Brucella suis biotype 1 was isolated from 13.1% of the pigs slaughtered in Kapuk Jakarta, West Java and from 15.09% of the pigs slaughtered in Surabaya, East Java. The prevalence of B. suis by means of the Rose Bengal Plate Test, was 22.3% for West Java and 14.9% for East Java. The Rose Bengal Plate Test detected more B. suis infected animals (73% of the infected animals) than did the Complement Fixation Test (41%) and the Serum Agglutination Test (54.5%). The high infection rate is a potential health danger for the abattoir workers.  相似文献

New and currently used serological procedures were evaluated using sera from cattle that were challenged with B. abortus S544 (S544) after vaccination with either B. abortus S19 (S19) or B. abortus 45/20 (S45/20) as calves or adults. In animals vaccinated with S19, titres to the indirect haemolysis test (IHLT) rose more slowly, declined more rapidly and involved fewer animals than did titres to the complement fixation test (CFT). In animals vaccinated with S45/20 the rough antigen complement fixation test (RCFT) showed persistent titres. At slaughter the IHLT and CFT were found to be more specific and more sensitive than the Rose Bengal Plate Test (RBPT) and Serum Agglutination Test (SAT) in the detection of cattle infected with B. abortus.  相似文献

The objective of this study was to describe the prevalence of brucellosis in various occupations in Tanga, Tanzania. During the year 2004, serum samples were collected from volunteers in various occupations and screened for brucella antibodies using Rose Bengal Plate Agglutination Test. Information on occupation, kind of work and contacts with livestock was collected and age and sex were recorded. The overall sero prevalence of antibodies to Brucella abortus was 5.52%. One variable in a multivariate regression model was significantly associated with seropositivity to B. abortus: abattoir workers. The prevalence of antibody was significantly higher (P < 0.05) among personnel employed in high-risk jobs such as cutting animal throats and cleaning slaughtered animals parts. None of the other factors investigated was associated with seropositivity to B. abortus. This study documents the serological evidence of B. abortus infection for the first time in abattoir workers in Tanzania.  相似文献

SUMMARY Sixty cows, 48 of which had been vaccinated with live Brucella abortus strain 19 (S19) or with killed B. abortus strain 45/20 (S45/20) and 12 of which were unvaccinated animals, were challenged with B. abortus strain 544. Ten of the 27 cattle found to be infected after challenge showed aberrant serological reactions to the Rose Bengal Plate test, serum agglutination test and/or complement fixation test. These 10 cattle were all previously vaccinated with S19 or S45/20. It was concluded that infection in cattle vaccinated with S19 or S45/20 may be more difficult to detect than infection in animals that have no history of vaccination.  相似文献

Swine brucellosis by Brucella suis biovar 2 is an emerging disease whose control is based on serological testing and culling. However, current serological tests detect antibodies to the O-polysaccharide (O/PS) moiety of Brucella smooth lipopolysaccharide (S-LPS), and thus lack specificity when infections by Yersinia enterocolitica O:9 and other gram-negative bacteria carrying cross-reacting O/PS occur. The skin test with the protein-rich brucellin extract obtained from rough B. melitensis B115 is assumed to be specific for discriminating these false positive serological reactions (FPSR). However, B115 strain, although unable to synthesize S-LPS, accumulates O/PS internally, which could cause diagnostic problems. Since the brucellin skin test has been seldom used in pigs and FPSR are common in these animals, we assessed its performance using cytosoluble protein extracts obtained from B. abortus rough mutants in manBcore or per genes (critical for O/PS biosynthesis) and B. melitensis B115. The diagnostic sensitivity and specificity were determined in B. suis biovar 2 culture positive and brucellosis free sows, and apparent prevalence in sows of unknown individual bacteriological and serological status belonging to B. suis biovar 2 naturally infected herds. Moreover, the specificity in discriminating brucellosis from FPSR was assessed in brucellosis free boars showing FPSR. The skin test with B. abortus ΔmanBcore and B. melitensis B115 allergens performed similarly, and the former one resulted in 100% specificity when testing animals showing FPSR in indirect ELISA, Rose Bengal and complement fixation serological tests. We conclude that O/PS-free genetically defined mutants represent an appropriate alternative to obtain Brucella protein extracts for diagnosing swine brucellosis.  相似文献

Madam:– I wish to advise of the recent isolation of Streptococcus suis 1 and 2 from pigs in New Zealand. Type 1 was isolated from brain and joint swabs from a three-week-old piglet. The piglet had meningitis, was laterally recumbent, paddling and had arthritis with grossly swollen joints. Type 2 was isolated from tonsils collected from apparently normal bacon pigs slaughtered at a meat works. Isolates were confirmed as either Streptococcus suis type 1 or 2 on the basis of biochemical characteristics(4) and Lancefield precipitin tests using Sand R antisera as originally used by De Moor.(2)  相似文献

Summary

Data are presented on the incidence of various streptococcal infections in pigs in the Netherlands. 314 Strains isolated in the course of routine post‐mortem diagnosis were examined. The most frequently occurring streptococcus was S. subacidus (bio) type II which was isolated in 31.2% of the cases.

S. suis type 2 (Serogroup R) and S. equisimilis (Serogroup C) constituted 16.2% and 13.7% of the isolates respectively. Besides meningitis, endocarditis and polyserositis S. suis type 2 infections may frequently be associated with pneumonia (42%).

The biochemical profiles of the various S. suis and S. subacidus (bio) types are presented. The profile of both species is almost identical. It seems justified to use the name S. suis for strains with this characteristic profile and to abandon the name S. subacidus. Haemolysis does not appear to be a suitable characteristic to screen for S. subacidus/S.suis types. In comparing three serological methods for typing S. suis type 2, gel precipitation using Fuller's extract and slide agglutination give an almost 100% correlation. A cross sectional sero-prevalence study was conducted on 1,595 cattle in Jimma zone, Ethiopia to investigate the status of bovine brucellosis and identify potential risk factors. Sera samples were analyzed using Rose Bengal Plate Test (RBPT) and Complement Fixation Test (CFT). The overall individual and herd level sero-prevalences were 3.1% (n = 1,595) and 15.0% (n = 227), respectively. The sero-prevalence of bovine brucellosis at individual animal level was significantly higher in non-pregnant (11.18%) than pregnant (2.77%) and lactating (22.35%) than non-lactating animals (2.46%). Moreover, significantly higher sero-prevalence was observed in herds of larger sizes. Individual animal sero-prevalence was also positively associated with the occurrence of abortion (26.98 and 1.54% in those with and without previous history of abortion, respectively). Generally, the sero-prevalence of bovine brucellosis found in Jimma area was not high and the sero-prevalence was closely associated with some of the risk factors considered at individual animal and herd level.  相似文献

A trial was conducted to assess the efficacy of a single intra-muscular injection of 25mg per kg of streptomycin in stopping leptospiruria in growing pigs exposed to a natural infection.

The investigation involved 54 pigs, 10–12 weeks old, that were free of serological evidence of infection with serotypes pomona or tarassovi at the start of the trial. The animals were kept in pens in a finishing house through which flowed effluent from other pens containing infected pigs. Urine samples were collected from each pig three times weekly until it was slaughtered for bacon at 20–24 weeks old.

Leptospiruria was first detected between 16 and 74 days after exposure. Twenty-one of 37 pigs which showed leptospiruria were injected with streptomycin. Leptospirae were detected in 11 of 185 (5.9%) subsequent urine samples from these 21 treated animals and serotype pomona was cultured from the kidney of 1 of the 10 animals that were examined from this group at slaughter.

Following the initial detection of leptospiruria in the 16 un-treated pigs, 179 of 211 (84.8%) subsequent urine samples con-tained leptospirae, which were also cultured from the kidneys of 4 out of 11 of these animals.  相似文献

This study was undertaken to investigate the seroprevalence of brucellosis in unvaccinated sheep from the flocks having previous abortion cases in Kars and around, Turkey and to compare the efficacy of each serological test used. Four hundred serum samples collected from 16 different flocks of sheep having a history of abortions in Kars and its surrounding area in Turkey were examined for the presence of antibodies raised against Brucella using Rose Bengal Plate Test (RBPT), Serum Agglutination Test (SAT), Rivanol Agglutination Test (RAT) and Complement Fixation Test (CFT). All animals were unvaccinated against Brucella. Of the serum samples tested, 147 (%36.7), 142 (%35.5), 139 (%34.75) and 135 (%33.75) were found positive by SAT, RAT, RBPT and CFT, respectively. No statistically significant difference was found between the serological tests used (p > 0.05). It is concluded from this study that brucellosis continues to be an important problem for ovine abortions and poses a risk both for human and other animals in this area. Therefore, adequate intervention measures should be implemented to control and eradicate brucellosis. In addition, if conventional serological tests are used at least two tests, RPBT for screening and CFT for the confirmation of the positive samples are preferable, should be used in parallel for detection of brucellosis effectively. A previous comparative study of 806 human serums showing the similarities and differences between Wright agglutination test, Renoux hem agglutination test and Rose Bengal Plate Test confirmed the practical interest of the easy and cheap Rose Bengal Plate Test in detecting human brucellosis.IRSA of Tours made with this test a systematic detecting of human brucellosis concerning 89 000 persons of agricultural occupation from the middle-west district of France.It revealed a brucellosis infection much more important than we expected among the adult agricultural population: about 2.5% and even 5 or 10% with some populations. The attention of doctors must be called to the so far unsuspected frequency of brucellisation among agricultural people.  相似文献

Porcine brucellosis due to Brucella suis biovar 1 raises important issues for pig breeders in French Polynesia. In this region, the disease is enzootic, spreads silently and engenders economic losses in infected farms as well as sporadic human cases. While serological tests are essential in surveillance and control programmes of animal diseases, to date none of the available tests have been shown to be reliable enough to be used as a gold standard in routine individual diagnosis of porcine brucellosis. Few studies about the estimation of the sensitivity and the specificity of porcine brucellosis screening tests have been published, none of them dealing with French Polynesia. The studied population included 1,595 pigs from French Polynesia. Five tests were evaluated: Rose Bengal test, fluorescence polarisation assay, indirect ELISA, and two competitive ELISAs (C-ELISA). The sensitivity and the specificity of each test were estimated. C-ELISA2 was the most sensitive test (Se C-ELISA2?=?0.954 [0.889; 0.992] 95 % credibility interval (CrI)) while both C-ELISA and Rose Bengal test (RBT) were the most specific ones (Sp C-ELISA1?=?0.856 [0.806; 0.915] 95 % CrI; Sp C-ELISA2?=?0.849 [0.817; 0.879] 95 % CrI; Sp RBT?=?0.853 [0.812; 0.898] 95 % CrI).  相似文献

Summary

The lymphocyte transformation test (using an in vitro whole‐blood lymphocyte stimulation procedure) and the Brucellin skin test were applied to five heifers infected with virulent Brucella abortus strain 544, five cows inoculated with Yersinia enterocolitica serotype 09, and four non‐exposed cows. Lymphocytes from Brucella‐inoculated animals persistently gave very high blastogenic reactions indicative of active Brucella infection. The test was persistently negative in Yersinia‐infected and non‐exposed cattle. Four of thefive cowsinfected with Yersinia enterocolitica type 09 and allfour control cattle were persistently negative to the delayed hypersensitivity skin reaction with brucellin. All cattle infected with Yersinia enterocolitica type 09 were strongly positive to the Rose Bengal, Serum agglutination, Complement fixation and Antibovine globulin tests using Brucella abortus antigens. One lactating cow infected with Yersinia enterocolitica type 09 was positive to Brucella milk ring test. These results indicate that standard Brucella serological tests are unreliable in differentiating the two infections in cattle and that both the Lymphocyte transformation and brucellin skin tests could be used to differentiate bovine brucellosis from yersiniosis.  相似文献

The intracheal inoculation of pigs with Haemophilus suis led to the production of Glasser's disease at every attempt without significant pulmonary involvement. Isolation of this organism from the experimental animals was possible only in the acute phase of the disease.

The indirect fluorescent antibody technique when applied to frozen sections of tissues obtained from the experimentally infected pigs at autopsy, revealed a few rod forms but mostly “round bodies” of H. suis in animals from which the organism was isolated, and “round bodies” only in the pigs from which the organism was not isolated.

Attention is drawn to the similarities between the lesions caused by H. suis and Mycoplasma hyorhinis, and to the confusion which may result therefrom. It is stressed that the laboratory diagnosis of these two diseases is complicated by the fact that both agents may not be isolated on the media commonly used in diagnostic laboratories. Both organisms necessitate the use of special media where the clinical and autopsy results indicate polyserositis and arthritis.

Seven of 18 elk on a deer farm were found by the official Rose‐Bengal agglutination test (RBT) and tube agglutination test to be brucellosis reactors/suspects. Evaluation with the competitive ELISA (C‐ELISA) and the fluorescence polarization assay (FPA) tests revealed that six and five sera were positive respectively. The seven reactors/ suspects were slaughtered and their blood and tissues were collected. Brucella species could be isolated from three of the slaughtered animals, with nine isolates being obtained from the popliteal, supramammary and submandibular lymph nodes, vaginal discharge, mammary tissue and spleen. Brucella genus‐specific PCR based on 16S rRNA and AMOS‐PCR, which is specific for differential Brucella species, revealed that all nine isolates were Brucella abortus. These nine were further confirmed to be B. abortus biovar 1 by classical biotyping scheme assays. This is the first report of an outbreak of brucellosis in domestic elk in Korea. Our observations suggest that deer should be included in the routine Brucella surveillance programme for the effective control and prevention of brucellosis in Korea.  相似文献

This study was carried out to investigate the prevalence of bovine brucellosis and infectious bovine rhinotracheitis (IBR) in organized dairy farms with history of abortion in India. ELISA and Rose Bengal Plate Test (RBPT) were used to detect the seropositive animals and the test results indicated that 22.18% and 13.78% animals were declared as sero-positive by ELISA and RBPT, respectively. Milk Ring Test (MRT) was carried out only in one farm and 12.82% of the tested animals were turned positive. Culture examination analysis of milk samples, two animals revealed the presence of organisms indistinguishable from Brucella spp. The organism was confirmed as brucella by morphological characteristics and biochemical tests. An overall sero-prevalence of antibodies against IBR was found to be 60.84%. None of the genital and nasal swab samples was found to be positive for presence of bovine herpesvirus -1 (BHV-1) on repeated passage in Madin-Darby Bovine Kidney (MDBK) cell lines. Brucella and IBR considered as the causal agent for abortions in these farms. The present study indicates the urgent need and the necessity for control of these infectious diseases which cause heavy economic losses to the organized farms.  相似文献

Despite the endemicity of brucellosis in Nigeria, reports on equine brucellosis are rare. The Rose Bengal Plate Test (RBPT) was used to determine the serologic prevalence of Brucella abortus antibodies amongst 75 horses from three stables in two States of northern Nigeria. The highest prevalence (22.7%) was recorded in stable C located in Plateau State and the lowest (6.7%) was found in stable B in Kaduna State. The overall prevalence determined was 14.7%. There is need for the inclusion of horses in brucellosis surveillance and control strategies in Nigeria to safeguard people at high risk.  相似文献

Summary

Growing pigs were placed on feeds with high (Group A), medium (B) and low (C) dietary energy and were infected with a virulent stock of T. brucei. Eight weeks later, the infected pigs were treated with isometamidium chloride at 1mg/kg live weight and all pigs were subsequently placed on a high energy diet to investigate their response to therapy.

Clearance of T. brucei from blood was completed 72h after treatment. There was no evidence of relapsed infection up to eight weeks after treatment. Red blood cell parameters returned to normal four to six weeks after treatment with responses being fastest in Group A, B and C had gained about two‐thirds of the live weight gains of their non‐infected pair‐fed controls.

It appears that the retarded weight gain as a result of the infection persisted after therapy since drug‐treated pigs did not gain as much weight as their non‐infected controls.  相似文献

A total of 413 pig faecal samples were collected from pre-weaners (119), starters (131), pre-growers (123) and sows (40) from a farm with a closed breeding system segmented into two breeding complexes and a growing complex in the region of Vysočina, Czech Republic and screened for the presence of Cryptosporidium using staining methods and genotyping (SSU rRNA). Cryptosporidium oocysts were detected by microscopy in the faeces of 21.1% of the samples (87/413). Sequence analyses and RFLP identified C. suis in 44, Cryptosporidium pig genotype II in 23 and C. muris in 2 samples. No mixed infections were found.Pigs under 7 weeks of age were infected with C. suis only. Cryptosporidium pig genotype II was found in animals from 7 weeks of age. No relationship was found between diarrhoea and any Cryptosporidium infection in any of the different age groups (P < 0.05). The pre-weaned pigs shed significantly more Cryptosporidium oocysts than older pigs and it was associated with C. suis infection.  相似文献

Mass vaccination with reduced dose 1/50 Rev 1 strain live vaccine (1–2 109 colony forming units), administered subcutaneously, over a four and a half year period reduced the prevalence of Brucella melitensis in Kuwait's small ruminant population from 5.8% in 1993 to 2.02% in 1997.Serological test results using the Rose Bengal Plate Test, Rivanol Agglutination Test and Complement Fixation showed no evidence of persistence of positive serology in animals nine or more months after vaccination. Questionnaires and post-vaccination flock inspections found that the effects on gestation (abortions) were minimal – and not proven to be due to the vaccine.The conclusion from these findings is that mass vaccination with reduced dose Rev 1 administered by the subcutaneous route is a practical field strategy for control of Brucella melitensis.  相似文献
