Greenhouse trials conducted in 2003 and 2004 investigated the impact of trichothecenes on the severity of seedling blight and root rot in common wheat ( Triticum aestivum ), durum wheat ( Triticum turgidum var. durum ), barley ( Hordeum vulgare ) and triticale (× Triticosecale 6x ) using two trichothecene-producing and two trichothecene-nonproducing Fusarium graminearum strains. In 2003 seedling emergence and survival following soil infestation of the trichothecene-producing strain (Gz3639) were significantly reduced compared with the trichothecene-nonproducing strain (GzT40), while root-rot incidence and severity were increased significantly. In 2004, two trichothecene-producing strains (Gz3639 and GzT106) reduced seedling emergence and survival ( P  ≤ 0·01) in eight of 10 crops/cultivars based on single-degree-of-freedom contrasts. However, when results from all strains were combined no significant differences were observed between two trichothecene-producing and two trichothecene-nonproducing F. graminearum strains. Inoculation with GzT106, a trichothecene-producing 'add-back' strain, resulted in more severe root rot symptoms in eight of 10 cultivars ( P  ≤ 0·01–0·05) and lower seedling emergence and survival in seven of 10 cultivars ( P  ≤ 0·01–0·10), compared with the wild-type parental strain Gz3639. 随着北京市小麦种植面积的逐步恢复, 小麦赤霉病和茎基腐病的发生范围也逐渐扩大。为明确北京地区小麦赤霉病及茎基腐病的病原群体组成, 于2023年从北京市6个区14个采样点采集小麦赤霉病和茎基腐病样品, 采用组织分离法得到290株小麦赤霉病菌株及163株小麦茎基腐病菌株。采用翻译延伸因子TEF1-α基因进行分子鉴定, 明确了北京地区不同区县小麦赤霉病菌和茎基腐病菌的群体组成。北京地区小麦赤霉病的病原主要是禾谷镰孢Fusarium graminearum和假禾谷镰孢F. pseudograminearum, 其中禾谷镰孢占群体总数的92.41%, 是整个北京地区的优势致病菌, 也是海淀、平谷、顺义、密云和延庆地区的优势致病菌, 假禾谷镰孢是怀柔地区的优势致病菌。北京地区小麦茎基腐病的病原主要是假禾谷镰孢(45.40%)和禾谷镰孢(37.42%), 还分离出层出镰孢F. proliferatum (1.23%)、中华镰孢F. sinense (7.36%)、锐顶镰孢F. acuminatum (4.29%)、三线镰孢F. tricinctum (3.07%)和木贼镰孢 F. equiseti (1.23%)。海淀和密云地区的小麦茎基腐病优势致病菌是假禾谷镰孢, 平谷、顺义和延庆地区的优势致病菌是禾谷镰孢。该研究结果为小麦赤霉病和茎基腐病的预测预报和精准防治提供依据。  相似文献

玉米赤霉烯酮(zearalenone,ZEN)是由镰孢菌产生的真菌毒素,该毒素具有雌性激素生物活性。PKS4基因是禾谷镰孢菌中玉米赤霉烯酮生物合成途径中的必需基因。根据PKS4基因序列,设计一对PKS4基因特异性引物,通过检测PKS4基因的存在间接检测ZEN毒素的产生。特异引物在禾谷镰孢菌菌株以及被禾谷镰孢菌侵染的小麦籽粒中都能稳定地扩增出大小为1 076 bp的特异片段,扩增片段与PKS4基因(DQ019316)相同区段序列相似性达99.63%。同时,酶联免疫吸附法(ELISA)验证了PCR的检测结果,证明了该技术的可靠性。  相似文献

为探明不同杀菌剂对小麦赤霉病和小麦籽粒DON毒素(包括DON、3-ADON和15-ADON)的控制效果, 采用菌丝生长速率法测定了12种药剂对禾谷镰刀菌野生型菌株PH-1的室内活性, 同时采用液相色谱-串联质谱法(LC-MS)测定了这些药剂对DON毒素的抑制效果, 并开展了小麦赤霉病及籽粒DON毒素的田间防治试验。结果表明, 12种原药对菌丝生长抑制活性强弱依次为氟唑菌酰羟胺>咪鲜胺>戊唑醇>丙硫菌唑>叶菌唑>氰烯菌酯>氟环唑>多菌灵>甲基硫菌灵>吡唑醚菌酯>嘧菌酯>井冈霉素A。氟环唑EC50和EC90离体胁迫均刺激DON毒素产生, 其他杀菌剂EC50和EC90胁迫均抑制DON毒素产生。田间试验结果表明, 200 g/L氟唑菌酰羟胺SC、30%丙硫菌唑OD和20%叶菌唑WP病指防效和DON防效为87.68%~94.77%; 430 g/L戊唑醇SC、25%氰烯菌酯SC、45%咪鲜胺EW、25%氟环唑SC、50%多菌灵WP和70%甲基硫菌灵WP病指防效和DON防效为57.63%%~85.49%; 250 g/L吡唑醚菌酯EC和250 g/L嘧菌酯SC病指防效分别为72.18%和51.98%, DON防效分别为43.06%和-7.96%; 24%井冈霉素A AS病指防效和DON防效分别为42.37%和62.87%。药剂离体和田间控毒效果不完全一致, 赤霉病有效防控是DON防控的前提, 病害防效与DON防效不完全一致, 本研究为小麦赤霉病及籽粒DON毒素防控提供了科学依据。  相似文献

小麦穗组织中脱氧镰刀菌烯醇毒素的免疫细胞化学定位   总被引:5,自引:0,他引:5  
 采用免疫细胞化学技术对禾谷镰刀菌(Fusarium graminearum)在侵染小麦穗部过程中产生的脱氧镰刀菌烯醇毒素(deoxynivalenol,DON)进行了定位分析。在接种后24h,当菌丝在外稃、内稃的内侧表面扩展而尚未侵入寄主细胞前,病菌已分泌DON,并且DON已扩散到寄主组织内。在菌丝细胞内,DON主要被定位于细胞质、线粒体及细胞壁上;在寄主细胞中DON主要分布于细胞壁、叶绿体、细胞质和内质网上。在侵染初期(接种后2 d),菌丝仅能在寄主细胞间隙扩展,随寄主组织中DON浓度的升高,寄主细胞相应发生了一系列病理变化。随寄主细胞坏死(接种后3~4d),病菌进入坏死的寄主细胞。上述结果表明,DON在禾谷镰刀菌的侵染、致病和定殖过程中起着重要的作用。毒素标记结果表明病菌产生的毒素可通过穗轴微管束组织从侵染部位向上、向下转输,毒素向上的转输量明显高于向下转输  相似文献

Soybean (Glycine max) is the most important crop in Argentina. At present Fusarium graminearum is recognized as a primary pathogen of soybean in several countries in the Americas, mainly causing seed and root rot and pre‐ and post‐emergence damping off. However, no information about infections at later growth stages of soybean development and pathogenicity of F. graminearum species complex is available. Therefore, the objectives of this study were to compare the pathogenicity of F. graminearum and F. meridionale isolates towards soybean under field conditions and to evaluate the degree of pathogenicity and trichothecene production of these two phylogenetic species that express different chemotypes. Six isolates of F. graminearum and F. meridionale were evaluated during 2012/13 and 2013/14 soybean growing seasons for pod blight severity, percentage of seed infected in pods and kernel weight reduction. The results showed a higher aggressiveness of both F. graminearum and F. meridionale species during the 2013/14 season. However, the differences in pathogenicity observed between the seasons were not reflected in a distinct trichothecene concentration in soybean seeds at maturity. Fusarium meridionale isolates showed similar pathogenicity to F. graminearum isolates but they were not able to produce this toxin in planta during the two field trials.  相似文献

Fusarium head blight (FHB) resistance of 50 cultivars from the National List of winter wheat cultivars approved for sale (or were undergoing trails for approval in 2003) in the UK was compared with 21 reference cultivars from continental Europe which had previously been characterized for resistance. Only three UK National List cultivars (Soissons, Spark and Vector) had stable resistance over trial sites that was significantly greater than that of the FHB susceptible cultivar Wizard. In addition, under moderate disease pressure, 21 of the National List cultivars had levels of the trichothecene mycotoxin deoxynivalenol (DON) above the proposed European Union limit of 1·25 ppm in grain. Surveys show that levels of FHB and DON in the UK crop are currently very low, however, should disease pressure increase for any reason, then an improvement in the overall levels of FHB resistance of UK winter wheat germplasm will be required. In order to infer the origin of resistance and to identify potentially novel resistance, allele sizes of microsatellite (simple sequence repeat, SSR) markers linked to quantitative trait loci (QTL) for FHB resistance were compared between the test cultivars and known, characterized resistance sources. The major FHB resistance QTL from the Chinese cv. Sumai-3 (3BS, 5A and 6B), the Romanian cv. Fundulea F201R (1B and 5A) and the French cv. Renan (5AL) were screened with 17 SSRs. No National List cultivar had haplotypes similar to any of these QTL. However, the highly resistant German reference cultivar Petrus had an identical haplotype to cv. Fundulea F201R on 1B indicating that this cultivar has an allelic FHB resistance QTL at that location.  相似文献

A series of experiments was conducted to determine whether type I resistance (resistance to initial infection) to fusarium head blight (FHB) in wheat could be assessed using fungal species/isolates that do not produce deoxynivalenol (DON), a mycotoxin critical to the spread of Fusarium graminearum in the wheat spike. It was shown that, while the non-toxin-producing species Microdochium nivale and M. majus could infect following spray inoculation of wheat spikes, they were unable to spread within the spike following point inoculation. However, although these species might reveal type I resistance, they are not highly pathogenic towards wheat. A nivalenol (NIV)-producing isolate of F. graminearum caused high levels of disease following spray inoculation, but spread only very slowly within the spike and rarely induced bleaching above the point of inoculation. It is proposed that spray inoculation with an appropriate, aggressive, non-DON-producing FHB pathogen may be used to characterize type I resistance to complement point inoculation with a DON-producing isolate to assess type II resistance (resistance to spread within the spike).  相似文献

本文分析了北京市与河北省小麦赤霉病菌群体遗传结构以及基础生物学特性。结果表明所有菌株均为禾谷镰刀菌(Fusarium graminearum),属于一个大的单一群体,群体内具有较高的遗传多样性。毒素化学型测定表明,北京与河北地区小麦真菌毒素污染的主要风险为DON与15ADON毒素。表型测定显示,与F.asiaticum群体相比,F.graminearum具有较高的产孢能力,而生长速率和产毒能力较低。该群体对主要杀菌剂多菌灵、戊唑醇和氰烯菌酯均无抗药性。  相似文献

 本文采用细胞化学方法, 对健康和禾谷镰刀菌(Fusarium graminearum)侵染的小麦穗组织中细胞壁主要成分进行了比较分析。电镜观察发现, 被侵穗部组织细胞壁中的主要成分如纤维素、木聚糖和果胶质的标记密度下降, 显著低于未接种的健康对照组织。结果表明病菌侵染和扩展过程中分泌产生了纤维素酶、木聚糖酶和果胶酶等细胞壁降解酶类, 造成寄主细胞壁成分的分解及细胞壁松弛, 从而有利于病菌在寄主穗部组织中的侵染和扩展。  相似文献

为探索生物炭对小麦赤霉病的防治效果和对产量的影响,设置小麦播种前、返青拔节期单施或两个时期均施生物炭处理,在小麦分蘖期人工接种病麦粒,在抽穗扬花期采用孢子捕捉器对穗层空气中的赤霉病菌孢子数量进行动态监测,并在乳熟期调查各处理的病穗率和病情指数,成熟期测定各处理的理论产量、实际产量和籽粒中脱氧镰刀菌烯醇(deoxynivalenol,DON)毒素的含量。结果表明,施用生物炭后穗层赤霉病菌孢子的数量显著降低,赤霉病病情指数降低,且以基施和返青拔节期追施13 500 kg/hm~2生物炭的处理最优;施用生物炭后小麦千粒重和株高均显著增加(P0.05),其中基施和追施13 500 kg/hm~2生物炭的处理较对照处理增产29.5%(P0.05)。  相似文献

BACKGROUND: Carbendazim (MBC) has failed to control wheat scab, caused by Fusarium graminearum Schwabe, on the eastern coast of China in recent years after about 30 years of application. RESULTS: MBC resistance was found to be common in pathogen populations on the eastern coast and along areas of the Yangtze River. EC(50) and minimum inhibitive concentration (MIC) values of MBC inhibiting mycelium growth of wild-type isolates were less than 0.9 and 1.4 microg mL(-1) respectively, while EC(50) values of resistant collections averaged 7.02 +/- 11.86 microg mL(-1). The slope of the MBC dosage-response curve (DRC) for resistant isolates of F. graminearum was flat: 1 < b < 2.8 for resistant isolates and 3.5 < b < 11 for sensitive isolates). Both field resistant and sensitive MBC strains shared similar temperature sensitivity, fitness and virulence on ears. Field resistant strains and UV-induced mutants showed positive cross-resistance to other benzimidazole derivatives and were mainly at intermediate MBC resistance level. Highly resistant field MBC strains rarely appeared, but only some of the highly resistant MBC UV mutants were insensitive to N-phenylaminecarbamates. No mutation in beta-tubulin was found in F. graminearum, in contrast to mutation in this tubulin which has led to MBC resistance in other plant pathogens. CONCLUSION: MBC(R) isolates have high fitness and competition in field, conferred by a novel molecular mechanism.  相似文献

为明确不同杀菌剂防治小麦赤霉病和减少小麦籽粒中DON(脱氧雪腐镰刀菌烯醇)毒素积累的效果,于大田人工接菌条件下进行了不同杀菌剂对小麦赤霉病的防治试验,并于收获期测定了不同药剂处理的小麦籽粒中DON毒素含量。结果表明:两种处理方式下,50%多菌灵WP,30%多·酮SC,25%戊唑醇WP,25%氰烯菌酯SC和70%甲基硫菌灵WP处理中,除先喷药后接菌条件下25%戊唑醇WP在病粒率和籽粒中DON积累上没有显著防治效果,接菌后1d喷药条件下50%多菌灵WP、25%戊唑醇WP在病粒率上没有显著防治效果;30%多·酮SC在病粒率和籽粒中DON积累上没有显著防治效果外,各处理对小麦的病穗率、病情指数、病粒率和籽粒中DON积累都有显著的防治效果(P0.05)。各处理的病穗率防效在42.4%~83.5%,病情指数防效在44.9%~88.2%,病粒率防效在34.7%~69.4%,籽粒中DON防效在48.0%~86.9%。上述几种杀菌剂具有明显的保护作用,其在先喷药后接菌条件下的防治效果均明显优于接菌后1d喷药的防治效果。两种处理方式下,30%己唑醇悬浮剂、25%嘧菌酯悬浮剂和12.5%烯唑醇可湿性粉剂处理在病穗率、病情指数、病粒率和籽粒中DON积累与清水对照处理没有显著性差异(P0.05)。  相似文献

The genetic diversity and pathogenicity of isolates of Fusarium graminearum and F. asiaticum isolated from wheat heads in China were examined and compared with those of isolates of F. graminearum , F. asiaticum and F. meridionale from Europe, USA and Nepal. Genetic diversity was assessed by SSCP (single strand conformation polymorphism) and AFLP (amplified fragment length polymorphism) analysis and by molecular chemotyping. SSCP analysis of the Fg16F/Fg16R PCR amplicon differentiated F. graminearum , F. asiaticum and F. meridionale and revealed three haplotypes among sequence-characterized amplified region (SCAR) type 1 F. graminearum isolates. AFLP analysis showed a high level of genetic diversity and clustered the majority of Chinese isolates in one group along with other isolates of Asian origin. The second cluster contained F. graminearum isolates from China, Europe and the USA. Of the Chinese isolates, 79% were F. asiaticum and 81% of these were of the 3-AcDON chemotype, with only 9·5% of either chemotype 15-AcDON or NIV. All the Chinese and USA isolates of F. graminearum were 15-AcDON, whereas among the isolates from Europe, 21% were NIV and 8% were 3-AcDON chemotype. No evidence was found for possible differences in aggressiveness between F. graminearum and F. asiaticum . Highly aggressive isolates were present in each region and no evidence was found for any association between aggressiveness and geographical origin or chemotype among the isolates examined. No difference was observed in pathogenicity towards wheat seedlings between Chinese isolates and those from Europe, the USA or Nepal.  相似文献

Fusarium graminearum species complexes (FGSCs), such as Fusarium asiaticum and F. graminearum, are important pathogens that cause Fusarium head blight (FHB) in several cereal crops worldwide. In this study, we collected 342 gramineous weed samples in the proximity of rice fields from May to June 2018 in Korea. Among the 500 Fusarium isolates from the weed samples, 13 species of Fusarium were identified, and F. asiaticum (41.2%), F. avenaceum (18.0%), F. acuminatum (16.4%) and F. graminearum (14.8%) were the most frequently isolated. The trichothecene genotype analysis showed that 206 F. asiaticum strains consisted of the nivalenol (NIV) genotype (n = 195, 94.7%) and 3-acetyldeoxynivalenol (3ADON) genotype (n = 11, 5.3%), whereas 74 F. graminearum strains consisted of the 15-acetyldeoxynivalenol (15ADON) genotype (n = 58, 78.4%) and 3ADON genotype (= 16, 21.6%). Geographical differences were observed in the FGSC and trichothecene genotype compositions, which appeared host-dependent between the southern provinces and mid-eastern provinces. The aggressiveness assessment of FHB showed that the 3ADON chemotype was most aggressive followed by the 15ADON and NIV chemotypes in wheat, while the NIV chemotype was most aggressive followed by the 3ADON and 15ADON chemotypes in rice. The F. asiaticum strains grew slowly and produced fewer conidia and perithecia than the F. graminearum strains, regardless of their chemotypes. The results of this study suggest that F. asiaticum with the NIV chemotype has a host preference for rice, and FHB-causing pathogens can be harboured in gramineous weeds, which play a role in the dispersal of FHB pathogens to rice and other cereal crops.  相似文献

为明确不同侵染时期对小麦赤霉病发生和籽粒中DON的积累情况,在2015年和2016年于河南省焦作市温县黄庄镇西虢村田间进行小麦(‘豫保1号’和‘周麦18’)不同生育期接种对病穗率、病情指数、病粒率和籽粒中DON毒素积累的影响试验。结果表明:小麦赤霉病菌在抽穗盛期(DAA=-6,Zadoks=55~57)至灌浆中期(DAA=14或15,Zadoks=75)都能侵染麦穗,但在始花期前3~4 d的齐穗期(DAA=-3或-4,Zadoks=59)至始花期后4 d的灌浆初期(DAA=4,Zadoks=71)处理的病穗率、病情指数和病粒率显著高于其他处理(P0.05),其中盛花末期(DAA=1,Zadoks=69)处理的侵染率最高,为小麦赤霉病菌侵染发病的关键生育期。此外,在套袋保湿2 d的条件下赤霉病菌在小麦始花期前6 d的抽穗盛期(DAA=-6,Zadoks=55~57)和始花期后10 d的乳熟期(DAA=10,Zadoks=73)也能有较高的侵染率。在籽粒中DON积累方面,‘豫保1号’所有处理的籽粒中DON含量与病穗率、病情指数和病粒率的趋势一致,都是盛花末期(DAA=1,Zadoks=69)处理的籽粒中DON含量最高(P0.05),且间隔时间离盛花末期(Zadoks=69)越近籽粒中DON含量越高;而‘周麦18’齐穗期(DAA=-4,Zadoks=59)至灌浆中期(DAA=14,Zadoks=75)处理的没有显著性差异,说明小麦赤霉病菌在始花期后14 d的灌浆中期侵染也能引起籽粒中DON的大量积累。  相似文献

Quantitative PCR analysis and visual disease assessment (VDA) were used to study Fusarium culmorum and F. poae ear blight of wheat and its fungicidal control in three glasshouse trials (1994–5 and 1996–7). VDA indicated high levels of both diseases in the trials, while PCR analysis showed that the amounts of F. poae detected in infected plants were low relative to the amounts detected for F. culmorum . PCR and VDA analysis both indicated that the demethylase-inhibiting (DMI) fungicides prochloraz and tebuconazole significantly decreased F. culmorum and F. poae ear blight. The PCR results, however, revealed levels of disease control by fungicide treatments that were consistently higher than those suggested by VDA. Overall, both fungicides appeared equally effective in controlling the two pathogens. PCR and VDA analysis indicated that the anilino-pyrimidine fungicide pyrimethanil had no significant effect on F. culmorum ear blight. Correlations between VDA at cereal growth stage 80 and PCR analysis were similar for F. culmorum and F. poae . Yield analysis, as measured by 1000 grain weight, indicated that DNA content more accurately predicted yield loss than did VDA scores. Inoculation with F. culmorum significantly reduced yield and significant relationships were observed between F. culmorum disease (as assessed visually or by PCR) and yield, with yield decreasing as disease increased. In contrast, inoculation with F. poae had no significant effect on yield and no significant relationships were observed between F. poae disease and yield. These results have important implications for field studies of Fusarium ear blight of wheat because they highlight the importance of identification of the causal organisms to the species level.  相似文献

为探究麦田前茬作物玉米秸秆和水稻秸秆上中国小麦赤霉病菌的两个优势种Fusarium graminearum sensu stricto (F.graminearum)和F.asiaticum子囊壳形成和发育成熟过程的差异,分别选择3株F.graminearum菌株(SE81,3-ADON型;LcA-2,15-ADON型;HX5-1,NIV型)和2株F.asiaticum菌株(M4A,3-ADON型;M31-2,NIV型),用其孢子悬浮液接种玉米秸秆和水稻秸秆,放置于花泥和地表两种环境条件下,比较接种不同菌株秸秆上子囊壳的形成和发育成熟情况及5个菌株对‘郑麦366’(高感小麦赤霉病品种)的致病力。结果表明:花泥环境下产生子囊壳的速度要快于地表,以玉米秸秆为载体子囊壳产生速度比水稻秸秆快且多数密度大,3个F.graminearum菌株(SE81、LcA-2和HX5-1)在同种秸秆相同环境下比2个F.asiaticum菌株(M4A和M31-2)产生子囊壳的速度快且子囊壳密度大。5个菌株在不同环境的不同秸秆上产生的子囊壳的成熟度均无规律,接种‘郑麦366’后其病情指数间有极显著差异(P<...  相似文献
