Potato seeds of 24 families consisting primarily ofSolanum tuberosum ssp.andigena with someS. vernei germplasm that were segregating for resistance to potato cyst nematodes (Globodera pallida races P4A and P5A andG. rostochiensis race R1A) were obtained from the International Potato Center. These seeds produced 468 clones with sufficient tubers for evaluation for resistance toG. pallida races P4A and P5A in pot tests at the International Potato Center. Twenty-six of these clones, selected for a high degree of resistance toG. pallida races P4A and P5A, were crossed with neotuberosum ×Solanum tuberosurn hybrids that had been selected for resistance toG. rostochiensis race R1A (golden nematode) and the viruses PVX and PVY. The resultant progenies possessed a high degree of resistance to all three races of the potato cyst nematode. From these progenies, 23 clones were selected for resistance to the three races (G. pallida P4A and P5A,G. rostochiensis R1A) of potato cyst nematodes. The better adapted clones with resistance to the 3 races of potato cyst nematodes will be used in the next cycle of back-crossing.  相似文献

Fusarium wilt incited by Fusarium oxysporum f. sp. melonis (Fom) causes severe losses in melon crops. Four physiological races of Fom have been identified: 0, 1, 2 and 1.2. In most cases, resistance to race 1.2 has been described as recessive, polygenic and not race specific. However, some evidence of race-specific effects within melon resistance to race 1.2 has been reported. In this work, we study these effects and assess whether they are due to race-specific resistance. Seeds were obtained from 14 melon accessions that exhibit some level of resistance to race 1.2, and from the lines ‘Charentais-Fom1’ (resistant to races 0 and 2), ‘Charentais-Fom-2’ (resistant to races 0 and 1), and ‘Dinero F1’ (with partial resistance to Fom race 1.2). Melon seedlings were artificially inoculated using two different procedures: ‘continuous shaking’ and ‘tray immersion’. Symptom severity was assessed on leaves using a rating scale from 0 (no symptoms) to 4 (death of the plant). Symptoms were recorded weekly over the four-week period following the first appearance of symptoms and the area under the disease progress curve (AUDPC) was calculated. Six Fom isolates (3 from pathotype Y and 3 from pathotype W) were used in the inoculation. The less aggressive ‘tray immersion’ procedure seems to be more appropriate for detecting the typically small resistance factors of this type of polygenic partial resistance. ‘Kogane Nashi Makuwa’, ‘BG-5384’, ‘Shiro Uri Okayama’, ‘C-211’ and the control, ‘Dinero F1’, showed a high level of resistance to all Fom isolates. However, some genotype × isolate effects were also detected. ‘Baza’, when inoculated with isolate Fom 9302, and ‘Korça, when inoculated with Fom 37mls.1.2W, showed resistance levels similar to that of ‘Dinero F1’; this effect was not observed when ‘Baza’ and ‘Korça’ were inoculated with other isolates. These results are characteristic of race-specific resistance and offer evidence for the presence of this type of resistance to Fom race 1.2 in melons.  相似文献

Twelve accessions ofSolarium fendleri collected in the United States and Mexico were inoculated with 5,000 eggs of host-races 1 and 2 ofMeloidogyne chitwoodi, Columbia root-knot nematode. The test seedlings that were derived from plant introduction true seed lots were maintained for 55 days before harvest and egg count. The results revealed that two accessions, PI 275162 and PI 275165, were non-hosts (final egg count/initial egg inoculation < 0.1) forM. chitwoodi race 1. The results were uniformly confirmed in the second experiment. No resistance was found to race 2. The tetraploidS. fendleri accessions were crossed to a nematode-susceptible cultivated diploid potato clone from aS. phurejastenotomum population. The triploid hybrids expressed resistance to race 1 at the non-host level. After somatic doubling, the resulting hexaploids also expressed non-host-level resistance to race 1. The two resistant accessions had been collected in southeastern Arizona, one each from the Huachuca and Chiricahua Mountains. Each range is an isolated island of high-elevation mesic flora surrounded by typical lowerelevation Sonora-type desert habitat. These accessions are the only known sources of resistance toM. chitwoodi from wildSolanum species in the USA.  相似文献

Introductions of 13 frost-resistant diploid (2n=2x=24)Solanum species (2 non-tuberous) were inter-crossed to produce F1 hybrids involving two frost resistance sources. Successful F1 combinations were: (i) inter-crossed to produce hybrids involving 3 to 4 frost resistance sources, (F1×F1); and (ii) crossed with haploids (2n=24) of frost-susceptible tetraploid (2n=4x=48)S. tuberosum subsp.tuberosum, (F1×tbr). Thirty-five of the 75 crosses between the 13 species produced viable seed. The F1 and F1×F1 progenies segregated to yield plants more resistant to freezing temperatures than the parental species. All attempts to cross these hybrids with 24-chromosometbr were successful. Although tuber production improved following the first cross totbr, the frequency of resistant plants decreased approximately 50%. Thus far, all first back-cross attempts, (F1×tbrtbr, have succeeded. Utilizing hybrids synthesized from several sources of frost resistance as the donor parents, and clonal selections of 24-chromosometbr as the recurring parents, may provide a high order of resistance throughout the backcrossing program and implement selection of frost-resistant, long-day tuberizing parental stocks acceptable to the potato breeder.  相似文献

Five cycles of phenotypic recurrent selection of a population ofSolanum tuberosum ssp.Andigena for adaptation to N. temperate growing conditions have been accomplished. There have been marked improvements in maturity and yield. Unselected F1 hybrids between this population and tuberosum clones yielded 15% more total yield than unselected intratuberosum progenies at Ithaca as spaced plants and 17% as twelve-hill plots. In Huancayo, Peru and Toluca, Mexico the hybrids produced total yields greater than tuberosum progenies and equal to the andigena progenies. In Colombia, the hybrids were intermediate in yield. It appears that the extent of heterotic advantage of the intergroup tuberosum-andigena F1 hybrids may be dependent upon the day length of where the crop is grown.  相似文献

Bacterial leaf blight of rice (BLB), caused by Xanthomonas oryzae pv. oryzae, is one of the most destructive diseases in Asian rice fields. A high-quality rice variety, LT2, was used as the recipient parent. IRBB21, which carries the Xa21 gene, was used as the donor parent. The resistance gene Xa21 was introduced into LT2 by marker-assisted backcrossing. Three Xoo races were used to inoculate the improved lines following the clipping method. Eleven BC3F3 lines carrying Xa21 were obtained based on molecular markers and agronomic performance. The 11 lines were then inoculated with the three Xoo races. All the 11 improved lines showed better resistance to BLB than the recipient parent LT2. Based on the level of resistance to BLB and their agronomic performance, five lines (BC3F3 5.1.5.1, BC3F3 5.1.5.12, BC3F3 8.5.6.44, BC3F3 9.5.4.1 and BC3F3 9.5.4.23) were selected as the most promising for commercial release. These improved lines could contribute to rice production in terms of food security.  相似文献

Three interspecific F3 hybrid clones ofSolarium tuberosum L. x 5.berthaultii Hawkes bearing both Type A and Type B glandular trichomes were assessed for resistance to the green peach aphid,Myzus persicae (Sulzer). In glasshouse trials, aphid mortality and population development on hybrid clones were similar to those on the aphid-resistant parent,S. berthaultii. Trichome exudate of the hybrid, B227-63, was as effective in aphid immobilization as that ofS. berthaultii. Field populations of the green peach aphid on hybrids were intermediate between those onS. berthaultii and susceptibleS. tuberosum cultivars.  相似文献

From 1993 to 1996 three groups of potato genotypes were evaluated for resistance toErwinia spp.: (1) 31 interspecific diploid hybrids (28 resistant and three susceptible), (2) five hexaploid or pentaploid somatic hybrids ofSolanum tuberosum (tbr) ×S. brevidens (brd), and (3) eight cultivars. Two evaluation methods were applied: tuber point inoculation with eitherErwinia carotovora ssp.atroseptica (Eca) orE. chrysanthemi (Ech) to test tuber soft rot resistance and stem base inoculation with Eca to test blackleg resistance. Some resistant diploid hybrids and somatic hybrids oftbr × brd were superior to cultivars for both tuber and stem resistance. Tuber and stem resistance toErwinia spp. in the most resistant diploid hybrids were equal to the highly resistant somatic hybrids oftbr xbrd. Tuber resistance to Eca was highly correlated to tuber resistance to Ech (r=0.815***). In two years of evaluation for stem resistance, three diploid hybrids and a derivative of one of the somatic hybrids (USA M 264) failed to develop symptoms of blackleg following inoculation with Eca, Analyses of variance for tuber and stem resistance indicated significant effects of genotype, year and genotype × year interaction. A positive relationship between tuber and stem resistance toErwinia spp. has been found, however the genetic control of resistance in tuber and stem is partially independent. In the case of Eca the correlation coefficient was r = 0.725***. Therefore it should be possible to obtain resistant genotypes to both blackleg and tuber soft rot. Several resistant diploid hybrids were selected from among those tested, which also have several other characters desirable for potato breeders.  相似文献

The leptine glycoalkaloids fromSolanum chacoense Bitter are potential resistance factors to Colorado potato beetle,Leptinotarsa decemlineata Say. A selected sample of 17 F1 hybrids (4x) from crosses of a 4xS. chacoense clone that synthesizes leptines (8380-1) with 4xS. tuberosum L. lines were selfed to generate F2 hybrids (4x) and were also backcrossed toS. tuberosum. Glycoalkaloid (GA) content in foliage and tubers was measured for 786 sibling genotypes from the two generations in the field at Beltsville, Maryland. Leptines were found in the foliage of all F2 hybrids and in 98% of the backcross genotypes. Leptines were not detected in tubers from either generation. Foliage concentrations of leptines for the F2 hybrids averaged 156 mg/100g fresh weight (FW). The proportion of the total GA content consisting of leptines averaged 42%. By backcrossing the F1 hybrids toS. tuberosum the average concentration of leptines was reduced to 44 mg/100g FW. The proportion of leptines averaged 16%. The tuber contents of solanine plus chaconine glycoalkaloids averaged 52 mg/100g FW for the F2 hybrids and 27 mg/100g for the backcross genotypes.  相似文献

Stripe rust, caused by Puccinia striiformis f. sp. tritici (Pst), is one of the most widespread and destructive diseases of wheat crop worldwide. Growing resistant cultivars is the preferred method of controlling diseases, but only a few effective genes are available. M8926-2, a translocation line developed from interspecific hybridization between Leymus mollis and a common wheat genotype 7182, is highly resistant to the prevalent Chinese Pst races. In order to identify gene(s) for stripe rust resistance, M8926-2 was crossed with susceptible genotype Mingxian 169, and seedlings of parents and F1, F2, and F2:3 progenies were tested with the most prevalent Pst race CYR32 under controlled greenhouse conditions. M8926-2 has a single dominant gene, designated as YrM8926, conferring all-stage resistance to CYR32. Genetic mapping was used to identify molecular markers linked to YrM8926. A linkage group of seven simple sequence repeat (SSR) markers was constructed for YrM8926 using 191 F2-2 population plants and the gene was located on wheat chromosome 2DS. The wheat-L. mollis translocation line background of M8926-2 and disease assessments indicated that YrM8926 is originally derived from L. mollis. The stripe rust resistance gene and closely linked molecular markers should be useful for pyramiding with other genes to develop wheat cultivars with high-level and long-term resistance to stripe rust.  相似文献

Summary Two races ofPhytophthora infestans, differing in virulence against four major (R) genes for resistance, were used to assess the field resistance of differential (R) genotypes, both singly and mixed together in equal proportions. The mixed inoculum led to an over-estimation of the foliage resistance of genotypes with which only one of the races was compatible, by up to two scoring categories of a 1–9 scale in field trials and, to a lesser extent, in glasshouse tests depending on the differential and the test. Assessment of tuber resistance was not significantly affected. The R10 and R2 differentials showed a higher level of field resistance in the foliage and tubers, respectively, than the other genotypes. It is concluded that assessments of genotypes which may possess R genes, using mixed inoculum, should be interpreted with caution, and apparent high levels of field resistance confirmed through genetic studies of the inheritance of the resistance.  相似文献

A survey of 24 wild Oryza accessions identified Oryza australiensis and Oryza rufipogon as potential sources of enhanced photosynthetic rate for introgression into cultivated rice. Photosynthetic capacity per unit leaf area (CER) was associated with leaf N content but not with leaf chlorophyll concentration, flag leaf area, or specific leaf area. Eight fertile, perennial F1 hybrids between O. sativa and O. rufipogon were grown in non-flooded soil, and CER was measured at flowering under saturating light. Two F1 hybrids had greater CER than the average of 26.1 μmol m2 s−1. The F2 progeny from these hybrids were screened for CER in the field, and segregants with even greater rates of photosynthesis were selected. The basis of high photosynthetic rate in the F2 populations was not leaf thickness or leaf chlorophyll content. One F2 line had exceptionally high CER and stomatal conductance. Broad-sense heritability on an individual plant basis for CER in two F2 populations was 0.44 and 0.37. A highly significant offspring-parent regression of 0.89 for CER was observed in a replicated field evaluation (four blocks, five plants per plot) of 20 vegetatively propagated F2 selections and their F3 seedling progeny. Broad-sense heritability for CER on a plot-mean basis was estimated as 0.74 for both selected F2:3 families and for the selected F2 clones. Genetic resources in the genus Oryza may represent a source of alleles to increase leaf photosynthetic rate in the cultivated species, which we have demonstrated to be a heritable, though environmentally variable, trait in an O. sativa/O. rufipogon population.  相似文献

为了研究披碱草属种间杂种抗旱性优势的遗传机理,以加拿大披碱草(J1)、老芒麦(L1)及其种间杂种F1为试验材料,经干旱胁迫处理,采用甲基化敏感扩增多态性(MSAP)方法,探究干旱胁迫后亲本及种间杂种F1的DNA甲基化变化规律。结果表明,亲本材料L1和J1的DNA平均甲基化水平分别为45.41%和59.53%,杂种F1的DNA甲基化水平为44.24%,说明杂种在形成过程中发生了DNA去甲基化作用。全甲基化在杂交种的DNA甲基化模式中占主导地位。干旱胁迫前亲本L1、J1和杂种F1的DNA甲基化程度分别为32.73%、41.54%和28.00%,杂种F1甲基化水平为亲本材料的75.40%;干旱胁迫后各材料的DNA甲基化程度均增加,平均增幅分别为14.99、20.44和19.78个百分点,说明干旱胁迫对不同材料DNA甲基化水平均有明显影响,且具有特异性。杂种F1的抗旱性最强,其DNA甲基化水平低于亲本,说明杂交种的抗旱性与DNA甲基化水平存在一定的负相关。  相似文献

Previously published data for potato foliage resistance to late blight were examined in a combined analysis to determine how the genetic backgrounds of the host and pathogen affect the occurrence of resistance QTLs. Data from 19 diploid populations and one tetraploid population that originated from at least 12 different tuber-bearingSolanum species were included. Comparative analysis across all populations revealed three highly active genomic regions on the distal parts of chromosomes 3,4, and 5. The region most consistently detected that conferred foliage resistance inSolanum was located on chromosome 5, near marker locus GP21. A previously identified cluster of three race-specific R-genes on chromosome 11 was not associated with polygenic resistance. Statistical examination of active QTLs indicates that congruence among QTLs is significantly affected by both genetic relatedness of mapping populations and race ofPhytophthora infestans used for resistance tests. The proportion of congruent QTLs for late blight resistance decreased from 0.52 detected in relatedSolanum populations tested with the same race ofPhytophthora infestans to 0.20 found in unrelated mapping populations tested with different races ofPhytophthora infestans. Analysis of resistance mapping tests provided statistical evidence for the occurrence of race-specific QTLs in tuber-bearingSolanum. The highly conserved genomic regions identified in the comparative analysis are likely to be good candidates for gene cloning or marker assisted selection in potato breeding programs.  相似文献

采用鉴别寄主鉴定技术,对2009~ 2010年采自河北省的120份玉米大斑病菌菌株进行生理小种组成及毒性分析.结果表明,共鉴定出15个生理小种致病类型,即0、1、3、N、12、13、1N、23、2N、3N、123、12N、13N、23N和123N,其中,0号和1号生理小种为优势小种,分别占总分离数的24.2%和20.8%;其次为1N和N号生理小种,分别占总分离数的10.0%和9.2%.春玉米区大斑病菌生理小种的结构较复杂,出现多个生理小种类型,夏玉米区大斑病菌生理小种的结构相对较简单.将各菌株进行毒性分析发现,河北省玉米大斑病菌对Htl和HtN基因的毒性较高,毒性频率分别为57.5%和37.5%;对Ht2和Ht3基因毒性较低.  相似文献

The inheritance of trichome density and droplet size was investigated in diploid and tetraploid crosses. In haploidTuberosum ×Solanum berthaultii and (S.phureja ×haploid Tuberosum) × S.berthaultii crosses, droplet size on type B trichomes was small or droplets were lacking on F1 hybrids. Parental phenotypes were recovered in the F2 and backcross generations. Trichome density was more variable within a clone that was droplet size. Density of both type A and type B trichomes appears to be under the control of small numbers of genes, as parental phenotypes were recovered in the F2 and backcross generations. The variation observed for each of the three traits studied could not be explained by hypotheses based on the segregation of one or two loci. The insect resistance ofSolanum berthaultii appears to be due to the interaction of several chemical and physical factors, thus potato breeders should handle it as a quantitatively inherited trait.  相似文献

The leaf phenolics of the potato speciesS. × ajanhuiri, S. stenotomum, S. megistacrolobum and the artificial F1 S. stenotomum × S. megistacrolobum hybrids have been compared using thin layer chromatography The wild potatoS. megistacrolobum produced unidentified phenolic compounds which were present in the artificialS. stenotomum × S. megistacrolobum hybrids and the YARI group ofS. × ajanhuiri, but which were absent inS. stenotomum. This further supports the hypothesis thatS. X ajanhuiri is of hybridogenic origin and was derived from natural crosses between the cultivatedS. stenotomum and the wildS. megistacrolobum.  相似文献

The existence of races within the species Nacobbus aberrans sensu Sher has been confirmed; however, there is no consensus on a consistent system for race determination. Four plant species previously used as differential hosts in some N. aberrans race tests (tomato, pepper, sugarbeet and potato) were tested for susceptibility to seven nematode populations from Argentina. Plants were inoculated with second-stage juveniles and kept under glasshouse conditions at 21 °C for 90 days. The parameters evaluated (root gall index, egg mass index, and reproduction factor) in different nematode populations and plants showed significant differences. The reproductive fitness of the nematode populations differed among the plants that showed some degree of susceptibility. The results showed that N. aberrans comprises populations differing in host preference. Based on the nematode populations’ behaviour on these hosts, two groups were distinguished: i) populations that multiplied on all hosts, ii) populations that did not multiply on potato. The present work contributes to the analysis of criteria for developing a differential host test.  相似文献

为明确冬小麦天867对国内当前流行条锈菌优势小种的抗性及遗传规律,并为在育种中对该品种的推广布局提供科学依据,本研究对其进行了田间混合和分小种接种、室内苗期接种和高温成株接种,并通过接种F2遗传群体,分析了天867对不同小种的抗性和遗传规律。结果表明,天867对CYR29、CYR30、CYR31、CYR33、Sun11-4和Sun11-11的抗性免疫到近免疫,其控制不同小种抗性的基因数目和互作方式不同,而对CYR32仅具有高温成株抗性。  相似文献

Traditional sweet corn is poor in provitamin-A, lysine and tryptophan, deficiency of which causes serious health problems. Here, parental lines of two shrunken2 (sh2) -based sweet corn hybrids viz., ASKH-1 and ASKH-2 were targeted for introgression of crtRB1 and opaque2 (o2) genes through marker-assisted backcross breeding. Gene-based markers; umc1066 (SSR) and 3′TE-InDel were utilized for foreground selection of o2 and crtRB1, respectively in BC1F1, BC2F1 and BC2F2 generations. Background selection employing 102–113 polymorphic SSRs led to >90% recovery of recurrent parent genome. Reconstituted hybrids recorded high mean provitamin-A (18.98 μg/g) with a maximum of 7.7-fold increase over original hybrids (3.12 μg/g). High mean lysine (0.39%) and tryptophan (0.10%) with an average enhancement of 1.71- and 1.79-fold, respectively was recorded among reconstituted hybrids over original versions (lysine: 0.23%, tryptophan: 0.06%). Improved hybrids exhibited high phenotypic resemblance with their original hybrids. The average cob yield (11.82 t/ha) and brix (17.66%) of improved hybrids was at par with their original versions (cob yield: 11.27 t/ha, brix: 17.04%). These biofortified sweet corn hybrids rich in provitamin-A, lysine and tryptophan hold immense significance as multinutrient-rich balanced food. This is the first report to stack sh2, crtRB1 and o2 genes to improve nutritional quality in sweet corn.  相似文献
