The aim of this study was to compare the antinociceptive effects of epidural buprenorphine (EB), epidural medetomidine (EM) or epidural buprenorphine–medetomidine (EBM). Eight cats were studied. Thermal thresholds (TT) were measured by increasing the temperature of a probe placed on the thorax. Mechanical thresholds (MT) were measured through inflation of a modified blood pressure bladder to the cat's forelimb. After baseline measurements, EB (0.02 mg/kg), EM (0.01 mg/kg) or half of the doses of each drug (EBM) were administered. Data were analysed using anova ( P  < 0.05) and 95% confidence interval (CI). TT increased from 30 min to 1 h after EB and at 45 min after EM. MT increased from 45 min to 2 h after EB, from 15 min to 1 h after EM and at 30, 45 min and at 2 h after EBM. MT were significantly lower after EB than EM at 30 min. TT were above the upper 95%CI from 15 min to 24 h after EB, from 15 min to 4 h after EM and from 15 min to 8 h after EBM. MT were above the upper 95%CI from 15 min to 5 h, and at 8, 12 and 24 h after EB, from 15 min to 6 h after EM and from 15 min to 6 h and at 12 and 24 h after EBM. All treatments had similar onset. Overall, EB presented longer period of action than EBM and EM. The same magnitude of analgesia was achieved, but with fewer side effects when EBM was compared with EM.  相似文献

Temperature of alfalfa silage was monitored for 28 d at three depths following ensiling in a vacuum bag system. For this silo system, silage was piled on one sheet of plastic and covered by another; the plastic sheets were connected on all sides by rolling and clamping around a polyvinylchloride pipe. The silo was evacuated with a vacuum pump attached to a perforated pipe placed on the bottom sheet of plastic prior to filling. Temperatures increased linearly as depths increased (0.3 to 0.6 and 0.9 m), but temperature measurements were <35.9oC at all depths. Storage cost per ton of silage ensiled was very sensitive to the quantity ensiled and marginally sensitive to DM losses. For a vacuum silo with 40 metric ton (mT) (DM) capacity, storage cost estimates per mT (DM) were $8.90, $9.50, $9.95, and $10.44 for 5, 11, 15 or 19% DM losses, respectively. Temperature differences attributable to post-ensiling days were detected. The performance of 48 midlactation (187+ 45 din milk) cows fed diets based on forages (45.8 to 61.4% DM) of corn (CS), alfalfa, or ryegrass silages (RS) with or without 7.0% cottonseed hulls (CSH) was compared in a 75-d lactation trial. Corn silage was ensiled in an upright silo; alfalfa silage was ensiled in both an Ag-Bag® (Ag-Bag International, Warrenton, OR) and vacuum bag system (BAL and VAL, respectively); RS was ensiled in an Ag-Bag® system. Cows fed CS without CSH produced more milk (21.1 kg/d) than did cows fed the other three silages (17.1 to 19.1 kg/d). No differences in milk production were detected for cows consuming silage from the different ensiling systems, indicating that the vacuum ensiling process can be a satisfactory method of making silage. Cows fed RS consumed less DM (11.0 to 12.1 kg/d) than did cows fed the other three silages (14.4 to 20.4 kg/d). Inclusion of CSH in the alfalfa silage-based diet increased DMI, milk production, and BW gain but had no effect on milk fat or protein percentage. A vacuum silo system may be a satisfactory method of ensiling crops, especially during emergency situations, for short-term ensiling, and for small herds. A dose-response curve was delineated for an acetylcholine-potassium chloride cocktail previously found to be highly effective in chemically defibrillating electrical induced ventricular fibrillation of thoracotomized dogs. The cocktail dosage responsible for the greatest percentage of chemical defibrillations was transthoracically given to nonthoracotomized dogs subjected to ventricular fibrillation; this resulted in 33% chemical defibrillation. Two clinical canine patients were transthoracically administered the acetylcholine-potassium chloride cocktail because they failed to respond to external electrical defibrillation; both were chemically defibrillated. Potassium citrate when substituted for potassium chloride, in the cocktail, or alone appeared to be more efficacious for chemical defibrillation. Magnesium chloride with acetylcholine failed to chemically defibrillate any dog administered this cocktail.  相似文献

OBJECTIVE: To compare hydromorphone with oxymorphone, with or without acepromazine, for preanesthetic sedation in dogs and assess changes in plasma concentration of histamine after drug administration. DESIGN: Randomized clinical study. ANIMALS: 10 healthy mixed-breed dogs. PROCEDURE: Dogs were treated IM with hydromorphone (group H), oxymorphone (group O), hydromorphone with acepromazine (group H/A), or oxymorphone with acepromazine (group O/A). Sedation score, heart rate, respiratory rate, systolic blood pressure, and oxygen saturation were recorded at baseline immediately after drug administration (T0) and every 5 minutes for 25 minutes (T25). Plasma histamine concentration was measured at baseline and T25. RESULTS: Sedation was similar between groups H and 0 at all times. Sedation was significantly greater for groups H/A and O/A from T10 to T25, compared with other groups. Systolic blood pressure was significantly reduced at T25 in group H/A, compared with group H, and in group O/A, compared with group O. Prevalence of panting at T25 was 50% for groups H and O, compared with 20% for group H/A and 30% for group O/A. By T25, heart rate was significantly lower in all groups. Oxygen saturation was unaffected by treatment. Mean +/- SD plasma histamine concentration was 1.72 +/- 2.69 ng/ml at baseline and 1.13 +/- 1.18 ng/ml at T25. There was no significant change in plasma histamine concentration in any group. CONCLUSIONS AND CLINICAL RELEVANCE: Hydromorphone is comparable to oxymorphone for preanesthetic sedation in dogs. Sedation is enhanced by acepromazine. Neither hydromorphone nor oxymorphone caused an increase in plasma histamine concentration.  相似文献

Effects of either sterigmatocystin or aflatoxin, alone or in combination, given orally to guinea pigs were studied. Sterigmatocystin and aflatoxin B1 given alone and in combination at 4.2 mg/day and 0.01 mg/day, respectively, markedly reduced body weight. Although changes in total serum protein were not marked in any of the guinea pigs in this study, sterigmatocystin given alone and aflatoxin given alone significantly ( less than 0.05) decreased alpha2-globulin. The combination of toxins significantly (P less than 0.01) increased albumin and significantly (P less than 0.01) decreased both alpha2- and beta-globulins. Sterigmatocystin depressed complement activity, although not significantly. However, the combination of sterimatocystin with 0.01 mg of aflatoxin B1/day (an amount that does not affect complement activity alone) significantly (P less than 0.01) reduced complement activity. Increased severity of lesions was not found in guinea pigs given aflatoxin at 0.01 mg of B1 equivalents/day in addition to the sterigmatocystin.  相似文献

Feed intake, digestion and digesta characteristics of cattle fed bermudagrass (BG) or orchardgrass (OG) alone or with supplemental ground corn or barley were determined in two 6 x 6 latin squares with 2 x 3 factorial treatment arrangements. In Exp. 1, beef cows (Hereford, Angus and Hereford-Angus; 452 kg) cannulated in the rumen and duodenum were fed BG (7.9% CP, 79% NDF and 8.7% ADL) or OG (9.8% CP, 79% NDF and 7.2% ADL) hays at 1.2% of BW per day either alone or with added ground barley (.64% BW) or ground corn (.60% BW daily). The increase in microbial OM flow with corn was greater for OG than for BG; corn elevated microbial OM flow more than did barley with OG but less than with BG (forage type x grain source interaction; P less than .10). The increase in total tract OM digestion with grain was greater for BG than for OG (supplementation effect and forage type x supplementation interaction; P less than 05). In Exp. 2, Holstein steers (228 kg) were fed BG and OG hays ad libitum either alone or with addition of either 1.07% of BW per day of barley or 1.00% BW of corn. Total DM intake was 2.19, 3.03 and 2.82% BW for BG and 2.14, 2.80 and 2.52% BW for OG alone or with barley or corn supplements, respectively, being affected by forage type, grain supplementation, grain type and a forage type x grain supplementation interaction (P less than .05). Organic matter digested daily (g/d) was higher for OG than for BG, higher with than without grain and higher for barley than for corn (P less than .05).  相似文献

Effects of dried distillers grains plus solubles (DDGS) on ruminal fermentation, degradation kinetics, and feeding behavior of steers offered annual (Eragrostis tef; TEFF) or perennial (Bothriochloa bladhii; OWB) grass hay were evaluated. Ruminally cannulated Angus crossbred steers (n = 6; body weight [BW] = 304 ± 11 kg) were assigned to a 4 × 6 unbalanced Latin square design with four treatments arranged as a 2 × 2 factorial: hay type (OWB or TEFF) and DDGS supplementation (0% or 0.5% BW [dry matter {DM} basis]). Steers had ad libitum access to hay. Periods consisted of a 14-d adaptation followed by 7 d of collection. Residues from the in situ incubations (0, 3, 6, 12, 24, 36, 48, 72, and 96 h post-feeding) were fitted to a first-order kinetics model using the NLIN procedure of SAS. The DDGS decreased (P < 0.01) TEFF DM intake (DMI) by 11.3%, while not affecting DMI of OWB. The greatest DMI was observed for steers supplemented with DDGS, regardless of forage, and least in steers consuming OWB without DDGS (hay type × DDGS; P = 0.03). Non-supplemented steers spent more (P < 0.01) time eating hay. Digestibility of DM tended (P = 0.06) to increase with DDGS supplementation. A hay type × DDGS interaction was observed (P ≤ 0.05) on ruminal effective degradable fractions. The rate of degradation, soluble fraction, and the potentially degradable fraction of organic matter (OM), neutral detergent fiber, and acid detergent fiber (ADF) increased (P ≤ 0.05), while the undegradable fraction of all components decreased (P ≤ 0.01) when steers were offered TEFF compared to OWB. Ruminal DM, OM, and ADF degradation lag-time increased (P ≤ 0.02) in steers offered OWB. Ruminal degradation kinetics were not (P ≥ 0.17) independently affected by DDGS supplementation. Average ruminal pH of steers offered TEFF (P < 0.01) and those offered DDGS (P < 0.01) were lower than OWB and non-supplemented steers. Total concentration of VFA tended (P = 0.09) to increase when DDGS was provided with OWB, while decreasing when TEFF was offered. The acetate:propionate increased (P < 0.01) with DDGS supplementation due to a decrease (P = 0.03) in propionate. Ruminal NH3-N was greater (P = 0.03) in steers offered TEFF compared to OWB, and those supplemented with DDGS (P = 0.03). An annual, in place of a conventional, perennial hay improved intake and digestion of nutrients, without affecting feeding behavior. The supplementation with DDGS appears to affect forage intake, ruminal degradation, and feeding behavior, although not independent of forage quality.  相似文献

ObjectiveTo evaluate the effects of methadone, administered alone or in combination with acepromazine or xylazine, on sedation and on physiologic values in dogs.Study designRandomized cross-over design.AnimalsSix adult healthy mixed-breed dogs weighing 13.5 ± 4.9 kg.MethodsDogs were injected intramuscularly with physiologic saline (Control), or methadone (0.5mg kg−1) or acepromazine (0.1 mg kg−1) or xylazine (1.0 mg kg−1), or acepromazine (0.05 mg kg−1) plus methadone (0.5 mg kg−1) or xylazine (0.5 mg kg−1) plus methadone (0.5 mg kg−1) in a randomized cross-over design, with at least 1-week intervals. Sedation, pulse rate, indirect systolic arterial pressure, respiratory rate (RR), body temperature and pedal withdrawal reflex were evaluated before and at 15-minute intervals for 90 minutes after treatment.ResultsSedation was greater in dogs receiving xylazine alone, xylazine plus methadone and acepromazine plus methadone. Peak sedative effect occurred within 30 minutes of treatment administration. Pulse rate was lower in dogs that received xylazine either alone or with methadone during most of the study. Systolic arterial pressure decreased only in dogs receiving acepromazine alone. When methadone was administered alone, RR was higher than in other treatments during most of the study and a high prevalence of panting was observed. In all treatments body temperature decreased, this effect being more pronounced in dogs receiving methadone alone or in combination with acepromazine. Pedal withdrawal reflex was absent in four dogs receiving methadone plus xylazine but not in any dog in the remaining treatments.Conclusions and clinical relevanceMethadone alone produces mild sedation and a high prevalence of panting. Greater sedation was achieved when methadone was used in combination with acepromazine or xylazine. The combination xylazine–methadone appears to result in better analgesia than xylazine administered alone. Both combinations of methadone/sedative were considered effective for premedication in dogs.  相似文献

ObjectiveTo evaluate the cardiorespiratory, sedative and antinociceptive effects of dexmedetomidine alone or in combination with methadone, morphine or tramadol in dogs.Study designExperimental, blinded, randomized, crossover study.AnimalsSix mixed breed dogs (two males and four females) weighing 10 ± 4 kg.MethodsThe animals were randomly divided into four treatments: D (10 μg kg?1 of dexmedetomidine), DM (dexmedetomidine 10 μg kg?1 and methadone 0.5 mg kg?1); DMO (dexmedetomidine 10 μg kg?1 and morphine 0.5 mg kg?1), and DT (dexmedetomidine 10 μg kg?1 and tramadol 2 mg kg?1). The combinations were administered intramuscularly in all treatments. The variables evaluated were heart rate (HR), respiratory rate (fR), rectal temperature (RT), systolic arterial pressure (SAP), sedation scale and pedal withdrawal reflex. These variables were measured at T0 (immediately before the administration of the protocol) and every 15 minutes thereafter until T105.ResultsA decrease in HR and fR occurred in all the treatments compared with T0, but no significant difference was observed between the treatments. The RT decreased from T45 onward in all the treatments. The SAP did not show a difference between the treatments, but in the DT treatment, the SAP was lower at T30 and T45 compared with T0. The D treatment had lower scores of sedation at T15 to T75 compared with the other treatments, and the DMO and DM treatments showed higher scores at T60 and T75 compared with DT.Conclusions and clinical relevanceThe treatments with morphine and methadone added to the dexmedetomidine showed higher sedation scores than the control treatment and the treatment with tramadol added to the dexmedetomidine showed no relevant differences in any of the variables evaluated in the study.  相似文献
