without crop residues. Base and optimum temperatures for germination rates were 12.8°C and 37°C respectively. At dispersal time, maximum percentage germination was 60–70% and this was generally recorded at 35°C/25°C in a 14-h photoperiod. Seed germination tended to increase in later seed collection dates. Seeds of A. quitensis showed seasonal changes in germinability in the soil. In winter, germination of retrieved seeds increased to over 90% until summer, after which there was a decrease until the following winter when germination was close to 40%. There were no differences in germinability between burial depths and crop residue levels. Germination requirements for alternating temperatures and light tended to disappear after burial. Initial viability was 99% and declined slightly during burial. Soil temperature seems to play a crucial role not only by regulating seasonal changes in dormancy, but also by defining the percentage and the germination rate in non-dormant seeds.  相似文献

本研究通过利用喷雾干燥技术,制备一种木霉菌微胶囊剂。采用单因素和正交试验,优化了喷雾干燥工艺和微胶囊剂配方的技术参数,进风和出风的温度(T-in,T-out)分别为(92±2)℃和(55±2)℃;微胶囊剂配方为:棘孢木霉菌(Trichoderma asperellum,Tr266B)发酵浓缩液含固量为20%,黄腐酸(YHA)与麦芽糊精的含量各为15%,阿拉伯胶和甘油的含量各为2%;喷雾干燥后的微胶囊含水量为7.5%~8.5%。同时,还研究了喷雾干燥温度、贮藏温度和微胶囊的含水量对孢子萌发率的影响。结果表明,在喷雾干燥过程中,T-in和T-out对孢子的萌发率有显著的影响。当T-in和T-out分别高于(95±2)℃和(65±2)℃时,孢子萌发率会下降10%~40%;当贮存温度4℃,微胶囊的含水量在7.5%~8.5%时,贮存6个月后,孢子的萌发率没有发生明显变化;当微胶囊的含水量低于6.5%时,随着贮存时间的延长,孢子的萌发率缓慢下降;当贮存在25℃条件下,6个月后孢子的萌发率不到10%。本研究确定了所开发的微胶囊壁材为黄腐酸、麦芽糊精和阿拉伯胶,保湿剂为甘油,这些材料与棘孢木霉菌Tr266B有良好的生物相容性,为产业化生产和贮存木霉菌微胶囊剂提供了依据。  相似文献

Shoot blights are common diseases of peach trees in Greece. This study is the first report of a shoot blight and canker disease of peach in Greece caused by the fungus Phoma glomerata (Corda) Wollenw. & Hochapfel. The pathogen caused distinct cankers with abundant gumming on shoots of peach trees. The rate of development of P. glomerata in vitro was reduced as temperatures increased from 25°C to 30°C, decreased from 25°C to 15°C, and was totally inhibited at 35 and 10°C. The rate of conidial germination and the germ tube elongation in vitro was reduced as temperatures increased from 25°C to 35°C, decreased from 25°C to 10°C, and was totally inhibited at 2–4°C. Pathogenicity tests showed that 24 peach and nectarine cultivars grown in Imathia Perfecture, Greece, were equally susceptible to P. glomerata. The fungicides thiophanate methyl, carbendazim, and tebuconazole were evaluated against the development of P. glomerata and disease symptoms. All fungicides inhibited the growth and conidial germination of P. glomereta and disease symptoms, and all 30 isolates tested were sensitive to the above fungicides. The disease caused by P. glomerata could be a threat to peach cultivation in Greece and its management should be investigated in the field.  相似文献

牛蒡叶内绿原酸经聚酰氨层析法纯化后,对7种植物病原真菌孢子萌发的影响及其对黄瓜枯萎病和辣椒疫病的防效研究表明,绿原酸对7种植物病原真菌孢子的萌发均有一定的抑制作用,抑制效果因病原菌的不同而存在着明显差异。绿原酸对疫霉菌和葫芦刺盘孢孢子萌发的抑制作用最为明显,在较低浓度时即可造成孢子萌发的明显畸变;对葡萄孢菌孢子萌发的影响最小,只能在较高浓度下延长其孢子萌发时间。盆栽防治试验表明,绿原酸对黄瓜枯萎病的相对防效可达50%以上,显著高于对照;大棚防治试验表明,绿原酸对黄瓜枯萎病和辣椒疫病也有良好的防治效果。  相似文献

Powdery mildew is an important disease of rubber trees worldwide. To assess the effects of temperature and leaf age on conidial germination and disease development, conidia were inoculated onto rubber tree seedlings with leaves at three phenological stages (copper bronze, colour-changing, and light green) and then incubated at six constant temperatures (10, 15, 20, 25, 30, and 35°C). Leaf age did not affect conidial germination (p = .296) whilst temperature did (p < .0001), although conidia were able to germinate at all tested temperatures. The estimated optimal temperature for conidial germination was 23.2°C. Leaf age, temperature, and their interactions had significant effects on conidial infection and hypha number (p < .0001). At 10 and 35°C, more than 2 and 4 days were needed for infection to complete, respectively, compared to <2, 1, 0.5, and 0.5 days for 15, 20, 25, and 35°C, respectively. Sporulation and mildew symptoms were only observed on those inoculated leaves of all stages at 20 and 25°C, and at the copper bronze stage only at 15°C. The latent period on the copper bronze leaves at 15°C was longer (9 days) than at 20 and 25°C (4 days). The latent period at 20 and 25°C increased from 4 to 7 days as the leaf development stage increased from copper bronze to light green. Therefore, temperature affected germination and postgermination growth of rubber tree powdery mildew, whereas leaf age primarily affected postgermination growth of the pathogen.  相似文献

Freshly-collected mature mericarps of Aethusu cynapium were dormant, but some germinated at alternating (16 h low/8 high) temperatures when the seed coverings were removed. Burial during winter increased percentage germination and the temperature range over which it took place. In late spring the range narrowed, first at low and then at higher temperatures, widening again in autumn. Moist storage at both low (4°C) and high (30°C) temperatures overcame dormancy, but exposure to 30°C inhibited subsequent germination at low temperatures. Germination of intact mericarps was consistently lower than that of de-coated seeds. The cyclic change in dormancy status of the seeds appears to interact with the restricting effects of the seed coverings and perhaps other factors in determining the consistent pattern of spring emergence in A. cynapium.  相似文献

禾谷镰孢分生孢子定量接种是研究作物抗禾谷镰孢的必要手段?本试验利用绿豆(mung bean, MB)与羧甲基纤维素(carboxylmethyl cellulose, CMC)液体培养基在相同条件下培养禾谷镰孢, 比较两种培养基对禾谷镰孢产孢效率的影响; 并用两种培养基诱导分生孢子制成相同浓度孢子悬浮液, 对75个玉米家系进行人工接种鉴定, 检测不同培养基所产分生孢子致病性差异?结果表明, CMC培养基诱导分生孢子增长率(k2=1.125)大于MB培养基诱导增长率(k1=0.844)?在培养第14天, MB培养基诱导获得的分生孢子平均浓度为2.51×105个/mL, CMC培养基诱导的为3.62×105个/mL, 比MB培养基诱导多44.22%?用两种培养基诱生的孢子进行田间接种, 玉米穗腐病发病程度无明显差异?CMC培养基具有产孢快?孢子浓度高的优点, 是一种适宜禾谷镰孢分生孢子诱生的高效液体培养基?  相似文献

A technique for inoculation of Brassica seedlings with single resting spores of Plasmodiophora brassicae is described. Three isolates derived from single spores were produced from one population of P. brassicae. They gave different reactions with the European Clubroot Differential (ECD) series, and two gave different reactions from those of the spore suspensions from which they were derived. When two isolates were mixed together, spores of one isolate restricted infection by spores of the other isolate.  相似文献
