Fourteen fall-born lambs were used to determine the effects of diet before and after weaning on intake, growth, serum hormones, and metabolite profiles. Before weaning, lambs were intensively sampled for 6 h at 35 and 42 d of age. Before sample collection, lambs were allowed to suckle, and milk intake was recorded. At 42 d of age, lambs were weaned and randomly allotted to ad libitum access to either alfalfa or grass hay. Blood samples were collected at 49 and 56 d of age for 6 h. Milk intake did not differ (P greater than .10) between groups. After weaning, lambs fed alfalfa hay consumed more (P less than .05) hay and had greater (P less than .05) ADG than lambs fed grass hay. Postweaning diet had no effect (P greater than .10) on serum insulin, growth hormone (GH), insulin:GH ratio, prolactin, cortisol, glucose, or nonesterified fatty acids (NE-FA) concentrations. Lambs consuming alfalfa had higher (P less than .05) serum urea nitrogen (SUN) at 49 and 56 d of age than lambs consuming grass. At 35 and 42 d of age, (P less than .05) serum insulin and insulin:GH ratio were higher (P less than .05) after milk intake than at 49 and 56 d of age after hay intake. The relationship of serum cortisol to immunoglobulin absorption and gut closure in cesarean-derived neonatal lambs was evaluated in two trials. In trial 1, 21 lambs were obtained on d 136 to 138 of gestation, and in trial 2, 17 lambs were obtained on d 140 to 142 of gestation. At birth, lambs were assigned randomly to four treatments: 1) control (CO), 1 ml saline/kg BW every 4 h; 2) a drug to lower cortisol (LC), 5 mg metyrapone/kg BW every 4 h; 3) single-peak cortisol (SP), 10 IU ACTH/kg BW at 0 h; or 4) elevated cortisol (HC), 5 mg cortisol/kg BW every 4 h in trial 1 or 10 IU ACTH/kg BW every 4 h in trial 2. The treatment period was 24 and 48 h after delivery for trial 1 and 2, respectively. Lambs were fed pooled bovine colostrum every 4 h for 48 h after birth at 2 and 3.5% BW for trial 1 and 2, respectively. Compared with CO, HC increased serum cortisol, LC decreased serum cortisol and SP elevated serum cortisol concentrations through at least 8 h for both trials. In trial 1, HC and SP lambs exhibited elevated serum IgG, IgM and IgA concentrations by 20 h compared with CO. However, no difference in serum immunoglobulin concentration was observed at 36 h among CO, HC and SP. Conversely, LC had the lowest immunoglobulin concentration at 36 and 48 h, and precocious closure to immunoglobulin absorption had occurred by 20 h (P less than .05).(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

A study was carried out to assess the bioavailabilities of several forms of vitamin E in lambs. A total of 40 lambs was allotted to eight dietary groups of five each and supplemented or not daily for 60 d with equimolar amounts of different vitamin E compounds as follows: 1) control, no supplemental vitamin E, 2) DL-alpha-tocopheryl acetate, 3) D-alpha-tocopheryl acetate, 4), D-alpha-tocopheryl succinate, 5) D-alpha-tocopheryl polyethylene glycol 1,000 succinate (TPGS), 6) DL-alpha-tocopheryl nicotinate, 7) DL-alpha-tocopheryl nicotinate+ TPGS, or 8) D-alpha-tocopheryl acetate + TPGS. During these 60 d, serum alpha-tocopherol concentrations in the control lambs remained constant and lower (P less than .05) than in lambs that received all treatments. Various indices of bioavailability, including Cmax-C(i) (concentration maximum-concentration initial), Ct-C(i) (concentration terminal-concentration initial), areas under the serum concentrations profiles, and pooled increment were higher (P less than .05) with D-alpha-tocopheryl acetate+ TPGS than in the other groups, suggesting a synergism between these forms. No such effect was observed between nicotinate and TPGS. For the TPGS, a water-soluble form of vitamin E, the indices of bioavailability were lower (P less than .05) than for the other groups. D-alpha-tocopheryl acetate resulted in a bioavailability that outranked all the other forms of vitamin E, except those of D-alpha-tocopheryl acetate + TPGS. Fifty-two Targhee twin-bearing ewes were used in a factorial arrangement of treatments to investigate the role of supplemental vitamin E (vit E); 0 (NE) vs 400 IU of vit E x ewe x (-1)d(-1) (E) and parainfluenza type 3 (PI3) vaccination; none (NP) vs PI3 vaccination (P) in immune function. Parainfluenza type 3 vaccination was used to evoke an immune response. Ewes receiving PI3 were vaccinated at 49 and 21 d before the expected lambing date. Ewes receiving vit E were orally dosed daily, 32 to 0 d before lambing. Blood was collected from ewes at the time of the initial PI3 vaccination and 4 h postpartum. Blood was collected from lambs (n = 104) at 3 d postpartum. Ewe and lamb sera were analyzed for anti-PI3 antibody titers, immunoglobulin G (IgG) titers, and vit E concentrations. Colostrum was collected 4 h postpartum and analyzed for IgG. The model for ewe and lamb analysis included the main effects of vit E and PI3, sex (lambs model only), and their interactions. No interactions were detected (P > 0.20) for any ewe or lamb variables. Serum anti-PI3 titers were greater (P < 0.01) in P ewes and their lambs than NP ewes and their lambs. Serum vit E concentrations were greater (P < 0.01) in E ewes and their lambs than NE ewes and their lambs. Colostral IgG titers and serum anti-PI3 titers did not differ (P > 0.20) between E and NE ewes. Serum IgG titers in E ewes and their lambs did not differ (P > 0.15) from IgG titers in NE ewes and their lambs. Lamb anti-PI3 titers did not differ (P = 0.76) between lambs reared by E and NE ewes. These results indicate that, although supplemental vit E to the ewe increased lamb serum vit E concentration, it had no effect on measures used in this study to assess humoral immunity in the ewe or passive immunity to the lamb.  相似文献

A possible role for endogenous opioid peptides (EOP) in the control of luteinizing hormone (LH) and prolactin (PRL) secretion was studied by injecting the opioid antagonist, naloxone (NAL), into postpartum ewes and cows. Twelve ewes that lambed during the fall breeding season and nursed their lambs were injected iv with NAL (1.0 mg/kg) on d 10, 14, 18, 22 and 26 postpartum. Blood samples were collected at 15-min intervals from 2 h before to 2 h after NAL, and serum concentrations of LH and PRL were quantified. Following treatment on d 10, suckling lambs were removed from 6 of the 12 ewes, creating non-suckled (NS) and suckled (S) treatment groups for subsequent study on d 14 through 26. On d 10, NAL treatment increased LH (P less than .01) but concentrations of PRL were not affected. When averaged across d 14 to 26, post-NAL concentrations of LH were greater (P less than .001) than pre-NAL concentrations (6.5 +/- .7 vs 1.9 +/- .4 ng/ml). In contrast, concentrations of PRL in the post-NAL period were lower (P less than .001) than pre-NAL concentrations (129 +/- 15 vs 89 +/- 10 ng/ml). Compared with S ewes over d 14 to 26, those in the NS group had similar pre-NAL concentrations of LH, tendencies for higher (P less than .10) post-NAL concentrations of LH, lower (P less than .001) mean serum concentrations of PRL (pre- and post-NAL) and similar pre-NAL vs post-NAL differences in serum PRL. Six suckled beef cows on d 24 to 35 were injected iv with either saline or NAL (.5 mg/kg) in a replicated crossover design. Injections of NAL increased serum concentrations of LH (P less than .05), when averaged over all 12 injections in the six cows, but serum PRL was not changed. However, three of six cows did not respond to NAL with increases in serum LH. These non-responding cows were similar to the responding cows in their pre-injection concentrations of LH and PRL, but they tended (P = .10) to have higher serum concentrations of cortisol than responding cows.  相似文献

Previous studies of the amino acid analogue, alpha-ketoisocaproate (KIC), indicate that it can stimulate lymphocyte blastogenesis and antibody responses of sheep. To determine whether KIC could overcome the effects of adrenocorticotropic hormone (ACTH)-induced lymphocyte suppression, 24 lambs were fed a control diet, a diet supplemented with 0.05% KIC, or a diet supplemented with 0.05% of the parent amino acid leucine. Immune status was monitored by determining lymphocyte blastogenic responsiveness to phytohemagglutinin-P (PHA), concanavalin A (conA), and pokeweed mitogen (PWM) and percentages of T-cell subsets in the blood, using monoclonal antibodies and a flow cytometer. Serum cortisol, insulin, and glucagon concentrations also were determined. After 60 days of consuming the respective diet, lambs were administered either saline solution or ACTH (100 IU) twice daily for 3 consecutive days. Administration of ACTH increased serum cortisol and insulin concentrations; however, no effects were seen for serum glucagon concentration. Compared with saline administration, ACTH administration significantly (P less than 0.05) suppressed mitogen-stimulated lymphocyte blastogenesis by approximately 50%, regardless of the mitogen used, and significantly (P less than 0.01) decreased the percentage of circulating T lymphocytes and decreased (P less than 0.01) the ratio of T4 to T8 cells. Lambs fed KIC had greater PHA- and conA-stimulated blastogenic responses and significantly (P less than 0.05) increased ratio of T4 to T8 cells in the blood, compared with lambs fed the leucine-supplemented diet or the control diet and given corresponding injections. These data indicate that ACTH decreased in vitro lymphocyte blastogenesis and altered the subset ratios of blood lymphocytes in sheep. These changes were partially prevented by feeding KIC.  相似文献

Three trials using 156 Yorkshire x Hampshire x Duroc crossbred pigs (avg initial wt, 7.9 kg) were conducted to evaluate the effects of two supplemental dietary vitamin E (11 vs 220 IU/kg of diet) and weaning age (21, 28 or 35 d) on performance and immunocompetence of pigs. Supplemental vitamin E (220 IU/kg of diet) increased (P less than .01) serum concentrations of vitamin E for all weaning ages compared with pigs fed 11 IU of vitamin E/kg of diet. However, supplemental vitamin E did not affect performance, serum cortisol concentration or the primary and secondary antibody response to sheep red blood cells. As weaning age increased, weekly ADG and avg daily feed intake increased linearly (P less than .01). Cortisol levels decreased during the 1st wk following weaning and then increased linearly (P less than .01) over time; pigs weaned at 35 d of age had higher (P less than .01) cortisol values initially and over time than pigs weaned at 21 and 28 d. Pigs weaned at 35 d had a higher (P less than .01) primary response to sheep red blood cells than pigs weaned at 21 and 28 d of age, but this effect was not observed for the secondary response. There were no interactive effects (P greater than .10) of dietary vitamin E level and weaning age. This study was conducted to determine the effect of dietary selenium (Se) and vitamin E (Vit E) on various blood characteristics and the primary and secondary humoral immune response of lambs challenged with parainfluenza3 virus (PI3 V). Treatments included: 1) +Se/+Vit E, 2) -Se/+Vit E, 3) +Se/-Vit E and 4) -Se/-Vit E. The basal diet (-Se/-Vit E) was deficient in Se and Vit E. Sodium selenite (.2 mg Se/kg diet) and alpha-tocopherol acetate (20 mg Vit E/kg diet) were added to +Se and +Vit E diets, respectively, to provide adequate levels of each according to NRC recommendations. Following a 10-wk dietary adaptation and depletion period, lambs in all treatment groups were intratracheally inoculated with PI3 V on d 0 and 35 of the 70-d study. Prior to inoculation, whole blood and plasma glutathione peroxidase (GSH-Px) activities were higher (P less than .01) for +Se lambs. Whole blood and plasma GSH-Px increased (P less than .01) after primary viral inoculation in +Se lambs but not in -Se lambs. Serum immunoglobulin M (IgM) concentrations were enhanced (P less than .05) by Se supplementation on d 14, 35 and 49 of the study. Selenium and (or) Vit E did not affect serum immunoglobulin G (IgG) levels. Serum PI3 V antibody titers increased after inoculation on d 0 and 35 in all treatment groups. The objective of this study was to determine the effect of feeding Ascophyllum nodosum (ANOD) to lambs at 0 (control), 0.5, 1, or 2% of DMI/d for 2 wk on lamb physiology in response to forced walking and transport during hot weather. Forty-four lambs (26 kg +/- 4.3) were used, and each lamb swallowed 3 gelatin capsules filled with ANOD or their normal 16% CP, pelleted grain ration twice daily, with the amount of ANOD dependent on the treatment. The amount of ANOD did not affect ear canal temperature or cortisol concentrations during 60 min of forced walking. The range between the minimum and maximum ear canal temperature for each lamb during 12 h of transport was narrower in lambs receiving the 2% ANOD than the control group (P = 0.05), and the 2% ANOD group also had lower (P = 0.05) ear canal temperatures than the control group during hot periods of transport. After 4 (P = 0.09) and 8 h (P = 0.05) of transport, the control group tended to have greater cortisol concentrations than the 2% ANOD group. Many differences among treatments were found in plasma protein, albumin, calcium, phosphorus, glucose, blood urea nitrogen, aspartate aminotransferase, magnesium, sodium, potassium, and chloride concentrations posttransport; mainly, the control and 0.5% ANOD groups had greater (P < 0.05) concentrations than the other 2 treatments. Aldosterone concentrations were greater in the control and 0.5% ANOD group than in the 1 and 2% ANOD groups before transport, whereas the concentrations were not different after transport, suggesting pretransport concentrations were lowered by supplementation. The 1 and 2% ANOD groups lost more BW than the control group as a result of transport (P = 0.04). After transport, no differences were observed in the latency for lambs to drink, eat, or lay. There was a suppression of the IgG and IgM antibody responses at 4 and 7 d after administration of ovalbumin, with greater ANOD supplementation rates suppressing antibody response the greatest. Although ANOD decreased ear canal temperature in hot periods of transport, stabilized electrolyte concentrations, and decreased cortisol throughout transport, it also suppressed the antibody response indicating that the effect of ANOD on immune function merits further investigation.  相似文献

Thirty-eight pregnant Suffolk ewes were assigned randomly to a control group or implanted with approximately 2 g of testosterone propionate (TP) when they were between d 40 and 60 of gestation. Implants were removed 3 wk prior to lambing. Five ewe lambs born to implanted ewes and ten ewe lambs born to nonimplanted ewes were utilized in this experiment. Ram lambs were not used in this trial. No differences (P greater than .10) were observed for fecal, urinary and total N excretion and amount of N absorbed. Nitrogen retained (percentage of N intake and g/d) was higher (P less than .05) in prenatally androgenized ewe lambs than in control ewe lambs. Plasma insulin concentrations averaged 99% higher (P less than .05) in prenatally androgenized ewe lambs. Plasma insulin-like growth factor I (IGF-I) concentrations averaged 29% higher (P less than .06) in ewe lambs treated prenatally with testosterone. Nonesterified fatty acid (NEFA) concentrations averaged 41% higher (P less than .05) in prenatally androgenized ewe lambs. Significant (P less than .05) treatment x time effects were observed in plasma thyroxine, glucose and urea N concentrations of prenatally androgenized vs control ewe lambs. These significant modifications in the plasma metabolite and endocrine status could be an important element of the physiological mechanism(s) by which prenatal androgenization improves growth performance and leanness of ewe lambs.  相似文献

AIM: To determine the effect of increasing doses of long-acting injectable vitamin B12 plus selenium (Se) given pre-mating on the vitamin B12 and Se status of ewes and their lambs from birth to weaning. METHODS: Four groups of 24 Poll Dorset ewes each were injected 4 weeks pre-mating with different doses of a long-acting vitamin B12 + Se product, containing 3 mg vitamin B12 and 12 mg Se per ml. The treatment groups received 5 ml (15 mg vitamin B12 + 60 mg Se), 4 ml (12 mg vitamin B12 + 48 mg Se), 3 ml (9 mg vitamin B12 + 36 mg Se), or no vitamin B12 or Se (control). Twelve of the twin-bearing ewes per group were selected for the study. Efficacy of the product was evaluated from changes in the concentrations of vitamin B12 in serum and liver, and of Se in blood, liver and milk in the ewes during gestation and lactation, and in their lambs from birth to weaning. Pasture samples in paddocks grazed by the ewes and lambs were collected at about 2-monthly intervals from 200-m transects. RESULTS: The flock was Se-deficient, as the mean initial concentration of Se in the blood of ewes was 182 (SE 20.3) nmol/L. Compared with untreated controls, all doses significantly (p < 0.01) increased concentrations of Se in the blood of ewes for at least 300 days. Selenium concentrations in milk were likewise increased throughout lactation, as were those in the blood and liver of lambs. The mean concentration of vitamin B12 in the serum of ewes was initially > 1,000 pmol/L, but this decreased within 28 days to < 460 pmol/L. Treatment with the 5-ml and 4-ml doses raised serum vitamin B12 concentrations of ewes for at least 176 days (p < 0.01), while their lambs had significantly greater concentrations of vitamin B12 in serum and liver for less than 37 days after birth. Tissue concentrations and duration of elevation of both vitamin B12 and Se were proportional to the dose administered. The mean concentrations of Se and cobalt (Co) in the pastures were 32 and 74 microg/kg dry matter (DM), respectively. CONCLUSIONS: Injecting ewes from a Se-deficient flock 4 weeks prior to mating with 48 or 60 mg Se and 12 or 15 mg vitamin B12 increased and maintained the Se status of ewes for at least 300 days, and of their lambs from birth to weaning. The vitamin B12 status of ewes was increased for at least 176 days and that of their lambs for less than 37 days. Due to the proportional nature of the response to increasing dosage, the dose rate of the formulation tested can be adjusted according to the severity of Se and Co deficiency in a flock. CLINICAL SIGNIFICANCE: A single subcutaneous injection of vitamin B12 + Se administered pre-mating to Se-deficient flocks is likely to prevent Se deficiency in ewes and their lambs until weaning, as well as increase the vitamin B12 status of ewes and their lambs until 5 weeks after lambing.  相似文献

The objective of this study was to determine whether feeding high doses of vitamin D3 7 d before slaughter would increase muscle Ca++ levels and result in more tender loin chops. Market lambs (n = 4 callipyge and 4 normal in Exp. 1, and n = 16 calipyge and 16 normal in Exp. 2) were randomly and equally assigned to feeding groups based on callipyge genotype and experimental diet, (vitamin D3 or control). Serum Ca++, muscle Ca++, Warner-Bratzler shear force, and troponin-T degradation data were analyzed. In Exp. 1, vitamin D3 was supplemented at 1 or 2 x 10(6) IU/d. The 2 x 10(6) IU dose resulted in the greatest serum Ca++ reponse and was chosen for Exp. 2. In Exp. 2, serum Ca++ concentration was higher (P < 0.05) for normal and callipyge lambs fed the vitamin D3 diet than for the control diet fed lambs. Muscle Ca++ concentrations, however, were not higher (P = 0.28) for the vitamin D3-fed lambs. Warner-Bratzler shear values were higher (P < 0.05) for callipyge than for normal lambs, but no differences were observed with vitamin D3 supplementation. These data were supported by results from Western blot analysis of troponin-T degradation, in which no differences were observed for vitamin D3 vs control diet lambs at 14 d postmortem. This experiment showed that feeding 2 x 10(6) IU/d of vitamin D3 to market lambs, callipyge or normal, raised serum Ca++ concentration, but did not increase muscle Ca++ concentration. This lack of response in muscle Ca++ was likely the reason that no differences were observed for Warner-Bratzler shear force values or troponin-T degradation data between the vitamin D3 and control loin chops. A higher dose of vitamin D3 may be required to improve tenderness.  相似文献

1. Effects of Ascaridia galli infection on the energy and nitrogen (N) metabolism were studied on groups of 5 cross-bred cockerels aged about 5 weeks and given a diet deficient or adequate in vitamin A at two levels of feeding in respiration chambers. 2. Metabolisability of dietary energy was 67% and N retention 33% in infected chickens compared with 71 and 41% respectively, in uninfected chickens. 3. Maintenance energy requirement of vitamin A-deficient birds was 882 kJ/kgW d compared with 998 kJ/kgW d for normal birds. N balance of the deficient chickens was also less when compared at the same energy balance. Infection did not affect maintenance energy requirement nor N balance. 4. Starvation heat production of infected chickens (619 kJ/kgW d) was higher than that of uninfected controls (586 kJ/kgW d). When infection treatments were combined, vitamin A-adequate chickens had a higher heat production (615 kJ/kg d) than the vitamin A-deficient (580 kJ/kgW d). Endogenous N excretion (mg/gW) was less in vitamin A-deficient than in adequate, starved birds. 5. Deficient chickens had undetectable liver reserves of vitamin A and only very low plasma concentrations. There was a difference in the length of larvae (17 d after infection) associated with vitamin A status, and with level of feeding.  相似文献

Medium wool ewes were injected with vitamin E and(or) Se over a 2-yr period to evaluate the influence of these treatments on reproduction. Ewes were divided randomly into four groups, consisting of a control, plus groups receiving monthly sc injections of either 272 iu vitamin E, 4 mg Se or 272 IU vitamin E plus 4 mg Se during pregnancy. Selenium administration increased (P less than .05) ewe blood Se concentrations, but had no effect (P greater than .10) on fertility (number of ewes lambing of ewes bred), prolificacy (number of lambs born/ewe lambing) or lamb sex ratio. Preweaning survival of lambs was increased (P less than .05) by ewe treatments with either Se or vitamin E and thus, treated ewes weaned approximately 20% more lambs/ewe mated than did control ewes.  相似文献

The influence of plane of nutrition, growth hormone (GH) treatment and dietary polyunsaturated fat on serum concentrations of GH and insulin (INS) and binding capacities of GH, INS and prolactin (PRL) in liver, mammary parenchyma and adipose tissue was assessed in prepubertal ewe lambs. Ten lambs were assigned to each of four treatment groups. Treatments included: (A) lambs with ad libitum access to a high-energy ration; (G) lambs fed as group A and treated with bovine GH (.08 mg/kg/d); (R) lambs with feed intake restricted to limit ADG to about 120 g; and (S) lambs with ad libitum access to a ration including formaldehyde-protected sunflower seed. Diets, all approximately isonitrogenous and isocaloric, were fed from about 7 to 22 wk of age. Weekly blood samples were collected during wk 6 to 14 of the trial. Averaged across sampling dates, mean serum concentrations of GH were elevated in G lambs (P less than .05) and INS concentrations differed in the order G greater than A greater than R = S (P less than .05). Crude membranes for binding assays were prepared from liver, mammary parenchyma and adipose tissue. Mean concentrations of GH receptors in liver and PRL receptors in mammary parenchyma were elevated in group S lambs (P less than .01). Dietary polyunsaturated fat increased the number of GH receptors in liver and PRL receptors in mammary parenchyma. Increased availability of receptors may mediate the stimulation of mammary growth observed in lambs fed polyunsaturated fat.  相似文献

Pregnant ewes were supplemented with dl-alpha-tocopheryl acetate, either as a single intramuscular dose (500 mg two weeks before lambing) or perorally (150 mg daily during 3-4 weeks before lambing). Ewes without such a supplementation were controls. The vitamin E supplemented ewes had nearly twice as high vitamin E (alpha-tocopherol) concentrations as the unsupplemented control ewes at lambing both in serum and in colostrum. The vitamin E concentration in colostrum was 5-11 higher than in milk 1 week after lambing. Both supplementations somewhat increased the vitamin E serum concentration of the newborn lambs, but the increase was negligible in comparison with the effect produced by the consumption of colostrum. All lambs had very low serum concentrations at birth. The lambs from the supplemented ewes had significantly higher serum values than the control lambs 24 h after birth. The ewes had somewhat higher selenium status at birth than their offsprings when evaluated by glutathione peroxidase (GSH-Px) in the erythrocytes. It seems reasonable that nutritional muscular degeneration may arise in newborn lambs with a normal selenium status if their vitamin E status is critical, either because of an inadequate consumption of colostrum or because of a vitamin E deficient diet during pregnancy with a low vitamin concentration of colostrum as a consequence.  相似文献

Monensin in ruminant diets increases production of propionic acid. We have tested the hypothesis that propionic acid may be elevated to such an extent by monensin that it cannot be optimally metabolized by the methyl malonyl-CoA pathway requiring vitamin B12 (B12) in the liver. Thus, the effects of weekly B12 injections (10 mg X head-1 X wk-1, intramuscularly) with and without dietary monensin (25 mg/kg diet) on average daily gain (ADG), dry matter intake (DMI), feed to gain ration (F/G), liver and serum B12 concentrations and liver activity of propionate metabolizing enzymes were examined in an 84-d trial. Sixteen lambs (27.5 kg average initial wt) were assigned randomly to one of four treatments in a factorial arrangement: monensin plus B12, monensin without B12, no monensin plus B12 and no monensin without B12. Lambs were fed an 80% concentrate diet and slaughtered at the end of the trial. Liver samples were obtained by biopsy on d 0 and at slaughter on d 84 to determine activity of propionate metabolizing enzymes and B12 concentrations. Serum samples were taken on d 0, 28, 56 and 84 to determine serum B12 concentration. Neither monensin nor B12 affected (P greater than .10) ADG, DMI or F/G. Lambs receiving B12 had higher (P less than .01) serum B12 concentrations, but this was not reflected (P greater than .10) in higher liver B12 concentrations. No difference (P greater than .10) in liver propionate metabolizing activity among treatments was detected; however, monensin decreased (P less than .05) fumarate and malate formation by liver homogenates. Liver B12 concentrations were highly correlated with endogenous propionate metabolizing activity at d 0 (r = .73, P less than .01) and d 84 (r = .51, P less than .05). Results suggest no advantage to providing supplemental B12 to lambs fed monensin-supplemented, high-concentrate diets.  相似文献

BACKGROUND: Selenium (Se) deficiency is an important etiological factor in Nutritional Myodegeneration Disease (NMD) of lambs, and Se is required for synthesis of thyroid hormones. HYPOTHESIS: That serum T4 and T3 concentrations in lambs with NMD will be abnormal. ANIMALS: Ten healthy lambs and 15 lambs with NMD were included in the study. METHODS: Serum thyroid hormone concentrations were measured by chemiluminescence assay in samples collected from control and NMD lambs before and after treatment with a preparation containing sodium selenite, vitamin E, and vitamin B1, which was administered subcutaneously to lambs with NMD twice, with a 2-week interval. RESULTS: Before treatment, serum concentrations of fT3 and TT3 in lambs with NMD were lower than those of control group (P < .001), while serum concentrations of fT4 and TT4 in lambs with NMD were significantly higher than those of control group (P < .001, P < .01, respectively). Similarly, serum creatine kinase (CK), lactate dehydrogenase (LDH), aspartate aminotransferase (AST), alanine aminotransferase (ALT), and alkaline phosphatase (ALP) values in lambs with NMD were significantly higher compared with those of control (P < .001, P < .001, P < .001, P < .01, and P < .05, respectively) before treatment. However, serum concentrations of Se and alpha-tocopherol in lambs with NMD were significantly lower compared with those of control (P < .001). After treatment (at day 30), none of the variables were significantly different between control and NMD lambs (P > .05). CONCLUSIONS AND CLINICAL IMPORTANCE: Nutritional myodegeneration in lambs is associated with abnormalities in serum thyroid concentrations. Abnormalities in serum thyroid concentrations can result from selenium deficiency in NMD lambs. Serum thyroid concentrations together with serum CK, LDH, AST, ALT, and ALP values can be considered additional tools in diagnosis and prognosis of NMD in lambs.  相似文献

In sheep, serum concentrations of leptin change congruently with increases or decreases in nutritional status, while intracerebroventricular infusions of leptin dramatically suppress feed intake in well-fed lambs, and may also increase growth hormone (GH), and/or luteinizing hormone (LH) in undernourished lambs. The objective of the present study was to determine the effects of peripherally delivered ovine leptin, via intravenous infusions, on feed intake and serum concentrations of GH, LH, insulin, IGF-1, cortisol, and thyroxine. Twelve ewe lambs weighing 29.4 +/- 0.7 kg were infused intravenously with a linearly increasing dose of leptin or saline (n = 6 per group) for 10 days, reaching a maximum dose delivered of 0.5mg/h on day 10. Feed intake was assessed twice daily, and blood samples were collected every 10 min for 6 h on days 0, 2, 5, 8, and 10. Serum concentrations of leptin increased in leptin-treated lambs by day 2 (P = 0.05), and continued to increase to concentrations 9-fold greater than saline-infused lambs by day 10 (P < 0.001). Despite the substantial increase in serum leptin, feed intake did not differ between leptin and saline-infused lambs except on day 3.5 (P = 0.01). Furthermore, intravenous infusions of leptin did not significantly influence serum concentrations of insulin, cortisol, IGF-1, thyroxine, LH, or GH. Collectively, these observations contrast with the potent hypophagic effects of leptin when delivered intracerebroventricularly into well-fed lambs. The reasons for the disparate response of lambs treated intravenously with leptin, versus that reported for lambs treated intracerebroventricularly with leptin are not known, but may provide insight into the mechanism(s) of leptin resistance.  相似文献

Twenty-four lambs, averaging 29 kg, were used to determine the effect of supplemental dietary Fe on performance and Cu, P, Zn and Mn utilization. Treatments consisted of supplemental Fe at 0, 300, 600 or 1,200 mg/kg diet as ferrous carbonate. The basal diet contained 154 mg Fe/kg diet and consisted of 90% Coastal bermudagrass pellets, 9.45% group corn, .5% sodium chloride and .05% vitamin mix. Lambs were slaughtered after having ad libitum access to diets for 98 to 121 d. Dietary Fe did not affect lamb gain or feed intake. Supplemental Fe increased Fe concentrations in liver (P less than .01), spleen (P less than .01) and bone (P less than .10), but not in kidney and muscle. Serum Fe concentrations and percentage transferrin saturation in serum were increased (P less than .01) by supplemental Fe at 28 and 84 d, but not at the termination of the study. Plasma Cu was decreased (P less than .01) at 56 d, whereas serum ceruloplasmin activity was reduced (P less than .01) at 28 d in lambs fed 1,200 mg Fe/kg diet compared with lambs fed 600 mg Fe/kg diet. Lower levels of Fe (300 and 600) reduced (P less than .01) ceruloplasmin by 56 d and plasma Cu by 84 d compared with controls. Liver Cu also was decreased (P less than .05) by supplemental Fe.
