Dry mycelium (DM) ofPenicillium chrysogenum and its water extract (DME) were examined for their effects on induced resistance against Verticillium wilt and plant growth of cotton in the greenhouse. Soil application of 0.1–5% DM or 0.5–5% DME provided significant protection against the wilt, relative to the control. As neither DM nor DME inhibited mycelial growth ofVerticillium dahliae in vitro, it is suggested that the disease-controlling effects of DM or DME are attributed to induced resistance. DME (5%), as well as DME treated with chloroform or cold acetone, were as effective as 2% DM in reducing disease severity of Verticillium wilt, implying that the resistance-inducing substance(s) in DM are mostly water-soluble, with neither proteins nor lipids likely to be responsible for the induction of resistance. No significant difference in root colonization withV. dahliae was found between control-inoculated and 2% DM- or 5% DME-inoculated plants. However, colonization of hypocotyls and epicotyls was drastically suppressed by either 2% DM or 5% DME relative to the control. Treatments with 2% DM or 5% DME significantly increased ionically-bound peroxidase (POX) activity in roots, hypocotyls and the second leaf of cotton plants, with the hypocotyls expressing the highest increase. Soil application of DM or DME increased plant height, fresh and dry weight of inoculated and non-inoculated cotton plants, relative to their corresponding controls. It is concluded that DM may be used in cotton crops to promote plant growth and to induce resistance againstV. dahliae. Dry fungal biomass ofPenicillium chrysogenum (dry mycelium), a waste product of the pharmaceutical industry, was extracted with water and applied to the roots of melon plants before or after inoculation withFusarium oxysporum f.sp.melonis (Font). Seedlings (4–6 days after emergence) treated with either acidic dry mycelium extract (DME) or neutralized dry mycelium extract (NDME) were protected against challenge infection withFom. A single drench with 2–5% DME applied 12–72 h before inoculation provided significant control of the disease compared with water-drenched, challenged seedlings. No protection was seen in plants treated 0–6 h before inoculation or 0–48 h after inoculation. Neither DME nor NDME (0.5–5%) had any effect on fungal growthin vitro, which implied that disease controlin vivo was mediated by induced resistance. The resistance induced by DME protected melon plants not only against race 1,2, but also against the three other races of the pathogen, indicating a race-non-specific resistance againstFom. Both DME and NDME significantly increased peroxidase activity and free L-proline content in seedlings 12 h and 48 h after soil drench, respectively. Resistance to Fusarium wilt was significantly associated with elevated levels of peroxidase activity but not with free L-proline content. Temporal variation in Fusarium oxysporum f. sp. vasinfectum (Fov) populations was determined by comparing the genetic diversity of pathogen isolates recovered from three consecutive cotton crops (2002, 2004 and 2006) in the Boggabilla area of New South Wales, Australia. A total of 288 isolates were collected, among which 25 distinct AFLP genotypes were identified. These genotypes were classified into two main groups corresponding to known vegetative compatibility groups (VCG)—01111 and 01112. The Fov populations were dominated by four genotypes (I-A, I-B, II-A, II-B) that accounted for 87.5% of the isolates. Significant temporal variation was observed in both sampled fields with 6.8% and 10.7% of total genetic variation being attributed to differences among collections in different years. Genetic diversity based on Nei’s gene diversity and the Shannon-Wiener index increased over time. Significant changes in the frequency of the dominant Fov genotypes were observed in one field, where genotype I-A declined from 84.8% to 40.0% over the study period (2002–2006), while genotype I-B increased from 7.6% to 35.4%. Strong inter-genotype competition was detected in glasshouse bioassays with 93.4% of symptomatic plants sampled from dual inoculation trials being infected by single genotypes. Competition was differentially mediated by cotton cultivars as the competitive ability of pathogen genotype I-B was enhanced on the resistant cultivar Sicot 189 relative to the susceptible cultivar Siokra 1–4. In the course of pre- and postharvest epidemiological studies on bulbs contamination byAspergillus niger, two Sudanese onion cultivars were tested: ‘Saggai Red’ and ‘El-Hilo White’.A. niger spores, whether seedborne, soilborne or airborne, were avirulent to the healthy growing onion plants. The fungus heavily contaminated the dead onion tissues, mainly the dead leaves followed by the dry scales, the dead roots and, to a lesser extent, the bulb necks, preferring the red-skinned cultivar to the white one. The initial spores carried from naturally contaminated field soil on the dead tissues could germinate and produce massive numbers of new spores on bulbs stored at average climatic conditions of Sudan (23–39°C, 29–93% relative humidity). Under laboratory-controlled conditions, optimal growth occurred at 75–85% r.h. on bulbs with dry scales and maximum losses occurred at 100% r.h. and ambient temperature. Differentin vivo resistance/susceptibility levels of 14 carnation cultivars toFusarium oxysporum f.sp.dianthi race 2, the causal agent of Fusarium wilt disease of carnation, were also expressed in anin vitro system and assayed as the degree of fungal colonization of callus cultures at 20° C. Temperature influenced thein vitro expression of carnation resistance. An incubation temperature of 27° C increased the colonization of calli derived from both the susceptible (‘Corrida’ and ‘Ambra’) and the resistant (‘Pulcino’ and ‘Pallas’) cultivars. At 15°C, the colonization of calli derived from Pulcino and Pallas diminished significantly more than for Ambra and Corrida. Inhibition of fungal growth on resistant calli was correlated to retardation in hyphal development. Both scanning electron microscopy and light microscopy observations showed that hyphae did not penetrate into carnation cells.  相似文献

Several new isolates ofBacillus thuringiensis Berliner have shown significantly higher levels of insecticidal activity than the previous best isolates (Bt24-entomocidus and IH-A-aizawai) against bothSpodoptera littoralis Boisd. andSpodoptera exempta Walker. The best ones, in the following descending order of potency, were: againstS. littoralis — K26–21, MF4B–2, MR1–37, K28–30, K26–8; againstS. exempta — Bt-3, K30–3, HD-133, MR1–37, Bt-17, K26–8, K26–21, MF4B-2, HD-593. In a study of date fruit damage caused byOligonychus spp., we investigated whether the cultivar affects phenology, and on what hosts the mites over-winter. Samples were taken from ‘Deglet Noor’, ‘Barhi’ and ‘Medjool’ trees from mid-April through mid-September during the years 1999–2002. In the ground-cover mites were monitored by collecting Bermuda grass (Cynodon dactylon) under each sampling tree. Over 99% of the mites collected on Deglet Noor and Barhi fruit were identified asO. afrasiaticus. Mean population levels ofO. afrasiaticus reached ten mites or more (initiation of infestation) on Medjool in the second half of May, whereas on Deglet Noor this did not occur before the first week of July. On Barhi the initiation of infestation varied between plots and years, ranging from the second half of May to the beginning of July, but always occurred earlier than Deglet Noor. Mite populations on the pinnae remained low from June through October, not exceeding seven mites per pinna, whereas on fruit strands they reached peak populations of approximately 4000 mites per strand. The sex ratio (proportion of females) ofO. afrasiaticus on fruit of all three cultivars was highly female-biased, usually above 0.85. Thirty-four isolates ofFusarium oxysporum f.sp.melonis (F.o.m.) obtained from 205 fields in melon-producing areas in the southeastern Anatolia Region of Turkey were identified on the basis of colony morphology and pathogenicity by the root dip method. In this region the mean prevalence of wilt disease was 88.1% and the mean incidence of disease was 47.5%. Physiologic races 0, 1, 2, and 1,2 of the pathogen were determined by their reactions on differential melon cultivars ‘Charentais T,’ ‘Isoblon’, ‘Isovac’ and ‘Margot’ in the greenhouse. Race 1,2, representating 58.8% (20/34) of all isolates, was widely distributed. Of the other pathogenic isolates, eight were identified as race 0, five as race 1, and one as race 2. This is the first report of physiologic races ofF.o.m. in Turkey. Field experiments were conducted during 1997 and 1998 to determine the effects of sugar beet cultivar susceptibility to Cercospora leaf-spot on the sensitivity ofCercospora beticola isolates to the triazole fungicide flutriafol. Four cultivars with different levels of disease resistance were treated in experimental plots with six spray applications of flutriafol. Disease assessments were carried out at 15-day intervals. Sensitivity to flutriafol was measured on isolates collected from the plots ∼15 days after the last flutriafol application. Measurements of disease severity and calculations of AUDPC (area under disease progress curve) values showed a distinct differentiation among cultivars, reflecting their level of disease resistance. Disease severity was significantly lower in cvs. ‘Bianca’ and ‘Areth’ than in ‘Univers’ and ‘Rizor’ both in the untreated and in the flutriafol-treated plots. Fungal isolates from flutriafol-treated plots were less sensitive to the fungicide than were isolates from untreated plots. However, no differences in isolate sensitivity were observed among the cultivars, as regards their level of disease resistance. Despite the fact that the use of resistant cultivars cannot eliminate selectively the resistant strains, it can eliminate both resistant and sensitive isolates. Forty-five onion (Allium cepa) cultivars were evaluated for disease resistance to the fungal pathogen Pyrenochaeta terrestris (pink root disease). Forty-three Iranian cultivars and two commercially exotic resistant cultivars (Texas Early Grano and Yellow Sweet Spanish) were grown in a glasshouse for two successive years (2003 and 2004) in Isfahan, Iran. Susceptibility of the cultivars was determined using a scale of 0–3. Six cultivars had significantly lower susceptibility of pink root compared to the exotic ones. High susceptibility tended to be associated with high mean scores (2–3), and the highly resistant cultivars had the lowest scores (0–1). In addition, the 45 examined cultivars were ranked from 1 to 45 according to their markedly differing reactions to P. terrestris, which differed markedly. Of 32 cultivars that were less resistant, 30 with an intermediate status or tolerance (1–2) differed considerably in their rank order. No pink root symptoms were seen in the early stages, but on blotter paper or wheat-straw agar some roots turned pink with no pycnidia formation, whereas on potato dextrose agar medium (PDA) with a few fresh healthy and sterile onion roots on the surface, mycelia and pycnidia were recovered. A rapid glasshouse‐based bioassay method to screen large numbers of cotton plants for responses to Fusarium oxysporum f. sp. vasinfectum (Fov) was developed. Different Fov inoculum concentrations and methods of inoculation were assessed using resistant and susceptible cotton cultivars. Cotton seeds were planted directly into Fov‐inoculated soil. Studies of seed germination, seedling establishment, seedling mortality and fusarium wilt symptoms (i.e. stunting, foliar symptoms and vascular browning) were performed to optimize the bioassay parameters. Growing seedlings in Fov‐inoculated soils at 5 × 104 or 1 × 105 CFU g?1 soil, in individual seedling tubes with 12 h at 28–30°C and 12 h at 15–18°C, gave consistent results when assessing Fov disease responses 6 weeks after inoculation. When fusarium wilt resistance ranks (FWRRs) and vascular browning index (VBI) means of 18 Australian and other cotton cultivars from the Fov glasshouse bioassay were compared against their fusarium field performance ranks (F‐ranks), assessed on adult plants for cotton cultivar release, Pearson’s correlation was highly significant for both comparisons. The level of congruence between field and glasshouse data indicated that this protocol should be an effective tool for large‐scale screening for Fov‐resistance responses in diverse germplasm and breeding populations and for advancing genetic research to develop molecular markers for Fov resistance in cotton.  相似文献

Phoma macdonaldii is one of the most important pathogens of sunflower (Heliantus annuus) in France. In order to determine the inheritance of resistance to the disease, five sunflower genotypes with wide genetic variability for resistance to two ‘collar’ and two ‘root’ Phoma isolates were crossed in a diallel programme. Four separate experiments were undertaken under controlled conditions. In each one, the response of parental genotypes and their F1 hybrids were evaluated with one of the four Phoma isolates. Analysis of variance was performed to determine the effects of genotype on disease severity score when inoculated with ‘collar’ or ‘root’ Phoma isolates and showed significant variability among parents and F1 hybrids for disease severity score. Diallel analysis showed that general combining ability (GCA) and specific combining ability (SCA) effects for resistance to ‘collar’ and ‘root’ Phoma isolates were highly significant for each of the four isolates indicating that both kinds of gene effects were important in controlling the resistance. The GCA/SCA ratios were more than one for three out of four isolates showing that additive genetic effects were more important than non-additive effects for resistance to three of the studied Phoma isolates. Hence, conventional breeding methods could be recommended to achieve genetic improvement to such ‘collar’ and ‘root’ Phoma isolates.  相似文献

Plants have developed mechanisms to resist secondary infection upon inoculation with a necrotizing pathogen, chemical treatment as well as treatment with some non-pathogenic microorganisms such as rhizosphere bacteria. This phenomenon has been variously described as induced systemic resistance (ISR) or systemic acquired resistance. In the present study, the chemical benzo(1,2,3)thiadiazole-7-carbothioic acid-S-methyl ester (BTH, acibenzolar-S-methyl), and the rhizobacteriaPseudomonas aeruginosa KMPCH andP. fluorescens WCS417 were tested for their ability to induce resistance toColletotrichum lindemuthianum in susceptible and moderately resistant bean plants (Phaseolus vulgaris L.). BTH induced local and systemic resistance when bean leaves were immersed in 10−3 to 10−7 M BTH 3 days before the challenge inoculation. At a high concentration (10−3 M), BTH induced resistance of the same order as resistance induced by the pathogenC. lindemuthianum, although at this high concentration BTH appeared to be phytotoxic. Soil and seed treatment with 1 mg kg−1 BTH protected beans against anthracnose. BTH-mediated induced resistance was effective in susceptible and moderately resistant plants.P. aeruginosa KMPCH induced resistance in bean againstC. lindemuthianum only in a moderately resistant interaction. Among 153 isolates ofRhizoctonia spp. obtained from 95 soil samples collected from different fields in the USA, 42 (27.5%) isolates were hypovirulent or non-pathogenic on cabbage (tested on tap water agar plus 250 μg/ml chloramphenicol plates). Of these, 14 (33.3% of the np-R) isolates protected >60% of the cabbage seedlings againstR. solani, and the best eight isolates protected 73–95% of the cucumber seedlings. The np-R isolates RU56-8 (AG-P) and RU89-1 [AG-B(o)] induced the highest resistance against hypocotyl challenge inoculation with virulentR. solani (38.3–85.7%), whereas most of the challenged control seedlings (85–100%) collapsed. Similarly, isolates RU56-8 and RU89-1 induced the highest resistance (22.2–87.5%) against hypocotyl challenge inoculation withPythium aphanidermatum, whereas most of the challenged control seedlings collapsed (90–100%). Isolates RU56-8 and RU89-1 significantly reduced the lesion numbers and area/leaf (to 8.9–42.0% of the control) caused by challenge inoculation of the first true leaves withPseudomonas syringae pv.lachrymans. No np-R isolate could be recovered from the upper hypocotyls or from the leaves, indicating that there was no contact between the inducer and the pathogen. A weed survey conducted in 2004 and 2005 in Aydin province of Turkey showed that Solanum nigrum, Xanthium strumarium, Amaranthus retroflexus, Portulaca oleracea, Sonchus oleraceus and Datura stramonium were the most prevalent weeds in the cotton fields exhibiting Verticillium wilt. Verticillium dahliae Kleb. was recovered from A. retroflexus and X. strumarium in those cotton fields. This is the first report of V. dahliae occurring naturally in A. retroflexus in Turkey. Pathogenicity tests on cotton and weeds showed that the virulence of V. dahliae isolates from weeds was higher on cotton plants than on weeds, with the disease severity ranging from 31.7% to 98.0%. Disease severity of V. dahliae isolates was 54.7–93.9% on eggplant, 23.7–51.6% on cucumber and 11.0–16.4% on tomato, whereas it did not cause any disease symptoms, or only low levels, on pepper and bell pepper. Two vegetative compatibility groups (VCGs) were identified among seven tested weed isolates: VCG2A (two isolates) and VCG2B (three isolates) using international reference strains.  相似文献

In nature, oviposition and larval growth of the olive fruit flyBactrocera oleae (Rossi) (Diptera: Tephritidae) occur only in the mesocarp of fruits of the genusOlea, including the cultivated olive. Here we report on its growth in tomatoes, in the laboratory, as affected by a number of factors. Caged flies from a colony reared for more than 100 generations on an artificial diet, oviposited in intact fruits of four tomato cultivars in an unheated greenhouse and in the open. In choice tests, at 25°C, 16L:8D and 65% r.h., medium (5–6 cm diam) and large (>6 cm diam) fruits received more eggs than small (3–4 cm diam) fruits, but differences were not significant in all cases. In all cultivars, more eggs were laid in greenhouse than in open-air tomatoes of three different degrees of maturity, the differences being significant in most cases. The smaller the resistance of a cultivar’s epicarp to penetration by flat ended 0.2 and 1 mm square probes, the greater the number of eggs a cultivar received. Green (unripe) fruits proved unsuitable for larval growth. Half-ripe and fully ripe (red) fruits of cvs. ‘Isidro’, ‘204’, ‘Tobo’ and ‘Ace’ yielded pupae of acceptable weight (mean 5.6 to 6.7 mg) with a high percentage of adult emergence, yet the yield was considerably lower than that of olive fruit. Fully ripe greenhouse Isidro tomatoes yielded an average of 3.2 adults per fruit and 20.4% of the eggs laid developed into adults, whereas the other categories and cultivars yielded less. The mean percentages of pupae over eggs were higher in fully ripe Isidro and 204 fruits, than in half-ripe Tobo and Ace ones. The mean time from oviposition to pupation ranged from 16.2 to 25 days, depending on the cultivar and degree of fruit maturity. In fully ripe Isidro and 204 fruits, the fewer the eggs laid per fruit, the higher the percent yield in pupae and the longer the time from oviposition to pupation. Greenhouse fully ripe fruits of cv. 204 maintained under a low light intensity of 50 lux gave a much lower yield in pupae and slower larval growth than under 1600 lux. The development time and parasitization rate ofDiaeretiella rapae (M’Intosh) onBrevicoryne brassicae (L.) feeding on differentBrassica cultivars was studied in the laboratory at 20°C. The shortest development time from egg to adult parasitoid was 11.6 days on cabbage cv. ‘Yalova 1’ and the longest was 12.1 days on turnip cv. ‘Antep’ and rapeseed cv. local variety. Females lived significantly longer than males on the host plants used in the study. Females and males had the shortest longevity on rapeseed at 11.1 and 5.1 days, respectively. The highest percent parasitism ofB. brassicae byD. rapae was found on cabbage (40.20%), and the lowest was recorded on turnip (32.64%). Our results demonstrate that parasitism rate could be influenced by the plant quality, probably due to the nutritional status of the aphids or to toxic compounds ingested through the plant. Grapevine virus A (GVA) is considered one of the viruses associated with rugose wood (RW), one of the most economically important diseases of grapevine. Thirty-seven GVA isolates collected from grapevine cultivars from Marche (central-eastern Italy), Apulia and Campania (southern Italy), were subjected to molecular characterization. The genetic and population diversity was studied in the coat protein (CP) gene by RT-PCR-RFLP analysis with three restriction enzymes (MseI, AluI, and AciI), and nucleotide sequencing. A new primer pair (CP1F/R) allowing amplification of the whole CP gene (621 bp) was developed. RFLP with AciI yielded the highest number of variants in GVA isolates, showing seven different ‘simple’ profiles (A, B, C, D, E, F, and G). ‘Complex’ profiles were also found, and the most common variant combination was A + B in 39% of isolates. The analysis of GVA sequences confirmed the presence of plants infected with more than one GVA variant and suggested that RT-PCR-RFLP is suitable for evaluating population diversity of GVA enabling a screening of different haplotypes. The distribution of RFLP profiles and the phylogenetic analysis were not correlated with the location of infected plants, showing the presence of a GVA population with genetic diversity in the average with those of RNA viruses.  相似文献

In a field experiment conducted over two growing seasons, the effectiveness and phytotoxicity of inorganic fungicides such as sulphur, lime sulphur, copper, silicon and Armicarb (a new formulation of potassium bicarbonate) was compared with water for the control of primary apple scab infections in Belgium on high, medium and low scab-susceptible cultivars (cvs ‘Pinova’, ‘Pirouette’ and ‘Reinette des Capucins’, respectively). In order to drastically reduce the amount of fungicide applied in the orchard, two approaches were used: (1) a strategy involving spraying during the infection process, before fungal penetration and (2) a tunnel sprayer machine for treatment applications. Under field conditions highly favourable for disease, low rates of elemental sulphur (31.8 and 38.6 kg ha−1 year−1 in 2005 and 2006, respectively) combined with low rates of copper (2.1 kg ha−1 year−1 in both years) provided the best scab control and reduced scab severity on the fruits of cv. ‘Pinova’ by 97 and 98% compared with water control in 2005 and 2006, respectively. Lime sulphur was much more effective than wettable sulphur and appeared to be efficient at temperatures below 10°C, but its effectiveness against apple scab decreased if the treatments were applied 12–24 h later than in the ‘during-infection’ spray strategy. Armicarb used alone significantly reduced apple scab severity on the leaves and fruits of the three cultivars compared with the water control. Its effectiveness was as good as wettable sulphur applied using the same timing and dosage. Silicon reduced apple scab on fruits very slightly, but not on leaves. The amounts of wettable sulphur, lime sulphur, copper, silicon and potassium bicarbonate used in this experiment to control apple scab were not phytotoxic, did not increase fruit russet, did increase the yield of each cultivar and did not affect summer density of the beneficial Typhlodromus pyri. The potential and limitations of ‘during-infection’ spraying as a protection strategy against apple scab in organic farming are discussed.  相似文献

The spider mites Tetranychus urticae Koch and Panonychus ulmi (Koch) (Tetranychidae) cause severe economic losses to vegetable farms and deciduous fruit orchards in Turkey. One of their predators, the ladybird beetle Stethorus gilvifrons (Muls.) (Col., Coccinellidae), aggregates on mite-infested patches of plants. The present study assessed whether there is a role for herbivore-induced plant volatiles (HIPVs) and/or odors emitted directly from these two mite species in the aggregative response of ladybird beetles. The olfactory responses of the predator females to volatiles from T. urticae- and/or P. ulmi-infested sweet pepper (four cultivars, viz. ‘Demre’, ‘Yalova Carliston’, ‘Kandil Dolma’ and ‘Yag Biberi’), kidney bean (cv. ‘Barbunya’) and apple (M9 rootstock) were investigated using a two-choice olfactometer. Our results showed that HIPVs emitted from both T. urticae- and P. ulmi-infested plants significantly attracted S. gilvifrons adults for all plants except the sweet pepper cv. Yag Biberi. In addition, it was found that volatiles from apple plants infested by T. urticae and, especially, P. ulmi are more attractive for S. gilvifrons females than those emitted by other infested plants. The results also suggest that the odors of T. urticae adults and their products might influence the attraction of S. gilvifrons females.  相似文献
