Molecular markers in some medicinal plants of the Apiaceae family   总被引:3,自引:0,他引:3  
The internal transcribed spacers ITS1 and ITS2 in the18S-5.8S-26S rDNA repeat units were amplified and cloned from Angelica gigas Nakai, Angelica acutiloba (Siebold & Zucc.) Kitagawa, A. dahurica Maxim, Angelica decursiva (Miq.) Franch. & Savat, Bupleurum falcatum L. and Peucedanum japonicum Thunb. Sequence analyses showed that ITS1 is approx. 215 bp, the 5.8S gene is 162 bp and the ITS2 approx. 221 bp in all six species. The sequences are deposited at the EMBL Nucleotide Database. By including these new sequences in the Apiaceae phylogenetic tree, a third branch consisting of P. japonicum, A. gigas, P. decursivum and A. decursiva is added to theAngelica clade. Peucedanum does not forma distinct branch. The sequence obtained from Angelica dahurica collected in S. Korea is identical to that reported for the same species originating from China. A Bupleurum clade of three species was added to the tree showing closer relationship to theDaucus Laserpitium clade than the Angelica clade. RAPD analysis of all six species showed that the 10-base primer OPC-17 only, out of the20 Kit-C primers from Operon gave polymorphic banding patterns. Summary Isolates of three species of rust (Puccinia allii, P. mixta and Uromyces ambiguus) were each inoculated onto a range of potential host plants, selected to include the major European crop Allium species together with the commoner wild species in the section Allium. Measurements of incubation period, lesion type, latent period and pustule quantity were made in a single environment. Each isolate was able to sporulate on a range of Allium species. There was evidence of specialisation to particular sections of the genus; all the leek isolates performed well on the majority of hosts in section Allium, whilst the chive isolate grew poorly on most members of that section. Exceptions to this are exemplified by A. fistulosum being attacked by all three rust species and A. sativum being attacked by both leek and chive isolates. A continuum of reaction types was found which, for the leek isolates, ranged from the low levels of quantitative resistance in leek cultivars, through higher quantitative levels within the A. ampeloprasum complex, to qualitative resistance in several interactions in non-host sections of Allium, where the pathogen was able to infect and form colonies of some size without sporulating.  相似文献

Bananas cultivated for export all belong to Cavendish cultivars and are all recognized as very susceptible to nematodes, particularly to the burrowing nematode Radopholus similis and the lesion nematode Pratylenchus coffeae. Even if there have been many changes in the management of banana nematodes in large commercial banana plantations, chemical control still remains most often the last resort method to manage the nematodes, although the number of registered products is definitely declining. Therefore, nematode control though genetic improvement is gaining new interest worldwide. In this study, 55 banana accessions mostly diploids from the Musa acuminata genome group (AA) but including some triploid accessions (AAA), some diploids of the Musa balbisiana genome group (BB) and some interspecific hybrids (AAB, AB) were evaluated for resistance to four nematode species R. similis, P. coffeae, Meloidogyne incognita and M. arenaria. These experiments were conducted in a growth chamber under controlled conditions. All banana accessions were susceptible to nematode species, although many different levels of susceptibility were detected. This study confirmed the good resistance status to R. similis of some cultivars from the Pisang jari buaya and Pisang batuau subgroups and the partial resistance of 17 diploid accessions significantly different from the susceptible reference cv. Grande Naine. This study also showed that 12 diploid accessions exhibited a partial resistance to P. coffeae, including some usual or potential genitors belonging to the wild diploids subspecies burmannica (cvs. Long Tavoy 1 and 2) and burmannicoides (cv. Calcutta 4). No source of resistance to Meloidogyne spp. was found. These screening results, combining for the first time four nematode species, are discussed within the scope of banana breeding in order to produce parental diploid lines with single or combined nematode resistances and further develop triploids that can substitute existing susceptible commercial cultivars.  相似文献

Summary Extensive pollinations were made in attempts to produce selected intergeneric or interspecific hybrids using Nicotiana and Petunia species that exhibit potential for somatic hybridization. Pollination between pairs of species was done by standard and bud-pollination methods. Nicotiana alata or N. tabacum when pollinated reciprocally with selected Petunia species failed to produce hybrids. Likewise, no interspecific hybrids were obtained between Petunia parviflora and 4 Petunia species or P. hybrida. The type of incongruity existing between these species is discussed in relation to the production of hybrids by standard In vitro techniques as compared to somatic hybridization.On leave from the Department of Horticulture, Michigan State University, E. Spodoptera litura (Fabr.), the tobacco caterpillar, is a major defoliator on sunflower in the tropics. Genetic variability for resistance to S. litura is limited in the cultivar germplasm of sunflower. In the present investigation, 43 accessions of 17 wild Helianthus species of the annual and perennial habit groups were evaluated along with cultivated sunflower under field and no-choice conditions in the laboratory for resistance to this pest. Under field conditions, H. occidentalis and H. argophyllus were found to be immune with no leaf damage and few accessions of the species belonging to section Divaricati were found to be resistant. Laboratory bioassays against neonate, two and 4-day-old larvae confirmed resistance both in terms of high larval mortality and low larval weight gain in eight species viz., H. occidentalis, H. argophyllus, H. tuberosus, H. maximiliani, H. mollis, H. simulans, H. divaricatus and H. hirsutus. Intra-accessional variability was observed and accessions of few species showed varied reaction (resistant, partially resistant to susceptible) to the target pest. Field evaluation of 224 backcross derived inbred lines from five cross combinations involving diploid species under high natural pest incidence revealed low damage in plants derived from crosses involving H. argophyllus and H. petiolaris.  相似文献

Screening for resistance to powdery mildew of Solanum melongena and wild related species was made in the field under natural infection conditions. A total of 172 accessions originating from several geographical parts of the world were tested. Single plant selection for resistance was carried out and open-pollination was used. Most S. melongena accessions were susceptible or highly susceptible to the disease. By S0 to S3 selection, an increase in the overall powdery mildew resistance level of S. melongena population was obtained and four S. melongena lines possessing a high level of resistance were obtained. Among the wild Solanum species, S. laciniatum and S. nigrum showed to be non-host plants of L. taurica. S. quinquangolare showed no symptoms of powdery mildew, S. linnaeanum, S. aculeatissimum, S. aviculare, S. pseudocapsicum were highly resistant, S. spinosissimum was resistant, S. gilo, S. capsicoides were susceptible or highly susceptible, and plants of S. sisymbriifolium showed a widely variable disease reaction. Four S. melongena resistant lines were obtained: PAVEG 10187 S3, PAVEG 10196 S3, P.I. 230279 S3 and P.I. 419198 S3. These S. melongena lines together with the resistant wild species could be used for genetic studies, classical breeding programs and biotechnological applications.  相似文献

Diversity in 28 accessions representing 12 species of the genus, Cajanus arranged in 6 sections including 5 accessions of the cultivated species, C. cajan, and 4 species of the genus Rhyncosia available in the germplasm collection at ICRISAT was assessed using RFLP with maize mtDNA probes. Cluster analysis of the Southern blot hybridization data with 3 restriction enzymes – 3 probe combinations placed the genus Rhyncosia in a major group well separated from all the species belonging to the genus Cajanus. Within the genus Cajanus, the 4 accessions of C. platycarpus belonging to section Rhynchosoides formed a separate group in contrast to those in other sections of pigeonpea. In the section, Cajanus all the 5 accessions of C. cajan were grouped together and C. cajanifolius belonging to the same section was in a subgroup by itself closer to the main group. The four accessions of C. scarabaeoides, were together and the other species belonging to section Cantharospermum were in different subgroups. The intra-specific variation was seen even within accessions of certain pigeonpea wild species such as C. scarabaeoides, C. platycarpus, C. acutifolius, and even the cultivated species of C. cajan. This study suggests that RFLP of mtDNA can be used for the diversity analysis of pigeonpea and it gives some indications on the maternal lineage among the species. The variations in the mitochondrial DNA hybridization patterns also suggest the extensive rearrangement of the organelle genome among the Cajanus species. Summary The past 25 years, 1686 potato accessions, representing 100 species in the genus Solanum L., subgenus Potatoe, section Petota, were evaluated for field resistance to one or more of the following insect pests: green peach aphid, Myzus persicae (Sulzer); potato aphid, Macrosiphum euphorbiae (Thomas); Colorado potato beetle, Leptinotarsa decemlineata (Say); potato flea beetle, Epitrix cucumeris (Harris); and potato leafhopper, Empoasca fabae (Harris). Accessions highly resistant to green peach aphid were identified within 36 species, to potato aphid within 24 species, to Colorado potato beetle within 10 species, to potato flea beetle within 25 species, and to potato leafhopper within 39 species. Resistance levels were characteristic within Solanum species. Insect resistance appears to be a primitive trait in wild potatoes. Susceptibility was most common in the primitive and cultivated Tuberosa. Insect resistance was also characteristic of the most advanced species. The glycoalkaloid tomatine was associated with field resistance to Colorado potato beetle and potato leafhopper. Other glycoalkaloids were not associated with field resistance at the species level. Dense hairs were associated with resistance to green peach aphid, potato flea beetle, and potato leafhopper. Glandular trichomes were associated with field resistance to Colorado potato beetle, potato flea beetle, and potato leafhopper. Significant correlations between insect score and altitude of original collection were observed in six of thirteen species. Species from hot and arid areas were associated with resistance to Colorado potato beetle, potato flea beetle, and potato leafhopper. Several annual and perennial species in the genus Glycine Willd., including G. soja, long-pod G. tomentella, short-pod G. tomentella and G. tabacina, collected in Taiwan and nearby islands were studied for variations of their seed proteins. SDS-PAGE and Western blotting were used to analyze the total proteins, the heat soluble proteins, six seed maturation proteins (GmPMs) and one seed storage protein. The various species had different patterns of seed heat soluble proteins. In addition,each species of Glycine collected in Taiwan exhibited unique seed maturation protein patterns. They had several cross-reactive polypeptides recognized by specific antibodies against GmPM1, GmPM2 and GmPM8, but only one polypeptide recognized by antibodies against GmPM4, GmPM5 and MP130. The long pod G. tomentella, which has been suggested as a new species and renamed as G. dolichocarpa, could be distinct from the short pod G. tomentella on the basis of the analysis of these biochemical markers. It is also indicated that these GmPM antibodies may be used to distinguish between and within other Glycine species. summary Meiotic chromosome studies of polyploid Cucumis species revealed the presence of bivalent chromosome configurations in the tetraploid C. aculeatus and C. zeyheri and hexaploid C. figarei, while a maximum of ten quadrivalents were observed in pollen mother cells of a tetraploid species, C. heptadactylus.Allotetraploidy was not accompanied by an increase in the number of pores per pollen grain, but the autotetraploid and hexaploid species showed a relatively high percentage of 4-porate pollen. An analysis of microsporogenesis was carried out in four populations of Pfaffia tuberosa (2n=4x=68) and in ten populations of P. glomerata(2n=2x=34). Monads, dyads and triads were reported at high frequency in all populations of P. tuberosa. In P. glomerata, on the other hand, only four populations presented monads, and at low frequency. Dyads and triads were observed in both species, albeit at a higher frequency in P. tuberosa populations. Certain abnormalities of spindles in the second division, such as sequential and tripolar spindles, led to triad formation by the rejoining of two chromosome sets. The precise causes of monad and dyad formation have not been found during meiosis, although evidence exists that dyads originated by omission of second meiosis and monads by omission of both meiotic divisions. Pea would benefit from the plasticity and adaptability of its cross-incompatible relatives Pisum fulvum and Lathyrus sativus L., and we have tested reciprocal sexual crossings by manually cross-pollinating plants of genotypes of these three species. Studies of in situ germination of pollen grains on stigmata showed that pollen tubes were generally unable to germinate or could not reach the ovary. A few putative hybrid pods were nevertheless harvested, with one grain per pod germinated in vitro, then micropropagated for flow cytometry, isoenzyme, molecular (ribosomal ITS PCR-RFLP) and genomic in situ hybridization (GISH) studies. One such grain was recovered from an inter-generic cross of P. sativum x L. sativus and four from an inter-specific P. sativum x P. fulvumcross. A strong cross-incompatibility was shown between pea and grass pea, where the putative hybrid turned out to be pea. Conversely, with the interspecific, P. sativum x P. fulvum cross, flow cytometry and isoenzymes with leaf tissues strongly suggested hybridity, while molecular approaches and GISH confirmed the production of inter-specific hybrids, and without the need for a wild type P. sativum accession as a bridging cross.  相似文献

Banana hybrids with resistance to Yellow Sigatoka and Black Leaf Streak disease were evaluated for resistance to the burrowing nematode Radopholus similis and to the lesion nematode Pratylenchus coffeae in a growth chamber at 24–28°. Plants produced by tissue culture were acclimatised for 6 weeks prior to inoculation. Forty-five days after inoculation with nematodes, the root systems were processed and nematode numbers assessed. Two cultivars of Grande Naine (Musa AAA, Cavendish subgroup, ITC1256 and cv902) and one cultivar of Yangambi Km5 (Musa AAA, Ibota subgroup, ITC1123) were used respectively as susceptible and resistant controls. Results based on multiplication rates and root infestations showed that three of these hybrids (FB918, FB919 and FB924) were not significantly different from the resistant control Yangambi Km5 with a lower multiplication of R. similis. Similarly four of these hybrids (FB918, FB919, FB920, FB924) showed a lower multiplication of P. coffeae, not significantly different from the same resistant control. This is the first study that shows a partial resistance to both nematode species, R. similis and P. coffeae within synthetic hybrids of M. acuminata, adding an important extra value to these dessert banana hybrids formerly bred to resist to Mycosphaerella leaf spot diseases.  相似文献

Summary Azolla accessions (section Azolla) from the germplasm collections of the International Rice Research Institute and Washington State University were fingerprinted and classified by enzyme electrophoresis and leaf trichome morphology. A. filiculoides was enzymatically distinctive and also reliably identified by its prominent one-celled trichomes. Neotropical accessions labelled as A. filiculoides proved to be members of other species. Two groups of isolates were designated A. rubra, but those from Japan were identified as A. filiculoides. The A. rubra of Australia-New Zealand was biochemically unique and possessed less protuberant trichomes than A. filiculoides. A. microphylla, A. mexicana, and A. caroliniana were phenetically similar, but a. microphylla was identifiable from the others in the banding patterns of certain enzymes. A. mexicana and A. caroliniana were closely related enzymatically. The two-celled leaf trichomes of these three species were similar in size and shape.  相似文献

Summary A collection of subterranean clover lines singled out from populations of the species Trifolium brachycalycinum and T. subterraneum collected in Sicily, Italy, was examined at two localities for flowering time and at one locality for oestrogen content and seed yield. The structure and variation of the populations of the two species were compared. The relationship between flowering time of the populations and some environmental features of their collection sites was examined to assess whether the maturity requirements of the two species were similar and to frame selection models focused on developing varieties of appropriate maturity. Populations of T. subterraneum were, on average, more complex than those of T. brachycalycinum, being characterized by higher number of lines per population and greater intra-population variation for flowering time, oestrogen content and seed yield. Furthermore, populations of T. subterraneum were, on average, about 15 days earlier than sympatric populations of T. brachycalycinum. Both the greater variation and the relative earliness of T. subterraneum occurred irrespective of the environments of origin of the populations. Inferences are drawn on the adaptive advantages that such features confer to T. subterraneum. Mean flowering time of the populations increased on increasing annual rainfall and altitude of the collection sites. However, the changes in maturity appeared almost exclusively related to variations in rainfall in T. subterraneum, while in T. brachycalycinum the effect of altitude was greater and that of rainfall less marked than in the former species.  相似文献

Summary The compatibility between gynoecia of different penicillarian millets (Pennisetum glaucum and P. purpureum) and pollens from some other Poaceae was evaluated in order to determine whether different penicillarian pistils showed the same pistil-pollen interactions with a same non-penicillarian pollen and to determine whether compatibility was dependent on the genetic divergence or the degree of sympatric evolution of the mating partners. Ten pearl millet lines (P. glaucum subsp. glaucum), six wild pearl millet accessions (P. glaucum subsp. monodii) and one P. purpureum accession were pollinated with five other Pennisetum species: P. pedicellatum, P. polystachyon, P. schweinfurthii, P. squamulatum, P. ramosum, and with three species from other genera: Cenchrus ciliaris, Panicum maximum and Zea mays. Each male species was represented by a unique accession. Pollen grain germination on the stigmata and pollen tube growth in the gynoecia were monitored by means of fluorescence microscopy after aniline blue staining. Significant compatibility differences were observed between a given pollen and gynoecia from different P. glaucum accessions. But there was no evidence of compatibility differences between the three female taxonomic groups. The eight pollen parents can be classified into three groups according to their mean degree of compatibility with the different penicillarian gynoecia. P. ramosum, P. schweinfurthii and P. squamulatum showed a high compatibility: more than 50% of penicillarian gynoecia had pollen tubes at the micropyle, six hours after pollination. Zea mays, Cenchrus ciliaris and Panicum maximum showed a low compatibility: pollen tubes were arrested in the style. P. pedicellatum and P. polystachyon showed nil compatibility: few pollen grains germinated on the stigmata and very rare tubes reached the entry of the style, where they stopped. It must be noted that such a strong gametic barrier has been observed only with pollinator species sympatric to pearl millet.  相似文献

Summary Atylosia scarabaeoides Benth. and A. platycarpa Benth., close relatives of the species Cajanus cajan (L.) Millsp., were screened for photoperiodic response. Four photoperiods ranging from 12 h 45 min to 19 h were studied in three environments. A. scarabaeoides flowered freely only in the first photoperiod. A. platycarpa not only flowered early (39 to 63 days after planting) in all four photoperiods, but also exhibited a relatively constant vegetative phase up to 16 h of illumination. Cajanus cajan, in which most cultivars-if not all-exhibit a response to photoperiod, was successfully hybridized with the two Atylosia species. It is suggested that genes for earliness and insensitivity to day-length could be transferred from A. platycarpa to C. cajan.  相似文献

Summary Accessions from exotic Solanum species, including diploid and tetraploid species, were screened for immunity to Clavibacter michiganensis subsp. sepedonicus, the causal agent of potato ring rot. The diploid species included S. infundibuliforme, S. lesteri, S. megistacrolobum, S. tuberosum Group Phureja, S. polyadenium, S. pinnatisectum, S. raphanifolium, S. sparsipilum, S. sanctae-rosae, S. tuberosum Group Stenotomum, S. toralapanum, and S. verrucosum. The tetraploid species included S. tuberosum Group Andigena, S. acaule, S. fendleri, S. hjertingii, S. oplocense, S. polytrichon, and S. stoloniferum. Apparent immunity was initially found in several diploid species, but was not present during subsequent retesting. Immunity was found in nine accessions of tetraploid S. acaule. These accessions maintained their immunity during testing over an eight-month period. S. acaule appears to be a good source of immunity for introgression studies.  相似文献

Summary Pollen germination and pollen tube growth after self- and intra- and interspecific pollinations of annual [M. scutellata (L.) Mill., M. disciformis DC., and M. rigidula (L.) All.] and perennial (M. sativa L.) Medicago species were studied using a Carl Zeiss microscope with a fluorescence filter attachment.Self-pollination of self-fertile annual and intraspecific pollination of cross-fertile perennials were characterized by high pollen germination and rapid normal pollen tube growth. In contrast pollen germination percentages were very low and many pollen tube growth abnormalities occurred following interspecific pollination and self-pollination of self-sterile plants. The time period from pollination to fertilization for interspecific crosses was about double that following self-pollination of the annual species. However, fertilization occurred frequently following interspecific pollinations. Summary Various wild relatives of pigeonpea,Cajanus cajan, namely some species ofAtylosia andRhynchosia, possess desirable characteristics that could be utilized for effecting genetic improvement of this crop. In total 73 cross combinations among two cultivars ofC. cajan and one accession each of eightAtylosia species and one ofRhynchosia were attempted. Twelve hybrids were obtained. Seven of these were analysed for F1 fertility and their utility for agronomic improvement of theC. cajan. Fertility behaviour of the different F1 hybrids varied and indicated that potential of gene transfer between the two genera,Atylosia andCajanus, was as good as within the genusAtylosia. From F2 and F3 families ofC. cajan × A. scarabaeoides andC. cajan × A. albicans, plants were selected with greater physiological efficiency and agronomic superiority. The prospects of transferring pod borer resistance and higher seed protein content from someAtylosia species to pigeonpea are discussed.  相似文献
