Blood samples were collected, at one day of age and at weekly intervals from one through 25 weeks of age, from seven white-tailed fawns (Odocoileus virginianus) to determine the effect of age upon serum biochemical and hematological values. Serum total protein concentration increased continually during the six month period. The rate of anabolism of serum gamma-globulin exceeded the rate of catabolism at about seven weeks of age. Serum cholesterol concentration more than doubled from one day to eight weeks of age. Blood hemoglobin concentration and packed cell volume increased markedly during the first eight weeks of life, but tended to remain constant from eight to 25 weeks of age. Blood erythrocyte count increased throughout the duration of the study; however, the relative increase was greater during the first eight weeks of age than thereafter.  相似文献

Nineteen serum biochemical and seven hematological parameters were determined for crossbred swine from birth through eight weeks of age. From birth (before nursing) to eight hours (after ingestion of colostrum) of age, there was an increase in concentrations of serum total protein, blood urea nitrogen and total bilirubin and increased activities of glutamic-oxaloacetic transaminase, alkaline phosphatase and lactic dehydrogenase. There was a decrease in serum sodium, chloride and potassium concentrations, hemoglobin concentration and packed cell volume during the same period. There was an increase in both serum potassium concentration and erythrocyte count from five (weaning) to six weeks of age. At the same time, there was a decrease in serum sodium and chloride concentrations. The mean concentration of serum cholesterol did not change during the first 24 hours of neonatal life; however, it increased during the 24 to 72 hour period with a linear decrease to six weeks of age.  相似文献

ObjectiveTo characterize the hemodynamic effects of dexmedetomidine in isoflurane-anesthetized cats.Study designProspective experimental study.AnimalsSix healthy adult female cats weighing 4.6 ± 0.8 kg.MethodsDexmedetomidine was administered intravenously using target-controlled infusions to maintain nine plasma concentrations between 0 and 20 ng mL?1 in isoflurane-anesthetized cats. The isoflurane concentration was adjusted for each dexmedetomidine concentration to maintain the equivalent of 1.25 times the minimum alveolar concentration, based on a previous study. Heart rate, systemic and pulmonary arterial pressures, central venous pressure, pulmonary artery occlusion pressure, body temperature, and cardiac output were measured at each target plasma dexmedetomidine concentration. Additional variables were calculated. Arterial and mixed-venous blood samples were collected for blood gas, pH, and (on arterial blood only) electrolyte, glucose and lactate analysis. Plasma dexmedetomidine concentration was determined for each target. Pharmacodynamic models were fitted to the data.ResultsHeart rate, arterial pH, arterial bicarbonate concentration, mixed-venous PO2, mixed-venous pH, mixed-venous hemoglobin oxygen saturation, cardiac index, stroke index, and venous admixture decreased following dexmedetomidine administration. Arterial blood pressure, central venous pressure, pulmonary arterial pressure, pulmonary arterial occlusion pressure, packed cell volume, PaO2, PaCO2, arterial hemoglobin concentration, mixed-venous PCO2, mixed-venous hemoglobin concentration, ionized calcium concentration, glucose concentration, rate-pressure product, systemic and pulmonary vascular resistance indices, left ventricular stroke work index, arterial oxygen concentration, and oxygen extraction increased following dexmedetomidine administration. Most variables changed in a dexmedetomidine concentration-dependent manner.Conclusion and clinical relevanceThe use of dexmedetomidine as an anesthetic adjunct is expected to produce greater negative hemodynamic effects than a higher, equipotent concentration of isoflurane alone.  相似文献

2-(l-Menthoxy)ethanol has been frequently employed as a flavoring agent; however, data regarding 2-(l-menthoxy)ethanol toxicity remain limited. We performed a 13-week subchronic toxicity study of 2-(l-menthoxy)ethanol in male and female F344 rats, with doses of 0, 15, 60, or 250 mg/kg body weight (BW)/day orally administered by gavage using corn oil as the vehicle. No significant toxicological changes in general condition, body weight, or food intake were observed in any groups. The hematological assessment showed decreased hemoglobin, hematocrit, mean corpuscular volume, and mean corpuscular hemoglobin and increased platelet count in the male 250 mg/kg group. Serum biochemistry revealed elevated total cholesterol in the 250 mg/kg group of male and female rats, reduced triglyceride in the female 250 mg/kg group, and increased total protein in the male 250 mg/kg group, indicating effects on lipid metabolism and protein synthesis. For organ weights, absolute and relative weights of the liver and adrenal glands were increased in the 250 mg/kg group of both sexes and the male 250 mg/kg group, respectively. Histopathological analysis showed chronic nephropathy in the male 15 mg/kg or higher groups, with increased absolute and relative kidney weights, as well as elevated serum creatinine, in the male 60 and 250 mg/kg groups. However, eosinophilic granules containing α2u-globulin were identified in proximal tubules, suggesting α2u-globulin nephropathy specific to male rats and without toxicological significance. These results indicated that the no-observed-adverse-effect level of 2-(l-menthoxy)ethanol was 60 mg/kg BW/day for both sexes.  相似文献

During infection with T. vivax (strain Y 58) four stages can be distinguished: (a) A prepatent period of about 1 week. (b) An acute or fulminating disease characterized by an almost continuous, fluctuating parasitaemia, a decrease of the number of thrombocytes, blood serotonin level, packed cell volume (PCV) and Hb, serum albumin and γ-globulin fractions, a low serum total lipid content and an initial rise with subsequent decrease of the blood bradykinin level. (c) A critical stage of about 4–5 weeks post infection (pi) during which the animal may develop a progressive parasitaemia and may die from disseminated intravascular coagulation. At necropsy microthrombi were a consistent and most significant observation. Accompanying features were oedema and haemorrhages in the various tissues, pulmonary oedema and lobular infarction, patches of necrosis in liver, spleen and adrenals and nephrotic changes in the kidneys. Perivascular mononuclear infiltrates, resulting in local granuloma formation (heart liver), were a constant feature. Related biochemical changes were an increase of the blood urea and serum lactate dehydrogenase levels. (d) A post-critical stage characterized by an ameliorating condition, periods with no or few trypanosomes in the peripheral blood alternating with relatively high peaks of parasitaemia. This was associated with increased blood bradykinin levels, an increase of the PCV and Hb, serum albumin and γ-globulin fractions, number of thrombocytes and blood serotonin level. A return to normal values of the serum total lipids was observed at this stage. Despite this clinical recovery of the animals, at necropsy (on days 90, 98, and 99 pi) there was a moderate to massive histiocyte-lymphocyte-plasma cellular infiltration of the heart, which led to cardiac insufficiency in two heifers.The rise of the d-amino laevulinic acid dehydratase levels of erythrocytes during the decline of the PCV and Hb values is an indication for the production of many young erythrocytes and thereby for the haemolytic nature of the anaemia.  相似文献

Administration of ground-up tick tissue stabilate (0.75 tick equivalent) by the subcutaneous route to crossbred calves aged 1 week to 1 month led to the development of acute theileriosis. Haematological studies revealed significant progressive decreases in haemoglobin concentration, packed cell volume and red blood cell count, whereas the total leukocyte count showed an initial non-significant leukocytosis followed by a significant leukopenia. Analysis of serum revealed significant increases in levels of alanine aminotransferase, aspartate aminotransferase, alkaline phosphatase, creatinine kinase and -glutamyltransferase, and in the concentrations of uric acid, blood urea nitrogen and bilirubin. The concentrations of total protein, albumin, glucose, cholesterol and calcium showed non-significant decreases, while phosphorus decreased significantly during the terminal stages of the disease.  相似文献

Peripartum mares and neonatal foals are physiologically unstable. Although hormonal changes around the parturition have been well studied in the field of endocrinology, hematological and biochemical changes have been studied little. The purpose of this study was to examine hematological and biochemical changes in peripartum mares and neonatal foals (n = 23; heavy draft horse). The number of white and red blood cells, hemoglobin concentration, hematocrit, mean cell volume, mean cell hemoglobin, mean cell hemoglobin concentration, platelet count in peripheral whole blood, and the concentration of glucose, nonesterified fatty acid, total cholesterol, triglyceride, total protein, albumin, globulin, blood urea nitrogen, creatinine, aspartate aminotransferase, gamma-glutamyltransferase, alkaline phosphatase, creatine kinase, iron, calcium, inorganic phosphate, magnesium, sodium, potassium, and chlorine in serum were measured. The main changes observed in peripartum mares suggested the following: (1) dehydration at the foaling, (2) physical stress by the foaling, (3) muscle damage by the foaling, and (4) change of energy metabolism associated with the beginning of lactation. The main changes observed in neonatal foals suggested the following: (1) dehydration (hemoconcentration) at the birth, (2) intake of colostrum, (3) beginning of urination, (4) functional change of hematopoiesis, (5) change of liver metabolism associated with the beginning of enteral nutrition, and (6) change of milk composition. This study revealed hematological and biochemical dynamics in peripartum mares and neonatal foals.  相似文献

Haematological and blood serum chemical changes were studied in two groups of specific pathogen free chickens infected at five weeks old with different field isolates of infectious bursal disease virus. Blood and serum components were determined five days after infection. There were significant decreases (P less than 0.05) in the total erythrocyte count, packed cell volume, haemoglobin concentration, albumin, albumin: globulin ratio, uric acid and glucose. Serum components which increased significantly (P less than 0.05) were globulins and cholesterol.  相似文献

Plasma biochemical changes were studied for 8 consecutive weeks in Sokoto Red goats experimentally infected by intravenous route injection of 1.6 x 10(7) Trypanosoma brucei. The strain 8/18 was highly infective. The mean packed cell volume significantly decreased from 1 to 8 weeks post-infection (PI) at P < 0.05. During this period, the mean plasma total bilirubin concentrations significantly increased (P < 0.05). The mean plasma direct and indirect bilirubin concentrations significantly increased from 2 to 8 weeks PI (P < 0.05). The mean plasma albumin concentrations did not vary significantly (P > 0.05), but the mean total plasma proteins and mean plasma globulin concentrations significantly increased between 5 and 8 weeks post infection (P < 0.05). There were no significant changes in the mean plasma bicarbonate, creatinine and cholesterol concentration (P > 0.05).  相似文献

The effect of water restriction in Awassi ewes was assessed under two physiological conditions: lactating and dry, over a 3-week-period (August–September, 2002). Eight dry and eight lactating Awassi ewes (with their lambs) were assigned to one of two watering regimes: watered once every three days and daily watering, respectively. Weather data, body weight and rectal temperature were recorded. Venous blood was sampled on the watering day of the water-restricted group, and subjected to haematological, biochemical and electrolytes analysis. Ewes under water restriction lost more weight and had higher packed cell volume, haemoglobin, serum cholesterol, urea, creatinine, total protein and albumin as compared to daily-watered animals. However, lactation seemed to have no significant effect on these serum components. As for blood pH and electrolytes, Na+ and Cl were significantly higher under water restriction; whereas significantly lower K+ and Ca++ but higher pH and Cl were observed in lactating animals. Finally, lower cortisol concentrations were observed in water-restricted animals, warranting further studies on the hormonal adaptation to long term water stress. It was concluded that water restriction induces similar changes in blood physiological indicators in dry and lactating Awassi ewes; while lactation beyond the first month post partum seems to mostly affect blood electrolytes.  相似文献

Background

Understanding mercury (Hg) distribution in blood and the importance of hair as an excretory pathway is critical for evaluating risk from long term dietary Hg exposure. The major objective of this study was to characterize changes in total Hg concentrations in specific blood compartments and hair over time due to long term piscivory.

Methods

Eight sled dogs (Canis lupus familiaris) were fed either a fish and kibble diet (n = 4), or a fish-free control diet (n = 4) for 12 weeks. Concentrations of Hg were monitored throughout the exposure period, and for 10 weeks post exposure, until Hg concentrations in all blood compartments of one of the exposed dogs dropped below detection limit. Additionally, foreleg hair was sampled during acclimation and weeks 0 and 12.

Results

Hg was detected primarily in whole blood and packed cells, although it was sporadically detected at low concentrations in plasma and serum in two of the fish fed dogs. Dogs ingested an estimated average of 13.4 ± 0.58 μg Hg per kg body weight per day. Hg was detectable in whole blood and packed cells within a week of exposure. Detected concentrations continued to rise until plateauing at approximately 3-6 weeks of exposure at a mean of 9.2 ± 1.97 ng/g (ppb) in whole blood. Hg concentration decreased post exposure following 1st order elimination. The mean half-life (t1/2) in whole blood for Hg was 7 weeks. Mean Hg in hair for the fish-fed dogs at week 12 was 540 ± 111 ppb and was significantly greater (about 7-fold) than the Hg hair concentration for the control dogs. The hair to blood ratio for Hg in fish-fed dogs was 59.0 ± 7.6:1.

Conclusions

This study found the sled dog model to be an effective method for investigating and characterizing blood Hg distribution (whole blood, serum, plasma, packed cells) and toxicokinetics associated with a piscivorous diet, especially for Hg-exposed fur bearing mammals (such as polar bears). Although hair excretion and hair to blood Hg ratios were not similar to human concentrations and ratios, the sled dog toxicokinetics of Hg in blood, was more similar to that of humans than traditional laboratory animals (such as the rat).  相似文献

Variation in haematological parameters of Nigerian native chickens was studied using 60 clinically normal frizzle-feathered, naked-neck, and normal-feathered native chickens. These included red blood cell count, haemoglobin, packed cell volume, white blood cell count, mean corpuscular volume, mean corpuscular haemoglobin concentration, serum glucose, urea, cholesterol, albumin, globulin and creatinine. Normal-feathered birds had higher (p < 0.05) mean values compared to frizzled and native neck genotypes except for albumin, red blood and white blood cells, and mean cell haemoglobin concentration. Males generally had higher mean values than their female counterparts across all genotypes. Correlation coefficients among the parameters were significant (p < 0.001) with r values ranging from 0.26 between red blood cell and mean corpuscular haemoglobin to 0.92 between red blood cell and cholesterol. Sufficient genetic variation therefore exists for haematological parameters among Nigerian native chickens that may represent indicator traits for further study. However, the application of molecular tools will provide better understanding and application of these differences.  相似文献

The aim of the present study was to fill the void in data related to hematological and biochemical parameters of donkey foals. Whole-blood and plasma samples were obtained from 16 Amiata donkey foals at birth, at 24 and 48 hours, and at 1, 2, 3, 4, 6, and 8 weeks of age. RBC, WBC, hemoglobin concentration (Hgb), mean corpuscular volume, mean corpuscular hemoglobin, mean corpuscular hemoglobin concentration, platelet count, glucose, creatinine, blood urea, triglycerides, total cholesterol, total bilirubin, aspartate aminotransferase, γ-glutamyl transferase, creatine-phosphokinase, alkaline phosphatase, total calcium, potassium, sodium, phosphorus, and albumin were measured. Similar to equine foals, values of RBC, Hgb, and Hct decreased significantly after the first 48 hours of life in Amiata donkey foals, reaching values similar to adult donkeys. No changes were found for mean corpuscular volume and mean corpuscular hemoglobin concentration during the study period. The WBC was low at birth when compared with subsequent measurements, but significantly increased in the subsequent surveys. Platelet count was lower in the first week, with a secondary peak 2 weeks later, and then a decline again up to the eighth week. In agreement with equine foals, electrolyte concentrations, triglycerides, and total cholesterol did not show significant differences. Creatinine, total bilirubin, and blood urea showed a trend similar to RBC, Hgb, and Hct. For the first time, data of hematological and biochemical parameters in Amiata donkey foals are provided.  相似文献

Sevoflurane has recently been introduced in feline anesthesia. However, its cardiovascular effects have not, to our knowledge, been reported in this species. Six healthy cats, aged 1.81 ± 0.31 years (mean ± SEM) and weighing 3.47 ± 0.11 kg, were studied. Anesthesia was induced and maintained with sevoflurane in oxygen. Body temperature was maintained between 38.5 and 39.55 °C. After instrumentation, end‐tidal sevoflurane concentration was randomly set at 1.25, 1.5, and 1.75 times the individual minimum alveolar concentration (MAC), determined in a previous study, according to a Latin Square Design. Thirty minutes of stabilization was allowed after each change of concentration. ECG and heart rate, systemic and pulmonary arterial pressures, central venous pressure (CVP), and core body temperature were continuously monitored and recorded. Inspired and end‐tidal oxygen, carbon dioxide, and sevoflurane concentrations were measured using a Raman spectrometer, calibrated every 80 minutes with three calibration gases of known sevoflurane concentration (1, 2, and 5%). Moreover, at selected times, pulmonary artery occlusion pressure and cardiac output (thermodilution) were measured, and arterial and mixed venous blood samples were collected for pH and blood gas analysis, hemoglobin concentration, hemoglobin oxygen saturation, packed cell volume (PCV) and total protein determination, and lactate concentration measurement. Cardiac index (CI), stroke index (SI), systemic and pulmonary vascular resistance indices, rate‐pressure product, left and right ventricular stroke work indices (LVSWI and RVSWI, respectively), arterial and mixed venous oxygen contents, oxygen delivery, oxygen consumption, and oxygen utilization ratio were calculated. Data were analyzed by a Repeated Measure Latin Square Design followed by a Tukey's test for 2 × 2 comparisons. Arterial pH significantly decreased from 7.40 ± 0.05 to 7.29 ± 0.07 with the administration of increasing concentrations of sevoflurane. Similarly, LVSWI decreased from 3.72 ± 0.60 to 2.60 ± 0.46 g m?2. Mean arterial pressure, PaO2, mixed venous pH, CI, SI, and oxygen delivery tended to decrease dose‐dependently, whereas CVP, PaCO2, Pv CO2, PCV, and arterial and mixed venous hemoglobin concentrations tended to increase dose‐dependently with the administration of sevoflurane. However, these trends did not reach statistical significance, possibly because of the limited number of animals studied. Sevoflurane seemed to induce dose‐dependent cardiovascular depression in cats.  相似文献

Twenty each of healthy lactating Friesian and Holstein cows were studied. They were of similar age and body weight, nonpregnant in their third lactation and 80–100 days post partum. The investigation was carried out for 8 weeks on two farms at the same time during the hot summer season in Egypt. The first farm was in Demietta (North east of the Nile Delta, 31° 40 N) on 20 Friesian cows and the second was in Fakous (East of the Nile Delta, 30° 40 N) on 20 Holstein cows. On each of the two farms, 10 cows were newly imported and 10 were born in Egypt.The average daily milk yield and total milk production in the third lactation of the newly imported cows were significantly higher than those of the locally born cows for both Friesians and Holsteins. At the same time, the T4, T3, urea-N, haematological values and AST enzyme activity in the newly imported cows were significantly lower than those in the locally born ones in both breeds, while the locally born cows showed significantly lower values for rectal temperature and respiration rate, as well as for AST and Alk-P enzyme activities, than the newly imported cows of either breed.Holstein cows surpassed the Friesians in milk production, as well as thyroid hormone secretion and cholesterol, haemoglobin, packed cell volume and erythrocyte count values. The opposite was found for serum total protein, urea-N and creatinine concentrations, leukocyte count and AST, ALT and Alk-P enzyme activities.  相似文献

The colonial formation of three serotypes of Leptospires on Cox's solid medium was promoted by microaerophilic incubation of one to three per cent of CO2 supplied by carbon dioxide cylinder, sodium carbonate oxalic acid, and candle method. In anaerobic incubation Leptospira pomona grew the same as with CO2 incubation.

The pH of the medium was an important influence on the rate of colonial formation of Leptospires.

Addition of hemoglobin and inactivation of rabbit serum was not an essential condition for rapid colonial formation. It was found that variation in the morphology of leptospiral colonies occurred with hemoglobin from different species and individuals.

The packed cell volume, haemoglobin concentration, erythrocyte sedimentation rate and specific gravity and percentage water of whole blood and plasma were determined at 4‐week intervals in male and female Pilgrim goslings from 1 to 20 weeks of age and in adult geese at 4‐ to 5‐week intervals from 34 to 59 weeks of age.

All parameters studied increased during the growing period except erythrocyte sedimentation rate which declined. There were no significant sex differences at any time during this period.

The packed cell volume of adult males remained relatively constant from 34 to 50 weeks while that of the females declined. After 50 weeks the female level began to rise in contrast to a decline in the males so that the values were about the same for both sexes at 59 weeks. Differences between sexes were highly significant between 43 and 50 weeks.

Changes in haemoglobin content and whole blood specific gravity were positively correlated with changes in packed cell volume while changes in erythrocyte sedimentation rate were negatively correlated.

Plasma specific gravity of adult males remained relatively constant whereas that of females decreased prior to commencement of egg production at 43 weeks, then increased significantly and remained significantly above the male level for the remainder of the adult period.  相似文献

Two different fluid solutions were infused through percutaneous cecal catheters in 6 healthy ponies to determine the effects on body weight; CBC; packed cell volume (PCV); total plasma protein concentration; plasma fibrinogen concentration; abdominal fluid analysis; concentrations of blood urea nitrogen (BUN), serum creatinine, Ca, total CO2 (TCO2), Na, CI, K, and P; and fractional clearance (FC) of Na, CI, K, and P. During intracecal administration of solution 1, FCNa and FCCI were significantly increased, whereas FCK and BUN were significantly decreased. During administration of solution 2, FCNa and serum P were significantly increased, while PCV was significantly decreased. All ponies developed peritonitis during the study. Complications included catheter-related problems, diarrhea, laminitis, and hypocalcemia. We concluded that hydration and electrolyte balance could be maintained by administration of crystalloid solutions intracecally, but that complications were associated with the procedure.  相似文献

In this study, African Catfish (also known as Sharptooth Catfish) Clarias gariepinus were exposed to sublethal concentrations of fenthion of 2.0, 4.0, and 8.0 mg/L for 21 d and allowed to recover for 7 d to investigate the potential for hematological changes. Whole blood was sampled on days 1, 7, 14, and 21 postexposure and after a 7-d recovery period. During exposure, fenthion caused a reduction in red blood cell counts, hemoglobin concentration, and packed cell volume. There was an increase in white blood cell counts but no significant difference in mean corpuscular hemoglobin concentration, mean corpuscular volume, and mean corpuscular hemoglobin. Both increases and decreases were observed in white blood cell differentials. After the 7-d recovery period, both increases and decreases were observed in the hematological parameters. These results reveal that sublethal concentrations of fenthion can cause hematological alterations in African Catfish and that the substance should be used with caution.

A study was conducted to evaluate the effects of three energy supply (E) levels [low (L), medium (M), high (H)], both pre‐partum (PRE) and post‐partum (POST), and their interactions on metabolic parameters and energy balance (EB) in dairy cows of three breeds. In both phases, E levels applied to a total of 81 multiparous cows of breeds Simmental (SI), Brown Swiss (BS) and Holstein–Friesian (HF;= 27 for each breed) were 75%, 100% and 125% of recommendations of the German Society of Nutrition Physiology, using a 3 × 3 factorial arrangement of treatments. During the pre‐calving period, serum concentrations of non‐esterified fatty acids (NEFA) were higher for LPRE cows, and glucose concentrations were elevated for HPRE cows. During the lactation period, NEFA concentrations were greatest for treatment LPOST. Mean concentrations of β‐hydroxybutyrate (BHB) were highest for cows of the LPOST treatment, intermediate for MPOST and lowest for HPOST. Glucose concentrations were lower for LPOST cows. SI cows had lower BHB concentrations both pre‐ and post‐calving and higher glucose concentrations during early lactation than the other breeds. BHB concentration POST was highest for BS cows. Restricted feeding PRE resulted in a better energy status of cows fed above energy requirements POST (EPRE × EPOST interaction). HF cows had a higher EB pre‐calving, whereas SI cows had a less negative EB during early lactation, compared with the other breeds respectively. Correlations of serum NEFA, BHB and glucose concentrations with EB were strongest during the transition period. Results suggest that controlling energy intake during the dry period might be advantageous for the energy status of dairy cows after calving, whereas energy restriction in early lactation leads to metabolic stress. Evidence is provided of a clear relationship between EB and the blood metabolites NEFA and BHB, especially in the transition period.  相似文献
