Thirty-two quarters, five of which harbored subclinical mastitis, were examined daily for one month. The usefulness of milk antitrypsin, BSA (bovine serum albumin), NAGase, somatic cells and bacteriological analysis in differentiating the inflamed quarters from the healthy control quarters was analysed. Inter-quarter evaluation clearly improved each indirect mastitis parameter; NAGase and antitrypsin were better indicators of differences between infected and non-infected quarters than BSA or the somatic cell count.  相似文献

Microplate analyzers for N-acetyl-beta-D-glucosaminidase (NAGase) and antitrypsin in milk were high-capacity systems for determination of subclinical mastitis on a mammary-quarter basis in a herd of 80 Ayshire cows. Infected quarters could be better differentiated from non-infected quarters by interquarter evaluation of milk NAGase and antitrypsin, than by adaptation of threshold values based on measured NAGase and antitrypsin values. This differentiation between healthy quarters and mastitic quarters was further improved by using absolute values and interquarter evaluations in combination. Therefore, quarter-based sample collection in combination with microplate technology and computerization is proposed as the method of choice for monitoring udder health in herds.  相似文献

The activities of plasmin, N-acetyl-β-d-glucosaminidase (NAGase) and antitrypsin in milk were determined during 1 week post partum. The concentration of NAGase and antitrypsin in milk decreased significantly during this period. A slight decrease in plasmin activity was also seen. Replication rates of E. coli and S. aureus in lactoserum were also determined. Both test bacteria showed a tendency for increasing growth rates towards the end of the period. Growth of E. coli was significantly (p < 0.01) enhanced in day 2 samples as compared with samples collected during the first day post partum.  相似文献

The cell wall of Staphylococcus aureus (strains 21 and Glaxo) was treated with deoxycholate and the insoluble residue was solubilised with lysozyme. The effect of the extract in modulating the inflammatory response due to infection of the lactating bovine udder was evaluated. Cows were infected with S. aureus strain 21 or Streptococcus agalactiae, with or without the cell wall extracts. The clinical response to infection was assessed, and milk samples collected up to 30 h were assayed for antitrypsin and NAGase levels, somatic cell count, and for the ability of whey to support bacterial growth. The extracts markedly reduced the inflammatory response elicited by both S. aureus and S. agalactiae, indicating the effect was non-specific. The extract from strain 21 was generally more effective than that from strain Glaxo.  相似文献

The objectives of this study were to determine factors affecting the reproductive performance of primiparous sows early weaned (EW; n = 35) at d 14 or conventionally weaned (CW; n = 35) at d 24 of lactation. Sow BW and backfat were recorded at farrowing, weekly until weaning, and at standing heat. Feed intake was controlled throughout lactation to standardize nutritional effects on subsequent reproductive performance. Litter size was standardized across treatments within 48 h after farrowing, and litter weight was recorded until weaning. In subsets of sows, blood samples were collected from 10 h before to 10 h after weaning, and then every 6 h until ovulation. Sows were heat checked twice daily and bred at 24-h intervals during standing heat using pooled semen. Ultrasonography every 6 h determined time of ovulation. Sows were either slaughtered within 24 h after ovulation to assess ovulation rate, fertilization rate, and embryonic development in vitro, or at d 28 of gestation to determine ovulation rate and embryonic survival. Compared with CW sows, EW sows had more backfat at weaning (15.9 +/- 0.5 vs. 14.7 +/- 0.5 mm; P < 0.001). Also, CW sows tended to lose more BW and to have lower IGF-I concentrations, indicating poorer body condition. Duration of lactation did not affect ovulation rate (EW = 17.6 +/- 0.7; CW = 18.7 +/- 0.6), fertilization rate (EW = 96.0 +/- 2.2; CW = 88.2 +/- 4.7%), or embryo survival to d 28 (EW = 62.5 +/- 4.5; CW = 63.1 +/- 5.0%). There was a marginal effect of duration of lactation on weaning-to-estrus interval (EW = 120 +/- 3; CW = 112 +/- 3 h; P < 0.06) and duration of estrus (EW = 52.4 +/- 2.3; CW = 46.3 +/- 2.2 h; P < 0.08). Overall, embryonic survival, not ovulation rate, seems to be the limiting factor for potential litter size in the second parity. Although fertility in both EW and CW sows studied was compromised, endocrine and metabolic data indicate that the mechanisms affecting reproductive performance may differ between the two weaning systems. The LH, FSH, and estradiol data from the EW sows are characteristic of animals with limited follicular development and incomplete recovery of the hypothalamic-pituitary-ovarian axis; consequently, the integrity of the uterine environment may be adversely affected and limit embryonic survival. In CW sows, variability in metabolic state seemed to be the key factor limiting the fertility, again adversely affecting embryonic survival.  相似文献

Three major antiprotease components in equine serum were identified and characterised. These were the acidic prealbumin Pr, the homologue of human alpha-1 antitrypsin and 2 protease binding proteins, the acidic prealbumin Xc and alpha-2 macroglobulin, both capable of inhibiting the proteolytic activity of trypsin, but with only limited inhibitory effect on its esterolytic activity. The possible role of these serum antiproteases in the onset of chronic obstructive pulmonary disease (COPD), analogous to the hereditary dysproteinaemia of alpha-1 antitrypsin in man, was investigated. There was no evidence of a genetically determined variation in the protease binding proteins but an increased frequency of the electrophoretically slower Pr antitrypsin alleles was present in horses affected with COPD. However, because of both the mixed breeding of the animals investigated and the lack of correlation with low serum trypsin inhibitory capacity, measured by inhibition of DL-BAPA hydrolysis, the significance of this observation could not be critically assessed.  相似文献

Twenty-one multiparous dairy cows were fed concentrates containing three levels (119, 154 and 191 g/kg DM) of crude protein (CP) during the first ten weeks of lactation. Part of the grain and molassed sugar beat pulp was substituted with 0% (RSM0), 15% (RSM15) or 30% (RSM30) repeseed meal. Wilted grass silage was fed ad libitum after calving. The average response between RSM0 and RSM15 was +1.66 kg milk/d per percentage unit change in concentrate CP content. No further response occurred between RSM15 and RSM30. The positive effect of RSM inclusion was seen throughout the experimental period and was associated with increased plasma non esterified fatty acids (NEFA) and decreased plasma insulin concentration one week after calving, and higher efficiency of metabolisable energy utilisation for milk production. Digestibility of the diet remained unaffected. Milk and plasma urea tended to increase with RSM30 indicating excessive supply of rumen degradable protein. Because of the limited potential of cows to compensate for a deficit in feed protein supply by mobilising tissue protein, a substantial milk yield response can be achieved with a moderate level of protein supplementation during early lactation.  相似文献

Several studies have shown that foot lesions and clinical lameness occur before first calving and develop further during the lactation period. Lameness may cause production losses, but the relationship between foot lesions, particularly in the claw horn, and lameness in heifers is unclear. The objectives of this study were to describe the development of and evaluate the relationship between lameness and foot lesions in Danish Holsteins before and after first calving. In a longitudinal study, 147 heifers were examined for lameness and foot lesions 2–5 times over an 18-month period. Lameness was assessed by means of a visual locomotion score and foot lesion severity was recorded.The prevalence of a locomotion score 3 was 25% before calving, and 90% at approximately 250 days in milk (DIM). Prevalence of moderate to severe sole haemorrhage (SH) was 27% before calving and 56% at 250 DIM, and that of moderate to severe white line lesion (WLL) 44% before calving with a peak of 70% at 200 DIM. There was one case of white line abscess but SH was seen throughout the entire study period. Digital dermatitis (DD) was prevalent prior to first calving (15%) and peaked at 39% at 0–100 DIM. Heel horn erosion (HHE) occurred in almost all cows (93–100%) and was strongly correlated with DD (r = 0.51). The correlation coefficient between SH and WLL was also high (0.42). The relatively high correlations between WLL and both DD and HHE were more surprising (0.38 and 0.35, respectively), those between SH and both DD and HHE were moderate (around 0.18). Interdigital dermatitis was significantly correlated with both HHE and DD, but completely unrelated to SH and WLL.The overall average locomotion score increased by about one-half of a score unit from 1 month prior to calving until 250 DIM, with a large difference between herds, although this was unsurprising as cows may alter their locomotion pattern with management factors (e.g. floor properties). DD and WLL were both associated with a locomotion score 3 but of the cows with severe WLL there was no clear association between a locomotion score 3 and DD. The highest locomotion scores occurred among cows with DD but without WLL.  相似文献

The objectives of the current study were to determine how intermittent suckling (IS) affects nursing behavior, litter activity, and general behavioral patterns during lactation, and whether IS during an extended lactation period results in behavioral patterns associated with piglet distress. Intermittent suckling was applied either with 6-h separation intervals (IS6) or with 12-h separation intervals (IS12) and was compared with the conventional treatment (CT). In the CT (n = 17 litters), sows were continuously present until weaning (d 21, d 0 = farrowing). In both IS6 and IS12, sows were separated from their litter for 12 h/d, beginning at d 14 and lasting until weaning (d 43 +/- 1 d). In IS6, litters (n = 14) and sows were separated from 0800 to 1400 and from 2000 to 0200; in IS12 litters (n = 14) and sows were separated between 0800 and 2000. In IS litters, the activity pattern over the 24-h cycle was markedly changed by IS; litter activity was lower (P < 0.001) during sow absence and greater (P < 0.001) during sow presence compared with the unweaned CT litters. Moreover, both total nursing frequency (P < 0.001) and the percentage (P < 0.002) of successful nursings were reduced by IS. Although total nursing frequency was greater in IS6 compared with IS12 (on d 21 and 28), no differences in the frequency of successful nursings existed between IS6 and IS12 from d 14 onward. Eating behavior was increased shortly after the onset of IS (d 17) in both IS6 (P = 0.059) and IS12 (P < 0.001) compared with the unweaned CT litters. The IS12 litters showed more eating behavior compared with IS6 and their exploratory behavior increased in time (P < 0.001), whereas IS6 showed more nursing behavior. Aggressive or manipulative behavior of both IS treatments was similar compared with the unweaned CT, and remained relatively unaltered with time in IS12 and IS6. Weaning in the CT resulted in more manipulative (P < 0.001) and aggressive (P = 0.004) behavior compared with pre-weaning values. Intermittent suckling may contribute to adaptation to the postweaning state by stimulating eating behavior, without causing obvious behavioral distress.  相似文献

The glucose tolerance test in the horse may be used to determine metabolic responses to diet, disease, or physiologic state. The objective of this study was to determine the effect of reproductive stage (gestation and lactation) and supplemental dietary energy source (sugar and starch [SS] or fiber and fat [FF]) on glucose metabolism in grazing mares using an oral glucose tolerance test. Twelve mares, six on each supplement, were examined on three occasions: one in the third trimester of pregnancy, the second in early lactation, and the third in late lactation. During each test, venous samples were taken at 30 and 1 min before, and 30, 60, 90, 120, 150, 180, 240, and 300 min after a nasogastric dose of glucose at 0.2 g/kg of BW. Plasma was assayed for glucose, insulin, and cortisol. Statistical analysis was a mixed model with repeated measures with horse, diet, and reproductive stage as fixed effects. The incremental glucose area under the curve (AUC) in response to oral glucose was lower in SS than in FF mares (P = 0.022). Mares tended to have a lower incremental glucose AUC in early lactation than in late gestation (P = 0.057), and insulin AUC was lower in early lactation than in late gestation (P = 0.002) and late lactation (P = 0.013). Glucose clearance was more rapid (P = 0.007) in SS than in FF mares. The glycemic response to the oral glucose tolerance test was consistent with adaptation to dietary sugar and starch as well as metabolic changes associated with pregnancy and lactation. Feeding twice-daily grain meals rich in SS influenced glucose metabolism in horses to an extent that the natural adaptation of glucose metabolism to pregnancy was moderated. Feeding a diet rich in FF more closely mimics the natural grazing state of pasture and allows for adaptation of glucose metabolism to pregnancy and lactation.  相似文献

First lactation milk production of Japanese Holstein cows was partitioned into ten stages. A quartic Legendre polynomial (k = 5) under an animal model was used to estimate the additive genetic covariance matrix between these ten stages. Various selection indexes with or without restrictions were constructed using the eigenvectors of the genetic covariance matrix. These eigen indexes were designed to improve lactation milk and persistency defined as difference in milk between DIM 280 and DIM 55. Two sets of economic weights between lactation milk and persistency and three levels of restriction on the intended gains in persistency were applied. The unrestricted index based on the first three eigenvectors achieved more than 99.9% of the net merit of the unrestricted index based on the first five eigenvectors. The advantage of unrestricted index over conventional selection based on lactation EBV increases with increasing economic weight assigned to persistency. The construction of the restricted eigen index does not require economic values between lactation milk and persistency and thus is more practical than the unrestricted eigen index which requires information on the economic values of both. The index coefficients of the eigen indexes reveal the relative selection emphasis among various stages of the lactation. When the degree of restriction or economic value for persistency increases, a greater selection emphasis was placed on the later part of the lactation than on the early part in order to realize the desired persistency. The developed eigen indexes provide useful information and easy understanding of the selection practice. A numerical example was given to illustrate the construction of the restricted eigen index.  相似文献

We conducted an experiment in dairy cows investigating the effects of calving season, milking frequency and nutrition on lactation persistency. Cows calved in the Spring (n=12) or Winter (n=12). Commencing in lactation week 9 one udder-half of each cow was milked thrice-daily and half of each calving group received additional concentrate at a fixed rate of 3kg per day above that of the control cows. As reported elsewhere, between lactation weeks 9 and 33 persistency (measured as the slope of decline in milk yield) was significantly improved by frequent milking (P<0.001), by calving in the Winter (P<0.001) and by additional concentrate (P<0.05). The cows were rebred after week 33. When analysis of persistency was extended up to week 20 of the recurring pregnancy only the frequency effect remained significant. Persistency was unaffected by the pregnancy up until pregnancy week 20 but was then greatly reduced (P<0.001). In this paper we report hormone concentrations. GH was unaffected by nutrition but was consistently elevated in the Winter calving group relative to the Spring. IGF1 and prolactin were both unaffected by nutrition and calving season, IGF1 tended to increase as lactation progressed but changes in prolactin were related to time of year more than stage of lactation. Insulin was not affected by nutrition and was lower in Winter calvers, but only during early lactation. Prior to rebreeding, lactation persistency was correlated (slightly) with [GH] but not with [IGF1] or [insulin] and was correlated significantly with changes in GH, IGF1 (both positive) and insulin (negative). In conclusion, whilst bovine lactation persistency is plastic and amenable to beneficial manipulation, the details of its endocrine control remain to be elucidated.  相似文献

Major changes in maternal physiology during pregnancy and lactation can have a large impact on the immune and neuroendocrine systems. One of the most significant changes, observed in rats and mice, is hyporesponsiveness of the hypothalamic pituitary adrenal axis (HPAA) in response to inflammation, restraint, and other psychological stressors during late pregnancy and lactation. This attenuation, however, has not been well characterized in ruminant animals and may be relevant to their susceptibility to inflammatory diseases during these periods. Thus, the intent of this study was to characterize responsiveness of the ovine HPAA to inflammatory challenge during pregnancy and lactation. Ewes from early (33 d), middle (55 d), and late (138 d) pregnancy, as well as early lactation (10 d), were challenged i.v. with a bolus dose of 400 ng of Escherichia coli lipopolysaccharide (LPS)/kg of BW or saline. A corresponding group of nonpregnant ewes was also challenged with LPS to serve as positive control animals for each pregnancy and lactation study. Responsiveness of the HPAA was assessed by measuring the 4-h change in serum cortisol concentration after LPS challenge. The cortisol increase after LPS challenge was elevated (P < 0.01) in pregnant ewes during late pregnancy over that of nonpregnant animals. In contrast, the characteristic temperature response associated with systemic LPS challenge was decreased (P < 0.01) during early pregnancy and lactation compared with nonpregnant or nonlactating animals. Serum IL-6 concentrations were measured to assess whether changes in HPAA responsiveness during pregnancy or lactation were attributed to changes in proinflammatory signaling to the HPAA. Interestingly, enhanced cortisol responsiveness during late pregnancy was correlated with increased (P < 0.01) serum IL-6 concentrations, indicating that IL-6 may contribute to enhanced HPAA responsiveness during this period. Serum IL-6 concentrations during early and midpregnancy did not increase in response to LPS challenge, indicating that HPAA activation during periods of pregnancy may be independent of IL-6 production.  相似文献

Colostral trypsin-inhibitor capacity was monitored during the first two weeks from parturition. The colostrum of the mare, sow, cow and ewe showed high antitrypsin activity at parturition, decreasing to about one hundredth during the first week. Canine milk remained on a relatively high antitrypsin level, and human milk was poor in antitrypsin from childbirth. The antitrypsin content seems to parallel the known changes in the colostral immunoglobulin levels of different species. The role of antitrypsin in protection of immunoglobulins from proteolytic damage during passive transfer of immunity to the newborn is obvious.  相似文献

Two varieties of avocado leaves (Persea americana var Guatemalan and var Mexican) were administered to lactating goats. The Mexican variety was without effect. The Guatemalan variety in doses exceeding 20 g fresh leaf per kg bodyweight, produced damage to the mammary gland with decreased milk production. The lesions were characterised by oedema and reddening, with clots in the large ducts. Microscopically, there was widespread degeneration and necrosis of the secretory epithelium, the necrotic cells sloughing into the lumen. There was no significant cellular inflammatory response. Concentrations of antitrypsin in the milk, indicating changes in vascular permeability, increased rapidly 15 h after a single high dose, coinciding with palpable oedema. Concentrations of NAGase, indicating cell damage, increased after 24 h. Goats given multiple doses followed a similar pattern but the initial response was delayed. The toxic principle, and its mode of action in selectively damaging mammary secretory cells, remains to be determined.  相似文献

Metabolic and production responses are reported for 72 cows treated with bovine somatotropin (BST) for 30 days starting at day 70 of lactation. Of these 72 cows, 48 had been exposed in the preceding lactation to long-term treatment with BST at 3 dosages and 24 (controls) had not been given BST. Approximately half of the cows in each group were parity-2 cows, the rest were older. Comparisons between groups were made separately for parity-2, and older cows. Analyses, using pretreatment values of each variable as a covariate, indicated that older cows, but not parity-2 cows, significantly (P less than 0.05) increased milk production during treatment. Parity-2 cows, however, had a significantly higher milk fat percentage than controls following treatment. Cows treated with 51.6 or 86 mg BST/d in both parity groups had significantly higher serum-free fatty acids than controls. Estimated net energy balances were significantly lower for older treated cows, but did not significantly differ from controls for parity-2 treated cows. Older cows in the 86 mg of BST/d group tended to have higher concentrations of blood glucose than did older control-group cows. Treatment with BST did not significantly increase serum ketone concentrations in any group of animals, and none of the cows developed clinical ketosis during this period. Estimated net energy balance (ENEB) during treatment was a significant (P less than 0.05) covariate for free fatty acid concentrations in older cows and for milk fat percentage in parity-2 cows. Covariate adjusted analyses, using ENEB during treatment as a covariate, indicated that lipolytic stimuli already acting may be enhanced by treatment with BST, but a negative energy balance was not a necessary precondition for free fatty acid concentrations to increase following somatotropin treatment. Similarly, milk fat percentages for parity-2 treated cows were significantly (P less than 0.05) higher during treatment than controls when ENEB during treatment was used as a covariate. Increased milk fat concentrations in parity-2 treated cows were not associated with significant increases in the ratio of C18:C4-10 milk fatty acids, indicating that increased milk fat resulted from either an increase in incorporation of C18 fatty acids into milk fat coupled with an increase in de novo mammary synthesis of C4-10 milk fatty acids or an increase in C12-16 fatty acids that may arise either from increased tissue mobilization, from diet, or from de novo mammary synthesis.  相似文献

AIM: To determine the prevalence of clinical mastitis in spring-calving dairy herds in the Waikato Region of New Zealand and to identify factors associated with variation in the prevalence of clinical mastitis between herds. METHOD: A total of 799 quarters from 595 dairy cows from 38 dairy herds were diagnosed by herd owners as having clinical mastitis between 8 July and 21 August 1997. Quarters diagnosed with clinical mastitis were sampled for bacterial culture and somatic cell count, and the presence of clots in the milk and the presence of udder oedema were assessed by a technician or veterinarian. RESULTS: Clinical mastitis was diagnosed in an average (+/-s.e.m.) of 9.9% (+/-0.8%, range 0.9-21.4%) of calved cows within the herds. Bacteria were not cultured from an average of 12.4 % (+/- 2.0%, range 0.0-45.5%) of cows and 22.3% (+/- 2.4%, range 0.0-54.0%) of quarters diagnosed as having clinical mastitis. There were significant differences between herds in the proportion of cows diagnosed with mastitis and in the proportion of clinical mastitis cases from which bacteria were not cultured. A decreased prevalence of clinical mastitis (p<0.001) was associated with an increased percentage of the herd treated with dry cow antibiotics. An increased prevalence of clinical mastitis (p<0.0001) was associated with both an increased percentage of cows treated in the previous season with lactating cow antibiotics and an increased percentage of heifers in the herd. Herds that were fed supplements before or during lactation had a higher prevalence of clinical mastitis than herds that were not fed supplements (p<0.001). An increased proportion of quarters diagnosed with clinical mastitis that did not culture bacteria was associated with an increased prevalence of clinical mastitis (p<0.001). The proportion of quarters that the technician or veterinarian found with evidence of clinical mastitis (i.e. a somatic cell count >500,000 cells/ml and the presence of either clots or udder oedema) within a herd was inversely related to the proportion of quarters within a herd from which no bacteria were isolated. CONCLUSION: There was a large variation in the prevalence of clinical mastitis and in the proportion of clinical quarters from which no bacteria were grown between herds. Management factors such as the use of dry cow therapy, feeding regimes and heifer replacement rates all affected the prevalence of clinical mastitis. Herd owners appear to differ in the sensitivity and specificity of their diagnosis of clinical mastitis, with bacteria not isolated from up to 50% of quarters diagnosed with clinical mastitis in some herds. Improvements in the specificity of herd owner diagnosis of clinical mastitis may reduce the use of antibiotics for mastitis during lactation and hence may reduce the risk of antibiotic contamination of milk supplied for human consumption.  相似文献

Growth of Escherichia coli and Staphylococcus aureus was analyzed by turbidometry in 456 selected whey samples. The samples had been graded mastitic or nonmastitic as determined by antitrypsin assay and bacteriologic examination. Whey samples from inflamed quarters (antitrypsin increased) and infected quarters significantly promoted bacterial growth as compared with whey samples from control quarters. The growth-stimulatory effect was limited to the diseased quarters, since control quarters from the same animals did not support enhanced bacterial growth. The observations indicate that in chronic mastitis, the inflammatory process leads to changes in the whey that promote bacterial growth in vitro.  相似文献
