The oocyte of Sterlet (Acipenser ruthenus) and Siberian sturgeon (Acipenser baeri) was evaluated after incubation with 17α, 20β‐Dihydroxyprogesterone (DHP) in artificial media. Concentration of testosterone (T), progesterone (P4) and 17‐β estradiol (E2) in blood and their relation to fertilization rate were measured during experimental period. The oocytes at the same developmental stages were incubated in SIS (based on sturgeon blood ionic composition), RM2 (Ringer solution modified for sturgeons) and L‐15 (Leibovitz medium) artificial mediums in the presence of 1 μg/ml of 17α, 20β‐Dihydroxyprogesterone for 12, 18 and 24 hr. The result of this study demonstrated that fertilization rate decreased with the increasing duration of incubation in all mediums. In the Siberian and Sterlet sturgeon, the highest fertilization rate was observed in the oocytes incubated in RM2 (0.81 ± 0.4) and SIS (0.44 ± 0.8) mediums for 12 hr respectively. In the Siberian sturgeon, fertilization rate decreased significantly (p < .05) with incubation time in all three mediums. In Starlet, fertilization rate decreased significantly (p < .05) with time and reached a minimum after 24 hr (0.16 ± 0.1) in L‐15 medium. No significant (p > .05) differences were observed between blood plasma hormones and fertilization rate. The results of this study indicated that 17α, 20β‐Dihydroxyprogesterone is essential for induction of in vitro oocyte maturation in these species.  相似文献

The effects of dopamine on the secretion of two sturgeon gonadotropins (stGTH I and stGTH II) in sexually mature male white sturgeon (Acipenser transmontanus) were evaluated. In Experiment I, sturgeon were given intraperitoneal injections of physiological saline (PS), dopamine (100 mg kg−1), the gonadotropin releasing hormone analog D-Ala6-des-Gly10-GnRH ethylamide (GnRHA) (10 μg/kg−1), and a combination of GnRHa and dopamine. Fish receiving only GnRHa had significantly higher (p < 0.05) concentrations of plasma stGTH I and stGTH II compared to fish receiving PS, dopamine, or a combination of GnRHa and dopamine. Two hours following its administration, dopamine was effective in decreasing plasma concentrations of both stGTHs that were previously elevated by GnRHa. Dopamine or PS administered by themselves did not alter plasma concentrations of either stGTH. In Experiment II, sturgeon injected intraperitoneally with a combination of GnRHa and pimozide had significantly higher (p < 0.05) concentrations of plasma stGTH I and stGTH II compared to males receiving GnRHa or pimozide alone. While this effect of GnRHAa + pimozide was observed in the spring, no such potentiation was seen in these fish during the summer (Experiment III). These results represent the first evidence of dopaminergic inhibition of GnRH-induced pituitary gonadotropin secretion in Chondrostean fish.  相似文献

Pectoral fin tissue of white sturgeon was investigated as a potential non-lethal sample source for the detection of white sturgeon iridovirus (WSIV) infection. Histopathology and polymerase chain reaction (PCR) results using fin tissue were compared with the standard lethal histopathology sampling method that utilizes head tissue. Tissues for each of the three sampling methods were collected weekly for 8 weeks from individual sturgeon undergoing an experimental cohabitation challenge with fish infected with the Abernathy isolate of WSIV. Non-lethal fin histopathological evaluation did not reveal infection during the first 3 weeks of sampling, while non-lethal PCR and the lethal method were variable. However, all three sampling methods were equally capable of identifying infection from 4 to 8 weeks post-exposure. Of the survivors tested, all were negative by PCR and the lethal method, and only one fish was identified as being positive by non-lethal fin histopathology. In another experiment, all three sampling methods were applied to asymptomatic WSIV carriers in a case study conducted at the Kootenai Tribal Sturgeon Conservation Hatchery. Results showed that both lethal and non-lethal fin histopathology were equally effective in detecting infection, but PCR was unable to identify this strain of WSIV. Depending on the virus isolate, these results suggest that non-lethal sampling of fin tissue (histopathology or PCR) is comparable with the lethal sampling method at identifying WSIV infection once infection is established, and under certain circumstances may provide an alternative to lethal sampling.  相似文献

Four 1‐week growth trials were conducted to determine the effects of feeding rates on the growth performances of white sturgeon (Acipenser transmontanus) fries 6–9 weeks after initiation of feeding. Six feeding rates with four replications were used in each of the four trials, and the feeding rates were 3.0–8.0, 2.0–7.0, 1.0–6.0 and 1.0–6.0% body weight (BW) per day in 1% increment, respectively. Number of fries per replicate and their initial BW (means ± SEM) were 60, 45, 30 and 30 and 2.8 ± 0.1, 4.5 ± 0.4, 8.5 ± 0.7 and 10.0 ± 0.7 g, respectively. The fries were kept at 18–19 °C and fed a commercial salmonid feed (488 g kg?1 protein and 123 g kg?1 fat). Mortality was low and unrelated to feeding rates. Final body weights, body weight increases, specific growth rates and feed efficiency were significantly (P < 0.05) affected by the feeding rates. Body moisture and lipid contents were significantly affected by feeding rates except body moisture content in trial II. Body protein contents were not affected by feeding rates except in trial III. Broken‐line analysis on specific growth rates indicated that the optimum feeding rates were 6.5 ± 0.4, 4.8 ± 0.2, 4.2 ± 0.1 and 3.8 ± 0.2% body weight per day, respectively, for white sturgeon fries 6–9 weeks after initiation of feeding.  相似文献

Plasma estradiol-17 (E2), testosterone (T), 17,20-dihydroxy-4-pregnen-3-one (DHP) and 17,20,21-tri-hydroxy-4-pregnen-3-one (20-S) levels were measured by radioimmunoassay (RIA) in white perch (Morone americana) and white bass (M. chrysops) that were induced to undergo final oocyte maturation (FOM) with human chorionic gonadotropin (hCG). Plasma DHP levels increased in females of both species in association with oocyte germinal vesicle migration (GVM) and germinal vesicle breakdown (GVBD) and decreased thereafter. Plasma 20-S levels also increased with oocyte GVM in white bass, but were several-fold lower than DHP levels. Circulating E2 and T levels were greatest during GVM and GVBD in both species and decreased to low levels during oocyte hydration and ovulation. Follicles from white perch and white bass which received a priming injection of hCG in vivo, produced both DHP and 20-S in vitro after exposure to hCG and their oocytes underwent GVBD. Ovarian incubates from unprimed fish of either species produced only E2 and T and their oocytes did not complete GVBD. Oocytes from unprimed bass, but not perch, matured when follicles were exposed to hCG in vitro. Both trilostane and cycloheximide blocked in vitro production of DHP and 20-S and oocyte GVBD by white perch follices. DHP and 20-S were equipotent inducers of FOM in the GVBD bioassay. None of several other structurally-related steroids tested were effective within a physiological range of concentrations. These results indicate a role for DHP and 20-S in the control of FOM in white perch and white bass.  相似文献

High levels of 11-ketotestosterone (11KT) were found (49 to 160 ng ml–1) in plasma of Siberian sturgeon females during the end of their reproductive cycle. These levels were measured either by specific radioimmunoassay, or both by specific radioimmunoassay and by UV absorption after HPLC (isocratic conditions, 33% methanol, 26% acetonitrile, 41% water). In order to find the origin of 11KT synthesis, ovaries were incubated (30 min and 2h at 20°C) with tritiated 17-hydroxyprogesterone (17OHP) or with tritiated androstenedione (A4). Testosterone (conversion rate from tritiated 17OHP: 4%) and 11-ketotestosterone (conversion rate from tritiated A4: 1.6%) were identified as metabolites of respectively 17OHP and A4 (TLC, HPLC and crystallization). 11-hydroxyandrostenedione (11OHA4) and 11-hydroxytestosterone (11OHT) were suggested to be intermediate metabolites. Besides interrenal and blood cells were incubated respectively with tritiated cortisol and tritiated A4. 11OHA4 was identified in interrenal incubation (yield from tritiated cortisol: 1.2%). 11KT in interrenal (yield from tritiated cortisol: 0.14%), and 11OHA4 and 11KT in blood cells (yield from tritiated A4: 1.6%), were suspected to be synthesized (TLC, HPLC, acetylation). No significant metabolization of tritiated cortisol could be found in liver. Outbreaks of an infectious disease affecting cultured white sturgeon (Acipenser transmontanus) were investigated. Clinical signs included erratic swimming, arching of the back and mortality. Necropsy findings included poorly demarcated yellow to dark-red and friable lesions in the epaxial muscle, ulcerative skin lesions and haemorrhages in the swim bladder and coelomic wall. Histological evaluation revealed areas of necrotizing and heterophilic myositis with aggregates of bacterial cocci. The lumen of blood vessels in the dermis, under ulcerated areas, and in the posterior kidney, was occluded by fibrin thrombi. Aggregates of Gram-positive cocci were observed in the muscle lesions and within the fibrin thrombi in the dermis and kidney. Genetically homogeneous Streptococcus iniae strains were recovered from affected fish from different outbreaks. The isolates shared high degree of similarity at gene locus (gyrB) with previously characterized S. iniae from cultured fish in California, confirming the emergence of this particular strain of S. iniae in US aquaculture.  相似文献

Juvenile white sturgeon (Acipenser transmontanus, Richardson) were fed or starved for 4 weeks, and their whole body amino acid composition determined at the beginning and the end of the study. The amino acid compositions of muscle, liver, gill and gastrointestinal tract, and the free amino acids in plasma of fed and starved fish were also determined. Very little variation was observed in the amino acid compositions of all the tissues examined after starvation. However, the plasma free amino acids were significantly (P < 0.05) decreased following starvation. On the basis of the amino acid composition and nitrogen retention values of the whole fish, daily indispensable amino acid requirements for protein accretion and obligatory nitrogen losses (mg amino acid 100 g body weighr?1 day?1) were estimated. The pattern of indispensable amino acids required for each of these is different, with the pattern for protein accretion containing much higher levels of lysine, but lower levels of tryptophan and sulfur amino acids. Individual amino acid retention rates varied from 33.3% of dietary tryptophan to 80.2% for arginine. The estimated dietary indispensable amino acid pattern for young growing white sturgeon (expressed as a percentage of total indispensable amino acids plus cystine and tyrosine) is: arginine, 14.0; histidine, 6.6; isoleucine, 8.8; leucine, 12.5; lysine, 15.8; methionine (plus cystine), 6.6; phenylalanine (plus tyrosine), 15.5; threonine, 9.7; tryptophan, 0.8; and valine, 9.7.  相似文献

Juvenile white sturgeon (Acipenser transmontanus) were fed three isonitrogenous and isoenergetic diets containing either 35% D-glucose (HC), a mixture of 20% dextrin and 10% cellulose (MC), or 23% cellulose (LC), to investigate the effects of dietary carbohydrate on liver function. After 8-week feeding, body weight gain of fish fed the HC diet was consistently higher than that of fish fed the MC and LC diets, but was not significantly different from the MC-fed fish. Fish fed the HC diet had significantly (p < 0.05) higher feed efficiencies and liver glycogen concentrations than fish fed the MC and LC diets. Sturgeon were injected intravenously with 10 mg kg-1 body weight of sulfobromophthalein (BSP) and post-injection blood taken from the caudal vein at 15, 30, 60, and 120 min. No significant differences in plasma BSP concentrations were found among the treatments at these times. Plasma hemoglobin and activities of aspartate and alanine aminotransferase were not affected by the diets. This study suggests that the HC diet does not adversely affect liver function or weight gain. Inclusion of high dietary levels of digestible and inexpensive carbohydrates in commercial sturgeon feeds seems promising, but long-term feeding trials should be conducted to confirm this assertion.  相似文献

In general little is known about hormones and the ontogeny of the stress response in the early developmental stages of chondrostean fishes and in particular of white sturgeon (Acipenser transmontanus, Richardson 1836). In this study, we measured for the first time cortisol and sex steroids (testosterone and estradiol) in eggs, larvae, post-larvae, and fry of white sturgeon by radioimmunoassay (RIA), to elucidate some endocrine aspects of its development. The cortisol, testosterone, and 17β-estradiol of maternal origin found in unfertilized eggs of white sturgeon probably regulate both growth and development of the embryo. Cortisol decreased after fertilization, whereas testosterone and 17β-estradiol did not significantly change. During the late stages of embryo development and immediately after hatching, endogenous production of cortisol and sexual steroids, respectively, occurred. Sex steroids may be physiological inducers of gonad sex differentiation in sturgeon. All steroids showed an increase 10 days post-hatch (dph), near the transition from an endogenous to an exogenous energy source. Cortisol maintained the same basal levels even after metamorphosis, whereas testosterone and 17β-estradiol declined significantly in post-larvae at 35 and 45 days post-hatch. In addition, to evaluate the ontogeny of a functional hypothalamic-pituitary-interrenal (HPI) axis, larvae and fry were submitted to acute stress. The HPI axis did not seem to be functional on the first day post-hatch, but became so from the third day post-hatch onward.  相似文献

The aim of this research was to evaluate the efficiency of diets with Spirulina and plant oils (POs) inclusion for white sturgeon weaning and their effects on the fatty acid (FA) composition of fish flesh. Three isoproteic (45%) and isoenergetic (21 MJ kg?1 DM) diets were formulated: one fish meal‐based diet integrated with fish oil (FMO) and two 40% Spirulina meal‐based diets integrated with corn (SPC) or soybean (SPS) oils respectively. One hundred and thirty‐five white sturgeon fingerlings (mean weight 17.5 g) were stocked randomly in nine fibreglass tanks. At the end of the trial, which lasted 71 days, the growth performance traits and somatic indexes were determined. The chemical composition, gross energy and FA profile were determined on the fish fillets. No significant effects were observed for the growth performances or fillet chemical composition. The FA profile of the fillets reflected those of the diets. In particular, the fillets of the fish fed with the SPC and SPS diets were lower in n‐3 FA, due to the substitution of fish oil (FO) with POs. It is possible to replace FO and meal in sturgeon; therefore, Spirulina meal integrated with POs could be a good alternative to sturgeon diet.  相似文献

用海星鞘神经提取液和二硫苏糖醇对刺参卵母细胞进行了体外人工促熟研究。结果显示,二硫苏糖醇对刺参卵母细胞具有明显的促熟作用。刺参卵母细胞包被在滤泡中发育,卵母细胞通过动物极的卵突起连接到单层细胞构成滤泡膜上,充分发育的卵母细胞处于生发泡期,染色质凝集在核仁周围,处于生发泡期的卵母细胞在海水中不会自发成熟。用二硫苏糖醇处理充分发育的卵母细胞20min,卵母细胞的生发泡开始移动,并锚定在卵母细胞的动物极,继而出现生发泡破裂。此后,减数分裂重新启动,第一、二极体分别排出。但海星鞘神经提取液对刺参卵母细胞没有促熟作用。实验表明,二硫苏糖醇浓度在10-5~10-1mol/L范围内,均可促使刺参卵母细胞发生生发泡破裂,10-2mol/L时诱导卵母细胞成熟率可达90%以上。  相似文献

The histochemical characteristics of the black-brown pigment granules accumulated during the lecitotrophic stage in the digestive tract of Siberian sturgeon, Acipenser baeri larvae, were studied in order to identify the nature of such pigments. Pigment granules appeared just after hatching in contact with the endodermal epithelium and in the periphery of the yolk-sac matrix, and increased in number with digestive system differentiation. At 3 days post hatch, the accumulation of pigment granules in the spiral valve formed a pigmented plug, which was ejected with first faeces after first feeding. Pigment granules were insoluble in all solubilising solutions tested (distilled water, H2O2, NH4OH, KMnO4, absolute methanol and different acid, basic and oxidising solutions). Pigment granules were completely bleached with the oxidising processes of Gomori and Casella (15 min) and with H2O2 (20%) during 48 h. Histochemical characteristics of bleached pigmented granules revealed that they were basophilic (Eosine negative), presented a moderate methachromasia (Toluidine Blue positive) and argentaffinity (Masson Fontana method positive), and contained carboxylated and sulphated (strongly and weakly ionised) glycoconjugated residues (Alcian Blue pH 0.5, 1.0, 2.5 positive). Bleached pigment granules also contained reductor groups (Ferric ferricyanide-Fe III positive), while they were negative for Bromophenol Blue, Perl's Prussian Blue, Red Oil O and Sudan Black B, indicating the absence of proteins, iron and lipidic substances respectively. According to such results, the black-brown pigment granules observed in the alimentary canal of Siberian sturgeon larvae during the lecitotrophic stage correspond to melanins.  相似文献

本试验在水温17±1℃的条件下,采用半静态式生物毒性试验方法,探讨了氟离子(F-)对西伯利亚鲟胚胎发育的影响。结果表明,100~600 mg/L氟暴露会导致胚胎孵化延迟,氟暴露组半数孵化时间(MHT)比对照组推迟9~22 h。氟暴露同时导致胚胎死亡率显著增加(P<0.05),出膜仔鱼活力减弱、存活时间缩短、畸形率显著升高(P<0.05),出现卵黄囊畸形、脊椎畸形、眼部充血等畸形症状。试验得出F-致西伯利亚鲟胚胎144 h半致死浓度(LC50)为447.61 mg/L,孵化安全浓度(SC)为4.48 mg/L,导致仔鱼畸形的半数效应浓度(EC50)为536 mg/L。氟对西伯利亚鲟胚胎的安全质量浓度低于我国部分高氟地区地下水、地表水及人为氟污染水域中氟含量,提示天然水体中氟可能会对鱼类胚胎产生负面影响,应引起重视。  相似文献

通过选取11个侧视形态特征度量进行主成分分析(principal component analysis,PCA),同时设置26个标记点进行相对扭曲分析(relative warp analysis,RWA),比较了中华鲟(Acipenser sinensis)和西伯利亚鲟(A.baerii)幼鱼在侧视形态上的差异以及相对扭曲分析和传统多元分析在鲟鱼形态研究中的优劣。结合前期对它们的有氧游泳能力比较结果(西伯利亚鲟比中华鲟强25%),分析了鲟鱼形态的水动力功能。主成分分析比较表明,中华鲟头长、头高、背鳍前基点之后的躯干高度、尾柄长、背鳍前缘长度均显著大于西伯利亚鲟(P<0.05),而尾鳍上叶显著短于西伯利亚鲟(P<0.05)。相对扭曲分析计算样本的几何信息并可视化统计结果,表明中华鲟的吻厚、吻长、头高、头长、躯干后半段高、背鳍前缘长显著大于西伯利亚鲟,而尾柄长和尾鳍上下叶长显著小于西伯利亚鲟(P<0.001)。上述形态特征直接影响着两种鲟鱼的游泳能力。  相似文献

以1998―2008年孵出的子一代(F1)中华鲟(Acipensersinensis)为材料,研究了人工养殖中华鲟的繁殖生物学特征。结果表明:年龄10~20龄的492尾人工养殖子一代中华鲟体重为30~169 kg,体长为140~258 cm,肥满度为0.77~1.26,体长(L)与体重(W)之间的关系式为W=1×10~(–5)L~(2.9658) (R~2=0.9076,n=492)。74尾中华鲟性腺发育成熟,成熟比例为15.04%,成熟个体中雄鱼体重[(60.73±14.53)kg]和体长[(172.27±13.46)cm]均小于雌鱼体重[(88.39±29.14)kg]和体长[(193.37±18.90) cm];雄鱼最小性成熟年龄为10龄,平均为(14.96±1.93)龄,雌鱼最小成熟年龄为12龄,平均为(17.84±1.80)龄。雄鱼催产成功率为76.36%,精子快速运动时间为(49.11±13.38) s,精子寿命为(220.75±56.47)s;雌鱼催产成功率为57.89%,产卵量为(13.43±6.79)万粒,卵径(3.97±0.15)mm,卵重(0.046±0.013)g,受精率为(42.72±27.82)%,孵化率为(51.61±32.41)%,出苗量为(4.44±5.67)万尾。与野生中华鲟相比,人工养殖中华鲟成熟个体体格、繁殖力和繁殖效果均有下降趋势,人工保种面临挑战。  相似文献

为了解中华鲟(Acipenser sinensis)免疫器官早期发育的形态和组织学特征,实验以1~300 dph中华鲟子二代的仔鱼、稚鱼和幼鱼为研究对象,使用连续石蜡切片技术和显微观察方法对免疫器官(头肾、胸腺和脾脏)的发育过程进行观察和记录。结果显示,培育水温为12.9~22.6℃时,中华鲟免疫器官原基出现的先后顺序为头肾(3 dph)、胸腺(7 dph)和脾脏(9 dph);免疫器官淋巴化先后顺序为胸腺(12 dph)、头肾(15 dph)和脾脏(33 dph)。仔鱼发育至15 dph可见头肾和胸腺间有淋巴细胞“桥”连接现象, 180 dph胸腺内可见哈氏小体结构,头肾、胸腺和脾脏内含有黑色素细胞或黑色素巨噬细胞中心(MMCs)。总体而言,中华鲟免疫器官发育具有原基出现时间早、发育速度慢和发育周期长的特点。由于仔稚幼鱼免疫系统发育不完善,建议在其培育过程中加强病害防治和早期疫苗的开发。  相似文献

Eight purified diets were fed to juvenile white sturgeon, Acipenser transmontanus Rick, for 9 weeks to investigate the effect of dietary lipids on the fatty acid composition of phospholipids and triglycerides from muscle, liver and brain. The diets contained 150 g kg?1 of oils from canola, corn, cod liver, lard, linseed, soybean, safflower, or a control mixture (corn oil/cod liver oil/lard, 1:1:1, by wt). Dietary lipids significantly (P≤ 05) affected the composition of tissue triglycerides and phospholipids. Tissue triglyceride fatty acid composition ranged widely, in parallel with the dietary lipids, while phospholipids changes were more conservative. Brain phospholipid fatty acid composition was less responsive to diet compared with that in muscle and liver. Considerable amounts of n-6 and n-3 long chain polyun-saturated fatty acids (> C20) were found in triglycerides and phospholipids with all diets, demonstrating that white sturgeon can desaturate and elongate linoleic acid (18:2n–6) and linolenic acid (18:3n–3). Further, the products of the Δ6 desaturase, i.e. 18:3 n–6 and 18:4n–3, were relatively abundant in triglyceride, suggesting that the Δ6 desaturase might not be a limiting step in the process in white sturgeon. Nevertheless, accumulation of both EPA and DHA was greater in the sturgeon fed fish oil than those fed linseed oil, indicating that muscle triglyceride EPA and DHA levels are best enhanced by diets rich in preformed EPA and DHA.  相似文献

为了确定中华鲟养殖基地一尾死亡子二代中华鲟的病原,利用传统病原菌分离方法,从濒死的中华鲟肝脏、脾脏和腹水中均分离到一株优势菌EM,采用16S r DNA基因序列分析、构建系统发育树并结合Biolog微生物自动分析系统进行鉴定,确定菌株EM为脑膜败血伊丽莎白菌。进一步对健康子二代达氏鲟进行人工感染试验确定其致病性;同时通过观察患病子二代中华鲟组织切片分析其病理特征;并对菌株EM进行药敏特性研究以期指导临床用药。结果显示,被感染子二代中华鲟肝脏、脾脏及肾脏组织中铁血黄素沉积、实质性细胞空泡变性,溶血严重。菌株EM人工感染健康子二代达氏鲟,发现其对达氏鲟具有致病性,且从人工感染患病的达氏鲟肝脏中再次分离到相同的病原菌。药物敏感性试验表明,中华鲟脑膜败血伊丽莎白菌对米诺环素、克林霉素、克拉霉素和麦迪霉素4种药物敏感,对四环素、头孢噻肟和头孢西叮3种药物中度敏感,对其余28种药物不敏感,呈多重耐药性。本研究首次发现脑膜败血伊丽莎白菌可感染中华鲟,并对达氏鲟也具有致病性,并初步研究了病原菌的生物学特性、病理特征和药物敏感性,以期为该病的诊断和防治提供理论依据。  相似文献
