Summary

The efficacy of the morantel sustained‐release bolus (MSRB) in controlling gastrointestinal parasites in first‐season grazing calves was evaluated on a dairy cattle farm in Belgium. The calves grazed a pasture which had been used by bolus‐treated animals in the three previous years. The effect of bolus administration was determined with respect to live weight gain, faecal egg shedding, herbage larval counts, serum pepsinogen levels and ELISA antibody titres.

In spite of an incomplete reduction of faecal egg shedding during the first months of the grazing season, bolus administration resulted in the prevention of parasitic gastro‐enteritis in the calves. A weight gain advantage of 35,2 kg of the bolus‐treated animals over the controls was noted already at two months after turnout. This weight gain advantage was maintained until housing.

The usefulness of serum pepsinogen values and ELISA antibody titres as parameters in prevention experiments is stressed. Both serological parameters gave more information concerning infection level than did the faecal egg output and the herbage larval counts.  相似文献

The Enzyme Linked Immunosorbent Assay (ELISA) provides a very efficient technique for detecting antibodies against Dictyocaulus vivparus in calves. Although low level cross reactions were found in animals with gastrointestinal nematodes, the specificity and sensitivity of the technique are sufficient for herd diagnosis of lungworm infections and for survey work. This conclusion is reached on the basis of artificially and naturally infected calves. ELISA titres correlate well with Indirect Haemagglutination titres, parasitological findings, and clinical observations.  相似文献

Vaccination of four calves with Leptavoid (Wellcome New Zealand Limited) gave rise to Leptospira interrogans serovars hardjo and pomona microscopic agglutination test titres that could not be distinguished in magnitude from post-infection titres. Vaccination of four calves with Lepto-3 (ICI Tasman Limited) gave rise to much lower titres.

Revaccination of cows with Leptavoid caused a rise in hardjo titres which was significantly greater than after use of Lepto-3.

The possibility that titres were due to the simultaneous infection with serovars pomona and hardjo of only the animals vaccinated with Leptavoid must he discounted.  相似文献

Three red deer calves (Cervus elaphus) died with a haemolytic disease associated with infection by Leptospira interrogans serovar pomona.

Infection within the herd was more prevalent than disease. Sera from 16 herd mates were tested by the microscopic agglutination test (MAT) and 12 had leptospiral titres, the majority to serovar pomona. A few calves had titres to balcanica and hardjo.

Urine was obtained for culture from six of these calves and serovar pomona was isolated from five with titres to pomona, and hardjo from one with a litre to hardjo but not pomona.

A fourth calf died with severe nephritis but a diagnosis of leptospirosis was not confirmed in this case.  相似文献

Four red deer calves (Cervus elaphus) died with severe nephritis apparently associated with infection by Leptospira interrogans serovar pomona. The sera of 12 in-contact red deer calves were examined for leptospiral agglutinins and nine showed titres to pomona consistent with recent infection. Two also showed titres of 1:100 to serovar hardjo. The urine of five of these in-contact calves was examined periodically over a period of nine months. All were initially leptospiruric, four being infected with pomona and one with hardjo. In four animals leptospiruria could only be detected for up to six months, but one animal infected with pomona was leptospiruric for at least eight months.

The apparent source of infection was from infected cattle, and it is suggested that deer are unlikely to act as maintenance hosts for serovar pomona.  相似文献

Summary

The activity of albendazole against gastrointestinal helminths in naturally infected calves in the Netherlands was tested. The calves were in their fist grazing season and kept in two groups of ten. One of these groups was grazed alternately with sheep. Five out of each group were drenched with albendazole (7.5 mg/kg) on the day they were housed (November 1). Before and 2, 14, and 28 days after treatment individual faecal samples were taken from all calves and larval cultures were made. Ten calves, six treated and four untreated, were killed for post mortem studies 14 days after treatment The remaining calves were slaughtered 14 days later.

The drug was highly effective in reducing the egg output, measured as the number of larvae cultured per gram of faeces. Compared with the untreated calves, the reduction was more than 99% two days after treatment, 100% at 14 days, and 99% after 28 days. It was shown that egg output 28 days after treatment came from worms which had developed from arrested larvae of Ostertagia ostertagi that had survived treatment. Post mortem results showed an efficacy of 100% against adult O. ostertagi, of almost 100% against Trichostrongylus axei, and 100% against adult and larval Cooperia oncophora.

Twenty‐eight days after treatment, the reduction of arrested early fourth stages of O. ostertagi was 85% in comparison with the untreated calves. Apparently less effect was found against Trichuris ovis at the given dose rate.  相似文献

Antibodies to P. haemolytica were detected in colostral sera of cows and blood sera of calves by means of indirect haemagglutination (IHA) and ELISA. The ELISA titres of the colostral sera of the dams and of the blood sera of the calves showed a significantly positive correlation. There is a positive correlation between the titres of the two serological methods. The results of the challenge infection with P. haemolytica A 1 do not show a correlation between the titres and the severity of the disease. Heifer calves, however, developed more severe pneumonias than calves from older cows.  相似文献

Calves vaccinated twice with one commercial pomona vaccine had serum MAT titres up to 1:300, warm CFT titres of 1:20 and cold CFT titres of up to 1:80. A second commercial vaccine did not elicit any detectable antibodies.

All calves, vaccinated as well as unvaccinated, showed a rapid rise in antibody levels, detectable with all three tests, following challenge with 38 × 108 pomona organisms.  相似文献

The use of the ELISA method for the detection of antibodies to B. besnoiti in cattle is described and compared to the IFAT technique. One hundred and twenty-one sera were examined, of which 61 were sera of calves experimentally infected with B. besnoiti, 52 sera from field animals and eight were sera with high titres of antibodies to other parasites. The specificity of both assays correlates but ELISA seemed to be more sensitive. The ELISA technique provides a rapid and reliable method for the screening of B. besnoiti infection in cattle.  相似文献

Summary

Calves (n = 2) born to dams with experimentally induced brucellosis, and calves (n = 4) born to dams with naturally occurring infection were examined by the delayed‐type hypersensitivity (DTH) test for possible B. abortus infection. The results were compared with the serum agglutination test, complement fixation test, and Coombs test. Five calves were nursed by their dams for 8–10 weeks after birth. One calf was separated from its dam and fed artificial milk.

Three to five months after birth, four calves tested seropositive in the serologic tests. Antibodies were detected in one calf as early as 1 week after birth. The calf fed on artificial milk was seronegative 4–5 weeks after birth. All calves reacted to the DTH test antigen from week 12 until the end of the experiment, even though serologic tests were negative. We conclude that the DTH test is a valuable technique for diagnosing Brucella in calves born to infected dams.  相似文献

Subsequent to a cross-sectional serological survey of Manawatu dairy farm workers, a case-control study was carried out to investigate the correlation between titres to leptospiral serovars in workers and those in cattle in their herds. A total of 52 herds was investigated, 25 of which were ‘high risk’ where milkers had titres of 1:96 or greater, and 27 were case-controls where milkers had no detectable agglutinin titres at a minimum serum dilution of 1:24.

The serological prevalence of titres to hardjo in cattle on ‘high risk’ farms (76.5%) was significantly higher (P<0.05) than on the case-control farms (60.0%). The geometric mean titres of seropositive cattle on ‘high risk’ farms were also significantly higher (P<0.0l) than in the cattle from the case-control farms, especially in the younger cohorts. These findings suggest that there was active endemic hardjo infection in the two- to three-year-old cattle on the ‘high risk’ farms.

Titres to pomona were demonstrated in only 5.2% of the cattle from both types of farm. Workers with titres to pomona tended to be from farms on which stock, especially calves, were bought-in and pigs were kept.

Conventional measures for protecting milkers from contact with infected urine appeared to be ineffective and it is concluded that prevention of leptospirosis in dairy farm workers can only be achieved by elimination of infection in the herd by vaccination of cattle.  相似文献

SUMMARY Bos indicus-crossbred calves exposed to natural Babesia bovis infection in wet and dry tropical environments in northern Queensland were tested for antibodies using the indirect haemagglutination (IHA) test. There was evidence of maternal antibodies suggesting endemicity of B. bovis in the 2 environments. Within one year 35.3 to 94.6% of the calves had acquired B. bovis infection. The highest titres and most rapid conversion rates were observed just after the wet season when the tick population was at its peak. Clinical disease was not observed.  相似文献

Twelve seronegative cows were vaccinated with an experimental bivalent Leptospira interrogans serovars hardjo and pomona vaccine late in their first pregnancy. Calves born of these dams were divided into 4 equal groups that received this vaccine at 4, 6, 10 and 18 weeks of age, respectively. Before vaccination the group geometric mean titres of maternally-derived circulating antibodies ranged from 2 to 25 for the microscopic agglutination (MA) test and 3 to 35 for enzyme-linked immunosorbent assay (ELISA) using a serovar hardjo outer envelope antigen. Post-vaccination peak titres were 645 to 1612 for MA and 562 to 1037 for ELISA, respectively. Calves vaccinated at the youngest age, had the highest pre-vaccination circulating maternal antibody titres, but showed the smallest rise in post-vaccination antibody titres. Circulating maternal antibody was detected in calves up to 13 weeks of age. All immunised calves were protected against a virulent challenge with serovar hardjo type Hardjobovis, regardless of their age or maternally-derived antibody titres. These findings indicate that calves as young as 4 weeks old, vaccinated in the presence of maternally-derived antibody, can be fully protected against homologous virulent challenge.  相似文献

An investigation was made into the prevalence of leptospiral infection in cattle. An area 50 km radius was selected in a region where leptospirosis was reputedly common. Farmers volunteered 250 herds with 39 500 cows for testing and 7 500 animals were selected and sampled. Twenty-nine cows (0.4%) on 14 (5.6%) of the farms had leptospiruria at the first examination. Leptospirae were cultured from the urines of nine of these animals and all were Leptospira interrogans serovar hardjo. Serologically 12.5% of cows had titres of 1:200 or greater to hardjo and 3.5% titres of 1:200 or greater to pomona. In the Spring of 1977, there was evidence of clinical leptospirosis in calves associated with only one of the herds and no clinical leptospirosis in the 250 lactating herds, although leptospiral titres were found in 88% of them. This indicated that clinical disease was much less common than infection. We concluded that leptospirosis was of minor economic importance in dairy cattle, although it could be significant in individual herds, and a health hazard to farm workers.  相似文献

Summary

A comparison was made between serological and parasitological techniques for the diagnosis of bovine trypanosomiasis in Zambia. Overall sero‐prevalence rates as determined by IFAT and ELISA were respectively 2.7‐fold and 2.9‐fold greater then the percentage of samples found positive with the dark ground/phase contrast buffy coat technique (DG). The results obtained by the two serological techniques were found to be closely correlated (94.2% agreement) and titres obtained by ELISA tended to be slightly higher than those obtained by IFAT. Linear regression analysis of the results obtained by the IFAT and DG techniques revealed a highly significant correlation. This finding would permit the use of only one of the techniques in an epidemiological survey and to extrapolate the results from the regression line.  相似文献

ABSTRACT

Aims: To compare detection by real-time PCR of DNA from Mycoplasma bovis on mucosal swabs taken from the palatine tonsillar crypt and the mainstem bronchi of clinically asymptomatic calves after slaughter.

Methods: We compared the sensitivity of mucosal swabs taken from two sites: the palatine tonsillar crypt and the mainstem bronchi. Paired samples were taken post-mortem at slaughter from 55 clinically well calves from an infected herd and were tested by real-time PCR for the presence of M. bovis-specific DNA.

Results: Mycoplasma bovis DNA was detected in 51 palatine tonsillar crypt swabs (92.7 (95% CI?=?82.4–98.0)%) and seven mainstem bronchial swabs (12.7 (95% CI?=?5.3–24.5)%). All seven calves with positive mainstem bronchial swabs also had positive palatine tonsillar crypt swabs.

Conclusions: When compared to mucosal swabs of the mainstem bronchi, mucosal swabs of the palatine tonsillar crypt were seven times more sensitive for the post-mortem detection of M. bovis DNA. The viability of detected M. bovis was not assessed, because any cattle carrying viable or non-viable M. bovis DNA were determined to be a potential risk to eradication. Palatine tonsillar crypt mucosa may be a useful anatomical site for real-time PCR detection of M. bovis DNA in naturally infected calves. More work is needed to define the persistence and viability of M. bovis at this anatomical site.

Clinical relevance: The results of this study helped form the basis of surveillance tools used in M. bovis control and eradication efforts. Familiarity with these results may help veterinarians better communicate with their clients about the science behind the eradication efforts.  相似文献

ABSTRACT

Aims: The main goal of the current study was to evaluate, on a commercial beef cattle farm, the impact of infection with gastrointestinal nematodes resistant to both ivermectin (IVM) and moxidectin (MXD) on the productivity of calves.

Methods: Male Aberdeen Angus calves, aged 9–11 months, with faecal nematode egg counts (FEC) ≥200 epg and body weight ≥190?kg, were allocated to two herds. Herd A (n?=?90) grazed a maize-winter forage crop rotation and Herd B (n?=?90) grazed a 2-year-old Agropyrum pasture. On Day 0 in each herd, calves were randomly allocated into five groups (n?=?18), which were treated with 0.2?mg/kg IVM; 0.2?mg/kg MXD; 3.75?mg/kg ricobendazole (RBZ), both IVM and RBZ, or remained untreated. Faecal samples collected on Days ?1 and 19 were used to determine the percentage reduction in FEC, and genera of the nematodes were determined by the identification of the third-stage larvae recovered from faecal cultures. Total weight gain was determined from body weights recorded on Days ?1 and 91.

Results: Overall mean reduction in FEC was 42% for IVM, 67% for MXD, 97% for RBZ and 99% for IVM?+?RBZ. The reduction in FEC for Cooperia spp. was ≤78% for IVM and MXD, and for Haemonchus spp. was 0 and 36% for IVM and MXD, respectively, confirming the presence of parasites resistant to both anthelmintics. Only IVM?+?RBZ treatment resulted in 100% efficacy against Haemonchus spp. The overall estimated mean total weight gain for calves treated with IVM was 15.7 (95% CI?=?11.9–19.7) kg and for calves treated with IVM?+?RBZ was 28.8 (95% CI?=?25–32.5) kg (p?<?0.001). Mean total weight gain for calves treated with MXD was 23.5 (95% CI?=?19.7–27.2) kg.

Conclusions and clinical relevance: In calves naturally infected with resistant nematodes, under the production system assessed here, weight gains were lower in calves treated with anthelmintics that were moderately or highly ineffective compared to those treated with highly effective anthelmintics. These results demonstrate to farmers and veterinarians the importance of a sustainable and effective nematode control under field conditions.  相似文献

Abstract

The performance of dairy calves allowed to suckle freely (FS) until eight weeks of age in a cubicle system with automatic milking was compared to automatic teat-feeding with low milk (LM) or high milk (HM) substitute allowances. Weight gain and feed intake of FS (n = 13), LM (n = 23) and HM (n = 22) calves were recorded until two weeks after abrupt weaning, and their behaviour was studied during weaning. FS calves had higher weight gain, but much lower solid feed intake until weaning, than LM and HM calves and their advantage in weight remained throughout the study despite very low post-weaning gain. During the first 24 hours after weaning, FS calves displayed more behavioural signs of stress. However, at 72 hours after weaning, behaviour was similar for all treatments. Methods to stimulate solid feed intake before weaning need to be developed to draw advantage of the FS system.  相似文献

Sir,—In his criticism of our recent paper in which we claimed to have demonstrated experimental leptospirosis in 19 of the 20 control calves, W. M. Webster claims that the evidence for infection in five of the control calves is unconvincing (“Standards in Scientific Contributions”, N.Z. vel. J., 6: 27). His main reasons for this opinion appear to be, first, because no significant agglutination-lysis titres were demonstrated in these calves and, secondly, the observed onset of leptospimria in three of them was later than nsnal. He further suggests that mistaken diagnosis of leptospirosis, due either to dark-ground examination of urine or to inadvertent infection in guinea pigs inoculated with calf kidney tissue, is a more probable explanation.  相似文献

Background: Natural autoantibodies (N(a)ab) were found in every species tested so far, and are likely important in maintaining homeostasis.

Objectives: (1) To determine N(a)ab in Bos taurus calves, (2) evaluate effects of diet and age on N(a)ab binding repertoires in calves, and (3) delineate bovine liver cell lysate (BLL) antigens related with variation in rumen score and body weight.

Animals and methods: Effects of age and diet on staining of BLL fragments by IgM and IgG antibodies in serum samples collected at 20 or at 26 weeks of age from bull calves either fed a restricted or ad libitum diet were analyzed using quantitative Western blotting. Correlations between fragments stained and grouping of calves were done by Principal Component Analysis (PCA). Redundancy analysis (RDA) was done to relate rumen score and body weight variation at slaughter at 27 weeks of age with stained BLL fragments.

Results: In sera from all calves IgM and IgG antibodies binding BLL antigens were found. Corresponding fragments were stained, but quantitative differences in staining intensities were related to diet and age for both IgM and IgG. PCA revealed that age had a greater influence than diet on BLL fragment staining. RDA suggested that staining by IgM or IgG of specific BLL fragments was related with variation in rumen score and body weight.

Conclusions and clinical importance: Analyses of N(a)ab in serum could be a potential tool to estimate the health status of cattle, and be used to evaluate effects of husbandry practices.  相似文献
